Since antiquity bitumen had been used as a construction material. Nowadays, most of the huge global production of bitumen each year, are functional to the roads paving industry where they are employed as binders for mineral aggregates to produce asphalt mixes. In the paving industry, a suitable bitumen should be fluid enough at to be pumpable and workable for a uniform covering of the mineral aggregates upon amalgamation. Furthermore, once the asphalt has been laid to build the roads, bitumen has to become sufficiently rigid at the highest pavement temperature to oppose rutting, depending on local climate conditions. Conversely, it must stay flexible enough at the lowest pavement temperature to resist cracking. For these purposes, additives such as polymers, acids, etc., are used to calibrate the operative range of bitumen. In addition asphalt industry is interested to reduce the costs of production, the environmental impact of the production and the safety condition for the paving workers. Bitumen is currently modeled as a dispersion of colloidal particles of asphaltenes, surrounded by a layer of stabilizing polar resins in a continuous oil phase (maltene). Although one can write a very simple definition of bitumen, its chemical composition is very complex and still not completely known. However the characterization of the bituminous materials for its convenient application, is still made by empirical standardized tests. This research project aimed to the deeper understanding of the behavior of bituminous systems, in order to correlate the macroscopic properties to the microstructure of the aggregates that constitute the bitumen colloidal network. We explored the possibility of taking advantage from chemical-physics techniques such as NMR, Rheology and AFM. In particular we have investigated the effect and overall the mechanism through which some chemical additives, already in use by paving companies, explicate their action to modulate the bitumen performances. Several samples different in nature and differently modified were analyzed. The rheological analysis, performed by the measurements under kinematic and dynamic control, helped to determine the material properties related to the structure of the system. The parameters thus obtained, being independent of the measurement conditions may be correlated with the microstructure of the sample investigated by the other techniques we used. As for NMR we exploited the spin-spin relaxation time measurement firstly to find the soften point of bituminous materials whether modified or not. As a novel approach to the understanding of the colloidal nature of the bitumen, the Inverse Laplace Transform (ILT) of the NMR spin-echo decay (T2) was applied. The ILT was used to draw the map of the macro-aggregates inside bitumen at different temperatures providing indication on the nature of the interaction between additives and the colloidal network. The efficiency of the ILT method was proved by atomic force microscopy images. As matter of fact collecting the AFM analysis, the ILT and the rheological results, we were able to describe the correlation between the aggregates at supra-molecular level inside fresh and doped bitumen. The removal of pollutants as well as the recovery of high added value molecules with low molecular weight is a current challenge in the wastewater treatment. Membrane processes can be considered as a viable option to solve these issues. In separations based membranes, high rejection of organic matter and water high permeability are two very important aspects that must be optimised. In general, the increase of the former comes at the expense of the latter because high rejection is cause of membrane fouling which in turns reduces dramatically the water permeability. Thus, membrane fouling constitutes one of the major limitations in membrane separation processes. The design of innovative materials which show high water permeability and at same time molecular rejection but with a low propensity to fouling is of fundamental relevance in this area. Mixed-matrix composite membranes where nanostructures such as Carbon Nano Tubes (CNTs) or Nano particles (NPs) are added, are receiving a huge focus since the properties of these nanostructures can confer an increase of the membrane efficiency in terms of permeability, selectivity, resistance and finally antifouling. The choice of the right nanomaterial can be highly accelerated by computational modelling. Thus, in this thesis an ab-initio modelling study in the frame of the Density Functional Theory (DFT) is carried out in order to investigate the structure-property relationships of nanostructures like CNTs and polyoxometalates NPs (POM). These nanostructures are used in the preparation of composite membranes in the frame of BioNexGen project [1] with the aim to propose novel membranes to be used in bioreactor for industrial waste water treatment. In particular, the rejection of CNTs towards organic solutes with low molecular weight coming from industrial waste water such as cosmetic, textile and oil olive was analysed in conjunction to the optimization of water permeability in CNTs. Also, the adsorption of POMs on polymeric membrane surface was analysed in order to understand if an efficient antifouling property can be imparted to the membranes by adding these NPs which possess excellent antibacterial and oxidants properties. While nanotube permeability has been extensively studied in both experimental and modelling works resulting in orders of magnitude water flow enhancements for tube diameters in the reverse osmosis and nanofiltration range [2], less work has been done on CNT selectivity of small organic solutes. Precise control of the CNTs synthesis allows for fine-tuning of the outer and inner diameters in the case of single- and multi-wall carbon nanotubes (SWNTs and MWNTs, respectively), offering the possibility of controlling their selectivity toward small solutes. In this thesis a modelling study was, therefore, addressed to define the optimal CNT internal diameter that should be used in order to achieve a total rejection of several target compounds