I ncreasing attention has been paid to health condition of the elderly as the fast growth of the elderly population. The Traditional Chinese Medicine external application has advantages of easy-to-use and low cost, and is highly accepted among patients. This paper summarized the current status of Traditional Chinese Medicine external application, such as moxibustion, auricular acupoint pressing therapy and Chinese Herbal medicine footbath, in integrated pension and medical service of Traditional Chinese Medicine for the elderly, and provides reference for the establishment and practice of pension and medical service with Traditional Chinese Medicine techniques. A novel amidoximated polyethylene nonwoven fabric (PE-g-AO) was successfully prepared by electron beam pre-irradiation grafting with glycidyl methacrylate, followed by reaction induced of ethylenediamine and maleic anhydride, AIBN-initiated polymerization of acrylonitrile, and finally, amidoximation. The chemical structure and morphology of the PE nonwoven fabric before and after modification were characterized by total reflection infrared spectroscopy, X-ray photoelectron spectroscopy, and scanning electron microscopy. Adsorption experiment results showed that the adsorption capacity of PE-g-AO for uranium is 71.92 mg/g (U(VI): 20 mg/L; NaF: 2.0 g/L; sorbent dosage: 0.2 g/L; adsorption time: 216 h). When the dosage of adsorbent is 1.0 g/L, the uranyl ions in the solution are basically adsorbed, and the residual uranium concentration is 0.04 mg/L, which meets the national emission standard. Furthermore, the material has a good adsorption selectivity for uranium in a solution containing UO22+, Ca2+, Mg2+, K+, and Na+ ions.

Objective: The main pathological change of myocarditis is an inflammatory injury of cardiomyocytes. Long noncoding RNAs (lncRNAs) are closely related to inflammation, and our previous study showed that differential expression of lncRNAs is associated with myocarditis. This study aimed to investigate the impact of lncRNAs on the onset of myocarditis. Methods: RNA expression was measured by quantitative reverse-transcription polymerase chain reaction (RT-qPCR). Lipopolysaccharide (LPS) was used to induce inflammation in human cardiomyocytes (HCMs). The expression of inflammatory cytokines and myocardial injury markers was detected by enzyme-linked immunosorbent assay (ELISA) and RT-qPCR. Cell viability and apoptosis were measured by the cell counting kit-8 assay and flow cytometry. The binding force between lncRNA NONHSAT122636.2 and microRNA miRNA-2110 was detected using the dual-luciferase assay. Results: NONHSAT122636.2 was dynamically expressed in patients with myocarditis and negatively correlated with inflammation severity. The overexpression of NONHSAT122636.2 improved inflammatory injury in LPS-stimulated HCMs. The study observed that there was a weak binding force between NONHSAT122636.2 and miR-2110. Conclusion: NONHSAT122636.2 attenuates myocardial inflammation and apoptosis in myocarditis. Additionally, its expression decreases in the peripheral blood of children suffering from myocarditis and in patients who are diagnosed for the first time showing higher diagnostic sensitivity and specificity. This decrease is negatively correlated with the degree of inflammation. Aims/Background Gestational diabetes mellitus is a common pregnancy complication that affects approximately 14% of pregnancies worldwide and can lead to adverse maternal and neonatal outcomes. This study aimed to investigate the trajectories of gestational weight gain among gestational diabetes mellitus patients and to inform the development of effective weight management strategies. Methods Demographic and antenatal examination data from 1421 pregnant women diagnosed with gestational diabetes mellitus were retrospectively analysed. Quantitative data comparisons were performed using Chi-square tests, Fisher's exact test, t-tests, and one-way analysis of variance. Group-based trajectory modelling was employed to identify the trajectories of gestational weight gain among patients with gestational diabetes mellitus. Results This study revealed that pre-pregnancy body mass index and types of gestational diabetes mellitus significantly influence gestational weight gain (p < 0.05). Group-based trajectory modelling identified three distinct gestational weight gain trajectories. Patients with gestational diabetes mellitus demonstrated a continuous weight gain throughout pregnancy, while women who were overweight or obese before pregnancy were