Health, distribution and life-history of aquatic crustaceans strongly depend on water hardness. However hardness is commonly correlated with alkalinity, which highlights the need to assess the joint effects of both hardness and alkalinity. This study aims to test the hypothesis that water hardness and alkalinity affect the life-history parameters (growth, reproduction and population growth rate) of D. longispina. Following this, life table experiments were carried out in order to study the effects of high levels versus low levels of water hardness and alkalinity. Low levels of hardness and alkalinity caused a significant reduction in the growth of daphnids after a 7-days period, which augmented during the 21-day-test period reaching a 14.5% reduction compared to high hardness and alkalinity. Allied to the reduced growth, daphnids reared at low hardness and alkalinity showed delayed reproduction, increased body length at first reproduction, reduced fertility at first brood and, consequently, a 36.6% reduction in total fertility, compared to daphnids reared at high hardness and alkalinity. Accordingly, daphnids with the same size produced smaller broods at low hardness and alkalinity, reflecting a direct effect of water chemistry on daphnids reproduction. The impaired growth and reproduction at low hardness and alkalinity levels was likely a consequence of increased maintenance costs, and was not related to changes in the feeding activity. Population growth rate of daphnids reared at low hardness and alkalinity was 13.4% lower than that of daphnids reared at high hardness and alkalinity. Thus, despite D. longispina can survive at low hardness and alkalinity, their life-history parameters are significantly affected. This study raises concerns about the effects of decreasing hardness and alkalinity, which has been reported in Europe and North America, on populations of D. longispina and, thus, on the structure of aquatic ecosystems.

There are a limited number of studies correlating the different stages of dendritic cells (DC) maturation with cytokines in individuals presented chronic periodontitis (CP). The aim of the study was to evaluate the correlation among the expression of IL-2, IL-10, IL-4, IL-6, IFN-γ, TNF-α, and IL-17A with the presence of DC and mild-moderate or advanced CP. Gingival samples were obtained from 24 individuals with CP and six samples of normal mucosa (NM) overlapping third molar for controls of the levels of cytokines. Periodontal examination was performed. Immunohistochemical staining was carried out, revealing CD1a+ immature, Fator XIIIa+ immature, and CD83+ mature DCs. The inflammatory infiltrate was counted, and the cytokines were measured by flow cytometry. Densities of DCs and inflammatory infiltrate, cytokines, subtypes of CP, and clinical periodontal parameters were correlated and compared. IL-6 expression was correlated positively with the increased numbers of CD1a+ immature DCs. Levels of IL-2, TNF-α, IFN-γ, IL-10, and IL-17A were increased when compared with NM. The percentage of sites with clinical attachment level (CAL)>3 were positively correlated with densities of inflammatory infiltrate and negatively correlated with densities of immature DCs. IL-6 can contribute to the increase of the immature DCs in the CP. Higher levels of IL-2, TNF-α, IFN-γ, IL-10, and IL-17A cytokines were observed in CP. Higher densities of inflammatory infiltrate as well as lower densities of immature DCs can result in a more severe degree of human CP.

Brain-derived neurotrophic factor (BDNF) is a member of the neurotrophic factor family. Outside the nervous system, BDNF has been shown to be expressed in various nonneural tissues, such as periodontal ligament, dental pulp, and odontoblasts. Although a role for BDNF in periodontal regeneration has been suggested, a function for BDNF in periodontal disease has not yet been studied. The aim of this study was to analyze the BDNF levels in periodontal tissues of patients with chronic periodontitis (CP) and periodontally healthy controls (HC). All subjects were genotyped for the rs4923463 and rs6265 BDNF polymorphisms. Periodontal tissues were collected for ELISA, myeloperoxidase (MPO), and microscopic analysis from 28 CP patients and 29 HC subjects. BDNF levels were increased in CP patients compared to HC subjects. A negative correlation was observed when analyzing concentration of BDNF and IL-10 in inflamed periodontium. No differences in frequencies of BDNF genotypes between CP and HC subjects were observed. However, BDNF genotype GG was associated with increased levels of BDNF, TNF-α, and CXCL10 in CP patients. In conclusion, BDNF seems to be associated with periodontal disease process, but the specific role of BDNF still needs to be clarified.

