- Abstract
Purpose:Adults with T-cell lymphoblastic lymphoma (T-LBL) generally benefit from treatment with acute lymphoblastic leukemia (ALL)-like regimens, but approximately 40% will relapse after such treatment. We evaluated the value of CpG methylation in predicting relapse for adults with T-LBL treated with ALL-like regimens.Experimental Design:A total of 549 adults with T-LBL from 27 medical centers were included in the analysis. Using the Illumina Methylation 850K Beadchip, 44 relapse-related CpGs were identified from 49 T-LBL samples by two algorithms: least absolute shrinkage and selector operation (LASSO) and support vector machine–recursive feature elimination (SVM-RFE). We built a four-CpG classifier using LASSO Cox regression based on association between the methylation level of CpGs and relapse-free survival in the training cohort (n = 160). The four-CpG classifier was validated in the internal testing cohort (n = 68) and independent validation cohort (n = 321).Results:The four-CpG–based classifier discriminated patients with T-LBL at high risk of relapse in the training cohort from those at low risk (P < 0.001). This classifier also showed good predictive value in the internal testing cohort (P < 0.001) and the independent validation cohort (P < 0.001). A nomogram incorporating five independent prognostic factors including the CpG-based classifier, lactate dehydrogenase levels, Eastern Cooperative Oncology Group performance status, central nervous system involvement, and NOTCH1/FBXW7 status showed a significantly higher predictive accuracy than each single variable. Stratification into different subgroups by the nomogram helped identify the subset of patients who most benefited from more intensive chemotherapy and/or sequential hematopoietic stem cell transplantation.Conclusions:Our four-CpG–based classifier could predict disease relapse in patients with T-LBL, and could be used to guide treatment decision.

56. Verteporfin attenuates cardiac fibrosis after myocardial infarction by suppressing the YAP-Smad2/3 signaling pathway

- Abstract
Purpose Excessive cardiac fibrosis and remodeling occur after myocardial infarction. Yes-associated protein (YAP) is a major transcriptional co-activator of the Hippo pathway and an important regulator of cardiac fibrosis. Verteporfin is a pharmacological inhibitor of YAP that effectively inhibits fibrosis and inflammatory responses. Therefore, this study aimed to explore the effects of verteporfin on cardiac fibrosis after myocardial infarction (MI) and its possible mechanisms. Methods Wild-type C57BL/6J mice were subjected to MI by ligating their left anterior descending coronary artery (LAD) and treating them with verteporfin (50 mg/kg/48 h) or phosphate-buffered saline for 2 weeks. Echocardiography was performed to evaluate cardiac function after 2 weeks, and hematoxylin and eosin and Masson staining were performed to evaluate the degree of myocardial fibrosis and inflammatory response. The protein expression levels of the YAP-Smad2/3 pathway, inflammatory factors, and fibrosis markers in the heart and in vitro were determined using western blotting and immunofluorescence staining. Results Compared to the MI group, verteporfin treatment improved cardiac function and fibrosis in mice post-MI. Moreover, myocardial YAP and SMAD2/3 expression were reduced in verteporfin-treated animals. Hematoxylin and eosin staining and molecular examination showed inflammatory factor and cardiac fibrosis marker expression in the heart sections. Conclusion Verteporfin can attenuate cardiac fibrosis and inflammatory responses and improve cardiac function by suppressing the YAP-Smad2/3 signaling pathway post-MI.

