Abstract

Laser nitriding is used for the fast and easy production of nitride coatings on iron and alloys. Here, first results of the laser nitriding process applied to stainless steel are reported. The laser treatment led to the appearance of additional lines in the Mossbauer spectra, which are attributed to γ-Fe(N) produced by the laser nitriding process. The Mossbauer results are discussed in connection with the results obtained from X-ray diffraction and resonant nuclear reaction analysis. Furthermore, the results of isochronical annealing treatments of laser nitrided iron are reported.

Abstract

We report on a search for second generation leptoquarks (LQ) produced in p (p) over bar collisions at root s = 1.8 TeV using the DO detector at Fermilab. Second generation leptoquarks are assumed to be produced in pairs and to decay to either mu. or nu and either a strange or a charm quark (q). Limits an placed on sigma(p (p) over bar --> LQ --> mu nu + jets) as a function of the mass of the leptoquark. For equal branching ratios to mu q and nu q, second generation scalar leptoquarks with a mass below 160 GeV/c(2), vector leptoquarks with anomalous minimal vector couplings with a mass below 240 GeV/c(2), and vector leptoquarks with Yang-Mills couplings with a mass below 290 GeV/c(2), are excluded at the 95% confidence level.

Abstract

Using the DZero detector at the 1.8 TeV pbarp Fermilab Tevatron collider, we have measured the inclusive dijet mass spectrum in the central pseudorapidity region |eta_jet| < 1.0 for dijet masses greater than 200 Gev/c^2. We have also measured the ratio of spectra sigma(|eta_jet| < 0.5)/sigma(0.5 < |eta_jet| < 1.0). The order alpha_s^3 QCD predictions are in good agreement with the data and we rule out models of quark compositeness with a contact interaction scale < 2.4 TeV at the 95% confidence level.

Abstract

We search for new physics using events with one high transverse energy photon, two or more jets, and an apparent imbalance in transverse energy, in p{ovr p} collisions at the Fermilab Tevatron at {radical} (s) =1.8 TeV . Such events are predicted for production of supersymmetric particles in some models. No excess is observed beyond expected background. For the parameter space of the minimal supersymmetric standard model with branching fraction B({tilde {chi}}{sup 0}{sub 2}{r_arrow}{gamma}thinsp{tilde {chi}}{sup 0}{sub 1})=1 and m{sub {tilde {chi}}{sup 0}{sub 2 }}{minus}m{sub {tilde {chi}}{sup 0}{sub 1}}{gt}20 GeV , we obtain a 95{percent} confidence level lower mass limit of 310thinspthinspGeV for equal mass squarks and gluinos. Abstract

We report on a search for $\mathrm{charge}\ensuremath{-}1/3$ third-generation leptoquarks (LQ) produced in $p\overline{p}$ collisions at $\ensuremath{\surd}s\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}1.8\mathrm{TeV}$ using the $\mathrm{D}0$ detector at Fermilab. Third-generation leptoquarks are assumed to be produced in pairs and to decay to a tau neutrino and a $b$ quark with branching fraction $B$. We place upper limits on $\ensuremath{\sigma}(p\overline{p}\ensuremath{\rightarrow}\mathrm{LQ}\overline{\mathrm{LQ}}){B}^{2}$ as a function of the leptoquark mass ${M}_{\mathrm{LQ}}$. Assuming $B\phantom{\rule{0ex}{0ex}}=\phantom{\rule{0ex}{0ex}}1$, we exclude at the 95% confidence level third-generation scalar leptoquarks with ${M}_{\mathrm{LQ}}l94\mathrm{GeV}{/c}^{2}$, and third-generation vector leptoquarks with ${M}_{\mathrm{LQ}}l216\mathrm{GeV}{/c}^{2}({M}_{\mathrm{LQ}}l148\mathrm{GeV}{/c}^{2}$) assuming Yang-Mills (anomalous) coupling.

Abstract

The shape of the transverse momentum distribution of W bosons (p_T(W)) produced in pbarp collisions at sqrt(s)= 1.8 TeV is measured with the DO detector at Fermilab. The result is compared to QCD perturbative and resummation calculations over the p_T(W) range from 0-200 GeV/c. The shape of the distribution is consistent with the theoretical prediction.

Abstract

We report a measurement of the W boson mass based on an integrated luminosity of 82 pb −1 from pp collisions at √s = 1.8 TeV recorded in 1994-1995 by the D0 detector at the Fermilab Tevatron. We identify W bosons by their decays to e∨ V and extract the mass by fitting the transverse mass spectrum from 28 323 W boson candidates. A sample of 3563 dielectron events, mostly due to Z→ee decays, constrains models of W boson production and the detector. We measure M W = 80.44±0.10(stat)±0.07(syst) GeV. By combining this measurement with our result form the 1992-1993 data set, we obtain M W = 80.43±0.11 GeV.

