The stability of many MOFs is not satisfactory, which severely limits the exploration of their potential applications. Given this, we have proposed a strategy to improve the stability of MOFs by introducing alkali metal K + capable of coordinating with metal nodes, which finally induces the interpenetrating uranyl-porphyrin framework to connect as a whole ( IHEP-9 ). The stability experiments reveal that the IHEP-9 has good thermal stability up to 400 °C and can maintain its crystalline state in the aqueous solution with pH ranging from 2 to 11. The catalytic activity of IHEP-9 as a heterogeneous photocatalyst for CO 2 cycloaddition under the driving of visible light at room temperature is also demonstrated. This induced interpenetration and fixation method may be promising for the fabrication of more functional MOFs with improved structural stability.

Introduction: Liver cancer-secreted serine protease inhibitor Kazal (LC-SPIK) is a protein that is specifically elevated in cases of hepatocellular carcinoma (HCC). We assessed the performance of LC-SPIK in detecting HCC, including its early stages, in patients with cirrhosis, hepatitis B virus (HBV), and hepatitis C virus (HCV)., Methods: We enrolled 488 patients, including 164 HCC patients (81 early HCC) and 324 controls in a blinded, prospective, case-control study. Serum LC-SPIK levels were determined by an enzyme-linked immunosorbent assay-based assay. The performance of serum LC-SPIK and α-fetoprotein (AFP), including area under the curve (AUC), sensitivity, and specificity, are compared. The performance of LC-SPIK was evaluated in an independent validation cohort with 102 patients., Results: In distinguishing all HCC patients from those with cirrhosis and chronic HBV/HCV, LC-SPIK had an AUC of 0.87, with 80% sensitivity and 90% specificity using a cutoff of 21.5 ng/mL. This is significantly higher than AFP, which had an AUC of 0.70 and 52% sensitivity and 86% specificity using a standard cutoff value of 20.0 ng/mL. For early-stage HCC (Barcelona Clinic Liver Cancer stage 0 and A), LC-SPIK had an AUC of 0.85, with 72% sensitivity and 90% specificity, compared with AFP, which had an AUC of 0.61, with 42% sensitivity and 86% specificity. In addition, LC-SPIK accurately detected the presence of HCC in more than 70% of HCC patients with false-negative AFP results., Discussion: The study provided strong evidence that LC-SPIK detects HCC, including early-stage HCC, with high sensitivity and specificity, and might be useful for surveillance in cirrhotic and chronic HBV/HCV patients, who are at an elevated risk of developing HCC.

In this study, six different types of residential areas in Guiyang were selected as the research objects, including high-rise high-density, high-rise low-density, middle-rise high-density, middle-rise low-density, low-rise high-density and low-rise low-density. The indices of sunshine compliance rate and the building's sunshine hour ratio were constructed to compare and analyze sunshine environment across those six different residential areas. The factors influencing sunshine environment in different residential areas were studied. The results showed that the average sunshine compliance rates of the six types of residential areas were 36.9%, 61.9%, 20.6%, 69.6%, 26.5% and 45.0%, respectively. The average sunshine compliance rate of low-density residential areas was 2.25 times higher than that of high-density residential areas within the same type, among which the sunshine environment of low-density residential areas was better. The sunshine environment of different types of low-density residential areas was different. The sunshine hours for high-rise low-density and middle-rise low-density forms were concentrated in 5-6 hours, while the building's sunshine hour ratio was 0.24 and 0.32, respectively. The sunshine hours for low-rise low-density forms were mainly 6-7 hours, with a building's sunshine hour ratio of 0.28. Compared with other types of residential areas, the low-rise low-density type of sunlight environment was the best. The plot ratio was significantly positively correlated with the building's sunlight ratio of 0-1 h sunlight hours in the residential area.

Herein we present a new viologen-based radical-containing metal-organic framework (RMOF) Gd-IHEP-7, which upon heating in air undergoes a single-crystal-to-single-crystal transformation to generate Gd-IHEP-8. Both RMOFs exhibit excellent air and water stability as a result of favorable radical-radical interactions, and their long-lifetime radicals result in wide spectral absorption in the range 200-2500 nm. Gd-IHEP-7 and Gd-IHEP-8 show excellent activity toward solar-driven nitrogen fixation, with ammonia production rates of 128 and 220 μmol h -1  g -1 , respectively. Experiments and theoretical calculations indicate that both RMOFs have similar nitrogen fixation pathways. The separation of actinides has a vital place in nuclear fuel reprocessing, recovery of radionuclides, and remediation of environmental contamination. Here we propose a new paradigm of nanocluster-based actinide separation, namely, nanoextraction, that can achieve efficient sequestration of uranium in an unprecedented form of giant coordination nanocages using a cone-shaped macrocyclic pyrogallol[4]arene as the extractant. The U 24 -based hexameric pyrogallol[4]arene nanocages with distinctive [U 2 (PG) 2 ] binuclear units (PG = pyrogallol) that rapidly assembled in situ in monophasic solvent were identified by single-crystal X-ray diffraction, MALDI-TOF mass spectrometry, NMR spectroscopy, and small-angle X-ray and neutron scattering. Comprehensive biphasic extraction studies showed that this novel separation strategy has enticing advantages such as fast kinetics, high efficiency, and good selectivity over lanthanides, thereby demonstrating its potential for efficient separation of actinide ions.

Elastic metal-organic materials (MOMs) capable of multiple stimuli-responsiveness based on dual-stress and thermally responsive triple-helix coordination polymers are presented. The strong metal-coordination linkage and the flexibility of organic linkers in these MOMs, rather than the 4 Å stacking interactions observed in organic crystals, causes the helical chain to act like a molecular spring and thus accounts for their macroscopic elasticity. The thermosalient effect of elastic MOMs is reported for the first time. Crystal structure analyses at different temperatures reveal that this thermoresponsiveness is achieved by adaptive regulation of the triple-helix chains by fine-tuning the opening angle of flexible V-shaped organic linkers and rotation of its lateral conjugated groups to resist possible expansion, thus demonstrating the vital role of adaptive reorganization of triple-helix metal-organic chains as a molecular spring-like motif in crystal jumping., (© 2020 Wiley-VCH Verlag GmbH & Co. Nicotine is proved to be an important factor for cardiac hypertrophy. Autophagy is important cell recycling system involved in the regulation of cardiac hypertrophy. Cilostazol, which is often used in the management of peripheral vascular disease. However, the effects of cilostazol on nicotine induced autophagy and cardiac hypertrophy are unclear. Here, we aim to determine the role and molecular mechanism of cilostazol in alleviating nicotine-induced cardiomyocytes hypertrophy through modulating autophagy and the underlying mechanisms. Our results clarified that nicotine stimulation caused cardiomyocytes hypertrophy and autophagy flux impairment significantly in neonatal rat ventricular myocytes (NRVMs), which were evidenced by augments of LC3-II and p62 levels, and impaired autophagosomes clearance. Interestingly, cathepsin B (CTSB) activity decreased dramatically after stimulation with nicotine in NRVMs, which was crucial for substrate degradation in the late stage of autophagy process, and cilostazol could reverse this effect dramatically. Intracellular ROS levels were increased significantly after nicotine exposure. Meanwhile, p38MAPK and JNK were activated after nicotine treatment. By using ROS scavenger N-acetyl-cysteine (NAC) could reverse the effects of nicotine by down-regulation the phosphorylation of p38MAPK and JNK pathways, and pretreatment of specific inhibitors of p38MAPK and JNK could restore the autophagy impairment and cardiomyocytes hypertrophy induced by nicotine. Moreover, CTSB activity of lysosome regained after the treatment with cilostazol. Cilostazol also inhibited the ROS accumulation and the activation of p38MAPK and JNK, which providing novel connection between lysosome CTSB and ROS/p38MAPK/JNK related oxidative stress pathway. This is the first demonstration that cilostazol could alleviate nicotine induced cardiomyocytes hypertrophy through restoration of autophagy flux by activation of CTSB and inhibiting ROS/p38/JNK pathway, exhibiting a feedback loop on regulation of autophagy and cardiomyocytes hypertrophy., Competing Interests: Competing Interests: The authors have declared that no competing interest exists., (© The author(s).)

