Inorganic and organic pollutants present a hazard to surface and groundwater resources. Peroxymonosulfate (PMS, HSO5−) has received increasing attention for in situ chemical oxidation (ISCO) capable of remediating contaminated sites. Considering that saline waters occur widely in natural environments, it is desirable to evaluate the effect of Cl− on the PMS oxidation of organic compounds. In this study, 2,4-dichlorophenol (2,4-DCP) was used as a model pollutant. At a PMS concentration of 2.0 mM, Cl− concentration of 50 mM, and solution pH of 7.0, 2,4-DCP was completely degraded by PMS in the presence of Cl− (PMS/Cl− system), while PMS alone exhibited almost no reactivity with 2,4-DCP. The degradation of 2,4-DCP was optimized at a solution pH of 8.4 and high concentrations of PMS and Cl−. Quenching experiments and degradation pathway analyses indicated that HClO was responsible for 2,4-DCP oxidation, and HClO was mainly generated by the interaction of Cl− with HSO5−, rather than SO52−. Consequently, the transformation from HSO5− to HClO appeared under a solution pH of 10.0 and was favored in an acidic solution. Given the ambient pH and Cl− concentrations of saline waters, a considerable amount of HClO may be produced by the interaction of PMS with Cl− in the oxidant delivery stage of ISCO processes. Interestingly, H2O2 and peroxydisulfate did not exhibit reactions similar to those of PMS. This research indicated that caution must be exercised when choosing an oxidant for ISCO processes in saline waters.

Membrane-bound talin head domain and kindlin bind to the �� integrin cytoplasmic tail, cooperating to activate the heterodimeric integrin, and the talin head domain induces integrin clustering in the presence of Mn$^{2+}$. Here we show that kindlin-1 can replace Mn2+ to mediate ��3 integrin clustering induced by the talin head, but not that induced by the F2���F3 fragment of talin. Integrin clustering mediated by kindlin-1 and the talin head was lost upon deletion of the flexible loop within the talin head F1 subdomain. Further mutagenesis identified hydrophobic and acidic motifs in the F1 loop responsible for ��3 integrin clustering. Modeling, computational and cysteine crosslinking studies showed direct and catalytic interactions of the acidic F1 loop motif with the juxtamembrane domains of ��- and ��3-integrins, in order to activate the ��3 integrin heterodimer, further detailing the mechanism by which the talin���kindlin complex activates and clusters integrins. Combining the star-shaped alkynyl ligands with low-nuclearity gold-copper triphosphane clusters produces 3D metallocage aggregates, which demonstrate room temperature phosphorescence in solution (max Φem =0.6). Their luminescence mainly originates from cluster-localized metal-to-ligand charge transfer excited state. These supramolecular assemblies can be easily converted into the isostructural gold-silver congeners by the direct exchange of the metal ions. Such modification of the terminal metal cores switches the emission to the intraligand (alkyne) electronic transitions of the triplet manifold, that represents an unusual optical functionality among the metallocycle/metallocage complexes.

The use of recombinant allergens is a promising approach in allergen-specific immunotherapy (AIT). Considerable limitation, however, has been the ability of recombinant allergens to activate effector cells leading to allergic reactions. Recombinant hypoallergens with preserved protein folding and capacity to induce protective IgG antibodies binding effectively to the native allergen upon sensitization would be beneficial for safer AIT. In this study, hypoallergen variants of the major horse allergen Equ c 1 were designed by introducing one point mutation on the putative IgE epitope region and two mutations on the monomer-monomer interface of Equ c 1 dimer. The recombinant Equ c 1 wild type and the variants were produced and purified to homogeneity, characterized by size-exclusion ultra-high performance liquid chromatography and ultra-high resolution mass spectrometry. The IgE-binding profiles were analyzed by a competitive immunoassay and the biological activity by a histamine release assay using sera from horse allergic individuals. Two Equ c 1 variants, Triple 2 (V47K + V110E + F112K) and Triple 3 (E21Y + V110E + F112K) showed lower allergen-specific IgE-binding capacity and decreased capability to release histamine from basophils in vitro when using sera from six allergic individuals. Triple 3 showed higher reduction than Triple 2 in IgE-binding (5.5 fold) and in histamine release (15.7 fold) compared to wild type Equ c 1. Mutations designed on the putative IgE epitope region and monomer-monomer interface of Equ c 1 resulted in decreased dimerization, a lower IgE-binding capacity and a reduced triggering of an allergic response in vitro.

This study examined side-stream generation from Finnish forest industry comprising data from 19 sawmills, 1 plywood mill, 1 particleboard mill, 10 paper and board mills, 1 semi-mechanical mill, and 15 chemical pulp mills. A structured questionnaire survey was carried out for this study. The results revealed that the generation of side stream in sawmills, plywood mills, paper and board mills, mechanical pulp mills, and chemical pulp mills accounted for 42.2%, 56.1%, 23.8%, 30.7%, and 56% of round wood log input, respectively, and 91.2%, 135.5%, 42.2%, 44.7%, and 238.5% of the finished product, respectively. Woodchips, sawdust, and bark were recognized as the significant side streams in sawmills and plywood mills, whereas fiber sludge was found the main side stream in paper, board and mechanical pulp mills. In chemical pulp mills, black liquor was found the greatest side stream, which comprises 43% of the wood input and 183.3% of the finished product. The study estimated that in 2016, Finnish forest industry generated a total 27.7 million tons of side streams, consisting of 49.2% black liquor, 28.5% solid wood-based waste, 14.1% sludge, 4.4% ashes, and 3.8% others. The encapsulation and/or surface modification can stabilize and protect the phosphorescence bio‐probes but impede their intravenous delivery across biological barriers. Here, a new class of biocompatible rhenium (ReI) diimine carbonyl complexes is developed, which can efficaciously permeate normal vessel walls and then functionalize the extravascular collagen matrixes as in situ oxygen sensor. Without protective agents, ReI‐diimine complex already exhibits excellent emission yield (34%, λ em = 583 nm) and large two‐photon absorption cross‐sections (σ 2 = 300 GM @ 800 nm) in water (pH 7.4). After extravasation, remarkably, the collagen‐bound probes further enhanced their excitation efficiency by increasing the deoxygenated lifetime from 4.0 to 7.5 µs, paving a way to visualize tumor hypoxia and tissue ischemia in vivo. The post‐extravasation functionalization of extracellular matrixes demonstrates a new methodology for biomaterial‐empowered phosphorescence sensing and imaging., Biocompatible rhenium (ReI) diimine carbonyl complexes functionalize the extravascular collagen matrixes as in situ oxygen sensor. After tail‐vein delivery and vessel penetration, the probe can bind extravascular collagens and achieve stabilization enhanced two‐photon phosphorescence yields. Synthetic micro- and nanomotors (MNMs) have emerged as a vibrant research field in multidisciplinary nanotechnology with proof-of-concept applications in various disciplines. In this tutorial review, an overview of the latest achievements towards light-driven MNMs is given and their propulsion mechanisms are introduced. The focus of the paper is on the autonomously propelled MNMs that exploit light-induced physical effects or chemical reactions. Light-induced body deformation, as a completely different, nature inspired concept that is found mostly in soft, polymeric MNMs, is also reviewed. In the end, a few applications of photocatalytic and light-driven MNMs for environmental remediation are presented and their potential is critically discussed.