Abstract

We have searched for first generation scalar leptoquark (LQ) pairs in the enu+jets channel using ppbar collider data (integrated luminosity= 115 pb^-1) collected by the DZero experiment at the Fermilab Tevatron during 1992-96. The analysis yields no candidate events. We combine the results with those from the ee+jets and nunu+jets channels to obtain 95% confidence level (CL) upper limits on the LQ pair production cross section as a function of mass and of beta, the branching fraction to a charged lepton. Comparing with the next-to-leading order theory, we set 95% CL lower limits on the LQ mass of 225, 204, and 79 GeV/c^2 for beta=1, 1/2, and 0, respectively.

Abstract

We have searched for the flavor-changing neutral current decay b -> s mu+ mu- in p pbar collisions at sqrt(s) = 1.8 TeV with the DO detector at Fermilab. We determine the 90% confidence level limit for the branching fraction to be B(b -> X_s mu+ mu-) < 3.2 x 10^{-4}. We argue that this limit is more stringent than the best published limit on this decay rate.

Abstract

The effect of the nitrogen take-up upon irradiation of iron or steel with excimer laser pulses in air or nitrogen atmosphere is well established. The resulting nitrogen depth profiles and phase compositions were measured by a combination of Rutherford backscattering spectrometry, resonant nuclear reaction analysis and conversion electron Mossbauer spectroscopy for various laser parameters. The accuracy of the nitrogen profiling via the 15 N(p, xy) 12 C reaction allowed the detection of very low nitrogen concentrations and also the resolving of them laterally across the laser spot. The laser-induced changes in surface topography were observed with optical microscopy and surface profilometry. The microhardness across the laser spot and as a function of the depth is compared with the nitrogen depth and lateral profiles and the phase composition. Thus the mechanical properties can be related to the microstructure and phase composition in the laser modified surfaces of iron and steel. A comparison is made between the effects for irradiation of pure iron and the carbon steel C80.

Abstract

Neurons and macroglia share the common, polarizing, morphological feature of multiple processes extending from a cell body, thereby defining two cellular domains. Frequently, specialized cellular activities occur within these processes, such as the dendrites of neurons and the myelin sheath of oligodendrocytes, which serve to define some of the functions of the cell. As a consequence, molecules involved in carrying out these functions need to be targeted to these domains, and mechanisms must exist for selecting and delivering these molecules to their appropriate locations. One mechanism that is emerging as increasingly important in targeting proteins to distal processes of neural cells is the translocation of the mRNAs encoding those proteins. In this review, we present many examples of such translocated mRNAs in neurons, astrocytes, and oligodendrocytes. There is a growing consensus that four major steps occur in mRNA targeting after transcription and exit of these molecules from the nucleus. These include 1) the assembly of mRNA into an RNA-protein granule, presumably around some translocation signal within the mRNA; 2) transport of the mRNA granule complex to distal sites via the cytoskeleton; 3) anchoring of the granule at the targeting site; and 4) translation of the localized mRNA to generate protein products in situ. It has become increasingly apparent that mRNA translocation is an active process, although many of the components of the translocation apparatus remain to be identified. Recent evidence also indicates that a number of factors can regulate the transport of mRNAs from within and without the cell. These include cell-cell contact, differentiation state, electrical activity, and trophic factors, which seem to exert their influence through signal transduction mechanisms that are only beginning to be defined. 64. Postnatal localization and morphogenesis of cells expressing the dopaminergic D2 receptor gene in rat brain: Expression in non-neuronal cells

Abstract

The cellular localization of the dopaminergic D2 receptor (D2R) mRNA and protein was determined during postnatal development, from birth to 35 days, in the rat neostriatum by in situ hybridization histochemistry and immunohistochemistry. To localize and identify more precisely the morphology of cells expressing the D2R mRNA, nonradioactive, digoxigenin in situ hybridization was performed. Throughout this period of development, D2R mRNA and protein were widely expressed by neostriatal cells, adjoining forebrain cells and small cellular processes. Within morphologically identifiable neurons, the expression of the D2 receptor appeared to occur after cell division ceased. D2R gene expression appeared during neuronal migration and followed the developmental pattern of neuronal settling within the neostriatum. Both D2R mRNA and protein appeared to colocalize in neostriatal cells and the labeling of both appeared to accumulate within the cells progressively with age. The structural phenotypes of neostriatal neurons bearing D2R mRNA and protein were diverse throughout postnatal development. The most frequently stained cells were a heterogeneous group of medium spiny and aspiny neurons. Large cells corresponding to aspiny neurons were less frequently stained. Both phenotypes exhibited considerable postnatal growth of their cell bodies. In addition to neurons, other cell types were also observed to express the D2R mRNA and protein over the developmental period studied. These other cells included patches of ciliated ependymal cells lining the lateral ventricles and many interfascicular oligodendroglia of forebrain fiber tracts. These results demonstrate the unexpected expression of the dopaminergic D2 receptor in non-neuronal cells within the brain. They provide a novel morphologic suggestion that the dopaminergic D2 receptor may support unrecognized, nonsynaptic functions in specific non-neuronal cell populations in the nervous system. J. Comp. Abstract

The effect of the nitrogen take‐up upon irradiation of iron or steel with excimer laser pulses in air or in nitrogen atmosphere is well established. The resulting phase compositions and nitrogen depth profiles were measured by a combination of simultaneous Conversion Electron Mossbauer Spectroscopy (CEMS), Conversion X‐ray Mossbauer Spectroscopy (CXMS), and Resonant Nuclear Reaction Analysis (RNRA) as a function of the nitrogen gas pressure during irradiation. A maximum nitrogen content and a maximum fraction of the ɛ-nitride was found at 0.1 MPa. This result is in accordance with hardness measurements performed by the nanoindentation technique.