very difficult to separate. Three novel algorithms [3, 4] were implemented during the thesis for reach the aforementioned objective. These algorithms do not make use of any adjustable parameters (i.e. fitting parameters) being based on geometry optimizations carried out in the frame of high level of quantum approach (DFT) and topological analysis of the considered systems. In addition, the CNTs geometrical characteristics should be optimized in order to get the best compromise between permeability and rejection. The functionalizing of the nanotubes tip with specific functional groups capable of hindering the passage of solutes while allowing water molecules to flow has been suggested as a solution to this problem [5, 6]. Thus, a theoretical study of CNT-composite membranes was carried out in this work with the aim of optimizing the CNTs tip, through their functionalization, to maximise both water permeability and solutes selectivity. This study is the basis of the third implemented algorithm [7]. Concerning the membrane fouling, the analysis of the adsorption of antibacterial anionic NPs (i.e. POM) on the surface of polymeric membranes was carried out in this thesis in order to achieve an optimal adsorption of these NPs. Noncovalent interactions between POM and commercial surfactants, used in polymer membrane preparations, were evaluated. Anionic POMs are in fact promising antibacterial agents [8], therefore, an efficient exchange with the bromide ions, used to counterbalance the positive charge of the commercial surfactants, would allow to increase the efficiency of the membrane by reducing the organic and bio fouling. In order to achieve this result, besides the aforementioned POM-surfactants noncovalent interactions the electronic hydration energy of POM and bromide anions was also evaluated in order to predict the probable exchange of these anions on the polymer surface. The conclusions of this thesis are achieved by using only ab-initio methods thus the results can be considered quite generals and homogeneous, free from empirical or fitting parameters. A validation of the theoretically prediction is provided concerning the POM-bromide exchange by means of an ad hoc designed experiment. In the past decade, plasmonic nanoparticles (NPs) have gained a lot of interest due to their exceptional and fascinating properties which have been accomplishing vital role in emerging science and technology towards multifunctional applications. The extensive current research efforts in nanoplasmonics trigger towards various opto-electronic and medical applications such as invisibility, perfect lens, increasing the efficiency of solar cells, designing and extra-sensitive single-particle detection of biomolecular recognition and in particular optical metamaterials. The negative real part and the low value of the imaginary part of dielectric permittivity are crucial for applications of nanoparticles as subunits of optical metamaterials. However, the performance of plasmonic nanostructures is significantly limited by the intrinsic and strong energy dissipation in metals, especially in the visible range. In fact, regardless of the challenges to synthesize plasmonic nanostructures, the path to use them as building blocks of optical metamaterial is crossing through the finding a solution to mitigate their optical losses. In this research thesis, it is demonstrated experimentally that the incorporation of gain material such as organic dye molecules and quantum dots in close proximity of enhanced local fields of various properly designed plasmonic systems makes it possible to induce resonant energy transfer processes from gain units to plasmonic nanoparticles, to accompanish loss compensation in optical metamaterials. Steady-state experiments and time resolved spectroscopy along with modification of Rayleigh scattering and optical transmission of a probe beam as a function of impinging energy are crucial evidences of mitigation of absorptive losses in different gain doped plasmonic systems The strategy that has been followed here towards mitigation of absorptive losses in optical metamaterials acts at different spatial scales from nano to macro (see Figure 1). The systems at nano-scale (10-100 nm) are based on dispersion of NPs, in particular, gain assisted (nanoparticle-dye dispersion), gain-functionalized core-shell gold NPs (encapsulated dye molecules into the shell) and dye grafted gold core multimeric nanostructures. The study of such nano-composites allows to recognize experimentally how the parameters such as the geometry and size of the metal nanostructures, inter-particle distance, overlap between emission spectrum of gain material and plasmon band of metal NPs, concentration and quantum yield of donor molecules are playing an important role to create more efficient nonradiative RET processes from donor molecules to acceptors. Figure 1 The followed spatial stages on this research study ranged from (a) nano-scale and (b) mesco scale towards (c) macro scale. The obtained results in nano-scale generate further motivations to move forward to study meso-scale (100-900 nm) plasmonic systems which include both dispersion (nanoshell composites) and bulk (periodic layers of gain materials and lossy metal elements) systems. The nanoshells which are consisted of dye doped dielectric core coated gold shell dispersed in ethanol solution are designed with an optimized ratio of core diameter and metallic shell thickness. The time resolved fluorescence spectroscopy results along with pump-probe experiments on nanoshells are convincing evidences for optical loss mitigation. Finally in third stage, the optical properties of gain-plasmon composites dispersed in PDMS host matrix as an example for bulk samples at the macroscopic scale (1 μm and beyond) have been investigated. The achieved results on this stage can help to design and fabricate such plasmonic structures that lead from fundamental physics towards practical applications. In this regard, the first four chapters provide the background concerning the main elements of this research work. The first chapter contains an