Kindergarten teachers' ability to master various information technologies for learning today is required absolutely. One of the important skills is to package teaching materials in the form of simple games, because kindergarten-age children really like games. Power Point is an application from Microsoft that can be applied to design teaching materials in the form of simple games. Training to improve the ability of kindergarten teachers in this application area has been held. This training was held with participants from the Association of Indonesian Kindergarten Organizing Organizations (GOPTKI) in Yogyakarta City. The target of this activity is to improve the ability of kindergarten teachers to package interesting teaching materials using the Power Point application. The method used is the lecture method, a simple game-making demo and game-making practice. The evaluation was carried out using a post-training questionnaire. The results of the evaluation show that this activity is very useful for teachers in preparing interesting teaching materials. Participants 86.4% were satisfied and 89% answered that the training was in accordance with the needs of the community., Kemampuan Guru TK untuk menguasai berbagai teknologi informasi bagi pembelajaran saat ini sangat diperlukan. Salah satu keterampilan yang penting adalah mengemas bahan ajar dalam bentuk game sederhana, karena anak-anak usia TK tentulah sangat menyukai game. Power Point adalah aplikasi dari Microsoft yang dapat digunakan untuk merancang bahan ajar dalam bentuk game sederhana. Pelatihan ini diselenggarakan dengan peserta dari Gabungan Organisasi Penyelenggara Taman Kanak-kanak Indonesia (GOPTKI) di Kota Yogyakarta. Sasaran dari kegiatan ini adalah meningkatkan kemampuan Guru TK untuk mengemas bahan ajar yang menarik menggunakan aplikasi Power Point. Metode yang digunakan adalah metode ceramah, demo pembuatan game sederhana dan praktek pembuatan game. Evaluasi dilakukan dengan kuesioner pasca pelatihan. Hasil evaluasi menunjukkan bahwa kegiatan ini sangat berguna bagi para guru dalam menyiapkan bahan ajar yang menarik. Peserta 86,4% merasa puas dan 89% menjawab bahwa pelatihan sesuai dengan kebutuhan masyarakat.

Breast cancer is the leading cause of cancer-related deaths in women. The aggressive breast cancer subtype is commonly linked to the genetic alterations in the TP53 tumor suppressor gene, predominantly the missense mutations. Robust experimental models are needed to gain better insights into these mutations’ molecular properties and implications in tumorigenesis. The generation of such models harboring the alterations is feasible with the CRISPR-based gene editing technology. Moreover, the development of new CRISPR applications, particularly DNA base and prime editing, has considerably improved the precision and versatility of gene editing. Here, we employed the prime editing tool to revert a TP53 missense C > T mutation (L194F) in a T47D luminal A breast cancer cell line. In parallel, this prime editing tool was also utilized to introduce the L194F mutation in HEK293T cells. To assess the prime editing efficiency in both cell lines, we first performed Sanger sequencing in the prime-edited cells pool and single cell-derived clones. However, the Sanger sequencing approach did not detect any base substitution in these cell lines. Next, by employing the more sensitive amplicon target sequencing, we managed to identify the expected substitution in these T47D and HEK293T cells, albeit the editing efficiency was low. In light of these findings, we discussed the technical aspects and provided suggestions for improve the prime editing workflow and efficiency for future experiments.

This study aims to determine the effect of the Guided Note Taking strategy with the aid of video media on student learning outcomes in grade IV SDN 5 Rambang subjects. The method used in this research is a quasi-experimental method in the form of One Group Fretest-posttest with a total sample of 44 students. The analytical technique used in this study is the normality test, homogeneity test, and t test using the SPSS 22 for windows software program. the posttest mean is 90.00 and the standard deviation is 6.43. Meanwhile, for the control class, the average fretest score was 67.6 and the standard deviation was 7.49, the posttest average was 83.5 and the standard deviation was 5.69. video has a significant effect on the learning outcomes of fourth grade students at SDN 5 Rambang., Penelitian ini bertujuan untuk mengetahui pengaruh strategi Guided Note Taking berbantu media video terhadap hasil belajar siswa pada Materi tematk kelas IV SDN 5 Rambang. Metode yang digunakan dalam penelitian ini adalah metode eksperimen semu bentuk One Group Fretest-posttest dengan jumlah sampel yang diteliti berjumlah 44 siswa. Teknik analisis yang digunakan dalam penelitian ini adalah uji normalitas, uji homogenitas, dan uji t dengan menggunakan program Sofware spss 22 for windows.Hasil penelitian menunjukan perolehan nilai rata-rata pretest kelas eksperimen sebesar 67,6 dan standar deviasinya 8,72, nilai rata-rata posttest 90,00 dan standar deviasinya 6,43. Sedangkan untuk kelas kontrol nilai rata-rata fretest sebesar 67,6 dan standar deviasinya 7,49, nilai rata-rata posttest 83,5 dan standar deviasinya 5,69.Berdasarkan hasil penelitian diatas, dapat disimpulkan bahwa penerapan strategi Guided Note Taking berbantu media vidio berpengaruh signifikan terhadap hasil belajar siswa kelas IV SDN 5 Rambang.
