Purpose The life quality of cervical cancer patients is severely compromised due to the vaginal stump contracture after surgery combined with radiotherapy. Therefore, our study focuses on ameliorating or preventing postoperative vaginal contracture. The objectives of this study were (1) to evaluate the method of ileal graft with vascular pedicle to extend the length of vagina and (2) to investigate the effect of the operation to prevent vaginal stump contracture after cervical cancer surgery combined with radiotherapy. Methods Twenty-five patients with vaginal stump contracture after cervical cancer radical resection with sequential radiotherapy were recruited for the study between 2011 and 2014. The therapy includes releasing the adhesion between vaginal stump and rectum as well as bladder, resecting the vaginal stump but reserving the exterior orifice of vagina, and extending the vaginal length using vascularized ileal graft. Results No postoperative complications such as infection and bleeding were observed in all 25 patients. The patients were satisfied with the length and width of vagina after extension; the discharge of reconstructed vagina was acceptable. All patients had sexual intercourse on the follow-up examination, and five patients complained of vaginal bleeding during or after sexual intercourse, although no abnormality was found on colposcopy. Conclusions The patients demonstrated a high level of safety, efficacy, and satisfaction with the ileal–vaginal extension for treating vaginal stump contracture after cervical cancer surgery combined with radiotherapy, suggesting that this method is valid for broad clinical application to improve the life quality of cervical cancer patients after surgery.

Polycystic ovary syndrome (PCOS) is a common age-related endocrinopathy that promotes the metabolic disorder of the liver. Growing evidence suggests that the pathophysiology of this disorder is closely associated with the interaction between the liver and its exosome. However, the underlying mechanism of the interactions remains unclear. In this study, we aimed to investigate the metabolite profiles of liver tissues and hepatic exosomes between normal (n = 11) and PCOS (n = 13) mice of young- and middle-age using gas chromatograph-mass spectrometry (GC-MS) based metabolomics analysis. Within the 145 identified metabolites, 7 and 48 metabolites were statistically different (p < 0.05, q < 0.05) in the liver tissue and exosomes, respectively, between PCOS and normal groups. The greater disparity in exosome indicated its potential to reflect the metabolic status of the liver. Based on hepatic exosome metabolome, the downregulations of glycolysis and TCA cycle were related to hepatic pathophysiology of PCOS independent of age. Fatty acids were the preferred substrates in young-age-PCOS liver while amino acids were the main substrates in middle-age-PCOS liver for the processes of gluconeogenesis. Overall, this study enables us to better understand the metabolic status of the PCOS liver at different ages, and exosome metabolomics shows its potential to gain the metabolic insights of parental cell or source organ.

The COVID-19 pandemic has deeply influenced the tourism and hospitality industry, and it has also reshaped people’s travel preferences and related behaviors. As a result, how prospective travelers perceive travel constraints and their effects on future travel behaviors may have changed to some extent. Besides, such perception arguably varies across gender. Therefore, this research examines the interplay between travel constraints, gender, and travel intentions for facilitating robust tourism recovery by revisiting the Leisure Constraints Model (LCM) from a gender perspective. Data were collected through a survey from 357 Malaysian prospective travelers. By conducting path analysis and multigroup analysis (MGA), it is found that structural and interpersonal constraints impose indirect effects on travel intentions (mediated by intrapersonal constraints), and gender moderating the effect of structural cost on intrapersonal constraints and effect of intrapersonal constraints on travel intentions. Based on these findings, this research provides theoretical and practical implications into how to adjust their marketing strategies and travel products during the era of “new normal” for tourism policy makers, destination marketers, and related businesses.

BackgroundColorectal cancer (CRC) is a common gastrointestinal tumor with high morbidity and mortality. At the molecular level, patients at different stages present considerable heterogeneity. Although the miRNA in exosome is an effective biomarker to reveal tumor progression, studies based on stage-associated exosome miRNA regulatory network analysis still lacking. This study aims to identify CRC stage-associated exosome miRNAs and reveal their potential function in tumor progression.MethodsIn this study, serum and cellular exosome miRNA expression microarrays associated with CRC were downloaded from GEO database. Stage-common (SC) and stage-specific (SS) differentially expressed miRNAs were extracted and their targets were identified based on 11 databases. Furthermore, miRNA SC and SS regulatory function networks were built based on the CRC phenotypic relevance of miRNA targets, and the corresponding transcription factors were identified. Concurrently, the potential stage-associated miRNAs were identified by receiver-operating characteristic (ROC) curve analysis, survival analysis, drug response analysis, ceRNA analysis, pathway analysis and a comprehensive investigation of 159 publications.ResultsTen candidate stage-associated miRNAs were identified, with three SC (miR-146a-5p, miR-22-3p, miR-23b-3p) and seven SS (I: miR-301a-3p, miR-548i; IIIA: miR-23a-3p; IV: miR-194-3p, miR-33a-3p, miR-485-3p, miR-194-5p) miRNAs. Additionally, their targets were enriched in several vital cancer-associated pathways such as TGF-beta, p53, and hippo signaling pathways. Moreover, five key hotspot target genes (CCNA2, MAPK1, PTPRD, MET, and CDKN1A) were demonstrated to associated with better overall survival in CRC patients. Finally, miR-23b-3p, miR-301a-3p and miR-194-3p were validated being the most stably expressed stage-associated miRNAs in CRC serum exosomes, cell exosomes and tissues.ConclusionsThese CRC stage-associated exosome miRNAs aid to further mechanism research of tumor progression and provide support for better clinical management in patients with different stages.

Ovarian cancer (OC) is the third most common malignant tumor of women accompanied by alteration of systemic metabolism, yet the underlying interactions between the local OC tissue and other system biofluids remain unclear. In this study, we recruited 17 OC patients, 16 benign ovarian tumor (BOT) patients, and 14 control patients to collect biological samples including ovary plasma, urine, and hair from the same patient. The metabolic features of samples were characterized using a global and targeted metabolic profiling strategy based on Gas chromatography-mass spectrometry (GC-MS). Principal component analysis (PCA) revealed that the metabolites display obvious differences in ovary tissue, plasma, and urine between OC and non-malignant groups but not in hair samples. The metabolic alterations in OC tissue included elevated glycolysis (lactic acid) and TCA cycle intermediates (malic acid, fumaric acid) were related to energy metabolism. Furthermore, the increased levels of glutathione and polyunsaturated fatty acids (linoleic acid) together with decreased levels of saturated fatty acid (palmitic acid) were observed, which might be associated with the anti-oxidative stress capability of cancer. Furthermore, how metabolite profile changes across differential biospecimens were compared in OC patients. Plasma and urine showed a lower concentration of amino acids (alanine, aspartic acid, glutamic acid, proline, leucine, and cysteine) than the malignant ovary. Plasma exhibited the highest concentrations of fatty acids (stearic acid, EPA, and arachidonic acid), while TCA cycle intermediates (succinic acid, citric acid, and malic acid) were most concentrated in the urine. In addition, five plasma metabolites and three urine metabolites showed the best specificity and sensitivity in differentiating the OC group from the control or BOT groups (AUC > 0.90) using machine learning modeling. Overall, this study provided further insight into different specimen metabolic characteristics between OC and non-malignant disease and identified the metabolic fluctuation across ovary and biofluids.