introduction to the metamaterials. Second chapter describes the optical properties of plasmonic nanostructures. In third chapter, gain materials and the optical processes beyond these materials have been investigated. The fourth chapter deals with the optical properties of hybrid systems consisted of active materials and nano-plasmonic elements. After providing a brief introduction regarding the applied setups and instruments in this research study in chapter five, the last three chapters represent the acquired experimental results in each mentioned spatial scale. In chapter six, the optical properties of nano-scaled gain-plasmon systems in solution including gain-assisted, gainfunctionalized and dye grafted multimeric samples are investigated. Chapter seven explores the optical characteristics of dispersion of nanoshell sample as an example of the study in mesoscale. Finally, the thesis is completed with the study of the optical features of macro-scaled bulk samples based on core–shell type quantum dots and gold NPs dispersed in PDMS, and a short conclusion of this research study. The study of the interaction between molecules, in particular biological molecules and liquid crystals (LC), has experienced a huge growth in the recent years because of the development in devices engineering applied not only in photonics but also in the biomedical eld. In order to design more e cient LC devices, it is rst necessary to understand the behavior and properties of newly-synthesized liquid crystals and to garner a more indepth understanding of currently-existing LCs in order to answer pending questions about them. The aim of this thesis work, is to better understand the interactions involved at the interface between liquid crystals and other materials, whatever is their nature, i.e. polymeric or biological. We started studying the e ect of di erent con ning surfaces on the alignment of a special class of lyotropic liquid crystals, called \chromonics", which, in addition of LC properties, are biocompatible. Di erently from the most common liquid crystals, i.e. thermotropic LC, the mesogens that constitute the chromonic LC phases are not amphiphilic, but they are \plan-like" aromatic compound. This class of molecules embraces not only dyes and drugs, but also DNA and its bases. Using the knowledge acquired with chromonic mesogen, we tried to understand a more complicate system, such as the phenomena involved at the biomolecules decorated-liquid crystals lms interfaces. More speci cally, it is possible to divide the work in two macro-parts. The rst part concerns the alignment of a chromonic molecule, \disodium cromoglycate" (DSCG). The study of chromonic LC behaviour in con ned geometries and its physical properties, could be a model for more complex biological assemblies. Hence, we demonstrated the role of alignment layer's surface energy in the alignment of nematic phase of DSCG, achieving both alignments and for the rst time, a stable-in-time homeotropic anchoring of this LC solution. With the knowledge acquired from DSCG, we were able to align also DNA bases liquid crystal solutions. In particular, guanosine monophosphate in pre-cholesteric and cholesteric liquid crystals phases were perfectly aligned homeotropically without means of external elds, as was done until now, and partially planar aligned. Moreover, we observed that if ionic and/or silver doped solutions are added to the LC guanosine phases, it is possible to control the pitch of the cholesteric phase, modifying the helix structure. Instead, varying the nature of the con ning surfaces, in such conditions, it is possible to obtain guanosine vesicles. Other studies have been carried out on new chromonic complexes, synthesized at Chemistry Department of UNICAL, with possible application as anticancer drugs. A complete characterization of these compounds were done (XRD, phase diagrams, etc) and also for these compounds, we developed a\route"to drive the alignment, particularly important for future application in biophotonic devices. The second part of the work is focused on LC based biosensors. From the biotechnological and biomedical applications point of view, the studies on interactions of proteins with lipids are an area of fundamental interest, due to enormous biological importance. In fact, studies on biosensor devices are tremendously increased in recent years, focusing the attention also on nding low cost raw materials with high e ciency: liquid crystals, thanks to their high sensitivity to the external conditions, represent the best candidate. It has been demonstrated that aqueous interface of LC has an instantaneous response when exposed to phospholipids. This is a good base to study the interaction between biomolecules using LC as probe. Starting from the results found in literature, we studied the e ect of phospholipids on protein decoratede-liquid crystal interfaces by means of optical microscopy and FT-IR measurements. The rst technique allowed us to observe the response of decorated LC lm when exposed to phospholipids vesicles, while the second, gave us insight on conformational changes involved in secondary structure of the protein in function of the time of interaction between protein and LC, and the pH of the surrounding environment. The results obtained show a new methods to report speci c binding of vesicles on protein decorated interfaces. Questo lavoro di ricerca è dedicato allo sviluppo di nuovi materiali molecolari basati su complessi ciclometallati di Pd(II) e di Ir(III) per la foto/elettro conversione. Negli ultimi quindici anni, al fine di ottenere dispositivi più flessibili, più leggeri e più facilmente processabili, sono stati studiati principalmente materiali a base organica. Tali materiali presentano però, proprio a causa della loro natura organica, molteplici svantaggi, tra i quali scarsa qualità delle interfacce formate, limitata stabilità chimico-fisica in fase di funzionamento del dispositivo, difficile trasporto di carica, scarso assorbimento nel visibile e basse efficienze di luminescenza. Al fine di migliorare le prestazioni finali dei dispositivi elettro-ottici, è stata