To treat the impact of trauma, most current evidence supports the use of trauma-focused psychotherapy as the first line approach. However, millions of individuals exposed to trauma worldwide seek Complementary and Integrative Health (CIH) therapies in hopes of achieving wellness above and beyond reducing symptoms. But what is the evidence for CIH interventions? What are potential pitfalls? Given the growing popularity of and strong interest in CIH, EJPT is featuring research on these approaches in this special issue. The papers range from common interventions such as mindfulness to the use of service dogs and scuba diving to alleviate trauma related symptoms. A featured editorial highlights the importance of defining when, where, and how placebo responses work. Nonspecific elements of treatment such as positive expectations, therapeutic rituals, healing symbols, and social interactions are identified as factors influencing treatment response and scientists looking to add to the CIH evidence base are encouraged to consider the impact and methodological challenges these elements present. CIH interventions more specifically recognize and harness some of these factors in addition to intervention-specific factors such as attention or emotion regulation along with focus on overall wellbeing. The body of work in this special issue supports the emerging evidence for meditative and relaxation-based interventions and illustrates a creative but nascent state of the field. Cross-intervention mechanisms that may play a role in achieving wellness, such as arousal reduction, emotion regulation, posttraumatic growth, and positive affect are highlighted. The trauma field would benefit from accumulation of evidence for promising CIH interventions, evaluation of potential mechanisms, and examination of health and wellbeing outcomes. With the paucity of high-quality trials, it would be premature to recommend CIH interventions as first-line treatments. However, the emerging literature on CIH continues to advance our understanding of what works and how these interventions exert their effects. Complementary and Integrative Health (CIH) interventions for trauma that target holistic wellness above and beyond symptom reduction are increasingly used in the real world, though the evidence base lags. Papers in this issue support the emerging evidence for efficacy of mindfulness or other meditative or relaxation-based interventions. Evidence in humans suggests a correlation between nicotine smoking and severe respiratory symptoms with COVID-19 infection. In lung tissue, angiotensin-converting enzyme 2 (ACE2) appears to mechanistically underlie viral entry. Here, we investigated whether e-cigarette vapor inhalation alters ACE2 and nicotinic acetylcholine receptor (nAChR) expression in male and female mice. In male lung, nicotine vapor inhalation induced a significant increase in ACE2 mRNA and protein, but surprisingly, these differences were not found in females. Further, both vehicle and nicotine vapor inhalation downregulated α5 nAChR subunits in both sexes, while differences were not found in α7 nAChR subunit expression. Finally, blood ACE2 levels did not differ with exposure, indicating that blood sampling is not a sufficient indicator of lung ACE2 changes. Together, these data indicate a direct link between e-cigarette vaping and increased ACE2 expression in male lung tissue, which thereby reveals an underlying mechanism of increased vulnerability to coronavirus infection in individuals vaping nicotine.

Technical challenges remain in the sequencing of RNA viruses due to their high intra-host diversity. This bottleneck is particularly pronounced when interrogating long-range co-evolved genetic interactions given the read-length limitations of next-generation sequencing platforms. This has hampered the direct observation of these genetic interactions that code for protein-protein interfaces with relevance in both drug and vaccine development. Here we overcome these technical limitations by developing a nanopore-based long-range viral sequencing pipeline that yields accurate single molecule sequences of circulating virions from clinical samples. We demonstrate its utility in observing the evolution of individual HIV Gag-Pol genomes in response to antiviral pressure. Background: Problems arising during menstruation include pain before or during menstruation. Progressive muscle relaxation (PMR) techniques and endorphin massage (EM) therapy are effective in controlling menstrual pain. This study aims to determine the difference in the effectiveness of pain intensity carried out by PMR therapy and EM therapy. Methods: This study was a quasi-experimental research with a two-group pretest and posttest design. Respondents were selected using a purposive sampling technique with inclusion and exclusion criteria. The inclusion criteria included students who experienced dysmenorrhea and do not have a genetic abnormality. The respondents who met the criteria were 80 respondents. The respondents were randomly selected using computerized to divide them into two groups. Forty respondents received PMR intervention and another 40 respondents received EM intervention. Bivariate analysis was performed using Wilcoxon test. Identification of respondents who fit the criteria, i.e. adolescents aged 13-14 years who experience menstrual pain, and do not have genetic abnormalities. Pain measurement was performed with a Numeric Rating Scale (NRS) and analyzed using the Mann-Whitney test. Results: PMR is more effective in lower intensity painful menstruation compared to EM proven that the average painful menstruation after PMR was 2.42±0.675, while the average painful menstruation after EM was 2.90±0955 with a difference of 4.8 points with a significance level of 0.000. Menstrual pain can be reduced by blocking nerve impulses from reaching the brain, including PMR therapy to control tension and anxiety. Conclusion: PMR therapy is more effective for controlling menstrual pain. PMR therapy can be done alone by adolescents when menstrual pain appears.

RNA molecules can fold into complex two- and three-dimensional shapes that are critical for their function. Chemical probes have long been utilized to interrogate RNA structure and are now considered invaluable resources in the goal of relating structure to function. Recently, the power of deep sequencing and careful chemical probe design have merged, permitting researchers to obtain a holistic understanding of how RNA structure can be utilized to control RNA biology transcriptome-wide. Within this review, we outline the recent advancements in chemical probe design for interrogating RNA structures inside cells and discuss the recent advances in our understanding of RNA biology through the lens of chemical probing.

