Objectives: The aim of this study is to indirectly compare and rank the different drugs that have been studied in randomized clinical trials (RCTs) in patients with tardive dyskinesia (TD) in terms of their efficacy in ameliorating the symptoms of TD and safety. Design: A network meta-analysis and a systematic review were registered prospectively on PROSPERO under the ID: CRD42023407823 and were conducted in accordance with the PRISMA-NMA guidelines. Data sources: PubMed, Scopus, The Cochrane Central Register of Controlled Trials (CENTRAL), Web of Sciences, and Clinicaltrials.gov were searched to identify relevant records. Eligibility criteria for selecting studies: Any parallel randomized blinded controlled clinical trials that studied the use of any medications in treating TD and assessed the symptoms using a functional scale that has been previously validated. Data extraction: The standardized mean difference of improvement along with the reported adverse events for each drug was extracted from each trial, and a network meta-analysis was conducted using a random-effects model. Results: One thousand eight hundred seventeen patients in 33 RCTs were included in the analysis. Twenty-three different drugs were compared to placebo in terms of reduction in TD symptoms. Among these, valbenazine 80 mg (SMD = − 1.66, 95%CI = [− 2.55; − 0.78]), valbenazine 40 mg (− 1.00, [− 1.89; − 0.11]), and vitamin E (− 0.77, [− 1.45; − 0.1]) significantly reduced TD symptoms in comparison to placebo, while deutetrabenazine 36 mg (− 1.00, [− 2.12; 0.11]) and reserpine (− 0.54, [− 1.09; 0.02]) did not significantly reduce symptoms. Some serious adverse events were reported for valbenazine and deutetrabenazine, which included mainly psychiatric symptoms such as depression, worsening of schizophrenia, and suicidal ideation, while mild adverse events were reported for other drugs, and their incidence in the treatment arms was comparable to those in the placebo arm. Conclusions: Valbenazine (80 and 40 mg) and vitamin E demonstrated efficacy in treating tardive dyskinesia. The present article provides a personalized account on CH-arylation reactions employed for the synthesis of heterocycles. The presence of a nitro group allowed for direct and regioselective CH-arylations of pyrazoles, imidazoles, indoles and a variety of purine analogues. Direct CH-arylations without the presence of an activating nitro-group were employed for inter- and intramolecular reactions of purine derivatives, which allowed for the synthesis of a great variety of polycyclic systems. Domino C–N coupling / hydroamination / CH-activation reactions of diarylacetylenes with anilines allowed for the synthesis of polycondensated N -heterocycles. Products include indolo- and azaindolo[1,2- f ]phenanthridines, quinolino[3′,4′:4,5]pyrrolo[1,2- f ]phenanthridines, pyrimido[5′,4′:4,5]pyrrolo[1,2- f ]phenanthridines, and benzothieno[2′,3′:4,5]pyrrolo[1,2- f ]phenanthridines. The reaction of N -heterocycles, such as indoles, with 1,1-difluoroalkenes resulted in a twofold addition-elimination reaction to give 1,1-diaminoalkenes, which were transformed by CH-arylation into various polycondensated heterocycles, such as indoloisoquinolines, thienoindolizines, oxepines and helicenes. Pyridofluoranthenes, diindenopyrene and azadiindenopyrenes were prepared by a combination of Pd-catalyzed cross-coupling reactions with acid-mediated cycloisomerizations and Pd-catalyzed intramolecular CH-arylations. The dithioacetalization of lactaldehyde derivatives with ethane-1,2-, propane-1,3-, butane-1,4-, and pentane-1,5-dithiols in the presence of 4 mol% of scandium triflate has been described. A series of cyclic dithioacetals were obtained with yields ranging from quantitative to 37%. The dithioacetalization of lactaldehyde derivatives with butane-1,4-dithiol and pentane-1,5-dithiol groups are accompanied by the formation of 14- and 16-membered macrocyclic sulfur structures with yields of 3% and 18%, respectively. In the case of a cyclic dithioacetal derivative with three methylene groups, a diastereoisomeric pair of enantiomers was obtained, the structure of which was confirmed by single-crystal X-ray diffraction analysis. The metal-free direct oxidative cross-dehydrogenative C–H bond amination of imidazopyridines with pyrazole in water as solvent has been developed under K2 S2 O8-promoted mild conditions. This strategy was applied to variable imidazoheterocycles to give amination products in moderate to good yields. The described protocol also offers scale-up synthesis for the construction of C–N bonds, which makes it suitable for the synthesis of biologically active compounds and pharmaceuticals. A phytochemical investigation on the 80% EtOH extract of the leaves of Paederia scandens (Lour.) Merr. resulted into the isolation of three undescribed iridoid glycosides, 10-O-trans-p-coumaroyl-(4R,6R)-3,4-dihydro-3α-methylthiopaederoside (1), 10-O-trans-feruloyl-(4S,6R)-3,4-dihydro-2′-O-3α-paederoside (2), and 10-O-trans-caffeoyl-paederosidic acid ethyl ester (3). The structures of the new compounds were elucidated by spectral methods such as 1D and 2D (1H-1H COSY, HMQC, and HMBC) NMR spectroscopy, as well as high resolution mass spectrometry. The isolated compounds were tested in vitro for cytotoxic activity against five endocrine tumor cell lines. Twenty 3-acyloxymaltol/ethyl maltol derivatives (7a-j and 8a-j) were synthesized and evaluated in vitro for their anti-oomycete activity against Phytophthora capsici, respectively. Among all of twenty derivatives, more than half of the compounds 7f, 7h, 8a-h and 8j had anti-oomycete activity higher than the positive control