Nicorandil, which is structurally a nitrate and also a nicotinamide, has a vasodilator action by stimulating cyclase and ATP-sensitive K+ channel. The aim of present study was to examine the effects of chronic oral administration of a high dose of nicorandil on in vitro vascular reactivity. Nicorandil (30 mg/kg), at a dose 6-10-times higher than to decrease blood pressure in rat, was orally administered 2-times daily for a 2-4 weeks to the rats. At the end of the administration period, thoracic aorta was isolated for in vitro study. Treatment with nicorandil for 4 weeks markedly reduced the relaxant effect of nicorandil itself and other vasodilators including sodium nitroprusside, nitric oxide, endothelium-derived relaxing factor released by carbachol, 8-Br-cyclic guanosine 3',5'-monophosphate (cGMP), a K+ channel opener, levcromakalim, and forskolin. Increase in cGMP content induced by nicorandil and sodium nitroprusside was less in the aorta from nicorandil-treated rat than in the vehicle-control rat. Chronic administration of nicorandil altered neither the contractile responses to norepinephrine nor the vasodilator effect of verapamil. On the other hand, a 4-week treatment with a dose of nicorandil (2 mg/kg) sufficient to decrease blood pressure in rat showed no change in aortic response. These results suggest that in vivo chronic treatment with a high dose of nicorandil inactivates not only the guanylate cyclase activity but also the mechanism mediated by cGMP; it also attenuates the sensitivity of K+ channels to levcromakalim. Prolonged activation of the specific site may desensitize its site of action.

It has been shown that oral administration of 0.038-0.15 mg/kg levcromakalim elicits a dose-related antihypertensive response in spontaneously hypertensive rats (Clapham et al., Arzneim. Forsch. 41 (1991) 385). In the present study, we examined the effects of long term administration of a high dose of levcromakalim on in vitro vascular contractility. Levcromakalim (2.25 mg/kg/day) was administered to the rats for 2 weeks and the thoracic aorta was then isolated. The levcromakalim treatment markedly reduced the relaxant effect of levcromakalim itself on norepinephrine-induced contraction. Relaxant effects of sodium nitroprusside and 8-bromo-cGMP were also attenuated by the levcromakalim treatment, although the relaxant effects of verapamil and forskolin were unchanged. The levcromakalim treatment decreased the threshold concentration for KCl and norepinephrine to induce contraction. The chronic levcromakalim treatment did not affect the cGMP production due to 3-isobutyl-1-methylxanthine and/or sodium nitroprusside. The aorta isolated from spontaneous hypertensive rats did not exhibit spontaneous activity in normal solution. After treatment with levcromakalim, however, the aorta showed spontaneous rhythmic contractions. Verapamil (10 microM) completely suppressed the spontaneous activity and decreased the basal tension below the original level. Similar to the effects of chronic treatment with levcromakalim, high-K+ solution (15.4 mM) augmented the contractile response to norepinephrine in the aorta of normotensive rats and induced rhythmic contractions in the aorta of spontaneously hypertensive rats. These results suggest that chronic treatment with a high dose of levcromakalim attenuates not only the effects of levcromakalim itself but also the cGMP-mediated relaxation, possibly by desensitizing the K+ channel.

From proteolytic digest of swine hemoglobin, we isolated four peptide, E-1 (Phe-Gln-Lys-Val-Val-Ala), E-2 (Phe-Gln-Lys-Val-Val-Ala-Gly), peptide 30-3 (Phe-Gln-Lys-Val-Val-Ala-Lys) and H-1 (Gly-Lys-Lys-Val-Leu-Gln). These peptides inhibited angiotensin I-converting enzyme activity with an IC50 of 5.8, 7.4, 2.1 and 1.9 microM, respectively. Oral administration of 50 mg/kg E-1 and 50 mg/kg H-1 decreased blood pressure in spontaneously hypertensive rats. In normotensive rats, oral administration of 500 mg/kg E-1 and 500 mg/kg H-1 inhibited the pressor effect of i.v. administrated 300 ng/kg angiotensin I, possibly by inhibiting its conversion to angiotensin II. These results suggest that these peptides are orally effective inhibitors of angiotensin I-converting enzyme that have a hypotensive effect.

In this study, we examined the effects of in vitro and in vivo treatment with isosorbide dinitrate (ISDN) on the in vitro response of isolated rat aorta. The in vitro treatment of isolated aorta with ISDN (100 μM) for 2 hr had no effect on the ISDN-induced relaxation of norepinephrine-induced contraction. In the aorta isolated from the rats treated with a high dose (90 mg/kg) of ISDN for 7–14 days, in contrast, the relaxant effect of ISDN was significantly reduced. However, the relaxant effect of sodium nitroprusside was only slightly attenuated by the treatment with a high dose of ISDN for 14 days; and the relaxant effects of 8-bromo-cGMP, levcromakalim and verapamil were unchanged. These results suggest that tolerance to ISDN was obtained only after the in vivo chronic treatment with a high dose of ISDN. ISDN may desensitize the nitric oxide-generating step rather than inactivate guanylate cyclase or the downstream pathways.

This study shows the effects of a selective endothelin ET(B) receptor agonist, IRL 1720 {Ac-[Ala11,15]endothelin-1-(8-21)}, on cardiovascular responses in anesthetized spontaneously hypertensive rats and Wistar-Kyoto rats. Single intravenous bolus injection of IRL 1720 caused a dose-related short-lasting fall in blood pressure, left ventricular pressure and myocardial contractility. However, repeated intravenous bolus injection of 10(-5) mol/kg IRL 1720 produced a biphasic response consisting of an initial short-lasting decrease followed by a sustained increase in these parameters. The initial decrease was reduced, whereas the following increase was enhanced with the repeated injections of IRL 1720. The cardiovascular pressor response was not inhibited by the endothelin ET(A) receptor antagonist, FR139317 ((R)2-[(R)-2-[(S)-2-[[1-(hexahydro-1H-azepinyl)]carbonyl] amino-4-+methylpentanoyl] amino-3-[3-(1-methyl-1H-indolyl)]propionyl]amino -3- (2-pyridyl)propionic acid). The effects of IRL 1720 were qualitatively similar but more potent in spontaneously hypertensive rats than in Wistar-Kyoto rats. These results suggest the existence of two types of endothelin ET(B) receptor for IRL 1720: a tachyphylactic endothelin ET(B) receptor that mediates cardiovascular depressor responses and a less tachyphylactic endothelin ET(B) receptor that mediates pressor responses in the rat.