In the present work, a multi-pin-to-plate pulsed gas-liquid hybrid discharge reactor coupled with graphene/CdS catalysts was evaluated to eliminate Bisphenol A (BPA) in wastewater. The optimization of series of process parameters including pulse peak voltage, air flow rate, electrode gap, catalyst dosage, initial BPA concentration, initial solution pH, and solution conductivity was performed in terms of BPA degradation performance. The experimental results demonstrated that BPA removal rate increased with increasing the discharge voltage and decreasing the initial BPA concentration or solution conductivity. Nevertheless, too high or too low air flow rate, electrode gap, catalyst dosage, and initial solution pH would lead to the decrease in BPA degradation. Moreover, optical emission spectroscopy (OES) was gained to understand the information of short-life reactive species formed from the pulsed gas-liquid hybrid discharge plasma (PHDP) system. The results indicated the existence of several highly oxidative free radicals like ·O and ·OH. Finally, the activation pathway of O3 on the catalyst surface was analyzed by density functional theory (DFT).

There are many emergency risks in the process of natural gas hydrate (NGH) drilling. In order to ensure the safe and efficient exploitation of NGH, it is urgent to establish an intelligent judgment method for the risks in the process of NGH drilling. In this paper, the response relationship between monitoring parameters and risk categories of NGH while drilling is established. Based on fuzzy analytic hierarchy process (FAHP), the comprehensive weights of 10 risk monitoring parameters are obtained, including gas production, wellbore instability, hydrate ice barrier, drill string fracture, sticking, bit balling, drilling tool piercement, gas seepage, seabed subsidence and seabed landslide. Besides, the comprehensive judgment weight matrix is constructed, and the reasonable fluctuation range of monitoring parameters is formed. Thus, the intelligent judgment method of NGH drilling risk is established. The intelligent judgment and alarm of NGH drilling risks can be realized quickly and accurately by this method, namely, it can monitor the risks in the process of operation and guarantee the construction safety of NGH drilling.

Three new compounds (1‒3), including a phenylpropanoid (1), a bistetrahydrofuran lignin (2), and a lignan dimer (3), and seven known lignans (4‒10), were isolated from an EtOH extract of the aerial parts of Triadica rotundifolia. The structures of the new compounds were elucidated by comprehensive spectroscopic analyses. New compounds 1−3 were evaluated for their antineuroinflammatory and antioxidant activities. Research on the antineuroinflammatory activity suggested that compounds 2 and 3 had significant inhibitory effects on NO production in LPS-induced BV-2 cells, with the IC50 values of 15.7 and 5.8 μM, respectively. Meanwhile, compounds 2 and 3 exhibited better DPPH free radical scavenging activities with EC50 values of 2.0 and 4.3 μM, respectively, than the positive control vitamin C (EC50 = 14.3 μM).

Purpose The risk psychological distress (PD) confers on mortality due to specific chronic diseases compared to suicide is unclear. Using the National Health Interview Survey (NHIS), we investigated the association between PD levels and risk of all-cause and chronic disease-specific mortality and compared the contribution of chronic disease-related mortality to that of suicide. Methods Data from 195, 531 adults, who participated in the NHIS between 1997 and 2004, were linked to the National Death Index records through to 2006. Multivariable Cox regression models were used to estimate hazard ratios (HRs) and their 95% confidence intervals (CIs) across four levels of PD, measured using the Kessler-6 scale. Outcomes included all-cause mortality, and mortality due to all CVDs and subtypes, all cancers and subtypes, diabetes mellitus, alcoholic liver disease and suicide. Results During a mean follow-up time of 5.9 years, 7665 deaths occurred. We found a dose–response association between levels of PD and all-cause mortality, with the adjusted HRs (95% CI) elevated for all levels of PD, when compared to asymptomatic levels: subclinical 1.10 (1.03–1.16), symptomatic 1.36 (1.26–1.46) and highly symptomatic 1.57 (1.37–1.81). A similar association was found for all CVDs and certain CVD subtypes, but not for cancers, cerebrovascular diseases diabetes mellitus. Excess mortality attributable to suicide and alcoholic liver disease was evident among those with levels of PD only. Conclusion PD symptoms, of all levels, were associated with an increased risk of all-cause and CVD-specific mortality while higher PD only was associated with suicide. These findings emphasise the need for lifestyle interventions targeted towards improving physical health disparities among those with PD. Background & Aims: Despite the increasing prevalence of metabolic-associated fatty liver disease (MAFLD) worldwide, the ethnic differences between Han Chinese and Caucasian inclinical features remainunknown. Methods: This is a cross-sectional study comparing Han Chinese MAFLD patients from 2 University-affiliated Medical Centers of fatty liver in south China from January 2015 to January 2022 and Caucasian MAFLD patients from UK Biobank database. Liver fat content was defined using the magnetic resonance imaging proton density fat fraction (MRI-PDFF). Results: Han Chinese MAFLD patients(n=588) had higher proportions of moderate and severe grades of steatosis than that of Caucasian MAFLD patients (n=829) (25.7% vs 16.4%, PConclusions: Compared with Caucasians, Han Chinese MAFLD patients with higher LFC tended to suffer from higher risk of liver injury.