BackgroundA better understanding of the factors and their correlation with clinical first-line nurses’ sleep, fatigue and mental workload is of great significance to personnel scheduling strategies and rapid responses to anti-pandemic tasks in the post-COVID-19 pandemic era.ObjectiveThis multicenter and cross-sectional study aimed to investigate the nurses’ sleep, fatigue and mental workload and contributing factors to each, and to determine the correlation among them.MethodsA total of 1,004 eligible nurses (46 males, 958 females) from three tertiary hospitals participated in this cluster sampling survey. The Questionnaire Star online tool was used to collect the sociodemographic and study target data: Sleep quality, fatigue, and mental workload. Multi-statistical methods were used for data analysis using SPSS 25.0 and Amos 21.0.ResultsThe average sleep quality score was 10.545 ± 3.399 (insomnia prevalence: 80.2%); the average fatigue score was 55.81 ± 10.405 (fatigue prevalence: 100%); and the weighted mental workload score was 56.772 ± 17.26. Poor sleep was associated with mental workload (r = 0.303, P < 0.05) and fatigue (r = 0.727, P < 0.01). Fatigue was associated with mental workload (r = 0.321, P < 0.05). COVID-19 has caused both fatigue and mental workload. As 49% of nurses claimed their mental workload has been severely affected by COVID-19, while it has done slight harm to 68.9% of nurses’ sleep quality.ConclusionIn the post-COVID-19 pandemic era, the high prevalence of sleep disorders and fatigue emphasizes the importance of paying enough attention to the mental health of nurses in first-class tertiary hospitals. Efficient nursing strategies should focus on the interaction of sleep, fatigue and mental workload in clinical nurses. In that case, further research on solutions to the phenomenon stated above proves to be of great significance and necessity.Clinical trial registration[https://clinicaltrials.gov/], identifier [ChiCTR2100053133].

Parkinson’s disease (PD) is the second most common neurodegenerative disease. PARK9 (also known as ATP13A2) is recognized as one of the key genes that cause PD, and a mutation in this gene was first discovered in a rare case of PD in an adolescent. Lewy bodies (LBs) formed by abnormal aggregation of α-synuclein, which is encoded by the SNCA gene, are one of the pathological diagnostic criteria for PD. LBs are also recognized as one of the most important features of PD pathogenesis. In this article, we first summarize the types of mutations in the ATP13A2 gene and their effects on ATP13A2 mRNA and protein structure; then, we discuss lysosomal autophagy inhibition and the molecular mechanism of abnormal α-synuclein accumulation caused by decreased levels and dysfunction of the ATP13A2 protein in lysosomes. Finally, this article provides a new direction for future research on the pathogenesis and therapeutic targets for ATP13A2 gene-related PD from the perspective of ATP13A2 gene mutations and abnormal aggregation of α-synuclein.

In order to reveal the influence of ammonium salts on the rare earth leaching process of weathered crust elution-deposited rare earth ores, ammonium acetate, ammonium chloride, and ammonium sulfate were used as leaching agents. The effects of the leaching agent on the rare earth leaching efficiency and the expansion, dissolution, and transformation behavior of clay minerals in the rare earth leaching process were studied. The results showed that rare earth leaching efficiency followed the order ammonium acetate > ammonium chloride > ammonium sulfate, with values of 90.60%, 85.96%, and 84.12%, respectively. The swelling ratio of clay mineral followed the order ammonium acetate < ammonium chloride < ammonium sulfate; the clay mineral swelling ratio was 2.09% when ammonium acetate was the leaching agent. Thermogravimetric analysis showed that the interlayer water content was the lowest when ammonium acetate was used as the leaching agent. Under the conditions of different leaching agents, the clay mineral contents changed from illite and halloysite to smectite and kaolinite. When ammonium acetate was used as the leaching agent, the relative conversion of illite was 1.49%, and that of smectite was only 0.17%. SEM analysis showed that the clay minerals expanded and dissolved obviously when ammonium chloride and ammonium sulfate were used as the leaching agents. Meanwhile, the clay mineral layered structure was relatively complete when ammonium acetate was used as the leaching agent. Therefore, when ammonium acetate was used as the leaching agent, it had the least effect on the swelling, dissolution, and transformation of clay minerals. This can provide a theoretical basis for the safe production of weathered crust elution-deposited rare earth ore, and for the screening of green and efficient leaching agents.

In recent years, the rapid development of the domestic alumina industry has greatly accelerated the consumption of high-alumina–silica-ratio bauxite resources in China. The development of an efficient and clean utilization technology applicable to low-grade bauxite in China is not only a requirement for resource and environmental protection, but also a powerful guarantee to maintain the sustainable development of China’s aluminum industry. Based on this, the authors’ team proposed a new process for the treatment of low-grade bauxite ore via a calcification–carbonization method from the perspective of equilibrium solid-phase reconstruction and achieved the first industrial-scale trial run on the basis of existing laboratory research. The results show that the mass fraction of Na2O in bauxite residue can be reduced to 0.95% in the treatment of typical diaspore bauxite, the A/S in the bauxite residue can be reduced to 0.85 after two-time carbonization–alumina dissolution, and the actual alumina dissolution rate can reach 81.32%. The relevant results verified the feasibility and advantages of the calcification–carbonization method in industrial production.

As an effective technique for fabricating conductive and thermally conductive polymer composites, a multi-filler system incorporates different types and sizes of multiple fillers to form interconnected networks with improved electrical, thermal, and processing properties. In this study, DIW forming of bifunctional composites was achieved by controlling the temperature of the printing platform. The study was based on enhancing the thermal and electrical transport properties of hybrid ternary polymer nanocomposites with multi-walled carbon nanotubes (MWCNTs) and graphene nanoplates (GNPs). With thermoplastic polyurethane (TPU) used as the matrix, the addition of MWCNTs, GNPs and both mixtures further improved the thermal conductivity of the elastomers. By adjusting the weight fraction of the functional fillers (MWCNTs and GNPs), the thermal and electrical properties were gradually explored. Here, the thermal conductivity of the polymer composites increased nearly sevenfold (from 0.36 W·m−1·k−1 to 2.87 W·m−1·k−1) and the electrical conductivity increased up to 5.49 × 10−2 S·m−1. It is expected to be used in the field of electronic packaging and environmental thermal dissipation, especially for modern electronic industrial equipment.

To reveal the regularity of variation in the rare earth occurrence states of weathered crust elution-deposited rare earth ores, ore samples from different weathering crust layers were obtained by performing the sequential extraction procedure. The order of rare earth contents firmly obeyed the following sequence: the weathered layer > humic layer > partly weathered layer. The occurrence states of rare earth elements were mainly the ion exchange state, carbonate bound state, iron–manganese oxide state, organic binding state and residual state. The proportions of rare earth elements found in the rare earth ion exchange state of the weathered layer, humic layer and partly weathered layer were 78.55%, 73.53% and 53.88%, respectively. The light rare earth elements (LREEs) found in the rare earth ion exchange state were enriched in the upper part of the weathering crust, while the heavy rare earth elements (HREEs) were enriched in the lower part. There were also obvious negative anomalies in the content of cerium in the ion exchange state. The content of rare earth elements found in the carbonate bound state was small, and the rare earth partition pattern was basically consistent with that of the ion exchange state, which had little effect on the differentiation of the rare earth elements. The iron–manganese oxide state was mainly enriched with cerium, and the content of cerium increased with the depth of the weathering crust. The iron–manganese oxide state was the main factor causing the phenomenon of the anomaly in the cerium content. Meanwhile, the iron oxides in the iron–manganese oxide state were mainly hematite and goethite. The organic binding state mainly beneficiated yttrium and cerium by complexation and certain adsorption. The content of elements found in the rare earth residual state was related to the degree of weathering and reflected the release sequence of rare earth elements in the mineralization process. Clarifying the rare earth occurrence states is conducive to better revealing the metallogenic regularity of weathered crust elution-deposited rare earth ores. In addition, the results can provide a valuable reference for expanding the available rare earth resources and the efficient comprehensive utilization of rare earth ore.

