Anabarella, a conspicuous taxon of early mollusc, is widely distributed in the early Cambrian strata and is considered an important link in the evolutionary lineage that reflects a transitional form from helcionelloids to bivalves. In South China, Anabarella has mainly been documented from Yunnan, Shaanxi, Sichuan, and Hubei provinces. However, the taxonomy of Anabarella is questionable, which has implications for the interpretation of the genus’ temporal and spatial distribution. New and abundant well-preserved specimens of the helcionelloid mollusc Anabarella were recovered from the Member 5 of the Yanjiahe Formation in the Three Gorges area. Through morphological study, these specimens can be definitely identified as A. plana. On the basis of this new material, the species of Anabarella previously reported in the literature from South China were taxonomically revised, and, with the exception of A. plana from the Yanjiahe Formation, other species should be assigned to Igorella. Therefore, at present, A. plana is the only valid species of the genus Anabarella in South China and is limited to Cambrian Stage 2. Study of the available specimens of A. plana reveal three types of microstructures: convex polygonal impressions, concave polygons, and lamello-fibrillar microstructure. In addition, the thicker shell of the sub-apical area and the three different structures of the sub-apical area provide more evidence that A. plana might have adapted a semi-infanual mode of life and indicate that Anabarella is a likely ancestor of Watsonella.

Hexangulaconulariids, a family of biradially symmetrical medusozoan cnidarians, have been widely reported from the Lower Cambrian of South China. The four currently recognized genera of hexangulaconulariids differ from each other mainly in the number of faces in the abapical region of the periderm. However, previously published illustrations of the monospecific type genus, Hexangulaconularia, clearly show two distinct morphotypes, one with six faces and the other with 10. Specimens with 10 faces are herein reassigned to the genus Decimoconularia. In addition, the new species D. anisfacialis is described from the Kuanchuanpu Formation (Cambrian Fortunian Stage) in the Kuanchuanpu and Shizhonggou sections in Ningqiang County, southern Shaanxi Province, China. Also described are additional specimens of H. formosa from the same formation in the Zhangjiagou section in Xixiang County, southern Shaanxi Province, and from Member 2 of the Yanjiahe Formation (Cambrian Fortunian Stage) in western Hubei Province. The discovery of D. anisfacialis extends the known stratigraphical range of Decimoconularia, now composed of two species, downward from Cambrian Stage 2 into the Fortunian Stage. Additionally, certain specimens previously assigned to H. formosa are reassigned to D. anisfacialis. The diagnoses of Hexangulaconularia, Decimoconularia, and Hexangulaconulariidae are emended accordingly. In accordance with the rule of time priority, the previously designated type genus and species, Hexaconularia He and Yang, 1986 and Hexaconularia sichuanensis He and Yang, 1986, are replaced herein by Hexangulaconularia He, in Xing et al., 1983.

The repair of critical-sized bone defects (CSBD) remains a significant challenge in clinical practice. Personalized 3D porous scaffolding is generally considered an ideal approach for directed regeneration of CSBD, which usually has irregular and complex 3D geometry. In this study, we designed a customized porous scaffold rabbit radius based on a body-centered cubic unit. Borate bioactive glass/polycaprolactone (BBG/PCL) composite scaffolds with different BBG contents (0%, 5%, 10%, 20%, and 40%) were fabricated using selective laser sintering. The pore geometry, porosity, mechanical strength, hydrophilicity, protein absorption, degradation behavior, in vitro cytocompatibility, and osteogenesis differentiation of the composite scaffolds were systematically characterized. The in vivo biological properties of the BBG/PCL composite scaffold for CSBD repair were assessed using a rabbit foreleg radius defect model. The results indicate that the BBG/PCL composite scaffold with 20% BBG content effectively promoted the proliferation of osteoblasts and repaired the rabbit radius defects. The mechanical strength of the scaffold was sufficient to maintain the integrity of the scaffold structure during a relatively long CSBD repair. Our results indicate that the BBG/PCL composite scaffold can be anticipated to be a promising biomaterial for CSBD repair.

Abstract Background The radiation of ecdysozoans (moulting animals) during the Cambrian gave rise to panarthropods and various groups of worms including scalidophorans, which played an important role in the elaboration of early marine ecosystems. Although most scalidophorans were infaunal burrowers travelling through soft sediment at the bottom of the sea, Selkirkia lived inside a tube. Results We explore the palaeobiology of these tubicolous worms, and more generally the origin and evolutionary significance of tube-dwelling in early animals, based on exceptionally preserved fossils from the early Cambrian Chengjiang Lagerstätte (Stage 3, China) including a new species, Selkirkia transita sp. nov. We find that the best phylogenetic model resolves Selkirkia as a stem-group priapulid. Selkirkia secreted a protective cuticular thickening, the tube, inside which it was able to move during at least part of its life. Partly based on measured growth patterns, we construe that this tube was separated from the trunk during a moulting process that has no direct equivalent in other scalidophorans. Although the ontogeny of Selkirkia is currently unknown, we hypothesize that its conical tube might have had the same ecological function and possibly even deep development origin as the lorica, a protective cuticular thickening found in larval priapulids and adult loriciferans. Selkirkia is seen as a semi-sedentary animal capable of very shallow incursions below the water/sediment interface, possibly for feeding or during the tube-secreting phase. Brachiopod epibionts previously reported from the Xiaoshiba Lagerstätte (ca. 514 Ma) also presumably occur in Selkirkia sinica from Chengjiang (ca. 518 Ma). Conclusions Our critical and model-based approach provides a new phylogenetic framework for Scalidophora, upon which to improve in order to study the evolution of morphological characters in this group. Tube-dwelling is likely to have offered Selkirkia better protection and anchoring to sediment and has developed simultaneously in other Cambrian animals such as hemichordates, annelids or panarthropods. Often lost in modern representatives in favour of active infaunal lifestyles, tube-dwelling can be regarded as an early evolutionary response of various metazoans to increasing environmental and biological pressure in Cambrian marine ecosystems.

