64. Highly active Mn2O3-Fe3O4catalyst with abundant metal-oxygen bonds for real time sensing of carbendazim in environmental samples

Abstract

In agriculture, a benzimidazole-based fungicide of carbendazim (CBZ) is widely used for disease control in fruits and vegetables. However, its toxicity raises concerns about potential harm to humans from consuming contaminated food. In this study, we developed an eco-friendly manganese iron oxide (Mn2O3-Fe3O4) catalyst via coprecipitation, employing with high temperature annealing (MFO-A) and calcination (MFO-C) techniques. Comparative physical and chemical characterization demonstrated that MFO-A performed better than MFO-C. The prepared catalyst was employed to modify a glassy carbon electrode (GCE) in order to enable electrochemical CBZ sensing through cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under the optimized experimental condition, the sensor achieving a lower limit of detection (LOD), limit of quantification (LOQ), and sensitivity are 0.027 ng/L, 0.084 ng/L, and 1.57 µA (ng/L)–1cm−2. The sensor demonstrated excellent selectivity, repeatability, reproducibility, and storage stability with acceptable recovery rate. The effectiveness of the present sensor was further validated through real-time analysis of environmental and food samples, highlighting its potential for enhancing environmental safety.

Abstract

Potato farming is a vital component of food security and the economic stability especially in the under developing countries but it faces many challenges in production, blackleg disease caused by Pectobacterium atrosepticum (Pa) is one of the main reason for damaging crop yield of the potato. Effective management strategies are essential to control these losses and to get sustainable potato crop yield. This study was focused on characterizing the Pa and the investigating new chemical options for its management. The research was involved a systematic survey across the three district of Punjab, Pakistan (Khanewal, Okara, and Multan) to collect samples exhibiting the black leg symptoms. These samples were analyzed in the laboratory where gram-negative bacteria were isolated and identified through biochemical and pathogenicity tests for Pa. DNA sequencing further confirmed these isolates of Pa strains. Six different chemicals were tested to control blackleg problem in both vitro and vivo at different concentrations. In vitro experiment, Cordate demonstrated the highest efficacy with a maximum inhibition zones of 17.139 mm, followed by Air One (13.778 mm), Profiler (10.167 mm), Blue Copper (7.7778 mm), Spot Fix (7.6689 mm), and Strider (7.0667 mm). In vivo, Cordate maintained its effectiveness with the lowest disease incidence of 14.76%, followed by Blue Copper (17.49%), Air One (16.98%), Spot Fix (20.67%), Profiler (21.45%), Strider (24.99%), and the control group (43.00%). The results highlight Cordate's potential as a most effective chemical against Pa, offering promising role for managing blackleg disease in potato and to improve overall productivity. Abstract

Cobalt oxide nanoparticles (Co3O4-NPs) and cobalt sulfide nanoparticles (Co9S8-NPs) have shown extensive potential in electrochemical sensing, catalysis, specific drug targeting, and resonance imaging. However, Co9S8-NPs have been rarely explored for biomedical applications as compared to their oxide counterparts. Thus, in the current study, biocompatible PEGylated Co9S8-NPs and Co3O4-NPs are explored and compared for biological and photocatalytic properties. PEGylation of both the NPs is achieved using a simple chemical co-precipitation method followed by characterization using UV visible spectroscopy (UV-Vis), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscopy (SEM), energy-dispersive X-ray analysis (EDX), and pH-responsive dispersion study. After thorough characterization, the NPs are evaluated and compared for various biological applications including antibacterial, antifungal, antileishmanial, antioxidant, and biocompatibility as well as photo-catalytic dye degradation studies. Both the NPs have shown excellent biological applications; however, Co9S8-NPs exhibit comparatively better antibacterial, antifungal, and antioxidant properties except antileishmanial potential where Co3O4-NPs show slight superiority. Furthermore, Co3O4-NPs indicate a higher degradation potential of methylene blue (MB) up to 46.93%. In comparison, Co9S8-NPs have a degradation ability of up to 42%, at 20 mg/ml within 3 h, which indicates considerable remediation potential of the NPs. 82. Bioprocess optimization of Penicillium oxalicumSM03 for the production of cellulases on freshwater alga Salvinia molestabiomass in solid-state fermentation

