Introduction Mushroom-forming fungi comprise diverse species that develop complex multicellular structures. In cultivated species, both ecological adaptation and artificial selection have driven genome evolution. However, little is known about the connections among genotype, phenotype and adaptation in mushroom-forming fungi. Objectives This study aimed to (1) uncover the population structure and demographic history of Lentinula edodes, (2) dissect the genetic basis of adaptive evolution in L. edodes, and (3) determine if genes related to fruiting body development are involved in adaptive evolution. Methods We analyzed genomes and fruiting body-related traits (FBRTs) in 133 L. edodes strains and conducted RNA-seq analysis of fruiting body development in the YS69 strain. Combined methods of genomic scan for divergence, genome-wide association studies (GWAS), and RNA-seq were used to dissect the genetic basis of adaptive evolution. Results We detected three distinct subgroups of L. edodes via single nucleotide polymorphisms, which showed robust phenotypic and temperature response differentiation and correlation with geographical distribution. Demographic history inference suggests that the subgroups diverged 36,871 generations ago. Moreover, L. edodes cultivars in China may have originated from the vicinity of Northeast China. A total of 942 genes were found to be related to genetic divergence by genomic scan, and 719 genes were identified to be candidates underlying FBRTs by GWAS. Integrating results of genomic scan and GWAS, 80 genes were detected to be related to phenotypic differentiation. A total of 364 genes related to fruiting body development were involved in genetic divergence and phenotypic differentiation. Conclusion Adaptation to the local environment, especially temperature, triggered genetic divergence and phenotypic differentiation of L. edodes. A general model for genetic divergence and phenotypic differentiation during adaptive evolution in L. edodes, which involves in signal perception and transduction, transcriptional regulation, and fruiting body morphogenesis, was also integrated here.

Background. As in philosophy of traditional Chinese medicine (TCM), the theory of “kidney governing bones” has been demonstrated by a series of scientific studies. Furthermore, many groups including ours have explored the molecular mechanisms related to bone development, growth, and regeneration using modern biology technologies, such as RNA sequencing (RNA-Seq) and isobaric tags for relative and absolute quantification (ITRAQ), and have demonstrated that the underlying molecular mechanisms were highly consistent with the “kidney governing bones” theory. Objective. Kidney-yang deficiency (YD), as a pathological condition, has a passive effect on the skeleton growth; more specifically, it is a state of skeletal metabolic disorder. However, the exact molecular mechanisms related to the “kidney governing bones” theory under the control of multiple organs and systems are still unknown. Methods. In this study, we performed RNA-Seq analysis to investigate and compare the gene expression patterns of six types of tissue (bone, cartilage, kidney, testicle, thyroid gland, and adrenal gland) from YD rats and normal rats and analyzed the interaction effects controlled by multiple functional genes and signaling pathways between those tissues. Results. Our results showed that, in the state of YD, the functions of bone and cartilage were inhibited. Furthermore, multiple organs involving the reproductive, endocrine, and urinary systems were also investigated, and our results showed that YD could cause dysfunctions of these systems by downregulating multiple functional genes and signaling pathways that positively regulate the homeostasis of these tissues. Conclusion. We ensure that “kidney governing bones” was not a simple change in a single gene but the changes in complex biological networks caused by functional changes in multiple genes. This also coincides with the holistic view of TCM, which holds that the human body itself is an organic whole and the functional activities of each organ coordinate with each other.

The objective of this study was to investigate the effects of increasing dietary total Ca/total P ratios on growth performance, digestibility of Ca and P, bone mineralization, and concentrations of Ca and P in urine and plasma in nursery pigs. There were six diets in a randomized complete block design, including one positive control and five diets corresponding to five total Ca/total P ratios: 0.55, 0.73, 0.90, 1.07, and 1.24 (analyzed as 0.58, 0.75, 0.93, 1.11, and 1.30). These five diets were deficient in P but supplemented with 1,000 phytase units/kg feed. Each diet was fed to six pens of eight pigs (four barrows and four gilts per pen). All diets contained 3 g/kg TiO2, and fecal samples were collected from each pen on days 5–7 of trial. At the end, one pig per pen was sacrificed to collect the right tibia and urine in the bladder. The results showed that increasing dietary Ca/P ratio to 0.93 increased gain:feed but then gain:feed decreased as the Ca/P ratio was increased to 1.30 (linear and quadratic, P < 0.05). Although average daily gain and final BW were unaffected by changing Ca/P ratio in diet, dry bone weight; weights of bone ash, Ca and P; and bone Ca/P ratio increased linearly (P < 0.001) with increasing dietary Ca/P ratio. The percent bone Ca showed a tendency to increase (P = 0.064). Increasing dietary Ca/P ratio decreased apparent total tract digestibility (ATTD) of Ca and P linearly (P < 0.05) and the concentration of digestible P linearly (P < 0.001), but increased the concentration of digestible Ca (linear and quadratic effects: P < 0.01) and the digestible Ca/P ratio (linear effect: P < 0.001). In plasma, the concentration of Ca increased both linearly (P < 0.01) and quadratically (P = 0.051), whereas the concentration of P tended (linear and quadratic, P < 0.10) to decrease with increasing dietary Ca/P ratio. Similarly, in urine, the concentration of Ca increased both linearly and quadratically (P < 0.05), whereas the concentration of P decreased linearly (P < 0.01). In conclusion, increasing the dietary Ca/P ratio reduced feed efficiency but increased bone mass and the amounts of Ca and P deposited in bone of nursery pigs fed diets supplemented with 1,000 FYT/kg phytase. The increases in bone growth led to a reduction of urinary P excretion that exceeded the decreased digestible P supplied in diet with the widening dietary Ca/P ratios.