The chicken gastrointestinal tract has a diverse microbial community. There is increasing evidence for how this gut microbiome affects specific molecular pathways and the overall physiology, nervous system and behavior of the chicken host organism due to a growing number of studies investigating conditions such as host diet, antibiotics, probiotics, and germ-free and germ-reduced models. Systems-level investigations have revealed a network of microbiome-related interactions between the gut and state of health and behavior in chickens and other animals. While some microbial symbionts are crucial for maintaining stability and normal host physiology, there can also be dysbiosis, disruptions to nutrient flow, and other outcomes of dysregulation and disease. Likewise, alteration of the gut microbiome is found for chickens exhibiting differences in feather pecking (FP) behavior and this alteration is suspected to be responsible for behavioral change. In chickens and other organisms, serotonin is a chief neuromodulator that links gut microbes to the host brain as microbes modulate the serotonin secreted by the host’s own intestinal enterochromaffin cells which can stimulate the central nervous system via the vagus nerve. A substantial part of the serotonergic network is conserved across birds and mammals. Broader investigations of multiple species and subsequent cross-comparisons may help to explore general functionality of this ancient system and its increasingly apparent central role in the gut-brain axis of vertebrates. Dysfunctional behavioral phenotypes from the serotonergic system moreover occur in both birds and mammals with, for example, FP in chickens and depression in humans. Recent studies of the intestine as a major site of serotonin synthesis have been identifying routes by which gut microbial metabolites regulate the chicken serotonergic system. This review in particular highlights the influence of gut microbial metabolite short chain fatty acids (SCFAs) on the serotonergic system. The role of SCFAs in physiological and brain disorders may be considerable because of their ability to cross intestinal as well as the blood-brain barriers, leading to influences on the serotonergic system via binding to receptors and epigenetic modulations. Examinations of these mechanisms may translate into a more general understanding of serotonergic system development within chickens and other avians.

Abstract A coupling dynamic model of a subway train and an embedded track is established to study the safety limits of track irregularities. The simulated vehicle system was a 74-degrees of freedom multi-rigid body model, and the rail was a Timoshenko beam. The slab was a three-dimensional solid finite element model. The sensitive wavelength irregularity was first studied, and then the safety limit of the sensitive wavelength was analyzed. The wheel-rail lateral force exhibited a substantial effect on the track alignment and gauge irregularity safety limit. The wheel-rail vertical force and the rate of wheel load reduction significantly affected the height and cross-level irregularity safety limit. The results demonstrate that the safety limits of the alignment, gauge, height, and cross-level embedded track geometric irregularity are 5.3 mm, [− 10.5, 8] mm, 5.6 mm, and 6 mm, respectively.

Sb2S3 is a promising photovoltaic absorber with appropriate bandgap, excellent light absorption coefficient and great stability. However, the power conversion efficiency (PCE) of Sb2S3 planar thin film solar cells is unsatisfactory for further commercial application due to low crystallinity and high resistivity of Sb2S3 film. Here, we introduce an additive of 4-Chloro-3-nitrobenzenesulfonyl Chloride (CSCl) to alleviate these problems. The CSCl molecular contains two terminal Cl with lone pair electrons, which have the interaction with Sb atoms. Thus, the Sb2S3 film with enhanced crystallization and low trap states has been obtained and the resistivity is also decreased. Furthermore, CSCl additive raises the Fermi level of the Sb2S3 film, thereby enhancing the transport of electron from Sb2S3 to TiO2. Consequently, the optimal PCE of Sb2S3 solar cells is raised from 4.20% (control device) to 5.84%. Our research demonstrates a novel additive to enhance the photoelectric performance of Sb2S3 solar cells.

The presence of numerous trap states on the perovskite surface severely affects the performance of inorganic CsPbI2Br perovskite solar cells. Surface modification has been proven to be an effective strategy to passivate the surface trap states of CsPbI2Br perovskite. However, most modifiers behave high volatility and insulation, not enough to further develop the CsPbI2Br solar cells. Herein, an ionic liquid of 1-viny-3-propionate ethyl imidazolium chloride ([PEVIM]Cl) is applied to modify the CsPbI2Br film surface, yielding a compact film with enhanced crystallinity. The surface trap states of CsPbI2Br film are effectively passivated via the interaction between carbonyl group of [PEVIM]Cl and uncoordinated metal cations of CsPbI2Br perovskite, leading to charge recombination suppression and charge transport enhancement. Consequently, the power conversion efficiency (PCE) of [PEVIM]Cl modified CsPbI2Br device is obviously enhanced from 12.49% to 14.19% with an improved open-circuit voltage of 1.160 V. Moreover, the non-encapsulated device presents excellent thermal stability, still maintaining 91% PCE when heated at 85 °C in nitrogen atmosphere for 360 h. Meanwhile, the non-encapsulated device degrades only 11% PCE after stored at 50% relative humidity for 960 h. This simple and efficient approach provides a promising direction to fabricate high-efficiency and stable inorganic perovskite devices.

Decabromodiphenyl ethane (DBDPE), a widely used novel brominated flame retardant, is gaining concerns due to rapidly increased contents in various environmental and biota samples. In the present study, zebrafish (Danio rerio) embryos were exposed to 2.91, 9.71, 29.14 and 97.12 μg/L of DBDPE until 120 h post-fertilization (hpf) to investigate the potential developmental neurotoxicity and underlying mechanisms. Chemical analysis revealed concentration-dependently increased body burdens of DBDPE in zebrafish larvae, with bioaccumulation factors (BCFs) ranging from 414 to 726. Embryonic exposure to DBDPE caused hyperactivity without affecting the development of secondary motoneuron axons and muscle fibers. However, further results implicated that DBDPE may affect the locomotor regulatory network via different mechanisms at lower and higher concentrations. On the one hand, embryonic exposure to 2.91 μg/L DBDPE transiently promoted spontaneous coiling contractions, but showed no effects on touch-response and swimming activity in zebrafish larvae. The whole-body contents of neurotransmitters were significantly decreased. Significant decreased protein abundances of α1-TUBULIN and SYN2a and molecular docking results pointed out possible interactions of DBDPE with these two proteins. However, these changes may be unconcerned with the transient hyperactivity, and the exact molecular mechanisms need further investigation. On the other hand, 29.14 and 97.12 μg/L DBDPE exposure caused longer-lasting effects in promoting spontaneous coiling contractions, and also touch-response and swimming activity. At the same time, increased ACh contents (without changes of other neurotransmitters) and ChAT activity and inhibited transcription of nAChRs were observed at higher concentrations. Molecular docking indicated direct interaction of DBDPE with ChAT. The results suggested that DBDPE induced hyperactivity at higher concentrations was probably involved with disrupted cholinergic system, with ChAT as a potential target. Given that the body burden of DBDPE in lower concentration group was comparable with those detected in wild fish, the current results may provide useful information for ecological risk assessment.