Using polarization-maintaining fiber (PMF) in dual-frequency heterodyne interferometry has the advantages of reducing the laser’s own drift, obtaining high-quality light spots, and improving thermal stability. Using only one single-mode PMF to achieve the transmission of dual-frequency orthogonal, linearly polarized beam requires angular alignment only once to realize the transmission of dual-frequency orthogonal, linearly polarized light, avoiding coupling inconsistency errors, so that it has the advantages of high efficiency and low cost. However, there are still many nonlinear influencing factors in this method, such as the ellipticity and non-orthogonality of the dual-frequency laser, the angular misalignment error of the PMF, and the influence of temperature on the output beam of the PMF. This paper uses the Jones matrix to innovatively construct an error analysis model for the heterodyne interferometry using one single-mode PMF, to realize the quantitative analysis of various nonlinear error influencing factors, and clarify that the main error source is the angular misalignment error of the PMF. For the first time, the simulation provides a goal for the optimization of the alignment scheme of the PMF and the improvement of the accuracy to the sub-nanometer level. In actual measurement, the angular misalignment error of the PMF needs to be smaller than 2.87° to achieve sub-nanometer interference accuracy, and smaller than 0.25° to make the influence smaller than ten picometers. It provides theoretical guidance and an effective means for improving the design of heterodyne interferometry instruments based on PMF and further reducing measurement errors.

Introduction: Volatile Organic Compounds (VOCs) are the most significant indoor pollutants that mainly emitted from various building materials and can do a great harm to human beings. The present study aims to the effect of different shapes of building materials on the emission characteristics of VOCs by using Computational Fluid Dynamics (CFD) simulation. Materials and methods: The indoor air flow is described by the continuity equation, the momentum equation and the standard k-ε model. The transport of VOCs in materials is described by a transient diffusion equation. ANSYS Fluent is used to solve the governing equations. Results: The emission of VOCs from planar building materials is validated based on the experimental data according to the literature. The numerical results have proved that the emission VOCs from hollow cylindrical building materials is faster than that of VOCs from planar building materials. The thinner the building materials and diffusion coefficient is bigger, the faster decrease rate of VOCs it is. Conclusion: The material shape has a significant impact on the emission of VOCs from building materials. When assessing indoor air quality, the realistic shape of materials should be used instead of the simplified planar materials.

BackgroundEosinophilic granulomatosis with polyangiitis (EGPA) is characterized by asthma-like attacks in its early stage, which is easily misdiagnosed as severe asthma. Therefore, new biomarkers for the early diagnosis of EGPA are needed, especially for differentiating the diagnosis of asthma.ObjectivesTo identify serum biomarkers that can be used for early diagnosis of EGPA and to distinguish EGPA from severe asthma.MethodData-independent acquisition (DIA) analysis was performed to identify 45 healthy controls (HC), severe asthma (S-A), and EGPA patients in a cohort to screen biomarkers for early diagnosis of EGPA and to differentiate asthma diagnosis. Subsequently, parallel reaction monitoring (PRM) analysis was applied to a validation cohort of 71 HC, S-A, and EGPA patients.ResultFour candidate biomarkers were identified from DIA and PRM analysis—i.e., serum amyloid A1 (SAA1), fibrinogen-α (FGA), and serum amyloid P component (SAP)—and were upregulated in the EGPA group, while cholesteryl ester transfer protein (CETP) was downregulated in the EGPA group compared with the S-A group. Receiver operating characteristics analysis shows that, as biomarkers for early diagnosis of EGPA, the combination of SAA1, FGA, and SAP has an area under the curve (AUC) of 0.947, a sensitivity of 82.35%, and a specificity of 100%. The combination of SAA1, FGA, SAP, and CETP as biomarkers for differential diagnosis of asthma had an AUC of 0.921, a sensitivity of 78.13%, and a specificity of 100%, which were all larger than single markers. Moreover, SAA1, FGA, and SAP were positively and CETP was negatively correlated with eosinophil count.ConclusionDIA-PRM combined analysis screened and validated four previously unexplored but potentially useful biomarkers for early diagnosis of EGPA and differential diagnosis of asthma.

This study aims to determine novel analytical lump solutions to the (2+1)-dimensional Hirota–Satsuma–Ito equation through conducting symbolic computations using the Hirota direct method. This nonlinear model describes the propagation of unidirectional shallow-water waves and interactions of two long waves with different dispersion forms. Some informative descriptions of physical behavior related to the solutions obtained in this article have also been included through several 3D figures and 2D contour plots. The acquired results in this research may be beneficial for better understanding the interaction phenomena of localized nonlinear waves in different research fields of nonlinear science. It is notable that the use of computer algebra in the calculations required by the article is inevitable. Accordingly, in this work, we have used the symbolic package Mathematica. Our results will be meaningful for the investigation of the future development of lump solitons in many physical systems.

Given the overlapping characteristics of poverty-stricken counties within key ecological function zones and ecologically fragile areas in China, this study takes the sample of 393 state-designated poverty-stricken counties from 2011 to 2018 and uses the differences-in-differences model to test the influence of forest resource protection on economic growth and its mechanism under the carbon neutrality target. The empirical research proves how to break the “green paradox” and “poverty curse” in the process of Chinese ecological civilization construction and targeted poverty alleviation and how policy tools integrate emission reductions with carbon-sequestration increase and achieve sustainable development. The results show that 1) the economic growth of poverty-stricken counties cannot be separated from industrialization, urbanization, and financial system; 2) China’s poverty-stricken counties have achieved an initial decoupling of carbon emissions from economic growth; 3) forest resource protection represented by forest coverage and stock changes the carbon emissions through emission reductions with carbon-sequestration increase and affect the economic growth of poverty-stricken counties; and 4) the reduction of total carbon emissions in poverty-stricken counties further enhances the role of forest resource protection in promoting county economic growth. Taking the 13th Five-Year Plan for eco-environment protection as the starting point, this research reveals different paths and impacts selected by Chinese local governments in poverty alleviation.

BackgroundMelanoma is a highly aggressive skin cancer with a poor prognosis and mortality. Immune checkpoint blockade (ICB) therapy (e.g., anti-PD-1 therapy) has opened a new horizon in melanoma treatment, but some patients present a non-responsive state. Cancer-associated fibroblasts (CAFs) make up the majority of stromal cells in the tumor microenvironment (TME) and have an important impact on the response to immunotherapy. There is still a lack of identification of CAFs-related predictors for anti-PD-1 therapy, although the establishment of immunotherapy biomarkers is well underway. This study aims to explore the potential CAFs-related gene panel for predicting the response to anti-PD-1 therapy in melanoma patients and elucidating their potential effect on TME.MethodsThree gene expression datasets from melanoma patients without anti-PD-1 treatment, in a total of 87 samples, were downloaded from Gene Expression Omnibus (GEO) as the discovery sets (GSE91061) and validation sets (GSE78220 and GSE122220). The CAFs-related module genes were identified from the discovery sets by weighted gene co-expression network analysis (WGCNA). Concurrently, we utilized differential gene analysis on the discovery set to obtain differentially expressed genes (DEGs). Then, CAFs-related key genes were screened with the intersection of CAFs-related module genes and DEGs, succeeded by supervised machine learning-based identification. As a consequence of expression analysis, gene set enrichment analysis, survival analysis, staging analysis, TME analysis, and correlation analysis, the multidimensional systematic characterizations of the key genes were uncovered. The diagnostic performance of the CAFs-related gene panel was assessed by receiver operating characteristic (ROC) curves in the validation sets. Eventually, the CAFs-related gene panel was verified by the expression from the single-cell analysis.ResultsThe six-gene panel associated with CAFs were finally identified for predicting the response to anti-PD-1 therapy, including CDK14, SYNPO2, TCF4, GJA1, CPXM1, and TFPI. The multigene panel demonstrated excellent combined diagnostic performance with the area under the curve of ROC reaching 90.5 and 75.4% ~100% in the discovery and validation sets, respectively.ConclusionConfirmed by clinical treatment outcomes, the identified CAFs-related genes can be used as a promising biomarker panel for prediction to anti-PD-1 therapy response, which may serve as new immunotherapeutic targets to improve survival outcomes of melanoma patients.

