57. An arf1delta synthetic lethal screen identifies a new clathrin heavy chain conditional allele that perturbs vacuolar protein transport in Saccharomyces cerevisiae

Abstract

ADP-ribosylation factor (ARF) is a small GTP-binding protein that is thought to regulate the assembly of coat proteins on transport vesicles. To identify factors that functionally interact with ARF, we have performed a genetic screen in Saccharomyces cerevisiae for mutations that exhibit synthetic lethality with an arf1[Delta] allele and defined seven genes by complementation tests (SWA1-7 for synthetically lethal with arf1[Delta]). Most of the swa mutants exhibit phenotypes comparable to arf1[Delta] mutants such as temperature-conditional growth, hypersensitivity to fluoride ions, and partial protein transport and glycosylation defects. Here, we report that swa5-1 is a new temperature-sensitive allele of the clathrin heavy chain gene (chc1-5), which carries a frameshift mutation near the 3[prime] end of the CHC1 open reading frame. This genetic interaction between arf1 and chc1 provides in vivo evidence for a role for ARF in clathrin coat assembly. Surprisingly, strains harboring chc1-5 exhibited a significant defect in transport of carboxypeptidase Y or carboxypeptidase S to the vacuole that was not observed in other chc1 is mutants. The kinetics of invertase secretion or transport of alkaline phosphatase to the vacuole were not significantly affected in the chc1-5 mutant, further implicating clathrin specifically in the Golgi to vacuole transport pathway for carboxypeptidase Y.

59. 98 Evaluation of bacteria antibiotic resistance in wastewater sludge from hospitals

Abstract

In hospital wastewater treatment plants, some bacteria and heavy metals are accumulated by direct or indirect sedimentation, which may influence the characteristics of bacteria antibiotic resistance. In the research, three kinds of hospital wastewater sludges were collected for subsequently analysis. A BD Phoenix-100 automated interpretation system was used to identify the bacteria species and antibiotic resistance. The metal compositions were analyzed by using ICP-OES.The results showed that most of the identified bacteria were gram positive coccus, i.e., Streptococcus agalactiae, Enterococcus faecalis, and Staphylococcus hominis. The sludge was found to contain high concentration of metals and most of them exceeded the national Water Quality Standards, for instance: Aluminum was averaged 4300 (mg/kg), Chromium 25 (mg/kg), Copper 70(mg/kg), Iron 6140(mg/kg), Nickel 16(mg/kg), Lead 29(mg/kg), and Zinc 618(mg/kg). Bacillus cereuswas found to resist antibiotic agents such as AM, FOX, and P; Streptococcus agalactiaewas found to resist to FA and GM; Enterococcus faecaliswas found to resist to FOX, CC, FA, GM, P, QDA, and SXT; Staphylococcus hominiswas found to resist to AM, FOX, OX, P, and SXT. Bacteria increase antibiotic resistance through cross-resistance or co-resistance mechanisms when encountered the environmental stress.From previous studies, when bacteria expose to excessive concentrations of heavy metals will stimulate the bacteria mutation and force bacteria to alter their genetic to survive, contributing to an increase the antibiotics resistance. Employees who work in wastewater treatment plants are recommended to wear respirators to avoid exposure to suspended sludge aerosols.

The optical feedback dynamics of two multimode InAs/GaAs quantum dot lasers emitting exclusively on sole ground or excited lasing states is investigated under the short delay configuration. Although the two lasers are made from the same active medium, their responses to the external perturbation are found not much alike. By varying the feedback parameters, various periodic and chaotic oscillatory states are unveiled. The ground state laser is found to be much more resistant to optical feedback, benefitting from its strong relaxation oscillation damping. In contrast, the excited state laser can easily be driven into very complex dynamics. While the ground state laser is of importance for the development of isolator-free transmitters, the excited one is essential for applications taking advantages of chaos such as chaos lidar, chaos radar, and random number generation.

Abstract

Food loss and waste have become a critical problem worldwide. Determining an appropriate promotional strategy for reducing waste and environmental impacts to ensure sustainability is essential in supply chains (SCs). Considering quality losses, this paper proposes an on-site direct-sale dynamic SC inventory model involving one retailer and one contracted supply centre for perishable foods. This model enables the determination of optimal price markdown decisions while maximizing the total channel profit. Regularly priced and markdown products are substitutable during the markdown period, and their demand rates are dependent on price, quality, and time. The optimal solution for the price markdown timing is derived analytically, and a sensitivity analysis of various model parameters is conducted. Numerical studies are also presented to illustrate the effectiveness of the theoretical results; three scenarios with different types of customers served by the retailer are considered (general customers, product-quality-oriented customers, and price-oriented customers). Abstract