X-linked agammaglobulinemia (XLA) is a monogenic primary immune deficiency characterized by very low levels of immunoglobulins and greatly increased risks for recurrent and severe infections. Patients with XLA have a loss-of-function mutation in the Bruton's tyrosine kinase (BTK) gene and fail to produce mature B lymphocytes. Gene editing in the hematopoietic stem cells of XLA patients to correct or replace the defective gene should restore B cell development and the humoral immune response. We used the clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 platform to precisely target integration of a corrective, codon-optimized BTK complementary DNA (cDNA) cassette into its endogenous locus. This process is driven by homologous recombination and should place the transgenic BTK under transcriptional control of its endogenous regulatory elements. Each integrated copy of this cDNA in BTK-deficient K562 cells produced only 11% as much BTK protein as the wild-type gene. The donor cDNA was modified to include the terminal intron of the BTK gene. Successful integration of the intron-containing BTK donor led to a nearly twofold increase in BTK expression per cell over the base donor. However, this donor variant was too large to package into an adeno-associated viral vector for delivery into primary cells. Donors containing truncated variants of the terminal intron also produced elevated expression, although to a lesser degree than the full intron. Addition of the Woodchuck hepatitis virus posttranscriptional regulatory element led to a large boost in BTK transgene expression. Combining these modifications led to a BTK donor template that generated nearly physiological levels of BTK expression in cell lines. These reagents were then optimized to maximize integration rates into human hematopoietic stem and progenitor cells, which have reached potentially therapeutic levels in vitro. The novel donor modifications support effective gene therapy for XLA and will likely assist in the development of other gene editing-based therapies for genetic disorders.

Although high-throughput RNA sequencing (RNA-seq) has greatly advanced small non-coding RNA (sncRNA) discovery, the currently widely used complementary DNA library construction protocol generates biased sequencing results. This is partially due to RNA modifications that interfere with adapter ligation and reverse transcription processes, which prevent the detection of sncRNAs bearing these modifications. Here, we present PANDORA-seq (panoramic RNA display by overcoming RNA modification aborted sequencing), employing a combinatorial enzymatic treatment to remove key RNA modifications that block adapter ligation and reverse transcription. PANDORA-seq identified abundant modified sncRNAs-mostly transfer RNA-derived small RNAs (tsRNAs) and ribosomal RNA-derived small RNAs (rsRNAs)-that were previously undetected, exhibiting tissue-specific expression across mouse brain, liver, spleen and sperm, as well as cell-specific expression across embryonic stem cells (ESCs) and HeLa cells. Using PANDORA-seq, we revealed unprecedented landscapes of microRNA, tsRNA and rsRNA dynamics during the generation of induced pluripotent stem cells. Importantly, tsRNAs and rsRNAs that are downregulated during somatic cell reprogramming impact cellular translation in ESCs, suggesting a role in lineage differentiation.

Diversity-generating retroelements (DGRs) vary protein sequences to the greatest extent known in the natural world. These elements are encoded by constituents of the human microbiome and the microbial 'dark matter'. Variation occurs through adenine-mutagenesis, in which genetic information in RNA is reverse transcribed faithfully to cDNA for all template bases but adenine. We investigated the determinants of adenine-mutagenesis in the prototypical Bordetella bacteriophage DGR through an in vitro system composed of the reverse transcriptase bRT, Avd protein, and a specific RNA. We found that the catalytic efficiency for correct incorporation during reverse transcription by the bRT-Avd complex was strikingly low for all template bases, with the lowest occurring for adenine. Misincorporation across a template adenine was only somewhat lower in efficiency than correct incorporation. We found that the C6, but not the N1 or C2, purine substituent was a key determinant of adenine-mutagenesis. bRT-Avd was insensitive to the C6 amine of adenine but recognized the C6 carbonyl of guanine. We also identified two bRT amino acids predicted to nonspecifically contact incoming dNTPs, R74 and I181, as promoters of adenine-mutagenesis. Our results suggest that the overall low catalytic efficiency of bRT-Avd is intimately tied to its ability to carry out adenine-mutagenesis.

The in vivo metabolism and pharmacokinetics of flunixin meglumine and phenylbutazone have been extensively characterized; however, there are no published reports describing the in vitro metabolism, specifically the enzymes responsible for the biotransformation of these compounds in horses. Due to their widespread use and, therefore, increased potential for drug-drug interactions and widespread differences in drug disposition, this study aims to build on the limited current knowledge regarding P450-mediated metabolism in horses. Drugs were incubated with equine liver microsomes and a panel of recombinant equine P450s. Incubation of phenylbutazone in microsomes generated oxyphenbutazone and gamma-hydroxy phenylbutazone. Microsomal incubations with flunixin meglumine generated 5-OH flunixin, with a kinetic profile suggestive of substrate inhibition. In recombinant P450 assays, equine CYP3A97 was the only enzyme capable of generating oxyphenbutazone while several members of the equine CYP3A family and CYP1A1 were capable of catalyzing the biotransformation of flunixin to 5-OH flunixin. Flunixin meglumine metabolism by CYP1A1 and CYP3A93 showed a profile characteristic of biphasic kinetics, suggesting two substrate binding sites. The current study identifies specific enzymes responsible for the metabolism of two NSAIDs in horses and provides the basis for future study of drug-drug interactions and identification of reasons for varying pharmacokinetics between horses.

