We investigate the coseismic and post-seismic deformation due to the 6 February 2018 M w 6.4 Hualien earthquake to gain improved insights into the fault geometries and complex regional tectonics in this structural transition zone. We generate coseismic deformation fields using ascending and descending Sentinel-1A/B InSAR data and GPS data. Analysis of the aftershocks and InSAR measurements reveal complex multifault rupture during this event. We compare two fault model joint inversions of SAR, GPS and teleseismic body waves data to illuminate the involved seismogenic faults, coseismic slip distributions and rupture processes. Our preferred fault model suggests that both well-known active faults, the dominantly left-lateral Milun and Lingding faults, and previously unrecognized oblique-reverse west-dipping and north-dipping detachment faults, ruptured during this event. The maximum slip of ∼1.6 m occurred on the Milun fault at a depth of ∼2–5 km. We compute post-seismic displacement time series using the persistent scatterer method. The post-seismic range-change fields reveal large surface displacements mainly in the near-field of the Milun fault. Kinematic inversions constrained by cumulative InSAR displacements along two tracks indicate that the afterslip occurred on the Milun and Lingding faults and the west-dipping fault just to the east. The maximum cumulative afterslip of 0.4–0.6 m occurred along the Milun fault within ∼7 months of the main shock. The main shock-induced static Coulomb stress changes may have played an important role in driving the afterslip adjacent to coseismic high-slip zones on the Milun, Lingding and west-dipping faults. The Mw 6.3 Jinghe earthquake struck Xingjiang Province, China, on 8 August 2017 (05:15:04 UTC); the epicenter was near the Kusongmuxieke Piedmont Fault (KPF) of the northern Tian Shan Mountains. We used multi-source and multi-track satellite Synthetic Aperture Radar (SAR) imagery and Interferometric SAR (InSAR) techniques to reconstruct the coseismic displacement field from different line-of-sight geometries. To reduce the phase artifacts, we employed multi-temporal scenes acquired by Sentinel-1, and reconstructed the coseismic deformation through a temporal averaging strategy. Together with a single interferometric pair obtained using the Phased Array type L-band Synthetic Aperture Radar 2 (PALSAR2) sensor aboard the Advanced Land Observing Satellite 2 (ALOS2), we obtained five displacement maps with slightly different viewing geometries; all of which were used to constrain a geodetic inversion to retrieve the fault geometry parameters and slip distribution. Based on the focal mechanism and regional geology, we constructed multiple fault models that differ in dip direction (south and north dipping), and various striking angles. Both models fit the InSAR displacement maps, but have slip distributions of different depths. The slip depth of the south dipping model, with a dip of ~42°, is the most consistent with the relocated earthquake sequence and regional geological structure. Through the geodetic inversion, the maximum slip (0.25 m) occurred at 14.05 km and the associated rake was 89.56°. The result implies that the seismogenic fault is a blind thrust fault north of KPF (towards the foreland). Considering the relative locations of the suggested blind fault, the KPF, and the continuing north to south (N–S) shortening of the Tian Shan Mountains, this fault could be formed by the northward propagation of the regional fold-thrust belt. Pediatric acute lymphoblastic leukemia (ALL) is the most common neoplasm and one of the primary causes of death in children. Its treatment is highly dependent on the correct classification of subtype. Previously, we developed a microarray-based subtype classifier based on the relative expression levels of 62 marker genes, which can predict 7 different ALL subtypes with an accuracy as high as 97% in completely independent samples. Because the classifier is based on gene expression rank values rather than actual values, the classifier enables an individualized diagnosis, without the need to reference the background distribution of the marker genes in a large number of other samples, and also enables cross platform application. Here, we demonstrate that the classifier can be extended from a microarray-based technology to a multiplex qPCR-based technology using the same set of marker genes as the advanced fragment analysis (AFA). Compared to microarray assays, the new assay system makes the convenient, low cost and individualized subtype diagnosis of pediatric ALL a reality and is clinically applicable, particularly in developing countries. Background: Neural precursor cell expressed developmentally down-regulated 4-like (NEDD4L) is a candidate gene for hypertension, and carriers of an intact NEDD4L C2-domain,encoded by the NEDD4L rs4149601 (G/A) GG genotype, together with the C-allele of the NEDD4L rs2288774 (C/T) polymorphism have been found be associated with hypertension both in African Americans and whites. However, there is no data on the relationship between polymorphism of NEDD4L rs4149601 and rs2288774 and hypertension in Chinese chronic kidney disease (CKD) patients. The purpose of the current study was to investigate the relationship between the variation of NEDD4L rs4149601, rs2288774 and hypertension in CKD patients.Methods: A total of 546 Chines Hans CKD patients were enrolled in our study. The SNPs were genotyped using PCR-based techniques. All patients underwent ambulatory blood pressure monitoring, and clinical data were also collected. Multivariate logistic regression analysis was used to identify the relationship between polymorphisms and hypertension.Results: 506 patients carried GG/GA genotype and 30 carried AA genotype. Rs4149601 AA genotype carriers had significantly higher rate of hypertension (68.3% vs 46.2%, P = 0.022) than GG/GA genotype carriers by Chi-squared test. AA genotype carriers also had a higher day-time and bedtime systolic blood pressure (142±16 vs 135±23, P=0.036; 137±18 vs 127±13, P=0.022, respectively) when compared with GG/GA genotype carriers. AA genotype [OR= 3.08, 95% CI (1.06-9.80)], lowever eGFR [OR=0.98, 95% CI (0.97-0.99)], older age [OR=1.03, 95% CI (1.01-1.05)] were independently associated with hypertension in CKD patients by multivariate logistic regression. However, No difference was found in blood pressure with rs2288774 TT/TC/CC genotypes, and no difference was found in the incidence of hypertension among patients with three genotypes.Conclusions: Our results suggested 4149601AA genotype of NEDD4L may be associated with hypertension in CKD patients, and further genetic and functional studies are required to understand its role in the manifestation of hypertension in Chinese CKD patients. Introduction: Interstitial cells are crucial to the development of kidney structure and function, although the mechanism underlying their role in it remains unclear to date. Our previous study identified cell clusters in human fetal kidney tissue, and we further analyzed the interstitial cell cluster within this context., Methods: We extracted the barcoded cDNA from tissue samples and prepared spatial transcriptome libraries. Sequencing data was quality-checked, normalized, and clusters were identified using Seurat. Single-cell and spatial data were integrated using MIA and cell types were deconvoluted. DEGs in interstitial cells were identified and functionally annotated using DAVID. CellPhoneDB was used to predict ligand-receptor interactions between cell types., Results: The results of the present study revealed that this cluster of interstitial cells appeared to be scattered in the junction between the cortical and medullary regions. The subsequent KEGG pathway analysis revealed that the differentially expressed genes (DEGs) in this cluster of interstitial cells were involved in the WNT signaling pathway. The Gene Ontology (GO) analysis revealed that these DEGs were involved in multiple pathways associated with kidney development, with six of the genes (NKD2, TCF21, WNT5A, WNT4, MDK, and SFRP1) associated with kidney development exhibiting significant upregulation. Accordingly, it was inferred that these interstitial cells might be involved in regulating epithelial cell differentiation, ureteral bud development, and morphogenesis. The subsequent cell-cell communication analysis revealed that the cellular crosstalk was primarily regulated mainly by ligand-receptor pairs. Additionally, 17 genes reported to be associated with kidney disease were focused on, and these genes were found to be predominantly expressed in a single cell type., Conclusion: In summary, the present study revealed the characteristics of a previously identified cluster of interstitial cells in the kidney tissue, thereby providing fresh insights into the process of kidney development., (The Author(s). Objectives: To evaluate the specificity of three-color flow cytometry in childhood acute lymphoblastic leukemia (ALL) immunophenotyping., Methods: Immunophenotyping was performed by three-color flow cytometry analysis using CD(45)/SSC gating., Results: The percentage of blasts was correlated better with leukemic cell count compared with that of FSC/SSC, and the false positive results were low. Among eighty six cases of ALL, 95.3% was B-ALL, in which common-ALL and Pro-B-ALL were 76.8% and 6.1%, respectively, and 2.3% was T-ALL. CD(34)(+) and myeloid-associated antigen expression were observed in 57.0% and 34.9% of the cases, respectively, among which Pro-B-ALL was the commonest. CD(33) was more commonly expressed than CD(13) in Pro-B-ALL cases, but no difference in the expression between these two antigens in other subtypes., Conclusion: Gating of CD(45)/SSC eliminated effection of normal cells to blasts in bone marrow, with which the immunophenotyping results were more reliable.