Fast pyrolysis oil (FPO) produced from short-rotation willow was characterized by using high-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry and various bulk chemical analyses. The pyrolysis experiments were carried out on a bench-scale bubbling fluidized bed pyrolysis reactor operating at the temperature of 500 °C. The resulting pyrolysis oil was a single-phase liquid with relatively low oxygen content and had pH and total acid number values of 3.0 and 129 mg/g, respectively. A wealth of compositional information of the willow FPO was obtained using FT-ICR MS in combination with electrospray ionization (ESI) and atmospheric-pressure photoionization (APPI) techniques. While ESI preferentially ionized acidic extractives, small phenolics and anhydrosugars, APPI ionized clearly larger compounds, including neutral lipophilic extractives, triterpenoids and high molecular mass pyrolytic lignin, most of which are beyond conventional gas chromatography–mass spectrometry analyses.

We present a novel crystal structure of the IlvD/EDD family enzyme, l-arabinonate dehydratase from Rhizobium leguminosarum bv. trifolii (RlArDHT, EC 4.2.1.25), which catalyzes the conversion of l-arabinonate to 2-dehydro-3-deoxy-L-arabinonate. The enzyme is a tetramer consisting of a dimer of dimers, where each monomer is composed of two domains. The active site contains a catalytically important [2Fe-2S] cluster and Mg2+ ion and is buried between two domains, and also at the dimer interface. The active site Lys129 was found to be carbamylated. Ser480 and Thr482 were shown to be essential residues for catalysis, and the S480A mutant structure showed an unexpected open conformation in which the active site was more accessible for the substrate. This structure showed the partial binding of L-arabinonate, which allowed us to suggest that the alkoxide ion form of the Ser480 side chain functions as a base and the [2Fe-2S] cluster functions as a Lewis acid in the elimination reaction.

The coordination chemistry of the tri- and tetradentate chelating phosphines (2-PPh2C6H4)2P(O)Ph (P3O) and (2-PPh2C6H4)3P (P4) with respect to d10 copper subgroup metal ions has been investigated. Depolymerization of (MC2R)n (M = Cu, Ag) with P4 affords the series of mono- and trinuclear complexes (P4)CuC2Ph (1), (P4)Cu3(C2Ph)3 (2), (P4)Ag3(C2Ph) (Hal)2 (Hal = Cl (3), Br (4), I (5)). Reactions of the M+ (M = Cu, Ag) ions with (M′C2R)n (M′ = Cu, Ag, Au) acetylides in the presence of P4 yield the family of dinuclear species [(P4)MM′(C2R)]+ (6–12), which comprise the Cu2/Ag2 (6, 7; R = Ph), AuCu (8–10; R = Ph, C(OH)Me2, C(OH)Ph2), and AuAg (11, 12; R = Ph, C(OH)Ph2) metal cores. Small zinc finger (ZnF) motifs are promising molecular scaffolds for protein design owing to their structural robustness and versatility. Moreover, their characterization provides important insights into protein folding in general. ZnF motifs usually possess an exceptional specificity and high affinity towards Zn(II) ion to drive folding. While the Zn(II) ion is canonically coordinated by two cysteine and two histidine residues, many other coordination spheres also exist in small ZnFs, all having four amino acid ligands. Here we used high-resolution mass spectrometry to study metal ion binding specificity and primary coordination sphere robustness of a designed zinc finger, named MM1. Based on the results, MM1 possesses high specificity for zinc with sub-micromolar binding affinity. Surprisingly, MM1 retains metal ion binding affinity even in the presence of selective alanine mutations of the primary zinc coordinating amino acid residues.