Five new polychlorinated bibenzyls (1–5) along with 3 known compounds (6–8) were isolated from the stems and leaves of Rhododendronminutiflorum. The chemical structures of all the isolates were determined by spectroscopic methods, and compounds 1 and 2 were further verified by single-crystal X-ray diffraction analyses. Compounds 1–5 were halogenated compounds which bear three to five chlorine atoms in their chemical structures. Biologically, compounds 2, 5 and 6 showed varying degrees of toxicity toward the Asian citrus psyllid (Diaphorinacitri) with LD50 values 27.15, 17.02 and 16.20 mg/L, respectively. These values were comparable to the positive control matrine (LD50 = 11.86 mg/L), which were calculated using observations on day 6. Meanwhile, compound 4 had α-glucosidase inhibitory activity with IC50 value of 17.87 ± 0.74 μM. Graphical Abstract

Three new tyramine-type alkamides (1-3), three new natural products (4-6), five new N-acylated/formylated aporphine alkamides with different ratios of rotational isomers (7-11), and 20 known alkamides (12-31) were isolated from an EtOH extract of the stems and leaves of Piper puberulum. The absolute configurations of compounds 7, 8, and 10 were determined by single-crystal X-ray diffraction analysis. In the biological activity assay, compounds 3, 5, and 10-23 displayed inhibitory effects against lipopolysaccharide-induced NO release in BV-2 microglial cells, exhibiting IC50 values of 0.93-45 μM.

The structures of the enantiomers were elucidated by comprehensive spectroscopic analyses. Their absolute configurations were assigned by single-crystal X-ray diffraction analysis as well as comparison of the experimental and calculated ECD spectra. The antineuroinflammatory activity research revealed that compounds 2a, 2b, 3a, 4a, and 5b could moderately inhibit NO production in LPS-induced BV-2 microglial cells with IC50 values ranging from 13.42 to 48.34 μM.

Two pairs of cycloneolignane enantiomers, piperhancins A and B (1 and 2, respectively), along with two enantiomeric pairs of biosynthetic related neolignanes, hancinone C (3) and piperhancin C (4), were isolated from the stems of Piper hancei. Compound 1 is an unprecedented 1',2:1,2'-dicyclo-8,3'-neolignane. Their structures and absolute configurations were elucidated by spectroscopic analyses, X-ray diffraction, and electronic circular dichroism calculations. Among all of the isolates, compounds (+)-1, (-)-1, (+)-2, (-)-2, and (+)-3 could significantly inhibit the production of nitric oxide secreted by lipopolysaccharide (LPS)-induced neuroinflammation in BV-2 microglial cells, with IC50 values of 1.1-26.3 μM. In addition, compound (-)-1 could decrease the mRNA levels of iNOS, IL-6, and TNF-α induced by LPS in BV-2 microglial cells.