Abstract

We have searched for the pair production of first generation scalar leptoquarks in the $\mathrm{eejj}$ channel using the full data set $(123{\mathrm{pb}}^{\ensuremath{-}1})$ collected with the D0 detector at the Fermilab Tevatron during 1992--1996. We observe no candidates with an expected background of approximately 0.4 events. Comparing the experimental 95% confidence level upper limit to theoretical calculations of the cross section with the assumption of a 100% branching fraction of $\mathrm{eq}$, we set a lower limit on the mass of a first generation scalar leptoquark of $225\mathrm{GeV}{/c}^{2}$. The results of this analysis rule out the interpretation of the excess of high ${Q}^{2}$ events at DESY HERA as leptoquarks which decay exclusively to $\mathrm{eq}$.

Abstract

The Golli-myelin basic protein (MBP) transcription unit gives rise to two sets of products. One set (i.e., the MBPs) is expressed exclusively in myelin forming cells and the other set (i.e., the golli isoforms) is expressed in both oligodendrocytes and neurons in the CNS. The two major golli proteins, generated from RNAs transcribed from the most upstream promoter of the gene, contain MBP peptide sequences in their C-terminal halves and are, therefore, structurally and immunologically related to the MBPs. We have examined the distribution and localization of golli proteins in the mouse peripheral nervous system (PNS) using immunocytochemistry with a golli-specific antibody. Golli immunoreactivity was first observed in sensory and motor fibers of the mouse at E11 during fiber tract extension, but prior to the maturation of terminal connections. Once neuromuscular junctions had formed, golli immunoreactivity appeared in motor endplates and persisted to the latest age examined, P60. Golli immunoreactivity was also observed in the cell bodies and processes of the dorsal root ganglia throughout development. Strong staining in the PNS of the dysmyelinating mutant shiverer suggested that the major golli protein in peripheral fibers was the BG21 isoform. Interestingly, golli immunoreactivity was also found in adrenal chromaffin cells, which share a common neural crest derivation with other postganglionic neurons that express golli protein. These results suggest that in addition to its role in early forming neuronal systems of the CNS, golli protein also plays a role in the early development and maintenance of neurons in the PNS. J. Abstract

We present a measurement of the ttbar production cross section in ppbar collisions at root(s) = 1.8TeV by the D0 experiment at the Fermilab Tevatron. The measurement is based on data from an integrated luminosity of approximately 125 pb^-1 accumulated during the 1992-1996 collider run. We observe 39 ttbar candidate events in the dilepton and lepton+jets decay channels with an expected background of 13.7+-2.2 events. For a top quark mass of 173.3GeV/c^2, we measure the ttbar production cross section to be 5.5+-1.8 pb.

Abstract

The myelin basic protein (MBP) gene locus is composed of two overlapping transcription units that share all of the MBP exons. One of these transcription units expresses the MBPs and the other expresses a family of proteins structurally related to the MBPs. This second transcription unit is called the Golli gene, and the entire complex is called the Golli-mbp gene. In this study, the expression of the Golli gene was examined in the human fetal central nervous system (CNS). By using reverse transcriptase-polymerase chain reaction cloning we have identified eight new members of the Golli gene family of transcripts expressed in the human CNS. Golli gene expression was examined by in situ hybridization and immunohistochemistry, and surprisingly, Golli products were found to be expressed in neurons as well as oligodendrocytes. Furthermore, the subcellular distribution of Golli immunoreactivity in fetal spinal cord interneurons shifted between the various laminae. Golli protein was localized within the nuclei of interneurons in the posterior horn, but was found in the cell bodies and processes of interneurons in the anterior horn. Within oligodendrocytes, Golli protein was detected in the cell bodies and processes, including processes which were wrapping axonal segments. Golli mRNA expression was also observed in neurons within the cerebral cortex between 18 and 20 weeks postconception, prior to myelination of this brain region. During this period, there was a striking developmental increase in the numbers and in the locations of neurons expressing Golli mRNAs within the cortical plate. The diverse distribution of Golli proteins within neurons and oligodendrocytes indicates that their function is quite different from that of the MBPs to which they are closely related.