introdotta una nuova classe di materiali che utilizza come specie attive i composti organometallici. Nell’ambito di questo lavoro di tesi, sono stati sintetizzati nuovi complessi organometallici di Pd(II) e Ir(III) e ne sono state studiate le proprietà chimico-fisiche alla base dello sviluppo di materiali efficienti per la foto/elettro conversione. La prima parte del lavoro di tesi ha riguardato la preparazione di complessi per la conversione dell’energia solare. In particolare sono stati sintetizzati e caratterizzati nuovi complessi fotoconduttori ciclopalladati di Rosso Nilo, contenenti basi di Schiff opportunamente funzionalizzate come leganti ancillari. Inizialmente si è scelto di utilizzare il colorante Rosso Nilo come legante ciclometallante per le sue ottime proprietà di assorbitore di luce visibile. Il suo utilizzo ha inoltre comportato, nei complessi ottenuti, anche la separazione fisica su scala molecolare degli orbitali di frontiera HOMO e LUMO. In particolare i due orbitali risultano essere prevalentemente localizzati su due diversi frammenti molecolari. Tale separazione induce un efficiente processo di fotogenerazione che è alla base delle ottime proprietà di fotoconduzione osservate per questa classe di composti in un ampio intervallo di lunghezze d’onda. Per finalizzare l’utilizzo di complessi ciclopalladati di Rosso Nilo a specifiche applicazioni optoelettroniche, sono stati introdotti opportuni gruppi funzionali sia sul legante ciclometallante che sul legante ancillare In particolare i nuovi gruppi funzionali hanno permesso di : i) indurre proprietà mesomorfiche caratterizzate da un ampio grado di ordine in un grande intervallo di temperature e di ottenere così una nuova classe di metallomesogeni fotoconduttori; ii) aumentare la solubilità dei complessi preparati rendendo possibile il loro utilizzo unitamente al PC61BM, nella costruzione di celle solari ad eterogiunzione dispersa; iii) preparare, tramite il processo di elettropolimerizzazione, film sottili fotoconduttori di elevata qualità su elettrodi modificati; iv) poter ancorare i complessi sintetizzati a substrati di TiO2 e costruire celle solari di tipo Dye Sensitized (DSSCs). La seconda parte del lavoro di tesi, ha invece riguardato, la sintesi e la caratterizzazione fotofisica ed elettrochimica di complessi di Ir(III) per lo sviluppo di dispositivi elettroluminescenti. Sebbene esista in letteratura un elevato numero di esempi di complessi cationici di Ir(III) solamente pochi complessi anionici di Ir(III) sono stati descritti finora. Inoltre, i pochi esempi riportati, contengono leganti ancillari monodentati che rendono tali complessi chimicamente instabili all’interno di dispositivi elettroluminescenti. Al fine di ampliare la classe di composti anionici di Ir(III) esistenti e di migliorane la stabilità chimica, sono stati sintetizzati nuovi complessi anionici aventi leganti ancillari bidentati di tipo catecolato e orotato. E’ stata inoltre preparata e caratterizzata una nuova serie di “soft salt” di Ir(III) contenenti i nuovi complessi anionici ottenuti accoppiati ad opportuni complessi cationici di Ir(III) già noti in letteratura. Sono stati infine preparati e caratterizzati nuovi complessi luminescenti neutri di Ir(III) elettropolimerizzabili. Utilizzando tali complessi sono stati anche ottenuti film sottili elettrogenerati di alta qualità. Il peperoncino (Capsicum) è da tempo riconosciuto come un’eccellente fonte di composti fitochimici bioattivi quali i carotenoidi, i polifenoli e i capsaicinoidi, noti per esercitare vari effetti biologici positivi e di svolgere un ruolo importante nella prevenzione di molte malattie croniche. Inoltre, composizione e concentrazione dei principi attivi possono essere influenzata dai metodi di processazione e di conservazione. La relazione tra il contenuto in fitocomposti e il loro assorbimento è molto importante, giacché questi prodotti sono valutati in funzione del loro contributo alla dieta umana. Col termine bioaccessibilità si descrive il potenziale di una sostanza a essere assorbita, tuttavia, vi sono pochissime informazioni in letteratura riguardanti la biodisponibilità delle molecole presenti nei peperoncini. Gli attuali metodi per stimare la biodisponibilità includono studi sia a breve sia a lungo termine, in esseri umani e in modelli animali. Tuttavia, questi studi in vivo hanno un certo numero di limitazioni, come il costo elevato, il limitato numero di soggetti che possono essere sottoposti a studio e la difficoltà nell’interpretazione dei dati, vista l'influenza di fattori ospite-correlati quali lo stato di salute dell'intestino, lo stato nutrizionale e il genotipo del soggetto sottoposto a esame. L'uso di metodi in vitro fornisce un'alternativa agli studi umani e animali ed è un modello maggiormente riproducibile. Il primo obiettivo del presente lavoro di tesi è stato quello di valutare l'effetto dei diversi metodi di processazione (congelamento, cottura ed essiccazione) sul contenuto iniziale, sul contenuto micellare e sulla bioaccessibilità dei carotenoidi violaxantina, neoxantina, anteraxantina, luteina, capsantina, zeaxantina, β-criptoxantina e β-carotene. Peperoncini rossi, arancio e gialli hanno confermato di essere una buona fonte di carotenoidi, ma si è visto che il colore, molto più della specie, ne è indicativo del contenuto. I frutti gialli, infatti, sono molto ricchi di luteina, che è scarsamente presente, invece, nei peperoncini rossi, nei quali abbondano la capsantina, i carotenoidi con azione di pro-vitamina A (β-carotene e β-criptoxantina) e, in misura minore, la zeaxantina. Dopo digestione il contenuto di carotenoidi nelle micelle diminuisce notevolmente, particolarmente nei peperoncini essiccati al sole. E’ noto che il trasferimento dei carotenoidi nelle micelle è inversamente proporzionale all’idrofobicità del carotenoide. La lipofilia della β-carotene ha, infatti, influenzato negativamente la sua incorporazione nelle micelle, mentre capsantina, zeaxantina e luteina hanno mostrato una maggiore bioaccessibilità. Sono state riscontrate differenze nel contenuto e nella bioaccessibilità dei carotenoidi nei frutti