58. Candidate gene (PHA-E) and phytohemagglutinin content in snap bean (Phaseolus vulgarisL.): an association study

Abstract

AbstractSnap bean (Phaseolus vulgarisL.) is low in fat and high in nutrients; however, it also contains toxigenic factors such as phytohemagglutinin (PHA), which is poisonous to humans. Here, we aimed to determine the distribution of PHA in natural populations of snap bean by analyzing a candidate gene (PHA-E) related to PHA content. Functional genome-wide single nucleotide polymorphisms (SNPs) were then scanned to identify SNPs related to PHA content. A total of 150 snap bean varieties were examined. PHA content and PHA-Eexpression levels were determined 10, 14, 18, 22, 26 and 30 days after anthesis. Then, association analyses were carried out to identify causal SNPs in PHA-E viagenotyping and association tests. PHA-Eexpression was positively-correlated with PHA content. Among 59 detected SNPs, the ratio of synonymous to nonsynonymous changes was 1:8. P190 was identified as a novel SNP associated with PHA content. The allele carrying the GG genotype at P190 was detected in 130 varieties, of which 86 had an above-average PHA content, while the allele carrying the AA genotype at P190 was detected in 20 varieties, of which 17 (85%) had a low PHA content. Analyses of the PHA content and gene expression in fresh pods of 150 snap bean cultivars revealed population-level distribution and genetic variation, providing a unique measure for screening of low-level or nontoxic cultivars and aiding future breeding of new cultivars.Supplemental data for this article is available online at https://doi.org/10.1080/13102818.2021.1985613.

Abstract

Quite often ESR dating of tooth enamel provides underestimated ages. The factors, which may account for the age suppression are studied. For dating of the samples of Early Pleistocene, thermal fading of the g =2.0018 signal during the burial time may also lead toage underestimation. Isothermal annealing of the g =2.0018 line was studied in this work and a mixed first- and second-order kinetics was observed. Its implication to the practical dating purpose is discussed. It is suggested that if no uranium leakage is detected, ESR EU ages of Early Pleistocene enamel samples should be considered only as the lower limits of the true ages. Combined ESR and magnetic dating of the Longgupo profile, Wushan, China, assigns the Olduvai subchron to the fossil hominid-bearing layers of 7–8 of this profile.

Abstract

BACKGROUND We present the genome sequence of the tammar wallaby, Macropus eugenii, which is a member of the kangaroo family and the first representative of the iconic hopping mammals that symbolize Australia to be sequenced. The tammar has many unusual biological characteristics, including the longest period of embryonic diapause of any mammal, extremely synchronized seasonal breeding and prolonged and sophisticated lactation within a well-defined pouch. Like other marsupials, it gives birth to highly altricial young, and has a small number of very large chromosomes, making it a valuable model for genomics, reproduction and development. RESULTS The genome has been sequenced to 2 × coverage using Sanger sequencing, enhanced with additional next generation sequencing and the integration of extensive physical and linkage maps to build the genome assembly. We also sequenced the tammar transcriptome across many tissues and developmental time points. Our analyses of these data shed light on mammalian reproduction, development and genome evolution: there is innovation in reproductive and lactational genes, rapid evolution of germ cell genes, and incomplete, locus-specific X inactivation. We also observe novel retrotransposons and a highly rearranged major histocompatibility complex, with many class I genes located outside the complex. Novel microRNAs in the tammar HOX clusters uncover new potential mammalian HOX regulatory elements. CONCLUSIONS Analyses of these resources enhance our understanding of marsupial gene evolution, identify marsupial-specific conserved non-coding elements and critical genes across a range of biological systems, including reproduction, development and immunity, and provide new insight into marsupial and mammalian biology and genome evolution.