Pharmaceutically active compounds (PhACs) are emerging pollutants causing serious challenges to wastewater treatment plants due to poor biodegradability. In this study, the enhanced removal of highly recalcitrant and commonly monitored PhACs, carbamazepine (CBZ) and diclofenac (DCF) by heterogeneous photocatalysis was investigated using 5% Ag2O/P-25 photocatalyst. The photocatalyst was characterized by scanning electron microscope (SEM-EDX), Brunauer-Emmett-Teller (BET), Fourier transfer infrared spectroscopy (FTIR) and UV–vis diffuse reflectance spectra (UV-DRS). The effects of catalyst dose, initial pollutants concentration, and mineralization during the photocatalytic degradation of PhACs were investigated. The matrix effect was assessed in deionized water (DW) and real membrane bioreactor effluent (RME). Optimal CBZ and DCF removals of 89.10% and 93.5%, respectively for 180 min of UV irradiation were achieved at catalyst dosage of 0.4 g L−1 in DW matrix. However, the optimal catalyst dosages for CBZ and DCF in RME matrix were increased by factor 2 and 1.5, respectively, to achieve the same degree of removal. Declining trends of removal rate were observed when initial concentrations of both the PhACs were increased under optimal catalyst dosages, and kinetics seem to fit the Langmuir-Hinshelwood model. Photo-induced holes and OH were the dominant oxidation species involved in the photocatalytic degradation of the PhACs. A plausible reusability of 5% Ag2O/P-25 photocatalyst was observed for both the PhACs. Moreover, various aromatic/aliphatic intermediates generated during the photodegradation CBZ were identified using fourier-transform ion cyclotron resonance (FT-ICR) mass spectrometry, and a possible multi-step degradation pathway was proposed. Coinage metal cyanides efficiently react with a triphosphine. PPh2C6H4-PPh-C6H4PPh2 (P(3)). to give the complexes M(P(3))CN, where M = Cu (1), Ag (2), and Au (3), which can further interact with coordinatively unsaturated metal centers [M(P(3))](+) to give the homobimetallic [(P(3))M-CN-M(P(3))](+)X(-) [M = Cu (4a with X(-) = CF3SO3(-) and 4b with X(-) = BF4(-)), Ag (5)] or heterometallic [(P(3))Au-CN-Ag(P(3))](+) (6) species. Extension of this approach also provided the trinuclear complex [(P(3))Cu-NC-Au-CN-Cu(P(3))](+) (7). Compounds 1-5 were characterized in the solid state by X-ray crystallography. The NMR spectroscopic studies revealed that all of the complexes except 6 retain their structures in solution. The title compounds are luminescent in the solid state, with quantum yields ranging from 8 to 87%. The observed photoemission originates mainly from the metal-to-ligand charge-transfer states according to time-dependent density functional theory computational studies. The crystalline bimetallic Cu complexes 4a/4b demonstrate extremely high sensitivity of the emission intensity to molecular O2 (KSV1 = 639 atm(-1) and LOD = 0.010% for 3 times the signal-to-noise ratio).

Manganese oxide (MnO2 ) based micromotors exhibiting a dual effect, that is, catalytic degradation and adsorptive bubble separation, were employed for water remediation. The dual effect of MnO2 microparticles led to a greater than 90 % of decolorization of non-biodegradable organic dyes in just 1 h, without the need for external agitation or bubble generation. These findings suggest high potential of MnO2 micromotors for decontamination of organic pollutants from wastewaters or natural water reserves.

Sin3A-associated protein 30-like (SAP30L) is one of the key proteins in a multi-subunit protein complex involved in transcriptional regulation via histone deacetylation. SAP30L, together with a highly homologous SAP30 as well as other SAP proteins (i.e., SAP25, SAP45, SAP130, and SAP180), is an essential component of the Sin3A corepressor complex, although its actual role has remained elusive. SAP30L is thought to function as an important stabilizing and bridging molecule in the complex and to mediate its interactions with other corepressors. SAP30L has been previously shown to contain an N-terminal Cys3 His type zinc finger (ZnF) motif, which is responsible for the key protein-protein, protein-DNA, and protein-lipid interactions. By using high-resolution mass spectrometry, we studied a redox-dependent disulfide bond formation in SAP30L ZnF as a regulatory mechanism for its structure and function. We showed that upon oxidative stress SAP30L undergoes the formation of two specific disulfide bonds, a vicinal Cys29-Cys30 and Cys38-Cys74, with a concomitant release of the coordinated zinc ion. The oxidized protein was shown to remain folded in solution and to bind signaling phospholipids. We also determined a solution NMR structure for SAP30L ZnF that showed an overall fold similar to that of SAP30, determined earlier. The NMR titration experiments with lipids and DNA showed that the binding is mediated by the C-terminal tail as well as both α-helices of SAP30L ZnF. The implications of these results for the structure and function of SAP30L are discussed.

Platinum-based micromotors have shown many exciting applications when functionalized using gold–thiol chemistry. However, thiols are known to bind to the Pt surface, which can lead to serious deactivation of the catalyst. In this paper, it is demonstrated that MnO2 can be used to protect Pt-based micromotors prior to the thiol treatment, thus fully avoiding the catalyst poisoning. This approach will greatly facilitate the use of the functionalized Pt micromotors for which the thiol toxicity has been a limiting factor.

Lignin is an important aromatic biopolymer with the structure based on phenolic units. Depolymerized lignin materials can be useful substitutes for phenol in phenol-formaldehyde resins. Our aim was to examine the potential of the base-catalyzed oxidation method to provide reactive oligomeric lignin fragments from softwood kraft lignin through the cleavage of the ether bonds. Three different bases, namely, NaOH, KOH, and NH4OH, were used as the catalyst in an aerobic oxidation, in which the influence of the reaction time was examined for 2 h and 6 h. Oxidized lignin materials were characterized with high-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry and nuclear magnetic resonance spectroscopy (13C, 1H-13C HSQC, and 31P NMR). The presence of methanol in the oxidation gas phase and formation of carboxylic acids indicate that some of the lignin structures were degraded in the oxidation. Guaiacyl units, the major phenolic structure of softwood kraft lignin, survived the oxidation. The oxidatively depolymerized lignin materials showed a reactivity close to that of pristine lignin in a reaction with formaldehyde, as determined by differential scanning calorimetry (DSC). Overall, the FT-ICR MS, NMR, and DSC results proved that the oxidized lignin materials maintained their oligomeric nature and possess the reactivity with formaldehyde.

Catechol oxidases and tyrosinases are coupled binuclear copper enzymes that oxidize various o-diphenolic compounds to corresponding o-quinones. Tyrosinases have an additional monooxygenation ability to hydroxylate monophenol to o-diphenol. It is still not clear what causes the difference in the catalytic activities. We solved a complex structure of Aspergillus oryzae catechol oxidase with resorcinol bound into the active site. Catalytic activity of A. oryzae catechol oxidase was studied, for the first time, by high-resolution FT-ICR mass spectrometry to shed light on the reaction mechanism. The enzyme was also found to catalyze monooxygenation of small phenolics, which provides a novel perspective for the discussion of differences in the catalytic activity between tyrosinases and catechol oxidases. According to the results, two binding modes for resorcinol are suggested and a reaction mechanism for coupled binuclear copper enzymes is discussed.