Abstract Colorectal cancer (CRC) is a common hereditary tumor that is often fatal. Its pathogenesis involves multiple genes, including circular RNAs (circRNAs). Notably, circRNAs constitute a new class of noncoding RNAs (ncRNAs) with a covalently closed loop structure and have been characterized as stable, conserved molecules that are abundantly expressed in tissue/development-specific patterns in eukaryotes. Based on accumulating evidence, circRNAs are aberrantly expressed in CRC tissues, cells, exosomes, and blood from patients with CRC. Moreover, numerous circRNAs have been identified as either oncogenes or tumor suppressors that mediate tumorigenesis, metastasis and chemoradiation resistance in CRC. Although the regulatory mechanisms of circRNA biogenesis and functions remain fairly elusive, interesting results have been obtained in studies investigating CRC. In particular, the expression of circRNAs in CRC is comprehensively modulated by multiple factors, such as splicing factors, transcription factors, specific enzymes and cis-acting elements. More importantly, circRNAs exert pivotal effects on CRC through various mechanisms, including acting as miRNA sponges or decoys, interacting with RNA binding proteins, and even translating functional peptides. Finally, circRNAs may serve as promising diagnostic and prognostic biomarkers and potential therapeutic targets in the clinical practice of CRC. In this review, we discuss the dysregulation, functions and clinical significance of circRNAs in CRC and further discuss the molecular mechanisms by which circRNAs exert their functions and how their expression is regulated. Based on this review, we hope to reveal the functions of circRNAs in the initiation and progression of cancer and highlight the future perspectives on strategies targeting circRNAs in cancer research.

Abstract Background A simple and accurate scoring system to guide perioperative blood transfusion in patients with coronary artery disease (CAD) undergoing cardiac surgery is lacking. The trigger point for blood transfusions for these patients may be different from existing transfusion guidelines. This study aimed to evaluate the safety and efficacy of a new scoring strategy for use in guiding transfusion decisions in patients with CAD. Methods A multicenter randomized controlled trial was conducted at three third-level grade-A hospitals from January 2015 to May 2018. Data of 254 patients in a Cardiac Peri-Operative Transfusion Trigger Score (cPOTTS) group and 246 patients in a group receiving conventional evaluation of the need for transfusion (conventional group) were analysed. The requirements for transfusion and the per capita consumption of red blood cells (RBCs) were compared between groups. Results Baseline characteristics of the two groups were comparable. Logistic regression analyses revealed no significant differences between the two groups in primary outcomes (1-year mortality and perioperative ischemic cardiac events), secondary outcomes (shock, infections, and renal impairment), ICU admission, and ICU stay duration. However, patients in the cPOTTS group had significantly shorter hospital stays, lower hospital costs, lower utilization rate and lower per capita consumption of transfused RBCs than controls. Stratified analyses revealed no significant differences between groups in associations between baseline characteristics and perioperative ischemic cardiac events, except for hemofiltration or dialysis and NYHA class in I. Conclusions This novel scoring system offered a practical and straightforward guideline of perioperative blood transfusion in patients with CAD. Trial registration chiCTR1800016561(2017/7/19).

Summary Here, we propose to develop microbiome‐based machine learning models to predict the response of biological wastewater treatment systems to environmental or operational disturbances or to design specific microbiomes to achieve a desired system function. These machine learning models can be used to enhance the stability of microbiome‐based biological systems and warn against the failure of these systems.

As a result of the difficulties brought by COVID-19 and its associated lockdowns, many individuals and companies have turned to robots in order to overcome the challenges of the pandemic. Compared with traditional human labor, robotic and autonomous systems have advantages such as an intrinsic immunity to the virus and an inability for human-robot-human spread of any disease-causing pathogens, though there are still many technical hurdles for the robotics industry to overcome. This survey comprehensively reviews over 200 reports covering robotic systems which have emerged or have been repurposed during the past several months, to provide insights to both academia and industry. In each chapter, we cover both the advantages and the challenges for each robot, finding that robotics systems are overall apt solutions for dealing with many of the problems brought on by COVID-19, including: diagnosis, screening, disinfection, surgery, telehealth, care, logistics, manufacturing and broader interpersonal problems unique to the lockdowns of the pandemic. By discussing the potential new robot capabilities and fields they applied to, we expect the robotics industry to take a leap forward due to this unexpected pandemic.

Vehicle exhaust prediction is of great importance for exhaust emissions control and public environment protection, and very challenging because it is influenced by various complex factors, including multi-component temporal dependencies (closeness, period, trend) and external factors, e.g., meteorological data and workday information. To address these issues, we propose a novel deep-learning-based framework, called multi-component fusion temporal networks (MCFT-Net), to collectively forecast emission concentration of carbon monoxide (CO). The proposed MCFT-Net consists of three parts, 1) a one-dimensional Convolution Neural Networks (1D-CNNs) module to capture temporal dependencies of vehicle emissions by merging closeness, period, and trend time sequences; 2) an artificial neural network (ANN) module to capture features of external factors, such as wind direction, weekday and weekend; and 3) a cascade fusion component to merge the temporal patterns, which are derived from temporal dependencies processed by multiple 1D-CNNs, and then assigned different weights according to an attention mechanism after fusion. The output of the attention operation is further combined with external features through sum and average operations to predict the final emission concentration. We evaluate the proposed MCFT-Net using two real-world vehicle exhaust datasets, ExhaustBMS and ExhaustHMS, which were collected from Beijing monitoring station (BMS) and Hefei monitoring station (HMS). The results demonstrate that our MCFT-Net outperforms 8 baseline methods, such as ARIMA, RNN, GRU, BiLSTM.

Banks’ primary goal is to gain profit for survival and to thrive. Therefore, they have to take various measures, such as data analysis, to maintain their sustainable competitiveness. Along with the rapid development of information technology, big data analytics capabilities (BDAC) is considered essential for banks in the highly dynamic market. To gain an in-depth understanding of the economic importance of BDAC in the banking sector in Malaysia, this research examines the relationship between BDAC and firm performance (i.e., market performance and operational performance) based on the resource-based view (RBV) and the contingent resource-based view (CRBV). The partial least squares structural equation modelling (PLS-SEM) was adopted to analyse the collected data from 162 bank managers in Malaysia. The findings verify that BDAC is composed of seven tangible/intangible resources and human skills, and it significantly influences firm performance in the banking sector.

