Birnaviridae is a family of double stranded (ds) RNA virus with non-enveloped virions and 2-segmented genomes. These viruses are known to cause diseases in many hosts. Virus of this family has affected the fish and poultry economy in a wide sector. Unevenness in the use of synonymous codons for a particular amino acid in the coding strand of DNA is known as codon usage bias (CUB). Codons that code the same amino acid are used with variable frequency in a variety of life forms. To understand the pattern of CUB in Birnaviridae, we carried out bioinformatics study to understand the properties of coding sequences of proteins. ENC value of Birnaviridae suggested low CUB. Nucleotide analysis revealed high GC content. Parameters such as RSCU values, nucleotide skewness, translational selection, parity plot and neutrality plot were studied to investigate the pattern of codon use and it was clear that both mutational pressure and natural selection contributed to the designing of CUB in Birnaviridae family. The neutrality plot revealed natural selection to dominate the structuring of CUB and hence remained the major CUB determinant in Birnaviridae. Outcome of our study exemplified the pattern of codon use in the Birnaviridae genomes and contributed the basic primary data for fundamental evolutionary research on them.

Many insect cell lines are persistently infected with insect-specific viruses (ISV) often unrecognized by the scientific community. Considering recent findings showing the possibility of interference between arbovirus and ISV infections, it is important to pay attention to ISV-infected cell lines. One example is the Entomobirnavirus, Culex Y virus (CYV). Here we describe the detection of CYV using a combination of small RNA sequencing, electron microscopy and PCR in mosquito cell lines Aag2, U4.4 and C7-10. We found CYV-specific small RNAs in all three cell lines. Interestingly, the magnitude of the detected viral RNA genome is variable among cell passages and leads to irregular detection via electron microscopy. Gaining insights into the presence of persistent ISV infection in commonly used mosquito cells and their interactions with the host immune system is beneficial for evaluating the outcome of co-infections with arboviruses of public health concern.

Mosquito cell lines have been used extensively in research to isolate and propagate arthropod-borne viruses and understand virus-vector interactions. Despite their utility as an in vitro tool, these cell lines are poorly defined and may harbor insect-specific viruses. Accordingly, we screened four commonly-used mosquito cell lines, C6/36 and U4.4 cells from Aedes albopictus, Aag2 cells from Aedes aegypti, and Hsu cells from Culex quinquefasciatus, for the presence of adventitious (i.e. exogenous) viruses. All four cell lines stained positive for double-stranded RNA, indicative of RNA virus replication. We subsequently identified viruses infecting Aag2, U4.4 and Hsu cell lines using untargeted next-generation sequencing, but not C6/36 cells. PCR confirmation revealed that these sequences stem from active viral replication and/or integration into the cellular genome. Our results show that these commonly-used mosquito cell lines are persistently-infected with several viruses. This finding may be critical to interpreting data generated in these systems. Bipolaris maydis is the causal agent of corn southern leaf blight. Here, we report a novel double-stranded RNA (dsRNA) mycovirus designated Bipolaris maydis botybirnavirus 1 (BmBRV1) from B. maydis strain JZ11 in Jingzhou, Hubei province of China. BmBRV1 has a genome consisting of two dsRNAs (dsRNA1 and dsRNA2) with a size of 6435 and 5987 bp, respectively, each of which contains a single open reading frame (ORF). The two polyproteins encoded by dsRNA1 and dsRNA2 share the highest amino acid identities of 81.8 and 75.3%, respectively, with the RdRp and coat protein of Sclerotinia sclerotiorum botybirnavirus 1 (SsBRV1), a tentative species of the genus Botybirnavirus. Phylogenetic analysis based on the amino acid sequences of RdRp indicated that BmBRV1 belongs to a distinct species of the newly proposed family Botybirnaviridae.

Chicken proventricular necrosis virus (CPNV) is a recently described birnavirus, which has been proposed to be the cause of transmissible viral proventriculitis (TVP). The understanding of the epidemiology of both the virus and the disease is very limited. A retrospective investigation on TVP and CPNV in broiler chicken submissions from the UK from between 1994 and 2015 was performed with the aims of assessing the longitudinal temporal evolution of TVP and CPNV, and to review the histological proventricular lesions in the studied chickens. Ninety-nine of the 135 included submissions (73.3%) fulfilled the TVP-diagnostic criteria, while the remaining 36 submissions (26.7%) displayed only lymphocytic proventriculitis (LP). The first detection of CPNV by PCR dated from 2009. Results showed a rise in the number of both TVP and positive CPNV RT-PCR submissions from 2009 with a peak in 2013, suggesting that they may be an emerging or re-emerging disease and pathogen, respectively. Twenty-two out of the 99 submissions displaying TVP lesions (22%) and four out of the 36 (11%) submissions with LP gave positive CPNV RT-PCR results, further supporting the association between CPNV and TVP and confirming that CPNV is present in a low proportion of proventriculi that do not fulfil the TVP-diagnostic criteria. In addition, intranuclear inclusion bodies were observed in 22 of the submissions with TVP. WOS: 000504656100001 Infectious pancreatic necrosis virus (IPNV; Birnaviridae,Aquabirnavirus)causes infectious pancreatic necrosis (IPN) in fish. IPN disease was first found in 2002 in Turkey. In this study, 10 IPNV isolates were isolated between 2005 and 2013 and propagated in RTG-2 cell cultures. RNAs obtained from cell lysates were used as template and VP1, VP2, VP3, VP4 and VP5 genes of the virus were amplified in full length. Sequence analyses of the genes were made. The obtained sequences were compared with international reference strains from GenBank and phylogenetic analyses and genogrouping of the viruses were conducted. Turkish isolates were found to show a genetic similarity of between 93.5% and 99.8% in terms of molecules. When segment A sequences were compared with each other, a similarity between 97.8% and 99.8% was found; when they were compared with international reference strains, they were found to have the highest similarity (99.3%) with France AJ622822 isolate and the lowest (65.7%) similarity with Canada NC001915 isolate. When segment B sequences of the isolates were compared with each other, a similarity between 93.5% and 99.5% was found; when they were compared with international reference strains, they were found to have the highest similarity (98.9%) with Canada M58757 isolate and the lowest (52.7%) similarity with Finland KY548519 strain. Much of the knowledge on viruses is focused on those that can be propagated using cell-cultures or that can cause disease in humans or in economically important animals and plants. However, this only reflects a small portion of the virosphere. Therefore, in this study, we explore by targeted next-generation sequencing, how the virome varies between Atlantic horse mackerels and gilthead seabreams from fisheries and aquaculture from the center and south regions of Portugal. Viral genomes potentially pathogenic to fish and crustaceans, as well as to humans, were identified namelyese included Astroviridae, Nodaviridae, Hepadnaviridae, Birnaviridae, Caliciviridae, and Picornaviridae families. Also bacteriophages sequences were identified corresponding to the majority of sequencese detected, with Myoviridae, Podoviridae, and Siphoviridae, the most widespread families in both fish species. However, these findings can also be due to the presence of bacteria in fish tissues, or even to contamination. Overall, seabreams harbored viruses from a smaller number of families in comparison with mackerels. Therefore, the obtained data show that fish sold for consumption can harbor a high diversity of viruses, many of which are unknown, reflecting the overall uncharacterized virome of fish. While cross-species transmission of bonafide fish viruses to humans is unlikely, the finding of human pathogenic viruses in fish suggest that fish virome can be a potential threat regarding food safety.