This phylum is distinctive among photosynthetic organisms in containing genes encoding uncharacterized cystathionine β-synthase (CBS)–chloroplast protein (CP12) fusion proteins. These consist of two domains, each recognized as stand-alone photosynthetic regulators with different functions described in cyanobacteria (CP12) and plants (CP12 and CBSX). Here we show that CBS–CP12 fusion proteins are encoded in distinct gene neighborhoods, several unrelated to photosynthesis. Most frequently, CBS–CP12 genes are in a gene cluster with thioredoxin A (TrxA), which is prevalent in bloom-forming, marine symbiotic, and benthic mat cyanobacteria. Focusing on a CBS–CP12 from Microcystis aeruginosa PCC 7806 encoded in a gene cluster with TrxA, we reveal that the domain fusion led to the formation of a hexameric protein. We show that the CP12 domain is essential for hexamerization and contains an ordered, previously structurally uncharacterized N-terminal region. We provide evidence that CBS–CP12, while combining properties of both regulatory domains, behaves different from CP12 and plant CBSX. It does not form a ternary complex with phosphoribulokinase (PRK) and glyceraldehyde-3-phosphate dehydrogenase. Instead, CBS–CP12 decreases the activity of PRK in an AMP-dependent manner. The chemical composition of slow pyrolysis oil made from unbarked Scots pine (Pinus sylvestris) was characterized by negative-ion electrospray ionization Fourier transform ion cyclotron resonance (ESI FT-ICR) mass spectrometry and various bulk analyses. The wood pieces were heated stepwise in a batch pyrolysis reactor to the target temperature of 320 °C. The resulting pyrolysis oil spontaneously separated into aqueous and oily phases. The water content of the aqueous phase was as high as 60.4 wt %, whereas for the oily phase it was only 8.1 wt %. The bulk O/C ratio (on a dry basis) was more than twice as high for the aqueous phase as compared to the oily phase (0.61 vs 0.29), suggesting different chemistry between the two phases. On the basis of pH and total acid number measurements, the aqueous phase was more acidic than the oily phase. A family of mono- and bidentate alkynylphosphanes has been used for the preparation of the rhenium(I) phenanthroline complexes [Re(phen)(CO)2(L)2]+ (L = monophosphane), [{Re(phen)(CO)3}2(L)]2+ (L = diphosphane), and [{Re(phen)(CO)2}2(L)2]2+ (L = diphosphane). The dicarbonyl compounds display intense luminescence at 298 K both in solution and in the solid state (Φem ≤ 44 %). The tricarbonyl complexes demonstrate a strong dependence of the emission efficiency on the ancillary ligand L through a pronounced π–π* contribution of the diphosphane to the electronic transitions, as confirmed by TDDFT studies. In the solid state, the complexes [Re(phen)(CO)2(L)2]+ display significant luminescence rigidochromism that is influenced by the structure of the phosphane substituents. The latter provide a means to fine-tune the photophysical properties of this class of luminophores through variation of the stereochemical characteristics of the ligands that are not directly involved in the electronic transitions.

We report on the preparation of trimetallic (Pt/Ni/Au) microcaps using silica particles as templates. Capping of silica particles was performed by simple sputter deposition of metal layers, followed by dissolution of templates to obtain a suspension of microcaps. The microcaps were bubble-propelled to very high speeds (up to ∼500 μm s−1 on average) in the presence of 6% hydrogen peroxide as fuel. While the Ni layer provides a directional control of microcaps in an external magnetic field, the Au layer allows surface modification for further applications. We also demonstrate capture and cargo transport of oil droplets by microcap motors.

Background Carbonic anhydrases (CAs) are ubiquitous, essential enzymes which catalyze the conversion of carbon dioxide and water to bicarbonate and H+ ions. Vertebrate genomes generally contain gene loci for 15–21 different CA isoforms, three of which are enzymatically inactive. CA VI is the only secretory protein of the enzymatically active isoforms. We discovered that non-mammalian CA VI contains a C-terminal pentraxin (PTX) domain, a novel combination for both CAs and PTXs. Methods We isolated and sequenced zebrafish (Danio rerio) CA VI cDNA, complete with the sequence coding for the PTX domain, and produced the recombinant CA VI–PTX protein. Enzymatic activity and kinetic parameters were measured with a stopped-flow instrument. Mass spectrometry, analytical gel filtration and dynamic light scattering were used for biophysical characterization. Sequence analyses and Bayesian phylogenetics were used in generating hypotheses of protein structure and CA VI gene evolution. A CA VI–PTX antiserum was produced, and the expression of CA VI protein was studied by immunohistochemistry. A knock-down zebrafish model was constructed, and larvae were observed up to five days post-fertilization (dpf). The expression of ca6 mRNA was quantitated by qRT-PCR in different developmental times in morphant and wild-type larvae and in different adult fish tissues. Finally, the swimming behavior of the morphant fish was compared to that of wild-type fish. Results The recombinant enzyme has a very high carbonate dehydratase activity. Sequencing confirms a 530-residue protein identical to one of the predicted proteins in the Ensembl database (ensembl.org). The protein is pentameric in solution, as studied by gel filtration and light scattering, presumably joined by the PTX domains. Mass spectrometry confirms the predicted signal peptide cleavage and disulfides, and N-glycosylation in two of the four observed glycosylation motifs. Molecular modeling of the pentamer is consistent with the modifications observed in mass spectrometry. Phylogenetics and sequence analyses provide a consistent hypothesis of the evolutionary history of domains associated with CA VI in mammals and non-mammals. Briefly, the evidence suggests that ancestral CA VI was a transmembrane protein, the exon coding for the cytoplasmic domain was replaced by one coding for PTX domain, and finally, in the therian lineage, the PTX-coding exon was lost. We knocked down CA VI expression in zebrafish embryos with antisense morpholino oligonucleotides, resulting in phenotype features of decreased buoyancy and swim bladder deflation in 4 dpf larvae. Four out of six officially recognized dog allergens are members of the lipocalin protein family. So far, a three-dimensional structure has been determined for only one dog allergen, Can f 2, which is a lipocalin protein. We present here the crystal structure of a second lipocalin allergen from dog, a variant of Can f 4. Moreover, we have compared and analyzed the structures of these two weakly homologous (amino acid identity 21%) dog allergens. The size and the amino acid composition of the ligand-binding pocket indicate that Can f 4 is capable of binding only relatively small hydrophobic molecules which are different from those that Can f 2 is able to bind. The crystal structure of Can f 4 contained both monomeric and dimeric forms of the allergen, suggesting that Can f 4 is able to form transient (weak) dimers. The existence of transient dimers in solution was confirmed by use of native mass spectrometry. The dimeric structure of Can f 4 is formed when the ends of four β-strands are packed against the same strands from the second monomer. The residues in the interface are mainly hydrophobic and the formation of the dimer is similar to the major horse allergen Equ c 1. Interestingly, the crystal structure of dog Can f 2 has been reported to show a different type of dimer formation. The capability of these allergens to form dimers may be important for the development of immediate allergic reaction (mast cell activation) because oligomeric allergens can effectively present multivalent epitopes.