Powdery mildews are notorious fungal plant pathogens but only limited information exists on their genomes. Here we present the mitochondrial genome of the grape powdery mildew fungus Erysiphe necator and a high-quality mitochondrial gene annotation generated through cloning and Sanger sequencing of full-length cDNA clones. The E. necator mitochondrial genome consists of a circular DNA sequence of 188,577 bp that harbors a core set of 14 protein-coding genes that are typically present in fungal mitochondrial genomes, along with genes encoding the small and large ribosomal subunits, a ribosomal protein S3, and 25 mitochondrial-encoded transfer RNAs (mt-tRNAs). Interestingly, it also exhibits a distinct gene organization with atypical bicistronic-like expression of the nad4L/nad5 and atp6/nad3 gene pairs, and contains a large number of 70 introns, making it one of the richest in introns mitochondrial genomes among fungi. Sixty-four intronic ORFs were also found, most of which encoded homing endonucleases of the LAGLIDADG or GIY-YIG families. Further comparative analysis of five E. necator isolates revealed 203 polymorphic sites, but only five were located within exons of the core mitochondrial genes. These results provide insights into the organization of mitochondrial genomes of powdery mildews and represent valuable resources for population genetic and evolutionary studies.

Huangzangsi Reservoir is the first comprehensive utilization reservoir in the upper reaches of the Heihe River. Supported by the new energy policy of Qinghai Province, if the Huangzangsi Reservoir is equipped with photovoltaic power stations, a complex complementary system of hydropower and photovoltaic power generation in the upper reaches of the Heihe River will be formed. This study used PVsyst software to complete the design of a 100 MW photovoltaic plant, and proposed a daily output clustering method based on the number of output curve fluctuations, maximum output change and output proportion as indicators. A daily optimal operation model for Huangzangsi Reservoir was established with the goal of stabilizing the output of the Hydro-PV complementary system and improving the power generation revenue of cascade hydropower stations and solved by using the NSGA-Ⅱ. The results indicate that cloudy weather, peak shaving power generation by cascade hydropower stations, installed capacity of Huangzangsi Reservoir and water volume limitations that can be regulated are the main factors affecting the stability of the output of the Hydro-PV complementary system; When the Huangzangsi Reservoir compensates for photovoltaic output, the outgoing flow has a small fluctuation, this fluctuation is absorbed by the Baopinghe Hydropower Station; When the Huangzangsi Reservoir compensates for the cascade hydropower station peaking and power generation, the upstream produces a large flow of incoming water, Longshou-Ⅱ Hydropower Station ensures that the Yingluoxia section has a stable flow throughout the day; The Hydro-PV complementary mode has a small impact on the hydropower benefits of Huangzangsi Reservoir and has a significant effect on stabilizing the fluctuation of photovoltaic output, however, the capacity configuration of photovoltaic power stations should be further optimized. Background: The period from 6 to 24 months in an infant's life presents a critical window for understanding feeding practices and for designing culturally appropriate interventions. However, little is known about the complementary feeding practices of Black mothers and how this period can be used to optimise the long‐term health of their children. The present study aimed to identify factors that influence the complementary feeding practices of low‐income Black mothers with children aged 6–24 months. Methods: Participants were recruited through Research Match, Facebook advertising, flyers, and snowballing techniques. Low‐income, Black mothers, with a 6–24‐month‐old infant, and who lived in Franklin County, Ohio, USA, were eligible for the study. A cross‐sectional design using in‐depth interviews was used. Reflexive thematic analysis was utilised to analyse and interpret the feeding practices of Black mothers. Results: Mothers (n = 8) were aged between 18 and 30 years old and most completed college or had some college education (n = 6). Half (n = 4) were married, employed, and rated their diet quality and their children's as very good. Three themes emerged: (a) complementary feeding at ≥ 6 months of age; (b) involvement of health care providers and service organisations in feeding decisions; and (c) use of responsive feeding cues. Conclusions: All mothers breastfed exclusively and most (n = 6) initiated complementary feeding at 6 months. Paediatricians, other health providers and service organisations were instrumental in helping Black mothers adopt complementary feeding practices. Mothers also engaged in responsive feeding practices. These findings point to the critical nature of access and education in helping Black mothers in the study achieve feeding recommendations for their infants. This scoping review aimed to examine the existing evidence on complementary and alternative medicine interventions for preeclampsia, emphasizing empowering midwives in postpartum treatment. Three databases (PubMed, Web of Science, Scopus) were used following the PRISMA-2020 guidelines to find articles from January 2012 to December 2022 and synthesized using Rayyan AI software. A total of 707 articles were initially identified, 7 were included in the review. Traditional Chinese Medicine combined with Western medicine showed promise in treating severe preeclampsia. Massage therapy reduced anxiety, aromatherapy positively affected postpartum well-being, and warm foot therapy reduced swelling associated with postpartum preeclampsia. Using herbal ingredients, lavender aromatherapy, magnesium sulfate, massage, and physical therapy contributed to successful postpartum preeclampsia management. Complementary and alternative medicine, collaboration with community health workers, clear guidelines, targeted training, and future research are crucial for effective postpartum preeclampsia management. This study aims to explain the indicators of human resource productivity management in Kerman executive agencies. It is a descriptive, correlational as well as a developmental study purposefully conducted as a survey in the field of human resources. The sample includes 30 experts in the field of management and administration. It was obtained by purposeful sampling method and included 524 employees of the Kerman executive agencies that were gathered by stratified sampling method. Two questionnaires were used by the experts to confirm the validity and reliability of the model and one questionnaire was used to answer the questions. The validity and reliability of all questionnaires were confirmed. AMOS and SPSS statistical software were used for data analysis. The 42 subscales of human resource productivity management are summarized in 5 factors (individual, organizational, complementary organizational, occupational, and extra-organizational). It is an applieddevelopmental study