Abstract

Intravenous Immunoglobulin (IVIg) are human IgG derived from plasma pools of healthy donors. Although there are studies in literature evaluating their effectiveness in different pathological animal models, there are no data about their possible role on Bortezomib (BTZ)-induced peripheral neuropathies. Female Wistar rats were treated following a preventive schedule (BTZ and IVIg co-treatment for 3 and 8 weeks) and a therapeutic schedule (4 weeks of BTZ treatment followed by a 4-week IVIg-BTZ cotreatment). Caudal nerve conduction velocity (NCV), plantar and dynamic tests were performed at different time points. Animals were sacrificed after 3ws (acute phase) or 8ws (chronic phase) and tissue samples (Dorsal Root Ganglias -DRG-, sciatic nerve, caudal nerve, skin) were collected for morphological, morphometrical and immunohistological analysis.In the preventive schedule, IVIg was not able to rescue caudal NCV reduction caused by BTZ neither after 3 nor after 8 weeks of co-treatment. Same results were observed in the therapeutic schedule. On the other hand, the evaluation of mechanical allodynia and cold hyperalgesia showed that IVIg injection protected from BTZ effect in both treatment schedules. Morphometric analysis evidenced that, even if not statistically significant only the preventive schedule has a tendency to protect the caudal nerve from BTZ damage. This result is consistent with the morphological evaluation of the nerve. Also, intraepidermal nerve fibers density was preserved in the preventive schedule but not in the therapeutic one. Finally, sciatic nerve and DRG macrophage infiltration levels tended to be reduced in the therapeutic schedule and were brought back to ctrl (rats not treated or injected with IVIg alone) levels in the preventive one. Abstract

In this paper we introduce %CEM, a macro package allowing researchers to automatically perform coarsened exact matching (CEM) in SAS environment. CEM is a non-parametric matching method widely used by researchers to avoid the confounding influence of pre-treatment control variables to improve causal inference in quasi-experimental studies. %CEM introduces a completely automated process which allows SAS users to efficiently perform CEM in fields in which large data sets are common and where SAS is the most popular statistical tool. In addition, such a macro may be used to test several coarsening combinations of numeric variables. This option also provides a visual representation of the matching frontier, thus enabling researchers to select the optimal setting which takes into account both the L1 imbalance and the percentage of matched units. The paper concludes with an empirical application comparing computational performance and results obtained using alternative available software (SAS, R and STATA) using multiple administrative data sets from a large regional database.

Abstract

%CEM is a SAS macro which allows researchers to perform the recently introduced Coarsened Exact Matching (CEM) technique. CEM is a non-parametric matching method to avoid the confounding influence of pre-treatment control variables directly improving causal inference in quasi experimental studies. The %CEM macro introduces a completely automated process wich allows SAS users to perform an exact matching over massive data. Several matching strategies automatically test the population’s imbalance providing the optimal variables binning an associated L1 imbalance measure. An empirical application estimating the causal effect of regional health systems on the intra-hospital mortality using multiple administrative datasets from a large regional database completes the paper.

Abstract

Studies on resource utilization by carnivores are essential to assess the role of the species in the ecosystem. These studies help us to understand the mechanisms that influence vertebrate community structure and the relationships between predators and prey. The aims of this study were to compare and describe the food habits of the sympatric Pampas fox (Lycalopex gymnocercus) and crab‐eating fox (Cerdocyon thous) in the Brazilian Pampa, and to evaluate the food niche overlap between them, and infer aspects of habitat preferences on the basis of their prey. For the diet analyses, 25 stomach contents of Pampas foxes and 33 of crab‐eating foxes were collected. We identified 80 prey items (49 for the Pampas fox and 63 for the crab‐eating fox) and grouped them into eight main categories. Mammals and invertebrates were the most frequent items in the diets of both canids, followed by fruits. The trophic niche breadth of the Pampas fox (Bsta = 0.56) was larger than that of the crab‐eating fox (Bsta = 0.47). The diet overlap between them was Ojk = 0.58. However, a PERMANOVA test showed that diets did not differ between species or season. Our results showed that both L. gymnocercus and C. thous had generalist diets, with a great diversity of prey, but based on similar main food items. Items that contributed the most to the diet of the Pampas fox were insects (Gryllotalpidae and Scarabaeidae) and cavies (Cavia aperea) and those that contributed the most to the diet of the crab‐eating fox were fruits of Syagrus romanzoffiana, beetles (Scarabaeidae), amphibians (Leptodactylus spp.) and cavies. This is the first study to describe and compare the food habits of these two species, occurring in sympatry in the Brazilian Pampa. Interspecific competition for food is a critical factor that determines species coexistence in carnivores. In this study we compare and describe the food habits of two sympatric foxes in southern Neotropics, the Pampas fox (Lycalopex gymnocercus) and the crab‐eating fox (Cerdocyon thous). Our results showed that both species had generalist diets, with a great diversity of prey, but based on similar main food items. The foraging strategies of these canids could be associated with the exploration of different microhabitats. This plasticity may be important for the survival of these species under the constant loss of their natural environments, persecution, and other sources of mortality. [ABSTRACT FROM AUTHOR]

Abstract

This study is aimed at describing the changes occurring in the entire peripheral nervous system sensory pathway along a 2-year observation period in a cohort of C57BL/6 mice. The neurophysiological studies evidenced significant differences in the selected time points corresponding to childhood, young adulthood, adulthood, and aging (i.e., 1, 7, 15, and 25 months of age), with a parabolic course as function of time. The pathological assessment allowed to demonstrate signs of age-related changes since the age of 7 months, with a remarkable increase in both peripheral nerves and dorsal root ganglia at the subsequent time points. These changes were mainly in the myelin sheaths, as also confirmed by the Rotating-Polarization Coherent-Anti-stokes-Raman-scattering microscopy analysis. Evident changes were also present at the morphometric analysis performed on the peripheral nerves, dorsal root ganglia neurons, and skin biopsies. This extensive, multimodal characterization of the peripheral nervous system changes in aging provides the background for future mechanistic studies allowing the selection of the most appropriate time points and readouts according to the investigation aims.

