Abstract

An innovative electrically conductive hydrogel was fabricated through the incorporation of silica nanoparticles (SiO2 NPs) and poly(aniline-co-dopamine) (PANI-co-PDA) into oxidized alginate (OAlg) as a biomimetic scaffold for bone tissue engineering application. The developed self-healing chemical hydrogel was characterized by FTIR, SEM, TEM, XRD, and TGA. The electrical conductivity and swelling ratio of the hydrogel were obtained as 1.7 × 10−3 S cm−1 and 130%, respectively. Cytocompatibility and cell proliferation potential of the developed scaffold were approved by MTT assay using MG-63 cells. FE-SEM imaging approved the potential of the fabricated scaffold for hydroxyapatite (HA) formation and bioactivity induction through immersing in SBF solution. Abstract

Objective(s): Encapsulation of essential oil in polymeric nanoparticles (NPs) increases their retention and improves their efficacy. Here, eucalyptus essential oil (EEO) encapsulate in the chitosan (CS) NPs increases its retention, and enhances the anticancer effect of EEO. Materials and Methods: The effects of pH, chitosan sodium tripolyphosphate ratio, and chitosan concentration on the size and charge NPs were evaluated. The success of EEO encapsulation was confirmed by FT-IR, UV-Vis spectroscopy, and GC techniques. The toxic effect of free EEO and CS-EEO NPs was investigated in MDA-MB-231 breast cancer cells and fibroblast normal cells. Results: The optimized obtained EEO -loaded chitosan nanoparticles (CS-EEO NPs) were spherical with an average diameter of 86 nm, a polydispersity index below 0.4, and positive zeta potential (+14.25 mV) as confirmed. Increasing the concentration and pH of the chitosan solution and decreasing the chitosan/sodium tripolyphosphate ratio, the size of NPs decreased. Loading capacity (LC) and encapsulation efficiency (EE) of EEO in the NPs were about 45% and 32-76%, respectively. The chitosan nanoparticles exhibited a biphasic release profile with the release of 87% of the EEO in the first 5 h, followed by a sustained release for the next 43 h. Conclusion: The free EEO was more toxic for MDA-MB-231 cells than fibroblast cells; however, CS-EEO NPs were non-toxic for fibroblast cells and more toxic for MDA-MB-231 cells compared to free EEO. Therefore, the CS-EEO NPs illustrate smart behavior in killing cancerous cells and will be suggested for breast cancer drug delivery. [ABSTRACT FROM AUTHOR]

Abstract

Despite the promising medicinal properties, berberine (BBR), due to its relatively poor solubility in plasma, low bio-stability and limited bioavailability is not used broadly in clinical stages. Due to these drawbacks, drug delivery systems (DDSs) based on nanoscale natural polysaccharides, are applied to address these concerns. Natural polymers are biodegradable, non-immunogenic, biocompatible, and non-toxic agents that are capable of trapping large amounts of hydrophobic compounds in relatively small volumes. The use of nanoscale natural polysaccharide improves the stability and pharmacokinetics of the small molecules and, consequently, increases the therapeutic effects and reduces the side effects of the small molecules. Therefore, this paper presents an overview of the different methods used for increasing the BBR solubility and bioavailability. Afterwards, the pharmacodynamic and pharmacokinetic of BBR nanostructures were discussed followed by the introduction of natural polysaccharides of plant (cyclodextrines, glucomannan), the shells of crustaceans (chitosan), and the cell wall of brown marine algae (alginate)-based origins used to improve the dissolution rate of poorly soluble BBR and their anticancer and antibacterial properties. Finally, the anticancer and antibacterial mechanisms of free BBR and BBR nanostructures were surveyed. In conclusion, this review may pave the way for providing some useful data in the development of BBR-based platforms for clinical applications.

Abstract

The advent of porous materials, in particular zeolitic nanoparticles, has opened up unprecedented putative research avenues in nanomedicine. Zeolites with intracrystal mesopores are low framework density aluminosilicates possessing a regular porous structure along with intricate channels. Their unique physiochemical as well as physiological parameters necessitate a comprehensive overview on their classifications, fabrication platforms, cellular/macromolecular interactions, and eventually their prospective biomedical applications through illustrating the challenges and opportunities in different integrative medical and pharmaceutical fields. More particularly, an update on recent advances in zeolite-accommodated drug delivery and the prevalent challenges regarding these molecular sieves is to be presented. In conclusion, strategies to accelerate the translation of these porous materials from bench to bedside along with common overlooked physiological and pharmacological factors of zeolite nanoparticles are discussed and debated. Furthermore, for zeolite nanoparticles, it is a matter of crucial importance, in terms of biosafety and nanotoxicology, to appreciate the zeolite-bio interface once the zeolite nanoparticles are exposed to the bio-macromolecules in biological media. We specifically shed light on interactions of zeolite nanoparticles with fibrinogen and amyloid beta which had been comprehensively investigated in our recent reports. Given the significance of zeolite nanoparticles’ interactions with serum or interstitial proteins conferring them new biological identity, the preliminary approaches for deeper understanding of administration, distribution, metabolism and excretion of zeolite nanoparticles are elucidated.