Electrocardiogram (ECG) signals are among the significant physiological signals that indicate the essential properties of the human body. In recent years, the measurement of ECG signals has become more portable thanks to the increasing usage of wearable health testing technology. However, the enormous amount of signal data gathered over a long period of time does impose a heavy load on medical professionals. In addition, false alarms might occur due to the potential for the detected signal to become jumbled with noise and motion perturbations. Therefore, analyzing the quality of the measured raw ECG signal automatically is a valuable task. In this paper, we propose a new single-channel ECG signal quality assessment method that combines the Resnet network structure and the principle of self-attention to extract ECG signal features using the principle of similarity between individual QRS heartbeats within a time slice of ten seconds. In addition, an improved self-attention module is introduced into the deep neural network to learn the similarity between features. Finally, the network distinguishes between acceptable and unacceptable ECG segments. The model test results indicate that the F1-score can approach 0.954, which leads to a more accurate assessment of the ECG signal quality.

Circular RNAs (circRNAs) have been shown to be closely linked to the tumorigenesis and treatment response of hematological malignancies. However, the biological functions and clinical implications of circRNAs in acute myeloid leukemia (AML) remain largely unknown. CircRNA microarray datasets were analyzed to screen differentially expressed circRNAs in AML patients. It was found that circZBTB46 was significantly upregulated in AML patients and AML cells. Moreover, the expression of circZBTB46 was associated with the stages of AML patients and showed high sensitivity and specificity for diagnosing AML. Silencing of circZBTB46 inhibited AML cell proliferation and induced cell cycle arrest. Importantly, the depletion of circZBTB46 notably increased ferroptosis and enhanced RSL3-induced ferroptosis in AML cells. Mechanistically, circZBTB46 upregulated the expression of stearoyl-CoA desaturase 1 (SCD) possibly by acting as a miRNA sponge. Finally, the circZBTB46 knockdown repressed the tumor growth of AML in vivo. In conclusion, circZBTB46 protects AML cells from ferroptosis and promotes the proliferation by upregulating SCD, thus suggesting that circZBTB46 may be a potential therapeutic target for AML.

In order to study the best technology of remediation of petroleum contaminated soil, naphthalene (NAP), a typical semi volatile organic compounds in petroleum contaminated soil was selected as the target pollutant, the simulated NAP contaminated soil was taken as the test soil, and the microwave(f=2.45 GHz) was used to restore the soil. On the basis of single factor experiment, the Box-Behnken experiment design of response surface method was adopted, and the removal rate of NAP was taken as the response value. A quadratic multiple regression model was established with microwave power, irradiation time, soil thickness and soil moisture content as the influencing factors. The ANOVA model reached a very significant level, in which the microwave power had the most significant effect. The best process parameters were obtained as follows: the wave power was 768 W, the irradiation time was 19 min, the soil thickness was 3.2 cm, the water content was 16%, and the removal rate of NAP was 97.3% under these conditions. The verification results show that the removal rate of NAP is 97.0%, which is only 0.31% different from the predicted results. The process parameters optimized by response surface methodology are reasonable and feasible, which may provide some reference for the industrial application of microwave remediation of soil.

ObjectiveCurrent advances in immunotherapy requires accurate tumor sub-classification due to the heterogeneity of lung adenocarcinoma (LUAD). This study aimed to develop a LUAD sub-classification system based on immune cell signatures and identified prognostic gene markers.MethodsSignatures related to the prognosis of TCGA-LUAD and 4 GSE cohorts were screened and intersected from 184 previously published immune cell signatures. The LUAD samples in the TCGA were clustered by ConsensusClusterPlus. Molecular characteristics, immune characteristics and sensitivity to immunotherapies/chemotherapies were compared. LDA score was established through Linear Discriminant Analysis (LDA). Co-expression module was constructed by Weighted Gene Co-Expression Network Analysis (WGCNA).ResultsFour LUAD subtypes with different molecular and immune characteristics were identified. Significant differences in prognosis among the four subtypes were observed. The IS1 subtype with the worst prognosis showed the highest number of TMB, mutant genes, IFN γ score, angiogenesis score and immune score. Twenty co-expression modules were generated by WGCNA. Blue module, sky blue module and light yellow module were significantly correlated with LUAD prognosis. The hub genes (CCDC90B, ARNTL2, RIPK2, SMCO2 and ADA and NBN) showing great prognostic significance were identified from the blue module. A total of 8 hub genes (NLRC3, CLEC2D, GIMAP5, CXorf65, PARP15, AKNA, ZC3H12D, and ARRDC5) were found in the light yellow module. Except for CXorf65, the expression of the other seven genes were significantly correlated with LUAD prognosis.ConclusionThis study determined four LUAD subtypes with different molecular and immune characteristics and 13 genes closely related to the prognosis of LUAD. The current findings could help understand the heterogeneity of LUAD immune classes.

The composition/content of aroma compounds in tobacco leaves in the different producing areas varies too much, and it is very meaningful to develop advanced analysis techniques to investigate the composition/content-producing area correlation. Here, gas chromatography-ion mobility spectroscopy (GC-IMS) was used to analyze the composition/content of aroma compounds in tobacco samples from eight different producing areas. With this technique, ion mobility spectrum, differential ion mobility spectrum, and fingerprint spectrum were constructed for two-dimensional analysis. Then, the principal component analysis (PCA) and similarity analysis were performed for the eight tobacco samples. The results showed that the GC-IMS can detect 197 volatile aroma compounds in tobacco leaves and 75 of them are well identified. The fingerprint spectrum directly showed the difference in the composition/content of volatile aroma compounds in tobacco leaves from different producing areas. PCA and similarity analysis can clearly distinguish tobacco samples from different producing areas. This work demonstrated that the application of GC-IMS in analyzing the composition/content of aroma compounds in tobacco leaves is efficient. GC-IMS is a very powerful tool to give a direct and visual comparison of the composition/content of aroma compounds in tobacco leaves from different producing areas. The relationship between the composition/content of aroma compounds and producing areas could be established by this advanced technology. This work offers the possibility of planting or grading tobacco with different taste and smell more precisely in the future.

The research aims to explore the changes of national identity among Chinese international students in the odd social context of the global pandemic. By conducting semi-structured interviews with 10 Chinese undergraduate and postgraduate students in a prestigious university located in London, UK, the study provides evidence of Western neo-racism against Chinese students and the rise of Chinese nationalism. More significantly, it is found that Western neo-racism and Chinese nationalism have a push and pull effect on the national identity enhancement of Chinese international students. The participants revealed that bottom-up popular nationalism is more than a shadow of top-down state nationalism in China, and is more influential on students’ national identity formation. The research also discusses the implications of these findings, limitations and future research directions.

BackgroundFerroptosis is a newly defined form of programmed cell death that plays an important role in many cancers. However, ferroptosis-related lncRNAs (FRLs) involved in the regulation of colon cancer are not thoroughly understood. This study aimed to identify a prognostic FRL signature in colon cancer and explore its potential molecular function.MethodsRNA-seq data and relevant clinical information were obtained from The Cancer Genome Atlas (TCGA) database, and a list of ferroptosis-related genes was extracted from the FerrDb website. Analysis of differentially expressed FRLs was performed using the ‘limma’ package in R software. By implementing coexpression analysis and univariate Cox analysis, we then identified prognostic FRLs. Using Cox regression analysis with the least absolute shrinkage and selection operator (LASSO) algorithm, we constructed a prognostic model based on 4 FRLs. We evaluated the prognostic power of this model using Kaplan–Meier (K-M) survival curve analysis and receiver operating characteristic (ROC) curve analysis. Moreover, the relationships between the signature and immune landscape, somatic mutation and drug sensitivity were explored. Finally, in vitro experiments were conducted to validate the functions of AP003555.1 and AC000584.1.ResultsA 4-FRL signature was constructed. Two risk groups were classified based on the risk score calculated by this signature. The signature-based risk score exhibited a more powerful capacity for survival prediction than traditional clinicopathological features in colon patients. Additionally, we observed a significant difference in immune cells, such as CD4+ and CD8+ T cells and macrophages, between the two groups. Moreover, the high-risk group exhibited lower IC50 values for certain chemotherapy drugs, such as cisplatin, docetaxel, bleomycin or axitinib. Finally, the in vitro experiments showed that ferroptosis processes were suppressed after AP003555.1 and AC000584.1 knockdown.ConclusionThe proposed 4-FRL signature is a promising biomarker to predict clinical outcomes and therapeutic responses in colon cancer patients.

