Abstract

Objective: To investigate the genetic architecture of internalizing symptoms in childhood and adolescence. Method: In 22 cohorts, multiple univariate genome-wide association studies (GWASs) were performed using repeated assessments of internalizing symptoms, in a total of 64,561 children and adolescents between 3 and 18 years of age. Results were aggregated in meta-analyses that accounted for sample overlap, first using all available data, and then using subsets of measurements grouped by rater, age, and instrument. Results: The meta-analysis of overall internalizing symptoms (INToverall) detected no genome-wide significant hits and showed low single nucleotide polymorphism (SNP) heritability (1.66%, 95% CI = 0.84–2.48%, neffective = 132,260). Stratified analyses indicated rater-based heterogeneity in genetic effects, with self-reported internalizing symptoms showing the highest heritability (5.63%, 95% CI = 3.08%–8.18%). The contribution of additive genetic effects on internalizing symptoms appeared to be stable over age, with overlapping estimates of SNP heritability from early childhood to adolescence. Genetic correlations were observed with adult anxiety, depression, and the well-being spectrum (|rg| < 0.70), as well as with insomnia, loneliness, attention-deficit/hyperactivity disorder, autism, and childhood aggression (range |rg| = 0.42–0.60), whereas there were no robust associations with schizophrenia, bipolar disorder, obsessive-compulsive disorder, or anorexia nervosa. Conclusion: Genetic correlations indicate that childhood and adolescent internalizing symptoms share substantial genetic vulnerabilities with adult internalizing disorders and other childhood psychiatric traits, which could partially explain both the persistence of internalizing symptoms over time and the high comorbidity among childhood psychiatric traits. Reducing phenotypic heterogeneity in childhood samples will be key in paving the way to future GWAS success.

Abstract

BackgroundThe interplay of genetic factors and environmental adversities in ADHD can be reflected by cell type-specific DNA methylation (DNAm). Previous methylome-wide association studies (MWAS) for ADHD have been performed in bulk tissue only, where meaningful DNAm signal could be left undetected. We aim to identify cell type-specific DNAm profiles associated with childhood ADHD symptoms.MethodsFirst, we performed array-based (<850.000 sites) cell type-specific MWAS meta-analyses of childhood ADHD symptoms (age-range 4-13 years) in cord- (N=1543) and peripheral blood (N=2009) from the PACE consortium. Then we expanded the coverage of the epigenome to 28 million sites based on methyl-binding-domain sequencing in peripheral blood (N=583; Great Smoky Mountain Study; mean-age 13.5 years). Using epigenomic deconvolution, DNAm associations in the five major blood cells were calculated.ResultsAssociations between DNAm and ADHD symptoms were identified in cord blood monocytes (PDE6B, FDR=0.012), CD8T cells (KCNA3, HAND2, both FDR=0.016), and NK cells (KIFC1, FDR=0.043). In peripheral blood, only CD8T cells (RPL31P11, FDR=4.48*10-6 and KCNJ5, FDR=0.0021) showed associations. In the broader coverage analysis, we detected 24 and 16 significant CpG sites (FDR<0.05) in monocytes and granulocytes, respectively. None of these hits were covered by the arrays, and some overlapping genes showed opposite effect sizes in the different cell types, which signal was masked in bulk tissue. Downstream analyses of gene ontology term enrichment showed consistent results between cord and peripheral blood.ConclusionsWe identified the first ADHD-DNAm associations at a cell type-specific level, bringing us closer to understanding the role of specific cell types in the etiology of ADHD.

Abstract

Background: Dietary habits were investigated as environmental risk factors for Attention-Deficit/Hyperactivity Disorder (ADHD). However, no previous studies explored the effects of dietary factors on modifying the role of genetic factors on ADHD. Methods: Based on a Swedish population-based twin study with 1518 twin pairs aged 20–47 years, we tested whether the importance of genetic and environmental effects on ADHD varied as a function of dietary habits. Self-reported dietary habits and ADHD symptoms were collected. Twin methods were used to test the degree to which high-sugar and unhealthy food intake moderated the genetic and environmental influences on ADHD symptoms. Results: In middle-aged adults, genetic influences on inattention symptoms were statistically significantly higher among individuals with higher levels of high-sugar (45%, 95%CI: 25–54%) and unhealthy food intake (51%, 95%CI: 31–60%), compared with those with lower levels of consumption of high-sugar (36%, 95%CI: 25–47%) and unhealthy foods (30%, 95%CI: 20–41%). Similar patterns were also found for the associations between hyperactivity/impulsivity and high-sugar/unhealthy food intake, even though the moderation effects were not statistically significant. Conclusion The present study suggests that genetic factors play a more prominent role in individual differences of ADHD symptoms in the presence of the high consumption of sugar and unhealthy foods. Future longitudinal studies with multiple assessments of ADHD and dietary habits are needed to replicate our findings.