considering the simultaneous identification of factors related to human resource productivity in line with the development indicators and can be used by all executive agencies throughout Iran. This paper focuses on the Linguistic Landscape (LL) in three geographical representative areas (Achrafieh, Bourj Hammoud, and Zahle) of Lebanon. The study investigates the state of multilingualism across the public and private signs in the selected geographical areas. The investigation is approached through a mixed design to unfold the bottom- up and top- down organization (Ben-Rafael et al., 2008) of the collected signs' images. The picturized items are arranged in accordance with Reh's (2004) framework into duplicating, overlapping, fragmentary, and complementary multilingual writings. The systematized pictures are accompanied with interviews, field notes, and participant observations to align with the Scollon and Scollon's (2003) qualitative study of discourses in place. A data quantification is followed to clarify the results at a computed state. The research concludes with disclosing the language choice in the selected areas, the ideology in each chosen area, and the reasons behind choosing the present versatile texts. Hypertension is one of the diseases with the highest risk in the world for cardiovascular disease, making it a primary factor in Indonesia as it causes damage to the heart, blood vessels, kidneys, lungs, and sensory-motor nerve cells, which may lead to death. This literature review aims to identify cupping therapy for clients' hypertension to decrease blood pressure according to results from studies from the last 5 years. Cupping therapy is one of the complementary therapies that is effective for prevention and treatment of some health problems in the body. The research method uses literature review designs as much as fifth journal from secondary databases, Google Scholar, and Science Direct on journals published between 2017 and 2022, according to keyword articles for a review. The article review uses the PICOS method in accordance with keyword of journal selection from the abstract, which include inclusion and exclusion criteria for the article to be reviewed. Overall, the journal research on cupping therapy intervention has shown results for patients with high blood pressure, so getting cupping therapy periodically can be effective in decreasing blood pressure. The smartphone is now a widespread trend that causes phone accessory markets to rise. The high penetration rate of smartphones has led to a saturated market. The accessory market not only has tremendous business opportunities but also high potential. In practice, the decision of whether to join the accessory market differentiates by each smartphone firm. This study analyzed and discussed the main product firms' optimal decisions of whether to enter the accessory market by using game theory as a construct model. Considering the different competition situations, the decision of whether the main product firm invests in the accessory market affected the market demand, pricing, and profit of all firms. Furthermore, we analyzed the differences in the external environment to determine what the best decision for the main product firm was. The results of the research show that complementary effects do not always have a positive influence on firms' willingness to join the accessory market. When weak main product firms have different market forces in the main product market, the dominant main product firms are not necessarily more willing to join the accessory market than weak firms. Depression represents one of the most common non-motor disorders in Parkinson's disease (PD) and it has been related to worse life quality, higher levels of disability, and cognitive impairment, thereby majorly affecting not only the patients but also their caregivers. Available pharmacological therapeutic options for depression in PD mainly include selective serotonin reuptake inhibitors, serotonin and norepinephrine reuptake inhibitors, and tricyclic antidepressants; meanwhile, agents acting on dopaminergic pathways used for motor symptoms, such as levodopa, dopaminergic agonists, and monoamine oxidase B (MAO-B) inhibitors, may also provide beneficial antidepressant effects. Recently, there is a growing interest in non-pharmacological interventions, including cognitive behavioral therapy; physical exercise, including dance and mind–body exercises, such as yoga, tai chi, and qigong; acupuncture; therapeutic massage; music therapy; active therapy; repetitive transcranial magnetic stimulation (rTMS); and electroconvulsive therapy (ECT) for refractory cases. However, the optimal treatment approach for PD depression is uncertain, its management may be challenging, and definite guidelines are also lacking. It is still unclear which of these interventions is the most appropriate and for which PD stage under which circumstances. Herein, we aim to provide an updated comprehensive review of both pharmacological and non-pharmacological treatments for depression in PD, focusing on recent clinical trials, systematic reviews, and meta-analyses. Finally, we discuss the pharmacological agents that are currently under investigation at a clinical level, as well as future approaches based on the pathophysiological mechanisms underlying the onset of depression in PD. We describe an RT-PCR protocol that allows high-resolution mapping of splicing products and isoforms using fluorescently labeled primers. Each species contains one fluorescent group allowing a direct comparison of the different isoforms despite size differences. A custom-size ladder enables the precise determination of cDNA lengths and discrimination of isoforms differing by less than five nucleotides on polyacrylamide gels. This protocol also allows the detection of products from in vitro splicing reactions, circumventing the need to use radiolabeled transcripts. PurposeThe use of complementary and integrative medicine (CIM) is on the rise among diverse populations of older adults in the USA. CIM is commonly perceived as safer, less expensive, and more culturally acceptable. There is a growing body of evidence to support the use of CIM, especially mind-body therapies, diet and nutritional supplements used for mental disorders of aging.Recent findingsWe summarize the results of the recent clinical trials and meta-analyses that provide the evidence for the role of CIM in treating older adults with mood or cognitive disorders that includes the use of diet and supplements, and mind-body therapies.SummaryDietary and mind-body therapies have become increasingly popular and show the strongest evidence of efficacy for mood and cognitive disorders. Although the use of vitamins and supplements is the most popular CIM practice, only mixed evidence supports their use with additional concerns for herb (supplement)-drug interactions. Despite increasing use of CIM by the general population, information to guide clinicians providing care for older adults remains limited with variable scientific rigor of the available RCTs for a large number of commonly used CIM interventions for the mental health of older adults.

