Diseases produced by helminth parasites occur frequently in underdeveloped countries where they present a serious public health problem. At the same time, in these regions, a lower rate of autoimmune and allergic diseases has been observed. Due to these observations, some researchers have proposed that some helminths, such as Trichinella spiralis or its proteins, have strong anti-inflammatory potential, or have assessed them as modulating agents of the immune response. T. spiralis shifts the host immune response from a Th1 profile, characterized by pro-inflammatory cytokines, to a Th2 profile, characterized by the release of different cytokines with anti-inflammatory properties. This parasite has shown high therapeutic potential in a wide variety of disease models. In one of the most promising, the experimental lupus model in mice, the release of anti-inflammatory cytokines IL-4 and IL-10 and delayed onset of the key clinical features of the experimental lupus model for at least 5 months were observed, when previously parasitized. This is the first study to date that focuses on the use of T. spiralis as an immunomodulator in lupus disease. In conclusion, further study of the immune response generated by the parasite is necessary to advance the development of new therapies for inflammatory diseases.

Summary • C assimilation (A) has been shown to limit the growth of young Lactuca sativa (lettuce) plants following an interruption in their external N supply. Further data from these plants were used to test two hypotheses: that N-limited growth of lettuce is associated with lower stomatal conductance (gs); and that reductions in gs result from adjustments to stomatal frequency or distribution. • The photosynthetic characteristics, nitrate and organic N-concentrations, as well as epidermal and stomatal distributions, were determined in leaves of hydroponically grown lettuce plants supplied continuously with N or with N removed for up to 14 d. • Although N-limited plants had lower maximum rates of A, comparisons at equivalent values of gs showed that A was not directly limited by organic-N but by gs. Reductions in gs under N-limiting conditions did not associate with adjustments to stomatal frequency or distribution. • Associations between plant N and A could arise either through stomata responding directly to signals induced by N deprivation or to increased CO2 partial pressure at the sites of carboxylation.

The clinical presentation of coronavirus disease 2019 (COVID-19), which is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), ranges between mild respiratory symptoms and a severe disease that shares many of the features of sepsis. Sepsis is a deregulated response to infection that causes life-threatening organ failure. During sepsis, the intestinal epithelial cells are affected, causing an increase in intestinal permeability and allowing microbial translocation from the intestine to the circulation, which exacerbates the inflammatory response. Here we studied patients with moderate, severe and critical COVID-19 by measuring a panel of molecules representative of the innate and adaptive immune responses to SARS-CoV-2, which also reflect the presence of systemic inflammation and the state of the intestinal barrier. We found that non-surviving COVID-19 patients had higher levels of low-affinity anti-RBD IgA antibodies than surviving patients, which may be a response to increased microbial translocation. We identified sFas and granulysin, in addition to IL-6 and IL-10, as possible early biomarkers with high sensitivity (73 %) and specificity (51 %) to discriminate between surviving and non-surviving COVID-19 patients. Finally, we found that the microbial metabolite d-lactate and the tight junction regulator zonulin were increased in the serum of patients with severe COVID-19 and in COVID-19 patients with secondary infections, suggesting that increased intestinal permeability may be a source of secondary infections in these patients. COVID-19 patients with secondary infections had higher disease severity and mortality than patients without these infections, indicating that intestinal permeability markers could provide complementary information to the serum cytokines for the early identification of COVID-19 patients with a high risk of a fatal outcome.

In the context of climate change and agrosystem complexification, process-based models of the reproductive phenology of perennial grasses are essential to optimize the agronomic and ecologic services provided by grasslands. We present a functional–structural model called L-GrassF, which integrates the vegetative and reproductive development of individual Lolium perenne plants. The vegetative development in L-GrassF was adapted from a previous model of perennial ryegrass where leaf elongation and tillering dynamics partially result from self-regulated processes. Significant improvements have been made to this vegetative module in order to deal with the whole growing cycle during which plants are exposed to contrasting temperatures. The reproductive module is a new functionality describing the floral induction of the individual tiller from daily temperature and photoperiod as well as its phenological state. From the interactions between the vegetative and reproductive developments, L-GrassF simulates the dynamics of plant architecture, the floral transition and heading date (HD) at tiller level. A sensitivity analysis was performed on L-GrassF and showed that parameters controlling the kinetics of leaf elongation and leaf appearance rate have a significant impact on HD. After calibration, L-GrassF was able to simulate the HD on seven L. perenne cultivars grown in a broad range of environmental conditions, as provided by an independent data set. We conclude that L-GrassF is a significant step towards better prediction of grassland phenology in contrasted conditions.