Abstract

Objective: Advanced parenthood increases the risk of severe neurodevelopmental disorders like autism, Down syndrome and schizophrenia. Does advanced parenthood also negatively impact offspring's general neurodevelopment?Method: We analyzed child-, father-, mother- and teacher-rated attention-problems (N = 38,024), and standardized measures of intelligence (N = 10,273) and educational achievement (N = 17,522) of children from four Dutch population-based cohorts. The mean age over cohorts varied from 9.73-13.03. Most participants were of Dutch origin, ranging from 58.7%-96.7% over cohorts. We analyzed 50% of the data to generate hypotheses and the other 50% to evaluate support for these hypotheses. We aggregated the results over cohorts with Bayesian research synthesis.Results: We mostly found negative linear relations between parental age and attention-problems, meaning that offspring of younger parents tended to have more attention problems. Maternal age was positively and linearly related to offspring's IQ and educational achievement. Paternal age showed an attenuating positive relation with educational achievement and an inverted U-shape relation with IQ, with offspring of younger and older fathers at a disadvantage. Only the associations with maternal age remained after including SES. The inclusion of child gender in the model did not affect the relation between parental age and the study outcomes.Conclusions: Effects were small but significant, with better outcomes for children born to older parents. Older parents tended to be of higher SES. Indeed, the positive relation between parental age and offspring neurodevelopmental outcomes was partly confounded by SES.

The genetic burden for ADHD and ASD on previously identified emotion recognition factors (speed and accuracy of visual and auditory emotion recognition) and classes (Class 1: Average visual, impulsive auditory; Class 2: Average-strong visual & auditory; Class 3: Impulsive & imprecise visual, average auditory; Class 4: Weak visual & auditory) was assessed using ASD and ADHD polygenic risk scores (PRS). Our sample contained 552 participants: 74 with ADHD, 85 with ASD, 60 with ASD + ADHD, 177 unaffected siblings of ADHD or ASD probands, and 156 controls. ADHD- and ASD-PRS, calculated from the latest ADHD and ASD GWAS meta-analyses, were analyzed across these emotion recognition factors and classes using linear mixed models. Unexpectedly, the analysis of emotion recognition factors showed higher ASD-PRS to be associated with faster visual emotion recognition. The categorical analysis of emotion recognition classes showed ASD-PRS to be reduced in Class 3 compared to the other classes (p value threshold [pT] = 1, p = .021). A dimensional analysis identified a high ADHD-PRS reduced the probability of being assigned to the Class 1 or Class 3 (pT = .05, p = .028 and p = .044, respectively). Though these nominally significant results did not pass FDR correction, they potentially indicate different indirect causative chains from genetics via emotion recognition to ADHD and ASD, which need to be verified in future research.

Abstract

Glycemic traits are used to diagnose and monitor type 2 diabetes and cardiometabolic health. To date, most genetic studies of glycemic traits have focused on individuals of European ancestry. Here we aggregated genome-wide association studies comprising up to 281,416 individuals without diabetes (30% non-European ancestry) for whom fasting glucose, 2-h glucose after an oral glucose challenge, glycated hemoglobin and fasting insulin data were available. Trans-ancestry and single-ancestry meta-analyses identified 242 loci (99 novel; P < 5 x 10-8), 80% of which had no significant evidence of between-ancestry heterogeneity. Analyses restricted to individuals of European ancestry with equivalent sample size would have led to 24 fewer new loci. Compared with single-ancestry analyses, equivalent-sized trans-ancestry fine-mapping reduced the number of estimated variants in 99% credible sets by a median of 37.5%. Genomic-feature, gene-expression and gene-set analyses revealed distinct biological signatures for each trait, highlighting different underlying biological pathways. Our results increase our understanding of diabetes pathophysiology by using trans-ancestry studies for improved power and resolution.