A laboratory-scale reactor was used to produce pyrolysis oils from Finnish silver birch hardwood (Betula pendula). The resulting wood distillates were characterized by ultrahigh-resolution (12 T) Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry coupled with negative-ion electrospray ionization (ESI). Two different pyrolysis temperatures were tested: 300 and 380 °C (the resulting oil samples were named as Oil-300 and Oil-380, respectively). The detected species were sorted on the basis of heteroatom class, carbon number, and double bond equivalent (DBE). About 1200 and 1400 unique compounds were identified from the ESI FT-ICR spectra of Oil-300 and Oil-380, respectively. These were mainly oxygen-containing compounds (Ox heteroatom classes, with x = 2–14), comprising up to 90% of all identified compounds. A series of 7-(2-oxoalkoxy)coumarins have been synthesized by conjugating substituted 7-hydroxycoumarins with different chloroketones. The anti-inflammatory properties of 7-(2-oxoalkoxy)coumarins were studied in LPS-induced inflammatory response in J774 macrophages. Western blot was used to determine the expression of iNOS and COX-2, NO was determined by measuring its metabolite nitrite by Griess reaction and IL-6 was measured by ELISA. Seventeen of the studied compounds inhibited NO and IL-6 production over 50% at 100 μM concentrations. IC50 values of the best inhibitors were 21 μM/24 μM (NO/IL-6) for compound 12 and 30 μM/10 μM (NO/IL-6) for compound 20. The main result was that the substitution with 7-(2-oxoalkoxy) group improved the anti-inflammatory properties of most of the investigated 7-hydroxycoumarins.

Microscale sample processing devices for application in bioanalysis and proteome related studies have undergone rapid advancements during the recent years. Several techniques for the preparation of chip-based and capillary-based microfluidic systems and their modification with desired functionalities have been reported. The use of microfluidic devices allow easy processing of extremely small sample quantities with low reagent consumption and offer high surface areas and surface-to-volume ratios. Microscale sample processing devices find application in various research fields such as chemical analysis, kinetic studies, and bioanalytical research. In MS-based proteomics, enzymatic digestion is one of the key steps in the identification of proteins and their posttranslational modifications. However, this is usually the slowest step in the analysis workflow. Thus, the development of suitable devices and methods capable of performing efficient protein digestion more rapidly has become an area of considerable interest. The immobilization of proteolytic enzymes on different kinds of stationary phases, such as monoliths, microparticles and nanoparticles and inner surface of sample transfer capillaries has shown promising results. On the other hand, problems associated with the fabrication and applicability of different stationary phases may pose some challenges, such as monolithic supports involve time consuming fabrication procedures. Similarly, particle-based supports usually involve multistep synthesis and require extended incubation times for protein digestion. Hence, in order to accelerate digestion, other techniques such as microwave irradiation have been employed, which will further complicate the experimental setup. Alternatively, open channel microreactors comprising immobilized enzymes have appeared as a promising alternative, offering minimal restriction over the flow rates. Krenkova et al. prepared open tubular enzyme-immobilized reactors by coupling of trypsin and pepsin to the inner wall of a fused silica capillary. Stigter et al. reported the development of an open tubular trypsin reactor based on amino-modified and carboxyl-modified dextran. RATIONALE Coumarins are naturally occurring, oxygen-containing heterocycles with considerable pharmaceutical potential. For structural elucidation of natural or synthetic coumarins, tandem mass spectrometry (MSn) represents an essential tool. In this study, fragmentation characteristics of twenty-two 7-acetonyloxycoumarins, having promising anti-inflammatory properties, were investigated with low-energy collision-induced dissociation (CID). METHODS Accurate mass measurements were performed on a 12-T Fourier transform ion cyclotron resonance (FT-ICR) instrument. Most CID-MSn measurements were performed on a quadrupole ion trap (QIT) instrument, except some additional CID-MS2 measurements performed on the FT-ICR instrument for further confirmation of some fragment ions. Positive-ion electrospray ionization (ESI) was employed throughout. Density functional theory (DFT) calculations (B3LYP) were carried out to analyze putative ion structures/fragmentation channels. RESULTS The most favourable dissociation channel for [M + H]+ ions of 7-acetonyloxycoumarins was the elimination of a C3H5O● radical (57 Da) from the 7-acetonyloxy group via homolytic bond cleavage. The resulting phenolic radical ion was the primary fragment ion for the most compounds studied. Losses of even-electron neutrals, C3H4O and C3H6O (56 and 58 Da), were also observed. These primary eliminations were accompanied with other characteristic neutral losses from the coumarin skeleton, including H2O, CO, CO2, and C2H2O (ketene). In addition, propene (C3H6) loss was also observed for 4-propyl or 3-ethyl-4-methyl-substituted compounds. CONCLUSIONS The studied coumarins showed interesting characteristics in low-energy CID due to the presence of a 7-acetonyloxy group, leading to both even- and odd-electron product ions. The main dissociation channels observed for each compound were highly dependent on the substituents in the benzopyranone ring. The present results will advance our knowledge on the dissociation characteristics of both synthetic and natural coumarins. Copyright © 2013 John Wiley & Sons, Ltd.