Abstract

BACKGROUND: Antineoplastic drugs, such as cisplatin (CDDP), induce disabling peripheral neuropathies, representing a hindrance to effective cancer treatments. The exact pathogenesis of CDDP-induced neuropathy is not yet understood, and the dysregulation of gene expression has been proposed. Valproate (VPA) is an antiepileptic drug recently discovered to remodel gene expression, with hypothetically putative neuroprotective effects. MATERIALS AND METHODS: VPA was tested in both, in vitro and in vivo models of CDDP-neurotoxicity. RESULTS: VPA administered in combination with CDDP promoted dorsal root ganglia (DRG) neurons survival. Moreover, this treatment induced in Wistar rats an improvement of body weight, sensory nerve conduction velocity, and DRG morphometric analysis. In contrast, VPA was not able to rescue CDDP pre-treated rats. CONCLUSION: When used in combination with CDDP, VPA displays a protective action against neuropathy, in our models, suggesting possible future clinical applications.

Abstract

Paclitaxel is an antineoplastic drug which acts by enhancing tubulin polymerization. The induction of peripheral neuropathy is the main dose-limiting side-effect of paclitaxel treatment. In this study, the neurotoxic effect of this drug in dorsal root ganglion (DRG) explants was analyzed by measuring the neurite length of DRG explants exposed to nerve growth factor (NGF). The neurotoxic effect of paclitaxel is dose- and time-dependent. Moreover, in DRG dissociated post-mitotic neurons, the molecular and morphological features of paclitaxel-induced cellular death were studied and the DRG neurons were observed to die by necrosis. On the contrary, the proliferating human neuroblastoma SH-SY5Y cells exposed to paclitaxel die by apoptosis, as reported for cortical neurons. The different response to the same stimulus of different neuronal populations underlines the importance of the biochemical and molecular phenotype of the neuronal population in determining cellular behavior and vulnerability to the same noxious stimulus.

Abstract

Cisplatin is one of the most effective metal-based anticancer agents, targeting a large number of solid tumours. Despite its efficacy, cisplatin treatment is still limited by severe side effects such as neuro-, hepato- and nephro-toxicity and by resistance phenomena, only partially overcome by the use of new platinum drugs (i.e. oxaliplatin and carboplatin). These problems have stimulated the research and development of alternative therapeutic strategies based on different heavy metals. In this work we investigated the in vitro activity and neurotoxicity of three anticancer complexes: [Cu(PTA)4]PF6, [Cu(thp)4]PF6 and [Au(PTA)4]PF6. Neurotoxicity was evaluated by embryonic rat dorsal root ganglia (DRG) organotypic culture model. Furthermore the extent of proteasome inhibition in rat embryonic DRG neurons was evaluated by fluorimetric assay. After 48 hours of treatment, both copper-based compounds were not neurotoxic even at higher concentrations with respect to the IC50 obtained in A549 and IGROV-1 human cancer cells while [Au(PTA)4]PF6 was neurotoxic at lower concentration than IC50 in cancer cell lines tested. Since the ubiquitin-proteasome system has been identified as molecular target in cancer cells for the heavy metal based-drug, we evaluated their ability to affect the proteasome machinery in DRG neurons. The copper-based compounds, that are not neurotoxic, do not inhibit proteasome activity in DRG neurons. On the contrary, the neurotoxic complex [Au(PTA)4]PF6, induces a significant inhibition of proteasome activity even at concentration lower than the IC50. Furthermore, based on the content in heavy metal-based atoms, the complexes used in this study are suitable candidates for Photon Activation Therapy (PAT). Pre-treatment of IGROV-1 cells with [Cu(PTA)4]PF6 induces an increase cell death with respect to drug or synchrotron (SR) alone. Furthermore the SR/[Cu(PTA)4]PF6 combinational treatment induced an increase in DNA damage with respect to single treatments

Abstract

The GAUDI database (Ground-based Asteroseismology Uniform Database Interface, http://sdc.laeff.esa.es/gaudi/) is a preparatory archive for the COROT (COnvection, ROtation and planetary Transits, http://www.astrsp-mrs.fr/projets/corot/) mission developed at LAEFF (Laboratory for Space Astrophysics and Theoretical Physics, http://www.laeff.esa.es). Its intention is to make the ground-based observations obtained in the preparation of the asteroseismology programme available in a simple and efficient way. It contains spectroscopic and photometric data together with inferred physical parameters for more than 1500 objects gathered since January 1998 in 6 years of observational campaigns. In this paper, the main functionalities and characteristics of the system are described. The observations have been collected at ESO-La Silla, Telescopio Nazionale Galileo, Observatoire de Haute-Provence, South African Astronomical Observatory, Tautenberg Observatory and Sierra Nevada Observatory., 12 pages, 3 figures (quality degraded).