Transcriptome profiling by RNA sequencing (RNA-seq) has been widely used to characterize cellular status, but it relies on second-strand complementary DNA (cDNA) synthesis to generate initial material for library preparation. Here we use bacterial transposase Tn5, which has been increasingly used in various high-throughput DNA analyses, to construct RNA-seq libraries without second-strand synthesis. We show that Tn5 transposome can randomly bind RNA/DNA heteroduplexes and add sequencing adapters onto RNA directly after reverse transcription. This method, Sequencing HEteRo RNA-DNA-hYbrid (SHERRY), is versatile and scalable. SHERRY accepts a wide range of starting materials, from bulk RNA to single cells. SHERRY offers a greatly simplified protocol and produces results with higher reproducibility and GC uniformity compared with prevailing RNA-seq methods.

High-throughput sequencing has become a standard tool for analyzing RNA and DNA. This method usually needs a cDNA/DNA library ligated with specific 5' and 3' linkers. Unlike mRNA, small RNA often contains modifications including 5' cap or triphosphate and 2'-O-methyl, requiring additional processing steps before linker additions during cloning processes; due to low expression levels, it is difficult to clone small RNA with a small amount of total RNA. Here we present a new strategy to clone 5' modified or unmodified small RNA in an all-liquid-based reaction carried out in a single PCR tube with as little as 20 ng total RNA. The 7-h cloning process only needs ∼1 h of labor. Moreover, this method can also clone mRNA, simplifying the need to prepare two cloning systems for small RNA and mRNA; the barcoded PCR primers are also compatible with non-cDNA cloning applications, including the preparation of genomic libraries. Not only is our method more convenient for cloning modified RNA than available methods, but it is also more sensitive, versatile, and cost-effective. Moreover, the all-liquid-based reaction can be performed in an automated manner.

This study examined the complementary dietary practices for infants among educated and non-educated women of Ife central local government area of Osun State, Nigeria. It was a descriptive survey research. A total of one hundred and fifty (154) respondents participated in this research. The research instrument used for data collection was a questionnaire on a 4-point scale. The major findings of the study revealed the complementary dietary practices for infants, identified possible factors affecting complementary dietary practice which are economic status, maternal education, employment status, residence type. It further revealed effects of inadequate complementary dietary practices which include sub-optimal growth and development and malnutrition, low intelligence level and others and revealed possible strategies to be adopted for adequate and proper complementary feeding practices which are provision of accurate dietary information to caregivers and family at large, introduction of developmental health system. Based on the findings, recommendations were made among which are adequate information should be given to upcoming mothers, hospitals, clinic and health center officials should educate mothers about the appropriate time to start feeding their children on complementary diet, mothers should breastfeed for at least six months of age before introducing semi-solid food to their children.

Objective: Fear of cancer recurrence among cancer survivors is a psychosocial concern that affects recovery and quality of life. They use complementary and alternative medicine to prevent the side effects of drugs and relieve anxiety and fear of cancer recurrence. This study aimed to examine the correlation between the use of complementary and alternative medicine and the fear of cancer recurrence in cancer survivors. Methods: This cross-sectional descriptive correlational study enrolled 280 cancer survivors referred to oncology centers and medical offices in Kerman using convenience sampling. The research tools included complementary and alternative medicine questionnaire and the fear of cancer recurrence inventory. IBM SPSS Statistics version 25 was used to analyze the data. Results: The study findings revealed that 78.2% of the participants used at least one type of complementary and alternative medicine in the last year; 71.8% used medicinal herbs, 19.6% used nutritional supplements, 7.5% used relaxation and meditation, 7.1% used dry cupping, and 5.7% used wet cupping. The mean score of fear of cancer recurrence was 80.72 ​± ​18.46, which was almost near the midpoint of the inventory score (84). The fear of cancer recurrence and its dimensions did not differ between users and nonusers of complementary and alternative medicine. Conclusions: Our results suggested that most of the survivors used at least one type of complementary and alternative medicine in the past year, and medicinal herbs and nutritional supplements were the most used types. Patients with cancer must be aware of the effects of different kinds of complementary and alternative medicine. A moderate level in the mean score of fear of cancer recurrence was found, and no difference was noted between users and nonusers of complementary and alternative medicine. Health managers and planners should conduct effective psychological interventions and strategies to minimize the fear of cancer recurrence among cancer survivors.