In the intestinal smooth muscle of guinea pig taenia caeci, acetylcholine and carbachol induced a transient contraction followed by a sustained contraction. The magnitudes of the transient and sustained contractions were similar when muscle was stimulated with acetylcholine (0.1 microM-1 mM) or a lower concentration (0.1 microM) of carbachol. However, higher concentrations of carbachol (1 - 100 microM) induced significantly smaller sustained contraction than the transient contraction. In the 45 mM KCI-stimulated strips, addition of 100 microM carbachol induced a transient increase followed by a sustained decrease in the contractile tension. In contrast, acetylcholine (0.1 microM-1 mM) showed only weak inhibitory effect on the high K(+)-induced contraction either in the absence or presence of a cholinesterase inhibitor, 0.5 microM diisopropylfluorophosphate. The same concentration of diisopropylfluorophosphate shifted the concentration-response curve for acetylcholine to lower concentrations. In the muscles pretreated with 3 microM phorbol 12-myristate 13-acetate for 24 hr to desensitize protein kinase C, sustained contractions induced by higher concentrations of carbachol (1-100 microM) were significantly greater than those in the strips without the treatment with phorbol ester. However, the transient contraction and the contraction induced by a lower concentration (0.1 microM) of carbachol were not changed by the treatment with phorbol ester. Pretreatment with phorbol ester attenuated the inhibitory effect of carbachol on the high K(+)-induced contraction. These results suggest that the inhibitory effects of carbachol is composed of two phases: protein kinase C-independent transient inhibition and protein kinase C-dependent sustained inhibition.

Activation of voltage-dependent Ca2+ channels by high K+ (40 mM) increased the cytosolic Ca2+ level ([Ca2+]i) (estimated by fura-PE3 fluorescence ratio) and force in myometrium isolated from pregnant (21 days after gestation) and non-pregnant (estrus) rats. 12-Deoxyphorbol 13-isobutyrate (DPB, 1 mM) decreased the high (K+)-stimulated [Ca2+]i and force in a concentration-dependent manner. The inhibitory effect was stronger in the pregnant myometrium than in the non-pregnant myometrium. In the pregnant myometrium, the increase in Ca2+ permeability by ionomycin (1 microM) greatly increased [Ca2+]i and force, which were only partially inhibited by verapamil (10 microM). DPB (1 microM) inhibited the verapamil-insensitive component of the increases in [Ca2+]i and muscle tension. Oxytocin (100 nM) and thapsigargin (1 microM) also induced a verapamil-insensitive increase in [Ca2+]i and force, and DPB (1 microM) inhibited these increments. Ca2+ sensitivity of contractile elements, estimated from the relationships between Ca2+ and muscle force in intact and alpha-toxin permeabilized muscle, was not significantly changed by DPB (1 microM). In summary, DPB inhibits the increase in [Ca2+]i more strongly in myometrium isolated from pregnant rats than that from non-pregnant rats without any change in the [Ca2+]i/tension relationship. Since DPB decreased [Ca2+]i-rise induced by three different mechanisms, DPB may activate Ca2+ extrusion, rather than to inhibit a specific influx pathway, to decrease [Ca2+]i.

Background We wished to investigate the possible mechanism of the protective effect of estrogen replacement on coronary atherosclerosis observed in postmenopausal women. Methods and Results Cytosolic Ca 2+ concentration ([Ca 2+ ] i ) and contraction were measured simultaneously in fura 2–loaded porcine coronary arterial strips stimulated by the thromboxane A 2 analogue U46619 and high-K + depolarization in the presence and absence of 17β-estradiol. Pretreatment with 17β-estradiol (30 nmol/L to 30 μmol/L) inhibited the sustained elevation of [Ca 2+ ] i and the sustained contraction induced by 300 nmol/L U46619. Higher concentrations of 17β-estradiol (1 to 100 μmol/L) also inhibited the U46619-induced transient increase in [Ca 2+ ] i and contraction in the absence of extracellular Ca 2+ . In the strips precontracted by 90 mmol/L K + , 17β-estradiol (30 μmol/L) inhibited the increases in [Ca 2+ ] i and contraction to resting levels. In contrast, 30 μmol/L 17β-estradiol only partially inhibited the U46619-induced sustained contraction, despite complete inhibition of the sustained increase in [Ca 2+ ] i . Verapamil (10 μmol/L) also strongly inhibited the sustained increase in [Ca 2+ ] i induced by 300 nmol/L U46619, with a partial inhibition of the U46619-induced sustained contraction. A subsequent addition of 30 μmol/L 17β-estradiol did not show an additional inhibitory effect on either the [Ca 2+ ] i or the tension after the addition of verapamil. 17β-Estradiol (10 μmol/L) also inhibited the increase in [Ca 2+ ] i and the contraction induced by cumulative addition of Ca 2+ in the strips pretreated with 90 mmol/L K + . However, 17β-estradiol did not change the slope of the [Ca 2+ ] i -tension curves. 17β-Estradiol (10 μmol/L) had no effect on the levels of cAMP and cGMP in the coronary strips. Conclusions 17β-Estradiol inhibits the contraction of coronary vascular smooth muscle mainly by inhibiting Ca 2+ influx without changing Ca 2+ sensitivity of contractile elements. The Ca 2+ channel blocker–like action of 17β-estradiol may explain at least a part of the antiatherosclerotic effect of estrogen.

Effects of endothelins on cytosolic Ca 2+ level ([Ca 2+ ]i) and contraction were examined in the swine pulmonary artery and vein. In the artery, endothelin-1 and endothelin-3, but not sarafotoxin S6c and IRL 1620 (300 nM each), induced transient increase followed by sustained increase in [Ca 2+ ]i and sustained contraction. These effects were inhibited by the ET A receptor antagonist, BQ-123. In the vein, endothelin-1 and endothelin-3 (300 nM each) induced sustained increase in [Ca 2+ ]i and sustained contraction whereas sarafotoxin S6c and IRL 1620 (300 nM each) transiently increased both [Ca 2+ ]i and contractile tension. The ET B receptor in the vein was desensitized by pretreatment with sarafotoxin S6c, abolishing the effects of sarafotoxin S6c and IRL 1620 without changing the effects of endothelin-1 and endothelin-3. In contrast, an ET B antagonist, RES-701-1, antagonized the effects of IRL 1620 without changing the effects of other stimulants. In both artery and vein, the maximum contraction induced by these stimulants was greater than that induced by KCl at a given [Ca 2+ ]i. In the absence of external Ca 2+ , endothelin-1 and endothelin-3 induced transient increase in [Ca 2+ ]i and slow susained contraction in both artery and vein. In the vein, sarafotoxin S6c induced small sustained contraction without changing [Ca 2+ ]i. In the permeabilized artery and vein, endothelin-1 augmented the contraction induced by Ca 2+ . These results suggest that the ET A receptors in the artery and vein are coupled to Ca 2+ release (which does not seem to trigger contraction), Ca 2+ influx and Ca 2+ sensitization. In contrast, the ET B receptor in the vein is coupled to Ca 2+ influx and Ca 2+ sensitization but not to Ca 2+ release.

Vascular smooth muscle contraction is regulated by various factors including membrane potential, receptor-linked signal transduction pathways, cytoplasmic Ca concentration, and Ca sensitivity of contractile elements. Vasodilator drugs are acting on either of these mechanisms to inhibit contraction. In 1922, 338 vasodilator drugs were sold in Japan. Among them, there were 67 Ca antagonists and one K channel opener. Mechanisms of action of other 270 drugs are not clarified yet (although 84 drugs increase cyclic AMP and 36 drugs increase cyclic GMP, mechanisms of action of these cyclic nucleotides are still ambiguous). To elucidate the mechanisms of vasodilator drugs will help not only to clarify the regulation mechanisms of smooth muscle contraction but also to develop new vasodilator drugs with novel sites of action including Ca sensitivity.