Zinc metalloproteins are one of the most abundant and structurally diverse proteins in nature. In these proteins, the Zn(II) ion possesses a multifunctional role as it stabilizes the fold of small zinc fingers, catalyzes essential reactions in enzymes of all six classes, or assists in the formation of biological oligomers. Previously, a number of database surveys have been conducted on zinc proteins to gain broader insights into their rich coordination chemistry. However, many of these surveys suffer from severe flaws and misinterpretations or are otherwise limited. To provide a more comprehensive, up-to-date picture on zinc coordination environments in proteins, zinc containing protein structures deposited in the Protein Data Bank (PDB) were analyzed in detail. A statistical analysis in terms of zinc coordinating amino acids, metal-to-ligand bond lengths, coordination number, and structural classification was performed, revealing coordination spheres from classical tetrahedral cysteine/histidine binding sites to more complex binuclear sites with carboxylated lysine residues. According to the results, coordination spheres of hundreds of crystal structures in the PDB could be misinterpreted due to symmetry-related molecules or missing electron densities for ligands. The analysis also revealed increasing average metal-to-ligand bond length as a function of crystallographic resolution, which should be taken into account when interrogating metal ion binding sites. Moreover, one-third of the zinc ions present in crystal structures are artifacts, merely aiding crystal formation and packing with no biological significance. Our analysis provides solid evidence that a minimal stable zinc coordination sphere is made up by four ligands and adopts a tetrahedral coordination geometry.

Bradavidin II is a biotin-binding protein from Bradyrhizobium japonicum that resembles chicken avidin and bacterial streptavidin. A biophysical characterization was carried out using dynamic light scattering, native mass spectrometry, differential scanning calorimetry, and isothermal titration calorimetry combined with structural characterization using X-ray crystallography. These observations revealed that bradavidin II differs from canonical homotetrameric avidin protein family members in its quaternary structure. In contrast with the other avidins, bradavidin II appears to have a dynamic (transient) oligomeric state in solution. It is monomeric at low protein concentrations but forms higher oligomeric assemblies at higher concentrations. The crystal structure of bradavidin II revealed an important role for Phe42 in shielding the bound ligand from surrounding water molecules, thus functionally replacing the L7,8 loop essential for tight ligand binding in avidin and streptavidin. This bradavidin II characterization opens new avenues for oligomerization-independent biotin-binding protein development.

Ultrahigh-resolution Fourier transform ion cyclotron resonance mass spectrometry was used for compositional analysis of polar and asphaltene fractions of complex oil residues. The samples were collected before and after the processing of oil in a residue hydrocracking unit, in which the feed oil was the vacuum distillation residue of the crude oil, and the product sample was the residue collected after the processing. From the asphaltene fraction, as many as ∼26 000 peaks were detected by atmospheric pressure photoionization and more than ∼33 000 peaks by positive-ion electrospray ionization (ESI), with up to 18 distinct heteroatom classes identified. Negative-ion ESI provided complementary information through selective ionization of acidic compounds. The syntheses and single crystal X-ray structures of [Ag(5-nitroquinoline)(2)]NO3 (1), [Ag(8-nitroquinoline)(2)]NO3 center dot H2O (2), [Ag(6-methoxy-8-nitroquinoline)(NO3)](n) (3), [Ag(3-quinolinecarbonitrile)(NO3)](n) (4), [Ag(3-quinolinecarbonitrile)(2)]NO3 (5), and [Ag(6-quinolinecarboxylic acid)(2)]NO3 (6) are described. As an alternative to solution chemistry, solid-state grinding could be used to prepare compounds 1 and 3, but the preparation of 4 and 5 in this way failed. The Ag(I) ions in the monomeric compounds 1, 2, 5, and 6 are coordinated to two ligands via the nitrogen atoms of the quinoline rings, thereby forming a distorted linear coordination geometry with Ag-N bond distances of 2.142(2)-2.336(2) angstrom and N-Ag-N bond angles of 163.62(13)degrees-172.25(13)degrees. The 1D coordination polymers 3 and 4 contain Ag(I) centers coordinating one ligand and two bridging nitrate groups, thereby forming a distorted trigonal planar coordination geometry with Ag-N bond distances of 2.2700(14) and 2.224(5) angstrom, Ag-O bond distances of 2.261(4)-2.536(5) angstrom, and N-Ag-O bond angles of 115.23(5)degrees-155.56(5)degrees. Hirshfeld surface analyses of compounds 1-6 are presented as d(norm) and curvedness maps. The d(norm) maps show different interaction sites around the Ag(I) ions, i.e., Ag center dot center dot center dot Ag interactions and possible O-H center dot center dot center dot O, C-H center dot center dot center dot O, C-H center dot center dot center dot N, and C-H center dot center dot center dot C hydrogen bonds. Curvedness maps are a good way of visualizing pi-pi tacking interactions between molecules. The antimicrobial activities of compounds 1, 2, and 6 were screened against 15 different multidrug-resistant strains of bacteria isolated from diabetic foot ulcers and compared to the antimicrobial activities of the clinically used silver sulfadiazine (SS). Compound 2 showed activity similar to SS against this set of test organisms, being active against all strains and having slightly better average silver efficiency than SS (5 vs 6 mu g Ag/mL). Against the standard nonresistant bacterial strains of Staphylococcus aureus, Pseudomonas aeruginosa, Proteus mirabilis, and Streptococcus pyogenes, compound 1 performed better than silver nitrate, with an average MIC of 6 mu g Ag/mL versus 18 mu g Ag/mL for the reference AgNO3. Electrospray ionization mass spectrometry (ESI-MS) analyses of compounds 3 and 6 in DMSO/MeOH confirm the two-coordinated Ag+ complexes in solution, and the results of the H-1 NMR titrations of DMSO solutions of 5-nitroquinoline and 8-nitroquinoline with AgNO3 in DMSO suggest that 5-nitroquinoline is more strongly coordinated to the silver ion.

