59. Molecular simulation of imperfect structure I CO2 hydrate growth in brine.

Abstract

In order to investigate the viability of carbon dioxide (CO2) storage in seawater, molecular dynamics techniques were employed to study the dynamic evolution of CO2 hydrate in saline water. The simulation was conducted under specific conditions: a temperature of 275 K, a pressure of 10 MPa and a simulated marine environment achieved using a 3.4 wt% sodium chloride (NaCl) solution. The total simulation time was 1000 ns. The results of the simulation indicate that the pre‐existence of CO2 hydrate crystals as seeds leads to rapid growth of CO2 hydrate. However, analysis of the F3 and F4 order parameters reveals that the hydrate does not meet the standard values of the perfect structure I (sI) type, confirming the existence of an imperfect structure during the simulation. Additionally, the changes in the number of different phase states of water molecules during the hydrate growth process shows that there are always some liquid water molecules, which means some water molecules fail to form solid water cages. Further investigation suggests that the presence of Na+ and Cl− hampers the hydrogen bonds between water molecules, resulting in incomplete cage structures. By analyzing the density variations in the system, it is observed that CO2 hydrate, with a density of around 1.133 g cm−3, forms rapidly, surpassing the average density of seawater. This density increase facilitates the efficient and swift containment of CO2 on the seabed, thereby supporting the feasibility of the CO2 storage theory. [ABSTRACT FROM AUTHOR]

Abstract

Summary A gel-plug system for temporary blocking technology is proposed in this paper to address the prevalent leakage of killing fluid in low-pressure wells; the low technical strength of existing gel plugs for temporary blocking in well killing; difficult-to-control crosslinking time; and gel embrittlement and the difficulty of breaking certain gel plugs. A mixture of etherified galactomannan plant gum, isooctanol polyoxyethylene ether surfactant, and oil phase was used as a thickener. An inorganic salt complex containing long-chain polyhydroxy alcohol was used as a crosslinker and the concentration of long-chain polyhydroxy alcohol far exceeds the theoretical amount required to complex the metal ion. A mixture of polyhydroxy alcohol with a small amount of weak acid was used as a crosslinking regulator. Finally, a mixture of sodium thiosulfate and long-chain quaternary ammonium salt surfactant was used as a stabilizer. Laboratory evaluations showed that this gel-plug system can be directly pumped into the wellbore after being mixed homogeneously, and the viscosity of the system on the surface can be controlled by the amount of crosslinking regulator. The viscosity of the gel-plug system after gelling was high (viscoelastic solid colloid); the initial viscosity reached 30 000 mPa·s at 120°C and retained a semisolid gel shape after aging for 72 hours. Right-angle thickening occurred when the gel warmed to target-zone temperature. The acidic liquid breaker acted quickly, and the viscosity of the broken fluid was lower than 5 mPa·s after 1 to 4 hours. This gel plug for temporary blocking and well-killing technology was successfully applied in a low-pressure, leakage-prone gas well. No gas, pressure, or liquid remained in the open well after killing, the wellhead was successfully replaced, and the tubing was successfully removed. The gel plug also exhibited self-healing: The hole formed by the tubing could be filled and sealed automatically by the gel plug in the annulus. The static friction (outer wall) of 73-mm tubing in the gel plug was 39.6 t/km; the dynamic friction (outer wall) after tubing removal was 7.2 t/km. This gel plug thus shows promise as a temporary blocking technology in workover operations of low-pressure, leakage-prone gas wells.

Abstract

A water-soluble nonionic surfactant hydrophobic monomer (AT100) was synthesized by esterification of acryloyl chloride with Triton X-100, after that, a nonionic hydrophobically associating polyacrylamide P(AM-AT100) was synthesized and used as drag reducer. They were characterized by IR and NMR. In fresh water, when the concentration of drag reducer reached 0.3 g·L−1 (so called slick water), the viscosity was 3.86 mm2·s−1 with a drag reduction rate up to 74.7%. After sodium dodecyl sulfate (SDS) was added into the slick water, hydrophobic chains of SDS and hydrophobic units in P(AM-AT100) aggregated and micelles were likely formed, and this process was just like hydrophobic association. The viscosity of the slick water increased significantly, while the drag reduction rate of the mixed system decreased. With SDS concentration at 1.5 g·L−1 and the drag reducer concentration at 0.3 g·L−1, the drag reduction rate was 71.72% and viscosity was 6.25 mm2·s−1, indicating that increasing viscosity had a significantly adverse effect on drag reduction performance. The proppant layup experiment confirmed that the sand-carrying capacity of low-viscosity slick water is limited, so proppant will settle inevitably. [ABSTRACT FROM AUTHOR]

99. Identification of microRNA expression profiles of CD44+ ovarian cancer stem cells.

Abstract

Purpose: The aim of our study was to investigate microRNA (miRNA) expression profiles in CD44+ ovarian cancer stem cells (ovarian CSCs). Methods: In this study, we enriched CD44+ ovarian CSCs using magnetic activated cell sorting (MACS). A combination of real-time quantitative PCR (qRT-PCR), western blot and sphere formation assays was used to demonstrate stem cell-like properties. RNA sequencing was used to detect the miRNA expression profiles in CD44+ ovarian CSCs. Transient transfection, qRT-PCR, western blot and sphere formation assays were further used to test the function of miR-181a-2-3p. Results: We found that CD44+ ovarian CSCs showed enhanced sphere formation and expression of stemness-associated genes (NANOG, OCT4, SOX2) compared to ovarian cancer cells. The RNA sequencing results showed that the miRNA expression profiles of CD44+ ovarian CSCs were different from those of ovarian cancer cells. GO and KEGG pathway analyses indicated that these miRNAs regulate stem cell-like properties in CD44+ ovarian CSCs. In addition, miR-181a-2-3p negatively regulates the stem cell-like properties of CD44+ ovarian CSCs by targeting EGR1. Conclusion: Our data suggest that miRNAs play important roles in regulating the stem cell-like properties of CD44+ ovarian CSCs. [ABSTRACT FROM AUTHOR]