The mechanism of the inhibitory effect of Li + on contraction was examined in guinea pig ileal longitudinal smooth muscle. Li + -substitution (68.4 mM) reversed contractions induced by high K + (45.4 mM), carbachol (1 μM) and histamine (1 μM) without changing the cytosolic Ca 2+ level. Li + also had no effect on the increase in 45 Ca 2+ uptake stimulated by high K + . High K + transiently increased myosin light chain (MLC) phosphorylation, reaching a peak at 6-9 sec. Li + -substitution inhibited the high K + -induced MLC phosphorylation. In permeabilized ileal strips, contraction induced by 1 μM Ca 2+ was inhibited by 10 mM Li + . The inhibitory effect was antagonized by increasing the concentration of Ca 2+ or calmodulin. In the permeabilized muscle in which MLC was previously thiophosphorylated with 1 mM ATPγS and 3 μM Ca 2+ , ATP induced contraction in Ca 2+ free buffer. Li + added during this contraction did not show an inhibitory effect. In contrast, when 30 mM Li + was added during the thiophosphorylation, the contraction induced by the subsequent addition of ATP was inhibited. Li + (30 mM) changed neither the rate of relaxation induced by removing external Ca 2+ in permeabilized muscle nor the rate of dephosphorylation of MLC induced by crude phosphatase extracted from the ileum. Li + (15 mM), on the other hand, inhibited the rate of phosphorylation of MLC caused by crude MLC kinase extracted from the ileum. Li + did not inhibit the calmodulin activity as measured with the (Ca 2+ +Mg 2+ )-ATPase activity of the erythrocyte membrane. These results suggest that the inhibitory effect of Li + on contractions is attributable to the inhibition of MLC kinase in guinea pig ileum

RES-701-1 (cyclic (Gly1-Asp9)(Gly-Asn-Trp-His-Gly-Thr-Ala-Pro-Asp-Trp-Phe- Phe-Asn-Tyr-Tyr-Trp)), a peptide isolated from Streptomyces sp., has been reported to inhibit the endothelin ETB receptor. We examined the effects of this peptide on the blood vessels and the small intestine. In isolated rat aorta without endothelium, 10 microM RES-701-1 did not affect the resting tone, nor did it attenuate the contractions induced by endothelin-1, endothelin-3 or norepinephrine. In the aorta with endothelium, 3 microM RES-701-1 shifted the concentration-response curves for the contractile effects of endothelin-1 and endothelin-3 to the left. Removal of endothelium showed a similar effect to 3 microM RES-701-1. In the norepinephrine-stimulated aorta, endothelium-dependent relaxation induced by endothelin-3 was antagonized by 0.3-10 microM RES-701-1 in a concentration-dependent manner. In the guinea pig ileum stimulated by carbachol, endothelin-3 induced a transient relaxation followed by sustained relaxation. RES-101-1 (3 microM) selectively inhibited the transient relaxation. Since it has been shown that the contractile effects of endothelins in the aorta are mediated by the endothelin ETA receptor whereas the endothelium-dependent relaxation and the ileal relaxation are mediated by the endothelin ETB receptor, it is suggested that RES-701-1 is a selective antagonist against the endothelin ETB receptor.

In rabbit saphenous vein, endothelin (ET)-1 and ET-3 induced sustained contractions whereas the selective agonists of the ETB receptor, sarafotoxin S6c (STXc) and IRL 1620, induced transient contractions. In the presence of an ETA antagonist, BQ-123, contractions induced by ET-1, STXc and IRL 1620 did not change whereas ET-3 induced only transient contraction. The ETB antagonists, RES-701-1 and IRL 1038, only weakly antagonized the effects of these stimulants. In the muscle pretreated with STXc, neither STXc nor IRL 1620 was effective whereas ET-3 induced sustained contraction at higher concentrations than ET-1. In the muscle pretreated with STXc, BQ-123 weakly antagonized the effect of ET-1 and abolished the effect of ET-3. These results suggest that there are two types of ET receptors; less tachyphylactic and isopeptide-selective ETA receptor, and tachyphylactic and isopeptide-nonselective ETB receptor. The ETA receptors may be further classified as a BQ-123-sensitive ETA1 and a BQ-123-insensitive ETA2 subtypes. The ETB receptors may also be subclassified as the ETB1 and ETB2 subtypes based on the sensitivity to the ETB antagonists.

The effects of isoquinoline derivatives, HA1077 (1-[5-isoquinolinesulfonyl]-homopiperazine) and H-7 (1-[5-isoquinolinesulfonyl]-2-methylpiperazine), on cytosolic Ca2+ levels ([Ca2+]i) and muscle tension were examined in vascular smooth muscle of rat aorta. High K+ (72.7 mM) and norepinephrine (1 microM) induced a sustained contraction with a sustained increase in [Ca2+]i. HA1077 and H-7 (3-10 microM) inhibited the increase in muscle tension more strongly than the increase in [Ca2+]i. Verapamil (10 microM) completely inhibited the increase in [Ca2+]i and the contraction induced by high K+ whereas it inhibited the increase in [Ca2+]i more strongly than the contraction due to norepinephrine. The verapamil-insensitive portion of the norepinephrine-induced contraction was inhibited by HA1077 or H-7. In Ca(2+)-free solution, 0.1 microM norepinephrine induced a transient increase in [Ca2+]i and muscle tension. The transient contraction was inhibited by 10 microM HA1077 or 10 microM H-7 without inhibiting the increase in [Ca2+]i. 12-Deoxyphorbol 13-isobutyrate (DPB) (1 microM) caused a sustained contraction, and this contraction was inhibited by HA1077 and H-7 at similar concentrations needed to inhibit the contractions induced by high K+ or norepinephrine. In rabbit mesenteric artery permeabilized with Staphylococcus aureus alpha-toxin, 100 microM HA1077 and 100 microM H-7 inhibited the contraction induced by 0.3 microM Ca2+. These results suggest that the inhibitory effects of isoquinoline derivatives, HA1077 and H-7, are due to a decrease in [Ca2+]i and in the Ca2+ sensitivity of contractile elements in vascular smooth muscle.

The effects of a non-selective P2-receptor agonist ATP and a selective P2x-receptor agonist alpha,beta-methylene-ATP on intracellular free Ca2+ level ([Ca2+]i) and force were examined in rat isolated aorta without endothelium. 2. Both ATP (1-1000 microM) and alpha,beta-methylene-ATP (0.1-100 microM) induced transient increase followed by small sustained increase in [Ca2+]i in a concentration-dependent manner. Compared with the force induced by a high concentration of KCl, the force induced by alpha,beta-methylene-ATP was smaller and that induced by ATP was much smaller at a given [Ca2+]i. 3. An L-type Ca2+ channel blocker, verapamil (10 microM), completely inhibited the high K(+)-stimulated [Ca2+]i and force. Verapamil partially inhibited the transient and sustained increases in [Ca2+]i induced by 10 microM alpha,beta-methylene-ATP and the sustained increase but not the transient increase induced by 1 mM ATP. 4. In the absence of extracellular Ca2+ (with 0.5 mM EGTA) 1 mM ATP caused transient increase in [Ca2+]i while 10 microM alpha,beta-methylene-ATP was ineffective 5. ATP, but not alpha,beta-methylene-ATP, increased the tissue adenosine 3':5'-cyclic monophosphate (cyclic AMP) level. 6. These data suggest that ATP and alpha,beta-methylene-ATP increase [Ca2+]i by an activation of both L-type and non-L-type Ca2+ channels. In addition, ATP, but not alpha,beta-methylene-ATP, increases [Ca2+]i by a release of Ca2+ from an intracellular Ca2+ store. Possible reasons are discussed as to why the increase in [Ca2+]i due to ATP and alpha,beta-methylene-ATP resulted in only a small contraction.