Abstract

Considering the hypothesis that androgen-independent but growth factor dependent epithelial cell division may be important in the development and progression of prostate cancer and that protein tyrosine kinases and phosphotyrosine protein phosphatases are key enzymes modulating the levels of specific phosphotyrosylated proteins implicated in several growth factor regulated signal transduction pathways, our aim was to study the cellular distribution of phosphotyrosine proteins in normal and hyperplastic dog prostates as well as in those of castrated dogs supplemented with either androgens or estrogens in order to modify the relative proportion of basal versus secretory epithelial cells.Following the determination of optimal conditions to specifically detect phosphotyrosine proteins by a rabbit polyclonal antibody directed against phosphotyrosine, immunohistochemistry was performed on prostate tissue sections from these experimental animals. In addition to morphological criteria, an antibody to high molecular weight cytokeratins and antisera against arginine esterase were used to selectively identify basal and secretory cells. Since prostatic acid phosphatase may be involved in the local regulation of phosphotyrosine proteins, its distribution was also evaluated with a human prostatic acid phosphatase antiserum.In all prostatic tissues examined, basal epithelial cells were preferentially and specifically stained with antiphosphotyrosine. The staining intensity per basal cell was highest in the estrogen-supplemented dogs. In addition, basal cells were numerically increased and all were highly immunoreactive for high molecular weight cytokeratins. In prostates displaying a well-differentiated glandular epithelium, the number of positive basal cells and their staining intensity varied in the following order: normalhyperplasticandrogen-supplemented dogs. At all times, the levels of phosphotyrosine proteins in prostatic acid phosphatase and arginine esterase positive cells (secretory) remained low. Fibroblasts and smooth muscle cells were unreactive to antiphosphotyrosine, even though estrogen supplementation increased the prostatic stromal volume.The preferential localization of phosphotyrosine proteins in basal cells, their increased level per cell and the number of positive cells in the different experimental animals support the concept that basal cells represent the stem cells of the prostate. The sex steroid-mediated up-regulation of protein phosphorylation on tyrosine residues in these cells suggests that their proliferation is likely to involve growth factor regulated signal transduction pathways. In this respect, the lack of maturation of basal cells and the differentiation of secretory cells induced by androgen deprivation, combined with estrogen stimulation, favors the activation of these pathways and cell growth. On the other hand, the activation of glandular cell differentiation and the increase of stromal volume do not alter the threshold level of protein tyrosine kinase and phosphatase activities in secretory cells, fibroblasts and smooth muscle cells.

Abstract

Five immortalized oligodendrocyte cell lines, representing a spectrum of different stages of oligodendrocyte maturation, were transplanted into neonatal shiverer mouse brains and examined for their ability to survive, multiply, and migrate in vivo. Each of the cell lines migrated to different regions of the brain with remarkable consistency when injected into the mouse forebrain, suggesting that the cells might be responding to different environmental cues present in the neonatal mouse brain. These results are consistent with the fact that cells at different stages in the oligodendrocyte lineage probably possess different sets of surface molecules and receptors. Significant differences were also observed in the survival and proliferation of the lines examined, even when the lines tested had quite similar in vitro phenotypes. Interestingly, the cell line with the most mature in vitro phenotype, N20.1, appeared to elaborate membranous processes when transplanted into the brain, reminiscent of oligodendrocytes ensheathing axonal segments. The experiments suggest that these immortalized cells could be useful models to study the cellular and molecular mechanisms involved in the development, maturation and possibly formation of myelin by oligodendrocytes in the mammalian brain.

Abstract

The astrocyte brain fatty acid binding protein (Fabp7) has previously been shown to have a coordinated diurnal regulation of mRNA and protein throughout mouse brain, and an age-dependent decline in protein expression within synaptoneurosomal fractions. Mechanisms that control time-of-day changes in expression and trafficking Fabp7 to the perisynaptic process are not known. In this study, we confirmed an enrichment of Fabp7 mRNA and protein in the astrocytic perisynaptic compartment, and observed a diurnal change in the intracellular distribution of Fabp7 mRNA in molecular layers of hippocampus. Northern blotting revealed a coordinated time-of-day-dependent oscillation for the Fabp7 mRNA poly(A) tail throughout murine brain. Cytoplasmic polyadenylation element-binding protein 1 (CPEB1) regulates subcellular trafficking and translation of synaptic plasticity-related mRNAs. Here we show that Fabp7 mRNA coimmunoprecipitated with CPEB1 from primary mouse astrocyte extracts, and its 3′UTR contains phylogenetically conserved cytoplasmic polyadenylation elements (CPEs) capable of regulating translation of reporter mRNAs duringXenopusoocyte maturation. Given that Fabp7 expression is confined to astrocytes and neural progenitors in adult mouse brain, the synchronized cycling pattern of Fabp7 mRNA is a novel discovery among known CPE-regulated transcripts. These results implicate circadian, sleep, and/or metabolic control of CPEB-mediated subcellular trafficking and localized translation of Fabp7 mRNA in the tripartite synapse of mammalian brain.