1. Introduction MicroRNAs (miRNAs) are small noncoding RNAs found in plants and animals which downregulate gene expression by binding to complementary sites on the 3' untranslated region (UTR) of target [...], MicroRNAs (miRNAs) downregulate gene expression by binding to the partially complementary sites in the 3' untranslated region (UTR) of target mRNAs. Several methods, such as Northern blot analysis, quantitative real-time RT-PCR, microarray, and the luciferase reporter system, are commonly used to quantify the relative level or activity of miRNAs. The disadvantage of these methods is the requirement for cell lysis, which means that several sets of wells/dishes of cells must be prepared to monitor changes in miRNA activity in time-course studies. In this study, we developed a multisampling reporter system in which two secretable bioluminescence-generating enzymes are employed, one as a reporter and the other as an internal control. The reporters consist of a pair of vectors containing the Metridia luciferase gene, one with and one without a duplicated miRNA targeting sequence at their 3'UTR, while the other vector coding for the secreted alkaline phosphatase gene is used as an internal control. This method allows miRNA activity to be monitored within the same population of cells over time by withdrawing aliquots of the culture medium. 76. Intra-auricular modification of facelift incision decreased the risk of Frey syndrome.

Abstract

Objective: Frey syndrome is a complication followed by parotidectomy which caused gustatory sweating and facial flush. There were several methods for the prevention of Frey syndrome, but most of them had no obvious effects. In this study, we compare the intra-auricular modification of facelift incision with the traditional lazy-S incision to see if it can decrease the risk of Frey syndrome. Materials and Methods: This is a retrospective study. From 2003 to 2009, a total of 61 patients with benign parotid tumor who received parotidectomy at Hualien Tzu Chi Hospital and were followed at outpatient department for at least 5 years were enrolled. Patients were divided into two groups according to the type of incisions during operation: (1) Group M: intra-auricular modification of facelift incision or (2) Group S: traditional lazy-S incision. All patients received the partial thickness sternocleidomastoid muscle flap. Clinical data including age, gender, pathologic result, presentation of Frey syndrome, size of tumor, length of operation, blood loss from surgery, length of placement of drain, total amount of drainage, and length of stay were collected and analyzed. Results: Sixty-one patients were enrolled. Eighteen patients were in Group M and forty-three were in Group S. There was no significant difference of age, gender, and size of tumor between the two groups. The pathologic results included parotitis, pleomorphic adenoma, Warthin's tumor, and others. No significant difference of pathologic results, blood loss from surgery, length of placement of drain, total amount of drainage, and length of stay between two groups was obtained. The length of operation was longer in Group M (P = 0.001) and the incidence of Frey syndrome was lower in Group M than Group S (P < 0.05). Conclusions: The use of intra-auricular modification of facelift incision can decrease the incidence of Frey syndrome. [ABSTRACT FROM AUTHOR]

Abstract

Rearrangements within the AUTS2region are associated with a rare syndromic disorder with intellectual disability, developmental delay, and behavioral abnormalities as core features. In addition, smaller regional variants are linked to wide range of neuropsychiatric disorders, underscoring the gene's essential role in brain development. Like many essential neurodevelopmental genes, AUTS2is large and complex, generating distinct long (AUTS2-l) and short (AUTS2-s) protein isoforms from alternative promoters. Although evidence suggests unique isoform functions, the contributions of each isoform to specific AUTS2-linked phenotypes have not been clearly resolved. Furthermore, Auts2is widely expressed across the developing brain, but cell populations most central to disease presentation have not been determined. In this study, we focused on the specific roles of AUTS2-l in brain development, behavior, and postnatal brain gene expression, showing that brain-wide AUTS2-l ablation leads to specific subsets of the recessive pathologies associated with mutations in 3′ exons (exons 8–19) that disrupt both major isoforms. We identify downstream genes that could explain expressed phenotypes including hundreds of putative direct AUTS2-l target genes. Furthermore, in contrast to 3′ Auts2mutations which lead to dominant hypoactivity, AUTS2-l loss-of-function is associated with dominant hyperactivity and repetitive behaviors, phenotypes exhibited by many human patients. Finally, we show that AUTS2-l ablation in Calbindin 1-expressing cell lineages is sufficient to yield learning/memory deficits and hyperactivity with abnormal dentate gyrus granule cell maturation, but not other phenotypic effects. These data provide new clues to in vivo AUTS2-l functions and novel information relevant to genotype–phenotype correlations in the human AUTS2region.