Changes in the cytoskeletal structure of cultured A10 smooth muscle cells induced by calyculin-A (CL-A), a potent inhibitor of types 1 and 2A protein phosphatases, were analyzed using indirect fluorescence techniques. In the presence of 1 × 10(−7) M CL-A the cells became round and subsequently detached from the substratum. The effect of CL-A was inhibited by a non-selective kinase inhibitor, K-252a, but not by EGTA. In rounded cells stress fibers were absent and staining for F-actin appeared in patches. Vinculin, one of the components of focal contacts, was localized at the periphery of control cells. CL-A treatment moved the focal contacts towards the inside of the cell along the stress fibers, and this was followed by the rounding up of the cell. In addition, rapid and marked changes in microtubule structure were observed in CL-A-treated cells. Many ‘nicks’ or ‘gaps’ were observed along the microtubules in the attached, spread cells. A filamentous network of microtubules was not observed in the detached cells, i.e. after longer exposure to CL-A. These results suggest that CL-A may change the structure of focal contacts, resulting in the rounding up of the cell, and inducing a microtubule-severing activity. These effects were independent of the external Ca2+ concentration. The changes in cytoskeletal structure may be caused by disturbing the balance of phosphorylation and dephosphorylation in the cell.

The inhibitory effects of felodipine, nifedipine and verapamil were compared in vascular smooth muscle. In rat aorta, these inhibitors attenuated the high K(+)-induced contraction with a parallel decrease in the cytosolic Ca2+ level ([Ca2+]i). Maximal inhibition was obtained with 10 nM felodipine, 100 nM nifedipine and 10 microM verapamil. The inhibitory effects were antagonized by an increase in external Ca2+ concentration to 6.5 mM and the addition of a Ca2+ channel activator, 100 nM Bay k 8644. These inhibitors also attenuated the contraction induced by norepinephrine although these effects were weaker than those on high K(+)-induced contraction. Furthermore, these inhibitors attenuated the norepinephrine-stimulated [Ca2+]i more strongly than contraction. In contrast, none of these inhibitors inhibited the transient increase in [Ca2+]i and muscle tension induced by norepinephrine in Ca(2+)-free solution and the Ca(2+)-induced contraction in permeabilized smooth muscle. These results suggest that felodipine, nifedipine and verapamil inhibit smooth muscle contraction by inhibiting Ca2+ channels at concentrations which do not change Ca2+ release or Ca2+ sensitivity of contractile elements.

In isolated rat aorta, endothelin-1 induced contractions at lower concentrations than endothelin-3. The contractile effects were augmented by removing the endothelium. In contrast, endothelin-1 and endothelin-3 at similar concentrations induced endothelium-dependent relaxation in norepinephrine-stimulated aorta. IRL 1038 ([Cys 11 , Cys 15 ]endothelin-1(11–21); 3 μM) augmented the contractile effects of endothelins only in the presence of the endothelium. IRL 1038 (0.3–3 μM) inhibited the endothelium-dependent relaxation induced by endothelins but not by carbachol. IRL 1038 itself did not change muscle tension. These results suggest that IRL 1038 is a novel antagonist of the ET B receptor responsible for the release of relaxing factor from the vascular endothelium.

The movement of a fluorescent intracellular Ca2+ indicator, fura-2, in smooth muscle was examined. Strips of rat and rabbit aortas and bovine trachea were loaded with the acetoxymethyl ester of fura-2 (fura-2/AM), followed by washing with normal physiological solution. Not only fura-2/AM but also fura-2 was detected in the washout solution. The amount of fura-2 in the cells, measured fluorometrically, decreased gradually during the washout. The decrease was fastest in rat aorta followed by rabbit aorta > bovine trachea. In rat aorta, fura-2 leakage was inhibited by an inhibitor of anion transport, probenecid, or by a decrease in bath temperature. The Ca2+ ionophore ionomycin (10 microM) increased the leakage of fura-2, which was not inhibited by probenecid, possibly because a high concentration of ionomycin nonselectively increased membrane permeability. These results suggest that fura-2/AM is cleaved to fura-2 in the cell which gradually leaked out of the cell mainly by an anion transport system. The amount of fura-2 in the cell seemed to be determined mainly by the rate of leakage of fura-2, which is the largest in rat aorta followed by rabbit aorta and bovine trachea.

To determine the role of beta-adrenoceptors in the regulation of intestinal smooth muscle, the action of isoproterenol (ISO) on cytoplasmic Ca2+ level ([Ca2+]cyt) and mechanical activity in the isolated guinea pig taenia caecum was examined. Spontaneous changes in [Ca2+]cyt and contraction were inhibited by ISO (0.1-1 microM) without changing resting [Ca2+]cyt. ISO more strongly inhibited the histamine-induced contraction than the high K(+)-induced contraction. ISO inhibited muscle tension more strongly than [Ca2+]cyt stimulated by high K+ and thus shifted the [Ca2+]cyt-tension curve to the lower-right. In the muscle stimulated by histamine, on the other hand, ISO inhibited both [Ca2+]cyt and tension. Salbutamol, a beta 2-selective agonist, showed similar effects as ISO on spontaneous, high K(+)- and histamine-stimulated [Ca2+]cyt and tension. Stimulation of beta-adrenoceptors by ISO increased cyclic AMP content without changing cyclic GMP content. These results suggest that activation of beta 2-adrenoceptors by ISO inhibits the contractions by two mechanisms of action: decrease in Ca2+ sensitivity of contractile elements in the muscle stimulated by K(+)-depolarization and decrease in [Ca2+]cyt in the muscle stimulated by histamine. These effects may be mediated by cyclic AMP.

In isolated rat aorta, endothelin (ET)-3 increased cytosolic Ca2+ ([Ca2+]i) in the endothelium at a concentration (100 nM) which had little effect on muscle resting tone. In the absence of external Ca2+, ET-3 still transiently increased endothelial [Ca2+]i. Verapamil (10 microM) did not change the effects of ET-3. In aortas stimulated with 100 nM norepinephrine, 100 nM ET-3 relaxed the muscle with an increase in endothelial [Ca2+]i. An inhibitor of nitric oxide synthase, 100 microM NG-monomethyl-L-arginine, inhibited the relaxant effect of ET-3 but not the increase in endothelial [Ca2+]i. In the absence of the endothelium or in the presence of an antagonist of ETB receptors, 3 microM IRL 1038, the ET-3-induced increase in endothelial [Ca2+]i and relaxation of norepinephrine-induced contraction were inhibited. Under these conditions, ET-3 increased smooth muscle [Ca2+]i and induced contraction, both of which were inhibited by an inhibitor of ETA receptors, 3 microM BQ123. These results suggest that ET-3 acts on ETB receptors in the vascular endothelium to increase [Ca2+]i by releasing Ca2+ from storage sites and by opening non-L type Ca2+ channels, activates nitric oxide synthase, releases nitric oxide and relaxes vascular smooth muscle. Although ET-3 also activates ETA receptors in smooth muscle to induce contraction, this effect is overcome by the relaxant effect mediated by ETB receptors.