Abstract

Recent transparency reforms in China are intended in part to help activists, journalists, and others to limit the ability of the local party-state to engage in practices that not only harm ordinary Chinese but also go against central government interests. However, implementing these reforms requires cooperation from the very actors who may be weakened by them. We show that even controlling for pollution, development, and industrialization, Chinese cities dominated by large industrial firms are less transparent than those with a less-concentrated industrial base. This effect is stronger when the city’s largest firm is in a highly polluting industry. We control for pollution using satellite-generated data in order to avoid relying on questionable official data that may be manipulated by local governments. While the negative economic consequences of the dominance of large state-connected firms have been widely discussed, our study shows that this growth strategy has important and deleterious political effects as well.

Abstract

We argue that interjurisdiction competition in authoritarian regimes engenders a specific logic for taxation. Promotion-seeking local officials are incentivized to signal loyalty and competence to their principals through tangible fiscal revenues. The greater the number of officials accountable to the same principal, the more intense political competition is, resulting in higher taxation; however, too many officials accountable to the same principal leads to lower taxation due to shirking by uncompetitive officials and the fear of political instability. Using a panel dataset of all Chinese county-level jurisdictions from 1999-2006, we find strong evidence for an inverse U-shaped relationship between the number of county-level jurisdictions within a prefecture — our proxy for the intensity of political competition — and fiscal revenues in most provinces but not so in politically unstable ethnic minority regions. The results are robust to various alternative specifications, including models that account for heterogeneous county characteristics and spatial interdependence.

Abstract

The unique structures of process-bearing cells in the central nervous system (CNS) present an ideal model with which to study the differential distribution of mRNA. We conducted a side-by-side examination of the intracellular distribution of nine neural mRNAs by in situ hybridization histochemistry in mammalian brain and observed four general types of mRNA distributions. (1) Some mRNA species were confined to cell somas and included those encoding the glial proteins, myelin proteolipid protein and 2'3'-cyclic nucleotide-3'-phosphodiesterase and the neuronal enzymes, neuron-specific enolase and glutamate decarboxylase-67. (2) Some mRNAs were found abundantly within the cell soma and were also located throughout cellular processes. These included myelin basic protein (MBP) mRNA, which was localized to the cell soma and myelin sheaths of oligodendrocytes, and glial fibrillary acidic protein (GFAP) mRNA, which was localized to the cell soma and processes of reactive and some non-reactive astrocytes in the adult brain and radial glia in embryonic brain. (3) Some mRNAs were found primarily in perinuclear cytoplasm but in some cells were also observed in cell processes. These included mRNAs encoding the protein kinase C/calmodulin-binding substrates, RC3 (neurogranin) and GAP-43, which were identified in the somas as well as within the proximal dendritic branches of specific forebrain neurons. (4) Some mRNAs were localized primarily within cell processes. These included MAP2 mRNA, which was identified by deep staining within dendritic fields but by only light staining within neuronal cell bodies. The data also indicated that the stage of cellular development and the regional location of a cell within the CNS had a profound influence on translocation events. MAP2 mRNA was found in the dendritic processes of most neurons but was confined to the soma of neurons in specific brainstem nuclei. MBP mRNA was confined to the perinuclear cytoplasm of immature oligodendrocytes and was then transported into the myelin sheath at a developmental stage corresponding to myelination. The distribution patterns of these mRNAs are likely to reflect the mechanism by which the protein products of these molecules are targeted within neurons and glia. In addition, mRNA movement may be influenced by cellular and regional factors not encoded solely within the structure of the translocated mRNA.

Abstract

According to the 'pace-of-life' syndrome hypothesis, differences in resting metabolic rate (RMR) should be genetically associated with exploratory behaviour. A large number of studies reported significant heritability for both RMR and exploratory behaviour, but the genetic correlation between the two has yet to be documented. We used a quantitative genetic approach to decompose the phenotypic (co)variance of several metabolic and behavioural measures into components of additive genetic, common environment and permanent environment variance in captive deer mice. We found significant additive genetic variance for two mass-independent metabolic measures (RMR and the average metabolic rate throughout the respirometry run) and two behavioural measures (time spent in centre and distance moved in a novel environment). We also detected positive additive genetic correlation between mass-independent RMR and distance moved (r(A) = 0.78 ± 0.23). Our results suggest that RMR and exploratory behaviour are functionally integrated traits in deer mice, providing empirical support for one of the connections within the pace-of-life syndrome hypothesis.