75. Reproductive success of Long-billed Curlews (Numenius americanus) in Northeastern Nevada hay fields

Abstract

Because of the continuing loss of native grasslands, many shorebirds that breed in the temperate zone have become increasingly dependent on non-native grasslands associated with farming for breeding habitat. But invasive land-management in non-native grasslands has placed ground-nesting shorebirds at considerable risk. We studied the reproductive success of Long-billed Curlews (Numenius americanus) breeding in northeastern Nevada, in a landscape dominated by hay fields, between 2003 and 2006. Observed nest density was as high as 9 nests per 100 ha, and mean nest success was 31%, with considerable interannual variation. Predation, predominantly by large mammalian predators such as Coyotes (Canis latrans), was the greatest cause of nest failure in Long-billed Curlews. Flooding of nests during irrigation of hay fields and trampling and disturbance by cattle also affected nest success, albeit to a lesser degree. Fledging success of radiomarked chicks averaged 47%, and hay fields were the preferred brood-rearing habitat, especially in dry years. Mammalian predation was the greatest cause of chick mortality, whereas ranching activities had no noticeable effect on chick survival. In all years, Long-billed Curlews exhibited a high propensity for renesting after a first nest failure. After taking renesting into account, per-female nest success averaged 45% and mean annual productivity was 0.70 female chicks hatched and 0.33 female chicks fledged per breeding female. These results suggest that despite potential negatives associated with high land-management intensity, hay fields in northern Nevada can be of great value in conservation of the Long-billed Curlew. Abstract

AbstractAimsType 2 diabetes (T2D) is a global health burden, more prevalent among individuals with attention deficit hyperactivity disorder (ADHD) compared to the general population. To extend the knowledge base on how ADHD links to T2D, this study aimed to estimate causal effects of ADHD on T2D and to explore mediating pathways.MethodsWe applied a two-step, two-sample Mendelian randomization (MR) design, using single nucleotide polymorphisms to genetically predict ADHD and a range of potential mediators. First, a wide range of univariable MR methods was used to investigate associations between genetically predicted ADHD and T2D, and between ADHD and the purported mediators: body mass index (BMI), childhood obesity, childhood BMI, sedentary behaviour (daily hours of TV watching), blood pressure (systolic blood pressure, diastolic blood pressure), C-reactive protein and educational attainment (EA). A mixture-of-experts method was then applied to select the MR method most likely to return a reliable estimate. We used estimates derived from multivariable MR to estimate indirect effects of ADHD on T2D through mediators.ResultsGenetically predicted ADHD liability associated with 10% higher odds of T2D (OR: 1.10; 95% CI: 1.02, 1.18). From nine purported mediators studied, three showed significant individual mediation effects: EA (39.44% mediation; 95% CI: 29.00%, 49.73%), BMI (44.23% mediation; 95% CI: 34.34%, 52.03%) and TV watching (44.10% mediation; 95% CI: 30.76%, 57.80%). The combination of BMI and EA explained the largest mediating effect (53.31%, 95% CI: −1.99%, 110.38%) of the ADHD–T2D association.ConclusionsThese findings suggest a potentially causal, positive relationship between ADHD liability and T2D, with mediation through higher BMI, more TV watching and lower EA. In de jaren dertig van de vorige eeuw hebben de houtvesters Jansen en Blokhuis, geïnspireerd door Duits voorbeeld, gemengde bosopstanden aangelegd in Drenthe. Een veranderend klimaat stimuleert beheerders tegenwoordig ook om weer te denken aan het aanleggen van gemengde bossen. Meer mengen betekent vaak ook het introduceren van ‘nieuwe soorten’. Maar wat zou je dan introduceren? De gewone zilverspar (Abies alba) is zo’n voor Nederland ‘nieuwe soort’ met potentie. Dit concluderen Chris Hartman en Stefan Poelman in hun afstudeeronderzoek aan de hogeschool Van Hall Larenstein. Zij verkenden de rol die de gewone zilverspar kan gaan spelen in de bossen van de toekomst. Dankzij Jansen en Blokhuis hebben we in Drenthe gemengde bossen waarin de gewone zilverspar al bijna 100 jaar staat. De ervaringen die daar zijn opgedaan geven een doorkijkje naar de toekomst.