more likely to follow a low-speed growth trajectory. Women in the rapid growth trajectory group were more inclined to deliver by caesarean section and were more likely to give birth to macrosomic infants. Conclusion Our research underscores the importance of identifying and distinguishing between different gestational weight gain trajectories in pregnant women, thereby identifying high-risk groups, which is crucial for improving the health conditions of both mothers and newborns. Despite extensive research highlighting the pivotal role of MYB transcription factors in regulating anthocyanin biosynthesis, the interactive regulatory network involving these MYB factors in pear fruits remains inadequately characterized. In this study, the anthocyanin-regulatory gene PbrMYB114 was successfully cloned from 'Yuluxiang' pear (Pyrus bretschneideri) fruits, and its influence on anthocyanin accumulation was confirmed through transient expression assays. Specifically, the co-transformation of PbrMYB114 with its partner PbrbHLH3 in pears served to validate the functional role of PbrMYB114. Subsequently, PbrMYB114 was employed as bait in a yeast two-hybrid screening assay, using a 'Yuluxiang' pear protein library, which led to the identification of 25 interacting proteins. Further validation of the interactions between PbrMYB114 and PbrMT2/PbrMT3 was conducted. Investigations into the role of PbrMT2 and PbrMT3 in 'Duli' seedlings (Pyrus betulaefolia) revealed their potential to enhance anthocyanin accumulation. The outcomes of these studies provide novel insights into the protein network that regulates pear anthocyanin biosynthesis, particularly the functional interactions among PbrMYB114 and associated proteins. There is a growing need for new antibiotics. Compounds that target the proton motive force (PMF), uncouplers, represent one possible class of compounds that might be developed because they are already used to treat parasitic infections, and there is interest in their use for the treatment of other diseases, such as diabetes. Here, we tested a series of compounds, most with known antiinfective activity, for uncoupler activity. Many cationic amphiphiles tested positive, and some targeted isoprenoid biosynthesis or affected lipid bilayer structure. As an example, we found that clomiphene, a recently discovered undecaprenyl diphosphate synthase inhibitor active against Staphylococcus aureus, is an uncoupler. Using in silico screening, we then found that the anti-glioblastoma multiforme drug lead vacquinol is an inhibitor of Mycobacterium tuberculosis tuberculosinyl adenosine synthase, as well as being an uncoupler. Because vacquinol is also an inhibitor of M. tuberculosis cell growth, we used similarity searches based on the vacquinol structure, finding analogs with potent (∼0.5-2 μg/mL) activity against M. tuberculosis and S. aureus. Our results give a logical explanation of the observation that most new tuberculosis drug leads discovered by phenotypic screens and genome sequencing are highly lipophilic (logP ∼5.7) bases with membrane targets because such species are expected to partition into hydrophobic membranes, inhibiting membrane proteins, in addition to collapsing the PMF. This multiple targeting is expected to be of importance in overcoming the development of drug resistance because targeting membrane physical properties is expected to be less susceptible to the development of resistance.

For problems of existing pedestrian detection methods based on deep learning such as large computation, detection efficiency relying on hardware performance heavily and so on, the pedestrian detection method based on SSD network was improved. A lightweight convolutional neural network based on DenseNet network is designed as basic network of SSD network to meet real-time detection requirements of underground video pedestrian, and an auxiliary network based on ResNet network is designed to strengthen feature extraction ability and improve correctness of pedestrian detection. An underground video pedestrian detection method based on the improved SSD network has been arranged in embedded platform Jetson TX2 for experiments. The results show that detection accuracy rate of the method for underground video pedestrian is 87.9% as well as nearly 100% for underground low-density pedestrian scene, and calculation speed achieves 48 frames per second, which is about 4.4 times as quick as the pedestrian detection method based on SSD network and meets real-time detection requirements of underground pedestrian.

