Abstract

An integrated methodology is proposed for the design of polluted water control systems for road infrastructures, based on hazard analysis concepts. The case of a simultaneous rainstorm and road accident with the dispersion of dangerous substances is considered. The risk function is computed with regard to the objective evaluation of severity of the event and to the stochastic calculation of the associate probability. A logical framework for the evaluation of severity is proposed by taking into consideration four possible circumstances for the dispersion of pollutants: surface and ground water, and natural and urban soil. A set of check lists have been devised for the assessment and design of the environmental protection control systems. The methodological approach is completed with the formulation of a stochastic model representing the statistical combination of the occurrence of the hydrological processes and the road accident dynamics. The stochastic model estimates the expected volume of contaminated rainfall for any defined probability of occurrence of the polluting event, i.e., return time of the hazard, in a coherent statistical setting. The proposed integrated methodology has been applied to a real case presently under construction: the Salerno-Reggio Calabria Highway located in Southern Italy. Abstract

Circulating biomarkers of myocardial damage represent a cornerstone for the diagnosis and for the stratification of patient’s risk after Acute Myocardial Infarction (AMI). The present work presents a novel mathematical model for the dynamic analysis of cardiac troponin T (cTnT) circulating into the bloodstream. Tests on patients’ data show the model is potentially useful, enabling the extraction of important clinical information.

61. Noise propagation in chemical reaction networks: Analysis of a molecular subtractor module

Abstract

The realization of embedded molecular control systems is a challenging aim in Synthetic Biology, where a major goal is to design synthetic biological circuits performing specific tasks. In this field, the novel emergent approach is to assemble the circuit in a modular fashion, possibly restraining reciprocal interactions from interconnected modules (zero-retroactivity). Within this framework, recent results have been proposed, dealing with the realization of an embedded subtractor module, with the idea of exploiting it in a more general chemical reaction network that resembles a classical control scheme. So far, this research has been carried out according to the deterministic approach. More sophisticated analysis requires the use of stochastic models, which play a paramount role in investigating noise propagation in chemical reaction networks, especially when the species copy number is low and the intrinsic stochasticity of the phenomena under investigation cannot be neglected. This note deals with a first analysis of the subtractor module, according to the stochastic approach. To this end, Chemical Master Equations are exploited to model one of the possible molecular circuits implementing the subtractor, and moment equations are written in order to evaluate how noise propagates with respect to different values of the inputs and different model parameter settings.

Abstract

Talks on rare diseases in the field of neurology often start with a statement like this: “About 80% of all rare diseases have a neurologic manifestation and about 80% of those are genetic in origin.” Although these numbers probably represent more of an estimate than well-documented evidence, rapidly advancing and cost-effective sequencing technologies have led to the quickly growing identification of patients with hereditary neurological diseases. Although the importance of genetics for diagnosis and genetic counseling is undisputed, the recent development of first genetargeted therapies entering clinical trial1,2 is adding an important new layer to the (re-)consideration of genetic testing in neurology. However, establishing accurate genotype– phenotype and genotype–treatment relationships requires large sample sizes. Systematic reviews can serve as instruments to combine information from several small samples, but unfortunately, this is often complicated by inconsistent and incomplete reporting of clinical and genetic data across studies. Thus, large multicenter approaches are necessary to systematically and uniformly characterize patients with genetic neurologic conditions and to eventually establish sizable clinical trial-ready cohorts.

