Abstract

Estrogens exert a key biological role in mammary gland epithelial cells and promote breast carcinogenesis and tumor progression. We recently identified a new large set of estrogen responsive genes from breast cancer (BC) cells by DNA microarray analysis of the gene expression profiles induced by 17beta-estradiol in ZR-75.1 and MCF-7 cells. The purpose of the present study was to test whether the expression pattern of hormone regulated genes from this set identifies estrogen receptor (ERalpha) positive, hormone responsive BC cells. To this aim, we carried out in silico metanalysis of ERalpha positive and ERalpha negative human BC cell line transcriptomes, focusing on two sets of 171 and 218 estrogen responsive genes, respectively. Results show that estrogen dependent gene activity in hormone responsive BC cells is significantly different from that of non-responsive cells and, alone, allows to discriminate these two cellular phenotypes. Indeed, we have identified 61 genes whose expression profile specifically marks ERalpha positive BC cells, suggesting that this gene set may be exploited for phenotypic characterization of breast tumors. This possibility was tested with data obtained by gene expression profiling of BC surgical samples, where the ERalpha positive phenotypes were highlighted by the expression profile of a subset of 27 such hormone responsive genes and four additional BC marker genes, not including ERs. These results provide direct evidence that the expression pattern of a limited number of estrogen responsive genes can be exploited to assess the estrogen signaling status of BC cells both in vitro and ex-vivo. 63. Evaluation of VOC measurements in the EXPOLIS study

Abstract

Personal exposures and microenvironment concentrations of 30 target VOCs were measured for 401 participants living in five European cities as a part of the EXPOLIS (Air Pollution Exposure Distributions within Adult Urban Populations in Europe) study. Measurements in Basel used an active charcoal (Carbotech) adsorbent as opposed to the Tenax TA used in the other study centres. In addition, within each centre, personal and microenvironment VOC sampling required different sampling pumps and, because of different sampling durations, different sampling flow rates. Thus, careful testing of the sampling and analysis procedures was required to ensure accuracy and comparability of collected data. Monitor comparison tests using Tenax TA showed a mean VOC concentration ratio of 0.95 between the personal and microenvironment monitors. The LODs for the target VOCs using Tenax TA ranged from 0.7 to 5.2 mug m(-3). The LODs for the 14 target compounds quantifiable using Carbotech ranged from 0.9 to 3.2 mug m(-3). Tenax TA field blanks showed no remarkable contamination with the target VOCs, except benzaldehyde, a known artefact with this adsorbent. Thus, the diffusion barrier system used prevented contamination of Tenax TA samples by passive diffusion during non-sampling periods. Abstract

The paper presents the main results obtained within the Research in Advanced Communications in Europe (RACE) project: 2087 TITAN (Tool for Introduction Scenarios and Techno-Economic Evaluation of The Access Network) regarding minimum-risk introductory routes for new narrowband and broadband services in the residential and small business market. The project developed a methodology and a tool for technoeconomic evaluation of advanced narrowband and broadband telecommunications networks and services. Results from four extensive access network studies are presented. For the narrowband services, optical access and radio in the local loop have been assessed and compared to conventional copper layout in a greenfield environment. Access network upgrade strategies for emerging new broadband services have been evaluated in three studies, with technological options ranging from enhanced copper to hybrid-fiber-coax (HFC) and broadband passive optical networks. An extensive study on different broadband access network upgrade alternatives for public network operators (PNO's) were carried out, taking into account the diversity of conditions met today by European countries. In another study, the effect of competition in the access network was modeled assuming two main operators sharing the market. Broadband upgrade alternatives were evaluated for a new cable operator, competing with the dominant public network operator. In addition, video-on-demand (VoD) and Internet access upgrades have been analyzed. The paper demonstrates the viability of techno-economic studies for the access network, including in-depth analysis of the time-dependence of component cost, tariffs, service-penetrations and market shares. This work intends to support establishing guidelines for strategic decisions regarding the development of the access network alternatives of different operators.

Abstract

TIR8, also known as single Ig IL-1-related receptor, is a member of the IL-1 receptor/Toll-like receptor (TLR) superfamily, which acts as an intracellular decoy for components of the signaling pathway. Here we report that Tir8 has a unique pattern of expression, which includes mucosal tissues and dendritic cells (DC). Tir8-deficient DC showed increased cytokine production in response to TLR agonists (lipopolysaccharide, CpG oligodeoxynucleotides). Tir8-deficient mice had normal susceptibility to systemic lipopolysaccharide toxicity and to i.p. or s.c. inflammation. However, Tir8-deficient mice were more susceptible to intestinal inflammation. Thus, TIR8 represents a negative pathway of regulation of the IL-1 receptor/TLR system, expressed in epithelial cells and DC, crucial for tuning inflammation in the gastrointestinal tract.