The mixed application of pesticides and foliar fertilizer has been widely used in the production of cucumber, however, their effects on plant growth and pesticide dissipation are still unclear. In this study, the effects of mixed application of pymetrozine, tebuconazole and foliar fertilizer on the cucumber plant growth and pesticide dissipation were investigated simultaneously. The results show that the mixed use of pymetrozine, tebuconazole, especially adding foliar fertilizer, improved the physiological indexes (i.e., area, nitrogen content and chlorophyll content of the leaves, and root growth) of cucumber plants compared to those with the application of single pesticide. Meanwhile, it can significantly affect the dissipation of pymetrozine even in the slower growth matrices (lower leaves, stems, and plants). The residue of tebuconazole in cucumber plants was affected by the combination of formulation type and foliar fertilizer. This study can provide data for scientifically guiding the mixed application of pesticide and fertilizer.

As a non-destructive technology for detection, hyperspectral imaging has potential for phenotyping plant diseases and providing information for plant protection. Currently, most studies based on hyperspectral imaging technology primarily focus on leaf diseases and less on stalk diseases. In this study, a set of automatic detection and diagnosis methods, combined with spectral and image analyses, was proposed for detecting a stalk disease, rice sheath blight (Rhizoctonia solani), which is widely distributed and deleterious to yield and quality of rice. This study proposed a stepwise method for detecting rice sheath blight. The procedure starts from removal of non-plant background using the k-means clustering algorithm. Then the rice anomalous regions are identified by applying Fisher linear discrimination on sensitive bands. Finally, the scabs of rice sheath blight are detected through a newly developed approach called Hyperspectral Feature Profile Scanning-based Scab Detection (HFPSSD). The validation results showed that the proposed method can effectively recognise disease scabs. The overall accuracy reached 98.42% at pixel level and 95.92% at patch level, which outperformed the traditional support vector machines (SVM) algorithm. The proposed method can potentially serve as a tool for high-throughput detection of plant stalk diseases in the field.

In the present work, we systematically studied in-situ generation and detection of single anionic ionosomes via SECE combined with a miniaturized interface between two immiscible electrolyte solutions (ITIES). Meta-lens has successfully been developed for a variety of optical functions. We demonstrate a light-field edge detection imaging system with a gallium nitride achromatic meta-lens array. It enables edge detection from one dimension to three dimensions. The designed meta-lens array consists of 60 by 60 achromatic meta-lenses, which operate in the visible range from 400 to 660 nm. All of the light field information of objects in the scene can be captured and computed. The focused edge images from one dimension to three dimensions are extracted with depth estimation by image rendering. Three dimensions edge detection is two dimensions edge imaging with depth information. The focused edge images can be obtained by the sub-image reconstruction of the light field image. Our multidimensional edge detection system by achromatic meta-lens array brings novel advantages, such as broadband detection, data volume reduction, and device miniaturization capacity. Results of our experiments show new insight into applications of biological diagnose and robotic vision.

In this paper, chronic inflammatory cell infiltration was seen during this period. The reason for the false-positive result may be the atypical proliferation of cells caused by chronic inflammatory stimulation with malignant cytology. 12 cases of false negative results, including 2 cases of type I nodules, 10 cases of type III nodules, postoperative histology and pathology were papillary carcinoma, there are many reasons for false negative results, including the size of the nodules, the puncture operator, the technical level, internal tissue composition and properties of the nodules all affect the accuracy of puncture and cytopathology results. The satisfaction of puncture specimens of each group was calculated and compared. For thyroid nodules with the maximum diameter ≤5 mm, the satisfaction rate of puncture specimens was 87.2%, and the diagnostic sensitivity and accuracy of ultras guided fine needle aspiration biopsy were relatively low. Cytological results were in poor agreement with postoperative histological and pathological results, so it was recommended to take active follow-up for thyroid nodules with the maximum diameter ≤5 mm to reduce unnecessary puncture. The overall puncture success rate is relatively high, ≤10 mm low-echo nodules or even ≤5 mm nodules can be cytological puncture, especially the kind of close to the capsule, trachea and recurrent laryngeal nerve nodules, for the largest diameter of >5 mm, the satisfaction rate of puncture specimens is relatively high is 93.2%. Moreover, the sensitivity and accuracy of ultrasound guided fine needle aspiration biopsy were relatively high, and the cytological results were in good agreement with the postoperative histological and pathological results. Therefore, it was feasible to confirm the diagnosis of suspected malignant nodules by ultrasound. The diagnostic in each group and the consistency between cytological diagnosis and histological diagnosis were analyzed by comparing the histological and pathological results after surgery. Ultrasound-guided fine needle aspiration biopsy can be used more effectively in clinical practice.