zoxamide (EC50 = 22.23 mg/L), and the EC50 values of 18.66, 20.32, 12.80, 16.18, 10.59, 14.98, 16.80, 10.36, 15.32, 12.64, and 13.59 mg/L, respectively. Especially, compounds 8c and 8f exhibited the best anti-oomycete activity against P. capsici with EC50 values of 10.59 and 10.36 mg/L, respectively. Purpose: Rare yeasts species are increasingly reported as causative agents of invasive human infection. Proper identification and antifungal therapy are essential to manage these infections. Candida blankii is one of these emerging pathogens and is known for its reduced susceptibility to multiple antifungals. Methods: To obtain more insight into the characteristics of this species, 26 isolates reported as C. blankii were investigated using genetic and phenotypical approaches. Results: Among the 26 isolates, seven recovered either from blood, sputum, urine, or the oral cavity, displayed substantial genetic and some phenotypical differences compared to the other isolates, which were confirmed as C. blankii. We consider these seven strains to represent a novel species, Tardiomyces depauwii. Phylogenomics assigned C. blankii, C. digboiensis, and the novel species in a distinct branch within the order Dipodascales, for which the novel genus Tardiomyces is erected. The new combinations Tardiomyces blankii and Tardiomyces digboiensis are introduced. Differences with related, strictly environmental genera Sugiyamaella, Crinitomyces, and Diddensiella are enumerated. All three Tardiomyces species share the rare ability to grow up to 42 °C, display slower growth in nutrient-poor media, and show a reduced susceptibility to azoles and echinocandins. Characteristics of T. depauwii include high MIC values with voriconazole and a unique protein pattern. Dinitropyridines are one of the promising classes of heterocyclic compounds. They are considered as useful precursors of explosives and energetic compounds, agrochemicals, biosensors and biologically active compounds with diverse properties: antitumor, antiviral, anti‐neurodegenerative. Preliminary analysis of literature data shows that the chemistry of polynitropyridines has been actively developing over the past few decades. This is evidenced by the steadily growing annual number of publications. Chromenes and 2H‐Chromenes are vital components of natural products, pharmaceuticals, and agrochemicals that have captured the attention of researchers worldwide. These heterocyclic compounds represent an essential class of structural scaffolds, and the quest for their synthesis in a mild and straightforward way has gained significant momentum. Chromenes are oxygen‐containing heterocycles that have been widely studied due to their potential medicinal properties. In the last two decades, researchers have made remarkable progress towards the synthesis of chromene based heterocyclic compounds using O‐propargylated benzaldehydes as a substrate. These substrates are highly versatile and contain functional groups that make them ideal for a wide range of reactions. This review article provides an in‐depth analysis of the latest advances in alkynes' functionalization, emphasizing the substrate scope, limitations, regioselectivity control, and applications of these reactions. Herein, we have developed a metal‐free, atom‐economical, intrinsically sustainable approach for synthesizing unprotected 4,5‐diamino pyrazoles from readily available 1,2‐diaza‐1,3‐dienes. This approach utilizes inexpensive hydrazine hydrate as the aminating source. Further, we have explored the reactivity of 4,5‐diamino pyrazole to synthesize fused pyrazolo[3,4‐d]‐1,2,3‐triazole heterocyclic compounds. We have also successfully demonstrated the synthetic utility of this methodology in synthesizing two drug analogs: analgesic difenamizole and anti‐psychotic CDPPB. In the current study, a straightforward high-performance liquid chromatographic (HPLC) approach was developed and validated to identify a novel drug compound called 5-(4-bromophenyl)-N-(2-chloro-4-nitrophenyl)-1,3,4-oxadiazol-2-amine (A3). By putting the compound's solution under hydrolytic, oxidative, and photolytic stress, the method's capacity to detect stability was put to the test. A gradient mobile phase of acetonitrile, orthophosphoric acid, and methanol (90:051:05 v/v), at a flow rate of 1.00 mL/min, was used for the chromatographic separation on a C18 column (Promosil, 5μ, 4.60 250 mm), which was maintained at 40°C and a photodiode array detector was used for detection. At concentrations between 10 and 100.00 μg/mL, Beer's rule was observed. The recovery (99.25–100%, standard deviation [SD] 5%), intraday accuracy and precision (98.62–99.91%, relative standard deviation [RSD] 5%), interday accuracy and precision (96.25–99.91%, RSD 5%), and intermediate accuracy and precision (98.10–99.91%, RSD 5%) all indicated that the developed method was reliable, repeatable, reproducible, and robust. In cases of thermal and moisture deterioration, respectively, the compound's peak resolution and selectivity factors from the nearest resolving peak revealed specificity and selectivity. The selected synthesized compound barely broke down under oxidative and alkaline hydrolytic stress. However, the compound was resistant to photolysis in neutral and acidic environments. The results of this study demonstrate the sensitivity, specificity, and selectivity of the established approach for quality control, stability testing, and preformulation investigations. ABSTRACT This study synthesized 1,6‐naphthyridine as a novel class of fused heterocyclic compounds with high efficiency using a multicomponent reaction. The reaction involved benzaldehyde or its