Protein hydrolysates (PHs) are employed in agriculture to increase the sustainability of farming systems, with positive results on crop productivity and response against environmental stressors. Nevertheless, the molecular mechanism(s) triggered by their specific activity is not clearly understood. In this work, five PHs obtained by enzymatic hydrolysis of different vegetal protein sources were tested for their root-promoting activity on tomato cuttings. All the treatments improved both root length and number when compared to negative controls. Distinctive metabolomic signatures were highlighted in response to treatments, indicating the triggering of different molecular processes in leaf tissues of tomato cuttings. PHs differentially modulated the biosynthesis of plant stress-protectants, such as alkaloids and phenylpropanoids. Moreover, metabolites involved in phytohormone biosynthesis were significantly impacted. In this context, a clear modulation of several compounds related to auxin homeostasis was observed. In addition, the differential modulation of SlIAA2 and SlIAA9 genes, which are involved in the IAA signalling pathway, might further suggest the auxin-like activity elicited by the PHs tested. Here we provide evidence that PHs can impact plant molecular level, positively affecting root development, most likely by affecting the signalling cascades activated in leaf tissues. The biostimulant activity was sustained by PH-specific response at the molecular level, likely ascribable to their heterogeneous botanical origins. In fact, our findings did not point out a clear universal response to PHs, and specific effects are to be investigated.

Abstract How the often highly endemic biodiversity of islands originated has been debated for decades, and it remains a fervid research ground. Here, using mitochondrial and nuclear gene sequence analyses, we investigate the diversity, phylogenetic relationships, and evolutionary history of the mayfly Baetis gr. rhodani on the three largest northwestern Mediterranean islands (Sardinia, Corsica, Elba). We identify three distinct, largely co‐distributed, and deeply differentiated lineages, with divergences tentatively dated back to the Eocene–Oligocene transition. Bayesian population structure analyses reveal a lack of gene exchange between them, even at sites where they are syntopic, indicating that these lineages belong to three putative species. Their phylogenetic relationships with continental relatives, together with the dating estimates, support a role for three processes contributing to this diversity: (1) vicariance, primed by microplate disjunction and oceanic transgression; (2) dispersal from the continent; and (3) speciation within the island group. Thus, our results do not point toward a prevailing role for any of the previously invoked processes. Rather, they suggest that a variety of processes equally contributed to shape the diverse and endemic biota of this group of islands.

Despite the scientific evidence supporting their biostimulant activity, the molecular mechanism(s) underlying the activity of protein hydrolysates (PHs) and the specificity among different products are still poorly explored. This work tested five different protein hydrolysates, produced from different plant sources using the same enzymatic approach, for their ability to promote rooting in tomato cuttings following quick dipping. Provided that all the different PHs increased root length (45–93%) and some of them increased root number (37–56%), untargeted metabolomics followed by multivariate statistics and pathway analysis were used to unravel the molecular processes at the basis of the biostimulant activity. Distinct metabolomic signatures could be found in roots following the PHs treatments. In general, PHs shaped the phytohormone profile, modulating the complex interaction between cytokinins and auxins, an interplay playing a pivotal role in root development, and triggered a down accumulation of brassinosteroids. Concerning secondary metabolism, PHs induced the accumulation of aliphatic glucosinolates, alkaloids, and phenylpropanoids, potentially eliciting crop resilience to stress conditions. Here, we confirm that PHs may have a hormone-like activity, and that their application can modulate plant growth, likely interfering with signaling processes. Noteworthy, the heterogenicity of the botanical origin supported the distinctive and peculiar metabolomic responses we observed across the products tested. While supporting their biostimulant activity, these findings suggest that a generalized crop response to PHs cannot be defined and that specific effects are rather to be investigated.