Red:far-red ratio (R:FR) plays an important role in the architectural dynamics of vegetation. The integration of its effects into the crop model and/or into modelling work on plant dynamics over years requires new methods for describing R:FR spatial and temporal variability. This study assesses the sensitivity of simulating plant morphogenesis to the methods of R:FR modelling. The approach consisted in using a generic individual-based legume model coupled with radiative transfer models allowing the computation of R:FR values. Three methods of computation of R:FR were evaluated based on reference radiative transfer model CANESTRA and on turbid-medium model RIRI. The effects of R:FR simulated by the three methods on the simulation of plant morphogenesis were evaluated for the petiole and internodes of two contrasted plant architectures (alfalfa and white clover) at different stages of plant development and density. Plant morphogenesis expressed by the simulated petiole and internode length was sensitive to R:FR values provided by the models, especially at an early stage of development. Contrasted plant architectures exhibit different ranges of sensitivity to R:FR computed by the different models. However, this sensitivity follows a similar pattern between the two types of plant architecture, also for other conditions such as density or stage of development. This study highlights that the choice of radiative transfer model is of main importance for modelling plant morphogenetical responses, in particular at an early stage of plant development. The role of coupling of the FSP and radiative transfer models to address photomorphogenetic issues in order to consider plant-to-plant interactions is discussed.