Rapid development of aquaculture industry and increasing demand of various inputs (especially antibiotics), are suspected to promote the occurrence and spread of ARGs in aquaculture related environments. However, the occurrences of ARGs under different freshwater aquaculture practices are rarely known. Here, we investigated the seasonal profiles of the main ARGs, intI1 and bacteria in waters from three kinds of predominant freshwater aquaculture practices around the Honghu Lake (China), as well as their co-occurrences and interrelationships with antibiotics, heavy metals and general water quality. The results indicate that quinolone resistance genes (qnrB), tetracycline resistance genes (tetB and tetX) and sulfonamide resistance genes (sul1 and sul2) were the top five predominant ARGs with seasonal variations of abundance. Fish ponds were of the highest absolute abundances of tested ARGs than the other two modes. Crayfish ponds and their adjacent ditches shared similar ARGs profile. Different subtypes of ARGs belonging to the same class of resistance were varied in abundances. Some bacteria were predicted to carry different ARGs, which indicating multi-antibiotic resistances. Moreover, the combined environmental factors (antibiotics, heavy metals and water quality) partially shaped the profiles of ARGs and bacteria composition. Overall, this study provides new comprehensive understanding on the characterization of ARGs contamination in different freshwater aquaculture practices from the perspectives of environmental chemistry, microbiology and ecology. The results would benefit the optimization of aquaculture practices toward environmental integrity and sustainability.

The heat treatment process is a vital step for manufacturing high-speed railway spring fasteners. In this study, orthogonal experiments were carried out to obtain reliable optimised heat treatment parameters through a streamlined number of experiments. Results revealed that a better comprehensive mechanical performance could be obtained under the following combination of heat treatment parameters: quenching temperature of 850 °C, holding time of 35 min, medium of 12% polyalkylene glycol (PAG) aqueous solution, tempering temperature of 460 °C, and holding time of 60 min. As one of the most important testing criteria, fatigue performance would be improved with increasing strength. Additionally, a high ratio of martensite to ferrite is proven to improve the fatigue limit more significantly. After this heat treatment process, the metallographic microstructure and mechanical properties satisfy the technical requirements for the high-speed railway practical operation. These findings provide a valuable reference for the practical forming process of spring fasteners.

This paper presents an in-depth study of the variable reference process to improve the organization and properties of selective laser melting TC4 specimens. A relationship equation between body energy density and stratification is proposed. This study aimed to look into how layer and volume energy density affect the surface, tensile characteristics, and microstructure of specimens. The test findings demonstrated that specimen densities rise as the power index falls, with the most excellent density reaching 99.42%. The number of secondary α’ phases declined as the energy density of the laminae slowed down. The tensile strength of these specimens reached 1190.84 MPa, and the yield strength came at 1103.87 MPa with the same interval of variable laser power. This offers a fresh avenue for research into SLM to enhance the specimens’ overall performance.

Infectious bone defects present a major challenge in the clinical setting currently. In order to address this issue, it is imperative to explore the development of bone tissue engineering scaffolds that are equipped with both antibacterial and bone regenerative capabilities. In this study, we fabricated antibacterial scaffolds using a silver nanoparticle/poly lactic-co-glycolic acid (AgNP/PLGA) material via a direct ink writing (DIW) 3D printing technique. The scaffolds’ microstructure, mechanical properties, and biological attributes were rigorously assessed to determine their fitness for repairing bone defects. The surface pores of the AgNPs/PLGA scaffolds were uniform, and the AgNPs were evenly distributed within the scaffolds, as confirmed via scanning electron microscopy (SEM). Tensile testing confirmed that the addition of AgNPs enhanced the mechanical strength of the scaffolds. The release curves of the silver ions confirmed that the AgNPs/PLGA scaffolds released them continuously after an initial burst. The growth of hydroxyapatite (HAP) was characterized via SEM and X-ray diffraction (XRD). The results showed that HAP was deposited on the scaffolds, and also confirmed that the scaffolds had mixed with the AgNPs. All scaffolds containing AgNPs exhibited antibacterial properties against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). A cytotoxicity assay using mouse embryo osteoblast precursor cells (MC3T3-E1) showed that the scaffolds had excellent biocompatibility and could be used for repairing bone tissue. The study shows that the AgNPs/PLGA scaffolds have exceptional mechanical properties and biocompatibility, effectively inhibiting the growth of S. aureus and E. coli. These results demonstrate the potential application of 3D-printed AgNPs/PLGA scaffolds in bone tissue engineering.

The fracture of viscoplastic materials is a complex process due to its time-dependent and plastic responses. Numerical simulation for fractures plays a significant role in crack prediction and failure analysis. In recent years, the phase field model has become a competitive approach to predict crack growth and has been extended to inelastic materials, such as elasto-plastic, viscoelastic and viscoplastic materials, etc. However, the contribution of inelastic energy to crack growth is seldom studied. For this reason, we implement the fracture phase field model coupled with a viscoplastic constitutive in a finite element framework, in which the elastic energy and inelastic energy are used as crack driving forces. The implicit algorithm for a viscoplastic constitutive is presented; this procedure is suitable for other viscoplastic constitutive relations. The strain rate effect, creep effect, stress relaxation effect and cyclic loading responses are tested using a single-element model with different inelastic energy contributions. A titanium alloy plate specimen and a stainless-steel plate specimen under tension are studied and compared with the experimental observations in the existing literature. The results show that the above typical damage phenomenon and fracture process can be well reproduced. The inelastic energy significantly accelerates the evolution of the phase field of viscoplastic materials. For cyclic loadings, the acceleration effect for low frequency is more significant than for high frequency. The influence of the weight factor of inelastic energy β on the force-displacement curve mainly occurs after reaching the maximum force point. With the increase of β, the force drops faster in the force-displacement curve. The inelastic energy has a slight effect on the crack growth paths.

In this study, the microstructure and performance of newly designed dual-phase steel (DP590) after joining by flash butt welding (FBW) for vehicle wheel rims was analysed and compared by two simulations, i.e., physical simulation and numerical simulation, due to the high acceptance of these two methodologies. Physical simulation is regarded as a thermal–mechanical solution conducted by the Gleeble 3500 simulator and which can distribute the heat-affected zone (HAZ) of the obtained weld joint into four typical HAZs. These are coarse-grained HAZ, fine-grained HAZ, inter-critical HAZ and sub-critical HAZ. A combination of ferrite and tempered martensite leads to the softening behaviour at the sub-critical HAZ of DP590, which is verified to be the weakest area, and influences the final performance due to ~9% reduction of hardness and tensile strength. The numerical simulation, relying on finite element method (FEM) analysis, can distinguish the temperature distribution, which helps us to understand the relationship between the temperature distribution and real microstructure/performance. Based on this study, the combination of physical and numerical simulations can be used to optimise the flash butt welding parameters (flash and butt processes) from the points of temperature distribution (varied areas), microstructure and performance, which are guidelines for the investigation of flash butt welding for innovative materials.