73. Spermatogenesis genes: sex chromosome linkage and evolution

Abstract

The human X-chromosome contains a disproportionately high number of sex- and reproduction-related genes, a large number of which function in spermatogenesis. Comparative gene mapping between marsupials and eutherian mammals showed that genes on the long arm of the human X-chromosome map to the X in marsupials and are believed to represent the ancestral mammalian X. However, genes on the human X-chromosome short arm map to the autosomes in marsupials and represent more recent additions to the X exclusively in the eutherian lineage. It was assumed that genes acquired their functions in sex and reproduction after the evolution of the X-chromosome due to its hemizygosity in males allowing direct selection of male advantage alleles. To test this hypothesis further, three human X-linked spermatogenesis genes, AKAP4, KAL1 and TGIFLX were investigated using the marsupial tammar wallaby as a model to examine how these X-lined genes evolved their functions. Two of these genes map to the ancestral X-chromosome, while one maps to the region contained on the autosomes in marsupials. In humans and mice, X-linked AKAP4 is the most abundant protein on the spermatozoa principal piece, essential for the formation of flagellum and signal transduction, regulating sperm motility. Tammar AKAP4 was also located on the X-chromosome and was expressed only in adult testis. AKAP4 mRNA was confined to the round and elongated spermatids. Tammar AKAP4 protein was predominantly detected in the sperm tail, as in the mouse. The similar expression pattern suggests that AKAP4 has a conserved function in mammalian spermatogenesis, in the formation of the fibrous sheath and in signal transduction regulating motility. To investigate this conserved function further, one sperm pro-survival factor of humans, Prolactin, was tested in the tammar. Abstract

Marsupials are especially valuable for comparative genomic studies of mammals. Two distantly related model marsupials have been sequenced: the South American opossum (Monodelphis domestica) and the tammar wallaby (Macropus eugenii), which last shared a common ancestor about 70 Mya. The six-fold opossum genome sequence has been assembled and assigned to chromosomes with the help of a cytogenetic map. A good cytogenetic map will be even more essential for assembly and anchoring of the two-fold wallaby genome. As a start to generating a physical map of gene locations on wallaby chromosomes, we focused on two chromosomes sharing homology with the human X, wallaby chromosomes X and 5. We devised an efficient strategy for mapping large conserved synteny blocks in non-model mammals, and applied this to generate dense maps of the X and 'neo-X' regions and to determine the arrangement of large conserved synteny blocks on chromosome 5. Comparisons between the wallaby and opossum chromosome maps revealed many rearrangements, highlighting the need for comparative gene mapping between South American and Australian marsupials. Frequent rearrangement of the X, along with the absence of a marsupial XIST gene, suggests that inactivation of the marsupial X chromosome does not depend on a whole-chromosome repression by a control locus.

87. The synergy of morphokinetic parameters and sHLA-G in cleavage embryo enhancing implantation rates.

Abstract

Objective: This study aimed to assess the relationship between implantation and soluble HLA-G (sHLA-G) expression in cleavage embryo culture medium (ECM) in conjunction with early developmental kinetics determined by time-lapse imaging (TLI). Methods: A retrospective, single-center study was conducted involving 238 embryos from 165 patients who underwent Frozen-thawed embryo transfer (FET) using autologous oocytes, with either single or double embryo transfer. TLI morphokinetic parameters (t2, t3, t4, t5, t6, t7, t8, cc2, s2, cc3, s3) of embryos were analyzed, and sHLA-G levels in D3 ECM were measured using an enzyme-linked immunosorbent assay (ELISA). A hierarchical classification model was developed to categorize embryos into five groups (A, B, C, D, E). The correlation between sHLA-G levels, TLI classification of embryos, and embryo implantation was investigated to establish a non-invasive method for evaluating implantation potential. Multivariate logistic regression analysis was performed to identify potential influencing factors, and receiver operating characteristic (ROC) curves were used to evaluate the predictive value for implantation. Results: Multivariate unconditional logistic regression analysis indicated that TLI parameters t5 and s3 and sHLA-G level in ECM were independent risk factors affecting embryo implantation. The implantation rate decreased from TLI classification A to E. The proposed classification model effectively assessed the implantation potential of embryos. The implantation rate was higher in the sHLA-G positive group compared to the sHLA-G negative group ( p < 0.001). The expression of sHLA-G in D3 ECM, combined with the TLI classification model, accurately evaluated the implantation potential of embryos with an AUC of 0.876. Abstract