Maisons-Alfort : Anses, 275 p.; Le contexte de cette expertise est présenté ci-après selon les termes de la saisine émanant de la DGAL, la DGS et la DGCCRF .« La présence de nitrites dans l’organisme peut conduire à l’oxydation de l'hémoglobine réduisant la capacité des globules rouges à transporter l'oxygène. Elle peut également contribuer à la formation d’autres composés, tels que les nitrosamines, dont certains sont cancérogènes.Il existe différentes sources d’exposition des consommateurs aux nitrites et notamment :i) La conversion de nitrates présents dans certaines denrées alimentaires, en nitrites.Les nitrates se retrouvent dans l'eau notamment en raison de leur utilisation en production primaire. Ils sont naturellement présents dans certaines denrées alimentaires, en particulier dans les légumes-feuilles comme les épinards ou la laitue. Chez l’Homme, une partie des nitrates consommés peut être convertie en nitrites par les bactéries présentes dans la cavitébuccale.ii) La présence non intentionnelle de nitrites dans les denrées alimentaires.Les nitrites peuvent notamment être présents dans les légumes mais à des teneurs généralement très inférieures aux nitrates.iii) L’utilisation de nitrites et de nitrates en tant qu’additifs dans les denrées alimentaires.Les nitrites de potassium et de sodium (E249, E250) et les nitrates de sodium et de potassium (E251, E252) sont couramment utilisés pour préserver la viande et d'autres produits périssables. Ils contribuent également à limiter la prolifération de microorganismes nuisibles, en particulier Clostridium botulinum, responsable du botulisme. Il s’agit d’additifs alimentairesautorisés dans l’Union européenne. A ce titre, ils ont fait l’objet de travaux scientifiques d’évaluation pour caractériser les risques liés à leur utilisation.Actuellement la réglementation européenne prévoit une teneur maximale d’incorporation de 150 mg/kg pour les produits de viande transformés, hors produits traditionnels. Pour les produits traditionnels dont la production est moindre en volume que les produits fabriqués par les industriels et qui contribuent donc moins à l’exposition des consommateurs, laréglementation prévoit une teneur de nitrites et nitrates résiduels pouvant aller jusqu’à 250 mg/kg. La teneur résiduelle varie selon différents facteurs liés au procédé de transformation (température de transformation, pH, présence d’acide ascorbique, etc.) et ne dépend pas uniquement de la teneur initialement incorporée. Les autorités danoises ont, pourleur part, souhaité maintenir un niveau d’emploi plus faible, à 60 mg/kg, qu’ils avaient fixé avant l’harmonisation européenne.L’association du fer-héminique avec les nitrites ajoutés a notamment été envisagée comme une explication du risque accru observé de développement de cancers du côlon ou du rectum lié à la consommation de certains produits carnés (données CIRC-INCa 2018)[2]. Le ferhéminique contenu dans la viande favoriserait la conversion des nitrites en nitrosamines, substances classées cancérogène probable (groupe 2A).Pour répondre à la demande des consommateurs, l’industrie a développé des solutions qu’elle présente comme alternatives à l’utilisation de sels nitrés (raccourcissement des DLC, bouillons de légumes, extraits de végétaux, etc.). En outre, des opérateurs s’engagent de manière plus globale à diminuer l’utilisation des additifs conformément aux objectifs définis par leprogramme national de l’alimentation et de la nutrition (PNAN).Dans le cadre de la réévaluation des additifs, l’Autorité européenne de sécurité des aliments (EFSA) a rendu des avis en juin 2017[3] et conclu, sur la base des éléments de preuve disponibles, que les niveaux de sécurité existants pour les nitrites et les nitrates ajoutés en tant qu’additifs à la viande et à d'autres aliments constituaient une protection adéquate pour les consommateurs.Plus précisément :■ S’agissant des nitrates en tant qu’additif, et en ayant recours à des données dites réalistes (c’est-à-dire, les niveaux de concentration effectivement observés dans les aliments), les experts ont estimé que l'exposition des consommateurs aux nitrates utilisés comme additifs alimentaires était inférieure à 5% de l'exposition globale aux nitrates dans les aliments et ne dépassait pas la DJA ;■ S’agissant des nitrites en tant qu’additif, les experts ont estimé que l'exposition se situait dans des limites sûres pour tous les groupes de population, à l'exception d'un léger dépassement chez les enfants dont le régime alimentaire est riche en aliments contenant ces additifs.De plus, le panel de l’EFSA soulignait, lorsque l’ensemble des sources d’expositions alimentaires (présence naturelle dans les aliments, contamination environnementale, utilisation en tant qu’additifs) étaient prises en compte :1. pour les nitrates, des dépassements de la DJA pour l’ensemble des groupes d’âges (hypothèse d’exposition moyenne à haute) ;2. pour les nitrites, des dépassements de la DJA pour l’ensemble des groupes d’âges sous l’hypothèse d’exposition haute, et pour des groupes d’âges spécifiques (nourrissons, jeunes enfants et enfants) sous l’hypothèse d’une exposition moyenne.L’EFSA a également émis des recommandations pour pallier certaines incertitudes. »En ce qui concerne les nitrates, des travaux ont été engagés à l’Anses afin de caractériser les expositions par voie alimentaire (notamment légumes et eau destinée à la consommation humaine) propres à la France dans le cadre de la saisine 2015-SA-0029. Ces travaux résultent d’une saisie de l’Anses par l’association Eaux et rivières de Bretagne (le 15/01/2015), par laCoordination rurale (le 19/01/2015) puis par la FNSEA (le 26/01/2015) sur les impacts sanitaires des nitrates présents dans l’alimentation et dans l’environnement. En effet, les habitudes alimentaires, de même que la présence de nitrates dans les aliments (légumes notamment) et l’eau, peuvent différer sensiblement des données européennes utilisées par l’EFSA. Cela est particulièrement vrai pour l’eau de consommation humaine distribuée dont les paramètres caractérisant sa qualité physico-chimique varient en fonction des ressources exploitées et des traitements mis en œuvre. Ces travaux avaient pour but d’apprécier, au plan national, la situation d’exposition de la population aux nitrates au regard des repères toxicologiques. Les résultats permettraient d’identifier les situations méritant attention, de façon à recommander des mesures, en particulier grâce à l’identification des aliments (y compris l’eau de boisson) les plus contributeurs à l’exposition aux nitrates en France. Ces travaux sont complétés par la présente expertise.