TheBirnaviridaefamily, responsible for major economic losses to poultry and aquaculture, are non-enveloped viruses with a segmented double-stranded (ds)RNA genome that replicate in discrete cytoplasmic virus factories (VFs). Reassortment is common, however, the underlying mechanism remains unknown given that VFs may act as a barrier to genome mixing. In order to provide new information on VF trafficking during dsRNA virus co-infection, we rescued two recombinant infectious bursal disease viruses (IBDVs) of strain PBG98 containing either a split GFP11- or Tetracysteine (TC)- tag fused to the VP1 polymerase (PBG98-VP1-GFP11 and PBG98-VP1-TC). DF-1 cells transfected with GFP1-10 prior to PBG98-VP1-GFP11 infection, or stained with ReAsH following PBG98-VP1-TC infection, had green or red foci in the cytoplasm respectively that co-localised with VP3 and dsRNA, consistent with VFs. We generated the first replication competent tagged reporter birnaviruses, infectious bursal disease viruses (IBDVs) containing a split GFP11 or tetracysteine (TC) tag and used the viruses to track the location and movement of IBDV VFs, in order to better understand the intracellular dynamics of VFs from two different strains of dsRNA virus during a co-infection. Discrete VFs initially formed from each virus that subsequently coalesced from 10 hours pi. We hypothesise that VF coalescence is required for the reassortment of dsRNA viruses and the potential for reassortment increases later in the replication cycle. This study provides new information that adds to our understanding of dsRNA virus VF trafficking.

Infectious bursal disease virus (IBDV) of the Birnaviridae family leads to immunosuppression of young chickens by destroying B cells in the bursa of Fabricius (BFs). Given the increasing number of variant IBDV strains, we urgently require a method to produce attenuated virus for vaccine development. To accomplish this goal, the dual-promoter plasmids in which the RNA polymerase II and RNA polymerase I (Pol I) promoters were placed upstream of the IBDV genomic sequence, which was followed by mouse Pol I terminator and a synthetic polyadenylation signal, were developed for rapid generation of IBDV. This approach did not require trans-supplementation of plasmids for the expression of VP1 and VP3, the main components of IBDV ribonucleoprotein (RNP). Based on the finding in this study that the IBDV RNP activity was partially retained by VP1-FLAG, we successfully rescued the replication-competent IBDV/1FLAG expressing VP1-FLAG. Compared with its parental counterpart, IBDV/1FLAG formed smaller size plaques in cultured cells and induced the same 100% immune protection in vivo. However, neither retarded development nor severe BFs lesion was observed in the IBDV/1FLAG-inoculated chickens. Collectively, this is the first report that viral RNP activity was affected by the addition of an epitope tag on the componential viral proteins. Furthermore, this work demonstrates the rapid generation of attenuated IBDV from dual-promoter plasmids via reducing viral RNP activity by a fused FLAG tag on the C terminus of VP1. This would be a convenient strategy to attenuate epidemic variant IBDV strains for rapid and efficient vaccine development. IMPORTANCE Immunosuppression in chickens as a result of infectious bursal disease virus (IBDV) infection leads to significant economic losses in the poultry industry worldwide every year. Currently, vaccination is still the best way to prevent the prevalence of IBDV. However, with the occurrence of increasing numbers of variant IBDV strains, it is challenging to develop antigen-matched live attenuated vaccine. Here, we first developed a dual-promoter reverse-genetic system for the rapid generation of IBDV. Using this system, the attenuated IBDV/1FLAG expressing VP1-FLAG, which displays the decreased viral RNP activity, was rescued. Moreover, IBDV/1FLAG inoculation induced a similar level of neutralizing antibodies to that of its parental counterpart, protecting chickens against lethal challenge. Our study, for the first time, describes a dual-promoter reverse-genetic approach for the rapid generation of attenuated IBDV while maintaining entire parental antigenicity, suggesting a potential new method to attenuate epidemic variant IBDV strains for vaccine development.

Infectious bursal disease virus (IBDV) is an important member of the Birnaviridae family, causing severe immunosuppressive disease in chickens. The major capsid protein VP2 is responsible for the binding of IBDV to the host cell and its cellular tropism. In order to find proteins that potentially interact with IBDV VP2, a liquid chromatography-mass spectrometry (LC-MS) assay was conducted, and the host chicken CD74 protein was identified. Here, we investigate the role of chicken CD74 in IBDV attachment. Coimmunoprecipitation assays indicated that the extracellular domain of CD74 interacted with the VP2 proteins of multiple IBDV strains. Knockdown and overexpression experiments showed that CD74 promotes viral infectivity. Confocal assays showed that CD74 overexpression allows the attachment of IBDV and subvirus-like particles (SVPs) to the cell surface of nonpermissive cells, and quantitative PCR (qPCR) analysis further confirmed the attachment function of CD74. Anti-CD74 antibody, soluble CD74, depletion of CD74 by small interfering RNA (siRNA), and CD74 knockdown in the IBDV-susceptible DT40 cell line significantly inhibited IBDV binding, suggesting a pivotal role of this protein in virus attachment. These findings demonstrate that CD74 is a novel important receptor for IBDV attachment to the chicken B lymphocyte cell line DT40. IMPORTANCE CD74 plays a pivotal role in the correct folding and functional stability of major histocompatibility complex class II (MHC-II) molecules and in the presentation of antigenic peptides, acting as a regulatory factor in the antigen presentation process. In our study, we demonstrate a novel role of CD74 during IBDV infection, showing that chicken CD74 plays a significant role in IBDV binding to target B cells by interacting with the viral VP2 protein. This is the first report demonstrating that CD74 is involved as a novel attachment receptor in the IBDV life cycle in target B cells, thus contributing new insight into host-pathogen interactions.