Nitrate (NO3-) is known to be actively involved in the processes of mineralization and heavy metal transformation; however, it is unclear whether and how it affects the bioavailability of antimony (Sb) in paddy soils and subsequent Sb accumulation in rice. Here, the effects of NO3- on Sb transformation in soil-rice system were investigated with pot experiments over the entire growth period. Results demonstrated that NO3- reduced Sb accumulation in brown rice by 15.6% compared to that in the control. After amendment with NO3-, the Sb content in rice plants increased initially and then gradually decreased (in roots by 46.1%). During the first 15 days, the soil pH increased, the oxidation of Sb(III) and sulfides was promoted, but the reduction of iron oxide minerals was inhibited, resulting in the release of adsorbed and organic-bound Sb from soil. The microbial arsenite-oxidizing marker gene aoxB played an important role in Sb(III) oxidation. From days 15 to 45, after NO3- was partially consumed, the soil pH decreased, and the reductive dissolution of Fe(III)-bearing minerals was enhanced; consequently, iron oxide-bound Sb was transformed into adsorbed and dissolved Sb species. After day 45, NO3- was completely reduced, Sb(V) was evidently reduced to Sb(III), and green rust was generated gradually. Thus, the available Sb decreased due to its enhanced affinity for iron oxides. Moreover, NO3- inhibited the reductive dissolution of iron minerals, which ultimately caused low Sb availability. Therefore, NO3- can chemically and biologically reduce the Sb availability in paddy soils and alleviate Sb accumulation in rice. This study provides a potential strategy for decreasing Sb accumulation in rice in the Sb-contaminated sites.

Our understanding of signaling pathways regulating the cell fate of human embryonic stem cells (hESCs) is limited. Calcineurin-NFAT signaling is associated with a wide range of biological processes and diseases. However, its role in controlling hESC fate remains unclear. Here, we report that calcineurin A gamma and the NFATc3/SRPX2 axis control the expression of lineage and epithelial-mesenchymal transition (EMT) markers in hESCs. Knockdown of PPP3CC, the gene encoding calcineurin A gamma, or NFATC3, downregulates certain markers both at the self-renewal state and during differentiation of hESCs. Furthermore, NFATc3 interacts with c-JUN and regulates the expression of SRPX2, the gene encoding a secreted glycoprotein known as a ligand of uPAR. We show that SRPX2 is a downstream target of NFATc3. Both SRPX2 and uPAR participate in controlling expression of lineage and EMT markers. Importantly, SRPX2 knockdown diminishes the upregulation of multiple lineage and EMT markers induced by co-overexpression of NFATc3 and c-JUN in hESCs. Together, this study uncovers a previously unknown role of calcineurin A gamma and the NFATc3/SRPX2 axis in modulating the fate determination of hESCs.

Copper selenide is widely considered to be a promising candidate for high-performance flexible thermoelectrics; however, most of the reported ZT values of copper selenides are unsatisfactory at a relatively low temperature range. Herein, we utilized some wet chemical methods to synthesize a series of copper selenides. XRD, SEM and TEM characterizations revealed that CuSe, Cu3Se2 and Cu2−xSe were prepared successfully and possessed different morphologies and sizes. Based on the analysis of their thermoelectric properties, Cu2−xSe exhibited the highest Seebeck coefficient and lowest thermal conductivity among the three samples owing to its unique crystal structure. After being sintered at 400 °C under N2 atmosphere, the electrical conductivity of Cu2−xSe enhanced considerable, resulting in a significant improvement of its ZT values from 0.096 to 0.458 at 30 to 150 °C. This result is remarkable for copper selenide-based thermoelectric materials at a relatively low temperature range, indicating its brilliant potential in the field of flexible thermoelectric devices.