sottoposti ai vari metodi di processazione. Generalmente la bollitura e il congelamento, diminuiscono il contenuto di carotenoidi. Nei frutti secchi, invece, la quantità di carotenoidi è stata generalmente superiore a quella riscontrato nei peperoncini freschi, bolliti e congelati. La quantità effettiva di carotenoidi incorporata nelle micelle è, però, un fattore predittivo più accurato del potenziale di assorbimento dei carotenoidi. Dallo studio si evince che la bioaccessibilità media di ogni carotenoide analizzato è aumentata dopo congelamento o bollitura, quindi, nonostante la diminuzione del contenuto dei carotenoidi dopo congelamento e cottura, il contenuto micellare non era significativamente più basso rispetto a quello dei peperoncini freschi e conseguentemente i valori di bioaccessibilità dei carotenoidi dei peperoncini processati sono risultati spesso simili o, più spesso, leggermente superiori a quelli non processati, equilibrando, in questo modo, la perdita di carotenoidi inizialmente provocata dalla processazione. Discorso diverso per i frutti essiccati, i quali, partendo da un contenuto nettamente più elevato nel frutto non digeritohanno evidenziato, nonostante la drastica diminuzione di carotenoidi dopo digestione, un ottimo contenuto micellare di pigmenti. Il secondo obiettivo della ricerca è stato quello di investigare la possibile influenza dei diversi processi tecnologici applicati ai peperoncini sulle proprietà antiossidanti e sul profilo fitochimico delle ventidue differenti varietà di peperoncino. Tramite i diversi approcci tecnologici adottati e grazie alle metodologie di studio applicate, è possibile concludere che i peperoncini secchi, seguiti da quelli freschi hanno il più altro contenuto di capsaicinoidi, polifenoli, flavonoidi e carotenoidi, dimostrando che le operazioni di congelamento e cottura influiscono negativamente sul contenuto di questi principi attivi. Dalla valutazione dell’attività antiossidante effettuata non sembra però esserci una diretta correlazione tra l’abilità antiossidante e la quantità di fitocomposti contenuti nei peperoncini. Le singole classi di composti sono debolmente associate con la capacità antiossidante rivelata, suggerendo che l'espressione dell'attività antiossidante degli estratti di peperoncino potrebbe essere piuttosto una conseguenza del sinergismo tra vari composti fenolici, o altre molecole qui non quantificate, e non semplicemente attribuibile a composti di una specifica classe chimica. The present work takes advance of nitrone chemistry flexibility in order to synthesize, on one hand, bisphosphonates containing N,O-carbocyclic nucleoside units with potential biological activity, starting from nitrones with functionalizable ester or methylen bisphosphonated groups. On the other hand, to synthesize suitable allyl cyclic nitrones undergoing 2-aza-Cope rearrangement in order to study the [3,3]-sigmatropic process that has been rarely detected until now with neutral molecules which are nitrones. The synthetic strategy that we used for the bisphosphonates compound involves the synthesis of suitable nitrones and subsequently 1,3-dipolar cycloaddition reaction between these substrates and various vinyl nucleobases that carries at formation of isoxazolidinyl nucleosides. We decided to synthesize them for their significant pharmacological properties that make them very appealing: they showed a considerable cytotoxic activity against several human cell lines and therefore they could be successfully employed as anticancer drugs. Furthermore, bisphosphonates can be considered as stable analogs of pyrophosphate, that is implied in the physiological regulation of bone calcification and resorption. Moreover, during the staying at the University of Zaragoza in Spain, a synthesis of suitable cyclic allyl nitrones was carried out. In general aza-Cope rearrangements have attracted great interest because of the ubiquitous presence of nitrogen-containing structures in natural and biological products as well as synthetic intermediates. NMR spectroscopy in weakly and highly orienting media is used as a route for dealing with orientational, positional, structural and conformational problems of a variety of small rigid and flexible organic molecules in solution. First, the very weak orientational order of a quasi-spherical molecule dissolved in a nematic phase is exploited for exploring the role of the different contributions to the observed dipolar coupling. In such a limit condition, a predominant effect of the non-rigid reorientationvibration coupling term emerges. Then, NMR data obtained from small rigid probes dissolved in smectic solvents are combined with a statistical thermodynamic density functional theory, in order to measure the positional order parameters of both solutes and solvent. The methodology gives good results when applied to a conventional smectic A liquid crystal and to the more delicate case of an interdigitated smectic Ad phase. The strategy is subsequently extended to the investigation of structure, order and conformational equilibrium of flexible bioactive or biomimetic molecules dissolved in various partially ordered NMR solvents. A first experimental and theoretical study is presented on the symmetric single-rotor molecule of biphenyl dissolved in a thermotropic liquid crystal. This test-case indicates molecular dynamics simulations are a promising tool for estimating a set of dipolar couplings of a solute in a thermotropic solvent, to be used as starting set of parameters in a standard operator-mediated NMR spectral analysis. Then, we report the conformational study of some single- and two-rotor nonsteroidal anti-inflammatory drugs, belonging to the families of salicylates and profens, dissolved in weakly orienting chiral nematic PBLG phases. A new pulse sequence, the Gradient Encoded heTeronuclear 1H-19F SElective ReFocusing NMR experiment (GET-SERF), is proposed here for the trivial edition of all 1H-19F couplings in one single NMR experiment, for a given fluorine atom. Starting from homo- and heteronuclear dipolar couplings, difficult to extract in thermotropic solvents because of