In this study, the pH-responsive API-CMCS-SA (ACS) polymeric nanoparticles (NPs) based on 1-(3-amino-propyl) imidazole (API), stearic acid (SA), and carboxymethyl chitosan (CMCS) were fabricated for the effective transport of curcumin (CUR) in liver cancer. CUR-ACS-NPs with various degrees of substitution (DS) were employed to prepare through ultrasonic dispersion method. The effect of different DS on NPs formation was discussed. The obtained CUR-ACS-NPs (DS SA =12.4%) had high encapsulation rate (more than 85%) and uniform particle size (186.2 ± 1.42 nm). The CUR-ACS-NPs showed better stability than the other groups. Drug release from the CUR-ACS-NPs was pH-dependent, and more than 90% or 65% of CUR was released in 48 h in weakly acid medium (pH 5.0 or 6.0, respectively). Additionally, the CUR-ACS-NPs increased the intracellular accumulation of CUR and demonstrated high anticancer effect on HepG2 cells compared with the other groups. CUR-ACS-NPs prolonged the retention time of the drug, and the area under the curve (AUC) increased significantly in vivo . The in vivo antitumor study further revealed that the CUR-ACS-NPs exhibited the capability of inhibiting tumor growth and lower systemic toxicity. Meanwhile, CUR, CUR-CS-NPs, and CUR-ACS-NPs could be detected in the evaluated organs, including tumor, liver, spleen, lung, heart, and kidney in distribution studies. Among them, CUR-ACS-NPs reached the maximum concentration at the tumor site, indicating the tumor-targeting properties. In short, the results suggested that CUR-ACS-NPs could act a prospective drug transport system for effective delivery of CUR in cancer treatment.

Abstract

Protein inhibitor of activated signal transducer and activator of transcription 2 (PIAS2) is a member of the PIAS protein family. This protein family modulates the activity of several transcription factors and acts as an E3 ubiquitin ligase in the sumoylation pathway. To improve understanding of the physiological roles of PIAS2, the current study used a yeast two‑hybrid system to screen mouse stem cell cDNA libraries for proteins that interact with PIAS2. The screening identified an interaction between PIAS2 and ubiquitously expressed transcript (UXT). UXT, also termed androgen receptor trapped clone‑27, is an α‑class prefoldin‑type chaperone that acts as a coregulator for various transcription factors, including nuclear factor‑κB and androgen receptor (AR). A direct interaction between PIAS2 and UXT was confirmed by direct yeast two‑hybrid analysis. In vitro evidence of the association of UXT with PIAS2 was obtained by co‑immunoprecipitation. Colocalization between PIAS2 and UXT was identified in the nucleus and cytoplasm of HEK 293T and human cervical carcinoma HeLa cells. The results of the current study suggested that UXT is a binding protein of PIAS2, and interaction between PIAS2 and UXT may be important for the transcriptional activation of AR.

The underlying molecular mechanisms that facilitate these actions are still being elucidated. In this study we determined the mechanism by which maspin mediates increased MCF10A cell adhesion. Utilizing competition peptides and mutation analyses, we discovered two unique regions (amino acid residues 190-202 and 260-275) involved in facilitating the increased adhesion function of maspin. In addition, we demonstrate that the urokinase-type plasminogen activator (uPA)/uPA receptor (uPAR) complex is required for the localization and adhesion function of maspin. Finally, we showed that maspin, uPAR, and β1 integrin co-immunoprecipitate, suggesting a novel maspin-uPA-uPAR-β1 integrin mega-complex that regulates mammary epithelial cell adhesion.

Abstract

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders; it is characterized by polycystic ovaries, hyperandrogenism and chronic anovulation. To obtain a global view of those genes that might be involved in the development of this complex clinical disorder, we used recently developed cDNA microarray technology to compare differential gene expressions between normal human ovary and ovaries from PCOS patients. A total of 9216 clones randomly selected from a commercial human ovary cDNA library were screened. Among them, 290 clones showed differential expressions, including 119 known genes and 100 known or unknown expressed sequence tags (ESTs). Among 119 known genes, 88 were upregulated and 31 downregulated in the PCOS ovary, as compared with normal human ovary. These differentially expressed genes are involved in various biologic functions, such as cell division/apoptosis, regulation of gene expression and metabolism, reflecting the complexity of clinical manifestations of PCOS. The molecular characteristics established from our study will further our understanding of the pathogenesis of PCOS and help us to identify new targets for further studies and for the development of new therapeutic interventions.