Infectious bursal disease virus (IBDV) is a member of the Birnaviridae family that infects B cells in chickens, leading to immunosuppression and mortality. Immunosuppression is known to exacerbate the colonisation and shedding of zoonotic gut bacteria, for example Campylobacter jejuni, Salmonella Enteritidis and Escherichia coli, for reasons that are poorly understood. In order to address this, we infected groups of chickens (n=6) with either a classical strain (F52-70) or a very virulent strain (UK661) of IBDV. At 3 days post-infection, both strains were found to replicate in the gut-associated lymphoid tissue of thecaecaltonsils. 16 s rRNA sequencing revealed that in birds infected with IBDV, regardless of strain, there was a decrease in bacterial diversity in the caecal tonsils, but an increase in diversity of bacteria shed from the cloaca. Secretary IgA binding to commensal bacteria is known to influence the composition of the microbiome, and we speculate that IBDV alters the repertoire of sIgA thereby altering the microbiome composition. Interestingly, we found the number of clostridial species was reduced following IBDV infection. Clostridial species have been shown to induce Treg populations in mice and we speculate that IBDV-mediated changes in the microbiome affect the population of different immune cells in the mucosa. Taken together, we hypothesise that IBDV infection directly affects the B cell population and indirectly affects other immune cell populations in the gut and alters the gut microbiome, which leads to a more favourable environment for zoonotic bacterial infections to colonise.

The Birnaviridae family is comprised of non-enveloped viruses with a double-stranded RNA genome that is divided into two segments, A and B. Birnaviruses are responsible for major economic losses to the poultry and aquaculture industries, and reassortment complicates their epidemiology and control. However, little is known about the nature of theirreplication in cells, or the molecular mechanism underpinning reassortment. Reassortment is one of the main mechanisms of evolution in dsRNA viruses with segmented genomes. It contributes to generate genetic diversity and plays an important role in the emergence and spread of new strains with altered virulence. Natural reassorment has been demonstrated among infectious pancreatic necrosis-like viruses (genus Aquabirnavirus, Birnaviridae). In the present study, coinfections between different viral strains, and genome sequencing by the Sanger and Illumina methods were applied to analyze the frequency of reassortment of this virus in vitro, the possible mechanisms involved, and its effect on virulence. Results have demonstrated that reassortment is a cell-dependent and non-random process, probably through differential expression of the different mRNA classes in the ribosomes of a specific cell, and by specific associations between the components to construct the ribonucleoprotein (RNP) complexes and/or RNP cross-inhibition. However, the precise mechanisms involved, known in other viruses, still remain to be demonstrated in birnaviruses.

IPNV is a salmonid birnavirus that possesses the ability to establish asymptomatic persistent infections in a number of valuable fish species. The presence of IPNV may interfere with subsequent infection by other viruses. In the present study we show that an IPNV-carrier cell line (EPC IPNV ) can induce an antiviral state in fresh EPC by co-cultivating both cell types in three different ways: a “droplet” culture system, a plastic chamber setup, and a transmembrane ( Transwell ® ) system. All three cell co-culture methods were proven useful to study donor/target cell interaction. Naive EPC cells grown in contact with EPC IPNV cells develop resistance to VHSV superinfection. The transmembrane system seems best suited to examine gene expression in donor and target cells separately. Our findings point to the conclusion that one or more soluble factors produced by the IPNV carrier culture induce the innate immune response within the target cells. This antiviral response is associated to the up-regulation of interferon (ifn) and mx gene expression in target EPC cells. To our knowledge this is the first article describing co-culture systems to study the interplay between virus-carrier cells and naive cells in fish.

Background Lates calcarifer, known as seabass in Asia and barramundi in Australia, is a widely farmed species internationally and in Southeast Asia and any disease outbreak will have a great economic impact on the aquaculture industry. Through disease investigation of Asian seabass from a coastal fish farm in 2015 in Singapore, a novel birnavirus named Lates calcarifer Birnavirus (LCBV) was detected and we sought to isolate and characterize the virus through molecular and biochemical methods. Methods In order to propagate the novel birnavirus LCBV, the virus was inoculated into the Bluegill Fry (BF-2) cell line and similar clinical signs of disease were reproduced in an experimental fish challenge study using the virus isolate. Virus morphology was visualized using transmission electron microscopy (TEM). Biochemical analysis using chloroform and 5-Bromo-2′-deoxyuridine (BUDR) sensitivity assays were employed to characterize the virus. Next-Generation Sequencing (NGS) was also used to obtain the virus genome for genetic and phylogenetic analyses. Results The LCBV-infected BF-2 cell line showed cytopathic effects such as rounding and granulation of cells, localized cell death and detachment of cells observed at 3 to 5 days’ post-infection. The propagated virus, when injected intra-peritoneally into naïve Asian seabass under experimental conditions, induced lesions similar to fish naturally infected with LCBV. Morphology of LCBV, visualized under TEM, revealed icosahedral particles around 50 nm in diameter. Chloroform and BUDR sensitivity assays confirmed the virus to be a non-enveloped RNA virus. Further genome analysis using NGS identified the virus to be a birnavirus with two genome segments. Phylogenetic analyses revealed that LCBV is more closely related to the Blosnavirus genus than to the Aquabirnavirus genus within the Birnaviridae family. Conclusions These findings revealed the presence of a novel birnavirus that could be linked to the disease observed in the Asian seabass from the coastal fish farms in Singapore. This calls for more studies on disease transmission and enhanced surveillance programs to be carried out to understand pathogenicity and epidemiology of this novel virus. The gene sequences data obtained from the study can also pave way to the development of PCR-based diagnostic test methods that will enable quick and specific identification of the virus in future disease investigations.