Tetranuclear AuI–FeII dimetallic and AuI–CuI–FeII trimetallic complexes bearing ferrocenyl (Fc) groups have been assembled by using two triphosphane ligands, namely, (PPh2CH2)2PPh (dpmp) and (PPh2)3CH (tppm). The compositions and structural type of the clusters are dependent on the stereochemistry of the P donor ligands. The complexes [tppmAu3Cu(C2R)3]PF6 [R = Fc (1) and 4-C6H4-Fc (2)] adopt a trigonal pyramidal {Au3Cu} arrangement of the coordinating metal core, whereas for the compounds with the linear triphosphane [Au4(dpmp)2(C2R)2](PF6)2 [R = Fc (3) and 4-C6H4-Fc (4)], a planar rhomboidal {Au4} framework was found. Clusters 1–4 were characterized by NMR spectroscopy and ESI-MS measurements. The solid-state structures of 1, 3, and 4 have been determined by X-ray crystallography. The electrochemical properties of 1–4 were studied and revealed redox processes attributed to the Fc functions. Oxidation of the Fc units in 1, 2, and 4 occurs independently. In 3, an electron transfer process mediated by the {Au4} metal core was observed.

Small zinc finger (ZnF) motifs are promising molecular scaffolds for protein design owing to their structural robustness and versatility. Moreover, their characterization provides important insights into protein folding in general. ZnF motifs usually possess an exceptional specificity and high affinity towards Zn(II) ion to drive folding. While the Zn(II) ion is canonically coordinated by two cysteine and two histidine residues, many other coordination spheres also exist in small ZnFs, all having four amino acid ligands. Here we used high‐resolution mass spectrometry to study metal ion binding specificity and primary coordination sphere robustness of a designed zinc finger, named MM1. Based on the results, MM1 possesses high specificity for zinc with sub‐micromolar binding affinity. Surprisingly, MM1 retains metal ion binding affinity even in the presence of selective alanine mutations of the primary zinc coordinating amino acid residues.

A potentially tridentate hemilabile ligand, PPh2-C6H4-PPh(O)-C6H4-PPh2 (P(3)O), has been used for the construction of a family of bimetallic complexes [MM'(P(3)O)2](2+) (M = M' = Cu (1), Ag (2), Au (3); M = Au, M' = Cu (4)) and their mononuclear halide congeners M(P(3)O)Hal (M = Cu (5-7), Ag (8-10)). Compounds 1-10 have been characterized in the solid state by single-crystal X-ray diffraction analysis to reveal a variable coordination mode of the phosphine-oxide group of the P(3)O ligand depending on the preferable number of coordination vacancies on the metal center. According to the theoretical studies, the interaction of the hard donor P[double bond, length as m-dash]O moiety with d(10) ions becomes less effective in the order CuAgAu. 1-10 exhibit room temperature luminescence in the solid state, and the intensity and energy of emission are mostly determined by the nature of metal atoms. The photophysical characteristics of the monometallic species were compared with those of the related compounds M(P(3))Hal (11-16) with the non-oxidized ligand P(3). It was found that in the case of the copper complexes 5-7 the P(3)O hybrid ligand introduces effective non-radiative pathways of the excited state relaxation leading to poor emission, while for the silver luminophores the P[double bond, length as m-dash]O group leads mainly to the modulation of luminescence wavelength.

Sustainable energy generation is of recent interest due to a growing energy demand across the globe and increasing environmental issues caused by conventional non-renewable means of power generation. In the context of microsystems, portable electronics and lab-on-a-chip based (bio)chemical sensors would essentially require fully integrated, reliable means of power generation. Microfluidic-based fuel cells can offer unique advantages compared to conventional fuel cells such as high surface area-to-volume ratio, ease of integration, cost effectiveness and portability. Here, we summarize recent developments which utilize the potential of microfluidic devices for energy generation.

The major protein of bovine seminal plasma, PDC-109, is a 109-residue polypeptide that exists as a polydisperse aggregate under native conditions. The oligomeric state of this aggregate varies with ionic strength and the presence of lipids. Binding of PDC-109 to choline phospholipids on the sperm plasma membrane results in an efflux of cholesterol and choline phospholipids, which is an important step in sperm capacitation. In this study, Fourier transform ion cyclotron resonance mass spectrometry was used to analyze PDC-109 purified from bovine seminal plasma. In addition to the previously known PDC-109 variants, four new sequence variants were identified by top–down mass spectrometry. For example, a protein variant containing point mutations P10L and G14R was identified along with another form having a 14-residue truncation in the N-terminal region. Two other minor variants could also be identified from the affinity-purified PDC-109. These results demonstrate that PDC-109 is naturally produced as a mixture of several protein forms, most of which have not been detected in previous studies. Native mass spectrometry revealed that PDC-109 is exclusively monomeric at low protein concentrations, suggesting that the protein oligomers are weakly bound and can easily be disrupted. Ligand binding to PDC-109 was also investigated, and it was observed that two molecules of O-phosphorylcholine bind to each PDC-109 monomer, consistent with previous reports. Treatment of Au(SC(4)H(8))Cl with a stoichiometric amount of hydroxyaliphatic alkyne in the presence of NEt(3) results in high-yield self-assembly of homoleptic clusters (AuC(2)R)(10) (R = 9-fluorenol (1), diphenylmethanol (2), 2,6-dimethyl-4-heptanol (3), 3-methyl-2-butanol (4), 4-methyl-2-pentanol (4), 1-cyclohexanol (6), 2-borneol (7)). The molecular compounds contain an unprecedented catenane metal core with two interlocked 5-membered rings. Reactions of the decanuclear clusters 1-7 with gold-diphosphine complex [Au(2)(1,4-PPh(2)-C(6)H(4)-PPh(2))(2)](2+) lead to octanuclear cationic derivatives [Au(8)(C(2)R)(6)(PPh(2)-C(6)H(4)-PPh(2))(2)](2+) (8-14), which consist of planar tetranuclear units {Au(4)(C(2)R)(4)} coupled with two fragments [AuPPh(2)-C(6)H(4)-PPh(2)(AuC(2)R)](+). The titled complexes were characterized by NMR and ESI-MS spectroscopy, and the structures of 1, 13, and 14 were determined by single-crystal X-ray diffraction analysis. The luminescence behavior of both Au(I)(10) and Au(I)(8) families has been studied, revealing efficient room-temperature phosphorescence in solution and in the solid state, with the maximum quantum yield approaching 100% (2 in solution). DFT computational studies showed that in both Au(I)(10) and Au(I)(8) clusters metal-centered Au → Au charge transfer transitions mixed with some π-alkynyl MLCT character play a dominant role in the observed phosphorescence.