Abstract

Purpose This paper will the describe the overall project goals, activities, preliminary findings, and future work on this project. The purpose of this project is to develop a model of Disciplinary Literacy Instruction (DLI) in engineering that can be used in both K-12 and undergraduate engineering settings. This model of DLI will be informed by knowledge about the ways practicing engineers across four disciplines of engineering (i.e., electrical/computer, mechanical/aerospace, civil/environmental, and chemical/biological) read, interpret, evaluate, and generate texts in the context of their work environment. This information will be translated into a model of DLI in engineering to teach students how to use authentic engineering literacy practices as they learn discipline specific engineering content. Project Activities During the first year of this project, we conducted on-site observations with two electrical engineers and two mechanical engineers. In addition, we held interviews and conducted think-aloud protocols that were informed by the observations with each engineer. From these data sources, we developed a codebook describing the types of texts that the engineers interpreted, evaluated, and generated at the workplace. worked with both mechanical and electrical engineering consultants to help refine and revise the codes and code definitions to enhance their authenticity to each discipline. To further ensure the quality of our data analysis procedures, we sought feedback on our codes from three advisory board consultants having expertise in disciplinary literacy, engineering, and K-12 engineering education. During the second year of this project, we analyzed the interview and think-aloud protocol transcripts from the electrical and mechanical engineers to generate themes that described the interpretive and evaluative frameworks the engineers used as they solved a technical problem or generated a solution for a client or customer. Similarly, we developed themes that described the socially situated activities in which the previously defined genres were embedded. Taken together, these frameworks and activities inform the development of the DLI model in engineering. We also began collecting observation, interview, and think aloud data with one civil and one environmental engineer during this year. Currently, in the third year of this project, we are analyzing the observation field notes and the interview and think-aloud protocol transcripts from the civil and environmental engineers. Simultaneously, we are generating data with the final pair of engineers: one biological and one chemical engineer. We continue to refine our codebook by adding new genres as they appear and merging any similar, existing genres to capture the range of texts with which the engineers engaged. Engineering consultants from both the civil and environmental disciplines will provide feedback on our codes. Combined data from this phase with previous phases will be used develop disciplinary specific curricular materials for K-12 and undergraduate engineering education. Future Activities The data collected and analyzed throughout the project will inform the development of a model for DLI in engineering that can be used by teachers in both undergraduate and K-12 educational settings. This model will provide a framework for teachers to instruct students on how to use the authentic reading and writing strategies that practicing engineers use while solving problems. By providing a diverse set of students with exposure to these literacy practices in school at a young age, a model of DLI in engineering has the potential to remove literacy-based barriers that may deter students from pursuing engineering pathways.

Abstract

Although a genetic basis of depression has been well established in twin studies, identification of genome-wide significant loci has been difficult. We hypothesized that bivariate analyses of findings from a meta-analysis of genome-wide association studies (meta-GWASs) of the broad depression phenotype with those from meta-GWASs of self-reported and recurrent major depressive disorder (MDD), bipolar disorder and schizophrenia would enhance statistical power to identify novel genetic loci for depression. LD score regression analyses were first used to estimate the genetic correlations of broad depression with self-reported MDD, recurrent MDD, bipolar disorder and schizophrenia. Abstract

Objective: Advanced parenthood increases the risk of severe neurodevelopmental disorders like autism, Down syndrome and schizophrenia. Does advanced parenthood also negatively impact offspring’s general neurodevelopment? Method: We analyzed child-, father-, mother- and teacher-rated attention-problems (N = 38,024), and standardized measures of intelligence (N = 10,273) and educational achievement (N = 17,522) of children from four Dutch population-based cohorts. The mean age over cohorts varied from 9.73–13.03. Most participants were of Dutch origin, ranging from 58.7%-96.7% over cohorts. Abstract

Subcortical brain structures are integral to motion, consciousness, emotions and learning. We identified common genetic variation related to the volumes of the nucleus accumbens, amygdala, brainstem, caudate nucleus, globus pallidus, putamen and thalamus, using genome-wide association analyses in almost 40,000 individuals from CHARGE, ENIGMA and UK Biobank. We show that variability in subcortical volumes is heritable, and identify 48 significantly associated loci (40 novel at the time of analysis). Annotation of these loci by utilizing gene expression, methylation and neuropathological data identified 199 genes putatively implicated in neurodevelopment, synaptic signaling, axonal transport, apoptosis, inflammation/infection and susceptibility to neurological disorders. This set of genes is significantly enriched for Drosophila orthologs associated with neurodevelopmental phenotypes, suggesting evolutionarily conserved mechanisms. Our findings uncover novel biology and potential drug targets underlying brain development and disease.