Fast charge transfer and lithium-ion transport in the electrodes are necessary for high performance Li–S batteries. Herein, a N-doped carbon-coated intercalated-bentonite (Bent@C) with interlamellar ion path and 3D conductive network architecture is designed to improve the performance of Li–S batteries by expediting ion/electron transport in the cathode. The interlamellar ion pathways are constructed through inorganic/organic intercalation of bentonite. The 3D conductive networks consist of N-doped carbon, both in the interlayer and on the surface of the modified bentonite. Benefiting from the unique structure of the Bent@C, the S/Bent@C cathode exhibits a high initial capacity of 1,361 mA h g−1 at 0.2C and achieves a high reversible capacity of 618.1 m Ah g−1 at 2C after 500 cycles with a sulfur loading of 2 mg cm−2. Moreover, with a higher sulfur loading of 3.0 mg cm−2, the cathode still delivers a reversible capacity of 560.2 mA h g−1 at 0.1C after 100 cycles.

Microvascular invasion (MVI) is a key pathological factor that severely affects the postoperative prognosis of patients with hepatocellular carcinoma (HCC). However, no MVI classification schemes based on standardized gross sampling protocols of HCC are available at present. 119 HCC specimens were sampled at multiple sites (3-, 7-, and 13 points) for the optimum MVI detection rate. 16,144 resected HCCs were graded as M0, M1 or M2 by adopting three-tiered MVI grading (MVI-TTG) scheme based on the seven-point sampling protocol (SPSP). Survival analyses were performed on 2573 patients to explore the advantages of MVI-TTG. The MVI detection rate determined by SPSP was significantly higher than that determined by the 3-point sampling method (34.5% vs. 47.1%, p = 0.048), but was similar to that determined by the 13-point sampling method (47.1% vs. 51.3%, p = 0.517). Among 16,144 resected HCCs, the proportions of M0, M1 and M2 specimens according to SPSP were 53.4%, 26.2% and 20.4%, respectively. Background: Inhibition of cancer cell growth and low in vivo toxicity are two important criteria for the development of anti-cancer drugs. Curcumin is a promising candidate for developing novel anti-cancer drug analogs. The research group designed the 3,5-bis-(3,4,5- trimethoxybenzylidene)-1-methyl-piperidin-4-one analog of curcumin that significantly inhibited the growth of esophageal cancer cells in vivo. In this study, 81 curcumin analogs were synthesized, analyzed both in vitro and in vivo, and their structure activity relationships (SARs) were determined. Methods: Based on the parent structure of curcumin, 81 analogs of 3,5-bis(substitutedbenzylidene)- piperidin-4-one compounds were designed and synthesized. Their anti-cancer activity in the human cancer cell lines was evaluated using the MTT assay, and in vivo toxicity was evaluated in mice. The SARs of selected compounds were analyzed. Results and Discussion: Among the designed curcumin analogs, 61 compounds exerted anti-cancer effects higher than the parent compound in vitro; 23 compounds inhibited cell growth in the human cancer cell line at low concentrations (IC50 values below 1 μM). The acute toxicity of curcumin analogs was tested in mice; 13 compounds were selected, which did not show any obvious toxicity at doses as high as 25.0 mg/kg. The SARs of these shortlisted curcumin analogs were determined. Conclusion: Twenty-three curcumin analogs exhibiting promising in vitro anti-cancer activity and low in vivo toxicity were designed. SAR analysis indicated the optimal functional groups in the molecule required for anti-cancer activity. This study not only suggested a useful strategy to design curcumin analogs for the development of anti-cancer drugs, but also revealed a group of curcumin analogs which could be further explored.

Motivated by novel graphyne-like carbon nanostructure C68-GY, spin-polarized DFT calculations with dispersion-correction were performed to investigate the hydrogen adsorption capacity of Li decorated C68-GY nanosheet. The binding energy between Li and C68-GY was larger than the cohesive energy of bulk metal, indicating Li atoms would prefer to separately attached on C68-GY. The ab initio molecular dynamics simulation has been performed to confirm the stability of Li/C complex. When five Li atoms decorated on C68-GY, 14H2 molecules were captured. The maximum hydrogen storage density was 8.04 wt% with an average hydrogen adsorption energy of −0.227 eV per H2. The positively charged Li atoms aroused electrostatic field and induced the polarization of H2. It was notable to observe strong hybridization between the main peak of H-1s orbitals with Li below Fermi level, which was responsible for the enhancement of hydrogen binding energy, indicating its potential application on hydrogen storage.