derivatives, 2 mol of malononitrile, and 4‐aminocumarin in an aqueous solution at room temperature in the presence of SiO2/Fe3O4@MWCNTs as a recyclable catalyst. This study examines the antioxidant properties of 1,6‐naphthyridine in addition to the other research undertaken in this work. The production process of 1,6‐naphthyridine demonstrated several advantageous features, including rapid reactions, high yields of the final product, and straightforward separation of the catalyst and product from the reaction mixture. Heterocycles containing both phosphorus and nitrogen have seen increasing use in recent years in luminescent materials, coordination chemistry and as building blocks for inorganic polymers, yet their chemistry is currently dominated by five- and six-membered derivatives. Seven-membered P/N heterocycles are comparatively scarce and lack general, high yielding syntheses. Here, we explore the synthesis and characterisation of 1,2,5-diazaphosphepines from azophosphines. The mechanism has been probed in detail with both computational and experimental studies supporting a stepwise mechanism to form a five-membered ring, and subsequent ring expansion to the diazaphosphepine. Regioselective synthesis of five- and seven-membered rings is possible using asymmetric alkynes. The Lewis acidic borane B(C6F5)3 could either catalyse the formation of the seven-membered ring (iPr derivative) or trap out a key intermediate via a frustrated Lewis pair (FLP) mechanism (tBu derivative). Derivatives of the first substituted 7‐imino‐1,7‐dihydrobenzo[e][1,2,4]triazines, namely 1‐(2,3,5,6‐tetrafluoropyridin‐4‐yl)‐N,3‐diphenyl‐7‐(phenylimino)‐1,7‐dihydrobenzo[e][1,2,4]triazin‐6‐amine and 1‐(pentafluorophenyl)‐N‐phenyl‐7‐(phenylimino)‐3‐(2,3,4,5‐tetrafluorophenyl)‐1,7‐dihydrobenzo[e][1,2,4]triazin‐6‐amine, were unexpectedly isolated as the only products in yields of about 20–30 % after an attempt to obtain Blatter radicals from the corresponding amidrazones bearing polyfluorinated electron‐withdrawing substituents. Over the last three decades, owing to labour cost, time and especially environmental issues, one‐pot synthesis of heterocyclic compounds involving tandem or multicomponent (sequential or consecutive) reactions has gained particular attention worldwide. Most of the marketed drug molecules hold one or more heterocyclic rings. Due to some side effects of several drugs along with the gradual emergence of antimicrobial resistance, the necessity for developing new heterocyclic molecules continues; special attention should be paid in this respect towards employing more one‐pot reactions for their syntheses. Pyrazolopyridines are fused heterocycles holding pyridine and pyrazole and possess a wide variety of biological and pharmaceutical properties. Several existing drug molecules also bear pyrazolopyridine scaffolds. The present perspective is an endeavour to encompass green syntheses of different isomeric pyrazolopyridine systems, most of these published during the past five years or more for a few cases. 5-endo-trig radical cyclization has long been recognized as one of the most straightforward ways for the construction of densely functionalized five-membered rings. Nevertheless, according to Baldwin's rules, the 5-endo-trig radical cyclization is kinetically disfavored due to stereoelectronic effects and thus usually proceeds via a slow rate, which renders its application a challenging task. In recent years, with the emergence of efficient radical generation methods and effective cyclization strategies, 5-endo-trig radical cyclization has been successfully accelerated to a synthetically useful rate and has been utilized in the access of diverse five-membered carbo- and heterocyclic compounds. This review comprehensively summarizes the methodologies involving the 5-endo-trig radical cyclization process, with particular emphasis on the elucidation of the promoting strategies, which include the polar effect, geometrical constraints, spin delocalization effect, and persistent radical effect. Each of these strategies is discussed in detail, with illustrative examples from recent literature studies to highlight their practical applications and effectiveness. It is anticipated that the in-depth understanding of the 5-endo-trig radical cyclization provided by this review would inspire further innovation of this privileged reaction mode and expand its applications. Recent studies strongly suggest that gut microbiome can influence brain functions and contribute to the development of Alzheimer's disease (AD). However, reported changes in the gut microbiomes in AD patients from different countries are not similar, and more research is needed to reveal the relationships between human microbiomes and AD in diverse ethnic populations. There is also an assumption that microbiome‐associated peripheral inflammation might drive the development of sporadic AD. This cross‐sectional study is aimed at analyzing the gut microbial profile and exploring potential associations with blood cytokines and some clinical parameters among individuals diagnosed with Alzheimer's in Kazakhstan. Consistent with previous studies, we have found that the microbial landscape in AD reveals specific alterations in the gut microbiome. Specifically, the AD patient group showed a decreased Firmicutes/Bacteroidetes ratio. The differential abundance analysis highlighted a dysbiosis in the gut microbiota of AD patients, marked by a reduced presence of Bifidobacterium, particularly B. breve. In our study, AD patients' altered gut microbiota composition notably features an increased presence of Pseudomonadota like Phyllobacterium and inflammatory