The mechanism by which dibutyryl cyclic AMP (db-cAMP) induces vasodilatation was examined in isolated rat aorta. The contraction induced by norepinephrine (NE) was more sensitive to the inhibitory effect of db-cAMP than that induced by high K+, and the contraction induced by lower concentrations of each stimulant was more sensitive to db-cAMP than that induced by higher concentrations. Db-cAMP at 10 microM inhibited the increases in muscle tension and cytosolic Ca2+ level ([Ca2+]i) without changing the [Ca2+]i-tension relationship, suggesting that the inhibitory effect is mainly due to a decrease in [Ca2+]i. A higher concentration (300 microM) of db-cAMP inhibited muscle tension more strongly than [Ca2+]i suggesting that db-cAMP decreases Ca2+ sensitivity of contractile elements. In contrast, 10 microM verapamil inhibited the NE-stimulated [Ca2+]i more strongly than the NE-induced contraction. The verapamil-insensitive portion of the NE-stimulated [Ca2+]i and contraction was inhibited by db-cAMP, suggesting that db-cAMP and verapamil act by different mechanisms. In Ca(2+)-free solution, 1 microM NE induced transient increases in muscle tension and [Ca2+]i. The transient contraction was inhibited by 1 mM db-cAMP more strongly than [Ca2+]i. An activator of adenylate cyclase, forskolin, showed inhibitory effects similar to those of db-cAMP. The inhibitory effects of db-cAMP and forskolin were inversely proportional to [Ca2+]i before the addition of these inhibitors. These results suggest that db-cAMP inhibits smooth muscle contraction by decreasing [Ca2+]i and the Ca2+ sensitivity of contractile elements, and that both of these effects are stronger when [Ca2+]i is lower.

Inhibitory effects of a novel smooth muscle relaxant, KT-362 (5-[3-([2-(3,4-dimethoxyphenyl)-ethyl]amino)-1-oxopropyl]-2,3,4,5- tetrahydro-1,5-benzothiazepine fumarate), on contraction and the cytosolic Ca2+ level ([Ca2+]cyt) in isolated vascular smooth muscle of rat aorta were examined. 2. KT-362 inhibited the contractions induced by high K+ and noradrenaline. The inhibitory effect was antagonized by an increase in external Ca2+ concentration. A Ca2+ channel activator, Bay K 8644, did not change the effect of KT-362 on high K+-induced contraction. 3. [Ca2+]cyt, measured with fura-2-Ca2+ fluorescence, increased during the contractions induced by high K+ or noradrenaline. KT-362 decreased [Ca2+]cyt and muscle tension stimulated by high K+ or noradrenaline. By contrast, a Ca2+ channel blocker, verapamil, inhibited the noradrenaline-induced increase in [Ca2+]cyt with only partial inhibition of the noradrenaline-induced contraction and KT-362 inhibited the verapamil-insensitive portion of the contraction without changing [Ca2+]cyt. 4. In a Ca2(+)-free solution, noradrenaline and caffeine induced a transient contraction following a transient increase in [Ca2+]cyt. KT-362 inhibited the increments due to noradrenaline but not those induced by caffeine. 5. These results suggest that KT-362 inhibits vascular smooth muscle contraction by inhibiting Ca2+ channels, receptor-mediated Ca2+ mobilization, and receptor-mediated Ca2+ sensitization of contractile elements.

Okadaic acid and nifedipine inhibited contractions induced by noradrenaline (NA), KCl and ATP in guinea-pig vas deferens. NA, KCl and ATP induced initial spike-like changes followed by a sustained increase in cytosolic Ca 2+ levels ([Ca 2+ ]cyt) and tension. Okadaic acid inhibited the sustained increments in [Ca 2+ ]cyt and muscle tension due to NA and ATP more than the initial spike-like changes, whereas nifedipine more strongly inhibited the initial spike changes. Okadaic acid also inhibited the KCl-induced contraction with only a small inhibition of the stimulated [Ca 2+ ]cyt. By contrast, nifedipine (10 −7 M) inhibited the increments in both [Ca 2+ ]cyt and muscle tension due to KCl. Okadaic acid markedly inhibited the maximal contractile response to Bay K 8644 but nifedipine only shifted the response curve to the right without affecting the maximum responses. In a Ca 2+ -free medium containing EGTA and nifedipine, okadaic acid did not inhibit the residual phasic contractile response to NA (10 −4 M) but the contractile response to a subsequent addition of Ca 2+ (1.2 mM) was suppressed. These results suggest that, in guinea-pig vas deferens, okadaic acid has an inhibitory effect on smooth muscle contraction but not on the intracellular Ca 2+ mobilization. The inhibitory effect may be due to the inhibition of Ca 2+ influx and the possible interference of contractile elements.

Effects of stimulants and relaxants on the cytosolic Ca2+ level ([Ca2+]cyt) and contraction were examined in isolated canine tracheal smooth muscle. High K+ and carbachol induced a sustained increase in [Ca2+]cyt and muscle tension. Cumulative addition of KCl induced a graded increase in [Ca2+]cyt and muscle tension. Cumulative addition of carbachol induced greater contraction than high K+ at a given [Ca2+]cyt. 12-Deoxyphorbol 13-isobutyrate (DPB) (50 nmol/l) induced a small sustained contraction with little effect on [Ca2+]cyt. A higher concentration (1 μmol/l) of DPB induced a larger sustained contraction with a decrease in [Ca2+]cyt. DPB (50 nmol/l) potentiated the KCl-induced contraction without or with only a small additional increase in [Ca2+]cyt. By contrast, 1 μmol/l DPB potentiated the high-K+-induced contraction with a decrease in [Ca2+]cyt. Addition of 50 nmol/l or 1 μmol/l DPB in the presence of carbachol inhibited both [Ca2+]cyt and muscle tension. Verapamil, isoprenaline and forskolin did not change or slightly decreased [Ca2+]cyt and muscle tension in resting trachea. Verapamil inhibited the contrction and [Ca2+]cyt stimulated by high K+ and carbachol. Isoprenaline and forskolin inhibited the high-K+-induced contraction without changing [Ca2+]cyt, whercas these inhibitors inhibited carbachol-induced contraction with a relatively small decrease in [Ca2+]cyt. These results suggest that (a) sustained contractions induced by high K+ and carbachol are due to the sustained increase in [Ca2+]cyt, (b) carbachol increases the sensitivity of contractile elements to Ca2+, and (c) isoprenaline and forskolin inhibit the contraction by the decrease in [Ca2+]cyt and also by the decrease in the sensitivity of contractile elements to Ca2+. The carbachol-induced Ca2+ sensitization might be attributable to the activation of protein kinase C following the stimulation of the phosphatidylinositol turnover.