Abstract

We recently reported evidence implicating fatty-acid binding protein (Fabp) in the control of sleep and memory formation. We used Drosophila melanogaster to examine the relationship between sleep and memory through transgenic overexpression of mouse brain-Fabp, Fabp7, or the Drosophila Fabp homolog, (dFabp). The key findings are that 1) a genetically induced increase in daytime consolidated sleep (naps) correlates with an increase in cognitive performance, and 2) a late “window” of memory consolidation occurs days after the traditionally understood “synaptic” consolidation. Exactly how Fabp-signaling may be involved in converting normal to enhanced long-term memory (LTM) is not known. Here we describe additional data which support relative subcellular compartmental localization of Fabp in regulating stage associations of different forms of memory in Drosophila. Anesthesia resistant memory (ARM) is a longer lasting memory that is produced by massed training, but unlike LTM produced by spaced training, it is insensitive to protein synthesis inhibitors and does not persist as long. We observed that the ratio of ARM to LTM performance index of Fabp7-transgenic flies is proportional to the relative cytoplasmic to nuclear Fabp7 expression level. These data suggest a common lipid-signaling cascade exists between phases of memory formation previously thought to be molecularly distinct.

Abstract

We present a measurement of the inclusive jet cross section in {ital {bar p}p} collisions at {radical}{ital s} =1.8 TeV at the Fermilab Tevatron using the Collider Detector at Fermilab. Good agreement is seen with the predictions of recent next-to-leading-order ({ital O}({alpha}{sub {ital s}}{sup 3})) QCD predictions. The dependence of the cross section on clustering cone size is reported for the first time. 84. Crisis Management in an Authoritarian Regime: Media Effects During the Sichuan Earthquake

Abstract

Exogenous shocks are said to play a key role in the breakdown of authoritarian regimes. This paper sheds light on the conditions under which crisis management play out to the advantage of authoritarian leaders, or not. By chance, a national probability survey of the Chinese population was conducted before and after the 2008 Sichuan earthquake. Based on a quasi-experimental design that allows us to match pre- and post-quake respondents, we find that citizens were highly responsive to media content. In the short run, a more open information policy helped shore up support for a government that exhibited a high degree of responsiveness to the crisis. We find, however, that this approach may have undermined the regime’s legitimacy. Whereas in the immediate aftermath of the quake the Chinese media emphasized political unity and harmony, over time increasing and unusual media criticisms of local governments in the media eroded public confidence. Even though support for political unity and harmony increased immediately after the quake, it quickly dropped below pre-quake levels. Our results are robust both to matching and to more standard parametric specifications.

Abstract

In spite of its technological importance, the basic mechanisms of laser nitriding of metals and alloys are hardly understood. The nitrogen depth profiles achieved by laser nitriding of pure iron were measured with high accuracy by resonant nuclear reaction analysis and described by two superimposed diffusion profiles. Using simple estimates, together with the results of marker experiments and laser treatments in 15N-isotopically enriched atmospheres, the development of these profiles with the number of pulses can be simulated in excellent agreement with the experimental results.

China, like many authoritarian regimes, struggles with the tension between the need to foster economic development by empowering local officials and the regime's imperative to control them politically. Landry explores how the Chinese Communist Party (CCP) manages local officials in order to meet these goals and perpetuate an unusually decentralized authoritarian regime. Using unique data collected at the municipal, county, and village level, Landry examines in detail how the promotion mechanisms for local cadres have allowed the CCP to reward officials for the development of their localities without weakening political control. His research shows that the CCP's personnel management system is a key factor in explaining China's enduring authoritarianism and proves convincingly that decentralization and authoritarianism can work hand in hand.

Abstract

Authoritarian regimes that allow political competition at the lowest level of local government do not to so in order to organize their own downfall but because they seek to enhance the legitimacy of the Party and, in turn, extend the duration of their regime. For ruling parties that are threatened by corruption and entrenched local elites, the parsimonious use of the ballot box allows the incorporation of social groups previously excluded from the political system, institutionalizes elite recruitment and rejuvenation, and more generally enhances the responsiveness of the ruling Party to pent-up social demands without risking its hold on national power. The KMT's strategy on Taiwan before 1987 is one of the most successful examples of adaptability to limited and carefully controlled political competition. Even though in the end the ruling party accepted a complete democratization process, the KMT was able to hold onto power for a considerable amount of time, while at the same time holding locally competitive elections. Since the late 1980's, the Chinese government attempted to rejuvenate grassroots politics and reinforce the legitimacy of the CCP through a series of reforms affecting the recruitment of local leaders. Communist Party functionaries now interact with elected village governments, a configuration of local power that would seem to be at odds with the principal–agent logic that guaranteed tight Party control over government units, as we examined in the previous chapters.

Abstract

In November 2002, Hu Jintao became the fourth general secretary of the Chinese Communist Party (CCP) of the “reform era,” which began in earnest in December 1978. The carefully orchestrated leadership transition was widely regarded as the most predictable and peaceful transfer of power in the history of the People's Republic. The contrast with the events of the late 1980s that rocked the communist world could not have been greater. When communism ended, first in Eastern Europe, then in the Soviet Union itself, the future of the Chinese regime seemed very much in doubt. The series of demonstrations during the spring of 1989 proved that the CCP was not immune to the kind of political instability that led to the destruction of communism elsewhere. Although by the summer of 1989 the Chinese leadership seemed to have “won,” scholars outside China ascribed the use of force against demonstrators to the desperation of a Party weakened by ten years of reforms; Deng's pyrrhic victory signified a “transition postponed,” but certainly not a precluded one (Shue, 1992; Pei, 1994). Fifteen years later, the transition has still not taken place. Instead, the post-Tiananmen leadership surprised the world by embracing a breathtaking series of politically difficult reforms: deeper integration with the world economy, culminating with World Trade Organization membership in 2001; the restructuring of the state sector, including massive layoffs; the privatization of much of the housing sector in urban areas; and the generalization of partially competitive elections at the village level.