Non-covalent complexation of mono- and bisphosphonates by hexaglucosylthioureidocalix[6]arene and octaglucosylthioureidocalix[8]arene was studied by electrospray ionisation Fourier transform ion cyclotron resonance mass spectrometry. Glucosylthioureidocalix[8]arene formed 1:1 and 1:2 complexes with bisphosphonates, with a marked preference towards risedronate, clodronate, zoledronate and etidronate. In contrast, up to four guest molecules were bound in the case of monophosphonates, suggesting a different type of binding in comparison to bisphosphonates. Hexaglucosylthioureidocalix[6]arene also formed complexes with phosphonates with a somewhat similar binding selectivity. Chemically powered micromotors represent an exciting research area in nanotechnology. Such artificial devices are typically driven by catalytic bubble formation, taking place at the solid-liquid interface. Platinum has been most frequently used for the fabrication of different micromotors due to its superior catalytic efficiency. Other materials typically suffer from slow speeds and require very high concentrations of chemical fuel. Here, we report preparation and characterization of fast moving micromotors based on manganese oxide (MnO2) with different geometrical shapes (tubes, rods, and spheres). On the basis of the results, the prepared micromotors reached the highest speeds (up to ∼900 μm s(-1) in 10% H2O2) reported to date for any MnO2-based micromotors. Moreover, they moved by bubble propulsion even at very low concentrations of peroxide fuel. Thus, MnO2 represents a promising material for the preparation of micromotors for various biomedical or environmental applications, where high speeds are desired.

Reactions of the homoleptic (AuC(2)R)(n) precursors with stoichiometric amount of diphosphine ligand PPh(2)C(6)H(4)PPh(2) (P^P) and Cu(+) ions lead to an assembly of a new family of bimetallic clusters [Au(6)Cu(2)(C(2)R)(6)(P^P)(2)](2+) (type I; R=9-fluorenolyl (1), diphenylmethanolyl (2), 2,6-dimethyl-4-heptanolyl (3), 1-cyclohexanolyl (4), Cy (5), tBu (6)). In the case of R=1-cyclohexanolyl, a structurally different complex [Au(6)Cu(2)(C(2)C(6)H(11)O)(6)(P^P)(3)](2+) (7, type II) could be obtained by treatment of 4 with one equivalent of the diphosphine, while for R=isopropanolyl only the latter type of cluster [Au(6)Cu(2)(C(2)C(3)H(7)O)(6)(P^P)(3)](2+) (8) was detected. Steric bulkiness of the alkynyl ligands and O···H-O hydrogen bonding are suggested to play an important role in stabilizing the type I and type II cluster structural motif, respectively. All the complexes exhibit intense photoluminescence in solution with emission parameters that depending on the geometrical arrangement of the octanuclear metal core. The clusters 1-4 and 6 show single emission band in a blue region (469-488 nm) with maximum quantum yield of 94% (4), while structurally different 7 and 8 emit yellow-orange (590 nm) with unity quantum efficiency. The theoretical DFT calculations of the electronic structures have been carried out to demonstrate that the metal-centered triplet emission within the heterometallic core plays a key role for the observed phosphorescence.

The noncovalent complexation of three glucosylcalix[4]arenes (1–3) towards 23 mono- and dicarboxylic acid anions were studied by ESI-FTICR mass spectrometry. Competitive complexation, collision-induced dissociation and gas-phase H/D-exchange experiments were performed to obtain information on selectivity of calixarenes towards carboxylates and characteristics of their complexes. The flexibility and number of glucose units of the host and the spatial disposition of the hydrogen bonding groups on the carboxylate guests were found to affect the selectivity of complexation strongly. The glucosylcalixarenes exhibited particular selectivity for dicarboxylic acid anions incorporating π-systems, and clear isomeric selectivity was observed for isophthalic among phthalic acid anions and for fumaric acid over maleic acid anion. Synthesis and X-ray single crystal structure analysis of the compound {[Ag2(μ2-bpym)(μ-O-NO3)2]}n, (1), (where bpym = 2,2′-bipyrimidine) are presented. Compound (1) has a (6,3)-2D honeycomb structure with a tetrahedral coordination geometry around the Ag(I) ion. In contrary to the solid state structural investigation, ESI-MS for (1) in solution shows a strong peak at m/z 423.0269 which indicates that the [Ag(bpym)2]+ cation is dominating instead of [Ag2(bpym)]2+. The anti-microbial activity of (1) was screened against 15 multi-drug resistant bacteria in comparison to silver(I) sulphadiazine and it showed a high activity against Burkholderia mallei which causes glanders; with a MIC value of 4 μg/ml.

The gas-phase behavior and stability of [RuX2(CO)2(dcbpy)], [OsX2(CO)2(dcbpy)], and [OsI2(CO)2(mcbpy)] (X = Cl, Br, I; dcbpy = 2,2′-bipyridine-4,4′-dicarboxylic acid; mcbpy = 2,2′-bipyridine-4-carboxylic acid) were studied by electrospray ionization (ESI) Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry. Negative-ion ESI produced abundant singly and doubly deprotonated molecules for all of the compounds, without apparent changes in the metal atom oxidation state or the ligand coordination. The characteristic fragment ions resulting from the decarboxylation (loss of CO2) in one of the carboxylic acid substituents of the dcbpy ligand were also observed. The gas-phase fragmentation was investigated by means of collision-induced dissociation (CID) and infrared multiphoton dissociation (IRMPD) techniques. Synthetic micro‐ and nanomotors (MNMs) are tiny objects that can autonomously move under the influence of an appropriate source of energy, such as a chemical fuel, magnetic field, ultrasound, or light. Chemically driven MNMs are composed of or contain certain reactive material(s) that convert chemical energy of a fuel into kinetic energy (motion) of the particles. Several different materials have been explored over the last decade for the preparation of a wide variety of MNMs. Here, the discovery of materials and approaches to enhance the efficiency of chemically driven MNMs are reviewed. Several prominent applications of the MNMs, especially in the fields of biomedicine and environmental science, are also discussed, as well as the limitations of existing materials and future research directions.