The study of nitrogen cycling in mountain areas has a long tradition, as it was applied to better understand and describe ecosystem functioning, as well as to quantify long-distance effects of human activities on remote environments. Nonetheless, very few studies, especially in Europe, have considered catchment features controlling nitrogen dynamics above the tree line with focus on running waters. In this study, relationships between some water chemistry descriptors – including nitrogen species and dissolved organic carbon (DOC) – and catchment characteristics were evaluated for a range of sites located above the tree line (1950–2650 m a.s.l.) at Val Masino, in the central Italian Alps. Land cover categories as well as elevation and slope were assessed at each site. Water samples were collected during the 2007 and 2008 snow free periods, with a nearly monthly frequency. In contrast to dissolved organic nitrogen, nitrate concentrations in running waters showed a spatial pattern strictly connected to the fractional extension of tundra and talus in each basin. Exponential models significantly described the relationships between maximum NO3 and the fraction of vegetated soil cover (negative relation) and talus (positive relation), explaining almost 90% of nitrate variation in running waters. Similarly to nitrate but with an opposite behavior, DOC was positively correlated with vegetated soil cover and negatively correlated with talus. Therefore, land cover can be considered one of the most important factors affecting water quality in high-elevation catchments with contrasting effects on N and C pools.

Introduction: This study aims to assess the quality of life (QoL) in patients who have undergone decompressive craniectomy (DC) for any pathology that has caused life-threatening intracranial hypertension. Similarly, it aims to evaluate QoL perceived by caregivers or external informants. In addition to that, the last purpose is to determine which clinical or therapeutic factors could correlate with a better QoL. Methods: A single-center cross-sectional study was designed. All patients over 18 years old who underwent a supratentorial DC at our department due to intracranial hypertension of any etiology, from January 2015 to December 2021, were retrospectively selected. Patients with incomplete follow-up (under 1 year fromthe event or those who died) or who declined to participate in the study were excluded. QoL was assessed with SF-36 and CAVIDACE scales. The correlation between clinical and therapeutic variables and SF-36 subscales was studied with Spearman's correlation and the Mann-Whitney U-test. Results: A total of 55 consecutive patients were recruited: 22 patients had died, three were missed for follow-up, and 15 declined to participate, thus 15 subjects were finally included. The mean follow-up was 47 months (IQR 21.5-67.5). A significant reduction in the "role physical" and "role emotional" subscales of SF-36 was observed compared with the general population. According to caregivers, a significant reduction was assigned to the "physical wellbeing" and "rights" domains. The "physical functioning" score was poorer in women, older patients, those with dominant hemisphere disease, those who required tracheostomy, and those with poor outcomes in the modified Rankin scale. A strong correlation was found between the QoL index at the CAVIDACE scale and the SF-36 subscales "physical functioning" and "role physical". Conclusion: Most patients and caregivers reported acceptable QoL after DC due to a life-threatening disease. A significant reduction in SF- 36 subscales scores "role limitation due to physical problems" and "role limitation due to emotional problems" was referred by patients. According to caregivers' QoL perception, only 25% of the survey's participants showed low scores in the QoL index of the CAVIDACE scale. Only 26.7% of the patients showed mood disorders. Temporary streams are those water courses that undergo the recurrent cessation of flow or the complete drying of their channel. The structure and composition of biological communities in temporary stream reaches are strongly dependent on the temporal changes of the aquatic habitats determined by the hydrological conditions. Therefore, the structural and functional characteristics of aquatic fauna to assess the ecological quality of a temporary stream reach cannot be used without taking into account the controls imposed by the hydrological regime. This paper develops methods for analysing temporary streams' aquatic regimes, based on the definition of six aquatic states that summarize the transient sets of mesohabitats occurring on a given reach at a particular moment, depending on the hydrological conditions: Hyperrheic, Eurheic, Oligorheic, Arheic, Hyporheic and Edaphic. When the hydrological conditions lead to a change in the aquatic state, the structure and composition of the aquatic community changes according to the new set of available habitats. We used the water discharge records from gauging stations or simulations with rainfall-runoff models to infer the temporal patterns of occurrence of these states in the Aquatic States Frequency Graph we developed. The visual analysis of this graph is complemented by the development of two metrics which describe the permanence of flow and the seasonal predictability of zero flow periods. Finally, a classification of temporary streams in four aquatic regimes in terms of their influence over the development of aquatic life is updated from the existing classifications, with stream aquatic regimes defined as Permanent, Temporary-pools, Temporary-dry and Episodic. While aquatic regimes describe the long-term overall variability of the hydrological conditions of the river section and have been used for many years by hydrologists and ecologists, aquatic states describe the availability of mesohabitats in given periods that determine the presence of different biotic assemblages. This novel concept links hydrological and ecological conditions in a unique way. All these methods were implemented with data from eight temporary streams around the Mediterranean within the MIRAGE project. Their application was a precondition to assessing the ecological quality of these streams.