Abstract

Introduction. The focus of this study is the paleoclimatic reconstruction of the southern Tyrrhenian between ~9.2 and 2.9 ka, through the study of planktonic foraminiferal assemblages and stable isotopes, and comparing data with other coeval intervals. Several authors have studied the climatic sensitivity of Holocene planktonic foraminifera in different parts of the Mediterranean. Planktonic foraminifera produce good records of Holocene climatic variability, especially as regards the suborbital events such as Bond events (Bond et al., 1997) and other cooling/warming oscillations. Therefore, the obtained eco-biostratigraphy has allowed us to analyze how climatic forcing influenced sea surface temperature (SST) and water column structure during the Holocene in this sector of the southern Tyrrhenian Sea. Methodology. A sedimentary core (196 cm long) was collected in the Gulf of Palermo at the base of the upper continental slope (990 m bsl) and sub-sampled every 2 cm. Micropaleontological and geochemical analyses were performed on 98 samples, and three AMS 14C dates were determined. Micropaleontological analyses consisted of qualitative and quantitative characterization of the planktonic foraminiferal assemblages, in the size fraction greater than 125 μm. Geochemical analyses were performed on 8–10 specimens of Globigerina bulloides. Results. The calibrated AMS 14C ages, together with planktonic foraminiferal fluctuations and G. bulloides oxygen isotope records, were used to develop an age model of the studied interval. In order to obtain additional age control points, the studied records were also tuned to the NGRIP δ18O (GICC05) (Svensson et al., 2008) and GISP2 ice core temperature (Alley, 2000) records. Twenty species and eco-morphotypes were recognized in the planktonic foraminiferal assemblage and grouped depending upon their climatic and feeding affinity. The warm-water species, minus the typical cold-water species, were used to obtain the paleoclimatic curve, whilst the herbivorous/ carnivorous ratio has permitted us to reconstruct the trophism of the southern Tyrrhenian. In particular, between ~8 and 6.4 ka, an important warming phase was recognized, clearly indicated by the increase of Globigerinoides gr. ruber and G. gr. quadrilobatus. This interval corresponds to the deposition of Sapropel S1b-equivalent stage (Sprovieri et al., 2003; Lirer et al., 2013; Siani et al., 2013), when high SSTs and oligotrophy characterized the entire Mediterranean region, especially during the summer seasons. Between ~5.9 and 4.2 ka, another warming phase occurred in the Gulf of Palermo, differing from the previous one due to the enhanced winter mixing of the water column, testified by the high abundance of deep dweller species, such as Globorotalia truncatulinoides and Globorotalia inflata left coilings. The last important warming phase corresponds to the Minoan Warm Period, briefly interrupted by a rapid cooling event (at ~3.5 ka). Among these warm climatic phases, several rapid cold-cool events were recognized. In particular, Neogloboquadrina pachyderma and Turborotalita quinqueloba represent the best cold-water indicators, increasing in abundance during the Bond events (B2, B3, B4, B5, B6) and other rapid cooling events that characterized the western (M3, M4, M5, M6, M7) (Frigola et al., 2007) and central Mediterranean (TC2, TC3, TC4) (Cacho et al., 2001) during the 9.2–2.9 ka interval. Conclusions. Planktonic foraminifera and oxygen isotope variations highlight the climatic oscillations of the relatively stable Holocene stage. These oscillations are linked to the North Atlantic millennial scale climatic variability that forced cool/high productivity periods and warm/oligotrophic conditions. In particular, during the ~8–6.4 (S1b-equivalent), ~5.9–4.2, and ~3.7–3.2 (Minoan Warm Period) ka intervals, warm surface waters characterized the Gulf of Palermo. On the contrary, during rapid climatic phases, centered at 9.1, 8.2, 7.2, 6.2–5.9, 5.4, 4.7, 3.9, 3.5, and 3 ka, cool/high productivity conditions occurred in this sector of the southern Tyrrhenian. Abstract

In this paper, we investigate some finite-time control problems for stochastic linear time-varying systems, described by an Itô type differential equation. In particular, the annular stochastic finite-time stability control problem is dealt with. In this context, it is required that the norm of the state variables remains bounded between an inner and an outer ellipsoid, during a finite-interval of time. The first contribution of the paper consists of two sufficient conditions for stability, which are derived by exploiting an approach based on time-varying quadratic Lyapunov functions. The first condition requires the solution of two generalised differential Lyapunov equations; the latter the existence of a feasible solution to a pair of differential linear matrix inequalities. As particular cases, we obtain sufficient conditions for stochastic finite-time stability (in this last case, the lower ellipsoid collapses to the origin). From the analysis conditions, a sufficient condition is derived for the annular stochastic finite-time stabilisation via both state and output feedback. Some numerical examples illustrate the application of the proposed methodology and show that our approach attains less conservative results than those obtainable with the existing literature. Moreover, an application of the annular stochastic finite-time stability approach to the stabilisation problem of a satellite with respect to the geomagnetic field is presented. Abstract

In this paper we investigate the stabilization over a finite region of two-dimensional (2D)-systems. Such class of systems plays an important role in the biomedical engineering context, with particular reference to the field of image processing. Often 2D-systems are defined over a finite domain of the plane, where each independent variable attains values into a given interval (for example they represent the coordinates of the pixels of a given image). On the basis of these considerations, it is quite straightforward the idea to exploit for 2D-systems the finite-time stability (FTS) theory developed in the context of the classical one-dimensional system framework. Indeed the FTS approach is a more practical concept than classical Lyapunov asymptotic stability, useful to study the behaviour of a system within a finite (possibly short) interval, and therefore it finds application whenever it is desired that the state variables do not exceed a given threshold during the transients.