Infectious bursal disease virus (IBDV) belongs to the Birnaviridae family and is the etiological agent of a highly contagious and immunosuppressive disease (IBD) that affects domestic chickens ( Gallus gallus ). IBD or Gumboro disease leads to high rates of morbidity and mortality of infected animals and is responsible for major economic losses to the poultry industry worldwide. IBD is characterized by a massive loss of IgM-bearing B lymphocytes and the destruction of the bursa of Fabricius. The molecular bases of IBDV pathogenicity are still poorly understood; nonetheless, an exacerbated cytokine immune response and B cell depletion due to apoptosis are considered main factors that contribute to the severity of the disease. Here we have studied the role of type I interferon (IFN) in IBDV infection. While IFN pretreatment confers protection against subsequent IBDV infection, the addition of IFN to infected cell cultures early after infection drives massive apoptotic cell death. Downregulation of double-stranded RNA (dsRNA)-dependent protein kinase (PKR), tumor necrosis factor alpha (TNF-α), or nuclear factor κB (NF-κB) expression drastically reduces the extent of apoptosis, indicating that they are critical proteins in the apoptotic response induced by IBDV upon treatment with IFN-α. Our results indicate that IBDV genomic dsRNA is a major viral factor that contributes to the triggering of apoptosis. These findings provide novel insights into the potential mechanisms of IBDV-induced immunosuppression and pathogenesis in chickens. IMPORTANCE IBDV infection represents an important threat to the poultry industry worldwide. IBDV-infected chickens develop severe immunosuppression, which renders them highly susceptible to secondary infections and unresponsive to vaccination against other pathogens. The early dysregulation of the innate immune response led by IBDV infection and the exacerbated apoptosis of B cells have been proposed as the main factors that contribute to virus-induced immunopathogenesis. Our work contributes for the first time to elucidating a potential mechanism driving the apoptotic death of IBDV-infected cells upon exposure to type I IFN. We provide solid evidence about the critical importance of PKR, TNF-α, and NF-κB in this phenomenon. The described mechanism could facilitate the early clearance of infected cells, thereby aiding in the amelioration of IBDV-induced pathogenesis, but it could also contribute to B cell depletion and immunosuppression. The balance between these two opposing effects might be dramatically affected by the genetic backgrounds of both the host and the infecting virus strain.

Birnaviruses are unconventional members of the group of double-stranded RNA (dsRNA) viruses that are characterized by the lack of a transcriptionally active inner core. Instead, the birnaviral particles organize their genome in ribonucleoprotein complexes (RNPs) composed by dsRNA segments, the dsRNA-binding VP3 protein, and the virally encoded RNA-dependent RNA polymerase (RdRp). This and other structural features suggest that birnaviruses may follow a completely different replication program from that followed by members of the Reoviridae family, supporting the hypothesis that birnaviruses are the evolutionary link between single-stranded positive RNA (+ssRNA) and dsRNA viruses. Here we demonstrate that infectious bursal disease virus (IBDV), a prototypical member of the Birnaviridae family, hijacks endosomal membranes of infected cells through the interaction of a viral protein, VP3, with the phospholipids on the cytosolic leaflet of these compartments for replication. Employing a mutagenesis approach, we demonstrated that VP3 domain PATCH 2 (P2) mediates the association of VP3 with the endosomal membranes. To determine the role of VP3 P2 in the context of the virus replication cycle, we used avian cells stably overexpressing VP3 P2 for IBDV infection. Importantly, the intra- and extracellular virus yields, as well as the intracellular levels of VP2 viral capsid protein, were significantly diminished in cells stably overexpressing VP3 P2. Together, our results indicate that the association of VP3 with endosomes has a relevant role in the IBDV replication cycle. This report provides direct experimental evidence for membranous compartments such as endosomes being required by a dsRNA virus for its replication. The results also support the previously proposed role of birnaviruses as an evolutionary link between +ssRNA and dsRNA viruses. IMPORTANCE Infectious bursal disease (IBD; also called Gumboro disease) is an acute, highly contagious immunosuppressive disease that affects young chickens and spreads worldwide. The etiological agent of IBD is infectious bursal disease virus (IBDV). This virus destroys the central immune organ (bursa of Fabricius), resulting in immunosuppression and reduced responses of chickens to vaccines, which increase their susceptibility to other pathogens. IBDV is a member of Birnaviridae family, which comprises unconventional members of dsRNA viruses, whose replication strategy has been scarcely studied. In this report we show that IBDV hijacks the endosomes of the infected cells for establishing viral replication complexes via the association of the ribonucleoprotein complex component VP3 with the phospholipids in the cytosolic leaflet of endosomal membranes. We show that this interaction is mediated by the VP3 PATCH 2 domain and demonstrate its relevant role in the context of viral infection.

The infectious pancreatic necrosis (IPN) caused by a non-enveloped virus of the Birnaviridae family is one of the most important loss factors in the salmonid aquaculture. Virus isolation in the sensitive cell cultures has been approved in the Russian Federation as the diagnostic method for determination of IPNV antigen. This work gives the results of the development of the diagnostic test to reveal IPNV using the antigen-bound ELISA (sandwich ELISA). The developed test supplements a new diagnostic method and verifies some disputable results obtained with classical methods.

In November 2010, a rainbow trout (Oncorhynchus mykiss) hatchery in Victoria reported increased mortality rates in diploid and triploid female fingerlings. Live and moribund fish were submitted for laboratory investigation. All fish showed hyperpigmentation of the cranial half of the body. Histological lesions were seen in all areas of skin examined despite the localised nature of the gross lesions. There was irregular hyperplasia and spongiosis, alternating with areas of thinning and architectural disturbance. Occasionally, particularly in superficial layers of epithe- lium, cells showed large, eosinophilic inclusions that obscured other cellular detail. A small num- ber of fish had necrosis in dermis, subcutis and superficial muscles. Bacteriological culture of skin and gills was negative for all bacterial pathogens, including Flavibacterium columnare, the agent of columnaris disease. Attempts at virus isolation from the skin of affected fish resulted in the development of a cytopathic effect in RTG-2 cell cultures suggestive of the presence of a virus. Negative contrast electron microscopy of cell culture supernatant demonstrated the presence of viral particles with the typical morphology of birnaviruses. Preliminary molecular characterisation identified an aquabirnavirus that differed from both the Tasmanian aquabirnavirus (TABV) and other aquabirnaviruses exotic to Australia. Previous isolates of aquabirnaviruses in Australia and New Zealand have been from healthy fish in a marine environment. This is the first report of an aquabirnavirus isolated from young salmonids at a freshwater hatchery in Australia. The role of the virus in the mortality event on the farm is uncertain as no further deaths attributable to this virus have occurred in the 4 yr since its initial discovery. The virus has been provisionally named Victorian trout aquabirnavirus (VTAB).

