In order to achieve rapid and accurate prediction of the hydrodynamic performance of propellers, a set of propeller hydrodynamic performance prediction model was established based on the improved residual connection network. The residual connection method greatly improves the depth of the model. In combination with the Inception structure to simultaneously extract data features from different scales, the depthwise separable convolution reduces the model parameters. The sample space for training the deep neural network is built based on the propeller geometric parameters and model test results. An improved beetle swarm antennae search algorithm is proposed to optimize the initial weights and thresholds of the model to further improve the prediction accuracy of the model. The research results indicate that the improved beetles swarm antennae algorithm significantly improves the accuracy of the model and solves the problem of overfitting of it. The prediction results of the model are in good agreement with the experimental values, and its prediction performance for the propellers which are not in the dataset is basically the same as that of the CFD method. The model has an excellent universality and its calculation period is extremely short, which can meet the requirements of real-time and accurate prediction of propeller open water performance.

V-domain Ig suppressor of T cell activation (VISTA), encoded by the human VSIR gene, is a B7 family checkpoint homologous to the programmed death-Ligand 1 sequence. In gynecologic malignancies, VISTA is abnormally expressed and regulates the tumor immune microenvironment, causing a high upregulation of VISTA expression in T-cells and myeloid cells in the tumor microenvironment and promoting tumor proliferation, progression, and immune tolerance. Here, we review the research progress of VISTA in ovarian, cervical, and endometrial cancers through its structure and immunomodulatory mechanism. The comprehensive study of VISTA is expected to improve the current problem of poor immunotherapeutic effects and provide new ideas for immune therapy in patients with gynecologic tumors.

Swine leukocyte antigen (SLA) class I molecule-restricted T-cell epitopes, which induce cytotoxic T lymphocyte (CTL) responses, play a critical role in the clearance of porcine reproductive and respiratory syndrome virus (PRRSV) and the development of efficient protective vaccines. The SLA-1*04:01:01, SLA-2*04:01, and SLA-3*04:01 alleles, assigned the Hp-4.0 haplotype, are highly prevalent and usually present in all pig breeds. However, the SLA Hp-4.0 haplotype-restricted CTL epitopes in the structural membrane (M) protein of PRRSV are still unknown. In this study, we predicted 27 possible 9-mer epitope peptides in M protein with high binding scores for SLA-1*04:01:01 using CTL epitope prediction tools. In total, 45 SLA class I complexes, comprising the predicted peptide, extracellular region of the SLA-I molecules, and β2-microglobulin, were constructed in vitro to detect the specific binding of these peptides to SLA-1*04:01:01 (27 complexes), SLA-2*04:01 (9 complexes), and SLA-3*04:01 (9 complexes), respectively. Our results showed that the M27 (T91WKFITSRC), M39 (N130HAFVVRRP), and M49 (G158RKAVKQGV) peptides bind specifically to SLA-1*04:01:01, SLA-2*04:01, and SLA-3*04:01, respectively. Subsequently, using peripheral blood mononuclear cells (PBMCs) isolated from the homozygous Hp-4.0 and Hp-26.0 haplotype piglets vaccinated with commercial PRRSV HuN4-F112 strain, we determined the capacities of these 27 potential peptides to stimulate their proliferation with a Cell Counting Kit-8 and their secretion and expression of interferon gamma (IFN-γ) with an ELISpot assay and real-time qPCR, respectively. The immunological activities of M27, M39, and M49 were therefore confirmed when they efficiently induced PBMC proliferation and IFN-γ secretion in PBMCs from piglets with the prevalent SLA Hp-4.0 haplotype. The amino acid sequence alignment revealed that M27, M39, and M49 are highly conserved among 248 genotype II PRRSV strains collected between 1998 and 2019. These findings contribute to the understanding of the mechanisms of cell-mediated immune responses to PRRSV. Our study also provides a novel strategy for identifying and confirming potential SLA haplotype-restricted CTL epitopes that could be used to develop novel peptide-based vaccines against swine diseases.

Abstract Paroxysmal sympathetic hyperactivity (PSH) mainly occurs after acquired brain injury (ABI) and often presents with high fever, hypertension, tachycardia, tachypnea, sweating, and dystonia (increased muscle tone or spasticity). The pathophysiological mechanisms of PSH are not fully understood. Currently, there are several views: (1) disconnection theory, (2) excitatory/inhibitory ratio, (3) neuroendocrine function, and (4) neutrophil extracellular traps. Early diagnosis of PSH remains difficult, given the low specificity of its diagnostic tools and unclear pathogenesis. According to updated case analyses in recent years, PSH is now more commonly observed in patients with stroke, with tachycardia and hypertension as the main clinical manifestations, which is not fully consistent with previous data. To date, the PSH Assessment Measure tool is optimal for the early identification of PSH and stratification of symptom severity. Clinical strategies for the management of PSH are divided into three main points: (1) reduction of stimulation, (2) reduction of sympathetic excitatory afferents, and (3) inhibition of the effects of sympathetic hyperactivity on target organs. However, use of drugs and standards have not yet been harmonized. Further investigation on the relationship between PSH severity and long-term neurological prognosis in patients with ABI is required. This review aimed to determine the diagnostic and management challenges encountered in PSH after ABI.

A thermal-mechanical control process (TMCP) followed by tempering at 600 °C was applied to produce 420 MPa grade high strength low alloy (HSLA) steel heavy plates. The microstructure and mechanical properties through the thickness were analyzed using various techniques including optical microscopy (OM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), electron back-scattered diffraction (EBSD), tensile and impact tests. Microscopy analyses showed that a fine-grained structure with an average grain size of 6–10 μm could be obtained by TMCP. After subsequent tempering, the microstructure near the surface of the plate was dominated by bainite, while the microstructure of the core was mainly characterized by aggregated carbides and ferrite. In addition, the density of high-angle grain boundaries (HAGB) on the surface was significantly higher than that in the core. Tensile tests showed a decrease in both yield and tensile strengths from 530.1 and 626.9 MPa on the surface to 384.3 and 496.7 MPa in the core, respectively. The contribution of different strengthening mechanisms to the variations in strengths along the thickness was clarified. In addition, the deformation of ferrite and the initiation of micro-voids during tensile tests were also analyzed.