The incorporation of a mechanically interlocked molecule such as pseudorotaxane into metal-organic coordination polymers has afforded plenty of new hybrid materials with special structures and unique properties. In this work, we employ a weakly bound cucurbit[6]uril (CB[6])-bipyridinium pseudorotaxane as a supramolecular precursor to assemble with uranyl, aiming to construct uranyl-rotaxane coordination polymers (URCPs) with intriguing structures. By adjusting the synthetic conditions, a new kinked-helix uranyl rotaxane compound ( URCP3 ), together with three other compounds URCP1 , URCP2 , and URCP4 varying from 1D chains to 2D interwoven networks, was obtained. Detailed structural analyses indicate that the pseudorotaxane ligand ( C8BPCA @CB[6]) shows great configuration diversity in the construction of URCPs, which is most probably due to the weak binding strength between the host and guest molecules. Specifically, based on the monodentate coordination of the end carboxyl groups of C8BPCA forced by the surrounding unilaterally-chelated oxalate, the entire flexible pseudorotaxane linker will be more likely to undergo conformational change, thereby binding to the uranyl center from both sides of the uranyl equatorial plane and promoting the formation of a kinked helix structure of URCP3 that is shaped like a Chinese knot along [001]. This work enriches the library of actinide-rotaxane compounds and provides a new approach to construct metal-organic compounds with complicated structures using weakly bonded pseudorotaxanes as well.

Scope: β-Cryptoxanthin (BCX) can be cleaved by both β-carotene 15,15'-oxygenase (BCO1) and β-carotene 9',10'-oxygenase (BCO2), generating biological active vitamin A and apocarotenoids. We examined whether BCX feeding could inhibit diethylnitrosamine (DEN)-initiated, highly refined carbohydrate diet (HRCD)-promoted hepatocellular carcinoma (HCC) development, dependent or independent of BCO1/BCO2 activity., Methods and Results: Two-week-old male wild-type (WT) and BCO1 -/- /BCO2 -/- double knockout (DKO) mice are given a single intraperitoneal injection of DEN (25 mg kg -1 body weight) to initiate hepatic carcinogenesis. At 6 weeks of age, all animals are fed HRCD (66.5% of energy from carbohydrate) with or without BCX for 24 weeks. BCX feeding increases hepatic vitamin A levels in WT mice, but not in DKO mice that shows a significant accumulation of hepatic BCX. Compared to their respective HRCD littermates, both WT and DKO fed BCX have significantly lower HCC multiplicity, average tumor size, and total tumor volume, and the steatosis scores. The chemopreventive effects of BCX are associated with increased p53 protein acetylation and decreased protein levels of lactate dehydrogenase and hypoxia-inducible factor-1α in tumors., Conclusion: This study suggests that BCX feeding may alleviate HRCD-promoted HCC progression by modulating the acetylation of p53, hypoxic tumor microenvironment, and glucose metabolism, independent of BCO1/BCO2., (© 2019 WILEY-VCH Verlag GmbH & Co. Two novel thorium-based organic frameworks (Th-IHEP-5 and Th-IHEP-6) were assembled from a hexanuclear thorium cluster, porphyrin derivative ligand and linear carboxylic acid ligands via a mixed-ligand strategy. As a stable heterogeneous catalyst, Th-IHEP-5 exhibited high photocatalytic activity for the oxidation of 2-chloroethyl ethyl sulfide (CEES) and the fixation of CO2. The good catalytic effect is attributed to the large conjugated system of porphyrin and the photosensitizer enhancing effects of bipyridine.

Template synthesis is one of the most feasible ways to explore new uranyl compounds with intriguing structures and properties. Here we demonstrate the preparation of six novel "sandwichlike" uranyl coordination polymers (UCPs) based on two-dimensional uranyl-terephthalate acid (H 2 TP ) networks using CB n ( n = 5, 6, 8) as template ligands in the presence of different cations (Na + , K + , Cs + , or H 2 N(CH 3 ) 2 + ). Compound 1 ([UO 2 ( TP ) 2 ][Na 2 (CB5)(H 2 O)](H 2 O) 5 ) is composed of layered uranyl- TP networks with the complex of CB5 and sodium cations as template ligands. In compound 2 ([(UO 2 ) 2 ( TP ) 3 ] 2 (CB6)(H 2 O) 10 ), CB6 located between uranyl- TP networks contacts them by π-π interactions and hydrogen bonds. Compound 3 ([(UO 2 ) 2 ( TP ) 3 ] 2 [Na 2 (H 2 O) 10 (CB6)]) is the same as compound 2 except for sodium cations bonding with CB6. Similarly in compound 4 ([(UO 2 ) 2 ( TP ) 3 ][Cs(H 2 O) 3 (CB6)]), CB6 is a capsulelike structure capped with two cesium cations and interacts with uranyl- TP networks through π-π and C-H···π interactions. Compound 5 ([(UO 2 ) 2 ( TP ) 3 (HCOO) 2 ][K(H 2 O) 2 (CB5)] 2 [H 2 N(CH 3 ) 2 ] 2 (CB6)(H 2 O) 6 ) consists of both templates of CB6 and CB5 in which each CB5 is capped with one potassium cation while the H 2 N(CH 3 ) 2 + cation is held at CB6 portals. In compound 6 ([(UO 2 ) 2 ( TP ) 3 ] 2 [UO 2 ( TP ) 2 (H 2 O) 2 ][Cs(CB8) 3 (H 2 O) 4 ](H 2 O) 16 ), CB8 ligands are connected by cesium cations to form a triangle motif and are further located between the uranyl- TP networks as template agents. All of the 2D layered structures with free CB n or cation-anchored CB n intercalate into the laminates of uranyl-terephthalate and shows a cucurbituril-mediated structural evolution. The regulating role of CB n as structure-directing template agents for the construction of layered UCPs through outer-surface interactions with layers of uranyl terephthalate is demonstrated, especially for the case of CB6 with contractive interlayer spacing.

Direct acting antiviral (DAA) treatments may reduce the elevated α fetoprotein (AFP), but data on how these treatments affect elevated AFP in patients with chronic hepatitis C (CHC) remain insufficient. In the present study, the frequency of baseline AFP elevations and their related factors, AFP dynamics during and after DAA treatment, and factors associated with AFP reduction was assessed. This retrospective study included 141 patients with CHC without hepatocellular carcinoma who received DAA and achieved sustained virological response. The details are as follows: mean post-treatment follow-up was 99 weeks (12-213); mean age, 57.8 years old; 52%, males; 79%, genotype (GT) 1; and 47%, cirrhosis. Pre-treatment AFP elevation (> 5.5 ng/mL) was seen in 48.2% patients. On multivariate analysis, baseline AFP > 5.5 was associated with the presence of cirrhosis (P =0.001), coexisting non-alcoholic steatohepatitis (NASH) (P = 0.035), and GT 1 (P = 0.029). AFP normalization was seen in 28.2% patients at treatment week 2, in 52% at the end of treatment, and in 73.4% at the end of follow-up. Post-treatment week 24 AFP normalization was associated with the absence of cirrhosis (P = 0.003), Child-Pugh score < 6 (P = 0.015), and baseline AFP < 10 (P = 0.015). AFP elevation is common in patients with CHC and independently associated with NASH, cirrhosis, and GT 1. DAA treatment resulted in AFP normalization as early as treatment week 2. Post-treatment week 24 AFP normalization is independently associated with the absence of cirrhosis, Child-Pugh score < 6, and baseline AFP < 10.

As well-known functional groups with excellent electro/photochromic and ion-exchange properties, bipyridinium motifs have been used in functionalized metal-organic coordination polymers, but they are still rarely applied to construct actinide coordination polymers. In this work, we utilized a bipyridinium-based carboxylic acid, 1,1'-bis(4-carboxyphenyl)-4,4'-bipyridinium bis(chloride) ([H 2 bcbp]Cl 2 ), as the organic ligand to assemble with uranyl cations. By the introduction of different kinds of auxiliary ligands and adjustment of the pH, five novel uranyl coordination compounds, 1 - 5 , have been synthesized through hydrothermal reactions. Starting from uranyl ions and terephthalic acid (H 2 TP) and H 2 bcbp ligands, [(UO 2 ) 2 (bcbp)(TP) 2 ]·3H 2 O ( 1 ) has a wave-shaped two-dimensional (2D) structure consisting of dinuclear units connected by terephthalate linkers and further supported by the longer H 2 bcbp ligands. [(UO 2 ) 2 (bcbp)(PA) 2 ]·4H 2 O ( 2 ) has a zigzag chain of dimeric uranium units, and [(UO 2 ) 2 (bcbp)(bpdc) 2 ]·5H 2 O ( 3 ) forms a one-dimensional ribbonlike structure. The 2D structures of [(UO 2 )(bcbp)(OH)(H 2 O)]·Cl ( 4 ) and [(UO 2 )(bcbp)Cl]·Cl ( 5 ) are similar, both of which are constructed from dinuclear uranyl units and bcbp 2- ligands. Furthermore, the performance for perrhenate removal of compound 4 with a cationic framework is assessed, and we found that compound 4 can efficiently remove ReO 4 - from an aqueous solution in a wide range of pH values. This work extends the library of viologen derivative-based uranyl coordination polymers, provides to some extent broader insights into actinide coordination chemistry of functionalized ligands, and may facilitate the ion-exchange applications of related coordination polymers.