Introduction: Complementary therapy has become an option for various diseases over the time. One kind of disease that can be given complementary intervention is anxiety. Nowadays, many young people suffer from anxiety disorders, and they don't know how to deal with them properly. Make them less than optimal in achieving their life goals. The type of complementary therapy that can be used to reduce anxiety is meditation therapy. This survey aims to get an overview of the trends and knowledge of the younger generation about meditation in reducing excessive anxiety. Methods: This study uses an analytical descriptive design to see an overview of the anxiety level on adolescents and their knowledge about meditation in reducing anxiety disorders. The sample of the study was 129 respondents. Online questionnaire was used with 9 questions that contain about the anxiety experienced and knowledge about the respondent's meditation. The analysis used a frequency distribution. Results: The results obtained from 129 respondents 85.3% of respondents said they had experienced excessive anxiety and as many as 63.6% of respondents said they had never done meditation. Conclusion: It can be said that the younger generation does not know about meditation and its benefits in reducing anxiety disorders. It is hoped that nurses can educate and introduce several therapies to reduce anxiety, including meditation therapy. [ABSTRACT FROM AUTHOR]

Establishing a wind–photovoltaic (PV)–hydro hybrid system is a new method to utilize wind and PV power, but how multi-energy complementation affects the working modes of cascaded hydropower plants (CHP) has not been fully revealed. In this study, a wind–PV–CHP system was considered as an example and a multi-objective optimal operation model was constructed considering the maximization of both the power generation and the minimum power output. Simulation and comparison results showed that (1) the regulation capability of the hydropower was mainly used for two purposes: one was to drive the early water storage (or water release reduction) of CHP to improve the overall utilization efficiency of water resources, and the other was to drive the hydropower output to cooperate with the non-regulatable wind–PV power output in order to achieve a complementary output; (2) the different minimum outputs determined the regulation capacity for complementing the wind–PV power output, which represents the degree of complementation; and (3) complementation would significantly affect the minimum output and slightly affect the average output of CHP. Overall, this study improves our understanding of wind–PV–hydro complementary operation and can serve as a reference for actual project operation.

Background: The sharing of health-related information has become increasingly popular on social media. Unregulated information sharing has led to the spread of misinformation, especially regarding complementary, alternative, and integrative medicine (CAIM). This scoping review synthesized evidence surrounding the spread of CAIM-related misinformation on social media during the COVID-19 pandemic. Methods: This review was informed by a modified version of the Arksey and O'Malley scoping review framework. AMED, EMBASE, PsycINFO and MEDLINE databases were searched systematically from inception to January 2022. Eligible articles explored COVID-19 misinformation on social media and contained sufficient information on CAIM therapies. Common themes were identified using an inductive thematic analysis approach. Results: Twenty-eight articles were included. The following themes were synthesized: 1) misinformation prompts unsafe and harmful behaviours, 2) misinformation can be separated into different categories, 3) individuals are capable of identifying and refuting CAIM misinformation, and 4) studies argue governments and social media companies have a responsibility to resolve the spread of COVID-19 misinformation. Conclusions: Misinformation can spread more easily when shared on social media. Our review suggests that misinformation about COVID-19 related to CAIM that is disseminated online contributes to unsafe health behaviours, however, this may be remedied via public education initiatives and stricter media guidelines. The results of this scoping review are crucial to understanding the behavioural impacts of the spread of COVID-19 misinformation about CAIM therapies, and can inform the development of public health policies to mitigate these issues.

A novel digital PCR (dPCR) platform combining off-the-shelf reagents, a micro-molded plastic microfluidic consumable with a fully integrated single dPCR instrument was developed to address the needs for routine clinical diagnostics. This new platform offers a simplified workflow that enables: rapid time-to-answer; low potential for cross contamination; minimal sample waste; all within a single integrated instrument. Here we showcase the capability of this fully integrated platform to detect and quantify non-small cell lung carcinoma (NSCLC) rare genetic mutants (EGFR T790M) with precision cell-free DNA (cfDNA) standards. Next, we validated the platform with an established chronic myeloid leukemia (CML) fusion gene (BCR-ABL1) assay down to 0.01% mutant allele frequency to highlight the platform's utility for precision cancer monitoring. Thirdly, using a juvenile myelomonocytic leukemia (JMML) patient-specific assay we demonstrate the ability to precisely track an individual cancer patient's response to therapy and show the patient's achievement of complete molecular remission. These three applications highlight the flexibility and utility of this novel fully integrated dPCR platform that has the potential to transform personalized medicine for cancer recurrence monitoring.

Aedes aegypti transmit pathogenic arboviruses while the mosquito itself tolerates the infection. We examine a piRNA-based immunity that relies on the acquisition of viral derived cDNA (vDNA) and how this pathway discriminates between self and non-self. The piRNAs derived from these vDNAs are essential for virus control and Piwi4 has a central role in the pathway. Piwi4 binds preferentially to virus-derived piRNAs but not to transposon-targeting piRNAs. Analysis of episomal vDNA from infected cells reveals that vDNA molecules are acquired through a discriminatory process of reverse-transcription and recombination directed by endogenous retrotransposons. Using a high-resolution Ae. aegypti genomic sequence, we found that vDNAs integrated in the host genome as endogenous viral elements (EVEs), produce antisense piRNAs that are preferentially loaded onto Piwi4. Importantly, EVE-derived piRNAs are specifically loaded onto Piwi4 to inhibit virus replication. Thus, Ae. aegypti employs a sophisticated antiviral mechanism that promotes viral persistence and generates long-lasting adaptive immunity.

