Abstract

We briefly review the current status of anthropological and genetic studies of isolated populations and of their micro-evolutionary and biomedical applications, with particular emphasis on European populations. Thereafter, we describe the ongoing collaborative research project "Isolating the Isolates: geographic and cultural factors of human genetic variation" regarding Italian extant geographical and/or linguistic isolates, aimed at overcoming the limitations of previous studies regarding geographical coverage of isolates, number and type of genetic polymorphisms under study and suitability of the experimental design to investigate gene-culture coevolutionary processes. An interdisciplinary sampling approach will make it possible to collect several linguistic isolates and their geographic neighbours from Trentino, Veneto, Friuli, Tuscany, Sardinia and Calabria. This will be coupled with a shared genotyping strategy based on mitochondrial and Y-chromosomal polymorphisms. The results will be analyzed with a focus on the role of geographical and cultural factors in shaping human biodiversity. The aims of the project go beyond the simple reconstruction of the genetic structure and history of the examined groups. In fact, the study will also include an assessment for future bio-medical studies and the development of genetic and bio-demographic databases. Ethical and educational aspects are also foreseen by the project, by using informed consents together with disseminating activities in loco, completed by the creation of a dedicated web site for both scientific and public audiences.

Contains fulltext : 153534.pdf (Publisher’s version ) (Open Access) INTRODUCTION: Adults with end-stage kidney disease (ESKD) treated with haemodialysis experience mortality of between 15% and 20% each year. Effective interventions that improve health outcomes for long-term dialysis patients remain unproven. Novel and testable determinants of health in dialysis are needed. Nutrition and dietary patterns are potential factors influencing health in other health settings that warrant exploration in multinational studies in men and women treated with dialysis. We report the protocol of the "DIETary intake, death and hospitalisation in adults with end-stage kidney disease treated with HaemoDialysis (DIET-HD) study," a multinational prospective cohort study. DIET-HD will describe associations of nutrition and dietary patterns with major health outcomes for adults treated with dialysis in several countries. METHODS AND ANALYSIS: DIET-HD will recruit approximately 10,000 adults who have ESKD treated by clinics administered by a single dialysis provider in Argentina, France, Germany, Hungary, Italy, Poland, Portugal, Romania, Spain, Sweden and Turkey. Recruitment will take place between March 2014 and June 2015. The study has currently recruited 8000 participants who have completed baseline data. Nutritional intake and dietary patterns will be measured using the Global Allergy and Asthma European Network (GA(2)LEN) food frequency questionnaire. The primary dietary exposures will be n-3 and n-6 polyunsaturated fatty acid consumption. The primary outcome will be cardiovascular mortality and secondary outcomes will be all-cause mortality, infection-related mortality and hospitalisation. ETHICS AND DISSEMINATION: The study is approved by the relevant Ethics Committees in participating countries. All participants will provide written informed consent and be free to withdraw their data at any time. The findings of the study will be disseminated through peer-reviewed journals, conference presentations and to participants via regular newsletters. We expect that the DIET-HD study will inform large pragmatic trials of nutrition or dietary interventions in the setting of advanced kidney disease.

Abstract

PurposeBreast cancer follow-up procedures after primary treatment are still a controversial issue. Aim of this study was to investigate, through a web-based survey, surveillance methodologies selected by Italian oncologists in everyday clinical practice.MethodsReferents of Italian medical oncology units were invited to participate to the study via e-mail through the SurveyMonkey website. Participants were asked how, in their institution, exams of disease staging and follow-up are planned in asymptomatic women and if surveillance continues beyond the 5th year.ResultsBetween February and May 2013, 125 out of 233 (53.6%) invited referents of Italian medical oncology units agreed to participate in the survey. Ninety-seven (77.6%) referents state that modalities of breast cancer follow-up are planned according to the risk of disease progression at diagnosis and only 12 (9.6%) oncology units apply the minimal follow-up procedures according to international guidelines. Minimal follow-up is never applied in high risk asymptomatic women. Ninety-eight (78.4%) oncology units continue follow-up in all patients beyond 5 years.ConclusionsOur survey shows that 90.4% of participating Italian oncology units declare they do not apply the minimal breast cancer follow-up procedures after primary treatment in asymptomatic women, as suggested by national and international guidelines. Interestingly, about 80.0% of interviewed referents performs the so called "tailored follow-up", high intensity for high risk, low intensity for low risk patients. There is an urgent need of randomized clinical trials able to determine the effectiveness of risk-based follow-up modalities, their ideal frequency and persistence in time.

Abstract

We retrospectively assessed tolerability and efficacy of paclitaxel plus gemcitabine combination in 259 patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) enrolled in three randomized SICOG trials according to their age (70 years) at study entry. Apart from age, demographic and clinical characteristics were similar in the two groups. Response rate of paclitaxel plus gemcitabine was similar in younger and in elderly (36% versus 30%). Chemotherapy was well tolerated, but severe neutropenia (12% versus 7%), anaemia (6.6% versus 1.8%), and vomiting (5% versus 0) were more frequent in elderly patients. Both median progression-free survival (PFS, 5.5 months versus 4.2 months), and overall survival (OS, 11.1 months versus 9.1 months) resulted slightly prolonged for younger patients. However, only stage and performance status resulted independently affecting PFS and OS. In conclusion, paclitaxel plus gemcitabine were similarly tolerated and active in younger and elderly patients. This regimen should be considered an option for the management of fit elderly patients.