Soft climbing robots have attracted much attention of researchers for their potential applications on the wall or inside the tube. However, making a soft robot climb on the outer surface of a rod or tube by agile and efficient motion has long been a challenge. Inspired by the winding climbing locomotion of arboreal snakes, a tethered pneumatic-actuated winding-styled soft rod-climbing robot that consists of two winding actuators and a telescopic actuator is proposed in this work. Based on constant curvature assumption, we develop a theoretical model to analyze the linear and bending motion of the actuators. We demonstrate that our robot can perform climbing locomotion similar to snakes, including turning around a corner along a rod, climbing a vertical rod with a maximum speed of 30.85 mm/s (0.193 body length/s), and carrying a larger payload (weight, 500 g, more than 25 times its self-weight) than existing soft climbing robots do on a vertical surface. In addition, the experimental tests exhibit the potential applications of the robot in special environments such as high-voltage cables, nuclear power plants, and underwater sites.

Purpose: Adults with T-cell lymphoblastic lymphoma (T-LBL) generally benefit from treatment with acute lymphoblastic leukemia (ALL)-like regimens, but approximately 40% will relapse after such treatment. We evaluated the value of CpG methylation in predicting relapse for adults with T-LBL treated with ALL-like regimens. Experimental Design: A total of 549 adults with T-LBL from 27 medical centers were included in the analysis. Using the Illumina Methylation 850K Beadchip, 44 relapse-related CpGs were identified from 49 T-LBL samples by two algorithms: least absolute shrinkage and selector operation (LASSO) and support vector machine–recursive feature elimination (SVM-RFE). We built a four-CpG classifier using LASSO Cox regression based on association between the methylation level of CpGs and relapse-free survival in the training cohort (n = 160). The four-CpG classifier was validated in the internal testing cohort (n = 68) and independent validation cohort (n = 321). Results: The four-CpG–based classifier discriminated patients with T-LBL at high risk of relapse in the training cohort from those at low risk (P < 0.001). This classifier also showed good predictive value in the internal testing cohort (P < 0.001) and the independent validation cohort (P < 0.001). A nomogram incorporating five independent prognostic factors including the CpG-based classifier, lactate dehydrogenase levels, Eastern Cooperative Oncology Group performance status, central nervous system involvement, and NOTCH1/FBXW7 status showed a significantly higher predictive accuracy than each single variable. Stratification into different subgroups by the nomogram helped identify the subset of patients who most benefited from more intensive chemotherapy and/or sequential hematopoietic stem cell transplantation. Conclusions: Our four-CpG–based classifier could predict disease relapse in patients with T-LBL, and could be used to guide treatment decision.

Background The prognosis of patients with biliary atresia (BA) after Kasai portoenterostomy (KPE) varies, and precisely predicting the outcomes of KPE before surgery is still challenging. Methods A total of 158 patients who underwent KPE in our hospital were included in this study. The patients in the training cohort were recruited from January 2012 to October 2017 (n = 118), and then, those in the validation cohort were recruited from November 2017 to April 2019 (n = 40). Combined nomogram models were developed based on two-dimensional shear wave elastography (2D SWE) values and other biomarkers. The utility of the proposed models was evaluated by C-index. Results 2D SWE played a potentially important role in predicting native liver survival (NLS) of BA patients with a C-index of 0.69 (0.63 to 0.75) in the training cohort and 0.76 (0.67 to 0.85) in the validation cohort. The nomogram A based on 2D SWE values, age, gamma-glutamyl transferase (GGT) and aspartate aminotransferase-to-platelet ratio (APRI) had a better C-index in the training cohort [0.74 (0.68-0.80) vs. 0.66 (0.60-0.73), P = 0.017] and in the validation cohort [0.78 (0.70-0.86) vs. 0.60 (0.49-0.71), P = 0.002] than the nomogram B (without 2D SWE). Using risk score developed from nomogram A, we successfully predicted 88.0% (22/25) of patients in the training cohort and 75.0% (9/12) in the validation cohort to have survival time of less than 12 months after KPE. Conclusion The combined nomogram model based on 2D SWE values, age, GGT and APRI prior to KPE can effectively predict NLS in BA infants.

