Abstract

A theology of infertility is needed to help couples and the broader ecclesial community understand the theological implications of infertility. Infertility raises questions about human freedom, finitude, embodiment, childlessness, and parenthood. In this article, dominant cultural assumptions surrounding each of these areas when considering reproductive technologies are sketched. Official Roman Catholic teaching on reproductive technologies (Donum Vitae), while rejecting most forms of such technologies, does provide a viable response to the presupposition that reproductive technologies resolve infertility. Given the dominant cultural assumptions and insights from Roman Catholic teaching, this article advocates for several ecclesial changes when considering infertility. Finally, theological resources for developing a theology of infertility are offered. Specifically, insights from Karl Rahner's theology of concupiscence are examined with an eye toward how they provide a framework for rethinking the cultural assumptions about freedom and finitude when considering reproductive technologies. Abstract

PM2.5 is an air pollutant that is widely associated with adverse health effects, and which tends to be disproportionately located near low-income communities and communities of color. We applied a community-engaged research approach to assess the distribution of PM2.5 concentrations in the context of community concerns and urban features within and around the city of Santa Ana, CA. Approximately 183 h of one-minute average PM2.5 measurements, along with high-resolution geographic coordinate measurements, were collected by volunteer community participants using roughly two dozen low-cost AtmoTube Pro air pollution sensors paired with real-time GPS tracking devices. PM2.5 varied by region, time of day, and month. In general, concentrations were higher near the city's industrial corridor, which is an area of concern to local community members. While the freeway systems were shown to correlate with some degree of elevated air pollution, two of four sampling days demonstrated little to no visible association with freeway traffic. Concentrations tended to be higher within socioeconomically disadvantaged communities compared to other areas. This pilot study demonstrates the utility of using low-cost air pollution sensors for the application of community-engaged study designs that leverage community knowledge, enable high-density air monitoring, and facilitate greater health-related awareness, education, and empowerment among communities. The mobile air-monitoring approach used in this study, and its application to characterize the ambient air quality within a defined geographic region, is in contrast to other community-engaged studies, which employ fixed-site monitoring and/or focus on personal exposure. The findings from this study underscore the existence of environmental health inequities that persist in urban areas today, which can help to inform policy decisions related to health equity, future urban planning, and community access to resources. Abstract

As the second millennium draws to a close, leadership educators should examine what can be learned from the evolution of leadership as modern societies have developed, and participate in creating new leadership paradigms for the future. Changes in government, families, knowledge/technology, business, health-care, social activism and religion will necessitate new leadership approaches in Millennium 2000. To begin discussion among leadership educators in preparing for these new paradigms, this article reviews the evolution of leadership, projects changes which may occur during the next century, and proposes alternative administrative, catalytic, collegial, humanitarian/activist, innovative, religiosity, and visionary leadership paradigms which may evolve.

Disorders of sex development (DSD) are a diverse group of conditions in which there is variation from the typical chromosomal, gonadal, or anatomical developmental pathway. While much has been learnt about the genetic aetiology of many of these disorders, a significant proportion of cases remain without a definitive diagnosis. This thesis consists of a series of studies designed to look at different aspects of DSD in order to identify causes and develop better ways to assess and research these conditions in the future. Chapter 1 is an extensive literature review of normal sex development, models to describe the sex phenotype, steroidogenesis, steroid hormone structure and physiological role, classification of DSD, clinical uses of progestogens and determinants of foetal growth. An understanding of these diverse subjects is essential to consider the topics investigated. Chapter 2 presents the rationale for, and specific aims of, this thesis. Chapter 3 describes a study using the I-DSD registry, the largest international register of cases of DSD, to identify associated conditions co-occurring in DSD. 649 cases with documented consent were identified and analysed from the registry, with further information obtained from the reporting clinician where necessary. Associated conditions were reported in 168 cases (26%), overall, and when considered according to karyotype were reported in 112 cases of 46,XY DSD (24%), 27 cases of 46,XX DSD (22%), 19 cases of 45,X/46,XY (45%), and 6 cases of 45,X (75%). In 46,XY DSD, which represents the largest group of cases in the Registry, small for gestational age (SGA), cardiac and CNS anomalies were the most commonly reported associated conditions. This study strengthens the recognised association between SGA and non-specific 46,XY DSD. Additionally, the data indicate a possible association between genetically confirmed AIS and skeletal and renal anomalies. 74. "What Happened to the Post-War Dream?": Nostalgia, Trauma, and Affect in British Rock of the 1960s and 1970s