Inwardly rectifying potassium (Kir) channels make it easier for K+ to enter into a cell and subsequently regulate cellular biological functions. Kir5.1 (encoded by KCNJ16) alone can form a homotetramer and can form heterotetramers with Kir4.1 (encoded by KCNJ10) or Kir4.2 (encoded by KCNJ15). In most cases, homomeric Kir5.1 is non-functional, while heteromeric Kir5.1 on the cell membrane contributes to the inward flow of K+ ions, which can be regulated by intracellular pH and a variety of signaling mechanisms. In the form of a heterotetramer, Kir5.1 regulates Kir4.1/4.2 activity and is involved in the maintenance of nephron function. Actually, homomeric Kir5.1 may also play a very important role in diseases, including in the ventilatory response to hypoxia and hypercapnia, hearing impairment, cardiovascular disease and cancer. With an increase in the number of studies into the roles of Kir channels, researchers are paying more attention to the pathophysiological functions of Kir5.1. This minireview provides an overview regarding these Kir5.1 roles.

Abstract Grain boundary (GB) dynamics are largely controlled by the formation and motion of disconnections (with step and dislocation characters) along with the GB. The dislocation character gives rise to shear coupling; i.e. the relative tangential motion of two grains meeting at the GB during GB migration. In a polycrystal, the shear coupling is constrained by the presence of other grains and GB junctions, which prevents large-scale sliding of one grain relative to the other. We present continuum equations of motion for GBs that is based upon the underlying disconnection dynamics and accounts for this mechanical constraint in polycrystals. This leads to a reduced-order (zero-shear constrained) model for GB motion that is easily implemented in a computationally efficient framework, appropriate for the large-scale simulation of the evolution of polycrystalline microstructures. We validated the proposed reduced-order model with direct comparisons to full multi-disconnection mode simulations.

Three-dimensional porous polycaprolactone (PCL) bone scaffolds prepared by selective laser sintering (SLS) have demonstrated great potential in the repair of non-load-bearing bone defects. The microgeometry and surface roughness of PCL scaffolds during the SLS process may change the biocompatibility and bioactivity of the scaffolds. However, in addition to the widely concerned mechanical properties and structural accuracy of scaffolds, there is still a lack of systematic research on how SLS process parameters affect the surface roughness of PCL scaffolds and the relationship between roughness and biocompatibility of scaffolds. In this study, we use the energy density model (EDM) combined with the thermodynamic properties of PCL powder to calculate the energy density range (Ed1–Ed3) suitable for PCL sintering. Five PCL scaffolds with different laser powers and scanning speeds were prepared; their dimensional accuracy, mechanical strength, and surface properties were comprehensively evaluated, and the bioactivities were compared through the attachment and proliferation of MC3T3-E1 cells on the scaffolds. It was found that the high energy density (Ed3) reduced the shape fidelity related to pore size and porosity, and the dense and smooth surface of the scaffolds showed poor cytocompatibility, while the low energy density (Ed1) resulted in weak mechanical properties, but the rough surface caused by incomplete sintered PCL particles facilitated the cell adhesion and proliferation. Therefore, the surface roughness and related biocompatibility of PCL bone scaffolds should be considered in energy-density-guided SLS parameter optimization.

Due to extensive COVID-19 prevention measures, millions of tons of chemicals penetrated into natural environment. Alterations of human viruses in the environment, the neglected perceiver of environmental fluctuations, remain obscure. To decipher the interaction between human viruses and COVID-19 related chemicals, environmental samples were collected on March 2020 from surroundings of designated hospitals and receivers of wastewater treatment plant effluent in Wuhan. The virus community and chemical concentration were respectively unveiled in virtue of virome and ultra-high-performance liquid chromatography-tandem mass spectrometry. The complex relationship between virus and chemical was ulteriorly elaborated by random forest model. As an indicator, environmental viruses were corroborated to sensitively reflect the ecological disturbance originated from pandemic prevention supplies. Chemicals especially trihalomethanes restrained the virus community diversity. Confronting this adverse scenario, Human gammaherpesvirus 4 and Orf virus with resistance to trihalomethanes flourished while replication potential of Macacine alphaherpesvirus 1 ascended under glucocorticoids stress. Consequently, human viruses lurking in the environment were actuated by COVID-19 prevention chemicals, which was a constant burden to public health in this ongoing pandemic. Besides, segments of SARS-CoV-2 RNA were detected near designated hospitals, suggesting environment as a missing link in the transmission route. This research innovatively underlined the human health risk of pandemic prevention supplies from the virus - environment interaction, appealing for monitoring of environmental viruses in long term.

Abstract Background In view of the ongoing coronavirus disease (COVID-19) pandemic, it remains unclear whether the severity of illness and time interval from symptom onset to release from quarantine differ between cases that originated from clusters and cases reported in other areas. This study aimed to assess epidemiological and intergenerational clinical characteristics of COVID-19 patients associated with cluster outbreaks to provide valuable data for the prevention and control of COVID-19. Methods We identified the first employee with COVID-19 at a supermarket and screened the close contacts of this index patient. Confirmed cases were divided into two groups according to the generation (first generation comprising supermarket employees [group A] and second or third generations comprising family members or friends of the supermarket employees [group B]). The epidemiological and clinical characteristics of the two groups were retrospectively compared. Results A total of 8437 people were screened, and 24 COVID-19 patients were identified. Seven patients (29.2%) were asymptomatic; three patients were responsible for six symptomatic cases. The interval from the confirmation of the first case to symptom onset in symptomatic patients was 5–11 days. The clinical manifestations of symptomatic patients upon admission were non-specific. All patients (including the seven asymptomatic patients) were admitted based on chest computed tomography features indicative of pneumonia. There were 11 cases in group A (first generation) and 13 cases in group B (second generation, 11 cases; third generation, 2 cases), with no significant differences in clinical and epidemiological characteristics between the two groups, except for sex, duration from symptom onset to hospitalization, and underlying disease (P > 0.05). Conclusions For cluster outbreaks, it is important to comprehensively screen close the contacts of the index patient. Special attention should be paid to asymptomatic cases. The clinical management of cluster patients is similar to that of other COVID-19 patients.

