Background The debilitating human disease schistosomiasis is caused by infection with schistosome parasites that maintain a complex lifecycle alternating between definitive (human) and intermediate (snail) hosts. While much is known about how the definitive host responds to schistosome infection, there is comparably less information available describing the snail's response to infection. Methodology/Principle findings Here, using information recently revealed by sequencing of the Biomphalaria glabrata intermediate host genome, we provide evidence that the predicted core snail DNA methylation machinery components are associated with both intra-species reproduction processes and inter-species interactions. Firstly, methyl-CpG binding domain protein (Bgmbd2/3) and DNA methyltransferase 1 (Bgdnmt1) genes are transcriptionally enriched in gonadal compared to somatic tissues with 5-azacytidine (5-AzaC) treatment significantly inhibiting oviposition. Secondly, elevated levels of 5-methyl cytosine (5mC), DNA methyltransferase activity and 5mC binding in pigmented hybrid-compared to inbred (NMRI)-B. glabrata populations indicate a role for the snail's DNA methylation machinery in maintaining hybrid vigour or heterosis. Thirdly, locus-specific detection of 5mC by bisulfite (BS)-PCR revealed 5mC within an exonic region of a housekeeping protein-coding gene (Bg14-3-3), supporting previous in silico predictions and whole genome BS-Seq analysis of this species' genome. Finally, we provide preliminary evidence for parasite-mediated host epigenetic reprogramming in the schistosome/snail system, as demonstrated by the increase in Bgdnmt1 and Bgmbd2/3 transcript abundance following Bge (B. glabrata embryonic cell line) exposure to parasite larval transformation products (LTP). Conclusions/Significance The presence of a functional DNA methylation machinery in B. glabrata as well as the modulation of these gene products in response to schistosome products, suggests a vital role for DNA methylation during snail development/oviposition and parasite interactions. Further deciphering the role of this epigenetic process during Biomphalaria/Schistosoma co-evolutionary biology may reveal key factors associated with disease transmission and, moreover, enable the discovery of novel lifecycle intervention strategies.

International audience; Les modifications épigénétiques sont transmises au cours des générations, cependant le mode de transmission est inconnu pour la plupart des organismes. Notre objectif c’est de comprendre l’implication des modifications épigénétiques dans le succès du parasite, et de déterminer ainsi leur héritabilité (mendélienne ou non-mendélienne) dans le modèle Schistososma mansoni avec son hôte intermédiaire Biomphalaria glabrata. Dans ce but, nous avons effectué des croisements entre deux isolats géographiques différents de Schistosoma mansoni. Le phénotype des hybrides de trois générations successives a été analysé au niveau des traits de vie, mais aussi au niveau moléculaire. Pour cela, nous avons focalisé sur une famille multigénique (SmPoMucs : Schistosoma mansoni Polymorphic Mucins), qui est essentielle pour le succès d’infestation du parasite et qui son expression est contrôlée par des marques épigénétiques. Nos résultats préliminaires suggèrent que la structure de la chromatine au niveau de ces gènes candidats est transmise plutôt d’une façon non-mendélienne. Nous espérons contribuer par ce travail à une meilleure compréhension des origines de la forte adaptation du parasite sur ses hôtes, dans le but de lutter contre cette maladie

Tout le monde a entendu parler de la théorie de l'évolution, mais en quoi consiste-t-elle vraiment ? De nos jours, elle est décrite comme un phénomène lent, durant lequel les individus disposant des meilleures caractéristiques (apparues aléatoirement par des mutations) sont favorisés, survivent, se reproduisent et transmettent ces caractéristiques à leur descendance. Indissociable de Charles Darwin et de son concept de sélection naturelle, cette théorie tire ses sources dans "L'origine des espèces" publié en 1859. Cependant, 50 ans avant lui, Jean-Baptiste Lamarck présentait la première théorie véritablement scientifique de l'évolution : le transformisme. Il défend l'hypothèse que les individus adaptent leurs caractéristiques à leur environnement, et ces adaptatations sont transmises à la descendance. C'est l'image de l'animal, qui petit à petit, de générations en générations, étire son cou pour manger les feuilles de l'arbre et devient au final une girafe. Cette vision de l'évolution est globalement réfutée et le nom de Lamarck est souvent synonyme de moqueries. Pourtant, ce ridicule est il vraiment mérité ? International audience; The digenetic trematode Schistosoma mansoni is a human parasite that uses the mollusc Biomphalaria glabrata as intermediate host. Specific S. mansoni strains can infect efficiently only certain B. glabrata strains (compatible strain) while others are incompatible. Strain-specific differences in transcription of a conserved family of polymorphic mucins (SmPoMucs) in S. mansoni are the principle determinants for this compatibility. In the present study, we investigated the bases of the control of SmPoMuc expression that evolved to evade B. glabrata diversified antigen recognition molecules. We compared the DNA sequences and chromatin structure of SmPoMuc promoters of two S. mansoni strains that are either compatible (C) or incompatible (IC) with a reference snail host. We reveal that although sequence differences are observed between active promoter regions of SmPoMuc genes, the sequences of the promoters are not diverse and are conserved between IC and C strains, suggesting that genetics alone cannot explain the evolution of compatibility polymorphism. In contrast, promoters carry epigenetic marks that are significantly different between the C and IC strains. Moreover, we show that modifications of the structure of the chromatin of the parasite modify transcription of SmPoMuc in the IC strain compared to the C strain and correlate with the presence of additional combinations of SmPoMuc transcripts only observed in the IC phenotype. Our results indicate that transcription polymorphism of a gene family that is responsible for an important adaptive trait of the parasite is epigenetically encoded. These strain-specific epigenetic marks are heritable, but can change while the underlying genetic information remains stable. This suggests that epigenetic changes may be important for the early steps in the adaptation of pathogens to new hosts, and might be an initial step in adaptive evolution in general

Phenotypic variability is defined as the capacity of a given population to produce phenotypic variants under theinfluence of the environment. Some of the phenotypic variants are heritable. It is generally assumed that thegenetic variations are the sole source of heritable phenotypic variants. However, recent studies have shown thatepigenetic variations can provide alternative source for phenotypic variants without change in DNA sequence. Inhost / parasite interactions, parasites impose selective pressures on their hosts and vice versa, leading to a genuinearms race between both partners. Such interaction leads to rapid adaptation where each partner has to evolve thecapacity to express new phenotypic variants. We propose that epigenetic variations play an important role in thegenesis of phenotypic variability. Schistosoma mansoni is a human parasite that causes intestinal schistosomiasis.During the PhD project, I was interested in the interaction of S. mansoni with its intermediate host Biomphalariaglabrata. The two main goals of this PhD project were: (i) To determine the relative weight of epigenetics andgenetics in the expression of phenotypic variants in the parasite. (ii) To initiate the investigation of epigeneticmechanisms in the host.The results show that histone modifications i.e. changes in the epigenetic information are indeed a source ofphenotypic variants in S. mansoni. These phenotypic variants confer a higher fitness to the parasite, by increasingits compatibility towards the mollusk. Finally, studying the heritability of epigenetic changes showed a nonmendeliansegregation.