bacteria such as Synergistetes and the Christensenellaceae family. The metabolic profiling of the AD microbiome reveals a predominant presence of pathways related to sugar, carrier molecules, tetrapyrrole, pyrimidine biosynthesis, and nucleic acid processing. This analysis also highlighted a marked reduction in SCFA, carbohydrate, polysaccharide, polyamine, and myo‐inositol degradation pathways. The increases in the proinflammatory cytokines IL‐1a, IL‐8, IL‐17A, IL‐12p40, TNF‐β, MCP‐1, IL‐2, and IL‐12p70 and the anti‐inflammatory cytokines IL‐10 and IL‐13 were observed in AD patients. Key variables driving the separation of AD and controls include inflammatory markers (IL‐1a and IL‐8), growth factors (EGF), lipids (LDL), BMI, and gut microbes, like genus Tyzzerella and Turicibacter and species Parabacteroides distasonis and Bacteroides eggerthii. We have also demonstrated that almost all cytokines strongly correlate with serum adiponectin levels and specific microbial taxa in AD patients. Thus, our findings identify potential microbial and inflammatory signatures in an ethnically distinct cohort of AD patients. Background: Breastfeeding offers significant health benefits, but its practice and success can vary. While research on induced lactation in cisgender women has been documented, there is limited research on lactation induction in transgender women. Case presentation: A 50-year-old transgender woman undergoing hormone therapy and living with a pregnant partner sought to co-feed using induced lactation. After approval by the hospital ethics committee, a regimen of estradiol, progesterone, and domperidone was initiated, accompanied by nipple stimulation. Lactation was successfully induced and maintained, with milk composition analysis indicating high levels of protein and other key nutrients. This case, the seventh reported, highlights the complexity of lactation induction in transgender women, considering factors such as age, obesity, and insulin resistance. The nutrient profile of the milk suggests its suitability for infant feeding, despite some differences from typical human milk. Conclusions: Induced lactation is feasible in transgender women, expanding the understanding of non-puerperal lactation and its potential in diverse family structures. The present review discusses the development of new metal‐free conditions for the hydroacylation of alkenes and alkynes under both thermal and photochemical conditions from 2018, and their application to the synthesis of various bioactive molecules over the past few years. The key point highlights the remarkable progress made by metal‐free catalytic systems in comparison to the conventional metal catalysts in the hydroacylation of alkenes, alkynes, and arenes. Both inter‐ and intramolecular hydroacylation reactions and their mechanistic pathways were discussed. A novel methodology for the synthesis of 2-pyridones bearing a 2-pyridyl group on nitrogen and carbon atoms, starting from 2-bromopyridines, was developed employing a simple Ru(II)–KOPiv–Na2CO3 catalytic system. Unsubstituted 2-bromopyridine was successfully converted to the penta-heteroarylated 2-pyridone product using this method. Preliminary mechanistic studies revealed a possible synthetic pathway leading to the multi-heteroarylated 2-pyridone products, involving consecutive oxygen incorporation, a Buchwald–Hartwig-type reaction, and C–H bond activation. An efficient method for the synthesis of novel phenylazo-containing moieties is described. The derivatives of 5-(4-(phenyldiazenyl)phenyl)-1,3,4-oxadiazole, substituted at position 2 of the heterocyclic scaffold with alkyl groups of different chain lengths, were prepared. The titled compounds were obtained using the appropriate 4-(5-alkyl-1,3,4-oxadiazol-2-yl)anilines, which were directed to diazotization and subsequently coupled to phenol, resorcinol, and N,N-dimethylaniline. Additionally, we report a mild and effective procedure for the preparation of 4-(5-alkyl-1,3,4-oxadiazol-2-yl)anilines via the selective reduction of the corresponding 2-alkyl-5-(4-nitrophenyl)-1,3,4-oxadiazoles using sodium borohydride-tin(II) chloride dihydrate as the reducing system. Simple Summary: WEE1 negatively regulates the G2/M cell cycle checkpoint, allowing sufficient DNA damage repair before mitosis. Cancer cells often have a defective G1/S checkpoint and rely on the G2/M checkpoint for DNA damage repair. In this study, we investigated the association between WEE1 expression and clinicopathological features of colorectal cancer (CRC) using surgical specimens. Using various CRC cell lines with different mutation statuses, we compared the effects of the WEE1 inhibitor adavosertib on cell proliferation, cell cycle, and apoptosis induction, especially focusing on p53 and KRAS mutations involved in the G1/S checkpoint. We also performed therapeutic experiments with adavosertib in an orthotopic transplanted CRC mouse model using two cell lines with different mutation statuses to evaluate its antitumor effect and impact on the tumor immune microenvironment. Our findings indicate that WEE1 inhibitors can be used for treating CRC, particularly in cases with p53 mutations. Inhibition of WEE1, a key regulator of the G2/M checkpoint of the cell cycle, induces apoptosis by initiating mitosis without repairing DNA damage. However, the effects of WEE1 inhibitors on the tumor immune microenvironment in colorectal cancer (CRC) remain unclear. Here, we investigated the association between WEE1 expression and CRC clinicopathological features using surgically resected CRC specimens and assessed the antitumor effects of a WEE1 inhibitor using CRC cell