This study aims to investigate how young Chinese consumers make purchase intentions towards Western brands under the influence of two conflicting values and CSR, which is insufficiently discussed in the current literature. Both value-attitude-behavior (VAB) and consumer cultural theories are adopted to construct the research framework. Data was collected from undergraduate students studying at a public university located in Guangzhou via WeChat and Tencent QQ. A total of 314 usable responses were analyzed by the partial least squares structural equation modeling (PLS-SEM). The empirical findings indicated that cosmopolitanism has a significantly positive effect on brand attitudes and purchase intentions; ethnocentrism has a significantly negative effect on purchase intentions, but no significant impact on brand attitudes; and corporate social responsibility (CSR) initiatives positively affect brand attitudes rather than purchase intentions. The results also revealed that brand attitudes mediate the relationship between cosmopolitanism/CSR and purchase intentions, but it does not have a mediating effect on the relationship between ethnocentrism and purchase intentions. These findings provide essential insights to the body of knowledge of international marketing in emerging markets and shed light on understanding how young Chinese consumers make purchase decisions towards Western brands. The results are useful for Western brands to effectively adjust their marketing strategies and advertising/promoting campaigns for business development purposes in the Chinese market.

Cutaneous squamous cell carcinoma (CSCC) is a malignant proliferation of cutaneous epithelium that has been observed to have an alarming rise in incidence. Numerous studies have demonstrated microRNAs (miRNAs or miRs) as important biomarkers in the diagnosis, prognosis, and treatment of CSCC. This study aims to investigate the effects of miR-203 on the behaviors of CSCC cells and possible mechanisms associated with protein regulator of cytokinesis-1 (PRC1) and Wnt/β-catenin signaling pathway. PRC1 was suggested as a target of miR-203 in squamous cell carcinoma cell line 1 (SCL-1) cells by dual-luciferase reporter gene assay. Based on the immunohistochemical staining and qRT-PCR, PRC1 was abundantly expressed while miR-203 was poorly expressed in CSCC tissues. miR-203 mimic or inhibitor was transfected into SCL-1 cells to upregulate or downregulate its expression. Upregulation of miR-203 downregulated PRC1 expression to block the Wnt/β-catenin signaling pathway. By conducting 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT), scratch test, and Transwell and flow cytometric analyses, miR-203 was witnessed to restrain SCL-1 cell proliferation, migration, and invasion while accelerating their apoptosis. The rescue experiments addressed that inhibition of the Wnt/β-catenin signaling pathway conferred the anti-tumor effect of miR-203. These results establish a tumor-suppressive role for miR-203 in CSCC cell line SCL-1. Hence, miR-203 has promising potential as a therapeutic target for CSCC.

Abstract Background Circular RNAs (circRNAs) are a novel class of noncoding RNAs. Increasing evidence indicates that circRNAs play an important role in the occurrence and development of tumors. However, the role of circRNA hsa_circ_0044556 in the progression of colorectal cancer (CRC) remains unclear. Methods First, we searched for differentially expressed circRNAs using a circRNA microarray in paired CRC and adjacent normal tissues. The circRNA hsa_circ_0044556 was screened out from the existing CRC circRNA microarray in the Gene Expression Omnibus database and our microarray. The clinical significance of hsa_circ_0044556 expression level in CRC patients was then investigated. Finally, the functions of the targets of this circRNA were determined in CRC cell lines. Results Hsa_circ_0044556 was highly expressed in CRC patients and was positively correlated with tumor stage and lymph node metastasis. In CRC cell lines, the proliferation, migration, and invasion of cancer cells were inhibited by knocking down hsa_circ_0044556 expression. Conclusion Hsa_circ_0044556 promoted the progression of CRC. It is possible that hsa_circ_0044556 will become a novel biomarker or therapeutic target for CRC.

Synergistic strengthening is increasingly explored for developing high-performance metal matrix composites. In the present work, copper matrix composites reinforced with carbon nanotubes (CNTs) and TiB2 microparticles were fabricated via electroless deposition, powder metallurgy, hot extrusion, and water quenching. The synergistic strengthening effect of the nano-and micro-reinforcements was elaborated and analyzed based on experimental investigation and theoretical calculation. (1CNT-4TiB2)/Cu composite exhibited a better combination of the tensile strength of 375 MPa with elongation of 32.4% than other composites, representing 86.3%, 71.5%, and 22.8% strength increment in comparison with pure Cu, 4TiB2/Cu composite, and 1CNT/Cu composite, respectively. The strengthening efficiency of CNTs in (1CNT-4TiB2)/Cu composite is markedly higher than that in 1CNT/Cu composite (126.7 and 82.3, respectively), confirming the synergistic strengthening effect in enhancing the mechanical properties. Quantitative analysis and experimental characterization indicated that the thermal mismatch and load transfer mechanism of CNTs were the main factors contributing to the tensile strength enhancement of the composite. The synergistic strengthening effect was attributed to that TiB2 microparticles improved the homogeneous dispersion of CNTs, promoting the proportion of thermal mismatch and load transfer mechanisms. The present work may provide a new sight into improving the synergistic strengthening effect in metal matrix composites.

Abstract Background GATA binding protein 3 (GATA3) and miR-29b are related to colorectal cancer (CRC). The current study explored the regulatory relationship between GATA3 and miR-29b, and the mechanism of the two in the drug resistance of CRC cells to oxaliplatin. Method Apoptosis of CRC cells induced by oxaliplatin at various doses was detected by flow cytometry. CRC cells were separately transfected with overexpression and knockdown of GATA3, miR-29b agomir and antagomir, and treated by oxaliplatin to detect the cell viability and apoptosis by performing Cell Couting Kit-8 (CCK-8) and flow cytometry. The expression levels of GATA3, caspase3 and cleaved caspase3 were determined by Western blot, and the expression of miR-29b was detected by quantitative real-time polymerase chain reaction (qRT-PCR). Animal experiments were performed to examine the changes of transplanted tumors in nude mouse xenograft studies and observed by in vivo imaging. TUNEL staining was performed to detect tumor cell apoptosis. Result Both GATA3 and miR-29b agomir inhibited the activity of the CRC cells, promoted apoptosis and Cleaved caspase3 expression, and reduced the resistance of the cells to chemotherapy drug oxaliplatin. Although GATA3 could up-regulate miR-29b expression, the tumor-suppressive effect of GATA3 was partially reversed by miR-29b antagomir. In vivo experiments showed that down-regulating the expression of GATA3 promoted the growth rate and volume of transplanted tumors, while overexpressing GATA3 had no significant effect on tumor growth. TUNEL staining results showed that knocking down or overexpression of GATA3 did not cause significant changes to apoptotic bodies of CRC cells, while oxaliplatin treatment increased the number of apoptotic bodies. Conclusion GATA3 inhibits the cell viability of CRC cells, promotes apoptosis, and reduces oxaliplatin resistance of CRC cells through regulating miR-29b.