Abstract

The wide prevalence of oxidative stress-induced diseases has led to a growing demand for antioxidant therapeutics worldwide. Nanozyme antioxidants are drawing enormous attention as practical alternatives for conventional antioxidants. The considerable body of research over the last decade and the promising results achieved signify the potential of nanozyme antioxidants to secure a place in the expanding market of antioxidant therapeutics. Nonetheless, there is no report on clinical trials for their further evaluation. Through analyzing in-depth selected papers which have conducted in vivo studies on nanozyme antioxidants, this review aims to pinpoint and discuss possible reasons impeding development of research toward clinical studies and to offer some practical solutions for future studies to bridge the gap between preclinical and clinical stages. "We did not experience these kinds of strange illnesses in the past." Everybody might have heard such a familiar sentence from their grandparents and asked themselves, why? The current paper aims to provide readers with one of the answers: "Oxidative stress", which happens when the body fails to neutralize damage caused by unstable molecules called free radicals. In this paper, the authors present the seriousness of oxidative stress-induced clinical conditions. They discuss one of the promising treatments, nanozyme antioxidants, these are mostly based on nano-sized materials with enzyme-like function, in other words, they can speed up chemical reactions. Despite significant results, nanozyme antioxidants have not been investigated in clinical studies. This paper intends to search for the main reasons for this and suggest possible solutions. © 2021 Future Medicine Ltd.

Abstract

Novel shell crosslinked (SCL) pH- and reduction-responsive micelles were fabricated through the grafting of DMAEMA and AA monomers onto a star-shaped PCL through ATRP followed by crosslinking with Cys. The chemical structures of the samples were investigated using 1H NMR and FTIR. The developed micelles were loaded with MTX as an anticancer drug and their drug loading and encapsulation efficiencies were calculated to be 31.53 ± 0.3% for both. The fabricated drug delivery system (DDS) exhibited slow and stimuli-triggered drug release behavior. The anticancer performance of the DDS was examined using MTT assay against MCF7 cells. Abstract

It is becoming increasingly accepted that various diseases have a capacity to alter the composition of plasma proteins. This alteration in protein composition may consequently change the targeting capacity of nanoparticles (NPs). In this study, the impact of a model targeting ligand's (i.e., Transferrin; Tf) concentration in human plasma on the targeting capacity of gold NPs (Au NPs), pre-conjugated with Tf, is investigated. Our findings demonstrate that the protein corona formation by both healthy and Tf depleted human plasma diminishes the targeting efficacy of Au NPs within human cancer cells despite a preservation of targeting ability by plasma with excess Tf (10-fold). Moreover, the plasma samples obtained from patients with various Tf levels (e.g., thalassemia major, sickle cell anemia, aplastic anemia, and iron deficiency anemia) have affected the accessibility of the targeting Tf in the corona layer and subsequently affected their targeting ability, which emphasizes the critical role of disease-specific protein corona on the efficacy of Au NPs. Ultimately, variations of protein concentration (e.g., due to disease occurrence and progress) in plasma affect its recruiting in corona formation, and in turn, affect the targeting and therapeutic efficacies of Au NPs.

Abstract

As a non-invasive method of local and systemic drug delivery, the administration of active pharmaceutical ingredients (APIs) via the pulmonary route represents an ideal approach for the therapeutic treatment of pulmonary diseases. The pulmonary route provides a number of advantages, including the rapid absorption which results from a high level of vascularisation over a large surface area and the successful avoidance of first-pass metabolism. Aerosolization of nanoparticles (NPs) is presently under extensive investigation and exhibits a high potential for targeted delivery of therapeutic agents for the treatment of a wide range of diseases. NPs need to possess specific characteristics to facilitate their transport along the pulmonary tract and appropriately overcome the barriers presented by the pulmonary system. The most challenging aspect of delivering NP-based drugs via the pulmonary route is developing colloidal systems with the optimal physicochemical parameters for inhalation. The physiochemical properties of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) have been investigated as a template for the synthesis of NPs to assist in the formulation of virus-like particles (VLPs) for pharmaceutical delivery, vaccine production and diagnosis assays. Abstract

Nanofibrous electrically conductive scaffolds based on gelatin, namely gelatin-grafted polythiophene (Gel-g-PTh) and gelatin-grafted polythiophene/poly(ε-caprolactone) (Gel-g-PTh/PCL), have been designed and fabricated using the electrospinning technique. The performances of the fabricated electrospun nanofibers as scaffolds in tissue engineering (TE) application were preliminarily investigated in terms of some physicochemical as well as biological features. The results revealed that both fabricated electrospun nanofibers have potential as scaffolds for use in TE applications that require electroactivity. It was found that the co-electrospinning of Gel-g-PTh with a small amount of PCL improves the nanofibers' uniformity as well as some biological characteristics of the resultant scaffold.
