Abstract Dietary lipids can affect metabolic health through gut microbiota-mediated mechanisms, but the influence of lipid-microbiota interaction on liver steatosis is largely unknown. We investigate the impact of dietary lipids on human gut microbiota composition and the effects of microbiota-lipid interactions on steatosis in male mice. In humans, low intake of saturated fatty acids (SFA) is associated with increased microbial diversity independent of fiber intake. In mice, poorly absorbed dietary long-chain SFA, particularly stearic acid, induce a shift in bile acid profile and improved metabolism and steatosis. These benefits are dependent on the gut microbiota, as they are transmitted by microbial transfer. Diets enriched in polyunsaturated fatty acids are protective against steatosis but have minor influence on the microbiota. In summary, we find that diets enriched in poorly absorbed long-chain SFA modulate gut microbiota profiles independent of fiber intake, and this interaction is relevant to improve metabolism and decrease liver steatosis.

Background: Schizophrenia (SCZ) is a heterogeneous neuropsychiatric disorder for which current treatment has insufficient efficacy and severe adverse effects. The modifiable gut microbiome might be a potential target for intervention to improve neurobiological functions through the gut-microbiome-brain axis. Methods: In this case-control study, gut microbiota of 132 patients with SCZ and increased waist circumference were compared with gut microbiota of two age- and sex-matched control groups, composed of 132 healthy individuals and 132 individuals with metabolic syndrome. Shotgun sequencing was used to characterize fecal samples at the taxonomic and functional levels. Cognition of the patients with SCZ was evaluated using the Brief Assessment of Cognition instrument. Results: SCZ gut microbiota differed significantly from those of healthy control subjects and individuals with metabolic syndrome in terms of richness and global composition. SCZ gut microbiota were notably enriched in Flavonifractor plautii, Collinsella aerofaciens, Bilophila wadsworthia, and Sellimonas intestinalis, while depleted in Faecalibacterium prausnitzii, Ruminococcus lactaris, Ruminococcus bicirculans, and Veillonella rogosae. Functional potential of the gut microbiota accounted for 11% of cognition variability. In particular, the bacterial functional module for synthesizing tyrosine, a precursor for dopamine, was in SCZ cases positively associated with cognitive score (ρ = 0.34, q ≤ .1). Conclusions: Overall, this study shows that the gut microbiome of patients with SCZ differs greatly from that of healthy control subjects or individuals with metabolic syndrome. Cognitive function of patients with SCZ is associated with the potential for gut bacterial biosynthesis of tyrosine, a precursor for dopamine, suggesting that gut microbiota might be an intervention target for alleviation of cognitive dysfunction in SCZ.

Abstract Background The RTOG 94-13 trial has provided evidence that patients with high risk prostate cancer benefit from an additional radiotherapy to the pelvic nodes combined with concomitant hormonal ablation. Since lymphatic drainage of the prostate is highly variable, the optimal target volume definition for the pelvic lymph nodes is problematic. To overcome this limitation, we tested the feasibility of an intensity modulated radiation therapy (IMRT) protocol, taking under consideration the individual pelvic sentinel node drainage pattern by SPECT functional imaging. Methods Patients with high risk prostate cancer were included. Sentinel nodes (SN) were localised 1.5–3 hours after injection of 250 MBq 99mTc-Nanocoll using a double-headed gamma camera with an integrated X-Ray device. All sentinel node localisations were included into the pelvic clinical target volume (CTV). Dose prescriptions were 50.4 Gy (5 × 1.8 Gy / week) to the pelvis and 70.0 Gy (5 × 2.0 Gy / week) to the prostate including the base of seminal vesicles or whole seminal vesicles. Patients were treated with IMRT. Furthermore a theoretical comparison between IMRT and a three-dimensional conformal technique was performed. Results Since 08/2003 6 patients were treated with this protocol. All patients had detectable sentinel lymph nodes (total 29). 4 of 6 patients showed sentinel node localisations (total 10), that would not have been treated adequately with CT-based planning ('geographical miss') only. The most common localisation for a probable geographical miss was the perirectal area. The comparison between dose-volume-histograms of IMRT- and conventional CT-planning demonstrated clear superiority of IMRT when all sentinel lymph nodes were included. IMRT allowed a significantly better sparing of normal tissue and reduced volumes of small bowel, large bowel and rectum irradiated with critical doses. No gastrointestinal or genitourinary acute toxicity Grade 3 or 4 (RTOG) occurred. Conclusion IMRT based on sentinel lymph node identification is feasible and reduces the probability of a geographical miss. Furthermore, IMRT allows a pronounced sparing of normal tissue irradiation. Thus, the chosen approach will help to increase the curative potential of radiotherapy in high risk prostate cancer patients.