Background: β-Cryptoxanthin (BCX), a provitamin A carotenoid shown to protect against nonalcoholic fatty liver disease (NAFLD), can be cleaved by β-carotene-15,15'-oxygenase (BCO1) to generate vitamin A, and by β-carotene-9',10'-oxygenase (BCO2) to produce bioactive apo-carotenoids. BCO1/BCO2 polymorphisms have been associated with variations in plasma carotenoid amounts in both humans and animals., Objectives: We investigated whether BCX feeding inhibits high refined-carbohydrate diet (HRCD)-induced NAFLD, dependent or independent of BCO1/BCO2., Methods: Six-week-old male wild-type (WT) and BCO1-/-/BCO2-/- double knockout (DKO) mice were randomly fed HRCD (66.5% of energy from carbohydrate) with or without BCX (10 mg/kg diet) for 24 wk. Pathological and biochemical variables were analyzed in the liver and mesenteric adipose tissues (MATs). Data were analyzed by 2-factor ANOVA., Results: Compared to their respective HRCD controls, BCX reduced hepatic steatosis severity by 33‒43% and hepatic total cholesterol by 43‒70% in both WT and DKO mice (P < 0.01). Hepatic concentrations of BCX, but not retinol and retinyl palmitate, were 33-fold higher in DKO mice than in WT mice (P < 0.001). BCX feeding increased the hepatic fatty acid oxidation protein peroxisome proliferator-activated receptor-α, and the cholesterol efflux gene ATP-binding cassette transporter5, and suppressed the lipogenesis gene acetyl-CoA carboxylase 1 (Acc1) in the MAT of WT mice but not DKO mice (P < 0.05). BCX feeding decreased the hepatic lipogenesis proteins ACC and stearoyl-CoA desaturase-1 (3-fold and 5-fold) and the cholesterol synthesis genes 3-hydroxy-3-methyl-glutaryl-coenzyme A reductase and HMG-CoA synthase 1 (2.7-fold and 1.8-fold) and increased the cholesterol catabolism gene cholesterol 7α-hydroxylase (1.9-fold) in the DKO but not WT mice (P < 0.05). BCX feeding increased hepatic protein sirtuin1 (2.5-fold) and AMP-activated protein kinase (9-fold) and decreased hepatic farnesoid X receptor protein (80%) and the inflammatory cytokine gene Il6 (6-fold) in the MAT of DKO mice but not WT mice (P < 0.05)., Conclusion: BCX feeding mitigates HRCD-induced NAFLD in both WT and DKO mice through different mechanisms in the liver-MAT axis, depending on the presence or absence of BCO1/BCO2., (Copyright © American Society for Nutrition 2019.)

Objective: The aim of this study was to investigate the effect of microRNA-150 on the regulation of myocardial fibrosis and ventricular remodeling in rats with acute myocardial infarction (AMI)., Materials and Methods: The AMI rats model was established by the ligation of the left anterior descending coronary artery (LAD) in vivo. After AMI procedures, the rats were injected with microRNA-150 lentivirus overexpression or negative control, respectively. Cardiac function of rats was evaluated by echocardiography. Hematoxylin and eosin (HE) staining and Masson trichrome were performed to evaluate myocardial fibrosis in each rat. Meanwhile, cardiomyocyte apoptosis was detected by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) method. The expression levels of microRNA-150, col1α1, col1α2, col3 and α smooth muscle actin (α-SMA) in the border zone of rat infarct myocardium were detected by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR) and Western blot, respectively., Results: MicroRNA-150 expression in the border zone of infarct myocardium decreased significantly at day 28 after AMI (p<0.05). Overexpressing microRNA-150 significantly improved cardiac function, decreased collagen volume fraction (CVF) and attenuated cardiomyocyte apoptosis in rats. Furthermore, the expression levels of col1ɑ1, col1ɑ2, col3 and α-SMA in the border zone of infarct myocardium were remarkably down-regulated in rats overexpressing microRNA-150 compared with those of controls (p<0.001)., Conclusions: MicroRNA-150 expression in the border zone of rat infarct myocardium decreased at day 28 after AMI. In addition, the upregulation of microRNA-150 in myocardial tissue could inhibit myocardial fibrosis and improve ventricular remodeling at post-AMI.

As a less explored avenue, actinide-based metal-organic frameworks (MOFs) are worth studying for the particularity of actinide nodes in coordination behaviour and assembly modes. In this work, an azobenzenetetracarboxylate-based anionic MOF supported by uranyl-carboxyl helical chain units was synthesized, incorporating linear uranyl as the metal centre. This kind of helical chain-type building unit is reported for the first time in uranyl-based MOFs. Structural analysis reveals that the formation of helical chain secondary units can be attributed to restricted equatorial coordination of rigid flat azobenzene ligand to uranyl centres. Meanwhile, this newly-synthesized anionic material has been used to remove Eu 3+ ions, as a non-radioactive surrogate of Am 3+ ion, through an ion-exchange process with [(CH 3 ) 2 NH 2 ] + ions in its open channels, as evidenced by a combination of 1 H NMR spectroscopy, EDS and PXRD., (© 2019 Wiley-VCH Verlag GmbH & Co. Background: Current diagnosis of hepatitis C virus (HCV) infection requires two sequential steps: testing for anti-HCV followed by HCV RNA PCR to confirm viremia. We have developed a highly sensitive and specific HCV-antigens enzyme immunoassay (HCV-Ags EIA) for one-step diagnosis of viremic HCV infection., Aim: To assess the clinical application of the HCV-Ags EIA in one-step diagnosis of viremic HCV infection in human immunodeficiency virus (HIV)-coinfected individuals., Methods: The study blindly tested HCV-Ags EIA for its performance in one-step diagnosing viremic HCV infection in 147 sera: 10 without HCV or HIV infection; 54 with viremic HCV monoinfection; 38 with viremic HCV/HIV coinfection; and 45 with viremic HCV and non-viremic HIV coinfection., Results: Upon decoding, it was 100% accordance of HCV-Ags EIA to HCV infection status by HCV RNA PCR test. In five sera with HCV infection, HCV RNA was as low as 50-59 IU/mL, and four out of five tested positive for HCV-Ags EIA. Likewise, it was also 100% accordance of HCV-Ags EIA to HCV infection status by HCV RNA PCR in 83 sera with HCV and HIV coinfection, regardless if HIV infection was active or not., Conclusion: The modified HCV-Ags EIA has a lower detection limit equivalent to serum HCV RNA levels of approximately 100 IU/mL. It is highly sensitive and specific in the setting of HIV coinfection, regardless of HIV infection status and CD4 count. Objectives: Chronic hepatitis B (CHB) can progress into liver fibrosis and cirrhosis with poor outcomes. Early and accurate diagnosis of liver fibrosis/cirrhosis is important to guide the preventive strategy of their related complications., Methods: A Chinese multicenter cross-sectional study was conducted to develop and validate a novel noninvasive program for staging liver fibrosis in untreated patients with CHB. Liver histology was evaluated independently by 2 pathologists. The alanine aminotransferase ratio, Hepascore, and aspartate aminotransferase to platelet index values were calculated. Liver stiffness measurement (LSM) and diameter of the spleen were measured. Logistic regression with ℓ1 penalty of regression coefficients was used to select the optimal predictors. The diagnostic accuracy for the stage of liver fibrosis was assessed by the area under the receiver operator characteristic curve with 95% confidence interval (CI)., Results: A total of 1,200 patients with CHB were included, of whom 800 and 400 were in training and validation sets, respectively. LSM, platelets, age, hyaluronic acid, and diameter of the spleen were the top 5 predictors associated with any stage of liver fibrosis and integrated into a novel noninvasive program, named as the Chin-CHB score. The area under the receiver operator characteristic curve of the Chin-CHB score was 0.893 (95% CI: 0.77-0.92) for diagnosing significant fibrosis, 0.897 (95% CI: 0.85-0.95) for advanced fibrosis, and 0.909 (95% CI: 0.87-0.95) for cirrhosis. The diagnostic performance of the Chin-CHB score was similar between training and validation sets. The Chin-CHB score had better diagnostic performance than aspartate aminotransferase to platelet index, alanine aminotransferase ratio, LSM alone, and Hepascore for diagnosing any stage of liver fibrosis., Conclusions: The Chin-CHB score had good diagnostic performance for any stage of liver fibrosis.