Objective. Interleukin-17 (IL-17) is a cytokine that induces neutrophil recruitment and the release of inflammatory mediators in several inflammatory conditions; nevertheless, the involvement of IL-17 gene polymorphisms in chronic periodontitis (CP) has not been addressed yet. Our aim was to evaluate the association between periodontal status and the polymorphisms IL-17A G197A and IL-17F C7488T in subjects with CP along with their impact on levels of inflammatory mediators. Material and Methods. Genomic DNA was obtained from 30 CP patients and 30 healthy controls (HCs). IL-17A G197A and IL-17F C7488T polymorphisms were determined using PCR-RFLP. Serum and periodontal tissues were collected and processed for ELISA, myeloperoxidase (MPO), and/or microscopic analysis. Results. The frequencies of genotypes in the CP group were significantly different from those of HC. Odds ratio indicated that increased risks for CP were associated with the -197A allele, not with the -7488T allele. In addition, the -197A allele was correlated with worse clinical parameters, higher MPO activity, and increased expression of inflammatory mediators (IL-17A and IL-8) than the other genotypes. Conclusions. These results indicate that the IL-17A -197A allele is associated with increased risk for CP, likely because this genotype relates to the enhanced inflammation in periodontal tissues.

O presente trabalho, desenvolvido no âmbito de um Núcleo de Pesquisa em Ensino de Ciências teve como objetivo central analisar o movimento de idéias do grupo formado por professores formadores, professores do Ensino Básico e alunos de Graduação e Mestrado. Foram analisados quatro encontros, nos quais se discutiram as expectativas dos participantes, conceitos científicos e projetos de ensino elaborados a partir de temas relevantes. Os dados foram analisados com categorias criadas a partir da leitura das transcrições registradas em VHS. A análise evidenciou o interesse/necessidade dos professores do Ensino Básico de aproximação com a Universidade, as dificuldades de compreensão de conceitos químicos e a necessidade de ajuda de parceiros mais experientes na elaboração de propostas de trabalho para as escolas. Apesar desses problemas, os professores elaboraram e executaram projetos o que levou a conclusão de que a formação continuada é um processo indispensável e que deve ser promovido constantemente.

Periapical lesions are inflammatory conditions of tooth periapical tissues, triggered by dental pulp infection and characterized by exudation of immune cells to the affected tissues and production of inflammatory mediators such as cytokines. The inflammatory periapical reaction is mainly driven by Th1, Th2, and Th17 responses, and such polarization may modulate progression of the disease and expression of bone proresorptive cytokines. IL-12 is a potent inducer of IFN-γ production, which stimulates Th1 effector cells. Many evidences have shown a positive correlation between the bone resorptive cytokine IL-1β and the production of IL-12 and IFN-γ. Furthermore, IL-12 may have a potential role in the release of bone resorptive mediators and blockade of Th2 cytokines, affecting the progression of periapical bone loss. Nevertheless, IL-12 and IFN-γ have also been described as suppressors of osteoclast differentiation and activation, favoring bone maintenance. This paper focuses on the controversial roles of IL-12 in periapical lesions.

The study aimed to evaluate the suitability of myeloperoxidase (MPO) content as a local indicator of chronic inflammation, using the periodontal disease model. Anesthetized adult male Holtzman rats had their second left maxilar molar tied by a thread for 11 days and were then killed. Blood samples and photographic images from histopathological inflamed and noninflamed (contralateral) neighboring gingivomucosal specimens were collected for cell counts and MPO level analysis. Diseased animals were also treated with pharmacological tools such as the anti-inflammatory drug celecoxib or the opioid morphine. Increased blood neutrophils and local cell numbers characterized diseased animals. However, local MPO content was increased in inflamed and noninflamed tissues from diseased animals. Celecoxib and morphine reduced blood neutrophils and bilateral MPO content, but only celecoxib reduced local cell numbers in diseased animals. It is concluded that MPO content is a good indicator of a systemic rather than a local inflammation in a chronic inflammatory condition.