The California poppy (Eschscholzia californica) is renowned for its brilliant golden-orange flowers, though white petal variants have been described. By whole-transcriptome sequencing, we have discovered in multiple white petal varieties a single deletion leading to altered splicing and C-terminal truncation of phytoene synthase (PSY), a key enzyme in carotenoid biosynthesis. Our findings underscore the diverse roles of phytoene synthase in shaping horticultural traits, and resolve a longstanding mystery of the regaled golden poppy.

Cell division is a highly regulated and carefully orchestrated process. Understanding the mechanisms that promote proper cell division is an important step toward unraveling important questions in cell biology and human health. Early studies seeking to dissect the mechanisms of cell division used classical genetics approaches to identify genes involved in mitosis and deployed biochemical approaches to isolate and identify proteins critical for cell division. These studies underscored that post-translational modifications and cyclin-kinase complexes play roles at the heart of the cell division program. Modern approaches for examining the mechanisms of cell division, including the use of high-throughput methods to study the effects of RNAi, cDNA, and chemical libraries, have evolved to encompass a larger biological and chemical space. Here, we outline some of the classical studies that established a foundation for the field and provide an overview of recent approaches that have advanced the study of cell division.

BACKGROUND: Group B enteroviruses are common causes of acute myocarditis, which can be a precursor of chronic myocarditis and dilated cardiomyopathy, leading causes of heart transplantation. To date, the specific viral functions involved in the development of dilated cardiomyopathy remain unclear. METHODS: Total RNA from cardiac tissue of patients with dilated cardiomyopathy was extracted, and sequences corresponding to the 5 termini of enterovirus RNAs were identified. After next-generation RNA sequencing, viral cDNA clones mimicking the enterovirus RNA sequences found in patient tissues were generated in vitro, and their replication and impact on host cell functions were assessed on primary human cardiac cells in culture. RESULTS: Major enterovirus B populations characterized by 5 terminal genomic RNA deletions ranging from 17 to 50 nucleotides were identified either alone or associated with low proportions of intact 5 genomic termini. In situ hybridization and immunohistological assays detected these persistent genomes in clusters of cardiomyocytes. Transfection of viral RNA into primary human cardiomyocytes demonstrated that deleted forms of genomic RNAs displayed early replication activities in the absence of detectable viral plaque formation, whereas mixed deleted and complete forms generated particles capable of inducing cytopathic effects at levels distinct from those observed with full-length forms alone. Moreover, deleted or full-length and mixed forms of viral RNA were capable of directing translation and production of proteolytically active viral proteinase 2A in human cardiomyocytes. CONCLUSIONS: We demonstrate that persistent viral forms are composed of B-type enteroviruses harboring a 5 terminal deletion in their genomic RNAs and that these viruses alone or associated with full-length populations of helper RNAs could impair cardiomyocyte functions by the proteolytic activity of viral proteinase 2A in cases of unexplained dilated cardiomyopathy. These results provide a better understanding of the molecular mechanisms that underlie the persistence of EV forms in human cardiac tissues and should stimulate the development of new therapeutic strategies based on specific inhibitors of the coxsackievirus B proteinase 2A activity for acute and chronic cardiac infections.

Citrus sudden death-associated virus (CSDaV) is a member of the genus Marafivirus in the family Tymoviridae, and has been associated with citrus sudden death (CSD) disease in Brazil. Difficulties in the purification of CSDaV from infected citrus plants have prevented progress in the investigation of the role of this virus in CSD and an understanding of its molecular biology. In this work, we have constructed a full-length cDNA clone of CSDaV driven by the 35S promoter (35SRbz-CSDaV). Agrobacterium tumefaciens-mediated inoculation of 35SRbz-CSDaV in Nicotiana benthamiana plants enabled a fast recovery of large amounts of virions from the agroinfiltrated leaves, which allowed a better molecular characterization of CSDaV. In vivo analyses of mutant versions of 35SRbz-CSDaV revealed the expression strategies used by CSDaV for production of the capsid proteins (CPs). We showed that CSDaV virions contain three forms of CP, each of which is generated from the same coding sequence, but by different mechanisms. The major CPp21 is a product of direct translation by leaky scanning from the second start codon in the subgenomic RNA (sgRNA), whereas the minor CPs, p25 and p23, are produced by direct translation from the first start codon in the sgRNA and by trans-proteolytic cleavage processing derived from the p25 precursor, respectively. Together, these findings contribute to advance our understanding of CSDaV genome expression strategies. In addition, the construction and characterization of the CSDaV infectious clone represent important steps towards the investigation of the role of this virus in CSD and of its use as a tool for citrus biotechnology.