lines and orthotopic transplantation mouse models. WEE1 expression was not correlated with the clinicopathological features of CRC. The WEE1 inhibitor suppressed cell proliferation in a concentration-dependent manner in all CRC cell lines. It also increased the percentage of cells in the G2/M phase and apoptotic cells, especially in cell lines with p53 mutations, but did not alter these cell percentages in most p53 wild-type cell lines. In the orthotopic mouse model of CRC, tumor volume was significantly reduced in the WEE1 inhibitor-treated group compared to that in the control group. RNA sequencing and immunohistochemistry analyses of mouse tumors revealed that treatment with the WEE1 inhibitor activated tumor immunity and suppressed stromal reactions. Anthranilic acids, salicylaldehydes and arylboronic acids reacted in EtOH/H2O (1/3) at 150 °C under microwave irradiation for 1 h to give, in excellent yields and purity, twenty-three bridgehead bicyclo[4.4.0]boron heterocycles via one-pot, three-component green synthesis. The scope and the limitations of the reactions are discussed in terms of the substitution of ten different anthranilic acids, three salicylaldehydes and three arylboronic acids. The replacement of salicylaldehyde with o-hydroxyacetophenone demanded a lipophilic solvent for the reaction to occur. Eight novel derivatives were isolated following crystallization in a toluene-containing mixture that included molecular sieves. The above one-pot, three-component reactions were completed under microwave irradiation at 180 °C within 1.5 h, thus avoiding the conventional prolonged heating reaction times and the use of a Dean–Stark apparatus. All derivatives were studied for their affinity to calf thymus DNA using proper techniques like viscosity and UV–vis spectroscopy, where DNA-binding constants were found in the range 2.83 × 104–8.41 × 106 M−1. Ethidium bromide replacement studies using fluorescence spectroscopy indicated Stern–Volmer constants between 1.49 × 104 and 5.36 × 104 M−1, whereas the corresponding quenching constants were calculated to be between 6.46 × 1011 and 2.33 × 1012 M−1 s−1. Background: Migraine is among the most prevalent and burdensome neurological disorders in the United States based on disability-adjusted life years. Cortical spreading depolarization (SD) is the most likely electrophysiological cause of migraine aura and may be linked to trigeminal nociception. We previously demonstrated, using a minimally invasive optogenetic approach of SD induction (opto-SD), that opto-SD triggers acute periorbital mechanical allodynia that is reversed by 5HT1B/1D receptor agonists, supporting SD-induced activation of migraine-relevant trigeminal pain pathways in mice. Recent data highlight hypothalamic neural circuits in migraine, and SD may activate hypothalamic neurons. Furthermore, neuroanatomical, electrophysiological, and behavioral data suggest a homeostatic analgesic function of hypothalamic neuropeptide hormone, oxytocin. We, therefore, examined the role of hypothalamic paraventricular nucleus (PVN) and oxytocinergic (OXT) signaling in opto-SD-induced trigeminal pain behavior. Methods: We induced a single opto-SD in adult male and female Thy1-ChR2-YFP transgenic mice and quantified fos immunolabeling in the PVN and supraoptic nucleus (SON) compared with sham controls. Oxytocin expression was also measured in fos-positive neurons in the PVN. Periorbital mechanical allodynia was tested after treatment with selective OXT receptor antagonist L-368,899 (5 to 25 mg/kg i.p.) or vehicle at 1, 2, and 4 h after opto-SD or sham stimulation using von Frey monofilaments. Results: Opto-SD significantly increased the number of fos immunoreactive cells in the PVN and SON as compared to sham stimulation (p < 0.001, p = 0.018, respectively). A subpopulation of fos-positive neurons also stained positive for oxytocin. Opto-SD evoked periorbital mechanical allodynia 1 h after SD (p = 0.001 vs. sham), which recovered quickly within 2 h (p = 0.638). OXT receptor antagonist L-368,899 dose-dependently prolonged SD-induced periorbital allodynia (p < 0.001). L-368,899 did not affect mechanical thresholds in the absence of opto-SD. Conclusions: These data support an SD-induced activation of PVN neurons and a role for endogenous OXT in alleviating acute SD-induced trigeminal allodynia by shortening its duration. Biofilm formation by Staphylococcus aureus is a major challenge in clinical settings due to its role in persistent infections. The AgrA protein, a key regulator in biofilm development, is a promising target for therapeutic intervention. This study investigates the antibiofilm potential of halogenated phenazine compounds by targeting AgrA and explores their molecular interactions to provide insights for drug development. We employed molecular docking, molecular dynamics simulations, and computational mutagenesis to evaluate the binding of halogenated phenazine compounds (C1 to C7, HP, and HP‐14) to AgrA. Binding free energy analysis was performed to assess the affinity of these compounds for the AgrA‐DNA complex. Additionally, the impact of these compounds on AgrA's structural conformation and salt bridge interactions was examined. The binding‐free energy analysis revealed that all compounds enhance binding affinity compared to the Apo form of AgrA, which has a ΔGbind of −80.75 kcal/mol. The strongest binding affinities were observed with compounds C7 (−113.84 kcal/mol), HP‐14 (−115.23 kcal/mol), and HP (−112.28 kcal/mol), highlighting their effectiveness. Molecular dynamics simulations demonstrated that these compounds bind at the hydrophobic cleft of AgrA, disrupting essential