Abstract Waterborne polyurethane (WBPU) has the advantages of environmental, excellent film‐forming property and high bonding strength. However, the mechanical strength of waterborne polyurethane in coating products is not enough, and its wear resistance and softness are poor. In this paper, the Cellulose Nanocrystals (CNC) AQ3 obtained by acid hydrolysis of microcrystalline cellulose was successfully dispersed in the waterborne polyurethane matrix to prepare nanocomposite films with different contents. The mechanical properties, thermal properties, wear resistance and softness of the nanocomposite films were studied by Thermogravimetric (TG), tensile test, dry friction test and KES test. The morphology of CNC dispersed in waterborne polyurethane matrix was studied by Scanning electron microscope (SEM). The results show that the lower content of CNC was uniformly dispersed in the waterborne polyurethane matrix and the interaction between the two forms hydrogen bonds. Compared with waterborne polyurethane, the initial degradation temperature of the nanocomposite films made by adding only 1 wt% CNC increased by 50°C, the tensile strength increased by 179.3%, the wear resistance increased by 19%, and the minimum bending stiffness B reached 0.161 (gf·cm2). The results of this study can be used for textile coating applications, such as polyurethane synthetic leather.

Linear induction motor (LIM) subway yard line often have very-small-radius curves and small turnouts with “S”-shaped curves in the middle. The lateral stability of the train is low, and wheel-climb (slipway) derailments often occur at a speed of 10 km/h. To study the derailment safety and speed limit of a linear motor subway train running through yard line, a coupling dynamic model of the train-track system is established, fully considering the driving characteristics of the linear motor, characteristics of the linear motor train, and track structure. The safety of train derailment is judged by the over-limit time of the derailment coefficient. Under the premise of a safe operation, the variations in the derailment coefficient, out-of-limit time, wheel unloading rate, and wheel-axle lateral force with the train running speed are studied. When the train runs at speeds below 25 km/h, for both new and worn rails, the derailment coefficient and lateral force of the wheel axle at each speed exceed the safety limits, and are less affected by changes in the speed. The duration of the derailment coefficient is within the safety limit because the speed of crossing the curve is low, and thus the train will not overturn and derail. Rail wear reduces the train derailment safety through the yard line, while the wheel wear improves the train derailment safety through the yard line. Considering the actual state of the yard line (wheel-rail wear, irregularity, etc.), the speed of the linear motor subway train running through the yard line should be limited to below 15 km/h.

Some small shelly fossils are important index fossils for global stratigraphic subdivisions and correlations of the Cambrian Terreneuvian. The first appearance datum (FAD) of the cosmopolitan mollusk Aldanella attleborensis has been suggested as one of the potential markers for defining the base of Cambrian Stage 2. Aldanella fossils were uncommon in South China, and if occurring, were often described as Aldanella yanjiaheensis, A. attleborensis, or indeterminate species in the literature, while A. yanjiaheensis was often taken as a junior synonym of A. attleborensis. Nevertheless, a detailed taxonomic revision of A. yanjiaheensis based on material from its type locality awaits to be made. In this study, we systematically re-evaluated A. yanjiaheensis based on the numerous specimens collected from the base of Member 5 of the Yanjiahe Formation in the Three Gorges area, western Hubei Province of South China. Detailed taxonomic comparison further substantiates that A. yanjiaheensis is a junior synonym of A. attleborensis, signifying its strong potential for a global correlation across paleocontinents. Morphological parameter analyses indicate that the length and width of shell tube of A. attleborensis shows allometric growth. The nearly cosmopolitan distribution and characteristic morphology of A. attleborensis indicate that it can play a significant role in the subdivision and correlation of Cambrian Stage 2. The co-occurrence of A. attleborensis and Watsonella crosbyi from the base of Member 5 of the Yanjiahe Formation corroborates that Member 5 belongs to Cambrian Stage 2.

In this study, a three-phase unified power quality conditioner (UPQC) topology based on a quadruple-active-bridge (QAB) is presented as an attractive solution to power quality issues. It is composed of three single-phase series converters, a three-phase shunt converter and a QAB converter located at the common DC-link. Since the bulky low frequency isolation transformers are replaced by the high frequency ones in QAB, it leads to higher power density, reduced weight and volume. The QAB converter could function as an energy router to provide multiple DC-link voltage access ports in the UPQC, which increase the flexibility of the UPQC. Finally, the MATLAB/Simulink simulation results and hardware-in-the-loop testing verify the correctness and effectiveness of the proposed topology and control method.

Monoclonal antibodies (mAbs) encoded by IGHV3-53 (VH3-53) targeting the spike receptor-binding domain (RBD) have been isolated from different COVID-19 patients. However, the existence and prevalence of shared VH3-53-encoded antibodies in the antibody repertoires is not clear. Using antibody repertoire sequencing, we found that the usage of VH3-53 increased after SARS-CoV-2 infection. A highly shared VH3-53-J6 clonotype was identified in 9 out of 13 COVID-19 patients. This clonotype was derived from convergent gene rearrangements with few somatic hypermutations and was evolutionary conserved. We synthesized 34 repertoire-deduced novel VH3-53-J6 heavy chains and paired with a common IGKV1-9 light chain to produce recombinant mAbs. Most of these recombinant mAbs (23/34) possess RBD binding and virus-neutralizing activities, and recognize ACE2 binding site via the same molecular interface. Our computational analysis, validated by laboratory experiments, revealed that VH3-53 antibodies targeting RBD are commonly present in COVID-19 patients’ antibody repertoires, indicating many people have germline-like precursor sequences to rapidly generate SARS-CoV-2 neutralizing antibodies. Moreover, antigen-specific mAbs can be digitally obtained through antibody repertoire sequencing and computational analysis.

ObjectiveEvaluation of the endoscopic features of Crohn’s disease (CD) and ulcerative colitis (UC) is the key diagnostic approach in distinguishing these two diseases. However, making diagnostic differentiation of endoscopic images requires precise interpretation by experienced clinicians, which remains a challenge to date. Therefore, this study aimed to establish a convolutional neural network (CNN)-based model to facilitate the diagnostic classification among CD, UC, and healthy controls based on colonoscopy images.MethodsA total of 15,330 eligible colonoscopy images from 217 CD patients, 279 UC patients, and 100 healthy subjects recorded in the endoscopic database of Tongji Hospital were retrospectively collected. After selecting the ResNeXt-101 network, it was trained to classify endoscopic images either as CD, UC, or normal. We assessed its performance by comparing the per-image and per-patient parameters of the classification task with that of the six clinicians of different seniority.ResultsIn per-image analysis, ResNeXt-101 achieved an overall accuracy of 92.04% for the three-category classification task, which was higher than that of the six clinicians (90.67, 78.33, 86.08, 73.66, 58.30, and 86.21%, respectively). ResNeXt-101 also showed higher differential diagnosis accuracy compared with the best performing clinician (CD 92.39 vs. 91.70%; UC 93.35 vs. 92.39%; normal 98.35 vs. 97.26%). In per-patient analysis, the overall accuracy of the CNN model was 90.91%, compared with 93.94, 78.79, 83.33, 59.09, 56.06, and 90.91% of the clinicians, respectively.ConclusionThe ResNeXt-101 model, established in our study, performed superior to most clinicians in classifying the colonoscopy images as CD, UC, or healthy subjects, suggesting its potential applications in clinical settings.