An increase in cytosolic Ca2+ level [( Ca2+]cyt) initiates smooth muscle contraction. Simultaneous measurements of [Ca2+]cyt and muscle tension provide insight into the Ca2+ regulation of smooth muscle. The photoprotein aequorin and the fluorescent Ca2+ indicator fura-2 have been used for this purpose. Although there are some differences between aequorin signal and fura-2 signal, comparison between [Ca2+]cyt and muscle tension in vascular smooth muscle indicates that high K+ and agonists that operate via specific receptors (receptor-agonists) induce sustained contraction whereas stimulated [Ca2+]cyt gradually decreases during prolonged exposure to agonists. Furthermore, greater contraction is produced by receptor-agonists than high K+ at a given [Ca2+]cyt. By contrast, cyclic AMP and cyclic GMP decrease muscle tension more strongly than [Ca2+]cyt. These results suggest that contractility of vascular smooth muscle is regulated not only by [Ca2+]cyt, but also by other mechanisms, possibly including Ca2+ sensitivity of the contractile elements. Ca2+ sensitization may be due to activation of protein kinase C.

1. Repeated oral administrations of tryptic hydrolysate of bovine milk casein (CEI) showed antihypertensive effect in spontaneously hypertensive rats. 2. Single oral administration of CEI antagonized the pressor response to angiotensin I. 3. Bovine milk casein hydrolysate inhibited the angiotensin I-converting enzyme (ACE) activity. Three peptides with ACE-inhibiting activity were isolated from CEI. 4. It is suggested that ACE-inhibiting peptides in the tryptic hydrolysate milk casein are absorbed from the intestinal tract and produce an antihypertensive effect.

Objective—Although adenovirus is a powerful tool for vascular research and therapy, endothelial impairment after infection has been reported. We investigated the mechanisms of this impairment and the effect of dexamethasone (DEX) on gene transfer into the vascular endothelial cells.Methods and Results—β-Galactosidase gene encoding adenovirus vector (β-gal-Ad) (7.5×108plaque-forming units/mL) transduced β-gal into the rabbit organ–cultured pulmonary endothelium, followed by an apoptosis and an impairment of endothelium-dependent relaxation (EDR). Endothelial cell infected by β-gal-Ad expressed proinflammatory genes mRNAs and suppressed endothelial nitric oxide synthase (eNOS) mRNA. Treatment with DEX dramatically increased β-gal protein expression in the endothelium, attenuated β-gal-Ad–induced apoptosis, and prevented the impairment of EDR. DEX also suppressed the mRNAs expressions of proinflammatory genes and recovered eNOS mRNA expression in organ-cultured vascular endothelium. In addition, we confirmed the DEX’s beneficial effects in an endothelial cell line (in vitro) and rat femoral artery (in vivo) experiments.Conclusion—These results suggest that adenovirus vector induces host-immune responses and apoptosis in vascular endothelial cells. DEX is found to be a useful and potent tool to prevent the Ad-induced impairments of the endothelium and to optimize gene expression efficiency by adenovirus vector at the protein translation level in both in vitro and in vivo experiments.

In the present study, we assessed the effects of dexamethasone on fetal bovine serum-induced dysfunction of mesenteric endothelial cells using an organ culture procedure. In rabbit mesenteric arteries cultured in the presence of 10% fetal bovine serum for 7 days, the endothelium-dependent, nitric oxide (NO)-mediated relaxations caused by substance P and ionomycin were decreased as compared to those in non-treated arteries. Dexamethasone (3 microM) inhibited the proliferative stimuli-induced endothelial dysfunction without affecting the contractility or NO susceptibility of smooth muscle cells. Cross-sectioned hematoxylin-eosin staining and whole-mount CD31 staining indicated that chronic proliferative stimulation induced detachment of endothelial cells from the tunica intima in some regions, and also caused thickening of the arterial wall and shortening of the internal diameter. Endothelial NO synthesis (eNOS) mRNA expression was also decreased by the treatment with fetal bovine serum. The dexamethasone treatment did not inhibit the smooth muscle hypertrophy, but it inhibited the peeling of endothelial cells and recovered the eNOS mRNA expression. These results suggest that DEX ameliorate the impairments of arterial relaxation induced by proliferative stimuli and that these beneficial effects may be mediated by maintaining the adhesion of endothelial cells to the vascular wall and/or by recovering eNOS mRNA expression.

We assessed the effects of dexamethasone (DEX) on hypoxia-induced dysfunction of the pulmonary endothelium using organ-cultured rabbit intrapulmonary arteries; 3-microM DEX inhibited the 7-day hypoxia (5% oxygen)-induced impairments of endothelial-dependent relaxation, cGMP accumulation, and increase in intracellular Ca(2+) level under substance P-stimulated conditions. Treatment with DEX over the final 3 days of the 7-day hypoxic exposure period also restored the decreased endothelium-dependent relaxation. Although chronic hypoxia did not change the mRNA expression of endothelial nitric oxide synthase (eNOS), 3 microM of DEX increased eNOS mRNA expression in both the hypoxic and normoxic (20% oxygen) pulmonary endothelium. On the other hand, eNOS protein expression was not changed in any of the arteries. We next assessed the effects of DEX on the eNOS activation pathway. Chronic hypoxia impaired eNOS phosphorylation and Akt phosphorylation under both the nonstimulated and substance P-stimulated conditions, and 3-microM DEX restored these phosphorylations. Morphologic study revealed that 3-microM DEX inhibited chronic hypoxia-induced atrophy of endothelial cells and eNOS protein condensation into plasma membranes. These results suggest that DEX exerts beneficial effects on chronic hypoxia-induced impairments of nitric oxide-mediated arterial relaxation by increasing eNOS mRNA expression and inhibiting hypoxia-induced impairments in eNOS activation pathway with atrophy of endothelial cells.

To investigate the role of actin filaments (F-actin) for human immunodeficiency virus type 1 (HIV-1) production in host cells, the effect of mycalolide B that is a novel actin-depolymerizing marine toxin was examined. Mycalolide B blocked the production of HIV-1 from primary infected T-lymphoblastoid and clonically infected monocytoid cells in a concentration-dependent manner. In the presence of 10 microM of mycalolide B, F-actins were disorganized and mostly disappeared in the host cells, and viral envelope- and capsid-proteins did not reach the plasma membrane, but were distributed in the cytoplasm forming aggregates. In electron micrographs, no HIV-1 virions were detected on the cell surface, but many lysosome-like vesicles containing electron dense granules were observed in the cytoplasm, implying that mycalolide B did not disturb the synthesis of viral proteins, but rather inhibited their transport processes of HIV-1 in the host cells.