Amaç: İnfeksiyoz Pankreatik Nekrozis Virüsü (IPNV; Birnaviridae, Aquabirnavirus) balıklarda İnfeksiyoz Pankreatik Nekrozis (IPN) hastalığını oluşturmaktadır. IPN hastalığı ülkemizde kültürü yapılan gökkuşağı alabalıkları ve kuluçkahanelerinde oldukça yaygın olarak görülmektedir. Bu çalışmada; Türkiye'de gökkuşağı alabalığı çiftliklerinden ve denizde yaşayan kalkan balıklarından izole edilen IPNV suşlarının moleküler karakterizasyonu ve genogruplarının belirlenmesi ile genomik farklılıkların patojenite üzerine etkilerinin olup olmadığının araştırılması amaçlanmıştır.Materyal ve Metot: IPNV izolatları RTG-2 hücre kültürlerinde üretildi ve hücre lizatlarından virüsün VP1, VP2, VP3, VP4 ve VP5 genleri tam uzunlukta amplifiye edildi. Genlerin sekansları yapıldı. IPNV Almus, Hah-4, Hatay07 ve Uşak05 izolatları ile 0,48 gram ağırlıktaki gökkuşağı alabalıklarında immersiyon yöntemi ile deneysel enfeksiyon oluşturuldu. Deneysel enfeksiyon sırasında ölen ve öldürülen balıklarda realtime PCR metodu ile IPNV viral nükleik asit varlığı ve miktarı araştırıldı. Bulgular: İncelenen izolatlarda filogenetik analizler ile % 93,5 ile % 99,8 oranında genetik benzerlik saptandı. Deneysel enfeksiyon sonucunda mortalite oranı; kontrol grubunda % 1,53, Hah-4 ve Hatay07 izolatlarında % 1,42 Almus izolatında % 2,85 olarak gerçekleşti. Uşak05 izolatında ise ölüm gerçekleşmedi. rRT PCR testi sonucunda ölen/öldürülen balık örneklerinde 1,213x106-1,961x102/µl arasında viral kopya sayısı tespit edildi. Balık organ örneklerinde en yoğun CT değeri 21,75 olarak kaydedildi. Hastalığa spesfik klinik belirti veya işaret enfekte edilen, ölen ve öldürülen balıklarda gözlemlenmedi.Sonuç: Türkiye izolatlarının A2 serotipi ve genogrup 5 içinde olduğu, avirülent oldukları ve gökkuşağı alabalığı frylarında persiste enfeksiyon oluşturudukları belirlendi. In this study, establishment of molecular characterization and identification of genogroups with isolated IPNV strains from rainbow trout farm and sea turbot. It was aimed to investigate the effects of genomic differences on pathogenicity. Material and Method: IPNV isolates were propogated in RTG-2 cell line and VP1, VP2, VP3, VP4 and VP5 genes were fully amplified from cell lysates. Sequences were made of the genes and with the isolates that Almus, Hah-4, Hatay07 and Usak05. Experimental infection were established at 0.48 g rainbow trout fry by immersion method. At the dying or killed fish during the experimental infection amount of IPNV viral nucleic acit was investigated by realtime PCR method.Results: Examined isolates were found genetically similar to between 93.5 – 99.8 % in phylogenetic analyses. Mortality rates were detected us in the observed 1.53 %, Hah-4 and Hatay07 1.42 % and Almus 2.85 %. There was no mortality from isolate Usak05. The number of viral copies in the fish samples (dead/killed) were found to be 1.213x106 - 1.961x102/µl by rRT-PCR. The highest CT value in the fish organ samples was recorded as 27.75. Specific clinical symtoms or signs of the disease were not observed in all of the fish.Conclusion: As a result of genetic analysis Turkey isolates were classified in genogroup 5 and serogroup A2 and we determined as avirulent in rainbow trout fry. Highlights • Summary of identified viruses associated with Antarctic animals. • Genomes of Antarctic animals viruses have only been determine in the last five years. • Limited knowledge of animal virology relative to environmental virology in Antarctica., The Antarctic, sub-Antarctic islands and surrounding sea-ice provide a unique environment for the existence of organisms. Nonetheless, birds and seals of a variety of species inhabit them, particularly during their breeding seasons. Early research on Antarctic wildlife health, using serology-based assays, showed exposure to viruses in the families Birnaviridae, Flaviviridae, Herpesviridae, Orthomyxoviridae and Paramyxoviridae circulating in seals (Phocidae), penguins (Spheniscidae), petrels (Procellariidae) and skuas (Stercorariidae). It is only during the last decade or so that polymerase chain reaction-based assays have been used to characterize viruses associated with Antarctic animals. Furthermore, it is only during the last five years that full/whole genomes of viruses (adenoviruses, anelloviruses, orthomyxoviruses, a papillomavirus, paramyoviruses, polyomaviruses and a togavirus) have been sequenced using Sanger sequencing or high throughput sequencing (HTS) approaches. This review summaries the knowledge of animal Antarctic virology and discusses potential future directions with the advent of HTS in virus discovery and ecology.

Although a wide range of viruses have been reported in marine molluscs, most of these reports rely on ultrastructural examination and few of these viruses have been fully characterized. The lack of marine mollusc cell lines restricts virus isolation capacities and subsequent characterization works. Our current knowledge is mostly restricted to viruses affecting farmed species such as oysters Crassostrea gigas, abalone Haliotis diversicolor supertexta or the scallop Chlamys farreri. Molecular approaches which are needed to identify virus affiliation have been carried out for a small number of viruses, most of them belonging to the Herpesviridae and birnaviridae families. These last years, the use of New Generation Sequencing approach has allowed increasing the number of sequenced viral genomes and has improved our capacity to investigate the diversity of viruses infecting marine molluscs. This new information has in turn allowed designing more efficient diagnostic tools. Moreover, the development of experimental infection protocols has answered some questions regarding the pathogenesis of these viruses and their interactions with their hosts. Control and management of viral diseases in molluscs mostly involve active surveillance, implementation of effective bio security measures and development of breeding programs. However factors triggering pathogen development and the life cycle and status of the viruses outside their mollusc hosts still need further investigations.

International audience; Birnaviridae is a family of viruses with bi-segmented dsRNA genomes totalling about 6 kbp forming icosahedral, non-enveloped virions. The family includes four genera, members of three of which (Aquabirnavirus, Avibirnavirus and Blosnavirus) infect vertebrates (excluding mammals), whereas members of the fourth genus (Entomobirnavirus) infect insects. Each genus includes 1-3 species. Infectious pancreatic necrosis virus of salmonids and infectious bursal disease virus of poultry are two economically important birnaviruses. International audience; Heparin is one of the main pharmaceutical products manufactured from raw animal material. In order to describe the viral burden associated with this raw material, we performed high-throughput sequencing (HTS) on mucus samples destined for heparin manufacturing, which were collected from European pigs. We identified Circoviridae and Parvoviridae members as the most prevalent contaminating viruses, together with viruses from the Picomaviridae, Astroviridae, Reoviridae, Caliciviridae, Adenoviridae, Birnaviridae, and Anelloviridae families. Putative new viral species were also identified. The load of several known or novel small non-enveloped viruses, which are particularly difficult to inactivate or eliminate during heparin processing, was quantified by qPCR. Analysis of the combined HTS and specific qPCR results will influence the refining and validation of inactivation procedures, as well as aiding in risk analysis of viral heparin contamination.