Abstract Background New strategies are urgently needed to deal with the growing problem of multidrug-resistant bacterial pathogens. As the natural viruses against bacteria, recently, bacteriophages have received particular attention. Here, we identified and characterized a novel peptidoglycan hydrolase named MMPphg by decoding the complete genome sequence of Meiothermus bacteriophage MMP17, which was isolated in Tengchong hot spring in China and contains a circular genome of 33,172 bp in size and a GC content of 63.4%. Findings We cloned the MMPphg gene, overproduced and purified the phage lytic protein, which contains a highly conserved M23 metallopeptidase domain and can be activated by Mg2+ and Zn2+. MMPphg is capable of withstanding temperatures up to 70 °C, and preserved more than 80% of its activity after a 30 min treatment between 35 and 65 °C. More interestingly, by disrupting bacterial cells, MMPphg exhibits surprising antimicrobial activity against both Gram-negative and Gram-positive pathogenic bacteria, especially antibiotic-resistant strains such as Escherichia coli O157, Staphylococcus aureus and Klebsiella pneumonia. Conclusions In the current age of mounting antibiotic resistance, these results suggest the great potential of MMPphg, the gene product of bacteriophage MMP17, in combating bacterial infections and shed light on bacteriophage-based strategies to develop alternatives to conventional antibiotics for human or veterinary applications.

The details of the immunological microenvironment and its clinical implications for pancreatic cancer are still unclear. In this study, we obtained data from public databases, such as the Gene Expression Omnibus, the Cancer Genome Atlas Program, the International Cancer Genome Consortium Data Portal, the ArrayExpress Data Warehouse, and the cBioPortal for Cancer Genomics. We used these data to evaluate the pattern of immune cells infiltration in pancreatic ductal adenocarcinoma (PDAC) tissues. We observed that the levels of M0 macrophages and activated dendritic cells in tumor tissues were significantly higher than that in para-tumor tissues. M0 macrophages, gamma delta T cells and naive CD4 T cells were independent predictive factors of a poor outcome for PDAC patients. An immune score determined by M0 macrophages, gamma delta T cells and naive CD4 T cells could predict the survival of patients. The results of this study suggest that the infiltration of immune cells, such as M0 macrophages, may be a possible target for the treatment of PDAC. However, these findings need to be confirmed by additional studies.

Abstract Background Rheumatoid arthritis (RA) is characterized by joint inflammation and damage to the cartilage and bone in collagen-induced arthritis (CIA). Mesenchymal stem cells (MSCs) can improve articular symptoms and reduce bone erosion in CIA rats; however, the underlying mechanism remains unknown. This study aimed to investigate the mechanism underlying MSC-induced improvement of bone destruction in CIA. Methods Wistar rats were divided into a normal group, CIA control group, MTX intervention group, and BMSC intervention group, each comprising 8 rats. Serum RANKL, OPG, and CXCL10 levels of all groups were determined via flow cytometry after 42 days of interventions. RANKL, OPG, TRAF6, CXCL10, and CXCR3 were detected on the synovial membrane via immunohistochemistry, and their relative mRNA levels were determined via RT-PCR analysis. BMSCs were labeled with GFP and administered to CIA rats via the tail vein. At different time points, the distribution of implanted GFP-MSCs in synovial tissues was observed using a fluorescence microscope, and the potential of GFP-MSCs to differentiate into chondrocytes was assessed via immunofluorescence analysis. Results BMSC transplantation improved joint inflammation and inhibited bone destruction in CIA rats. BMSCs inhibited the expression of serum CXCL10 and CXCL10 and CXCR3 expression at the synovial membrane. Moreover, protein and mRNA expression analyses revealed that BMSCs potentially regulated RANKL/OPG expression levels in the serum and synovial tissue. Upon implantation into CIA rats, GFP-MSCs were traced in the joints. GFP-positive cells were observed in the cartilage tissue from day 11 and until 42 days after transplantation. Anti-type II collagen/GFP double-positive cells were observed in the articular cartilage (especially damaged cartilage) upon immunofluorescence staining of anti-type II collagen. Conclusions BMSCs improve bone destruction in CIA by inhibiting the CXCL10/CXCR3 chemotactic axis, regulating the RANKL/OPG ratio, and directly differentiating into chondrocytes.

ABSTRACTThe Zika virus (ZIKV) is a mosquito-borne flavivirus that causes neonatal abnormalities and other disorders. Antibodies to the ZIKV envelope (E) protein can block infection. In this study, next-generation sequencing (NGS) of immunoglobulin heavy chain (IgH) mRNA transcripts was combined with single-cell PCR cloning of E-binding monoclonal antibodies for analysing antibody response in a patient from the early stages of infection to more than one year after the clearance of the virus. The patient's IgH repertoire 14 and 64 days after symptom onset showed dramatic dominant clonal expansion but low clonal diversity. IgH repertoire 6 months after disease-free status had few dominant clones but increased diversity. E-binding antibodies appeared abundantly in the repertoire during the early stages of infection but quickly declined after clearance of the virus. Certain VH genes such as VH5-10-1 and VH4-39 appeared to be preferentially enlisted for a rapid antibody response to ZIKV infection. Most of these antibodies require relatively few somatic hypermutations to acquire the ability to bind to the E protein, pointing to a possible mechanism for rapid defence against ZIKV infection. This study provides a unique and holistic view of the dynamic changes and characteristics of the antibody response to ZIKV infection.

Oral cancer is a major global health issue accounting for 177,384 deaths in 2018 and it is most prevalent in low- and middle-income countries. Enabling automation in the identification of potentially malignant and malignant lesions in the oral cavity would potentially lead to low-cost and early diagnosis of the disease. Building a large library of well-annotated oral lesions is key. As part of the MeMoSA®(Mobile Mouth Screening Anywhere) project, images are currently in the process of being gathered from clinical experts from across the world, who have been provided with an annotation tool to produce rich labels. A novel strategy to combine bounding box annotations from multiple clinicians is provided in this paper. Further to this, deep neural networks were used to build automated systems, in which complex patterns were derived for tackling this difficult task. Using the initial data gathered in this study, two deep learning based computer vision approaches were assessed for the automated detection and classification of oral lesions for the early detection of oral cancer, these were image classification with ResNet-101 and object detection with the Faster R-CNN. Image classification achieved an F1 score of 87.07% for identification of images that contained lesions and 78.30% for the identification of images that required referral. Object detection achieved an F1 score of 41.18% for the detection of lesions that required referral. Further performances are reported with respect to classifying according to the type of referral decision. Our initial results demonstrate deep learning has the potential to tackle this challenging task.