salt bridge interactions between His174‐Glu163 and His174‐Glu226. This disruption led to structural conformational changes and reduced DNA binding affinity, aligning with experimental findings on biofilm inhibition. The halogenated phenazine compounds effectively inhibit biofilm formation by targeting AgrA, disrupting its DNA‐binding function. The study supports the potential of these compounds as antibiofilm agents and provides a foundation for rational drug design targeting the AgrA‐DNA interaction. Future research should focus on further optimizing these lead compounds and exploring additional active sites on AgrA to develop novel treatments for biofilm‐associated infections. The chemical transformations of substituted chromones 1a–c were examined toward some nucleophiles namely dimedone (R1), 4-hydroxycoumarin (R2) and 4-hydroxy-1-methylquinolin-2(1H)-one (R3). DFT computation at the B3LYP/6-311 G(d,p) level of theory was used to perform the theoretical computations for the produced compounds. HOMO and LUMO analyses were performed to determine the electronic charge distribution and reactivity of the molecules. Molecular electrostatic potential (MEP) surface analysis was utilized to predict the molecule's reactive sites. Moreover, the studied compounds showed NLO characteristics, where they have first order hyperpolarizability greater than urea. In addition, the GIAO method was used to estimate the 1H-NMR and 13C-NMR chemical shifts; and the findings were compared with experimental values. Testing the generated compounds for antibacterial and anticancer activities revealed varied degrees of inhibitory effect. Graphite carbon nitride (g-C3N4) is a two-dimensional nano-sheet with electronic properties, which shows unique characteristics with high chemical and thermal stability in its structure. The functionalization of these compounds through covalent bonding is an important step towards significantly improving their properties and capabilities. To achieve this goal, a novel strategy for the covalent functionalization of Fe3O4@g-C3N4 with thiamine hydrochloride (vitamin B1) via cyanuric chloride (TCT), which is a divalent covalent linker, was presented. The efficiency of Fe3O4@gC3N4@Thiamine as a heterogeneous organic catalyst in the synthesis of spirooxindole-pyran derivatives and 2-amino-4H-pyran under solvent-free conditions was evaluated and the yields of high-purity products were presented. In addition, easy recycling and reuse for seven consecutive cycles without significant reduction in catalytic activity are other features of this catalyst. Moreover, the performance of the prepared sorbent in the microextraction technique (herein, magnetic solid phase extraction) was studied. The tebuconazole was selected as the target analyte. The target analyte was extracted and determined by HPLC–UV. Under the optimum condition, the linear range of the method (LDR) was estimated in the range of 0.2–100 μg L−1 (the coefficient of determination of 0.9962 for tebuconazole). The detection limit (LOD) of the method for tebuconazole was calculated to be 0.05 µg L−1. The limit of quantification (LOQ) of the method was also estimated to be 0.16 µg L−1. In order to check the precision of the proposed method, the intra-day and inter-day relative standard deviations (RSD%) were calculated, which were in the range of 1.5- 2.8%. In this work, we synthesized 10 new intermediates 4‐chlorobutyl 1‐aryl‐1H‐pyrazole‐4‐carboxylates 3(a–j) from 1‐aryl‐1H‐pyrazole‐4‐carbonyl chlorides, obtained from 1‐aryl‐1H‐pyrazole‐4‐carboxylic acids 4(a–j), and 4‐chlorobutan‐1‐ol 1, generated by THF ring‐opening promoted by hydrochloric acid. Compounds 3(a–j) promoted N‐alkylation of 4(5)‐nitro‐1H‐imidazole to afford the targets 4‐(4‐nitro‐1H‐imidazol‐1‐yl)butyl 1‐aryl‐1H‐pyrazole‐4‐carboxylates 2(a–j) in 18%–85% yields. A catalyst-free one-pot synthetic protocol is presented for the preparation of CF3-containing benzofuran, indolin, and benzothiophene derivatives using readily available aromatic aldehydes and ketones. 2-Bromo-3,3,3-trifluoropropene was employed as a non-corrosive and environmentally benign trifluoromethylacetylene synthon for incorporation of the CF3 group. The reaction proceeds via the formation of a suitably substituted 4,4,4-trifluoro-1-phenylbut-2-yn-1-ol intermediate, which undergoes 5-exo-dig cyclization, resulting in an exocyclic double bond with Z-configuration. Heteropolycyclic molecular systems, which are essential components in the fields of materials and pharmacology, frequently consist of 2D extended organic aromatic rings. Here, we introduce a type of inorganic-organic hybrid 3D conjugates by merging an aromatic boron cluster with a phosphine and a π-conjugated unit. To achieve this, a couple-close synthetic strategy via B–H activation of nido-carboranes with alkynes has been developed, which leads to diverse boron cluster-extended phosphoniums in a twisted structure with high yields under mild conditions. Experimental and theoretical results reveal that the fusion between the boron cluster and the formed borophosphonium heterocycle facilitates electron delocalization throughout the structure. The unusual framework demonstrates distinct properties from bare boron clusters and pure aromatic ring-extended counterparts, such as improved thermal/chemical stability and photophysical properties. Thus, the boron cluster-based 3D conjugates expand the library of aromatic-based heterocyclics, showcasing great potential in functional materials. 