Erosion control materials with long-term effective, economical and environmentally friendly properties play a critical role in controlling erodible and unstable sandy slopes deformed under rainfall. In this study, a novel cellulose-based curing agent was developed and applied as a sustainable sand fixation materisal. The novel cellulose-based curing agent was a dual-polymer composite material (DPCM), which was prepared by mixing two kinds of organic polymer materials: carboxymethyl cellulose (CMC) and polyacrylamide (PAM). The microstructure and composition of DPCM were characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The adsorption kinetics of the curing agent on sand particles was discussed by fitting with pseudo-first-order and pseudo-second-order kinetic models. Water resistance and the compressive strength of the sand-fixing specimen at different curing ages were studied. Finally, the DPCM stabilization layer’s resistance to water erosion was investigated using a rainfall simulation test. The results demonstrated that the adsorption kinetic behavior of DPCM adsorbed on sand particles followed pseudo-second-order kinetics. Additionally, the interaction between the DPCM and sand particles was primarily hydrogen bonding, covalent bonds, and physical adsorption. The compressive stress was in the range from 53 to 915 kPa. Sandy slopes treated by DPCM showed excellent resistance to water erosion under a rainfall intensity of 120 mm/h.

Mast cells (MCs) are the major effector cells of allergic rhinitis (AR). The present study aimed to investigate the effects of C-C chemokine receptor type 3 (CCR3) on the proliferation, apoptosis, chemotaxis and activated degranulation of mouse MCs. Mouse bone marrow-derived MCs were cultured in vitro, purified and identified using toluidine blue staining and flow cytometry. Three different CCR3-short hairpin (shRNA) lentiviral vectors were constructed and transfected into MCs, and the mRNA and protein expression levels of CCR3 were assessed by reverse transcription-quantitative PCR and western blotting. Proliferation and apoptosis of the MCs were measured using Cell Counting kit-8 (CCK-8) assays and flow cytometry, respectively. MC chemotaxis was assessed by Transwell assay and quantified using flow cytometry. The activation of MC degranulation was examined using ELISAs. The results demonstrated that MCs were appropriately isolated, and identified that CCR3-shRNA2 presented the higher knockdown effect among the three shRNAs tested. Following 96 h of transfection, the results of CCK-8 and flow cytometry assays demonstrated that CCR3-shRNA2 inhibited MC proliferation and promoted MC apoptosis. The results from the Transwell assay indicated that CCR3-shRNA2 restrained MC chemotaxis, whereas ELISA results demonstrated that CCR3-shRNA2 suppressed MC degranulation. In conclusion, CCR3-shRNA2 effectively downregulated CCR3 mRNA and protein expression levels in mouse MCs. In addition, CCR3-shRNA2 promoted MC apoptosis and suppressed the proliferation, chemotaxis and degranulation of mouse MCs, suggesting that CCR3-shRNA2 may serve as a therapeutic tool for the treatment of allergic rhinitis.