Aiming at the problems of unbalanced energy consumption, redundant links, short life cycle in underwater sensor networks, a topology control algorithm for underwater wireless sensor networks based on potential-game and optimal rigid sub-graph is proposed. Firstly, based on the potential game theory, the topology control model of underwater sensor network is constructed, which considers network coverage, connectivity, transmission energy consumption, end-to-end delay, transmission success rate, node residual energy and so on. It is proved that the model is an ordinal potential game and has a Nash equilibrium solution. Then, the link weight function of node load and node residual energy is introduced, and the redundant links in the network are eliminated by using the principle of optimal rigid sub-graph. Simulation experiments and contrast analysis show that compared with other network models, the network topology model constructed in this paper has lowed node load and stronger energy balance, prolongs the life cycle of the network and more conforms to the underwater environment.

Fat greenling (Hexagrammos otakii) is a kind of economic fish that is widely consumed by human, and its intensive farming technology is making important progress. However, high-density farming may cause the occurrence of diseases in H. otakii. Cinnamaldehyde (CNE) is a new feed additive for aquatic animals and has a positive effect on disease resistance. In the study, dietary CNE was evaluated on the growth performance, digestion, immune response, and lipid metabolism of juvenile H. otakii (6.21±0.19 g). Six experimental diets were formulated containing CNE at levels of 0, 200, 400, 600, 800, and 1000 mg/kg for 8 weeks. The percent weight gain (PWG), specific growth rate (SGR), survival (SR), and feeding rate (FR) were significantly increased by including CNE in fish diets regardless of the inclusion level (P

Cnidarians diverged very early in animal evolution; therefore, investigations of the morphology and trophic levels of early fossil cnidarians may provide critical insights into the evolution of metazoans and the origin of modern marine food webs. However, there has been a lack of unambiguous anthozoan cnidarians from Ediacaran assemblages, and undoubted anthozoans from the Cambrian radiation of metazoans are very rare and lacking in ecological evidence. Here, we report a new polypoid cnidarian, Nailiana elegans gen. et sp. nov., represented by multiple solitary specimens from the early Cambrian Chengjiang biota (∼520 Ma) of South China. These specimens show eight unbranched tentacles surrounding a single opening into the gastric cavity, which may have born multiple mesenteries. Thus, N. elegans displays a level of organization similar to that of extant cnidarians. Phylogenetic analyses place N. elegans in the stem lineage of Anthozoa and suggest that the ancestral anthozoan was a soft-bodied, solitary polyp showing octoradial symmetry. Moreover, one specimen of the new polyp preserves evidence of predation on an epifaunal lingulid brachiopod. This case provides the oldest direct evidence of macrophagous predation, the advent of which may have triggered the emergence of complex trophic/ecological relationships in Cambrian marine communities and spurred the explosive radiation of animal body plans. Although fossil evidence suggests that various animal groups were able to move actively through their environment in the early stages of their evolution, virtually no direct information is available on the nature of their muscle systems. The origin of jellyfish swimming, for example, is of great interest to biologists. Exceptionally preserved muscles are described here in benthic peridermal olivooid medusozoans from the basal Cambrian of China (Kuanchuanpu Formation, ca. 535 Ma) that have direct equivalent in modern medusozoans. They consist of circular fibers distributed over the bell surface (subumbrella) and most probably have a myoepithelial origin. This is the oldest record of a muscle system in cnidarians and more generally in animals. This basic system was probably co-opted by early Cambrian jellyfish to develop capacities for jet-propelled swimming within the water column. Additional lines of fossil evidence obtained from ecdysozoans (worms and panarthropods) show that the muscle systems of early animals underwent a rapid diversification through the early Cambrian and increased their capacity to colonize a wide range of habitats both within the water column and sediment at a critical time of their evolutionary radiation.

Study of the association between single nucleotide polymorphisms (SNPs) of methotrexate (MTX) pathway genes and MTX-related toxicity in the treatment of hematological malignancies is popular. Here, we studied the association between SNPs of MTHFR and ABCB1 and MTX-related toxicity in 157 adult Chinese patients diagnosed with hematological malignancies. Patients were genotyped for MTHFR rs1801131, MTHFR rs1801133, and ABCB1 rs1045642 by fluorescence in situ hybridization. Patients with MTHFR rs1801133T allele had a significantly higher risk of hematopoietic toxicity compared with those with CC genotype (p=0.003). With respect to MTHFR rs1801131, patients with CC and AC genotypes had significantly lower frequency of hematopoietic toxicity than patients with AA genotype (p=0.044). In conclusion, we identified an important influence of the SNPs of ABCB1 and MTHFR on MTX-related hematopoietic toxicity in adults with hematological malignancies. To optimize high-dose (HD)-MTX therapy and reduce related hematopoietic toxicity, it is necessary to detect the SNPs of MTHFR and ABCB1 before initiating HD-MTX and deciding the optimal dose of MTX and duration of leucovorin rescue, according to genetic tests and disease type in adults with hematological malignancies.

Various microfossils from the early Cambrian provide crucial clues for understanding the Cambrian explosion and the origin of animal phyla. However, specimens with important anatomical structures are extremely rare and the efficiency of retrieving such fossils by traditional manual selection under a microscope is quite low. Such a contradiction has hindered breakthroughs in micropaleontology for a long time. Here, we propose a solution for identifying specific taxa of Cambrian microfossils using only a few available specimens by transferring a model pre-trained on natural image datasets to the field of paleontological artificial intelligence. The method employs a 34-layer deep residual neural network as the underlying framework, migrates the ImageNet pre-trained model, freezes the low-layer network parameters and retrains the high-layer parameters to build a microfossil image recognition model. We built training sets with randomly selected images of varied number for each taxon. Our experiments show that the average recognition accuracy for specific taxa of Cambrian microfossils (50 images for each taxon) is higher than 0.97 and it can reach 0.85 with only three training samples per taxon. Comparative analyses indicate that our results are much better than those of various prevalent methods, such as the transpose convolutional neural network (TCNN). This demonstrates the feasibility of using natural images (ImageNet) for the training of microfossil recognition models and provides a promising tool for the discovery of rare fossils.