Abstract

We briefly review the current status of anthropological and genetic studies of isolated populations and of their micro-evolutionary and biomedical applications, with particular emphasis on European populations. Thereafter, we describe the ongoing collaborative research project "Isolating the Isolates: geographic and cultural factors of human genetic variation" regarding Italian extant geographical and/or linguistic isolates, aimed at overcoming the limitations of previous studies regarding geographical coverage of isolates, number and type of genetic polymorphisms under study and suitability of the experimental design to investigate gene-culture coevolutionary processes. An interdisciplinary sampling approach will make it possible to collect several linguistic isolates and their geographic neighbours from Trentino, Veneto, Friuli, Tuscany, Sardinia and Calabria. This will be coupled with a shared genotyping strategy based on mitochondrial and Y-chromosomal polymorphisms. The results will be analyzed with a focus on the role of geographical and cultural factors in shaping human biodiversity. The aims of the project go beyond the simple reconstruction of the genetic structure and history of the examined groups. In fact, the study will also include an assessment for future bio-medical studies and the development of genetic and bio-demographic databases. Ethical and educational aspects are also foreseen by the project, by using informed consents together with disseminating activities in loco, completed by the creation of a dedicated web site for both scientific and public audiences.

Abstract

One way to alleviate the environmental problem associated with the disposal of granite processing waste is to utilize this material as aggregate in concrete manufacturing or as raw material for producing concentrates to be used in the ceramics industry. Based on the results of material characterization and amenability tests conducted on Sardinian granite waste, the paper demonstrates the potential utilization of this type of material as a concrete ingredient and in cement blends. Likewise, the application of basic mineral processing techniques, such as magnetic separation, integrated where necessary with flotation, showed that the beneficiated products of the same granite waste as well as of road tunnelling waste should find a ready market in the ceramics industry.

80. Spatial distribution of PCDD/F in surface sediments of Lake Maggiore (Italy)

Abstract

Lake Maggiore (LM) is the second largest Italian lake in terms of volume, surface and depth, with the northern third residing in Swiss territory. The most important tributaries are River Ticino (Northern inlet) and River Toce (Western inlet). The River Ticino leaves LM at the southern end and connects LM to the River Po. Particular concern arose from the contamination with DDT and its metabolites, which exceeded Italian and EU levels for consumption in fish, and was detected in bird eggs from fish feeding species as well. As a consequence the local Health Authorities prohibited commercial fishing of a series of contaminated fish species. The source of DDT contamination was a DDT production site, closed down in 1996. This plant was located on the Marmazza River, a tributary of River Toce entering LM from the westernmost part of LM, close to Baveno. Besides the contamination with DDT, PCBs, including some dioxin like congeners, were found at concentrations exceeding 3-7 times the levels of other sub alpine lakes in Italy. Levels of HCB and HCH in the LM ecosystem instead gave no rise to concern. The aim of the present study is to supplement the existing data on POPs with sediment levels of PCDD/Fs, where so far only few data are available for LM. The PCDD/F concentration in the 50 surficial sediment samples taken at water depths between 5 and 60 m in LM display a broad range of concentrations, from 0.13 to 32 pg WHO TEQ g-1 d.w.. The PCDD/Fs show a consistent congener pattern. In the sample taken at the inlet of River Bardello the congener distribution was dominated by Octachloro- followed by Heptachloro-dioxins; the Heptachloro- and Octachloro –furans occurred at a level of only 0.80 pg WHO TEQ g-1 d.w..The spatial homogeneity of the pattern along the whole lake (Figure 2) underlines the absence of local sources and important riverine inputs into LM. Moreover, the pattern is similar to that of PCDD/Fs in atmospheric particulate matter and bulk deposition

Abstract

Among the various industrial minerals occurring in Sardinia (Italy), the perlite minerals are not to be overlooked. Mining of perlite deposits is however hampered by the presence of obsidian and lesser amounts of mica as well as other accompanying minerals. In order to obtain a perlite material as free as possible of the above contaminants, a systematic investigation has been carried out for exploring the potential applicability of different beneficiating methods. This paper describes the basic studies conducted on the perlite and obsidian for determining the surface electric charge acquired on contact with various materials. The results of separation tests performed in a climate chamber using electric separators that rely on ionic charging are discussed for varying feed size and ambient conditions. The findings demonstrate the potential of electric techniques for separating the perlite from obsidian.

Abstract

The increasingly restrictive regulations foreseen for landfills and the consequent need to reduce the amount of fly ash to be disposed of in this way, makes beneficiation essential for increasing both the use and value of this by-product. Electrostatic separation offers a potentially promising solution to this problem as with this technique it is possible to treat the hot, dusty fly ash in the dry state. In particular the present paper describes a new separator designed with a view to overcoming the drawbacks of conventional machines. The new separator consists essentially of a grounded metal conveyor belt onto which the particle stream is spread in a thick layer by means of a vibrating feeder. Some significant results of experimental tests carried out on samples collected from Italian power stations are described and discussed.

The volume of pulp directly agitated is increased and the distribution of bubbles improved. Flotation tests on coal during continuous operation showed that the hydrodynamic regime in the cell is further improved and combustible recovery is considerably higher than that previously achieved. Abstract

Item does not contain fulltext, Mutations in the human Crumbs homologue 1 (CRB1) gene cause severe retinal dystrophies. CRB1 is homologous to Drosophila Crumbs, a protein essential for establishing and maintaining epithelial polarity. We have isolated the mouse orthologue, Crb1, and analyzed its expression pattern in embryonic and post-natal stages. Crb1 is expressed exclusively in the eye, and the central nervous system. In the developing eye, expression of Crb1 is detected in the retinal progenitors, and later on becomes restricted to the differentiated photoreceptor cells where it remains active up to the adult stage. In the developing neural tube, expression of Crb1 is restricted to its most ventral structures, coinciding with the expression domain of Nkx2.2. In the adult brain, Crb1 expression is defined to areas where the production and migration of neurons occurs in adulthood.