Most individuals infected with Mycobacterium tuberculosis (Mtb) have latent tuberculosis (TB), which can be diagnosed with tests (such as the QuantiFERON-TB Gold test [QFT]) that detect the production of IFN-γ by memory T cells in response to the Mtb-specific antigens 6 kDa early secretory antigenic target EsxA (Rv3875) (ESAT-6), 10 kDa culture filtrate antigen EsxB (Rv3874) (CFP-10), and Mtb antigen of 7.7 kDa (Rv2654c) (TB7.7). However, the immunological mechanisms that determine if an individual will develop latent or active TB remain incompletely understood. Here we compared the response of innate and adaptive peripheral blood lymphocytes from healthy individuals without Mtb infection (QFT negative) and from individuals with latent (QFT positive) or active TB infection, to determine the characteristics of these cells that correlate with each condition. In active TB patients, the levels of IFN-γ that were produced in response to Mtb-specific antigens had high positive correlations with IL-1β, TNF-α, MCP-1, IL-6, IL-12p70, and IL-23, while the proinflammatory cytokines had high positive correlations between themselves and with IL-12p70 and IL-23. These correlations were not observed in QFT-negative or QFT-positive healthy volunteers. Activation with Mtb-soluble extract (a mixture of Mtb antigens and pathogen-associated molecular patterns [PAMPs]) increased the percentage of IFN-γ-/IL-17-producing NK cells and of IL-17-producing innate lymphoid cell 3 (ILC3) in the peripheral blood of active TB patients, but not of QFT-negative or QFT-positive healthy volunteers. Thus, active TB patients have both adaptive and innate lymphocyte subsets that produce characteristic cytokine profiles in response to Mtb-specific antigens or PAMPs. These profiles are not observed in uninfected individuals or in individuals with latent TB, suggesting that they are a response to active TB infection.

Objectives To describe the kinetics of circulating cytokines and chemokines in humans with ZIKAV infection. Methods Serum levels of different immune mediators in patients with ZIKAV infection were measured at distinct stages of the disease, as well as in culture supernatants from human monocytes infected with a clinical ZIKAV isolate. We also looked for clinical features associated with specific immune signatures among symptomatic patients. Results We evaluated 23 ZIKAV-infected patients. Their mean age was 32 ± 8.3 years and 65% were female. ZIKAV patients showed elevated IL-9, IL-17A, and CXCL10 levels at acute stages of the disease. At day 28, levels of CCL4 and CCL5 were increased, whereas IL-1RA, CXCL8 and CCL2 were decreased. At baseline, IL-7 was increased among patients with headache, whereas CCL2, and CCL3 were decreased in patients with bleeding and rash, respectively. Our clinical ZIKAV isolate induced a broad immune response in monocytes that did not resemble the signature observed in ZIKAV patients. Conclusions We showed a unique immune signature in our cohort of ZIKAV-infected patients. Our study may provide valuable evidence helpful to identify immune correlates of protection against ZIKAV.

