Abstract

Summary: Bacterial DnaK belongs to the Hsp70 chaperone family, which plays a critical role in maintaining proteostasis by catalyzing protein folding, and is a proposed antibacterial target in the pathogen Mycobacterium tuberculosis. Here, we describe an experimental toolbox for evaluating inhibitors against the mycobacterial DnaK chaperone network: a coupled-enzymatic assay to monitor ATPase activity, a proteolytic cleavage assay to study DnaK conformational changes upon ligand addition, as well as a protein renaturation assay to assess chaperone function.For complete details on the use and execution of this protocol, please refer to Hosfelt et al. Abstract

Physical activity is essential to maintain physical and mental well-being. During the COVID-19 pandemic, in-person physical activity opportunities were limited. This paper describes a telephone-based physical activity support strategy among racially/ethnically diverse patients during the COVID-19 pandemic. Adult patients at a large, Federally Qualified Health Center with an on-site exercise facility referral were eligible to transition to telephone support with personal fitness advisors during the pandemic stay-at-home orders. Baseline surveys assessed physical activity and environmental characteristics; follow-up phone calls used motivational interviewing and physical activity goal setting strategies. From March 23-July 23, 2020, 72 patients participated in 270 phone calls, or 3.8 (±2.1) calls per participant. Participants were, on average, aged 51.3 (±11.6) years, 87.5% female, 31.9% Hispanic/Latino, and 47.2% non-Hispanic Black. Abstract

The article focuses on issues concerning computer privacy in the U.S. Privacy in the digital age is dead. At best, one of the most cherished rights as members of a free society is on life support, kept alive by cybercratic ramblings of an electronic fringe unable to come to grips with reality. Readers are certainly savvier than the newly minted America Online Inc. subscriber. Intimate knowledge of e-mail's inherent lack of privacy is a dangerous thing. It leads to complacency. Given that, most not know how to launch a drive to unionize the workplace using the company email system, to abstain from saying "bad things" about the boss, or sending dirty jokes. Scores of employees, from product managers to chief executive officers, are now busy defending emails they wrote years ago in the courtroom where the government is prosecuting Microsoft Corp. for alleged antitrust violations. Indeed, much of the government's case is drawn from the electronic paper trail of corporate email.

Abstract

29th Aerospace Sciences Meeting January 7-10, 1991/Reno, Nevada Dynamic stall over an oscillating airfoil in compressible flow was studied using a real-tine interferometry technique. Instantaneous flow field data was obtained for various unsteady as well as steady flow conditions. The details of the dynamic stall vortex, including its formation and development have been revealed by the interferograms, resulting in the first documentation of the complete dynamic stall flow field under compressible flow conditions. Comparison of steady flow interferograms with those taken in unsteady flow reveal significant delay in development of leading edge suction peaks in the unsteady case. The interferograms permit detailed analysis of the leading edge pressure field; as many as 13 pressure values have been obtained around the leading edge in the first 1% of the airfoil chord. The results offer significant new insight into the character of the dynamic stall vortex, and the stall delay that is observed during dynamic notions.