Staphylococcus aureus can cause chronic infections which are closely related to persister formation. Purine metabolism is involved in S. aureus persister formation, and purN, encoding phosphoribosylglycinamide formyltransferase, is an important gene in the purine metabolism process. In this study, we generated a ΔpurN mutant of the S. aureus Newman strain and assessed its roles in antibiotic tolerance and virulence. The ΔpurN in the late exponential phase had a significant defect in persistence to antibiotics. Complementation of the ΔpurN restored its tolerance to different antibiotics. PurN significantly affected virulence gene expression, hemolytic ability, and biofilm formation in S. aureus. Moreover, the LD50 (3.28 × 1010 CFU/mL) of the ΔpurN for BALB/c mice was significantly higher than that of the parental strain (2.81 × 109 CFU/mL). Transcriptome analysis revealed that 58 genes that were involved in purine metabolism, alanine, aspartate, glutamate metabolism, and 2-oxocarboxylic acid metabolism, etc., were downregulated, while 24 genes involved in ABC transporter and transferase activity were upregulated in ΔpurN vs. parental strain. Protein-protein interaction network showed that there was a close relationship between PurN and GltB, and SaeRS. The study demonstrated that PurN participates in the formation of the late exponential phase S. aureus persisters via GltB and regulates its virulence by activating the SaeRS two-component system.

The isolated sclerites of the Ocruranus and Eohalobia group are abundant among the early Cambrian small shelly fossil assemblages, which were recently assigned to the same scleritome as an early member of the polyplacophoran (chiton) stem lineage. However, the scleritome reconstruction and zoological affinities of these sclerites are still controversial due to the lack of exceptionally preserved articulated specimens with in-situ sclerites. Herein, we report new specimens of Ocruranus and Eohalobia sclerites from Member 5 of the Yanjiahe Formation, which provide new insights into the reconstruction of the original scleritome. The Eohalobia sclerites from the Yanjiahe Formation have an extended and upfolded proximal field with dense wrinkles, which seems to be a weakly mineralized structure and acted as a joint with another sclerite, Ocruranus. Comparing the butterfly-shaped proximal field on a unique sclerite of Eohalobia with the sub-apical field on Ocruranus sclerites suggests that the original scleritome of this group may consist of only two types of sclerites: the Ocruranus-type and the Eohalobia-type. The polygonal structure on the internal mold of Eohalobia sclerites is interpreted herein as the muscle attachment zone; their distribution corresponds well with that of the modern chitons, which provides strong evidence to support the close relationship between the Ocruranus–Eohalobia group and the Polyplacophora.

Treatment of mucosa-associated lymphoid tissue (MALT) lymphoma has recently received considerable attention. Here, we report a case of large esophageal MALT lymphoma that was successfully en bloc resected using endoscopic submucosal dissection (ESD). A 77-year-old woman was admitted to our hospital with progressive dysphagia for more than 2 months. Upper gastrointestinal endoscopy revealed a large rounded submucosal mass covered by normal mucosa, located at the lower esophagus. Endoscopic ultrasonography (EUS) showed a well-demarcated hypoechoic mass chiefly located in the esophageal wall, but the layers of the esophageal wall were not clear. ESD was performed for diagnostic and treatment purposes. No complications occurred during or after ESD. The resected specimen measured 4.3 cm × 2.8 cm × 1.5 cm. The histologic findings were diagnostic of esophageal MALT lymphoma. Infiltration of neoplastic cells in the lateral margins of the resected specimen was not observed. After 5 months, the patient was still under surveillance and free of recurrent disease. Resection with ESD of such a large mass of MALT in the esophageal region has rarely been reported before in the literature.

Mucosa-associated lymphoid tissue (MALT) lymphoma arises in extra-nodal sites from the malignant transformation of B lymphocytes that are mainly triggered by infection or autoimmune process. MALT lymphoma is frequently detected in the gastrointestinal tract. As the causal relationship between Helicobacter pylori (H. pylori) infection and gastric MALT lymphoma, it was well-established that early-stage gastric MALT lymphoma could be cured by H. pylori eradication, and about 50–95% of cases achieved complete response with anti-H. pylori treatment. Compared to the stomach which is the most involved site due to the high prevalence of H. pylori infection, the colorectum is rarely affected. Primary rectal MALT lymphoma is a rare malignancy, and there are no specific therapeutic strategies so far. Here we report a case of rectal MALT lymphoma successfully resected by endoscopic submucosal dissection (ESD). ESD serves as a novel strategy to cure small localized rectal MALT lymphomas to avoid unnecessary surgery or chemo-radiotherapy.

High-entropy alloys (HEAs) are a new type of multi-component alloy. The design of the compositions breaks the design ideas of traditional alloys and shows many excellent properties. Therefore, an Al0.5FeCoCrNi HEA with face-centered cubic (FCC) and body-centered cubic (BCC) dual-phase structure was used in this paper. During the additive manufacturing process, the heat dissipation rate gradually changes with the increase in wall height. As a result, the composition of the phases changes, resulting in differences in mechanical properties. Here, we designed laser melting deposition (LMD) on T-beams of different heights to change the heat dissipation rate of the wall, and the effects of the heat dissipation rate on the microstructure and mechanical properties of Al0.5FeCoCrNi HEAs were studied. The experimental results showed that increasing the height of the T-beam would gradually slow down the heat dissipation rate of the wall. The above phenomena not only led to a gradual reduction of the BCC phase under the influence of heat accumulation but also increased the length of columnar crystals in the wall with the slowing of heat dissipation. Heat accumulation hindered the nucleation during solidification and eventually led to the growth of grains across the deposition layer. Furthermore, the slow heat dissipation rate changed the grain number and BCC phase content, which gradually decreased the strength and hardness, while the ductility of the samples improved.

To optimize the microstructure and properties of TC4 specimens formed by selective laser melting (SLM), the test program of formed specimens by the variable parameter forming process (VPFP) was designed based on the quantitative parameter forming process (QPFP). The purpose of this study is to explore the influence of the VPFP on the surface morphology, tensile properties, and microstructure of the specimens. The test results show that the surface morphology and tensile properties of the specimens were better formed by the VPFP. The internal holes of the specimens formed by the VPFP were small in volume and occupied a relatively small proportion, and the density could reach 99.7%. When the laser power was 300 W–260 W and equally divided into six hierarchies, the tensile strength could reach 1185.214 MPa by VPFP, but the elongation had no obvious change. The number of secondary acicular martensite α’ phases was decreased in the microstructure of the specimens formed with VPFP. With the superposition of the hierarchy, the length of the primary acicular martensite α’ phase became shorter, the width became larger, and the width of the columnar crystal β phase became smaller. The VPFP is used to change the inherent method of forming specimens with the same parameters, which provides a new idea for SLM-forming structures; the test provides data and yields a theoretical research basis for forming the specimens process method.