a too complex spectral analysis, the torsional distributions of such molecules can be satisfactory described by the Additive Potential model combined with the Direct Probability Description of the torsional distribution in terms of Gaussian functions (AP-DPD approach). Finally, the conformational and orientational study of two stilbenoids displaying cooperative torsions is discussed in both a highly and weakly ordering liquid crystal phase. This comparative study allows to draw some conclusions on reliability, accuracy and accessibility of desired data in the two phases. Overall, this work proves NMR in liquid crystals is a flexible and meaningful tool for studying order, structure and conformation and it can greatly benefit from the availability of several aligning media inducing a different degree of order. Fruits and vegetables are widely recognized as an important source of beneficial compounds for the human nutrition. Citrus and juices are widely consumed in the world. Their health benefits have been recognized for a long time now since first cultivated in Asia. The market of oranges is growing constantly, because consumers have become more convenience-oriented and health conscious. Consequently, efforts by researchers and industrial organizations have been made to develop processes more efficient, putting emphasis in food quality and safety. In addition, with the increasing emphasis on water pollution control, industries are establishing programs that will permit disposal of their wastewater without damaging the environment. The citrus fruit industry is no exception. Citrus wastes, such as wet peels and whole rejected fruit, which a great amount of organic matter that can be associated with environmental and health problems due to water runoff and uncontrolled fermentation. Therefore, the development of optimized systems for the treatment of citrus wastes is a crucial factor in the rationalization of the traditional industrial cycle. Membrane operations in the last years have shown their potentialities in the rationalization of production systems. Their intrinsic characteristics of efficiency, operational simplicity and flexibility, relatively high selectivity and permeability for the transport of specific components, low energy requirements, good stability under a wide spectrum of operating conditions, environment compatibility, easy control and scale-up have been confirmed in a large variety of applications and operations in the food industry. Recent R&D efforts aim to convert the potential of wastes into profitable products creating new segments of production and offsetting the disposal costs. Orange peels and pulp contain several bioactive compounds, such as flavonoids and phenolic acids, recognized for their beneficial implications in human health due to their antioxidant activity and free radical scavenging ability. These semi-solid wastes can be pressed to obtain a liquid rich in soluble sugars, named press liquor, that can be concentrated up to citrus molasses grade or alternatively exploited as a natural source for the extraction of phenolic compounds which can be used as natural antioxidants mainly in pharmaceutical, cosmetic and foods industry. This PhD dissertation is based on the opportunity to develop a technical alternative to solve some problems related to the disposal of citrus wastes from an economic and environmental point of view, specifically in terms of recovery of attractive compounds present in orange press liquor. The general aim of the work was devoted to the recovery of phenolic compounds by using an integrated membrane process, in which microfiltration (MF) or ultrafiltration (UF), nanofiltration (NF) and osmotic distillation (OD) processes were involved. MF or UF processes were used for the clarification of the press liquor in order to separate macromolecules, such as pectins, and to obtain a clarified press liquor as permeate stream. In the second step the clarified press liquor obtained from the MF/UF process was treated by NF in order to obtain a retentate stream enriched in phenolic compounds with a low content of sugars. Finally, OD was used as final step to concentrate the NF retentate stream. In the first Chapter, a general overview of membrane operations, including methodologies to evaluate their performance, is reported. The industrial transformation of citrus fruits and the associated production of citrus wastes is reported in Chapter 2. In this Chapter the chemico-physical characterization of citrus press liquor used for the experimental activities performed on membrane laboratory plants is also included. Chapter 3 is focused on the experimental activities concerning the clarification of citrus press liquor by using MF or UF processes. In Chapter 4 the fractionation of clarified press liquor by using NF membranes is analysed and discussed. In recent years, particular interest has been addressed by researchers in the synthesis and study of silicates such as enstatite MgSiO3 and talc Mg3Si4O10(OH)2. The first one is useful for several technological applications such as substrates in electronics, high frequency insulators, thermal insulators in high temperatures applications, and as luminescent materials in laser technology. The latter, because of the low cost and good properties (i.e. resistant to heat and acids, hydrophobic, electrical insulating) is widely used in many different products such as ceramics, papers, cosmetics, foods, polymers and filler in composites. The usual presence of foreign ions (e.g., Mn, Ti, Ni, etc.) and their inconstant amounts in natural enstatite and talc hinder the use of these minerals as high-performance materials. For these reasons, in recent years pure and doped enstatite and talc have been grown and characterized in several different ways. Nevertheless, there are still various problems to be solved in order to obtain very high quality crystals and the desired changes in the physical and chemical properties of them when they are doped with metal elements. In this work, Zn-doped enstatite, Mn-doped enstatite, Zn-doped talc and Mn-doped talc have been grown and characterized