Abstract

The success or failure of a career depends upon character and capability; it has nothing to do with the desirability of the post assigned. Huang Liuhong ([1694] 1984: 76) The decentralization of personnel policy raises several theoretical and empirical issues for our understanding of the evolution of the Chinese political system. Pessimists have good reasons to expect failure: Local management of cadres requires myriad principals to monitor and reward an even greater number of agents posted in thousands of local government and Party organizations. Even if we assume that localities comply with central directives most of the time, the sheer number of bureaucratic layers severely reduces the likelihood that a central directive will be implemented at the lowest level. With China's five levels of administration, the odds are higher than 50% (namely, a coin toss) if all local agents comply 88% of the time! This harsh mathematical proposition conforms to the often-heard saying that “the sky is high and the Emperor is far away,” as cadres sometimes put it when discussing the gap between Beijing's expectations and the more prosaic realities of their localities. But what if the emperor has good ears, a few carrots, and uses a stick from time to time? We already know that JES respondents believe that cadre policy has been carried out in a fashion that is broadly consistent with the central government's stated objectives.

Abstract

During the reform era, the center of gravity of the Chinese political economy tilted decisively toward cities. The unprecedented pace of China's economic transformation favored urban growth, which in turn increased the political relevance of municipalities and the officials who rule them. Cities now control a far greater share of the country's resources than at any point in the history of the People's Republic. In 2000, municipalities accounted for 51.8% of China's GDP, 50.1% of its industrial output, and 76% of the value of services (Jiang and Cui, 2001). This increased economic might was largely purposeful. The center aimed some of its boldest reform initiatives at municipalities, symbolized by the early creation of special economic zones as early as 1979 (Crane, 1990; Kleinberg, 1990). After 1984, economic decentralization was generalized to other areas, but Premier Zhao Ziyang stressed that coastal cities would enjoy economic privileges that would not be extended to the less developed and more rural hinterland (D. Yang, 1990). The leadership's urban bias survived the crisis of 1989: Until 2002, Jiang Zemin and Zhu Rongji — both former mayors and Party secretaries in Shanghai — presided over further reform initiatives that benefited cities (Naughton, 1995; Wang and Hu, 1999). In parallel with their rapid economic transformation, cities also enjoy greater formal institutional weight.

Abstract

Brain fatty acid binding protein (Fabp7), which is important in early nervous system development, is expressed in astrocytes and neuronal cell precursors in mature brain. We report here that levels of Fabp7 mRNA in adult murine brain change over a 24 hour period. Unlike Fabp5, a fatty acid binding protein that is expressed widely in various cell types within brain, RNA analysis revealed that Fabp7 mRNA levels were elevated during the light period and lower during dark in brain regions involved in sleep and activity mechanisms. This pattern of Fabp7 mRNA expression was confirmed using in situ hybridization and found to occur throughout the entire brain. Changes in the intracellular distribution of Fabp7 mRNA were also evident over a 24 hour period. Diurnal changes in Fabp7, however, were not found in postnatal day 6 brain, when astrocytes are not yet mature. In contrast, granule cell precursors of the subgranular zone of adult hippocampus did undergo diurnal changes in Fabp7 expression. These changes paralleled oscillations in Fabp7 mRNA throughout the brain suggesting that cell-coordinated signals likely control brain-wide Fabp7 mRNA expression. Immunoblots revealed that Fabp7 protein levels also underwent diurnal changes in abundance, with peak levels occurring in the dark period. Of clock or clock-regulated genes, the synchronized, global cycling pattern of Fabp7 expression is unique and implicates glial cells in the response or modulation of activity and/or circadian rhythms.

Abstract

The distribution of beta-S100 mRNA within the brain and skull of the 14-day rat embryo was determined using in situ hybridization. In nonnervous tissue, signal corresponding to beta-S100 mRNA was prominent within cartilaginous regions. Signal appeared to be confined to chondrocytes and was not observed within undifferentiated cells of the perichondrium. Within the skull, expression of beta-S100 mRNA was prominent in the nasal region and maxillary process as well as within Meckel's cartilage. Signal was also observed within the cranial base of the embryonic skull. In the embryonic brain, beta-S100 mRNA was prominent within the ventricular region of the myelencephalon and could be localized to a layer of cells lining the ventricle. Although beta-S100 mRNA has been shown to accumulate postnatally in the central nervous system, the present results clearly identify high levels of beta-S100 mRNA within specific regions of the embryonic brain and skull.