2D π-conjugated systems incorporated with heteroatoms can suffer from stability issues and aggregation-induced quenching effects. The functionalization of small aromatic heterocycles is a straight and efficient method to access large polycyclic conjugated molecules. However, due to its unique reactivity, the derivatization of acridone usually gives the product in the angular shape and is difficult to obtain the linear ones. Here we report the direct functionalization of acridone to generate its linearly fused derivatives containing cyclobutadienoid via the palladium‐catalyzed arene‐(oxa)norbornene annulation followed by the dehydrated aromatization. The experimental characterizations demonstrate that the antiaromatic cyclobutadienoid ring can extend the molecular conjugation. Theoretical calculations reveal the weakened aromaticity of neighboring benzenoid rings of cyclobutadienoid with no influence on the electronic structure of central pyridone. Quinazolines are biologically potent heterocyclic compounds; however, their synthesis poses high challenges. In this present study, we have developed an inexpensive cobalt metal‐catalyzed acceptorless dehydrogenative pathway for the sequential synthesis of alkenylated quinazolines from benzhydrol via dehydrogenation, cyclization, and subsequent dehydrogenative sp3 C−H functionalization. This transformation holds potential owing to its commercially available key starting materials, good functional tolerance, moderate reaction conditions, environmental friendliness, and applicability for gram‐scale synthesis. Furthermore, the synthetic utility of the synthesized derivatives has been explored through various reactions, including reduction, halogenation, methoxylation, Michael addition, and spiro cyclization. Additionally, the photophysical properties indicate that the nitrogen atom of the synthesized derivatives readily undergoes a reversible protonation, leading to significant changes in colour. In the present study a library of five (2‐(amino)‐1,3‐thiazin‐6‐one) compounds was synthesized and investigated as against glioblastoma cells in vitro and in vivo in the mice model. The results revealed that all of the five 1,3‐thiazin‐6‐one compounds (4a, 4b, 4c, 4d and 4e) exhibited cytotoxicity against U87MG and 9 L brain cancer cell lines. Moreover, it was found that 9 L cells showed slightly higher sensitivity towards the compounds 4a, 4b, 4c, 4d and 4e compared to U87MG cells. It was observed that the compounds 4a, 4b, 4c, 4d and 4e showed a time‐dependent increase in uptake efficiency by U87MG and 9 L cells. Furthermore, the data revealed that uptake of the compounds and therefore internalization was maximum during initial 1 h of the treatment. Among the synthesized compounds, compound 4c containing trifluoromethyl moiety showed higher uptake efficiency compared to the compound 4a, 4b, 4d and 4e. Treatment of U87MG and 9 L cell tumor spheroids with compound 4c significantly (P<0.05) inhibited the tumor growth compared to the control spheroids. In vivo data revealed that treatment with compound 4c led to a significant (P<0.05) decrease in glioblastoma growth in mice in dose‐dependent manner. Growth of glioblastoma in mice was almost completely inhibited after 28 days of treatment with 2 mg/kg dose of compound 4c. Pharmacokinetic studies showed that compound 4c remained in circulation for longer duration in mice and its terminal half‐life was found to be 6.5 h. Treatment of U87MG and 9 L cells with compound 4c led to a prominent decrease in expression of CYR61 protein. In conclusion, findings of the present study suggest that compound 4c acts as a potential therapeutic agent for the treatment of advanced brain cancer through activation of Hippo pathway. The versatility of aza- and oxa-heterocyclic compounds has garnered significant attention in recent times. A large number of currently approved pharmaceutical products include nitrogen- and oxygen-containing heterocycles. Recent nanotechnological developments have propelled an upsurge in the applications of nanocatalysis for heterocyclic synthesis. Metal nanoparticles (MNPs) have emerged as promising catalysts for the synthesis of aza- and oxa-heterocycles owing to their unique physicochemical properties. Various MNPs including gold, silver, nickel and palladium have been evaluated for their catalytic activities in different reaction types, including cyclisation, coupling, hydrogenation and oxidative transformations. The MNPs have exhibited remarkable catalytic efficiency when utilised under optimal conditions. These catalysts have showcased high reusability and recyclability, yielding satisfactory amounts of the desired heterocyclic compound. The present work provides a detailed overview of recent advances in the area of MNP-assisted synthetic construction of aza- and oxa-heterocycles, published during the previous calendar year, 2022. Background and purpose: Oral candidiasis is one of the most common infections affecting the mucosal surfaces and skin, specifically involving the oral mucosa. This disease is primarily caused by various species of Candida, including Candida albicans. Current treatments with polyene and azole antifungal drugs can affect patient cells, and there are also several limitations in treating fungal infections. These limitations include the shortage and high cost of antifungal drugs, their side effects, and the increasing resistance to azole drugs among Candida species. Given these challenges, researchers are exploring new antifungal drugs, particularly heterocyclic compounds. Heterocyclic compounds are widely used in the pharmaceutical industry. Thiazolidinone derivatives can weaken the fungal and bacterial resistance caused by many antibiotics and drugs. Additionally, oxindoles show significant pharmacological and biological