In order to elucidate the signal transduction pathways of vascular smooth muscle contractions induced by stimulation of receptors for 5-hydroxytryptamine (5-HT) and thromboxane A2 (TXA2), both of which are released from activated platelets, we examined whether protein kinases, such as tyrosine kinase, p38 mitogen-activated protein kinase (MAPK) and protein kinase C (PKC), are involved in the contraction produced by either 5-HT or U46619 (an analog of TXA2) in the rat aorta. Both 5-HT and U46619 induced sustained contractions, which were markedly reduced in the absence of extracellular Ca2+. Verapamil (a L-type Ca2+ channel blocker) markedly inhibited the contractile response to 5-HT, while the U46619-induced contraction was only slightly inhibited by verapamil. Both contractile responses to 5-HT and U46619 were significantly inhibited by calphostin C (a PKC inhibitor). On the other hand, both genistein (5 microM, a tyrosine kinase inhibitor) and SB203580 (a p38 MAPK inhibitor) significantly inhibited 5-HT-induced contractions but had little effects on the contractions induced by U46619. These results suggest that the signal transduction mechanisms involved in the contractions mediated via 5-HT and TXA2 receptors are different as follows. Both the tyrosine kinase and p38 MAPK pathways are involved in 5-HT contraction but not in TXA2 contraction, while both contractions are strongly dependent on transplasmalemmal Ca2+ entry. The contractile responses to both 5-HT and TXA2 involve voltage-dependent Ca2+ channels and PKC.

We investigated the role of Rndl, a member of the small GTP-binding Rho protein family, in the change in Ca 2 + sensitivity of contractile element in rat myometrium at estrus, gestation, and postpartum stages. In the permeabilized muscles, GTPyS or carbachol with GTP increased Ca 2 + sensitivity of contractile force in non-pregnant myometrium at the estrus stage, whereas these stimuli were ineffective in pregnant myometrium at day 21. After postpartum, the reduced Ca 2 + sensitization was recovered. Semi-quantitative RT-PCR analysis indicated that the expressions of RhoA, ROCKI, and ROCKII were not significantly different between non-pregnant and pregnant myometria. In contrast, the expression of Rndl was increased during the course of pregnancy, reaching a maximal at day 21, and rapidly declined after the delivery. On the other hand, Ca 2 + sensitization of contractile elements was decreased during the progress in gestation. These results suggest that Rndl may have an important role as a negative-feedback control of uterine contraction during gestation through the inhibition of RhoA-mediated increase in the Ca 2 + sensitivity of contractile elements.

1. Activation of protein kinase C (PKC) by phorbol 12,13-dibutylate (PDBu, 1 microm) induced sustained contractions with no increase in [Ca2+]i in nonpregnant and pregnant human myometria. The contractile effects of PDBu in pregnant myometrium were much greater than those in nonpregnant myometrium, and the contractions in pregnant myometrium were accompanied by an increase in myosin light chain (MLC) phosphorylation at Ser19. 2. The contraction induced by PDBu in pregnant myometrium was inhibited by the inhibitors of conventional PKC isoforms, bisindolylmaleimides and indolocarbazole, such as Go6976, Go6983, and Go6850 (1 microM). LY333531 (1 microM), a specific inhibitor of PKC beta, also inhibited the PDBu-induced contraction in the pregnant myometrium. 3. In the pregnant myometrium permeabilized with alpha-toxin, PDBu increased the contractions induced at fixed Ca2+ concentration (0.3 microM) both in nonpregnant and pregnant myometria, indicating Ca2+ sensitization of contractile elements. 4. Western immunoblot analysis indicated that pregnant myometrium contained PKC isozymes such as conventional PKC (alpha, beta, gamma), novel PKC (delta, epsilon, theta), and atypical PKC (zeta but not iota and lambda). RT-PCR and real-time RT-PCR analysis indicated that, among the conventional PKC, the levels of mRNA of beta isoform in pregnant human myometrium were greater than those in nonpregnant myometrium. 5. CPI-17 is a substrate for PKC, and the phosphorylated CPI-17 is considered to inhibit myosin phosphatase. The levels of CPI-17 mRNA and protein expression were also greater in the pregnant myometrium. 6. These results suggest that the PKC-mediated contractile mechanism is augmented in human myometrium after gestation, and that this augmentation may be attributable to the increased activity of the beta PKC isoform and CPI-17.

In the mast cell signaling pathways, the binding of immunoglobulin E (IgE) to FcϵRI, its high-affinity receptor, is generally thought to be a passive step. In this study, we examined the effect of IgE alone, that is, without antigen stimulation, on the degranulation in mast cells. Monomeric IgE (500–5,000 ng/ml) alone increased cytosolic Ca2+level ([Ca2+]i) and induced degranulation in rat basophilic leukemia (RBL)-2H3 mast cells. Monomeric IgE (5,000 ng/ml) alone also increased [Ca2+]iand induced degranulation in bone marrow-derived mast cells. Interestingly, monomeric IgE (5–50 ng/ml) alone, in concentrations too low to induce degranulation, increased filamentous actin content in RBL-2H3 mast cells. We next examined whether actin dynamics affect the IgE alone-induced RBL-2H3 mast cell activation pathways. Cytochalasin D inhibited the ability of IgE alone (50 ng/ml) to induce de novo actin assembly. In cytochalasin D-treated cells, IgE (50 ng/ml) alone increased [Ca2+]iand induced degranulation. We have summarized the current findings into two points. First, IgE alone increases [Ca2+]iand induces degranulation in mast cells. Second, IgE, at concentrations too low to increase either [Ca2+]ior degranulation, significantly induces actin assembly, which serves as a negative feedback control in the mast cell Ca2+signaling and degranulation.

Studies were conducted in C57BL/6N Crj male mice and in cultured hepatocytes to clarify the relationship between galactosamine (GaIN) induced apoptosis and [Ca2+]i kinetics. Chlorpromazine (CPZ), a Ca(2+)-calmodulin antagonist, and verapamil (VR), a Ca(2+)-channel blocker each inhibited GaIN-induced DNA fragmentation and the appearance of apoptotic bodies. The kinetics of calcium uptake were evaluated using a calcium analyzer with the acetoxymethyl ester of fura-PE3 (fura-PE3/AM, 2.5 microM) as the calcium reporter. An increase in [Ca2+]i was detected in the cultured hepatocytes within 3 hours after treatment with 20 mM GaIN; this increase was inhibited by pretreatment with either 20 microM CPZ or 30 microM VR. Ca2+ imaging by confocal laser scanning microscopy showed that increase in [Ca2+]i after treatment with GaIN was initially localized around nuclei, while [Ca2+]i signals were later diffuse and observed throughout the cytoplasm. The activities of lactate dehydrogenase (LDH) and serum glutamate-pyruvate transaminase (sGPT), used as indicators of plasma membrane damage and leakage, however, were not reduced by pretreatment with CPZ or VR. From these findings, we infer that the DNA fragmentation in GaIN-induced hepatocyte apoptosis is associated with an elevation in the perinuclear concentration of Ca2+, but GaIN-induced necrotic cell death is triggered through pathway(s) that are insensitive to blockage of Ca2+ influx and therefore appear to occur independently of elevation in [Ca2+]i. These results help to clarify the role of calcium flux in hepatocyte apoptosis and necrosis induced by exposure to hepatotoxins in vivo and in vitro.

Calponin (h1 or basic) is an actin-binding protein that is expressed abundantly in smooth muscle. Our previous study using h1 calponin-null mutant mice demonstrated that h1 calponin inhibits the shortening velocity of smooth muscle contraction without significantly affecting the amplitude of force production. Furthermore, early onset of osteogenesis and increased bone formation have been reported in mutated mice. In the present study, we examined the effect of h 1 calponin depletion on the metabolism and behavior of mice and found that the mutated mice showed increased locomotor activity, as well as increased intake of food and water, associated with the decreased number of neurons in the paraventricular nucleus of the hypothalamus (PVN).