Abstract

The control of biochemical processes is a major goal in many fields, encompassing metabolic engineering and synthetic biology. Current approaches are based on ad-hoc designs, whereas a general and modular framework would be highly desirable, in order to exploit the well-assessed methods of control theory. A well-known problem when dealing with complex biosystems is represented by the retroactivity effect, which can significantly modify the dynamics of interconnected subsystems, with respect to the behaviour they exhibit when disconnected from each other. In the present work an implementation of a zero retroactivity Chemical Reaction Network Subtractor (CRNS) is proposed and its effectiveness is investigated through singular perturbations analysis. The proposed CRNS represents a first step towards the modular designing of embedded feedback biochemical controllers.

Abstract

Nonlinear dynamical systems theory is a suited framework to describe and analyse, rigorously, cellular state transitions. The basic idea is to represent cellular states as attractors of dynamical systems. However, due to the inherent complexity of biological networks, there is a lack of general methods for the characterisation of the parameters and state spaces of these mathematical models. This work presents a novel approach, based on the combination of several nonlinear analysis techniques, for the investigation of biological bistable switches. A crucial feature of the proposed approach is the capability to perform a robust analysis, i.e. it can take into account the uncertainty of biochemical parameters (deriving from interindividual variability) and the intrinsic environmental noise (which causes fluctuations of the species concentrations).

Abstract

Multigene delivery and subsequent cellular expression is emerging as a key technology required in diverse research fields including, synthetic and structural biology, cellular reprogramming and functional pharmaceutical screening. Current viral delivery systems such as retro- and adenoviruses suffer from limited DNA cargo capacity, thus impeding unrestricted multigene expression. We developed MultiPrime, a modular, non-cytotoxic, non-integrating, baculovirus-based vector system expediting highly efficient transient multigene expression from a variety of promoters. MultiPrime viruses efficiently transduce a wide range of cell types, including non-dividing primary neurons and induced-pluripotent stem cells (iPS). We show that MultiPrime can be used for reprogramming, and for genome editing and engineering by CRISPR/Cas9. Moreover, we implemented dual-host-specific cassettes enabling multiprotein expression in insect and mammalian cells using a single reagent. Our experiments establish MultiPrime as a powerful and highly efficient tool, to deliver multiple genes for a wide range of applications in primary and established mammalian cells., Current viral gene delivery systems are limited in the amount of foreign DNA they can deliver to cells. Here the authors develop MultiPrime, a baculovirus-based vector system capable of multigene delivery into a wide variety of cells, and use Multiprime for genome engineering by CRISPR/Cas9.

Abstract

The neutron time of flight (n-TOF) facility at CERN is a spallation source characterized by a white neutron spectrum. The innovative features of the facility, in the two experimental areas, (20 m and 185 m), allow for an accurate determination of the neutron cross section for radioactive samples or for isotopes with small neutron capture cross section, of interest for Nuclear Astrophysics. The recent results obtained at n-TOF facility are presented.

The clinical efficacy of SLIT has been established in a number of clinical trials and meta-analyses. However, because SLIT is self-administered by patients without medical supervision, the degree of patient adherence with treatment is still a concern. The objective of this study was to evaluate the perception by allergists of issues related to SLIT adherence.Methods: We performed a questionnaire-based survey of 296 Italian allergists, based on the adherence issues known from previous studies. The perception of importance of each item was assessed by a VAS scale ranging from 0 to 10.Results: Patient perception of clinical efficacy was considered the most important factor (ranked 1 by 54% of allergists), followed by the possibility of reimbursement (ranked 1 by 34%), and by the absence of side effects (ranked 1 by 21%). Patient education, regular follow-up, and ease of use of SLIT were ranked first by less than 20% of allergists.Conclusion: These findings indicate that clinical efficacy, cost, and side effects are perceived as the major issues influencing patient adherence to SLIT, and that further improvement of adherence is likely to be achieved by improving the patient information provided by prescribers.Keywords: adherence, sublingual immunotherapy, efficacy, cost, side effects

The use of by-products in animal nutrition is known since long time and considerable importance, from a quantitative point of view, is gived to straws from cereals in particular (Cianci et al., 1982; Vazhapilly et al., 1984). Straws are used as litter material or in paper industry but they are also utilised in ruminant nutrition even though the very low content in fermentable sugars. Dietary fat is one of the three main energy providing macronutrient groups. Humans require a certain amount of polyunsaturated fatty acids which are essential for life and health, but which cannot be synthesised in the body. These are the essential fatty acids.