Bipyridine organic bases are beneficial to the synthesis of novel uranyl-organic hybrid materials, but the relationship between their molecular structures and specific roles as structure-directing agents, especially for the semirigid dicarboxylate systems, is still unclear. Here we demonstrate how the bipyridine ligands direct the coordination assembly of uranyl-organic compounds with a semirigid dicarboxylate linker, 4,4'-dicarboxybiphenyl sulfone (H 2 dbsf), by utilizing a series of bipyridine ligands, 1,10-phenanthroline (phen), 2,2'-bipyridine (2,2'-bpy), 5,5'-dimethylbipyridine (5,5'-dmbpy), 4,4'-bipyridine (4,4'-bpy), or 1,3-di(4-pyridyl)propane (bpp). Under hydrothermal conditions, eight uranyl-organic coordination polymers (UCPs), four of which [[UO 2 (dbsf)(phen)] (1), [UO 2 (dbsf)(phen)]·H 2 O (1'), [U 4 O 10 (dbsf) 3 ] 2 [H 2 bpp] 2 (6), and [U 4 O 10 (dbsf) 3 ] 2 [H 2 bpp] (6')] were reported previously, were synthesized and divided into two types based on the chelate or template effect of these bipyridine ligands. 1, 1', [UO 2 (dbsf)(2,2'-bpy)] (2), and [(UO 2 ) 2 (dbsf) 2 (5,5'-dmbpy) 2 ] (3) are springlike triple helices with bipyridine ligands (phen, 2,2'-bpy, or 5,5'-dmbpy) as chelate ligands, while [U 4 O 10 (dbsf) 3 ][H 2 (4,4'-bpy)] (4), [U 4 O 10 (dbsf) 3 ] 2 [H(4,4'-bpy)] 2 [Ni(H 2 O) 6 ] (5), 6, and 6' are tetranuclear uranyl-mediated 2-fold-interpenetrating networks with 4,4'-bpy or bpp as template ligands and charge-balancing agents. The participation or not in uranyl coordination of different bipyridine ligands promotes not only diversity in uranyl speciation and final topological structures among different classes of organic bases but also consistency for the same types of bipyridine ligands, which thus endows the possibility of the rational design of UCPs based on semirigid dicarboxylate ligands with the aid of cautiously selected bipyridine ligands.

Efficient anion recognition is of great significance for radioactive 99 TcO 4 - decontamination, but it remains a challenge for traditional sorbents. Herein, we put forward a tactic using soft crystalline cationic material with anion-adaptive dynamics for 99 TcO 4 - sequestration. A cucurbit[8]uril-based supramolecular metal-organic material is produced through a multi-component assembly strategy and used as a sorbent for effective trapping of TcO 4 - . Excellent separation of TcO 4 - /ReO 4 - is demonstrated by fast removal kinetics, good sorption capacity and high distribution coefficient. Remarkably, the most superior selectivity among metal-organic materials reported so far, together with good hydrolytic stability, indicates potential for efficient TcO 4 - removal. The structure incorporating ReO 4 - reveals that the supramolecular framework undergoes adaptive reconstruction facilitating the effective accommodation of TcO 4 - /ReO 4 - . The results highlight opportunities for development of soft anion-adaptive sorbents for highly selective anion decontamination.

Pseudorotaxane-type ligands with tunable structural dynamics offer an opportunity in the exploration of new actinide hybrid materials. In this work, we utilized a weakly bonded pseudorotaxane ligand involving CB[6] and 1, 1'-(heptane-1, 7-diyl)bis(4-(ethoxycarbonyl)pyridin-1-ium) bromides ([C7BPCEt]Br 2 @CB[6]) to assemble with uranyl ion, and we systematically investigated the effect of different factors including pH and competing ligands on the hydrothermal synthesis of URCPs. Nine uranyl-rotaxane coordination polymers (URCPs) with diversity in coordination mode and topological structure were successfully prepared (two previously reported complexes, URCP1 and URCP2 are also included). The results indicate that sulfate, bromide, CB[6], and C7BPCA (the hydrolyzate of [C7BPCEt]Br 2 ) show a combined influence on the obtained URCPs. At low pH, both CB[6] and C7BPCA can bond with uranyl centers and produce interwoven structures in URCP1, URCP2, and URCP6; at high pH, C7BPCA and competing anions (sulfate and bromide) have priority to coordinate with uranyl ions in URCP3-URCP5 and URCP7-URCP9. Notably, for the first time, bromide anion with lower affinity to uranyl ions is also observed in solid-state uranyl coordination polymer (URCP7-URCP9), which has been demonstrated by both energy dispersive X-ray spectroscopy and single-crystal X-ray structure analysis. In addition, a spontaneously single-crystal-to-single-crystal transformation from URCP3 to URCP4, which is driven by thermodynamics, was observed and explained by computational study. Moreover, it reveals that sulfate with stronger coordination ability can inhibit the hydrolysis of uranyl ion to some extent with only a rarely reported pentanuclear uranyl center found in URCP5 obtained at pH 5.67. These results indicate that the combined effect of competing ligands and pH has great significance in the formation of URCPs in terms of uranyl coordination and speciation and can be an alternative way to design and synthesize uranyl coordination polymers with new topologies.

Background: Limited data exist with regard to treatment outcomes in Asian Americans with chronic hepatitis C (CHC). We evaluated sofosbuvir (SOF)-based regimens in a national cohort of Asian Americans., Methods: Eligible Asian Americans patients with CHC who had posttreatment follow-up of 24 weeks for SOF -based therapies from December 2013 to June 2017 were enrolled from 11 sites across the United States. The primary endpoint was sustained virologic response (SVR) rates at posttreatment weeks 12 and 24. Secondary endpoints were to evaluate safety by tolerability and adverse events (AEs)., Results: Among 231 patients screened, 186 were enrolled. At baseline, 31% (57/186) patients were cirrhotic, 34% (63/186) were treatment experienced. Most of the subjects (42%, 79/186) received ledispavir/SOF therapy. The overall SVR12 was 95%, ranging from 86% in genotype (GT) 1b on SOF+ribavirin to 100% in GT 1b patients on ledipasvir/SOF at subgroup analyses. SVR12 was significantly lower in cirrhotic than in noncirrhotic patients [88% (50/57) vs. 98% (126/129), P<0.01]. Stratified by GT, SVR12 were: 96% (43/45) in GT 1a; 93% (67/72) in GT 1b; 100% (23/23) in GT 2; 90% (19/21) in GT 3; 100% (1/1) in GT 4; 83% (5/6) in GT 5; and 100% (16/16) in GT 6. Cirrhotic patients with treatment failure were primarily GT 1, (GT 1a, n=2; GT 1b, n=4) with 1 GT 5 (n=1). Patients tolerated the treatment without serious AEs. Late relapse occurred in 1 patient after achieving SVR12., Conclusions: In Asian Americans with CHC, SOF-based regimens were well tolerated without serious AEs and could achieve high SVR12 regardless of hepatitis C viral infection GT.

The fluorescence of uranyl originated from electronic transitions (S 11 -S 00 and S 10 -S 0v , v = 0-4) of the ligand-to-metal charge transfer (LMCT) process is an intrinsic property of many uranyl coordination compounds. However, light-induced regulation on fluorescence features of uranyl hybrid materials through photoactive functional groups is less investigated. In this work, the photoactive vinyl group-containing ligands, ( E)-methyl 3-(pyridin-4-yl)acrylate and ( E)-methyl 3-(pyridin-3-yl)acrylate, have been used in the construction of uranyl coordination polymers in the presence of 1,10-phenanthroline (phen). Five compounds (UO 2 ) 3 (μ 3 -O)(μ 2 -OH) 2 (L 1 ) 2 ( phen) 2 (1), (UO 2 ) 3 (μ 3 -O)(μ 2 -OH) 3 (L 1 )( phen) 2 (2), (UO 2 ) 3 (μ 3 -O)(μ 2 -OH) 3 (L 2 )( phen) 2 (3), [(UO 2 ) 2 (μ 2 -OH) 2 (L 2 ) 2 ( phen) 2 ]·2H 2 O (4), and (UO 2 )Zn(SO 4 )(phen)(H 2 O)(OH) 2 (5) were obtained under hydrothermal conditions. Compounds 1-4 are polynuclear uranyl structures with abundant π-π interactions and hydrogen bonds contributed to the 3D crystal packing of them. As model compounds, 1 and 3 are selected for exploring photoresponsive behaviors. The emission intensities of these two compounds are found to decrease gradually over the exposure time of UV irradiation. X-ray single crystal structural analysis suggests that the fluorescence attenuation can be explained by the slight rotation of pyridinyl groups around the carbon-carbon double bond during UV irradiation, which is accompanied by the change of weak interactions, i.e., π-π interactions and hydrogen bonds in strength and density. This feature of light-induced fluorescence attenuation may enable these two compounds to act as potential photoresponsive sensor materials.