Antimicrobial immune defense is evolutionarily preserved in all organisms. Mammals have developed robust, protein-based antiviral defenses, which are under constant investigation. Studies have provided evidences for the various fish immune factors sharing similarity with those of mammals. In this study, we have identified an ortholog of mitochondrial antiviral signaling protein from rock bream, Oplegnathus fasciatus. RbMAVS cDNA possesses an open reading frame (ORF) of 1758 bp coding for a protein of 586 amino acids with molecular mass of approximately 62 kDa and isoelectric point of 4.6. In silico analysis of RbMAVS protein revealed a caspase recruitment domain (CARD), a proline rich domain and a transmembrane domain. RbMAVS protein also contains a putative TRAF2 binding motif, 319PVQDT323. Primary sequence comparison of RbMAVS with other orthologues revealed heterogeneity towards the C-terminus after the CARD region. RbMAVS transcripts were evident in all the examined tissues. RbMAVS expression was induced in vivo after poly I:C challenge in peripheral blood cells, liver, head kidney and spleen tissues. Over-expression of RbMAVS potently inhibited marine birnavirus (MABV) infection in rock bream heart cells and induced various cytokines and signaling molecules in vitro. Thus, RbMAVS is an antiviral protein and potentially involved in the recognition and signaling of antiviral defense mechanism in rock bream.

Infectious pancreatic necrosis virus (IPNV) is a non-enveloped virus belonging to the Birnaviridae family. IPNV produces an acute disease in salmon fingerlings, with high mortality rates and persistent infection in survivors. Although there are reports of IPNV binding to various cells, the viral receptor and entry pathways remain unknown. The aim of this study was to determine the endocytic pathway that allows for IPNV entry. We observed that IPNV stimulated fluid uptake and virus particles co-localysed with the uptake marker dextran in intracellular compartments, suggesting a role for macropinocytosis in viral entry. Consistent with this idea, viral infection was significantly reduced when the Na+/H+ exchanger NHE1 was inhibited with 5-(N-Ethyl-N-isopropyl) amiloride (EIPA). Neither chlorpromazine nor filipin complex I affected IPNV infection. To examine the role of macropinocytosis regulators, additional inhibitors were tested. Inhibitors of the EGFR pathway and the effectors Pak1, Rac1 and PKC reduced viral infection. Together, our results indicate that IPNV is mainly internalized into CHSE-214 cells by macropinocytosis.

Projede, IPNV' yi üretmek için rainbow trout gonad (RTG-2) hücre kültürü kullanıldı. Hücreler kültüre edildikten sonra IPNV inokule edilerek CPE oluşumu takip edildi. Projede kullanılan antioksidan madde olan kuersetinin RTG-2 hücresindeki nonsitotoksik dozu Cell Counting Kit-8 (CCK-8, Sigma-Aldrich) ile belirlendi. Kuersetinin, IPNV replikasyonu üzerindeki etkisinin belirlenmesinde titrasyon testi, sitopatojenik etki redüksiyon testi ve qRT-PCR testleri kullanıldı. Üç tekrarlı sitotoksisite denemelerinden elde edilen verilerin ortalama değeri hesaplandı ve kuersetinin farklı dozlarının uygulandığı gruplar arasındaki titre farklılıkları ise % olarak ifade edildi.RTG-2 hücrelerinde kuersetinin non toksik dozunun 50 µmol/L olduğu, 50μmol/L kuersetinin RTG-2 hücrelerinde üretilen IPNV' nin titresini 10-7' den10-5' edüşürürken, aynı dozdaki kuersetinin viral yükü %40 oranında azalttığı belirlendi. The purpose of this master thesis was to investigate the effect of quercetin on replication in cell cultures of infectious pancreatic necrosis virus (IPNV), which is important for the fish industry. In the project, rainbow trout gonad (RTG-2) cell culture was used to produce IPNV, after cells were cultured, IPNV was inoculated followed by CPE formation. The noncitotoxic dose of quercetin in the RTG-2 cell, the antioxidant used in the project, was determined by Cell Counting Kit-8 (CCK-8, Sigma-Aldrich). Titration test, cytopathogenic effect reduction test and qRT-PCR tests were used to determine the effect of quercetin on IPNV replication. The mean value of the data from the triplicate cytotoxicity assays was calculated and the titer differences between the groups to which different doses of quercetin were administered were expressed as %. The non-toxic dose of quercetin in RTG-2 cells was 50µmol/L, and 50 μMol/L quercetin lowered the titre of IPNV produced in RTG-2 cells to 10-7 μM 10-5, while the same dose of quercetin reduced the viral load by 40%. In this thesis, it is shown that quercetin, a potent antioxidant and antiinflammatory agent, inhibits IPNV replication in in vitro conditions and that IPN may be an option in the treatment of the disease. 50

Infectious bursal disease virus (IBDV) is a Birnaviridae family member of economic importance for poultry. This virus infects and destroys developing B lymphocytes in the cloacal bursa, resulting in a potentially fatal or immunosuppressive disease in chickens. Naturally occurring viruses and many vaccine strains are not able to grow in in vitro systems without prior adaptation, which often affects viral properties such as virulence. Primary bursal cells, which are the main target cells of lymphotropic IBDV in vivo, may represent an attractive system for the study of IBDV. Unfortunately, bursal cells isolated from bursal follicles undergo apoptosis within hours following their isolation. Here, we demonstrate that ex vivo stimulation of bursal cells with phorbol 12-myristate 13-acetate maintains their viability long enough to allow IBDV replication to high titres. A wide range of field-derived or vaccine serotype 1 IBDV strains could be titrated in these phorbol 12-myristate 13-acetate -stimulated bursal cells and furthermore were permissive for replication of non-cell-culture-adapted viruses. These cells also supported multistep replication experiments and flow cytometry analysis of infection. Ex vivo-stimulated bursal cells therefore offer a promising tool in the study of IBDV.