Relato de caso de interação de miocardite crônica e Síndrome Geral de Adaptação (estresse) causando ruptura da parede ventricular esquerda em uma fêmea jovem da espécie Cygnus atratus (cisne-negro), que veio a óbito no Zoológico Municipal de Taboão da Serra, em São Paulo, durante período de quarentena, após longa viagem em condições desfavoráveis de temperatura e ventilação.

Relatam-se cinco casos confirmados de Cinomose de um total de 13 óbitos de animais silvestres brasileiros das espécies Galictis vittata, Crisocyon brachyurus e Cerdocyon thous, mantidos em cativeiro no Zoológico Municipal de Taboão da Serra - SP - Brasil. À histopatologia observam-se as principais lesões da virose, inclusive a presença de corpúsculos de inclusão diferentes localizações. A sobrevivência de um único indivíduo, vacinado contra Cinomose, reforça a importância desta medida profilática no controle da doença.

Objective: We aimed to understand the potential therapeutic and anti-inflammatory effects of the phosphodiesterase-4 (PDE4) inhibitor roflumilast in models of pulmonary infection caused by betacoronaviruses. Methods: Mice were infected intranasally with murine hepatitis virus (MHV-3) or severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Roflumilast was given to MHV-3-infected mice therapeutically at doses of 1 mg/kg or 10 mg/kg, or prophylactically at 10 mg/kg. In SARS-CoV-2-infected mice, roflumilast was given therapeutically at a dose of 10 mg/kg. Lung histopathology, chemokines (CXCL-1 and CCL2), cytokines (IL-1β, IL-6, TNF, IFN-γ, IL-10 and TGFβ), neutrophil immunohistochemical staining (Ly6G+ cells), macrophage immunofluorescence staining (F4/80+ cells), viral titration plaque assay, real-time PCR virus detection, and blood cell counts were examined. Results: Therapeutic treatment with roflumilast at 10 mg/kg reduced lung injury in SARS-CoV-2 or MHV-3-infected mice without compromising viral clearance. In MHV-3-infected mice, reduced lung injury was associated with decreased chemokines levels, prevention of neutrophil aggregates and reduced macrophage accumulation in the lung tissue. However, the prophylactic treatment strategy with roflumilast increased lung injury in MHV-3-infected mice. Conclusion: Our findings indicate that therapeutic treatment with roflumilast reduced lung injury in MHV-3 and SARS-CoV-2 lung infections. Given the protection induced by roflumilast in inflammation, PDE4 targeting could be a promising therapeutic avenue worth exploring following severe viral infections of the lung. Hair straighteners are among the most technically complex products to be safely and effectively developed, and this challenge has increased even more with the higher incidence of resistant hair among consumers. This underscores the importance of studying new active ingredients, combinations and carrier formulations to improve performance without compromising safety. In this research, we compared eight hair-straightening formulations with different active ingredients and/or concentrations to develop new, safer and more effective texture modifiers. Eight formulations were developed and compared with each other and to controls (virgin and bleached hair) regarding mechanical and thermal resistance, cuticle morphology, hair shine and fiber diameter. Results showed that all formulations were safe and effective at straightening hair. Specifically, 13.3% and 9.4% ammonium thioglycolate (G03 and G04) were more suitable for wavy and curly hair, 12.5% and 7.9% amino methyl propanol thioglycolate (G05 and G06) for finer or chemically processed hair, 5% and 4% sodium cysteamine (G07 and G08) for curly and tight curly hair to control volume, and 2% and 1% of a combination of ammonium thioglycolate with sodium thioglycolate (G09 and G10) for more resistant wavy and curly hair. Abstract: The Cerrado is one of the world's biodiversity hotspots and Brazil’s second largest biome. Many native species of the Brazilian Cerrado provide fruits that have unique sensory characteristics and high nutritional value. This study aimed at characterizing the pulp of Eugenia klotzschiana O. Berg, concerning its proximal composition, bioactive compound content and antioxidant activity. The pulp under study had high moisture (89.47 g kg-1) and caloric (96.07 kcal kg-1) values whereas its contents of protein (0.59 g kg-1) and lipids (2.35 g kg-1) were low. The cerrado pear pulp also had high iron content (16.5 mg kg -1) and dietary fiber (6.45 g kg-1), besides 0.034-0,055 mg kg-1 carotenoids, 8.66 mg kg-1 ascorbic acid and 0.66 mg kg-1 total chlorophyll. Total phenolic compounds (333.41-566.33 mg EAG kg-1) and flavonoids (225-50 mg EQ kg-1) were found by extraction methods named Method 1 (water) and Method 2 (acetone+methanol), respectively. Thus, the cerrado pear can be an alternative to improving nutrient intake and to providing sustainable use of the native flora in the Cerrado.