In this work, a new orthorhombic organic-inorganic hybrid single crystal (C2H10N2)2MnBr4.2Br with-Zero-dimensional structure was synthesized by slow evaporation method at 75 °C. The crystal structure and intermolecular interactions were performed by single crystal X-ray diffraction and Hirshfeld surface. Optical properties of (C2H10N2)2MnBr4.2Br were systematically studied by Raman, infrared spectrum, UV–vis, photoexcitation spectra, and photoluminescence spectra. The optical band gap (Eg = 2.61 eV) were calculated from the absorption spectra of (C2H10N2)2MnBr4.2Br. This hybrid materials also had shown good thermal stability (decomposition temperature: 288–660 °C). Finally, photoluminescence measurements showed a strong excitation line at 362 nm and a strong fluorescence at 537 nm which makes it a promising material in luminescence field. Natural tissues contain highly organized cellular architecture. One of the major challenges in tissue engineering is to develop engineered tissue constructs that promote cellular growth in physiological directionality. To address this issue, micro-patterned polydimethylsiloxane (PDMS) substrates have been widely used in cell sheet engineering due to their low microfabrication cost, higher stability, excellent biocompatibility, and most importantly, ability to guide cellular growth and patterning. However, the current methods for PDMS surface modification either require a complicated procedure or generate a non-uniform surface coating, leading to the production of poor-quality cell layers. A simple and efficient surface coating method is critically needed to improve the uniformity and quality of the generated cell layers. Herein, a fast, simple and inexpensive surface coating method was analyzed for its ability to uniformly coat polydopamine (PD) with or without collagen on micro-grated PDMS substrates without altering essential surface topographical features. Topographical feature, stiffness and cytotoxicity of these PD and/or collagen based surface coatings were further analyzed. Results showed that the PD-based coating method facilitated aligned and uniform cell growth, therefore holds great promise for cell sheet engineering as well as completely biological tissue biomanufacturing. Introduction: The rabbit brain model is commonly applied in neuroscience, including basic science, neuro medicine, and neural network, etc. The rabbit brain is more suitable than the mouse one for a microscopic neuroanatomy atlas study due to its suitable size and shape. Lateral ventricles are essential landmarks that enable a three-dimensional structure understanding of the rabbit cerebrum. Knowledge about how the nervous structures outside the neural tube amplify and contort around the lateral ventricles would help in the understanding of the rabbit cerebrum's three-dimensional organization. Materials & methods: Ten rabbits were used in this study. After fixation for at least two weeks, rabbits’ heads were dissected and micro-operative anatomy photographs were taken using the stereomicroscope camera. Results: All figures were divided into three parts: lateral ventricles body; choroid fissure and the thalamus-cerebrum space; partition and connection between the left and right lateral ventricles. Under the stereoscopic microscope, it can be observed that: rabbit cerebrum left and right lateral ventricles were wrapped by the grey and white matter layers; hippocampal formation, relatively older cortical structures in the cerebrum, coiled inward by newer cortex; the curly edges of the cortex made the choroid fissure, namely the potential opening between the lateral ventricles (left and right) and cerebrum surface. The three-dimensional structure atlas also showed cortex folding and coil and that cortex with the hippocampal formation was centered in the thalamus, while the thalamus was in a direct extension relationship with the cortex-hippocampus formation. Conclusion: The relationship between the thalamus and the cortex-hippocampus formation should be noticed in neural network research as it suggests that the thalamus may be at the core of the cerebrum network. If we could have a correct understanding of rabbit cerebrum three-dimensional structure via microscopic anatomical atlas under the microscope before the section research, we would be able to assembly the local neural network structures in the brain constructed by section research into a complete cerebrum network properly.

Objective: The present study aims to investigate the incidence and predictors of atrial high-rate events (AHREs) in patients with permanent pacemaker implants.Methods: A total of 289 patients who were implanted with a dual-chamber pacemaker due to complete atrioventricular block or symptomatic sick sinus syndrome (SSS) and had no previous history of atrial fibrillation were included in the present study. AHREs are defined as events with an atrial frequency of ≥175 bpm and a duration of ≥5 min. The patients were divided into two groups according to whether or not AHREs were detected during the follow-up: group A (AHRE+, n = 91) and group N (AHRE–, n = 198).Results: During the 12-month follow-up period, AHREs were detected in 91 patients (31.5%). The multivariate Cox regression analysis revealed that patient age [odds ratio [OR] = 1.041; 95% confidence interval [CI], 1.018–1.064; and P < 0.001], pacemaker implantation due to symptomatic SSS (OR = 2.225; 95% CI, 1.227–4.036; and P = 0.008), and the percentage of atrial pacing after pacemaker implantation (OR = 1.010; 95% CI, 1.002–1.017; and P = 0.016) were independent AHRE predictors.Conclusion: The AHRE detection rate in patients with pacemaker implants was 31.5%. Patient age, pacemaker implantation due to symptomatic SSS, and the percentage of atrial pacing after pacemaker implantation were independent AHRE predictors.

Transcription factors are frequently aberrantly reactivated in various cancers, including colorectal cancer (CRC). However, as a transcription factor, the role of GMEB2 in cancer is still unclear, and further studies are needed. Here, we aimed to identify the function and mechanism of GMEB2 in regulating the malignant progression of CRC. GMEB2 was found to be highly expressed in online data analyses. We demonstrated that GMEB2 was markedly upregulated at both the mRNA and protein levels in CRC cells and tissues. GMEB2 knockdown inhibited CRC cell growth in vitro and in vivo. Mechanistically, as a transcription factor, GMEB2 transactivated the ADRM1 promoter to increase its transcription. Rescue experiments showed that ADRM1 downregulation partially reversed the promoting effects of GMEB2 on CRC growth in vitro. Moreover, the GMEB2/ADRM1 axis induced nuclear translocation of NF-κB, thus activating NF-κB signalling. Finally, we further revealed that YTHDF1 recognized and bound to the m6A site on GMEB2 mRNA, which enhanced its stability. Taken together, our findings reveal the crucial role and regulatory mechanism of GMEB2 in CRC for the first time and provide a novel potential therapeutic target for CRC therapy.

Background: Colorectal cancer (CRC) is one of the most commonly diagnosed cancers with high incidence and poor prognosis worldwide. Circ_RNF13 is upregulated in CRC; however, the biological roles and downstream signaling of circ_RNF13 remain undefined. Methods: The characterization of circ_RNF13 was determined by Sanger sequencing, qRT-PCR, subcellular fractionation assay, and RNA FISH. Western blot analysis and qRT-PCR were employed to detect the expression of the key molecules and stemness markers in CRC tumor samples and cells. The stem-like activities of CRC cells were assessed by sphere formation assay, flow cytometry, and immunofluorescence (IF). Cell viability was monitored by CCK-8 assay. The chemosensitivity of CRC cells was assessed by colony formation and cell apoptosis assays. Bioinformatics analysis, RIP assay, RNA pull-down assay, and FISH/IF staining were used to detect the association between circ_RNF13 and TRIM24. The transcriptional regulation of DDX27 was investigated by ChIP assay, and the post-translational regulation of TRIM24 was detected by Co-IP. The in vitro findings were verified in a xenograft model. Results: circ_RNF13 and DDX27 were elevated in CRC tumor samples and cells. Knockdown of circ_RNF13 or DDX27 inhibited stemness and increased chemosensitivity in CRC cells. Mechanistically, circ_RNF13 regulated DDX27 expression via TRIM24-mediated transcriptional regulation, and circ_RNF13 stabilized TRIM24 via suppressing FBXW7-mediated TRIM24 degradation. In vivo studies revealed that the knockdown of circ_RNF13 impaired stemness and enhanced the chemosensitivity of CRC in the xenograft model. Conclusion: circ_RNF13 regulated the stemness and chemosensitivity of CRC by transcriptional regulation of DDX27 mediated by TRIM24 stabilization.