With the rise in antibiotic resistance, there is interest in discovering new drugs active against new targets. Here, we investigate the dynamic structures of three isoprenoid synthases from Mycobacterium tuberculosis using molecular dynamics (MD) methods with a view to discovering new drug leads. Two of the enzymes, cis-farnesyl diphosphate synthase (cis-FPPS) and cis-decaprenyl diphosphate synthase (cis-DPPS), are involved in bacterial cell wall biosynthesis, while the third, tuberculosinyl adenosine synthase (Rv3378c), is involved in virulence factor formation. The MD results for these three enzymes were then compared with previous results on undecaprenyl diphosphate synthase (UPPS) by means of active site volume fluctuation and principal component analyses. In addition, an analysis of the binding of prenyl diphosphates to cis-FPPS, cis-DPPS, and UPPS utilizing the new MD results is reported. We also screened libraries of inhibitors against cis-DPPS, finding ~1 μm inhibitors, and used the receiver operating characteristic-area under the curve (ROC-AUC) method to test the predictive power of X-ray and MD-derived cis-DPPS receptors. We found that one compound with potent M. tuberculosis cell growth inhibition activity was an IC(50) ~0.5- to 20-μm inhibitor (depending on substrate) of cis-DPPS, a ~660-nm inhibitor of Rv3378c as well as a 4.8-μm inhibitor of cis-FPPS, opening up the possibility of multitarget inhibition involving both cell wall biosynthesis and virulence factor formation.

Hand pose estimation is one of the important research directions of computer vision, which plays an important role in human-computer interaction, virtual reality, robot control and other application fields. At present, hand pose estimation has the problem of single feature representation method. This paper proposes a feature construction method of hand key point connection relationship and a key point feature aggregation enhancement method based on hand motion semantic relationship to improve the hand feature representation and information sharing ability. Aiming at the occlusion problem in hand target detection and image segmentation, a hand contour feature extraction method is designed to improve the preprocessing effect. Based on the proposed multi- feature representation and enhancement method, a depth learning neural network model based on full convolution structure is constructed to avoid the problem of spatial information loss caused by direct regression calculation of 3D pose information, thus effectively improving the accuracy of 3D hand pose estimation. Compared with the SOTA model on DO, ED, RHD datasets, it has achieved a competitive effect, and the average AUC result has reached 93.3%, indicating that the proposed method also has good universality. A water‐soluble polymeric pyrene‐based polythioacetal (PTA‐Py) with thioacetal units in the main chain is simply synthesized by direct polycondensation of 3, 6‐dioxa‐1, 8‐octanedithiol, 1‐pyrene formaldehyde, and mPEG2k‐SH. The probe PTA‐Py shows a good fluorescence response to Hg2+ ions due to the Hg2+‐promoted deprotection reaction of thioacetal groups to regenerate the original 1‐pyrene formaldehyde compound. After adding Hg2+ to the PTA‐Py solution, the fluorescence intensity (FI) gradually increases with increasing concentrations of Hg2+. Compared with other metal ions, the probe exhibits high sensitivity, good selectivity, and rapid response to Hg2+. The low detection limits are 12.3 nm in ethanol‐PBS buffer and 13.3 nm in water, respectively. The results imply that the simply synthesized water‐soluble polymeric probe had potential applications in the rapid detection of Hg2+ ions in aqueous solutions. Moreover, the polymeric PTA‐Py shows high sensitivity for CH3Hg+ with detection limits of 26.5 nm in ethanol/PBS buffer. In addition, PTA‐Py can efficiently detect Hg2+ ions in HeLa cells. The results demonstrate that a valuable method is developed for biocompatible polymeric sensors for Hg2+ ions in biological and environmental samples. [ABSTRACT FROM AUTHOR]