Abstract

Background Cues predictive of food availability are powerful modulators of appetite as well as food-seeking and ingestive behaviors. The neurobiological underpinnings of these conditioned responses are not well understood. Monitoring regional immediate early gene expression is a method used to assess alterations in neuronal metabolism resulting from upstream intracellular and extracellular signaling. Furthermore, assessing the expression of multiple immediate early genes offers a window onto the possible sequelae of exposure to food cues, since the function of each gene differs. We used immediate early gene and proenkephalin expression as a means of assessing food cue-elicited regional activation and alterations in functional connectivity within the forebrain. Results Contextual cues associated with palatable food elicited conditioned motor activation and corticosterone release in rats. This motivational state was associated with increased transcription of the activity-regulated genes homer1a, arc, zif268, ngfi-b and c-fos in corticolimbic, thalamic and hypothalamic areas and of proenkephalin within striatal regions. Furthermore, the functional connectivity elicited by food cues, as assessed by an inter-regional multigene-expression correlation method, differed substantially from that elicited by neutral cues. Specifically, food cues increased cortical engagement of the striatum, and within the nucleus accumbens, shifted correlations away from the shell towards the core. Exposure to the food-associated context also induced correlated gene expression between corticostriatal networks and the basolateral amygdala, an area critical for learning and responding to the incentive value of sensory stimuli. This increased corticostriatal-amygdalar functional connectivity was absent in the control group exposed to innocuous cues. Conclusion The results implicate correlated activity between the cortex and the striatum, especially the nucleus accumbens core and the basolateral amygdala, in the generation of a conditioned motivated state that may promote excessive food intake. The upregulation of a number of genes in unique patterns within corticostriatal, thalamic, and hypothalamic networks suggests that food cues are capable of powerfully altering neuronal processing in areas mediating the integration of emotion, cognition, arousal, and the regulation of energy balance. As many of these genes play a role in plasticity, their upregulation within these circuits may also indicate the neuroanatomic and transcriptional correlates of extinction learning.

The purpose of this study was to determine the effects work roles (patient managementsservice provider, administratorsmanager, researcher, educator, and student) and their associated tasks have on the choice of information sources used to meet private practice dentists' information needs. Additionally, the study investigated how the Internet has affected the information seeking of dentists. Using Leckie, Pettigrew, and Sylvain's (1996) model of the information seeking of professionals as the conceptual framework, vignette-based, in-depth interviews were conducted with 12 dentists in the metropolitan areas of Seattle, Tacoma, and Everett, Washington. Follow-up interviews were used to investigate dentists' use of the Internet. Findings revealed that the type of work role-related task significantly shapes dentists' choices of information sources; the Internet emerged as a significant information source because it provides up-to-date information in a convenient and timely manner; the Internet is a complement to traditional information sources, not a replacement for them; and the Leckie and associates (1996) general model of professional information seeking is supported by this study. © 2006 Wiley Periodicals, Inc.

Abstract

Diurnal changes in Chordc1 mRNA were recently described in mouse hypothalamus. This report shows that Chordc1 mRNA changes rhythmically throughout the entire adult brain with highest expression levels occurring around the dark–light transition. The rhythmic cycling pattern of Chordc1 was retained under various light–dark schedules and analysis of adult whole brain revealed diurnal patterns that were different than young animals (postnatal day (P) 6). Analysis of adult hippocampus, prefrontal cortex and cerebellum confirmed these observations and a comparison between adult and P6 animals using in situ hybridization indicated that Chordc1 underwent coordinated but altered diurnal changes in mRNA abundance during development. Further, a developmental profile of Chordc1 expression beginning at embryonic day 17 revealed a regional distribution of Chordc1 consistent with its adult pattern. These results suggest that Chordc1 mRNA is under complex and widespread transcriptional regulation during development and implicate Chordc1 in circadian and/or homeostatic mechanisms in mammalian brain.

Abstract

The transcription coactivator CITED1 is an important mediator of transcriptional events regulated by estrogen or TGF-β. We used in situ hybridization to delineate the distribution of CITED1 mRNA in the adult and developing murine brain and found robust CITED1 expression in ventral hypothalamus and midbrain raphe. The distribution of CITED1 in these regions overlapped the reported expression of estrogen receptors α and β. Less intense expression of CITED1 was also evident in medial preoptic area, subfornical organ, thalamus and cerebral cortex. CITED1 mRNA in the arcuate nucleus (an area of active transcriptional modulation by TGF-β) was evident in postmigratory neurons as early as embryonic day 16. Expression of CITED1 in arcuate continued throughout postnatal development. CITED1 in developing cerebellum was first evident in external granule cells and was transiently expressed in the Purkinje cell/granule cell layer in a temporal pattern similar to estrogen receptor-β. The spatial and temporal distribution of CITED1 mRNA reported here is consistent with a role for CITED1 in the modulation of transcriptional events mediated by steroid hormone and cytokine signaling pathways.
