The prediction of the heat transfer coefficient commonly relies on the development of new empirical prediction equations when operating conditions and refrigerants change from the existing equations. Creating new prediction equations is expensive and time-consuming; therefore, recent attention has been given to machine learning approaches. However, machine learning requires a large amount of data, and insufficient data can result in inadequate accuracy and applicability. This study showed that using mini-channel data as highly relevant data for the micro-fin heat transfer coefficient yields high prediction accuracy, even when the experimental dataset of interest is limited. In the proposed method, we added dimensionless numbers assumed to significantly influence heat transfer coefficients calculated from experimental data to the training dataset. This allowed efficient learning of the characteristics of thin liquid films present in mini-channels and micro-fins. By combining distinctive physical mechanisms related to heat transfer coefficients with DNN/GPR/Fine-tuning, the proposed method can predict 96.7% of the data points within ±30% deviation. In addition, it has been confirmed that the dryout quality and post-dryout heat transfer coefficients were predicted with high accuracy. Additionally, we utilized visualization techniques to investigate the contents of the black-box machine learning models.

The suprachiasmatic nucleus (SCN) of the hypothalamus is the master circadian clock in mammals. SCN neurons exhibit circadian Ca2+ rhythms in the cytosol, which is thought to act as a messenger linking the transcriptional/translational feedback loop (TTFL) and physiological activities. Transcriptional regulation occurs in the nucleus in the TTFL model, and Ca2+-dependent kinase regulates the clock gene transcription. However, the Ca2+ regulatory mechanisms between cytosol and nucleus as well as the ionic origin of Ca2+ rhythms remain unclear. In the present study, we monitored circadian-timescale Ca2+ dynamics in the nucleus and cytosol of SCN neurons at the single-cell and network levels. We observed robust nuclear Ca2+ rhythm in the same phase as the cytosolic rhythm in single SCN neurons and entire regions. Neuronal firing inhibition reduced the amplitude of both nuclear and cytosolic Ca2+ rhythms, whereas blocking of Ca2+ release from the endoplasmic reticulum (ER) via ryanodine and inositol 1,4,5-trisphosphate (IP3) receptors had a minor effect on either Ca2+ rhythms. We conclude that the in-phasic circadian Ca2+ rhythms in the cytosol and nucleus are mainly driven by Ca2+ influx from the extracellular space, likely through the nuclear pore. It also raises the possibility that nuclear Ca2+ rhythms directly regulate transcription in situ.

Pterygoplichthys pardalis is an endemic as well as economically important fish species in the Amazon basin. Thirty P. pardalis specimens were purchased at assorted fairs in the city of Manaus. Fish were necropsied, and then, had their organs investigated for parasites. The identified parasites were observed under Light bright field microscopy and that of Scanning Electron Microscopy (SEM). Through the analyses it was observed 219 metazoan parasite specimens, belonging to four taxonomic groups: Monogenoidea, Digenea, Acanthocephala, and Copepoda. Six metazoan parasitic species were identified: Monogenoidea Unilatus unilatus Mizelle and Kritsky, 1967 and Copepoda Therodamas elongatus (Thatcher, 1986) from the gills, Digenea Austrodiplostomum compactum (Lutz, 1928), Megacoelium spinicavum Szidat, 1954 and Diplostomum sp. from the eyes, stomach, and gonads, respectively, and Acanthocephala Gorytocephalus elongorchis Thatcher, 1979 from the intestine. Gorytocephalus elongorchis was the one presenting the highest prevalence value, followed by digeneans M. spinicavum, Diplostomum sp., and A. compactum. The present study has come to provide the very first SEM M. spinicavum, G. elongorchis, and T. elongatus images, thus improving on the up to now available data addressing P. pardalis parasitic fauna, which has been found to be infecting, the fish species presently being marketed in Manaus. Moreover, the current survey has demonstrated the identified parasite species pose no public health concern at all.

Summary: Does the circadian clock keep running under such hypothermic states as daily torpor and hibernation? This fundamental question has been a research subject for decades but has remained unsettled. We addressed this subject by monitoring the circadian rhythm of clock gene transcription and intracellular Ca2+ in the neurons of the suprachiasmatic nucleus (SCN), master circadian clock, in vitro under a cold environment. We discovered that the transcriptional and Ca2+ rhythms are maintained at 22°C–28°C, but suspended at 15°C, accompanied by a large Ca2+ increase. Rewarming instantly resets the Ca2+ rhythms, while transcriptional rhythms reach a stable phase after the transient state and recover their phase relationship with the Ca2+ rhythm. We conclude that SCN neurons remain functional under moderate hypothermia but stop ticking in deep hypothermia and that the rhythms reset after rewarming. These data also indicate that stable Ca2+ oscillation precedes clock gene transcriptional rhythms in SCN neurons.

