Abstract

Background: Epidemiological studies that examined the association between Parkinson's disease (PD) and cancers led to inconsistent results, but they face a number of methodological difficulties. Objective: We used results from genome-wide association studies (GWASs) to study the genetic correlation between PD and different cancers to identify common genetic risk factors. Methods: We used individual data for participants of European ancestry from the Courage-PD (Comprehensive Unbiased Risk Factor Assessment for Genetics and Environment in Parkinson's Disease; PD, N = 16,519) and EPITHYR (differentiated thyroid cancer, N = 3527) consortia and summary statistics of GWASs from iPDGC (International Parkinson Disease Genomics Consortium; PD, N = 482,730), Melanoma Meta-Analysis Consortium (MMAC), Breast Cancer Association Consortium (breast cancer), the Prostate Cancer Association Group to Investigate Cancer Associated Alterations in the Genome (prostate cancer), International Lung Cancer Consortium (lung cancer), and Ovarian Cancer Association Consortium (ovarian cancer) (N comprised between 36,017 and 228,951 for cancer GWASs). We estimated the genetic correlation between PD and cancers using linkage disequilibrium score regression. We studied the association between PD and polymorphisms associated with cancers, and vice versa, using cross-phenotypes polygenic risk score (PRS) analyses. Results: We confirmed a previously reported positive genetic correlation of PD with melanoma (Gcorr = 0.16 [0.04; 0.28]) and reported an additional significant positive correlation of PD with prostate cancer (Gcorr = 0.11 [0.03; 0.19]). There was a significant inverse association between the PRS for ovarian cancer and PD (odds ratio [OR] = 0.89 [0.84; 0.94]). Abstract

Background: Previous studies showed that lifestyle behaviors (cigarette smoking, alcohol, coffee) are inversely associated with Parkinson's disease (PD). The prodromal phase of PD raises the possibility that these associations may be explained by reverse causation. Objective: To examine associations of lifestyle behaviors with PD using two-sample Mendelian randomisation (MR) and the potential for survival and incidence-prevalence biases. Methods: We used summary statistics from publicly available studies to estimate the association of genetic polymorphisms with lifestyle behaviors, and from Courage-PD (7,369 cases, 7,018 controls; European ancestry) to estimate the association of these variants with PD. We used the inverse-variance weighted method to compute odds ratios (ORIVW) of PD and 95%confidence intervals (CI). Significance was determined using a Bonferroni-corrected significance threshold (p = 0.017). Results: We found a significant inverse association between smoking initiation and PD (ORIVW per 1-SD increase in the prevalence of ever smoking = 0.74, 95%CI = 0.60-0.93, p = 0.009) without significant directional pleiotropy. Associations in participants =67 years old and cases with disease duration =7 years were of a similar size. No significant associations were observed for alcohol and coffee drinking. In reverse MR, genetic liability toward PD was not associated with smoking or coffee drinking but was positively associated with alcohol drinking. Conclusion: Our findings are in favor of an inverse association between smoking and PD that is not explained by reverse causation, confounding, and survival or incidence-prevalence biases. Genetic liability toward PD was positively associated with alcohol drinking. Conclusions on the association of alcohol and coffee drinking with PD are hampered by insufficient statistical power.

Abstract

Background: Previous prospective studies highlighted dairy intake as a risk factor for Parkinson's disease (PD), particularly in men. It is unclear whether this association is causal or explained by reverse causation or confounding. Objective: The aim is to examine the association between genetically predicted dairy intake and PD using two-sample Mendelian randomization (MR). Methods: We genotyped a well-established instrumental variable for dairy intake located in the lactase gene (rs4988235) within the Courage-PD consortium (23 studies; 9823 patients and 8376 controls of European ancestry). Results: Based on a dominant model, there was an association between genetic predisposition toward higher dairy intake and PD (odds ratio [OR] per one serving per day = 1.70, 95% confidence interval = 1.12–2.60, P = 0.013) that was restricted to men (OR = 2.50 [1.37–4.56], P = 0.003; P-difference with women = 0.029). Conclusions: Using MR, our findings provide further support for a causal relationship between dairy intake and higher PD risk, not biased by confounding or reverse causation. Further studies are needed to elucidate the underlying mechanisms. © 2022 International Parkinson and Movement Disorder Society.

Abstract

Background: Genetic variability in LRRK2 has been unequivocally established as a major risk factor for familial and sporadic forms of PD in ethnically diverse populations. Objectives: To resolve the role of LRRK2 in the Indian population. Methods: We performed targeted resequencing of the LRRK2 locus in 288 cases and 298 controls and resolved the haplotypic structure of LRRK2 in a combined cohort of 800 cases and 402 controls in the Indian population. We assessed the frequency of novel missense variants in the white and East Asian population by leveraging exome sequencing and densely genotype data, respectively. We did computational modeling and biochemical approach to infer the potential role of novel variants impacting the LRRK2 protein function. Finally, we assessed the phosphorylation activity of identified novel coding variants in the LRRK2 gene. Results: We identified four novel missense variants with frequency ranging from 0.0008% to 0.002% specific for the Indian population, encompassing armadillo and kinase domains of the LRRK2 protein. A common genetic variability within LRRK2 may contribute to increased risk, but it was nonsignificant after correcting for multiple testing, because of small cohort size. The computational modeling showed destabilizing effect on the LRRK2 function. In comparison to the wild-type, the kinase domain variant showed 4-fold increase in the kinase activity. Conclusions: Our study, for the first time, identified novel missense variants for LRRK2, specific for the Indian population, and showed that a novel missense variant in the kinase domain modifies kinase activity in vitro. © 2018 International Parkinson and Movement Disorder Society.