with different techniques. The starting materials and the final products were characterized and studied by binocular microscope, powder crystal X-ray diffraction (XRPD), scanning electron microscopy with energy-dispersive spectrometry (SEM/EDS), single-crystal X-ray diffraction (XRD), micro-Raman (μ-R), cathodoluminescence (CL), differential scanning calorimetry, thermogravimetric analysis (DSC-TG) and Fourier transform infrared spectroscopy (FT-IR). Zn- and Mn-doped enstatite was successfully produced by slow-cooling flux growth method, using MoO3, V2O5, Li2CO3 as melting agent. Several starting mixtures, with different MnO or ZnO concentrations, were first held at 1350 °C, 1250 °C 1050 °C and 950 °C and then slowly cooled down to 700 °C or 600 °C with different cooling rate (3.75 °C/h, 2.1 °C/h, 1.8 °C/h 1.7 °C/h). Enstatite crystallizes in the orthorhombic and monoclinic systems as revealed by XRD and Raman spectra. Transparent Zn-doped enstatite ( max length of 3.5 mm) and reddish Mn-doped enstatite (max length of 8 mm) single crystals are euhedral in form, not homogeneous in width and inclusion free. Maximum content of Mn-dopant is 14.52 wt %, while the maximum amount of Zn-dopant is 10.49 wt%. Crystals grow under equilibrium conditions only when the dopant content is maintained at low value. When either Zn or Mn is totally substituted for Mg in the starting material, no enstatite is produced. The presence of the dopant in the enstatite structure causes a decrease in unit cell volume respect to the pure one and strongly affects the CL-signal and micro-Raman spectra. CL spectrum of Mn-doped enstatite contains a broad emission located at 677 nm and attributed to the 4T1g(G)→6A1g(S) transition of octahedral Mn2+ centres. The presence of Zn in enstatite induces very remarkable peak broadening by the mode at 133 cm-1 and 343 cm-1 in the Raman spectra; for these modes a strong component of metal ion displacement must be postulated. Raman spectra of Mn-enstatite show: i) a general decrease of Raman intensity due to the increase in surface reflection when the MnO dopant concentration increases; ii) a widening and a down shifting of the peak positions indicating changes in vibrational modes because of the increasing presence of MnO. Zn- and Mn-doped talc was successfully synthesized in hydrothermal conditions at temperatures of 300, 500 and 650 °C, under constant pressure of 2 kbar and reaction time of 160 hours. Talc morphology and content of dopant within the crystals show strong dependence on crystallization temperature. Talc exhibits a cabbage-like morphology, its classical hexagonal tabular morphology and fibrous morphology. The best temperature to obtain the highest abundance of Zn- and Mn-doped talc is 650 °C. A decrease in temperature from 650 to 300 °C: i) worsens the reactions and poorly crystallized Zn- and Mn-doped talc is obtained; ii) increases the content of zinc or manganese dopant. Talc only grows when Mg is not totally substituted by either zinc or manganese in the starting mixture. Zn-doped talc formation is increased by treating the starting mixture with H2O + HCl; conversely, the reactant H2O + CaCl2 inhibits the growth of talc. In order to increase doped talc yield, large amounts of aqueous solution is as crucial as high temperatures. The presence of varying amounts of metal elements replacing Mg in talc influences its temperature decomposition. Indeed, the thermal stability of Zn- and Mn-doped talc decreases with respect to pure one. Zn-dopant in talc mainly affected the hydroxyl stretching fundamental peak (3674 cm-1), splitting itself into as many as four peaks with respect to non-doped talc, which displayed only a sharp band. The splitting appears to be dependent on the degree of substitution of the magnesium in the octahedral layer and related to the electronegativity difference between Zn and Mg. Future studies will be carried out on these materials to have a better knowledge of other physical properties, useful in novel applications. Lo scopo del presente lavoro di tesi è la sintesi di materiali polimerici innovativi funzionalizzati con una porzione antiossidante legata alla catena laterale da utilizzare nella conservazione dei cibi, in particolar modo per prolungare la “shelf life” dei cibi della IV gamma cui appartengono frutta e verdura già lavata e tagliata, pronta all’uso. Generalmente questo genere di alimenti deperisce velocemente e ha una durata limitata a pochi giorni. L’ossidazione è una delle più importanti reazioni di degradazione dei cibi e poichè ne riduce la “shelf-life”, ne limita anche la conservazione con conseguente diminuzione delle vendite. L’active packaging prolunga la “shelf-life” dei cibi e allo stesso tempo ne migliora la sicurezza e le proprietà organolettiche. L’idea innovativa rispetto alle tecniche tradizionali è la preparazione di film bioattivi in cui la molecola antiossidante non è adsorbita sulla superficie del film con procedure di spraying, immersione o rivestimento, ma è legata covalentemente, tramite un linker, alla matrice polimerica. In questo studio è stata evidenziata la sintesi di un PET modificato innovato che reca una catena laterale funzionalizzata con una molecola antiossidante quale il ter-butilidrochinone (TBHQ) o l’idrossitirosolo (Tyr-OH). L’attività antiossidante dei monomeri di PET modificato funzionalizzati con il TBHQ e il Tyr-OH è stata testata tramite test del DPPH che ha confermato l’elevata capacità scavenger dei monomeri stessi. Un altro obiettivo del lavoro di ricerca è stata la caratterizzazione tramite spettrometria di massa di polimeri e copolimeri funzionalizzati. Questo studio è stato svolto in collaborazione con l’Università di Akron, Ohio, USA. Lo scopo del lavoro è stato sviluppare un protocollo di analisi di spettrometria di massa che fornisce informazioni riguardo la composizione dei poliesteri copolimeri, dei gruppi terminali, delle sequenze e della loro architettura. Più nello specifico, l’oggetto dello studio è stato il confronto tra il copolimero lineare poli(caprolattone)-poli(etilene glicole) (PCL-PEG) e l’omopolimero poli(caprolattone). Inoltre è stato