Nanocomposite coatings functionalized with antimicrobial nanoparticles could be a promising alternative for the postharvest preservation of fruits. This study aimed to develop nanocomposite coatings based on pectin incorporated with zinc oxide (NPZ) nanoparticles to preserve the postharvest quality of papaya fruits. The coatings were prepared using pectin (3%) and NPZ (0%–0.4%). The materials were characterized for water-related properties (water solubility and water vapor permeability) as well as physical, mechanical, morphological, rheological, and structural properties. The coatings were applied to papaya fruits, which were analyzed for weight loss, firmness, titratable acidity, and soluble solids over nine days of storage. Incorporating NPZ (0%–0.4%) did not affect the films' water solubility and vapor permeability. However, films with NPZ exhibited lower mechanical properties than pure pectin films. Rheological behavior testing indicated that the pectin solution was a Newtonian fluid, whereas pectin solutions with zinc nanoparticles were non-Newtonian fluids. The pectin coating with 0.2% NPZ was the most effective in preserving the postharvest quality of papaya by reducing fruit weight loss and acidity content. A safe and effective vaccine with long-term protection against SARS-CoV-2 variants of concern (VOCs) is a global health priority. Here, we develop lipid nanoparticles (LNPs) to provide safe and effective delivery of plasmid DNA (pDNA) and show protection against VOCs in female small animal models. Using a library of LNPs encapsulating unique barcoded DNA (b-DNA), we screen for b-DNA delivery after intramuscular administration. The top-performing LNPs are further tested for their capacity of pDNA uptake in antigen-presenting cells in vitro. The lead LNP is used to encapsulate pDNA encoding the HexaPro version of SARS-CoV-2 spike (LNP-HPS) and immunogenicity and protection is tested in vivo. LNP-HPS elicit a robust protective effect against SARS-CoV-2 Gamma (P.1), correlating with reduced lethality, decreased viral load in the lungs and reduced lung damage. LNP-HPS induce potent humoral and T cell responses against P.1, and generate high levels of neutralizing antibodies against P.1 and Omicron (B.1.1.529). Our findings indicate that the protective efficacy and immunogenicity elicited by LNP-HPS are comparable to those achieved by the approved COVID-19 vaccine from Biontech/Pfizer in animal models. Together, these findings suggest that LNP-HPS hold great promise as a vaccine candidate against VOCs.Here the authors screen different lipid nanoparticle (LNP) formulations for intramuscular delivery of plasmid DNA and uptake by antigen-presenting cells. to evaluate the effects of the red and near-infrared wavelength lasers in isolated and simultaneous way on the modulation of inflammatory cytokines produced by human keratinocytes (HaCaT) challenged by cytokines of human monocytes stimulated by lipopolysaccharide from Escherichia coli. HaCaT cells was previously exposed to the laser with wavelengths red (660 nm), near-infrared (808 nm). Then, HaCat cells were stimulated with the supernatant of lipopolysaccharide-challenged peripheral blood cells. The cytokines expressed by HaCat cells were measured using multiplex CBA assay. HaCaT cells increased the production of inflammatory cytokines when stimulated with infrared laser compared to the control group (IFN-α2, IFN-γ, TNF-α, MCP-1, IL-6, IL-8, IL-10, IL -12p70, IL -17A, IL-23, IL-33), the red laser group (IFN-γ and IL-23) and the group of two lasers used simultaneously (IFN-α2, IFN-γ, IL-6 and IL-8, IL-17A, IL-18 and IL-23) (p < 0.05). The red laser also stimulated an increase in the expression of IFN-α2 by HaCaT cells in relation to the control group (p < 0.05).