Gene correction in human long-term hematopoietic stem cells (LT-HSCs) could be an effective therapy for monogenic diseases of the blood and immune system. Here we describe an approach for X-linked sSevere cCombined iImmunodeficiency (SCID-X1) using targeted integration of a cDNA into the endogenous start codon to functionally correct disease-causing mutations throughout the gene. Using a CRISPR-Cas9/AAV6 based strategy, we achieve up to 20% targeted integration frequencies in LT-HSCs. As measures of the lack of toxicity we observe no evidence of abnormal hematopoiesis following transplantation and no evidence of off-target mutations using a high-fidelity Cas9 as a ribonucleoprotein complex. We achieve high levels of targeting frequencies (median 45%) in CD34+ HSPCs from six SCID-X1 patients and demonstrate rescue of lymphopoietic defect in a patient derived HSPC population in vitro and in vivo. In sum, our study provides specificity, toxicity and efficacy data supportive of clinical development of genome editing to treat SCID-Xl.

Complementary horticultural therapy has the potential to improve gross motor skills in children. This therapy also provides hands-on experience and trains children's gross motor movements. Stimulated motor development in the form of movements of the hands, fingers, feet and limbs. The purpose is to carry out community service regarding the Implementation of Holticultural Complementary Therapy for Preschool Age Children at Nurul Ikhsan Integrated PAUD. The implementation method is carried out using Guided Training, FGD and Role Play techniques. The implementation of this community service activity was carried out for 2 days directly. The number of PAUD students involved in the activity was 32 people. Community Service Activities are carried out through 3 stages consisting of the preparation stage, implementation stage and evaluation stage. The results of community service activities show that the implementation of community service activities are carried out properly according to the plan. Evaluation of the results of the average value in the group given complementary horticultural therapy, namely pre-test 8.38 and post-test 11.06 and the average value in the group not given complementary horticultural therapy, namely pre-test 8.94 and post-test 9.06. These results can be concluded that there is an increase in gross motor skills in children.

The diversity of antibody variable regions makes cDNA sequencing challenging, and conventional monoclonal antibody cDNA amplification requires the use of degenerate primers. Here, we describe a simplified workflow for amplification of IgG antibody variable regions from hybridoma RNA by a specialized RT-PCR followed by Sanger sequencing. We perform three separate reactions for each hybridoma: one each for kappa, lambda, and heavy chain transcripts. We prime reverse transcription with a primer specific to the respective constant region and use a template-switch oligonucleotide, which creates a custom sequence at the 5' end of the antibody cDNA. This template-switching circumvents the issue of low sequence homology and the need for degenerate primers. Instead, subsequent PCR amplification of the antibody cDNA molecules requires only two primers: one primer specific for the template-switch oligonucleotide sequence and a nested primer to the respective constant region. We successfully sequenced the variable regions of five mouse monoclonal IgG antibodies using this method, which enabled us to design chimeric mouse/human antibody expression plasmids for recombinant antibody production in mammalian cell culture expression systems. All five recombinant antibodies bind their respective antigens with high affinity, confirming that the amino acid sequences determined by our method are correct and demonstrating the high success rate of our method. Furthermore, we also designed RT-PCR primers and amplified the variable regions from RNA of cells transfected with chimeric mouse/human antibody expression plasmids, showing that our approach is also applicable to IgG antibodies of human origin. Our monoclonal antibody sequencing method is highly accurate, user-friendly, and very cost-effective.

Multiple Sclerosis pattients suffer from physiological, psychological and social depending on the type and course of the disease. lower urinary tract symptoms is one of these physiological problems. These symptoms affect the person physiologically, psychologically and socially. Problems such as urinary incontinence, frequency, urgency, incontinence, retantion completely affect daily life of patients. These patients are socially isolated from the society because of frequency, incontinence. However, they limit fluid intake. Multiple sclerosis patients have to restrict shopping, meeting with friends and other social activities due to urinary symptoms. Urinary tract infection associated with low fluid intake and retention is more common in multiple sclerosis than the normal population. Lower urinary symptoms that seen in multiple sclerosis limit the patients life and negatively affect quality of life. In order to eliminate these complaints and increase the quality of life of the patients, these complaints can be minimized by initially bladder training, pelvic floor muscle exercises, nerve stimulation or catheter use. While bladder training extends urination intervals of the patients, pelvic floor muscle exercises reduce the lower urinary tract symptoms by strengthening the muscles. Neuromodulation aims to reduce lower urinary truct symptoms by suppressing neural impulses. Catheter use takes an important role in patients with urinary retention or urinary incontinence. With the catheter used, lower urinary truct symptoms are reduced.

The study aimed to formulate and optimize complementary weaning flours for better antioxidants and sensory acceptability. D-optimal constrained mixture design was used to generate Eighteen formulations with a range of malted sorghum 40–60%, blanched soybean 20–30%, boiled karkade seeds 10–20%, and 10% premix (5% figl leaves, 4.5% sugar, and 0.5% salt) added in equal proportion to all formulations were evaluated. The sensory properties of the gruel made from the selected complementary flour were evaluated using untrained panellists. The result indicated that high malted sorghum supplement levels in the formula were associated with increased bioactive compounds and antioxidant capacity. The optimum values of bioactive compounds, antioxidant capacity, and functional properties of the formulated product were 51.0%, 22.0%, and 17.0% for sorghum, soybean, and karkade seeds flour with 10.0% premix, respectively.