1. Xestospongin-C isolated from a marine sponge, Xestospongia sp., has recently been shown to be a membrane-permeable IP(3) receptor inhibitor. In this study we examined the effects of this compound on smooth muscle from guinea-pig ileum. 2. In guinea-pig ileum permeabilized with alpha-toxin, xestospongin-C (3 microM) inhibited contractions induced by Ca(2+) mobilized from sarcoplasmic reticulum (SR) with IP(3) or carbachol with GTP, but not with caffeine. 3. In intact smooth muscle tissue, xestospongin-C (3-10 microM) inhibited carbachol- and high-K+-induced increases in [Ca(2+)](i) and contractions at sustained phase. 4. It also inhibited voltage-dependent inward Ba(2+) currents in a concentration-dependent manner with an IC(50) of 0.63 microM. Xestospongin-C (3-10 microM) had no effect on carbachol-induced inward Ca(2+) currents via non-selective cation channels; but it did reduce voltage-dependent K+ currents in a concentration-dependent manner with an IC(50) of 0.13 microM. 5. These results suggest that xestospongin-C inhibits the IP(3) receptor but not the ryanodine receptor in smooth muscle SR membrane. In intact smooth muscle cells, however, xestospongin-C appears to inhibit voltage-dependent Ca(2+) and K+ currents at a concentration range similar to that at which it inhibits the IP(3) receptor. Xestospongin-C is a selective blocker of the IP(3) receptor in permeabilised cells but not in cells with intact plasma membrane.

In the pulmonary artery isolated from 1-week hypoxia-induced pulmonary hypertensive rats, endothelial NO production stimulated by carbachol was decreased significantly in in situ visualization using diaminofluorescein-2 diacetate and also in cGMP content. This change was followed by the decrease in carbachol-induced endothelium-dependent relaxation. Protein expression of endothelial NO synthase (eNOS) and its regulatory proteins, caveolin-1 and heat shock protein 90, did not change in the hypoxic pulmonary artery, indicating that chronic hypoxia impairs eNOS activity at posttranslational level. In the hypoxic pulmonary artery, the increase in intracellular Ca(2+) level stimulated by carbachol but not by ionomycin was reduced. We next focused on changes in Ca(2+) sensitivity of the eNOS activation system. A morphological study revealed atrophy of endothelial cells and a peripheral condensation of eNOS in hypoxic endothelial cells preserving co-localization between eNOS and Golgi or plasma membranes. However, eNOS was tightly coupled with caveolin-1, and was dissociated from heat shock protein 90 or calmodulin in the hypoxic pulmonary artery in either the presence or absence of carbachol. Furthermore, eNOS Ser(1177) phosphorylation in both conditions significantly decreased without affecting Akt phosphorylation in the hypoxic artery. In conclusion, chronic hypoxia impairs endothelial Ca(2+) metabolism and normal coupling between eNOS and caveolin-1 resulted in eNOS inactivity.

Background and aims: The endothelin ETB receptor null rat (ETB(−/−)R) has an intestinal segment without ganglia, and this rat is characterised by intestinal obstruction similar to that observed in human Hirschsprung's disease. In the present study, we have examined the myogenic mechanism responsible for obstruction in the ETB(−/−)R. Results: The ETB(−/−)R had an enlarged belly and the average lifespan was 18.1 days. The bowel from the rectum to the lower part of the small ileum was constricted whereas the upper region was dilated with faecal stasis and thus presented as megaileum. The constricted muscle segments without ganglia had a greater increase in absolute force when stimulated by carbachol, high K+, and endothelin-1 compared with that of normal siblings. In contrast, in the dilated part with ganglia, the absolute contractile force due to these stimulants in the ETB(−/−)R was not different from that in the ETB(+/+)R. Such a functional hypertrophy of the musculature was observed in parts of the colon, caecum, and distal ileum without ganglia but not in the part of the proximal ileum and jejunum with ganglia. Morphological study demonstrated that the thickness of the circular and longitudinal muscle layers was greater in the constricted part of the intestine in the ETB(−/−)R, and these changes were associated with an increase in the number of smooth muscle cells. Conclusions: Our findings suggest that both increased contractility of smooth muscle and increased thickness of the intestinal muscular wall may contribute to the intestinal obstruction in the ETB(−/−)R.

An autolysate of bonito bowels was treated with ultrafiltration, loose RO concentration, ion-exchange chromatography, and reverse phase chromatography to increase its potency to inhibit angiotensin I-converting enzyme (ACE) activity by 16-fold. Oral administration of the partially purified autolysate decreased the systolic blood pressure of spontaneously hypertensive rats (SHR) in a dose-dependent manner at the doses of 1 g peptides/kg or higher. The relationship between the antihypertensive activity (in vivo) of the partially purified preparation and its ACE inhibitory activity (in vitro) in comparison with previously reported ACE inhibitory peptides is discussed.

The phosphatase inhibitor okadaic acid at 100 nM slowly but completely inhibited high K(+)-induced contraction in the rat aorta (t1/2 = 118.9 min). High K(+)-induced contraction was partially inhibited (to 37-65%) by 1 microM forskolin, 100 microM dibutyryl cyclic AMP, 100 nM atrial natriuretic peptide, 1 microM nitroglycerin, 10 nM sodium nitroprusside, 300 pM nicardipine or 100 nM verapamil. The rate of relaxation due to okadaic acid became faster when the contraction was partially inhibited by these compounds. Augmentation of the relaxation was greater with forskolin and dibutyryl cyclic AMP than with the other inhibitors. These results support the suggestion that okadaic acid inhibits phosphatase to augment the phosphorylation due to cyclic AMP-dependent kinase, resulting in smooth muscle relaxation.

We have attempted to determine the chronic effects of doxorubicin, a commonly used anticancer agent, on vascular endothelium using an organ culture system. In rabbit mesenteric arteries treated with 0.3 microM doxorubicin for 7 days, rounding and concentrated nuclei and TUNEL-positive staining were observed in endothelial cells, indicating DNA damage and the induction of apoptosis. However, the endothelium-dependent relaxation induced by substance P and the expression of mRNA encoding endothelial NO synthase (eNOS) did not differ from those in control arteries. In arteries treated with a higher concentration (1 microM) of doxorubicin, apoptosis and damage to nuclei occurred in the endothelial cells at the third day of treatment, and the detachment and excoriation of endothelium from the tunica interna of the vascular wall were also observed. The impairment of endothelium-dependent relaxation was observed at the fifth day of the treatment with 1 microM doxorubicin. Additionally, apoptotic change in the smooth muscle layer was observed at this concentration of doxorubicin. Apoptotic phenomena were further confirmed by DNA fragmentation using isolated bovine aortic endothelial cells (BAECs) and A7r5 vascular smooth muscle cells, and it was revealed that BAECs are more sensitive than A7r5 to the apoptotic effect of doxorubicin. These results suggest that chronic treatment with doxorubicin at therapeutic concentrations induces apoptosis and excoriation of endothelial cells, which diminishes endothelium-dependent relaxation.