Both incidence and death rate due to liver cancer have increased in the United States. Higher consumption of lycopene-rich tomato and tomato products is associated with a decreased risk of cancers. β-Carotene-15, 15'-oxygenase (BCO1), and β-carotene-9', 10'-oxygenase (BCO2) cleave lycopene to produce bioactive apo-lycopenoids. Although BCO1/BCO2 polymorphisms affect human and animal lycopene levels, whether dietary tomato consumption can inhibit high-fat diet (HFD)-promoted hepatocellular carcinoma (HCC) development and affect gut microbiota in the absence of BCO1/BCO2 is unclear. BCO1/BCO2 double knockout mice were initiated with a hepatic carcinogen (diethylnitrosamine) at 2 weeks of age. At 6 weeks of age, the mice were randomly assigned to an HFD (60% of energy as fat) with or without tomato powder (TP) feeding for 24 weeks. Results showed that TP feeding significantly decreased HCC development (67%, 83%, and 95% reduction in incidence, multiplicity, and tumor volume, respectively, P < 0.05). Protective effects of TP feeding were associated with (1) decreased hepatic inflammatory foci development and mRNA expression of proinflammatory biomarkers (IL1β, IL6, IL12α, monocyte chemoattractant protein-1, and inducible NO synthase); (2) increased mRNA expression of deacetylase sirtuin 1 and nicotinamide phosphoribosyltransferase involving NAD + production; and (3) increased hepatic circadian clock genes (circadian locomotor output cycles kaput, period 2, and cryptochrome-2, Wee1). Furthermore, TP feeding increased gut microbial richness and diversity, and significantly decreased the relative abundance of the genus Clostridium and Mucispirillum , respectively. The present study demonstrates that dietary tomato feeding independent of carotenoid cleavage enzymes prevents HFD-induced inflammation with potential modulating gut microbiota and inhibits HFD-promoted HCC development., (©2018 American Association for Cancer Research.)

Uranyl-organic frameworks (UOFs) have recently been the object of many research endeavors due to the unique coordination mode of uranyl ions and their attractive physicochemical properties. Here, a new (3,4)-connected UOF (U-IHEP-4) assembled from uranyl and porphyrin ligand tetrakis(4-carboxyphenyl)porphyrin (H4TCPP) is reported, which represents the first case of actinide porphyrinic MOFs. Adsorption experiments in DMF solution demonstrated that U-IHEP-4 selectively adsorbs positively charged dyes, which is in line with its negatively charged framework and large pore volume ratio (90 %). Remarkably, U-IHEP-4 exhibited high catalytic activity for the dehydrogenation of N-heterocycles to synthesize the corresponding aromatic heterocycles and it can be used as an efficient heterogeneous catalyst., (© 2018 Wiley-VCH Verlag GmbH & Co. As an emerging type of actinide hybrid material, uranyl-rotaxane coordination polymers (URCPs) with new coordination patterns and topological structures are still desired. In this work, we propose a new strategy to construct URCPs by promoting the simultaneous coordination of both the wheel and axle moieties in pseudorotaxane linkers with metal nodes. Starting from a series of cucurbit[6]uril (CB[6])-based pseudorotaxane ligands, C nBPCA@CB[6] [C nBPCA = 1,1-(α,ω-diyl)bis[4-(ethoxycarbonyl)pyridin-1-ium] bromides, where n = 5-8] with slightly deformed CB[6], four new URCPs (URCP1, URCP3, URCP4, and URCP5) with interwoven network structures, as well as another noninterwoven polymer(URCP2), have been successfully prepared. According to single-crystal structure analysis, we attribute the interwoven structures of the URCPs to the distortion of CB[6] in pseudorotaxane ligands with shorter or longer spacers (C5, C7, and C8). This indicates that the deformation could effectively diminish the steric hindrance around the portals, thus endowing the "inert" CB[6] host with coordination ability like the string molecule. Besides, the participation of water molecules and sulfate anions in the uranyl coordination sphere is also found to have a great influence on the final structures of the obtained URCPs. The successful preparation of interwoven URCPs in this work gives some new insights into the metal coordination of supramolecular entities and could facilitate other new applications of CB[6]-based pseudorotaxane ligands. Most importantly, the strategy proposed in this work provides some hints in the controllable design of metal-organic rotaxane frameworks with unique topologies.

Background and Aims: Portal hypertension is a serious adverse event of liver cirrhosis. Recently, we developed a simple novel technique for EUS-guided portal pressure gradient (PPG) measurement (PPGM). Our animal studies showed excellent correlation between EUS-PPGM and interventional radiology-acquired PPGM. In this video we demonstrate the results of the first human pilot study of EUS-PPGM in patients with liver disease., Methods: EUS-PPGM was performed by experienced endosonographers using a linear echoendoscope, a 25-gauge FNA needle, and a novel compact manometer. The portal vein and hepatic vein (or inferior vena cava) were targeted by use of a transgastric or transduodenal approach. Feasibility was defined as successful PPGM in each patient. Safety was based on adverse events captured in a postprocedural interview., Results: Twenty-eight patients underwent EUS-PPGM with 100% technical success and no adverse events. PPG ranged from 1.5 to 19 mm Hg and had excellent correlation with clinical parameters of portal hypertension, including the presence of varices ( P  = .0002), PH gastropathy ( P  = .007), and thrombocytopenia ( P  = .036)., Conclusion: This novel technique of EUS-PPGM using a 25-gauge needle and compact manometer is feasible and appears safe. Given the availability of EUS and the simplicity of the manometry setup, EUS-guided PPG may represent a promising breakthrough for procuring indispensable information in the management of patients with liver disease.

Background and Aims: Hepatitis C virus (HCV) infection results in hepatocytic injury with elevation of both alanine aminotransferase (ALT) and aspartate aminotransferase (AST). It remains to be determined if direct-acting antiviral treatment can terminate hepatocytic injury following virologic response. To this end, we evaluated the pattern and predicting factors of ALT and AST normalization during and after direct-acting antiviral treatment with sustained virologic response at 12 weeks (SVR12). Methods: Single-center retrospective study on 115 HCV-infected patients who achieved SVR12 was performed. Results: At treatment week 2, 100% and 45.9% showed decline in HCV RNA to <700 IU/mL and undetectable levels, respectively, and this was associated with 85.5%, 83.9% and 77.4% ALT normalization, AST normalization and ALT and AST normalization. At end of treatment, 85.6% of patients with baseline elevation of both ALT and AST had normalization of both ALT and AST. At posttreatment weeks 12 and 24, 90.8% and 94.8% had normalization of both ALT and AST. HCV clearance also resulted in further decline of both ALT and AST in those with baseline <40 IU. Univariate analysis showed baseline Child-Pugh score of <6, model for end-stage liver disease score of <10, HCV genotype 1, and HCV RNA of <500 IU/mL at treatment week 2 were associated with sustained normalization of both ALT and AST at posttreatment week 12. On multivariate analysis, baseline model for end-stage liver disease score of <10 was significantly associated with normalization of both ALT and AST at posttreatment week 12, independent of baseline Child-Pugh score <6, HCV genotype 1, and HCV RNA of <500 IU/mL at treatment week 2. Conclusions: During direct-acting antiviral therapy, 85.5% and 83.9% had normalization of both ALT and AST as early as in week 2, providing biochemical evidence of hepatocytic injury resolution. Sustained normalization of both ALT and AST was seen in 90.8% at posttreatment weeks 12, and was independently associated with baseline model for end-stage liver disease score of <10., Competing Interests: Dr. Hu is on the Speaker Bureau for Gilead and Merck. β-Carotene-15, 15'-oxygenase (BCO1) and β-carotene-9', 10'-oxygenase (BCO2) are essential enzymes in carotenoid metabolism. While BCO1/BCO2 polymorphisms have been associated with alterations to human and animal carotenoid levels, experimental studies have suggested that BCO1 and BCO2 may have specific physiological functions beyond the cleavage of carotenoids. In the present study, we investigated the effect of ablation of both BCO1/BCO2 in the development of non-alcoholic fatty liver disease (NAFLD) and its underlying molecular mechanism(s). BCO1/BCO2 double knock out (DKO) mice developed hepatic steatosis (8/8) and had significantly higher levels of hepatic and plasma triglyceride and total cholesterol compared to WT (0/8). Hepatic changes in the BCO1/BCO2 DKO mice were associated with significant: 1) increases in lipogenesis markers, and decreases in fatty acid β-oxidation markers; 2) upregulation of cholesterol metabolism markers; 3) alterations to microRNAs related to TG accumulation and cholesterol metabolism; 4) increases in an hepatic oxidative stress marker (HO-1) but decreases in anti-oxidant enzymes; and 5) decreases in farnesoid X receptor (FXR), small heterodimer partner (SHP), and sirtuin 1 (SIRT1). The present study provided novel experimental evidence that BCO1 and BCO2 could play a significant role in maintaining normal hepatic lipid and cholesterol homeostasis, potentially through the regulation of the FXR/miR-34a/SIRT1 pathway., (Copyright © 2018. As an extension of actinide-rotaxane complexes from uranium to transuranium, we report the first crystal structure of a neptunium-rotaxane complex, NRCP-1, in which an interwoven neptunium(v)-rotaxane coordination network incorporating a mechanically-interlocked [c2]daisy chain unit is promoted via the simultaneous coordination of cucurbituril (CB6) and axle molecules in [2]pseudorotaxane to Np V .