Collisional electrochemistry between single particles and a biomimetic polarized micro-liquid/liquid interface has emerged as a novel and powerful analytical method for measurements of single particles. Using this platform, rapid detection of liposomes at the single particle level is reported herein. Individual potassium, sodium, or protonated dopamine-encapsulated (pristine or protein-decorated) liposomes collide and fuse with the polarized micro-liquid/liquid interface accompanying the release of ions, which are recorded as spike-like current transients of stochastic nature. The sizing and concentration of the liposomes can be readily estimated by quantifying the amount of encapsulated ions in individual liposomes via integrating each current spike versus time and the spike frequency, respectively. We call this type of nanosensing technology "Faradaic counter". The estimated liposome size distribution by this method is in line with the dynamic light scattering (DLS) measurements, implying that the quantized current spikes are indeed caused by the collisions of individual liposomes. The reported electrochemical sensing technology may become a viable alternative to DLS and other commercial nanoparticle analysis systems, for example, nanoparticle tracking analysis.

Clinical treatment of Osteoarthritis (OA) remains a challenge due to the poor self-regeneration ability of cartilage. Deer antler is the only cartilage tissue that can completely regenerate each year. Insulin-like growth factor 1 (IGF-1) is one of the major active components in the deer antler that participate in regulating the rapid regeneration of deer antler cartilage. This has led us to speculate that deer IGF-1 might potentially become a candidate drug for reducing damage and inflammation of OA. Thus, we aimed to explore the underlying mechanism of deer IGF-1 in chondrocyte proliferation, differentiation, and inflammation response. Deer, mouse, and human IGF-1 amino acid sequences and protein structures were aligned using CLUSTAL and PSIPRED. The underlying molecular mechanism of deer IGF-1 on primary chondrocytes was investigated by RNA-sequencing (RNA-seq) technology combined with various experiments. Cytokine interleukin-1β (IL-1β) was used to induce the inflammation response of primary chondrocytes. We found that deer IGF-1 was more similar to human IGF-1 than mouse IGF-1. qRT-PCR and immunofluorescence assay indicated that deer IGF-1 had stronger effects than mouse IGF-1. We also found that the deer IGF-1 enhanced the expression of cell proliferation, differentiation, and extracellular matrix (ECM)-related genes, but decreased the expression of ECM-degrading genes. Deer IGF-1 also attenuated the IL-1β-induced inflammatory and ECM degradation in chondrocytes. This study provides insight into the molecular mechanisms of deer IGF-1 on primary chondrocyte viability and presents a candidate for combatting inflammatory responses in OA development.

The objective of this study was to investigate the effects of two dietary total Ca/P ratios on available P release by phytase, measured using growth performance and bone mineralization with 528 barrows and gilts according to a randomized complete block design. Three were 11 diets in a factorial of 2 by 4 plus 3, including 3 reference diets consisting of 0.25% (control), 0.70%, or 1.15% monocalcium phosphate (MCP) and 8 diets from combining 4 phytase doses (500, 1,000, 2,000, and 3,000 FYT/kg) with 0.25% MCP and 2 dietary Ca/P ratios (1.05 and 1.20). Each diet was fed to 6 pens of 8 pigs. All diets contained 3 g/kg TiO2, and fecal samples were collected from each pen on d 13–15 of trial. At the end of trial, one pig per pen was sacrificed to collect a tibia and urine in the bladder. The results showed that MCP improved growth performance linearly (P < 0.01), whereas both a linear and quadratic response was observed with the addition of phytase. The MCP increased the percent bone ash and weights of bone ash, Ca, and P linearly (P < 0.01). At both Ca/P ratios, increasing supplementation of phytase increased the percent bone ash and weights of bone ash, Ca, and P both linearly and quadratically (P < 0.05). Both MCP and phytase significantly increased digestibility of Ca and P as well as digestible Ca and P in diets and reduced the digestible Ca/P ratio. The dietary Ca/P ratio of 1.20 resulted in poorer feed utilization efficiency, more digestible Ca, greater percent bone ash, Ca, and P and heavier weights of bone Ca and P than the ratio of 1.05 (P < 0.05). The ratio of 1.20 elicited numerically higher available P release values from phytase, with percent bone ash and bone P weight as the response variables, but significantly lower values with gain:feed. The urinary concentration of Ca increased linearly (P < 0.01) with increasing digestible Ca/P ratios whilst urinary concentration of P decreased quadratically (P < 0.01). In conclusion, fixing the same total Ca/total P ratio in diets supplemented with increasing phytase dosing created an imbalance of digestible Ca and P, which could have an adverse effect on bone mineralization and thus compromise the phytase efficacy relative to mineral P.