The infectious bursal disease virus (IBDV) is a major health threat to the world’s poultry industry despite intensive controls including proper biosafety practices and vaccination. IBDV (Avibirnavirus, Birnaviridae) is a non-enveloped virus with a bisegmented double-stranded RNA genome. The virus is traditionally classified into classic, variant and very virulent strains, each with different epidemiological relevance and clinical implications. Recently, a novel worldwide spread genetic lineage was described and denoted as distinct (d) IBDV. Here, we report the development and validation of a reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assay for the specific detection of dIBDVs in the global poultry industry. The assay employs a TaqMan-MGB probe that hybridizes with a unique molecular signature of dIBDV. The assay successfully detected all the assessed strains belonging to the dIBDV genetic lineage, showing high specificity and absence of cross-reactivity with non-dIBDVs, IBDVnegative samples and other common avian viruses. Using serial dilutions of in vitro-transcribed RNA we obtained acceptable PCR efficiencies and determination coefficients, and relatively small intra- and inter-assay variability. The assay demonstrated a wide dynamic range between 103 and 108 RNA copies/reaction. This rapid, specific and quantitative assay is expected to improve IBDV surveillance and control worldwide and to increase our understanding of the molecular epidemiology of this economically detrimental poultry pathogen.

Thosea asigna virus (TaV) is a positive-sense, single-stranded RNA (ssRNA) virus that belongs to the Permutotetravirus genera within the recently created Permutotetraviridae family. The genome of TaV consists of an RNA segment of about 5.700 nucleotides with two open reading frames, encoding for the replicase and capsid protein. The particular TaV replicase does not contain N7-methyl transferase and helicase domains but includes a structurally unique RNA-dependent RNA polymerase (RdRp) with a sequence permutation in the domain where the active site is anchored. This architecture is also found in double-stranded RNA viruses of the Birnaviridae family. Here we report the purification and preliminary crystallographic studies TaV RdRp. The enzyme was crystallized by the sitting-drop vapour diffusion method using PEG 8K and lithium sulfate as precipitants. Two different crystal forms were obtained: native RdRp crystallized in space group P2 12 12 and diffracts up to 2.1 Å and the RdRp-Lu 3+ derivative co-crystals belong to the C222 1 space group, diffracting to 3.0 Å resolution. Espirito Santo virus (ESV) is a newly discovered virus recovered as contamination in a sample of a virulent strain of dengue-2 virus (strain 44/2), which was recovered from a patient in the state of Espirito Santo, Brazil, and amplified in insect cells. ESV was found to be dependent upon coinfection with a virulent strain of dengue-2 virus and to replicate in C6/36 insect cells but not in mammalian Vero cells. A sequence of the genome has been produced by de novo assembly and was not found to match to any known viral sequence. An incomplete match to the nucleotide sequence of the RNA-dependent RNA polymerase from Drosophila X virus (DXV), another birnavirus, could be detected. Mass spectrometry analysis of ESV proteins found no matches in the protein data banks. However, peptides recovered by mass spectrometry corresponded to the de novo -assembled sequence by BLAST analysis. The composition and three-dimensional structure of ESV are presented, and its sequence is compared to those of other members of the birnavirus family. Although the virus was found to belong to the family Birnaviridae , biochemical and sequence information for ESV differed from that of DXV, the representative species of the genus Entomobirnavirus . Thus, significant differences underscore the uniqueness of this infectious agent, and its relationship to the coinfecting virus is discussed.

Diseases are an important cause of losses and decreased production rates in freshwater eel farming, and have been suggested to play a contributory role in the worldwide decline in wild freshwater eel stocks. Three commonly detected pathogenic viruses of European eel Anguilla anguilla are the aquabirnavirus eel virus European (EVE), the rhabdovirus eel virus European X (EVEX), and the alloherpesvirus anguillid herpesvirus 1 (AngHV1). In general, all 3 viruses cause a nonspecific haemorrhagic disease with increased mortality rates. This review provides an overview of the current knowledge on the aetiology, prevalence, clinical signs and gross pathology of these 3 viruses. Reported experimental infections showed the temperature dependency and potential pathogenicity of these viruses for eels and other fish species. In addition to the published literature, an overview of the isolation of pathogenic viruses from wild and farmed A. anguilla in the Netherlands during the past 2 decades is given. A total of 249 wild A. anguilla, 39 batches of glass eels intended for farming purposes, and 239 batches of farmed European eels were necropsied and examined virologically. AngHV1 was isolated from wild A. anguilla yellow and silver eels from the Netherlands from 1998 until the present, while EVEX was only found sporadically, and EVE was never isolated. In farmed A. anguilla AngHV1 was also the most commonly isolated virus, followed by EVE and EVEX.

The differential gene expression profile of survivors of unvaccinated Atlantic salmon challenged by cohabitation (81% mortality) with a highly virulent strain of IPNV was studied using suppressive subtractive hybridisation (SSH). RNA was extracted from head-kidney from fish prior to IPNV challenge and 4 weeks post challenge (survivors). Five hundred and seventy six cDNA clones were randomly selected for sequencing, BLAST N and BLAST X analyses were performed. 180 unique genes were found, out of these 167 were singletons and 13 were contigs. These genes were grouped according to their putative functions using Blast2Go and TrEMBL. Ten out of 14 genes identified by SSH were confirmed by quantitative real time PCR. Representatives of almost all the biological functional groups were found, including immune and stress response, transcription, translation, metabolism and protein transport and all were upregulated in the survivor fish. Viral Haemorrhagic Septicaemia Virus induced gene 2 (Vig-2) was among the genes found to be upregulated in the survivors and this prompted us to assess the expression level of IFN-α and Mx, which were both found to be significantly upregulated, alongside inflammatory markers (P-selectin) and proteolysis genes (proteasome subunit alpha type 2). In summary the results obtained point towards innate responses being induced in surviving fish combined with factors involved in protein degradation and yet un-classified genes with regard to antiviral functions (Vig-2).

Atlantic salmon is one of the most important aquaculture fish species in the world. A cDNA library was constructed from splenic leukocytes of Atlantic salmon challenged with infectious pancreatic necrosis virus, and sequenced to collect genomic information and identify genes involved in immune defense response. Sequencing of 1360 clones yielded 1043 high quality ESTs (expressed sequence tags) and these ESTs were assembled into 823 unigenes. BLAST analysis revealed that 203 unigenes were novel genes that had no significant matches to any protein sequences in the public databases. The remaining 586 unigenes were closely matched to the known genes or sequences deposited in public databases and 34 unigenes matched to unknown genes. Among them, GO and KEGG analyses identified several functional categories involved in the innate immune response, with immune response and cytokine activity being predominant. These results provided a useful resource for gene discovery for further research of this species.