Les substances naturelles à usage biostimulant (SNUB) sont définies dans le cadre de la réglementation applicable au niveau national. Conformément au point 4° de l'article L. 255-5 du code rural et de la pêche maritime, les SNUB sont dispensées d'une autorisation de mise sur le marché. Toutefois, elles sont soumises à une procédure d’autorisation et d’évaluation, dont les modalités sont fixées par voieréglementaire et codifiées à l'article D. 255-30-11 du code rural et de la pêche maritime.Ainsi, une SNUB est autorisée, le cas échéant avec des prescriptions particulières d’utilisation, par son inscription sur une liste annexée à l'arrêté du 27 avril 20162 et publiée par arrêté du ministre chargé de l’agriculture lorsque :- elle est d’origine végétale, animale ou minérale, à l’exclusion des micro-organismes, et n’est pas génétiquement modifiée ;- elle est obtenue par un procédé accessible à tout utilisateur final correspondant à une absence de traitement ou à un traitement reposant exclusivement sur des moyens manuels, mécaniques ou gravitationnels, la dissolution dans l’eau ou dans l’alcool, la flottation, l’extraction par l’eau ou par l’alcool, la distillation à la vapeur ou le chauffage uniquement pour éliminer l’eau ;- à l’exception des cas où la substance est mentionnée à l’article D. 4211-11 du code de la santé publique, elle a fait l’objet d’une évaluation par l’Anses qui révèle son absence d’effet nocif sur la santé humaine, sur la santé animale et sur l’environnement.Cette inscription intervient à l’issue d’une évaluation individuelle conduite par l’Anses, qui révèle l’absence d'effet nocif sur la santé humaine, sur la santé animale et sur l'environnement. Il est à noter qu’aucune évaluation de l’efficacité n’est requise dans ce cadre réglementaire.Par ailleurs, les SNUB issues de parties consommables de plantes utilisées en alimentation animale ou humaine sont dispensées de cette évaluation individuelle lorsqu'elles entrent dans la composition d'une PNPP3 conforme à un cahier des charges. Ainsi, conformément à l’arrêté le 14 juin 20214 visant notamment l’avis de l’Agence du 28 juillet 20205, les parties consommables de plantes utilisées enalimentation animale ou humaine, qui sont préparées selon le cahier des charges “CDC PLANTES CONSOMMABLES”, sont autorisées en tant que SNUB en dispense d’évaluation individuelle.Dans ce cadre de cette saisine, il est demandé à l’Anses de donner son avis sur la possibilité d’inscrire à la liste annexée à l'arrêté du 27 avril 2016, les 4 substances suivantes :- la prêle des champs ;- le saule ;- l’huile essentielle de menthe des champs ;- la léonardite.A l’appui de cette demande 4 dossiers (1 dossier par substance) préparés par l’ITAB6 ont été mis à disposition de l’Anses. Aucun autre document n’a été mis à disposition de l’Anses. Cet avis présente l’évaluation du saule et de la prêle. L’huile essentielle de menthe des champs et la léonardite feront l’objet d’un avis ultérieur de l’agence.

La presente sequencia didactica tiene como objetivo diseñar experiencias de aprendizaje a través de la creación de ecosistemas bilingües, mediados por el ecoturismo, enmarcado en la perspectiva del trabajo social en el contexto de una escuela primaria rural en la comunidad de La Honda. En Colombia, la educación de idiomas en áreas rurales es una de las diversidades sociales menos exploradas en este campo (Bonilla & Cruz-Arcila, 2013). Esta situación motivó a los implementadores a centrar su atención en las oportunidades de educación lingüística que la comunidad La Honda merece tener. El proyecto ha sido diseñado para realizarse con alumnos multigrado de la escuela primaria Institución Educativa Combia, sector La Honda. Dado que la escuela se basa en el modelo de Escuela Nueva, la metodología estará orientada a seguir sus principios, permeados por el uso del aprendizaje experiencial. La creación de este proyecto les permitió a los investigadores concluir que las experiencicias de aprendizaje diseñadas pueden generar una mejora en la vida de los participantes, en virtud de la adquisición de habilidades lingüisticas y la apreciación de los recursos culturales y ambientales que esta comunidad rural tiene. It was possible to undertake this journey thanks to the learning experience that we have had throughout the whole program. Various adaptive mechanisms can ensure that seedlings are established at the most favourable time and place. These mechanisms include seed dormancy i.e., incapacity to germinate in any environment without a specific environmental trigger and inhibition i.e., incapacity to germinate in an unfavourable environment (water availability, temperature: thermoinhibition and light). The objective of this research was to study in the temperate range for germination of forage and turf grass species perennial ryegrass, if the thermal requirements for germination are under genetic controlled and could be selectively bred. Two divergent selections of three cycles were realized on a natural population: one to select for the capacity to germinate at 10°C vs. the impossibility to germinate at 10°C, and one to select for the capacity to germinate at 32°C vs. the impossibility to germinate at 32°C. Seeds of all the lots obtained from the two divergent selections were then germinated at constant temperatures from 5 to 35°C to evaluate their germination ability. Concerning the positive selection, the first cycle of positive selection at 10°C was highly efficient with a very strong increase in the germination percentage. However, afterward no selection effect was observed during the next two cycles of positive selection. By contrast, the positive selection at 32°C was efficient during all cycles with a linear increase of the percentage of germination at 32°C. Concerning the negative selection, we observed only a large positive effect of the first cycle of selection at 10°C. These findings demonstrate that seed thermoinhibition at 10 and 32°C observed in a natural population of perennial ryegrass has a genetic basis and a single recessive gene seems to be involved at 10°C.