Abstract

The freshwater weed Salvinia molestawas used as a substrate for the production of cellulase in solid-state fermentation. The lignocellulose material was used as the sole source of carbon and energy. The decaying Salvinia molestaplant was used as the source of cellulase-producing fungi, and 12 strains were isolated. The strain SM03 exhibited maximum cellulase (9.1 ± 0.1 U/mL carboxymethyl cellulase, 10.4 ± 0.08 U/mL β-glucanase, and 0.72 ± 0.09 U/mL filter paperase) production. The strain SM03 was characterized as Penicillium oxalicumSM03 based on 18S rDNA gene sequencing. Solid-state fermentation was performed using P. oxalicumSM03, and culture conditions were optimized for maximum cellulase production. The present findings revealed that maximum cellulase production was achieved at 1% ammonium sulphate, 30 °C incubation temperature, and after 96-h incubation. A central composite design experiment was performed to determine the optimum pH level, fermentation period, and ammonium sulphate concentration. Response surface methodology improved twofold production of carboxymethyl cellulase (p= 0.0033), 1.9-fold increased enzyme yield in β-glucanase (p= 0.0004), and 2.1-fold increased filter paperase activity (p= 0.0004). The saccharification efficacy of cellulase on chemically pretreated S. molestabiomass was tested. The fungal cellulases improved, reducing sugar yield from 0.187 ± 0.02 to 0.417 ± 0.03 g/g.

Abstract

Wheat is an important crop, used as staple food in numerous countries around the world. However, wheat productivity is low in the developing world due to several biotic and abiotic stresses, particularly drought stress. Non-availability of drought-tolerant wheat genotypes at different growth stages is the major constraint in improving wheat productivity in the developing world. Therefore, screening/developing drought-tolerant genotypes at different growth stages could improve the productivity of wheat. This study assessed seed germination and seedling growth of eight wheat genotypes under polyethylene glycol (PEG)-induced stress. Two PEG-induced osmotic potentials (i.e., -0.6 and -1.2 MPa) were included in the study along with control (0 MPa). Wheat genotypes included in the study were ‘KLR-16’, ‘B6’, ‘J10’, ‘716’, ‘A12’, ‘Seher’, ‘KTDH-16’, and ‘J4’. Data relating to seed germination percentage, root and shoot length, fresh and dry weight of roots and shoot, root/shoot length ratio and chlorophyll content were recorded. The studied parameters were significantly altered by individual and interactive effects of genotypes and PEG-induced osmotic potentials. Seed germination and growth parameters were reduced by osmotic potentials; however, huge differences were noted among genotypes. A reduction of 32.83 to 53.50% was recorded in seed germination, 24.611 to 47.75% in root length, 37.83 to 53.72% in shoot length, and 53.35 to 65.16% in root fresh weight. The genotypes, ‘J4’, ‘KLR-16’ and ‘KTDH-16’, particularly ‘J4’ better tolerated increasing osmotic potentials compared to the rest of the genotypes included in the study. Principal component analysis segregated these genotypes from the rest of the genotypes included in the study indicated that these can be used in the future studies to improve the drought tolerance of wheat crop. The genotype ‘J4’ can be used as a breeding material to develop drought resistant wheat genotypes.

Abstract

In-vitro chemical characterization and mosquitocidal screening of essential oil derived from Mikania scandens (L.) Willd. (Asteraceae) (CVO-MS) against the malarial vector Anopheles gambiae was investigated. GC-MS analysis evidenced a total of 12 bio-active compounds, and maximum at α-bisabolol (39.34%) followed by stigmasterol (13.45%) respectively. The larvicidal activity of CVO-MS against the malarial vector An. gambiae evidenced that the mortality rate was prominent at the maximum dosage of 1000 ppm. The Median Lethal Concentration (LC50) of CVO-MS was established at 488±2.45 ppm, respectively. Enzyme inhibition assay showed that the CVO-MS delivers a significant upsurge in the level of CYP450, GST, and a decline in the level of α-β carboxylesterase activity in both the third and fourth instar (p ≤ 0.0001). The gut-histological examination of CVO-MS showed that there is severe damage in the mid-gut tissues, especially epithelial layer (EL), gut-lumen (GL), and peritrophic membrane (pM). The non-target screening against the mosquito predators (A. bouvieri, D. indicus, and third instar larvae of Tx. splendens) suggests that CVO-MS showed less toxicity as compared to Pestanal® (1 ppm) respectively. The present study has paved a new insight in understanding the bioactivity of CVO-MS as a potential larvicidal agent of malarial vector and non-toxic against mosquito predators.