For electrically conductive adhesives (ECAs), high electrical conductivity generally conflicts with excellent mechanical properties because electrical conductivity increases while desirable mechanical properties decrease with the increase in loading of conductive fillers or removal of lubricants on the silver filler surface. In this work, a method was developed to improve both the electrical conductivity and mechanical properties of the ECAs by introducing double bonds. Itaconic acid (IA) was used to replace the lubricant on the silver flake (Ag-F) surface, and acrylic acid (AA) was used as part of the ECA resin matrix. IA can replace the lubricant on the Ag-F surface, similar to other short-chain dibasic acids, to improve the ECA electrical conductivity. Furthermore, IA can be polymerized with AA to form covalent bonds between the silver flakes and the resin matrix, enhancing the mechanical properties of ECA. Compared with ECA filled with commercial silver flakes, the electrical conductivity of ECA filled with IA-treated silver flakes increased by ~ 21%, and the lap shear strength increased by ~ 29%.

Tyrosinase plays important roles in many different disease related processes, and the development of its inhibitors is particularly important in biotechnology. In this study, thirty-nine 3-/4-alkoxyphenylethylidenethiosemicarbazides were synthesized as novel tyrosinase inhibitors based on structure-based molecular design. Our experimental results demonstrated that thirty-one of them possess remarkable tyrosinase inhibitory activities with IC50 value below 1 µM, and 5a, 6e, 6g and 6t did not display any toxicity to 293T cell line at the concentration of 1000 µmol/L. According to the inhibitory activities, several compounds were selected for detail investigation on the structure-activity relationships (SARs), mechanisms of enzyme inhibition, inhibitory kinetics and cytotoxicity. In particular, the interaction between the selected inhibitors and the active center of tyrosinase was considered and discussed in detail based on their structural characteristics. Taken together, the results presented here demonstrated that the newly designed compounds are promising candidates for the treatment of tyrosinase-related disorders and further development of them may have significant contribution in biomedical science.

The hydrogen storage capacity of yttrium decorated graphyne nanotubes is calculated using spin polarized DFT method. The stabilities, electronic properties and the structures of Y attachment on graphyne tube are investigated. It is revealed that Y can be separately adsorbed on graphyne tube with the binding energy of 6.76 eV and the clustering of metal atoms is hindered. The geometry optimization shows that Y atoms decorated graphyne tube can capture 42H2 molecules through Dewar-Kubas like interaction and the polarization under the electrostatic potential formed by Y and graphyne tubes. The weight percent capacity is 5.71 wt%, with an average hydrogen adsorption energy of −0.153 eV per H2, indicating its potential application on hydrogen storage candidates.

We aimed to establish a discriminative gene-expression-based classifier to predict survival outcomes of T-cell lymphoblastic lymphoma (T-LBL) patients. Human embryonic stem cells (hESCs) can self-renew infinitely and differentiate into the cell types of all lineages of our body, holding great promise for investigating early human embryo development and providing functional cells for disease treatment. For the full application of hESCs, it is necessary to elucidate how hESCs maintain their identity. Recent studies have shown that glycolysis and mitochondrial respiration are linked to pluripotency states. However, the function of mitochondrial respiration in hESCs has not been fully understood. Herein, we report that the adenosine triphosphate (ATP) production rate is comparable between mitochondrial respiration and glycolysis, suggesting an important contribution of mitochondrial respiration to ATP production in conventionally cultured hESCs. To investigate the function of mitochondrial respiration, we silence OGDH expression in hESCs by the inducible CRISPRi method, and find that OGDH knockdown (KD) results in disrupted TCA (tricarboxylic acid) cycle, and diminished mitochondrial respiration activity and total ATP level. Moreover, OGDH KD leads to hESC death and aberrant transcriptional program. Interestingly, blockage of the electron transport chain (ETC) by small molecule inhibitors gives rise to the phenotype similar to that observed in OGDH deficient hESCs. Therefore, genetic and pharmacological perturbations of the mitochondrial respiration impair identity of hESCs. Collectively, our study highlights the pivotal role of the mitochondrial respiration activity for the stemness maintenance of primed hESCs, and unveils OGDH as a key regulator for the proper production of ATP and TCA cycle metabolites in primed hESCs.