Drug resistant tuberculosis (DR-TB) is an important public health issue in different parts of the world. Mycobacterium tuberculosis complex variants (MTBC vars) preferentially infect certain hosts, limiting their distribution to different ecosystems. However, MTBC vars can infect other hosts beyond their preferred target potentially contributing to persistence of drug resistance (DR) in other niches. Here, we performed a comprehensive intra-host genetic analysis for the identification of DR-related mutations among all MTBC minor vars whole genome sequences (8,095 strains) publicly available worldwide. High confidence drug-resistance mutations in katG (isoniazid), rpsL (streptomycin), pncA (pyrazinamide), rpoB (rifampicin) and gyrA (fluoroquinolones) genes were identified among intrahost minor sub-populations in 197 different strains (2.43%) belonging to vars africanum, bovis, caprae, microti, orygis and pinnipedii. In addition, a three-dimensional structure modeling analysis to assess the role of novel mutations was also performed. Our findings highlight the importance of detecting discrete intra-host populations carrying DR mutations.

La Chaparrala river is the most representative in Andes (Antioquia), in 2014 the physicochemical and biological characterization of the water was performed. The turbidity, COD, alkalinity, nitrate, N-ammonia, total nitrogen, total phosphorus, orthophosphate and total hardness showed high variability and exceeded the bounds of acceptable quality for human consumption. 37 genera of aquatic insect belonging to 27 families and five phyla were found. Diptera were the most abundant order with 25.9% of the total and the families Chironomidae (38.77%), Baetidae (21.03%), Simuliidae (12.03%) and Physidae (7.74%) account for 79.56% of all organisms founded. Although the body of water has a dubious biological quality (BMWP / medium Col = 46.33, EPT = 48.83%) and shows a progressive decline in water quality, we observed the presence of families Leptophlebiidae, Perlidae and Hydropsychidae who live in clean and oxygenated water. It was evident that the river is still capable of self-purification. La quebrada La Chaparrala surte al acueducto municipal de Andes-Antioquia, en 2014 se realizo la caracterizacion fisicoquimica y biologica de sus aguas. La turbiedad, DQO, alcalinidad, nitratos, N-amoniacal, nitrogeno total, fosforo total, ortofosfatos y dureza total, mostraron variabilidad alta y sobrepasaron los limites de calidad admisibles para el consumo humano. Se encontraron 37 generos de insectos acuaticos pertenecientes a 27 familias y cinco phyla. Los dipteros fueron el orden mas abundante con un 25.9% del total de individuos y las familias Chironomidae (38.77%), Baetidae (21.03%), Simuliidae (12.03%) y Physidae (7,74%) representan un 79.56% de todos los organismos encontrados. Aunque el cuerpo de agua presenta una calidad biologica dudosa (BMWP/Col medio=46.33, EPT=48.83%), se evidencia una disminucion progresiva en la calidad del agua, se observo la presencia de las familias Leptophlebiidae, Perlidae e Hydropsychidae que habitan en aguas limpias y oxigenadas. Se evidencio que la quebrada aun es capaz de auto depurarse.