Abstract This sentinel node (SN) biopsy trial aimed to assess its effectiveness in identifying predictive factors of micrometastases and to determine whether elective neck dissection is necessary in oral squamous cell carcinoma. This retrospective study included 55 patients from three previous trials, with positive SNs. The relationship between the sizes of the metastatic focus and metastasis in non-sentinel node (NSN) was investigated. Four of the 55 largest metastatic focus were isolated tumor cells, and the remaining 51 were ranged from 0.2 to 15 mm, with a median of 2.6 mm. The difference of prevalence between 46 negative- and 9 positive-NSN was statistically significant with regard to age, long diameter of primary site and number of cases with regional recurrence. Abstract Focal cortical dysplasia is the most common malformation during cortical development, sometimes excised by epilepsy surgery and often caused by somatic variants of the mTOR pathway genes. In this study, we performed a genetic analysis of epileptogenic brain malformed lesions from 64 patients with focal cortical dysplasia, hemimegalencephy, brain tumors, or hippocampal sclerosis. Targeted sequencing, whole-exome sequencing, and single nucleotide polymorphism microarray detected four germline and 35 somatic variants, comprising three copy number variants and 36 single nucleotide variants and indels in 37 patients. One of the somatic variants in focal cortical dysplasia type IIB was an in-frame deletion in MTOR, in which only gain-of-function missense variants have been reported. In focal cortical dysplasia type I, somatic variants of MAP2K1 and PTPN11 involved in the RAS/MAPK pathway were detected. The in-frame deletions of MTOR and MAP2K1 in this study resulted in the activation of the mTOR pathway in transiently transfected cells. In addition, the PTPN11 missense variant tended to elongate activation of the mTOR or RAS/MAPK pathway, depending on culture conditions. We demonstrate that epileptogenic brain malformed lesions except for focal cortical dysplasia type II arose from somatic variants of diverse genes but were eventually linked to the mTOR pathway.

Global warming has caused such problems as the poor coloration of grape skin and the decreased production of high-quality berries. We investigated the effect of synephrine (Syn) on anthocyanin accumulation. Anthocyanin accumulation in cultured grape cells treated with Syn at concentrations of 1 mM or higher showed no significant difference, indicating that the accumulation was concentration-independent. On the other hand, anthocyanin accumulation was dependent on the compound used for treatment. The sugar/acid ratio of the juice from berries treated with Syn did not differ from the control. The expression of anthocyanin-biosynthesis-related genes, but not phytohormones, was increased by the treatment with Syn at 24 h or later. The Syn treatment of cultured cells increased SOD3 expression and hydrogen peroxide (H2O2) production from 3 to 24 h after treatment. Subsequently, the expression of CAT and APX6 encoding H2O2-scavenging enzymes was also increased. Treatment of cultured cells with Syn and H2O2 increased the expression of the H2O2-responsive gene Chit4 and the anthocyanin-biosynthesis-related genes mybA1 and UFGT 4 days after the treatment and increased anthocyanin accumulation 7 days after the treatment. On the other hand, the treatment of berries with Syn and H2O2 increased anthocyanin accumulation after 9 days. These results suggest that Syn increases anthocyanin accumulation through H2O2 production without changing phytohormone biosynthesis. Syn is expected to improve grape skin coloration and contribute to high-quality berry production.

Cellular immunity is critical for the regulation of viral diseases, including coronavirus disease 2019 (COVID-19), and is generally considered immature in childhood. However, the details of cellular immunity against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection among children are unclear. We assessed cellular immunity in eight children post-vaccination against SARS-CoV-2 and 11 children after SARS-CoV-2 infection using the T-SPOT®.COVID assay for the spike (S) and nucleocapsid (N) proteins. In the vaccinated group, the T-SPOT®.COVID assay for the S protein yielded positive results in seven children. In the post-infection group, the assay for the N protein was positive for 5 of 11 children, with 3 of these 5 children requiring hospitalization, including 2 who needed mechanical ventilation. The T-SPOT®.COVID assay is thus valuable for assessing cellular immunity against SARS-CoV-2, and most children infected with SARS-CoV-2 may not develop such immunity unless the disease severity is significant.

