54. Re-expression of a developmentally restricted potassium channel in autoimmune demyelination: Kv1.4 is implicated in oligodendroglial proliferation

Abstract

Mechanisms of lesion repair in multiple sclerosis are incompletely understood. To some degree, remyelination can occur, associated with an increase of proliferating oligodendroglial cells. Recently, the expression of potassium channels has been implicated in the control of oligodendrocyte precursor cell proliferation in vitro. We investigated the expression of Kv1.4 potassium channels in myelin oligodendrocyte glycoprotein-induced experimental autoimmune encephalomyelitis, a model of multiple sclerosis. Confocal microscopy revealed expression of Kv1.4 in AN2-positive oligodendrocyte precursor cells and premyelinating oligodendrocytes in vitro but neither in mature oligodendrocytes nor in the spinal cords of healthy adult mice. After induction of myelin oligodendrocyte glycoprotein-induced experimental autoimmune encephalomyelitis, Kv1.4 immunoreactivity was detected in or around lesions already during disease onset with a peak early and a subsequent decrease in the late phase of the disease. Kv1.4 expression was confined to 2',3'-cyclic nucleotide 3'-phosphodiesterase-positive oligodendroglial cells, which were actively proliferating and ensheathed naked axons. After a demyelinating episode, the number of Kv1.4 and 2',3'-cyclic nucleotide 3'-phosphodiesterase double-positive cells was greatly reduced in ciliary neurotrophic factor knockout mice, a model with impaired lesion repair. In summary, the re-expression of an oligodendroglial potassium channel may have a functional implication on oligodendroglial cell cycle progression, thus influencing tissue repair in experimental autoimmune encephalomyelitis and multiple sclerosis.

Abstract

Pile-up and dead-time are two main causes of nonlinearity in the response of a PET scanner as a function of activity in the field of view (FOV). For a given scanner and acquisition system, pile-up effects depend on the material and size of the object being imaged and on the distribution of activity inside and outside the FOV, because these factors change the singles-to-coincidences ratio (SCR). Thus, it is difficult to devise an accurate correction that would be valid for any acquisition. In this work, we demonstrate a linear relationship between SCR and effective dead-time, which measures the effects of both dead-time (losses) and pile-up (gains and losses). This relationship allows us to propose a simple method to accurately estimate dead-time and pile-up corrections using only two calibration acquisitions with, respectively, a high and low SCR. The method has been tested with simulations and experimental data for two different scanner geometries: a scanner with large area detectors and no pile-up rejection, and a scanner composed of two full rings of smaller detectors. Our results show that the SCR correction method is accurate within 7%, even for high activities in the FOV, and avoids the bias of the standard single-parameter method. © 2013 Institute of Physics and Engineering in Medicine.

Abstract

The aim of this study is to investigate the acid-base balance when performing a maximal field test of professional soccer players. During the practice of physical exercise in a wide range of intensity, the pH is maintained close to resting values. At higher intensities the pH drops in inverse proportion. The different buffer systems of the body are responsible for these variations. The variables that determine the pH can be divided into independent and dependent, being the study of them essential to understanding the changes in acidbase balance. In this study we analyzed three variables that determine the changes in pH: the partial pressure of CO2 (pPCO2), the concentration of bicarbonate (HCO3 -) and lactate concentration. The determinations were performed on capillary blood in nineteen players while performing an incremental exercise., El objetivo de este estudio es investigar el equilibrio ácido-base al realizar un test de campo máximo en futbolistas profesionales. Durante la práctica de un ejercicio físico, en un amplio rango de intensidad, el pH se mantiene muy próximo a los valores de reposo. A intensidades más elevadas el pH desciende en relación inversa. Los diferentes sistemas de amortiguación del organismo son los responsables de estas variaciones. Las variables que determinan el pH pueden ser divididas en independientes y dependientes, siendo el estudio de las mismas esencial para comprender las variaciones en el equilibrio ácidobase. En el presente trabajo se analizaron tres de las variables que determinan las modificaciones en el pH: la presión parcial de CO2 (pPCO2), la concentración de bicarbonato (HCO3 -) y la concentración de lactato. Las determinaciones fueron realizadas en sangre capilar en diecinueve jugadores durante la realización de un ejercicio incremental.

Abstract

Effective medicines exist to treat or alleviate many diseases which predominate in the developing world and cause high mortality and morbidity rates. Price should not be an obstacle preventing access to these medicines. Increasingly, drug donations have been established by drug companies, but these are often limited in time, place or use. Measures exist which are more sustainable and will have a greater positive impact on people's health. Principally, these are encouraging generic competition; adopting into national legislation and implementing TRIPS safeguards to gain access to cheaper sources of drugs; differential pricing; creating high volume or high demand through global and regional procurement; and supporting the production of quality generic drugs by developing countries through voluntary licenses if needed, and facilitating technology transfer.