Scope: We have previously shown that apo-10'-lycopenoic acid (ALA), a derivative of lycopene through cleavage by carotene-9',10'-oxygenase, inhibits tumor progression and metastasis in both liver and lung cancer animal models. The underlying mechanism remains unknown. We hypothesized that ALA inhibits cancer cell motility and angiogenesis by up-regulating peroxisome proliferator-activated receptor γ (PPARγ) which is involved in controlling angiogenesis, tumor progression and metastasis., Methods and Results: ALA treatment, in dose-dependent manner, was effective at inhibiting migration and invasion of liver and lung cancer cells (HuH7 and A549) in both Transwell and wound-healing models, as well as suppressing actin remodeling and ruffling/lamellipodia formation in HuH7 and immortalized lung BEAS-2B cells. ALA treatment resulted in suppression of angiogenesis in both tube formation and aortic ring assays and inhibition of matrix metalloproteinase-2 expression and activation in both HuH7 and A549 cells. Additionally, ALA dose-dependently increased the mRNA expression and protein levels of PPARγ in human THLE-2 liver cells., Conclusion: ALA inhibits cancer cell motility and angiogenesis and induces PPARγ expression, which could be one of the potential mechanisms for ALA protecting against tumor progression., (Copyright © 2018. A bifunctional ligand, 2,2'-bipyridine-4,4'-dicarboxylic acid (H 2 bpdc), has been used in the investigation of constructing bimetallic uranyl organic frameworks (UOFs). Seven novel uranyl-transition metal bimetallic coordination polymers, [(UO 2 )Zn(bpdc) 2 ] n (1), [Cd(UO 2 )(bpdc) 2 (H 2 O) 2 ·2H 2 O] n (2), [Cu(UO 2 )(bpdc)(SO 4 )(H 2 O) 3 ·2H 2 O] n (3), [CuCl(UO 2 )(bpdc)(Hbpdc)(H 2 O) 2 ·H 2 O] n (4), [Cu(UO 2 )(bpdc) 2 (H 2 O)] n (5), [Co 2 (UO 2 ) 3 (bpdc) 6 ] n (6), and [Co 3 (UO 2 ) 4 (bpdc) 8 (Hbpdc)(H 2 O) 2 ] n (7), have been successfully constructed through the assembly of various transition-metal salts, uranyl ions, and H 2 bpdc ligands under hydrothermal conditions. UOFs 1, 5, 6, and 7 adopt three-dimensional (3D) frameworks with different architectures; UOFs 2 and 3 exhibit two-dimensional (2D) wavelike and stairlike layers, respectively, while UOF 4 is a one-dimensional (1D) chain assembly. These UOFs include a wide range of dimensionalities (1D-3D), interpenetrated frameworks, and cation-cation interaction species, suggesting that anion-dependent structure regulation based on the metalloligand [M(bpdc) m ] n- motifs, the coordination modes of the metal centers and bpdc 2- ligands, along with the reaction temperature, has a remarkable influence on the formation of bimetallic UOFs, which could be a representative system for the structural modulation of UOFs with various dimensionalities and structures. Furthermore, the thermal stability and luminescent properties of compounds 1, 3, and 6 are also investigated.

Background: Our recent study indicated the possible presence of detectable hepatitis C virus antigens (HCV-Ags) after denaturation of sera with resolved HCV (R-HCV) infection. The present study determined and characterized persistent HCV-Ags-specific immune complexes (ICs) in these patients., Methods: Sixty-eight sera with R-HCV and 34 with viremic HCV (V-HCV) infection were tested for free and IC-bound HCV-Ags using HCV-Ags enzyme immunoassay (EIA), the presence of HCV-Ags-specific ICs by immunoprecipitation and Western blot (IP-WB), HCV ICs containing HCV virions using IP and HCV RNA RT-PCR, and correlation of HCV ICs with clinical presentation in these patients., Results: Using HCV-Ags EIA, we found 57.4% of sera with R-HCV infection were tested positive for bound, but not free HCV-Ags. Using pooled or individual anti-HCV E1/E2, cAg, NS3, NS4b, and/or NS5a to precipitate HCV-specific-Ags, we confirmed persistent HCV-Ags ICs specific to various HCV structural and non-structural proteins not only in V-HCV infection, but also in R-HCV infection. Using IP and HCV RNA PCR, we then confirmed the presence of HCV virions within circulating ICs in V-HCV, but not in R-HCV sera. Multivariable analysis indicated significant and independent associations of persistent circulating HCV-Ags-specific ICs with both age and the presence of cirrhosis in patients with R-HCV infection., Conclusions: Various HCV-Ag-specific ICs, but not virions, persist in 57.4% of patients who had spontaneous or treatment-induced HCV clearance for 6 months to 20 years. These findings enriched our knowledge on HCV pathogenesis and support further study on its long-term clinical relevance, such as extrahepatic manifestation, transfusion medicine, and hepatocarcinogenesis.

We report the syntheses and characterization of four new heterometallic uranyl-silver compounds from isonicotinic acid derivatives with a stepwise ortho chlorination of carboxyl group, that is, isonicotinic acid (H-PCA), 3-chloroisonicotinic acid (H-3-MCPCA), and 3,5-dichloroisonicotinic acid (H-3,5-DCPCA). Compound 1, (UO 2 )Ag 4 (3,5-DCPCA) 6 (3,5-DCPy) 2 , from H-3,5-DCPCA displays a heterometallic three-dimensional (3D) framework through the connection of 3,5-DCPCA and in situ-formed 3,5-dichloropyridine (3,5-DCPy) with the aid of multiple argentophilic interactions. Compounds 2 ((UO 2 )Ag(3-MCPCA) 3 ) and 3 ((UO 2 )Ag 2 (3-MCPCA) 4 ), which differ from each other in coordination modes of uranyl center, are both heterometallic 3D reticular frameworks from 3-MCPCA based on highly coordinated silver nodes. All these heterometallic uranyl-silver compounds are different from the hydrothermal products from chlorine-free H-PCA ligand in the presence of uranyl and silver ions, U-Ag-PCA ((UO 2 Ag(OH)(PCA) 2 )) and 4 ((UO 2 )Ag 2 (OH)(H 2 O) 2 (PCA) 4 ) due to highly coordinated silver ions found in 1-3, among which carboxyl groups of isonicotinate expected to coordinate with uranyl are the biggest contributors. Detailed structural analysis reveals that the inclination of the carboxyl group of isonicotinate driven by large steric hindrance from bulky ortho chlorine atoms at its ortho positions enables it to participate in the coordination sphere of silver ion and promote the formation and structure variance of 3D heterometallic uranyl-silver frameworks.

We report five novel uranyl coordination polymers, [(CH 3 ) 2 NH 2 ]UO 2 (BTPCA) (1), [(CH 3 ) 2 NH 2 ]UO 2 (BTPCA) (2), [(CH 3 ) 2 NH 2 ] 2 [UO 2 (BTPCA)][UO 2 (BTPCA)]·(H 2 O) 5.5 (3), [(CH 3 ) 2 NH 2 ] 2 (UO 2 ) 2 (BTPCA) 2 ·(H 2 O) 3 (4), and [(CH 3 ) 2 NH 2 ]UO 2 (BTPCA) (5), by the utilization of semirigid ligand 1,1',1″-(benzene-1,3,5-triyl)tripiperidine-4-carboxylic acid (H 3 BTPCA) and uranyl nitrate through solvothermal reactions. Single-crystal X-ray diffraction analysis reveals that the five compounds share a similar structure composition and local coordination mode to the exclusion of disordered water or DMF molecules. Each UO 2 (COO) 3 - motif is connected to six neighboring units through three BTPCA 3- ligands, generating an infinite uranyl honeycomb (6, 3) net. The structures of all the five compounds consist of 2D honeycomb nets of various degrees of distortions, which are induced by the flexibility of piperidine rings. The dimethylamine cations and solvent molecules fill in the space between layers. Therefore, these five compounds are isomers in a broad sense. Notably, both compounds 3 and 4 possess 2-fold interpenetrated structures. For compound 5, the distance between the neighboring 2D honeycomb nets is 7.253 Å. This is the largest distance between the 2D honeycomb nets in uranyl-based coordination polymers, to the best of our knowledge. In addition, compounds 1, 2, and 4 are also characterized by infrared spectroscopy (IR), thermogravimetric analysis (TG), powder X-ray diffraction (PXRD), and luminescence properties.