Abstract

The widespread popularity of British rock musicians in the early 1960s presented the world with a postwar-dream version of Britain as a modern, thriving cultural force in the global economy. Over the course of the decade, however, many of these musicians increasingly turned toward the nation’s war-torn past as a source for musical inspiration. No longer were they only popular culture ambassadors for a modern Britain, but they also came to serve as representatives of a generation scorched by their nation’s history, particularly by the aftershocks of World War II. Although members of pioneering groups such as the Beatles, the Kinks, the Who, and Pink Floyd were all born in the early 1940s—much too late to have had experience on the frontline—memories of World War II haunted them. The rubble left from buildings decimated during the German Blitz attacks, family war stories, and national rationing into the late 1950s were among the constant reminders of near defeat during the war and the struggle to survive afterward amidst a crumbling empire. This dissertation explores how British rock musicians sought to engage with their identities as members of the postwar generation and to challenge their inherited national history. By applying theories addressing identity, memory, and nostalgia from Svetlana Boym, Pierre Nora, and Janelle Wilson, chapter 1 demonstrates how the Beatles in the film and soundtrack Magical Mystery Tour (1967) constructed a world deeply rooted in postwar nostalgia. Drawing on the works of Clay Routledge and Lawrence Kramer, chapter 2 illustrates how the Kinks employed the affective ambiguity of nostalgia as a tool for cultural criticism in their concept album Arthur: Or the Decline and Fall of the British Empire (1969). Theories on trauma and denial by Julie P. Sutton and Judith Herman inform chapter 3, which examines how the Who’s rock opera Tommy (1969) reflects a culture of postwar traumatic denial. Concluding the dissertation, chapter 4 incorporates trauma theory from works by Shoshana Felman, Cathy Caruth, and Maria Cizmic, to show how Pink Floyd’s rock opera The Wall (1979) functions as a traumatic narrative through musical disruption, creates a soundscape built from traumatic events, and emphasizes the role of the listener as witness.

Abstract

Context:\ud The focus of care in disorders of sex development (DSD) is often directed to issues related to sex and gender development. In addition, the molecular etiology remains unclear in the majority of cases.\ud Objective:\ud To report the range of associated conditions identified in the international DSD (I-DSD) Registry.\ud Design, Setting, and Patients:\ud Anonymized data were extracted from the I-DSD Registry for diagnosis, karyotype, sex of rearing, genetic investigations, and associated anomalies. If necessary, clarification was sought from the reporting clinician.\ud Results:\ud Of 649 accessible cases, associated conditions occurred in 168 (26%); 103 (61%) cases had one condition, 31 (18%) had two conditions, 20 (12%) had three conditions, and 14 (8%) had four or more conditions. Karyotypes with most frequently reported associations included 45,X with 6 of 8 affected cases (75%), 45,X/46,XY with 19 of 42 cases (45%), 46,XY with 112 of 460 cases (24%), and 46,XX with 27 of 121 cases (22%). In the 112 cases of 46,XY DSD, the commonest conditions included small for gestational age in 26 (23%), cardiac anomalies in 22 (20%), and central nervous system disorders in 22 (20%), whereas in the 27 cases of 46,XX DSD, skeletal and renal anomalies were commonest at 12 (44%) and 8 (30%), respectively. Of 170 cases of suspected androgen insensitivity syndrome, 19 (11%) had reported anomalies and 9 of these had confirmed androgen receptor mutations.\ud Conclusions:\ud Over a quarter of the cases in the I-DSD Registry have an additional condition. These associations can direct investigators toward novel genetic etiology and also highlight the need for more holistic care of the affected person.

84. Hu8F4-CAR T cells with mutated Fc spacer segment improve target-specificity and mediate anti-leukemia activity in vivo .

Abstract

Hu8F4 is a T cell receptor (TCR)-like antibody with high affinity for leukemia-associated antigen PR1/HLA-A2 epitope. Adapted into a chimeric antigen receptor (CAR) format, Hu8F4-CAR is comprised of the Hu8F4 scFv, the human IgG1 CH2CH3 extracellular spacer domain, a human CD28 costimulatory domain, and the human CD3ζ signaling domain. We have demonstrated high efficacy of Hu8F4-CAR-T cells against PR1/HLA-A2-expressing cell lines and leukemic blasts from AML patients in vitro . Previous studies have shown that modification of the Fc domains of IgG4 CH2CH3 spacer regions can eliminate activation-induced cell death and off-target killing mediated by mouse Fc gamma receptor (FcgR)-expressing cells. We generated Hu8F4-CAR(PQ) with mutated Fc receptor binding sites on the CH2 domain of Hu8F4-CAR to prevent unwanted interactions with FcgR-expressing cells in vivo . The primary human T cells transduced with Hu8F4-CAR(PQ) can specifically lyse HLA-A2 + PR1-expressing leukemia cell lines in vitro . Furthermore, both adult donor-derived and cord blood-derived Hu8F4-CAR(PQ)-T cells are active and can eliminate U937 leukemia cells in NSG mice.