Currently, using semantic segmentation networks to distinguish dynamic and static key points has become a mainstream designing method for semantic SLAM systems. However, the semantic SLAM systems must have prior semantic knowledge of relevant dynamic objects, and their processing speed is inversely proportional to the recognition accuracy. To simultaneously enhance the speed and accuracy for recognizing dynamic objects in different environments, a novel SLAM system without prior semantics called DGFlow-SLAM is proposed in this paper. A novel grid segmentation method is used in the system to segment the scene flow, and then an adaptive threshold method is used to roughly detect the dynamic objects. Based on this, a deep mean clustering segmentation method is applied to find potential dynamic targets. Finally, the results of grid segmentation and depth mean clustering segmentation are jointly used to find moving objects accurately, and all the feature points of the moving objects are removed on the premise of retaining the static part of the moving object. The experimental results show that on the dynamic sequence dataset of TUM RGB-D, compared with the DynaSLAM system with the highest accuracy for detecting moderate and violent motion and the DS-SLAM with the highest accuracy for detecting slight motion, DGflow-SLAM obtains similar accuracy results and improves the accuracy by 7.5%. In addition, DGflow-SLAM is 10 times and 1.27 times faster than DynaSLAM and DS-SLAM, respectively.

In this paper, we address the finite-time stability problem of Caputo nabla fractional-order switched linear systems with α∈(0,1). Firstly, the monotonicity of the discrete Mittag-Leffler function is proposed. Secondly, under the per-designed switching rules, the form of the solution for Caputo nabla fractional-order switched linear systems is obtained by using the discrete unit step function. On the above basis, some sufficient conditions of finite-time stability for Caputo nabla fractional-order switched linear systems are proposed, according to the discrete Grönwall inequality and the monotonicity of the discrete Mittag-Leffler function. Finally, simulation verification is carried out via three numerical examples.

Superhydrophobic films have been successfully prepared on various substrates by finely controlling surface micro/nanostructure's roughness and reducing surface energy. However, there is no unified conclusion about the correlation of surface roughness and 3D volume parameters to wettability. Herein, we fabricated a superhydrophobic film composed of hierarchical Cu cone-flowers via facile one-step pulse electrodeposition with a 30% duty cycle for 20 min. Based on this film, the relationship between roughness/volume parameters and hydrophobicity was well studied. The resulting cone-flower film exhibits excellent corrosion resistance and self-cleaning properties due to a high water contact angle (WCA) of ~160° and a low sliding angle (SA) of ~3°. The large roughness skewness (Rsk = 0.7–1.3) and void volume ratio (Vvc/Vmc > 1.5) are shown to be more desirable to achieve superhydrophobic surfaces. Moreover, a new hydrophobic model is proposed to explain the above results, which means that the nanoscale air cushions share most of the gravity of water droplets to help microscale air cushions closer to the Cassie state. Further experiments by adding or destroying the nanostructures to observe the change of hydrophobicity well verified our conjecture.

Regardless of its good electron-transfer ability and chemical stability, pure Zn2SnO4 (ZSO) still has intrinsic deficiencies of a narrow spectral response region, poor absorption ability, and high photo-activated carrier recombination rate. Aiming to overcome the deficiencies above-mentioned, we designed a facile hydrothermal route for etching ZSO nanoparticles in a dilute acetic acid solution, through which efficient oxygen vacancy defect engineering was accomplished and SnO2−x nanocrystals were obtained with an ultrafine particle size. In comparison with the untreated ZSO nanoparticles, the specific surface area of SnO2−x nanocrystals was substantially enlarged, subsequently leading to the notable augmentation of active sites for the photo-degradation reaction. Aside from the above, it is worth noting that SnO2−x nanocrystals were endowed with a broad spectral response, enhancing light absorption capacity and the photo-activated carrier transfer rate with the aid of oxygen vacancy defect engineering. Accordingly, SnO2−x nanocrystals exhibited significantly enhanced photoactivity toward the degradation of the organic dye rhodamine B (RhB), which could be imputed to the synergistic effect of increasing active sites, intensified visible-light harvesting, and the separation rate of the photo-activated charge carrier caused by the oxygen vacancy defect engineering. In addition, these findings will inspire us to open up a novel pathway to design and prepare oxide compound photocatalysts modified by oxygen vacancy defects in pursuing excellent visible-light photoactivity.

Materials that can be designed with programmable properties and which change in response to external stimuli are of great importance in numerous fields of soft actuators, involving robotics, drug delivery and aerospace applications. In order to improve the interaction of human and robots, materials with variable stiffness are introduced to develop their compliance. A variable stiffness composite has been investigated in this paper, which is composed of liquid metals (LMs) and silicone elastomers. The phase changing materials (LMs) have been encapsulated into silicone elastomer by printing the dual materials alternately with three-dimensional direct ink writing. Such composites enable the control over their own stiffness between soft and rigid states through LM effective phase transition. The tested splines demonstrated that the stiffness changes approximately exceeded 1900%, and the storage modulus is 4.75 MPa and 0.2 MPa when LM is rigid and soft, respectively. In the process of heating up, the stretching strain can be enlarged by at least three times, but the load capacity is weakened. At a high temperature, the resistance of the conductive composites changes with the deformation degree, which is expected to be applied in the field of soft sensing actuators.

KATNAL2 mutations have been associated with autism spectrum disorder (ASD) and other related neurodevelopmental disorders (NDDs) such as intellectual disability (ID) in several cohorts. KATNAL2 has been implicated in brain development, as it is required for ciliogenesis in Xenopus and is required for dendritic arborization in mice. However, a causative relationship between the disruption of Katnal2 function and behavioral defects has not been established. Here, we generated a katnal2 null allele in zebrafish using CRISPR/Cas9-mediated genome editing and carried out morphological and behavioral characterizations. We observed that katnal2-/- embryos displayed delayed embryonic development especially during the convergence and extension (CE) movement. The hatched larvae showed reduced brain size and body length. In the behavioral tests, the katnal2-/- zebrafish exhibited reduced locomotor activity both in larvae and adults; increased nocturnal waking activity in larvae; and enhanced anxiety-like behavior, impaired social interaction, and reduced social cohesion in adults. These findings indicate an important role for katnal2 in development and behavior, providing an in vivo model to study the mechanisms underlying the ASD related to KATNAL2 mutations.

MicroRNAs (miRNAs) and their target genes have been shown to play an important role in gastric cancer but have not been fully clarified. Therefore, our goal was to identify the key miRNA–mRNA regulatory network in gastric cancer by utilizing a variety of bioinformatics analyses and experiments. A total of 242 miRNAs and 1080 genes were screened from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO), respectively. Then, survival-related differentially expressed miRNAs and their differentially expressed target genes were screened. Twenty hub genes were identified from their protein–protein interaction network. After weighted gene co-expression network analysis was conducted, we selected miR-137-3p and its target gene, COL5A1, for further research. We found that miR-137-3p was significantly downregulated and that overexpression of miR-137-3p suppressed the proliferation, invasion, and migration of gastric cancer cells. Furthermore, we found that its target gene, COL5A1, could regulate the expression of another hub gene, FSTL1, by sponging miR-137-3p, which was confirmed by dual-luciferase reporter assays. Knockdown of COL5A1 inhibited the proliferation, invasion, and migration of gastric cancer cells, which could be rescued by the miR-137-3p inhibitor or overexpression of FSTL1. Ultimately, bioinformatics analyses showed that the expression of FSTL1 was highly correlated with immune infiltration.