Scope: Beta-carotene-15,15'-oxygenase (BCO1) and beta-carotene-9',10'-oxygenase (BCO2) metabolize lycopene to biologically active metabolites, which can ameliorate nonalcoholic fatty liver disease (NAFLD). We investigate the effects of tomato powder (TP containing substantial lycopene (2.3 mg/g)) on NAFLD development and gut microbiome in the absence of both BCO1 and BCO2 in mice., Method and Results: BCO1 -/- /BCO2 -/- double knockout mice were fed a high fat diet (HFD) alone (n = 9) or with TP feeding (n = 9) for 24 weeks. TP feeding significantly reduced pathological severity of steatosis and hepatic triglyceride levels in BCO1 -/- /BCO2 -/- mice (p < 0.04 vs HFD alone). This was associated with increased SIRT1 activity, nicotinamide phosphoribosyltransferase expression and AMP-activated protein kinase phosphorylation, and subsequently decreased lipogenesis, hepatic fatty acid uptake, and increasing fatty acid β-oxidation (p < 0.05). TP feeding significantly decreased mRNA expression of proinflammatory genes (tnf-α, il-1β, and il-6) in both liver and mesenteric adipose tissue, which were associated with increased plasma adiponectin and hepatic adiponectin receptor-2. Multiplexed 16S rRNA gene sequencing was performed using DNA extracted from cecum fecal samples. Background: Hepatitis B virus (HBV) infection is common with major clinical consequences. In Asian Americans, the HBsAg carrier rate ranges from 2% to 16% which approximates the rates from their countries of origin. Similarly, HBV is the most important cause of cirrhosis, hepatocellular carcinoma (HCC) and liver related deaths in HBsAg positive Asians worldwide., Aim: To generate recommendations for the management of Asian Americans infected with HBV., Methods: These guidelines are based on relevant data derived from medical reports on HBV from Asian countries as well as from studies in the HBsAg positive Asian Americans. The guidelines herein differ from other recommendations in the treatment of both HBeAg positive and negative chronic hepatitis B (CHB), in the approach to HCC surveillance, and in the management of HBV in pregnant women., Results: Asian American patients, HBeAg positive or negative, with HBV DNA levels >2000 IU/mL (>10 4 copies/mL) and ALT values above normal are candidates for anti-viral therapy. HBeAg negative patients with HBV DNA >2000 IU/mL and normal ALT levels but who have either serum albumin <3.5 g/dL or platelet count <130 000 mm 3 , basal core promoter (BCP) mutations, or who have first-degree relatives with HCC should be offered treatment. Patients with cirrhosis and detectable HBV DNA must receive life-long anti-viral therapy. Indications for treatment include pregnant women with high viraemia, coinfected patients, and those requiring immunosuppressive therapy. In HBsAg positive patients with risk factors, life-long surveillance for HCC with alpha-fetoprotein (AFP) testing and abdominal ultrasound examination at 6-month intervals is required. In CHB patients receiving HCC treatments, repeat imaging with contrast CT scan or MRI at 3-month intervals is strongly recommended. These guidelines have been assigned to a Class (reflecting benefit vs. risk) and a Level (assessing strength or certainty) of evidence., Conclusions: Application of the recommendations made based on a review of the relevant literature and the opinion of a panel of Asian American physicians with expertise in HBV treatment will inform physicians and improve patient outcomes., (© 2018 The Authors. A series of novel uranyl coordination polymers have been synthesized by hydrothermal reactions. Both complexes 1 and 2 prosess two ipbp - ligands (H 2 ipbpCl=1-(3,5-dicarboxyphenyl)-4,4'-bipyridinium chloride), one uranyl cation, and two coordination water molecules, which can further extend to 2D networks through hydrogen bonding. In complex 1, two sets of equivalent nets are entangled together, resulting in a 2D + 2D → 3D polycatenated framework. In complex 2, the neighbouring equivalent nets interpenetrate each other, forming a twofold interpenetrated network. Complexes 3 and 4 are isomers, and both of them are constructed from (UO 2 ) 2 (OH) 2 dinuclear units, which are connected with four ipbp - ligands. The 3D structures of complexes 3 and 4 are similar along the b axis. Similar to other viologen-based coordination polymers, complexes 3 and 4 exhibit photochromic and thermochromic properties, which are rarely observed in actinide coordination polymers. Unlike the monotonous coordination mode in complexes 1-4, the ipbp - ligands feature a μ 3 -bridge through two kinds of coordination modes in complex 5. Although the capability of supramolecular pseudorotaxane/rotaxane systems as ligands for coordination with actinides has been identified by the on-going emerging of uranyl-organic polyrotaxane compounds, it is, however, still unknown how supramolecular inclusion affects the coordination assembly of the simple "axle" ligand with uranyl species. Herein, a semi-rigid organic dicarboxylate compound [BzBPCEt]Br 2 (L 1 ) is selected as a small-molecule "axle" ligand and the corresponding cucurbit[7]uril (CB7)-based [2]pseudorotaxane ligand, [BzBPCEt]Br 2 @CB7 (L 1 @CB7) has been also synthesized through CB7-based inclusion in this work. A detailed comparison between uranyl complexes from the "axle" ligand L 1 and those from pseudorotaxane L 1 @CB7 has been conducted, demonstrating the significant role of CB7-based inclusion in distinguishing supramolecular pseudorotaxane ligands from small-molecule dicarboxylates in uranyl coordination assembly. Notably, the impact of supramolecular inclusion on the "axle" linker in the system with cucurbituril macrocycles involved is established for the first time. Although the prosperity of rotaxane coordination polymers with rotaxane molecules serving as main-chain linkers is known, side-chain metal-organic polypseudorotaxanes incorporating macrocyclic host molecules have not been reported to date. Herein a new type of coordination-driven cucurbit[6]uril-bearing side-chain polypseudorotaxane, with two-dimensional trimeric uranyl-oxalate as main chains, has been synthesized. This was carried out through hydrothermal reactions of uranyl components with an in situ-formed carboxylated pseudorotaxane ligand in the presence of oxalate co-ligands. Varying the substitution site of coordination groups led to two different supramolecular isomers. Further mechanistic analysis indicated that condition-dependent hydrolysis of the cyano groups of the pseudorotaxane ligand, as well as the participation of oxalate groups into the coordination sphere of uranyl moieties, contributes to the formation of this new type of side-chain polypseudorotaxane., (© 2017 Wiley-VCH Verlag GmbH & Co. The first reversible solid-state single-crystal-to-single-crystal isomerisation mediated by the change of uranyl-ligand coordination modes, that is from seven-coordinated uranium(vi) of α-UP to six-coordinated uranium(vi) of the supramolecular isomer, β-UP, has been achieved in the uranyl polyrotaxane system by a temperature-induced strategy.

We herein report the synthesis, crystal structure and magnetic properties of nine new heterotrimetallic complexes. [Ni(Me 2 valpn)] (H 2 Me 2 valpn = N,N'-bis(3-methoxysalicylidene)-2,2-dimethyl-1,3-diaminopropane) was used as the precursor to construct phenoxo-bridged [Ni 2 Ln] 3+ (Ln 3+ = Dy, Tb, Gd and Y) species that were respectively connected by two [M(CN) 6 ] 3- (M = Cr, Fe or Co) anions to form octanuclear cyclic complexes, i.e. {[Ni(Me 2 valpn)] 2 Ln(H 2 O)M(CN) 6 } 2 in which Ln = Y, Gd, Tb and Dy and M = Cr, Fe or Co (1-9). Each of the complexes contains many lattice-bound molecules of solvation. The Ni(ii) ions are penta-coordinate, while the Ln(iii) ions are nine coordinated with a muffin geometry. The fitting to the χ m T vs. T curves of complexes 6-9 gave the parameters of J NiCr = 11.82 cm -1 , J NiGd = 0.94 cm -1 and g = 2.04 for complex 6, J NiFe = 10.58 cm -1 , J NiGd = 1.24 cm -1 and g = 2.03 for complex 7, J NiCr = 9.4(1) cm -1 , zJ' = -0.050(2) cm -1 and g = 2.06(1) for complex 8 and J NiFe = 4.9(7) cm -1 , zJ' = -0.35(2) cm -1 and g = 2.24(1) for complex 9, respectively. The dynamic magnetic investigations demonstrate that complexes 1-5 display single-molecule magnet properties with an effective energy barrier (U eff ) of 38.9 K (1, M1 = Dy, M2 = Cr), 37.2 K (2, M1 = Dy, M2 = Cr), 24.4 K (3, M1 = Dy, M2 = Co), 21.9 K (4, M1 = Tb, M2 = Cr) and 29.6 K (5, M1 = Tb, M2 = Fe), respectively. Complex 1 shows the highest energy barrier among the octanuclear [Ni 4 LnM 2 ] (Ln = Dy or Tb, M = Fe, Cr, Co or W) system. Although the [Ni 4 Dy 2 Cr 2 ] complexes have U eff higher than that of [Ni 4 Dy 2 Fe 2 ], complex [Ni 4 TbCr 2 ] shows lower U eff than that of [Ni 4 TbFe 2 ]. The results indicate that besides the M-C[triple bond, length as m-dash]N-Ni magnetic coupling the lanthanide ions can significantly affect the magnetic performances of heterotrimetallic SMMs as well. Moreover, the SMMs are achieved when diamagnetic Co(iii) was substituted by paramagnetic Cr(iii) or Fe(iii) in the [Ni 4 Tb 2 M 2 ] system, suggesting that the trimetallic strategy is effective in the construction of new 3d-4f SMMs.