The infectious pancreatic necrosis virus (IPNV) belongs to the Birnaviridae family of viruses and causes acute contagious diseases in a number of economically important freshwater and marine fish. In this study, we infected zebrafish embryonic cells (ZF4) with IPNV and analyzed the gene expression patterns of normal and infected cells using quantitative real-time PCR. We identified a number of immune response genes, including ifna, ifng, mx, irf1, irf2, irf4, tnfa, tnfb, il-1b, il-15, il-26, ccl4 and mmp family genes, that are induced after viral infection. Transcriptional regulators, including cebpb, junb, nfkb and stat1, stat4 and stat5, were also upregulated in IPNV-infected cells. In addition, we used Pathway Studio software to identify TNFα as having the greatest downstream influence among these altered genes. Treating virus-infected cells with an siRNA targeting TNFα inhibited NF-κB expression. To further interrupt the TNFα/NF-κB-mediated pathway, the expression levels of cytokines and metalloproteinases were inhibited in IPNV-infected cells. These data suggest that, during IPNV infection, the expression of cytokines and metalloproteinases might be initiated through the TNFα/NF-κB-mediated pathway. The modulation of TNFα/NF-κB-related mechanisms may provide a therapeutic strategy for inhibiting viral infection in teleosts.

Chicken proventricular necrosis virus (CPNV), isolate R11/3, previously was isolated from transmissible viral proventriculitis-affected chickens and was determined to be the likely etiology of this disease. CPNV was identified as a birnavirus on the basis of virion size and morphology (icosahedral, approximately 75 nm in diameter, nonenveloped); buoyant density in cesium chloride (1.32 g/ml); a genome comprising bisegmented, double-stranded RNA (approximately 3.8 and 3.4 kilobase pairs); and nucleotide sequence analyses. Nucleotide sequencing of CPNV RNA, segment B, identified a single large open reading frame that encodes a 903-amino acid protein. The 903-amino acid protein was identified as the putative VP1, the viral RNA-dependent RNA polymerase (RdRp), on the basis of sequence homologies with other birnavirus VP1 proteins. The CPNV VP1 possessed the unique permuted RdRp sequence motif arrangement characteristic of birnaviruses; however, phylogenetic analyses based on VP1 demonstrated that CPNV is deeply divergent from other birnaviruses.

The delivery of foreign epitopes by a replicating nonpathogenic avian infectious bursal disease virus (IBDV) was explored. The aim of the study was to identify regions in the IBDV genome that are amenable to the introduction of a sequence encoding a foreign peptide. By using a cDNA-based reverse genetics system, insertions or substitutions of sequences encoding epitope tags (FLAG, c-Myc, or hepatitis C virus epitopes) were engineered in the open reading frames of a nonstructural protein (VP5) and the capsid protein (VP2). Attempts were also made to generate recombinant IBDV that displayed foreign epitopes in the exposed loops (P BC and P HI ) of the VP2 trimer. We successfully recovered recombinant IBDVs expressing c-Myc and two different virus-neutralizing epitopes of human hepatitis C virus (HCV) envelope glycoprotein E in the VP5 region. Western blot analyses with anti-c-Myc and anti-HCV antibodies provided positive identification of both the c-Myc and HCV epitopes that were fused to the N terminus of VP5. Genetic analysis showed that the recombinants carrying the c-Myc/HCV epitopes maintained the foreign gene sequences and were stable after several passages in Vero and 293T cells. This is the first report describing efficient expression of foreign peptides from a replication-competent IBDV and demonstrates the potential of this virus as a vector.

Development of rapid antiviral assays can expedite the process of screening potential agents against viral pathogens. In the present study, fluorescent quantum dots (QDs) incorporated with infectious pancreatic necrosis virus (IPNV) were used as imaging nanoprobes to detect the threshold amount of poly I:C (an interferon inducer) required to induce zebrafish cells into an antiviral state against IPNV. QD-IPNV hybrids were formed by colloidal clustering of negatively charged QDs and IPNV, using the cationic polymer polybrene (50 μg/mL). To test the screening potential of the QD-IPNV hybrids for anti-IPNV drug candidates, zebrafish ZF4 cells primed with the immunostimulant poly I:C at concentrations of 1, 5, and 10 μg/mL for 6 h were used as a model system. After poly I:C treatment, cells were exposed to the QD-IPNV hybrids for 6 h at a multiplicity of infection (MOI) of 5. The anti-IPNV effectiveness of poly I:C was assessed via fluorescence intensity of the QDs. Our results showed that ZF4 cells primed with poly I:C at 10 μg/mL were highly protected from IPNV challenge (i.e., no detection of QD fluorescence). In summary, a rapid and efficient cell-based imaging platform has been developed for assessing the anti-IPNV activity of poly I:C on ZF4 cells using QD-IPNV hybrids. This approach may be applied to a wider range of fish species and fish pathogenic viruses.

Infectious pancreatic necrosis virus (IPNV) is a major viral pathogen of salmonid fish and causes serious economic losses to salmonid aquaculture. Previously, we demonstrated that the IPNV capsid protein, VP2, expressed in yeast self-assembles into subviral particles (SVPs) and injection of these IPNV rVP2 SVPs into rainbow trout elicits an immune response. Immunized fish had reduced viral loads compared to unimmunized fish when challenged with IPNV. To evaluate the suitability of IPNV rVP2 SVPs for future development of multivalent vaccines, a linear epitope of a human oncogene, c-myc, was cloned into the IPNV rVP2 SVP backbone as a model epitope and expressed in yeast. Western blot analyses with anti-c-myc and anti-IPNV antibodies provided positive identification of both the c-myc and VP2 epitopes on the c-myc VP2 SVPs. Transmission electron microscopy of purified chimeric c-myc VP2 SVPs revealed the formation of approximately 20nm particles. Rainbow trout immunized with c-myc VP2 SVPs elicited both anti-c-myc and anti-IPNV immune responses. When immunized fish were challenged with IPNV, the viral load in the c-myc VP2 SVP immunized fish was significantly lower than the sham-vaccinated controls. The results indicate that IPNV rVP2 SVPs can tolerate the insertion of foreign epitopes without affecting either the antigenic potential of the epitopes of the backbone protein or the inserted foreign epitope. This opens the possibility of using the IPNV rVP2 SVP platform to express epitopes of other viruses, which could pave the way for development of multivalent subunit vaccines or novel marker vaccines.