activity. Given these factors and the notable biological properties of thiazolidinones and oxindoles, their potential for developing new antifungal drugs has gained interest among researchers. Therefore, this study aims to investigate the effectiveness of synthetic indole-hydrazono-thiazolidinones on Candida isolates obtained from oral infections. Materials and methods: Antifungal susceptibility testing was conducted on 100 clinical isolates of Candida, including 53 Candida albicans isolates, 22 Candida tropicalis isolates, 17 Candida parapsilosis isolates, and 8 Candida glabrata isolates. The susceptibility of these isolates to 4a and 4b indole-hydrazonothiazolidinone derivatives and nystatin was assessed using the broth microdilution method, based on CLSIM27S4 guidelines for determining drug sensitivity in yeasts. Species identification was performed using phenotypic chrome agar and genetic PCR-RFLP methods. The compounds and antifungal drugs were tested at a dilution range of 0.016–16 μg/ml. The minimum inhibitory concentration (MIC) was defined as the concentration that resulted in at least 50% inhibition of growth compared to the positive control group. Data were analyzed using SPSS version 20 software, with a significance level of P<0.05. Results: The results showed a significant difference in MIC values between the indole-hydrazonothiazolidinone derivatives 4a and 4b, and nystatin against Candida albicans, Candida tropicalis, and Candida parapsilosis. There was also a significant difference in the average MIC values of the derivatives. However, no significant difference was observed in MIC values between these derivatives and nystatin against Candida glabrata. Conclusion: The antifungal activity of the indole-hydrazono-thiazolidinone derivatives was lower than that of nystatin against Candida albicans, Candida tropicalis, and Candida parapsilosis. However, the activity against Candida glabrata was comparable to that of nystatin. Introduction: Hypertensive heart failure (HHF) has a high incidence and poor prognosis. Aim: This article evaluated the efficacy and safety of Vericiguat in HHF and analyzed the relationship between C-reactive protein (CRP) levels and patient prognosis. Methods: 110 HHF patients were divided into Placebo and Vericiguat groups. Cardiac function was assessed by echocardiography and 6-minute walk test (6MWT). Blood samples were collected to detect the levels of N-terminal pro-brain natriuretic peptide (NT-proBNP), cardiac troponin I (cTnI), endothelin (ET-1), nitric oxide (NO), and CRP. Results: Left ventricular end systolic diameter (LVESD) and left ventricular end diastolic dimension (LVEDD) were reduced, the left ventricular ejection fraction (LVEF) and 6MWT were increased, and the serum levels of NT-proBNP, cTnI, ET-1, NO, and CRP were decreased in Vericiguat group as against Placebo group; The total effective rate was 76.4% in Placebo group and 92.7% in Vericiguat group (P < 0.05). The adverse reaction rate was 10.9% and 9.1% (P > 0.05). The proportion of persons with poor prognosis and no improvement of cardiac function in patients with highly expressed CRP before treatment was higher as against patients with low expression of CRP (P < 0.05). A regioselective synthesis of 4H‐pyrano[2,3‐b]quinoline derivatives via DABCO‐mediated Knoevenagel condensation/heterocyclization sequence has been executed. In this way some new fused heterocyclic compounds were synthesized from 2‐chloroquinoline‐3‐carbaldehyde as a versatile and efficient synthetic building block. The one‐pot three‐component reaction of 2‐chloroquinoline‐3‐carbaldehyde and diverse cyclic active methylenes for construction of highly substituted quinolines scaffold has been accomplished under mild condition. In the absence of any head-to-head trials comparing the two products, this article aims to outline the similarities and differences of these two agents. Though moxitafortide has a more favorable pharmacokinetic profile in comparison to plerixafor, in clinical trials, the agents demonstrated similar efficacy. Jurkat T cells expressing Vpr-HiBiT were infected with NL4- 3, and the amount of virus release was quantified indirectly by the amount of Vpr-HiBiT incorporated into the progeny virions. Subsequently, the candidate compounds that suppressed viral release were used to synthesize the heterocyclic compound, HT-7, which reduces HIV-1 release with less cellular toxicity. Notably, HT-7 increased cell surface BST2 coupled with HIV-1 release reduction in Jurkat cells but not Jurkat/KO-BST2 cells. Seemingly, HT-7 impeded simian immunodeficiency virus (SIV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) release. Triazole derivatives of fluorescein-containing N,N-dimethylaminopropyl fragments and their ammonium salts were synthesized with yields of 74–85%. The resulting compounds exhibit fluorescent properties in the green region of the visible spectrum. The critical aggregation concentration (CAC) was estimated using a pyrene fluorescent probe corresponding to a range of 0.28–1.43 mM, and at concentrations above the CAC, the compounds form stable aggregates ranging from 165 to 202 nm. A relative quantum yield of 5–24% has been calculated based on fluorescence and UV spectra. The best value is shown by a derivative containing a tetradecyl substituent. When studying the photocatalytic properties of synthesized compounds through the reaction between N-substituted 1,2,3,4-tetrahydroisoquinoline and malonic ester, the mono-tetradecyl derivative demonstrated the best results. According to gas chromatography–mass spectrometry (GC-MS) data, the conversion of the initial heterocycle reached 95%.