Background and Aims: Portal hypertension (PH) is a serious adverse event of liver cirrhosis. The hepatic venous pressure gradient or portal pressure gradient (PPG) accurately reflects the degree of PH and is the single best prognostic indicator in liver disease. This is usually obtained by interventional radiology (IR), although it is not routinely performed. Recently, we developed a simple novel technique for EUS-guided PPG measurement (PPGM). Our animal studies showed excellent correlation between EUS-PPGM and IR-PPGM. We present the first human pilot study of EUS-PPGM in patients with liver disease., Methods: EUS-PPGM was performed by experienced endosonographers using a linear echoendoscope, a 25-gauge fine-needle aspiration needle, and a novel compact manometer. The portal vein and hepatic vein (or inferior vena cava) were targeted using a transgastric-transduodenal approach. Clinical parameters of PH were evaluated in each patient. Feasibility was defined as successful PPGM in each patient. Safety was based on adverse events captured in a postprocedural interview., Results: Twenty-eight patients underwent EUS-PPGM with 100% technical success and no adverse events. PPG ranged from 1.5 to 19 mm Hg and had excellent correlation with clinical parameters of portal hypertension including the presence of varices (P = .0002), PH gastropathy (P = .007), and thrombocytopenia (P = .036). PPG was increased in patients with high clinical evidence of cirrhosis (P = .005)., Conclusion: This novel technique of EUS-PPGM using a 25-gauge needle and compact manometer is feasible and appears safe. Given the availability of EUS and the simplicity of the manometry setup, EUS-guided PPG may represent a promising breakthrough for procuring indispensable information in the management of patients with liver disease., (Copyright © 2017. A mixed-ligand system provides an alternative route to tune the structures and properties of metal-organic compounds by introducing functional organic or inorganic coligands. In this work, five new uranyl-based polyrotaxane compounds incorporating a sulfate or oxalate coligand have been hydrothermally synthesized via a mixed-ligand method. Based on C6BPCA@CB6 (C6BPCA = 1,1'-(hexane-1,6-diyl)bis(4-(carbonyl)pyridin-1-ium), CB6 = cucurbit[6]uril) ligand, UPS1 (UO 2 (L) 0.5 (SO 4 )(H 2 O)·2H 2 O, L = C6BPCA@CB6) is formed by the alteration of initial aqueous solution pH to a higher acidity. The resulting 2D uranyl polyrotaxane sheet structure of UPS1 is based on uranyl-sulfate ribbons connected by the C6BPCA@CB6 pseudorotaxane linkers. By using oxalate ligand instead of sulfate, four oxalate-containing uranyl polyrotaxane compounds, UPO1-UPO4, have been acquired by tuning reaction pH and ligand concentration: UPO1 (UO 2 (L) 0.5 (C 2 O 4 ) 0.5 (NO 3 )·3H 2 O) in one-dimensional chain was obtained at a low pH value range (1.47-1.89) and UPO2 (UO 2 (L)(C 2 O 4 )(H 2 O)·7H 2 O)obtained at a higher pH value range (4.31-7.21). By lowering the amount of oxalate, another two uranyl polyrotaxane network UPO3 ((UO 2 ) 2 (L) 0.5 (C 2 O 4 ) 2 (H 2 O)) and UPO4 ((UO 2 ) 2 O(OH)(L) 0.5 (C 2 O 4 ) 0.5 (H 2 O)) could be acquired at a low pH value of 1.98 and a higher pH value over 6, respectively. The UPO1-UPO4 compounds, which display structural diversity via pH-dependent competitive effect of oxalate, represent the first series of mixed-ligand uranyl polyrotaxanes with organic ligand as the coligand. Moreover, the self-assembly process and its internal mechanism concerning pH-dependent competitive effect and other related factors such as concentration of the reagents and coordination behaviors of the coligands were discussed in detail.

Two highly symmetrical (3,4)-connected uranyl-organic frameworks (UOFs) were synthesized by a judicious combination of D 3h -symmetrical triangular [UO 2 (COO) 3 ] - and T d symmetrical tetrahedral tetrakis(4-carboxyphenyl)methane (H 4 MTB). These two as-synthesized UOFs possess similar structural units and coordination modes but totally different topological structures, namely ctn net and bor net. Solvent-induced interpenetration and a morphology change are observed. The two compounds exhibit crystal transformation via a dissolution-crystallization process. Adsorption experiments in CH 3 OH solution indicate that both of them can selectively remove positively charged dyes over negatively charged and neutral dyes. Despite the consistent association between a higher intake of the provitamin A carotenoid β-cryptoxanthin (BCX) and a lower risk of lung cancer among smokers, potential mechanisms supporting BCX as a chemopreventive agent are needed. We first examined the effects of BCX on 4-[methyl nitrosamino]-1-[3-pyridyl]-1-butanone (NNK)-induced lung tumorigenesis in A/J mice. BCX supplementation was given daily to the mice starting 2 weeks prior to the injection of NNK and continued 16 weeks after NNK injection. BCX supplementation resulted in a dose-dependent increase of BCX concentration in both serum and lungs of the mice without a significant alteration of vitamin A (retinol and retinyl palmitate) concentration. BCX significantly reduced the multiplicity of the NNK-induced lung tumor by 52% to 63% compared with the NNK-treated mice without BCX supplementation. The protective effect of BCX in the lungs was associated with reductions of both mRNA and protein of the homopentameric neuronal nicotinic acetylcholine receptor α7 (α7-nAChR), which has been implicated in lung tumorigenesis. We then conducted an in vitro cell culture study and found that BCX treatment suppressed α7-nAChR expression and inhibited the migration and invasion of α7-nAChR-positive lung cancer cells but not in cells lacking α7-nAChR. The activities of BCX were significantly attenuated by activators of α7-nAChR/PI3K signaling or by overexpression of constitutively active PI3K. Collectively, the results suggest that BCX inhibits lung tumorigenesis and cancer cell motility through the downregulation of α7-nAChR/PI3K signaling, independent of its provitamin A activity. Therefore, BCX can be used as a chemopreventive agent or a chemotherapeutic compound against lung cancer. The first 3D actinide polyrotaxane framework (named IHEP-URCP-2) has been obtained based on windmill-like six-connected high-nuclear oligomeric uranyl nodes under hydrothermal conditions. Notably, the in situ formed pseudorotaxane ligand simultaneously plays dual roles of both a bulky pseudorotaxane linker and a supramolecular guest.

Early epidemiologic studies have reported that tobacco smoking, which is causally associated with liver cancer, is an independent risk factor for non-alcoholic fatty liver diseases (NAFLD). Lycopene from tomatoes has been shown to be a potential preventive agent against NAFLD and hepatocellular carcinoma (HCC). In the present study, we investigated whether the tobacco carcinogen 4-(N-methyl-N-nitrosamino)-1-(3-pyridyl)-1-butanone (NNK) induces lesions in both lungs and livers of ferrets with or without lycopene intervention. Male ferrets (6 groups, n = 8-10) were treated either with NNK (50 mg/kg BW, i.p., once a month for four consecutive months) or saline with or without dietary lycopene supplementation (2.2 and 6.6 mg/kg BW/day, respectively) for 26 weeks. Results demonstrate that NNK exposure results in higher incidences of lung tumors, HCC and steatohepatitis (which is characterized by severe inflammatory cell infiltration with concurrent fat accumulation in liver, hepatocellular ballooning degeneration and increased NF-κB expression), as well as elevations in bilirubin and AST levels in ferrets. Lycopene supplementation at two doses prevented NNK-induced expressions of α7 nicotinic acetylcholine receptor in the lung and NF-κB and CYP2E1 in the liver and attenuated the NNK-induced mortality and pathological lesions in both the lungs and livers of ferrets. The present study provided strong experimental evidence that the tobacco carcinogen NNK can induce both HCC and steatohepatitis in the ferrets and can be a useful model for studying tobacco carcinogen-associated NAFLD and liver cancer.