Abstract

This paper presents competitive algorithms for a novel class of online optimization problems with memory. We consider a setting where the learner seeks to minimize the sum of a hitting cost and a switching cost that depends on the previous $p$ decisions. This setting generalizes Smoothed Online Convex Optimization. The proposed approach, Optimistic Regularized Online Balanced Descent, achieves a constant, dimension-free competitive ratio. Further, we show a connection between online optimization with memory and online control with adversarial disturbances. Abstract

We report the discovery and analysis of the planetary microlensing event OGLE-2017-BLG-0406, which was observed both from the ground and by the ${\it Spitzer}$ satellite in a solar orbit. At high magnification, the anomaly in the light curve was densely observed by ground-based-survey and follow-up groups, and it was found to be explained by a planetary lens with a planet/host mass ratio of $q=7.0 \times 10^{-4}$ from the light-curve modeling. The ground-only and ${\it Spitzer}$-"only" data each provide very strong one-dimensional (1-D) constraints on the 2-D microlens parallax vector $\bf{\pi_{\rm E}}$. When combined, these yield a precise measurement of $\bf{\pi_{\rm E}}$, and so of the masses of the host $M_{\rm host}=0.56\pm0.07\,M_\odot$ and planet $M_{\rm planet} = 0.41 \pm 0.05\,M_{\rm Jup}$. The system lies at a distance $D_{\rm L}=5.2 \pm 0.5 \ {\rm kpc}$ from the Sun toward the Galactic bulge, and the host is more likely to be a disk population star according to the kinematics of the lens. The projected separation of the planet from the host is $a_{\perp} = 3.5 \pm 0.3 \ {\rm au}$, i.e., just over twice the snow line. The Galactic-disk kinematics are established in part from a precise measurement of the source proper motion based on OGLE-IV data. A study of excitation dynamics and two-photon properties in new branched poly(p-phenylenevinylene) structures is presented, which are viewed as interlinked structural units producing localized interacting electronic excitations. It was found that PPV derivatives exhibited strong two-photon pumped up-conversion emission when excited by both nanosecond and femtosecond laser pulses.

Abstract

Relative spacecraft navigation based on Global Navigation Satellite System (GNSS) has been already successfully performed in low earth orbit (LEO). Very high accuracy, of the order of the millimeter, has been achieved in postprocessing using carrier phase differential GNSS (CDGNSS) and recovering the integer number of wavelength (Ambiguity) between the GNSS transmitters and the receiver. However the performance achievable on-board, in real time, above LEO and the GNSS constellation would be significantly lower due to limited computational resources, weaker signals, and worse geometric dilution of precision (GDOP). At the same time, monocular vision provides lower accuracy than CDGNSS when there is significant spacecraft separation, and it becomes even lower for larger baselines and wider field of views (FOVs). In order to increase the robustness, continuity, and accuracy of a real-time on-board GNSS-based relative navigation solution in a GNSS degraded environment such as Geosynchronous and High Earth Orbits, we propose a novel navigation architecture based on a tight fusion of carrier phase GNSS observations and monocular vision-based measurements, which enables fast autonomous relative pose estimation of cooperative spacecraft also in case of high GDOP and low GNSS visibility, where the GNSS signals are degraded, weak, or cannot be tracked continuously. In this paper we describe the architecture and implementation of a multi-sensor navigation solution and validate the proposed method in simulation. We use a dataset of images synthetically generated according to a chaser/target relative motion in Geostationary Earth Orbit (GEO) and realistic carrier phase and code-based GNSS observations simulated at the receiver position in the same orbits. We demonstrate that our fusion solution provides higher accuracy, higher robustness, and faster ambiguity resolution in case of degraded GNSS signal conditions, even when using high FOV cameras.

Abstract

We report the discovery of a cold Super-Earth planet (m(p) = 4.4 +/- 0.5 M-circle plus) orbiting a low-mass (M = 0.23 +/- 0.03 M-circle dot) M dwarf at projected separation a(perpendicular to) l = 1.18 +/- 0.10 a.u., i.e., about 1.9 times the distance the snow line. The system is quite nearby for a microlensing planet, D-L = 0.86 +/- 0.09 kpc. Indeed, it was the large lens-source relative parallax pi(rel) = 1.0 mas (combined with the low mass M) that gave rise to the large, and thus well-measured, "microlens parallax" pi(E) proportional to (pi(rel)/M)(1)(/2) that enabled these precise measurements. OGLE-2017-BLG-1434Lb is the eighth microlensing planet with planet-host mass ratio q < 1 x 10(-4).\ud \ud We apply a new planet-detection sensitivity method, which is a variant of "V/V-max", to seven of these eight planets to derive the mass-ratio function in this regime. We find dN/ d lnq proportional to q(P) , with p =1.05(-0.68)(+0.78), which confirms the "turnover" in the mass function found by Suzuki et al. relative to the power law of opposite sign n = -0.93 +/- 0.13 at higher mass ratios q greater than or similar to 2 x 10(-4). We combine our result with that of Suzuki et al. to obtain p = 0.73(-0.34)(+0.42.)