Abstract Background Targeting of the apelin–apelin receptor (Apj) system may serve as a useful therapeutic intervention for the management of chronic kidney disease (CKD)‐induced skeletal muscle atrophy. We investigated the roles and efficacy of the apelin–Apj system in CKD‐induced skeletal muscle atrophy. Methods The 5/6‐nephrectomized mice were used as CKD models. AST‐120, a charcoal adsorbent of uraemic toxins (8 w/w% in diet), or apelin (1 μmol/kg) was administered to CKD mice to investigate the mechanism and therapeutic potential of apelin on CKD‐induced skeletal muscle atrophy. The effect of indoxyl sulfate, a uraemic toxin, or apelin on skeletal muscle atrophy was evaluated using mouse myoblast cells (C2C12 cells) in vitro. Study aim: To determine the difference in joint angles of the shoulder, hip, and trunk (angle of the upper torso and lower torso) during vaulting between male pole vaulters with and without chronic low back pain (LBP) and to examine the relationship between the range of motion (ROM) and maximum angle of the same joint during vaulting in all participants.

Water–copper is one of the most common combinations of working fluid and heating surface in high-performance cooling systems. Copper is usually selected for its high thermal conductivity and water for its high heat transfer coefficient, especially in the two-phase regime. However, copper tends to suffer oxidation in the presence of water and thus the heat flux performance is affected. In this research, an experimental investigation was conducted using a cooper bare surface as a heating surface under a constant mass flux of 600 kg·m−2·s−1 of deionized water at a subcooled inlet temperature ΔTsub of 70 K under atmospheric pressure conditions on a closed-loop. To confirm the heat transfer deterioration, the experiment was repeated thirteen times. On the flow boiling region after thirteen experiments, the results show an increase in the wall superheat ΔTsat of approximately 26% and a reduction in the heat flux of approximately 200 kW·m−2. On the other hand, the effect of oxidation on the single phase is almost marginal.

To overcome the trade-off relationship between tensile strength and elongation of dual-phase steels, three exploratory techniques were utilized: Bayesian optimization (BO), BoundLess Objective-free eXploration (BLOX), and one-class support vector machine (OCSVM). The BO is an optimization method using Gaussian process regression, and the BLOX and OCSVM are designed for finding out-of-trend materials. Initially, a large number of synthetic phase distributions of ferrite and martensite were prepared by the Gaussian random field method. Feature importance analysis was then performed to extract microstructural descriptors used as explanatory variables in the explorations. The results revealed that the volume fraction of martensite, the higher-order principal component of the spatial correlation function, and the first principal component of the persistent homology are important in predicting the product of strength and elongation (TS × EL). In the three exploration methods, crystal plasticity finite element analyses with a ductile damage model were repeatedly performed to predict strength and elongation. The BO and OCSVM successfully found microstructures with TS × EL much higher than those of random search in a limited number of iterations. The microstructures discovered in the explorations indicated that two approaches to material design are effective in improving tensile properties of dual-phase steels: uniformly dispersing the fine hard phase and constructing a lamellar structure of the hard and soft phases.

Abstract Nivolumab, an immune checkpoint inhibitor is the first-line therapy for platinum-resistant recurrent/metastatic head and neck cancer, and highly effective for some patients. However, no factors have been identified that could predict response or prognosis after nivolumab administration. We retrospectively investigated the association between tumor infiltrating lymphocytes (TILs) of initial pathology and prognosis in patients treated with nivolumab. Twenty-eight patients with human papilloma virus and Epstein–Barr virus unrelated head and neck squamous cell carcinoma were enrolled. CD8+cells, FoxP3+cells and FoxP3−CD4+cells in the tumoral and peritumoral stromal area and PD-L1 were measured. In result, FoxP3−CD4+TIL, FoxP3+TIL, and CD8+TIL were not correlated with survival in either intratumoral and stromal area. In univariate analysis, objective response was significant prognostic factor both in progression-free survival and overall survival (p = 0.01, 0.006, respectively). PD-L1 was also significant prognostic factor both in progression-free survival and overall survival (p = 0.01, 0.01, respectively). ECOG Performance status was a significant prognostic factor in overall survival (p = 0.0009). In the combined analysis of stromal CD8+TIL and PD-L1, PD-L1 positive with high stromal CD8+TIL subgroups had a better prognosis than PD-L1 negative with low stromal CD8+TIL subgroups in progression-free survival (p = 0.006). Although these results require a further investigation, PD-L1 and ECOG Performance status and the combination of stromal CD8+TIL and PD-L1 positivity have potential as useful prognostic markers in patients of virus unrelated head and neck squamous cell carcinoma treated with nivolumab.