Background: Assessment of age-associated disorders has become increasingly important. Methods: In a clinical setting, people with intellectual disability with and without dementia were assessed retrospectively using the Neuropsychological Test Battery (NTB) and the Dementia Questionnaire for People with Learning Disabilities (DLD) at two different times to analyse neuropsychological changes and diagnostic validity. One group (n = 44) was assessed with both instruments, while the DLD was applied in 71 patients. Results: In the NTB (n = 44), only patients with dementia (n = 26) showed a decline in the NTB total score and three subscales. Receiver operating characteristic analysis revealed a diagnostic sensitivity of 0.67, a specificity of 0.81, and an area under the curve (AUC) of 0.767. In the DLD group (n = 71), only those with dementia displayed a decrease in the cognitive and social scale; diagnostic sensitivity and specificity values were low (0.61/0.63) and the AUC was 0.704. Conclusions: Neuropsychological assessment was sensitive to detect cognitive changes over time. Sensitivity values of both instruments suggest a reassessment at a later time point.

Platinum dissolution in PEM fuel cells is an increasingly important indicator for the state-of-health and lifetime prediction of fuel cells in real applications. For this reason, portable online analysis tools are needed that can detect and quantify platinum with high sensitivity, selectivity, and accuracy in the product water of fuel cells. We validated the hanging mercury drop electrode (HMDE) and non-toxic bismuth film electrodes for the voltammetric determination of platinum for this purpose. Bismuth films were prepared by reductive deposition on both a glassy carbon solid state electrode and on a screen-printed electrode (film on-chip electrode). Both bismuth film electrodes could be successfully validated for the determination of platinum by adsorptive stripping voltammetry. An LOD of 7.9 μg/L and an LOQ of 29.1 μg/L were determined for the bismuth film solid state electrode, values of 22.5 μg/L for the LOD and of 79.0 μg/L for the LOQ were obtained for the bismuth film on-chip electrode. These numbers are still much higher than the results measured with the HMDE (LOD: 0.76 ng/L; LOQ: 2.8 ng/L) and are not sufficient to detect platinum in the product water of a fuel cell run in different load tests. The amount of dissolved platinum produced by a 100 W fuel cell stack upon dynamic and continuous high load cycling, respectively, was in the range of 2.9–4.1 ng/L, which could only be detected by the HMDE.

In der Corona-Pandemie kommt der Krisenkommunikation der 16 deutschen Landesregierungen eine entscheidende Rolle zu. Der Beitrag untersucht diese Krisenkommunikation im ersten Jahr der weltumspannenden Gesundheitskrise und analysiert hierzu über 27000 Pressemitteilungen, Tweets und Facebook-Posts. Mittels der Methode der (teil-)automatisierten Inhaltsanalyse kann gezeigt werden, dass sich die Inhalte je nach Kommunikationskanal unterscheiden: Neue Infektionszahlen werden schnell über Social Media verbreitet, die wirtschaftlichen Folgen der Pandemie eher via Pressemitteilungen. Mittels Topic Modeling kann außerdem gezeigt werden, dass die Landesregierungen durchaus jeweils eigene thematische Schwerpunkte setzen. Über den gesamten Untersuchungszeitraum im Jahr 2020 hinweg zeigt sich, dass dabei einige Themen dem wellenförmigen Verlauf der Pandemie in Deutschland folgen, andere einen gegenteiligen Verlauf haben.

Purpose The health of parents prior to conception, a woman’s health during pregnancy and the infant’s environment across their first months and years collectively have profound effects on the child’s health across the lifespan. Since there are very few cohort studies in early pregnancy, gaps remain in our understanding of the mechanisms underpinning these relationships, and how health may be optimised. ‘BABY1000’, a pilot prospective longitudinal birth cohort study, aims to (1) identify factors before and during pregnancy and early life that impact longer-term health and (2) assess the feasibility and acceptability of study design to inform future research.Participants Participants were based in Sydney, Australia. Women were recruited at preconception or 12 weeks’ gestation, and data were collected from them throughout pregnancy and postpartum, their children until the age of 2 years, and dietary information from a partner (if able) at the last study visit. The pilot aimed to recruit 250 women. However, recruitment ceased earlier than planned secondary to limitations from the COVID-19 pandemic and the final number of subjects was 225.Findings to date Biosamples, clinical measurements and sociodemographic/psychosocial measures were collected using validated tools and questionnaires. Data analysis and 24-month follow-up assessments for children are ongoing. Key early findings presented include participant demographics and dietary adequacy during pregnancy. The COVID-19 pandemic and associated public health and research restrictions affected recruitment of participants, follow-up assessments and data completeness.Future plans The BABY1000 study will provide further insight into the developmental origins of health and disease and inform design and implementation of future cohort and intervention studies in the field. Since the BABY1000 pilot was conducted across the COVID-19 pandemic, it also provides unique insight into the early impacts of the pandemic on families, which may have effects on health across the lifespan.

Este ensaio é uma crítica à Sociologia contemporânea no Brasil. O ponto de partida é uma revisão de manuais do ensino médio que permite transparecer uma antítese oculta entre preocupações com a distribuição das riquezas e a desigualdade social versus o desinteresse pelas causas da produção econômica e do desenvolvimento. Sugerindo atenção a estes fatores, advogamos pela reinserção da Sociologia do desenvolvimento, passando por temas correspondentes, como o evolucionismo, o capital social, a economia e o método interdisciplinar como facilitador. Nessa perspectiva de justificação da renovação sociológica, correlacionamos tais preocupações com os pressupostos da Base Nacional Comum Curricular – BNCC, que é o documento norteador da Educação brasileira a partir de 2018.
