Abstract

Introduction: The impact of water quality on the survival of human norovirus (NoV) was determined in irrigation water field run-off (tail water) and well water from a representative Central Coast vegetable production site in the Salinas Valley, California. Methods: Tail water, well water, and ultrapure water samples were inoculated separately with two surrogate viruses for human NoV--Tulane virus (TV) and murine norovirus (MNV)--to achieve a titer of 1×105 plaque forming units (PFU)/ml. Samples were stored at 11, 19, and 24°C for 28 days. Additionally, inoculated water was applied to soil collected from a vegetable production site in the Salinas Valley or to the surface of growing romaine lettuce leaves, and virus infectivity was evaluated for 28 days in a growth chamber. Results: Virus survivalwas similar for water stored at 11, 19, and 24°C and there was no difference in infectivity based on water quality. After 28 days, a maximum1.5 log reduction was observed for both TV and MNV. TV decreased by 1.97-2.26 log and MNV decreased by 1.28-1.48 logs after 28 days in soil; infectivitywas not influenced bywater type. Infectious TV and MNV were recovered from lettuce surfaces for upto 7 and 10 days after inoculation, respectively. Across the experiments there was no significant impact of water quality on the stability of the human NoV surrogates. Discussion: Overall, the human NoV surrogates were highly stable in water with a less than 1.5 log reduction over 28 days and no difference observed based on the water quality. In soil, the titer of TV declined by approximately 2 logs over 28 days, while MNV declined by 1 log during the same time interval, suggesting surrogatespecific inactivation dynamics in the soil tested in this study. A 5-log reduction in MNV (day 10 post inoculation) and TV (day 14 post inoculation) was observed on lettuce leaves, and the inactivation kinetics were not significantly impacted by the quality of water used. These results suggest that human NoV would be highly stable in water, and the quality of the water (e.g., nutrient content, salinity, and turbidity) does not significantly impact viral infectivity. 60. Genetic variability of kernel phenolics in maize (Zea maysL.) inbreds with differing levels of resistance to gibberella ear rot

Abstract

A comprehensive study was performed to investigate the kernel phenolics content and profile of a broad range of maize inbreds varying in disease resistance to gibberella ear rot. Three phenolic fractions (soluble free, soluble conjugated, and insoluble bound) at different kernel developmental stages in two growing seasons were analyzed. The phenolics content and profile revealed a genotype dependency. The highest amount and diversity of phenolics were at day 11 after self‐pollination in both growing seasons. This was confirmed by a distinctive PCA biplot clustering of phenolics at day 11 compared to other harvesting days for all fractions. A strong negative correlation was found between the conjugated and bound phenolics in both growing seasons (in 2016, r= 0.93, p< 0.0001; and in 2018, r= 0.94, p< 0.0001). In both growing seasons, the main phenolic compounds in the free, conjugated, and bound fractions were chlorogenic acid, caffeic acid, and t‐ferulic acid, respectively. Comprehensive results were presented for how the phenolics evolved over the period of kernel development and how their content changed. The PCA‐biplot analysis revealed different patterns of clustering for phenolic associations in different fractions as well as in different growing seasons, reflecting the evolution process of individual phenolics during kernel filling period. Overall, genotypes with higher disease resistance possessed higher phenolics in the bound and conjugated fractions at blister stage. This study provides a useful resource of maize inbred phenolics profiles that could be applied in future breeding efforts. The phenolics distribution in free, conjugated, and bound fractions during maize kernel development was documented.The evolution of phenolic compounds during maize kernel development was revealed.Most changes in the phenolics profile occurred during the early stages of kernel development with high heritability.Genotypes with higher disease resistance possessed higher bound and conjugated phenolics at blister stage.

Abstract

Food manufacturers are required to obtain scientific and technical evidence that a control measure or combination of control measures is capable of reducing a significant hazard to an acceptable level that does not pose a public health risk under normal conditions of distribution and storage. A validation study provides evidence that a control measure is capable of controlling the identified hazard under a worst-case scenario for process and product parameters tested. It also defines the critical parameters that must be controlled, monitored, and verified during processing. This review document is intended as guidance for the food industry to support appropriate validation studies, and aims to limit methodological discrepancies in validation studies that can occur among food safety professionals, consultants, and third-party laboratories. The document describes product and process factors that are essential when designing a validation study, and gives selection criteria for identifying an appropriate target pathogen or surrogate organism for a food product and process validation. Guidance is provided for approaches to evaluate available microbiological data for the target pathogen or surrogate organism in the product type of interest that can serve as part of the weight of evidence to support a validation study. The document intends to help food manufacturers, processors, and food safety professionals to better understand, plan, and perform validation studies by offering an overview of the choices and key technical elements of a validation plan, the necessary preparations including assembling the validation team and establishing prerequisite programs, and the elements of a validation report.

Abstract

The Salmonella Syst-OMICS consortium is sequencing 4,500 Salmonella genomes and building an analysis pipeline for the study of Salmonella genome evolution, antibiotic resistance and virulence genes. Metadata, including phenotypic as well as genomic data, for isolates of the collection are provided through the Salmonella Foodborne Syst-OMICS database (SalFoS), at https://salfos.ibis.ulaval.ca/. Here, we present our strategy and the analysis of the first 3,377 genomes. Our data will be used to draw potential links between strains found in fresh produce, humans, animals and the environment. The ultimate goals are to understand how Salmonella evolves over time, improve the accuracy of diagnostic methods, develop control methods in the field, and identify prognostic markers for evidence-based decisions in epidemiology and surveillance.