condotto uno studio sull’energia di collisione necessaria alla frammentazione per il copolimero PCL-PEG, e gli omopolimeri PCL e PEG. Questo studio è stato condotto utilizzando spettrometri di massa MALDI Q/ToF e MALDI ToF/ToF/.Essendo questi polimeri ampiamente usati in moltissimi campi e per differenti applicazioni, in particolare applicazioni in campo biologico, è stato condotto uno studio di degradazione in mezzo acquoso per capire come e con quale velocità questi polimeri degradano e frammentano in presenza di acqua. Per effettuare questo studio i “frammenti” provenienti dalla degradazione sono stati separati per cromatografia liquida attraverso l’uso di uno strumento UPLC la cui uscita è accoppiata ad uno spettrometro di massa per poter facilmente individuare ed assegnare i vari frammenti. Astroparticle physics provide a fundamental tool to investigate the astrophysical structures at different scales. In this thesis the results are presented of three years of research focused on time variations on solar neutrino flux, galactic cosmic rays and fine-structure constant as observed from quasar spectra. These topics are nowadays crucial to investigate structures which otherwise result difficult to probe. In particular time variations in solar neutrinos can give a real time description of the dynamics which take place in the inner regions of the Sun, while the study of galactic cosmic rays through the observation of muons produced by the interaction of the formers with the atmosphere, may give us precious informations on the interaction between TeV particles and heliospheric magnetic fields. Finally the recent observations of variations in the fine-structure constant value from the analysis of quasar absorption spectra, offer new possibilities to probe physical laws at cosmological scales. The thesis is structured as follows. In Chapter One are discussed quasi-biennial oscillations observed on solar neutrino flux and a phenomenological model is proposed to interpret the observed time variations. In the scenario described by the model, the interaction between solar neutrinos and solar matter mediated by magnetogravity modes, is modulated by a background magnetic field varying in time on quasi-biennial time scales. In Chapter Two the 11-yr component recently observed in cosmic muon data from Gran Sasso experiments of MACRO, LVD and Borexino has been reconstructed through the EMD analysis technique. In Chapter Three the data of fine-structure constant variations as observed in the quasar absorption spectra have been analyzed to search for temporal coherent structures. The thesis is completed by the description in appendix of a new algorithm based on Monte Carlo methods to assign a confidence interval to the IMFs extracted with the EMD analysis, and the application of this method to study the proton and neutrino distributions used in the OPERA experiment for the neutrino velocity measurement. Another appendix is dedicated to detailed description of the effects induced on the magnetogravity spectrum by a background magnetic field with a low-pass bandwidth, thus generalizing the theory of magneto-gravity modes in presence of a time-dependent background magnetic field. The main objective of this study is active plasmonics. The work has been focussed on the design, characterization and theoretical interpretation of novel systems. Top-down and bottom-up, self-assembling, approaches have been utilized to realize devices where spherical gold nanoparticles have been periodically and randomly arranged. Two main paths have been followed to achieve this goal. In the first one, by utilizing a rigid periodic structure as a host platform for soft-matter (cholesteric liquid crystals) mixed with plasmonic nano-entities, was possible to obtain a chirally-organized tuneable plasmonic system. The tunability of the obtained device has been induced by applying temperature changes or external electric fields. In the second one, the surface of an elastomeric platform has been randomly covered by gold-nanoparticles. Controlled nano-chemistry processes have been successively applied to the nanoparticles (immobilized on the surface) to locally increase their size. The elastic properties of the template together with the increased size of particles have allowed a systematic study of the coupling between near-fields of the spherical nanostructures. Colloidal nano-chemistry technics have been utilized both to synthetize spherical gold nano-particles and to increase their sizes. Spectroscopic analysis has been used to analyse the response of obtained structures under electrical, thermodynamical and mechanical stimuli. SEM and TEM imaging have been exploited to study the morphology of devices, the shape of nano-structures and to measure their sizes. Negli ultimi anni le applicazioni dei nanotubi di carbonio nel settore microelettronico sono notevolmente aumentate date le proprieta’ uniche. In particolare la conducibilita’ dei nanotubi fa si che essi trovino interessanti utilizzi nel settore fotovoltaico. Questo lavoro si concentra sulla progettazione di una cella solare ibrida a base di nanotubi di carbonio e dye di Rutenio. Nella prima parte sono stati studiati diversi metodi di realizzazione dello starto omogeneo di nanotubi come starto conduttivo della cella. E’ stata proposta la molecola 1-Pyrenemethanol che contiene i gruppi pirenico e ossidrilico, per creare il contatto tra i nanotubi ed il dye. Nella seconda parte del lavoro lo starto di nanotubi e’ stato formato mediante elettroforesi per single-walled nanotubes (SWNTs) e metal-organic chemical vapor deposition (CVD) per multi-walled nanotubes (MWNTs). Entrambi i metodi hanno dato la possibilita’ di creare campioni con ampia area superficiale, dai quali sono state fabbricate le celle solari. Sono state studiate determinate le dipendenze tra i fattori dei metodi e le proprieta’ delle celle prodotte. The present thesis is focused on the development of novel nancomposite membranes, prepared by the incorporation of two-dimensional inorganic layered structures such as (i) smectite clays (synthetic and natural), (ii) graphene oxide (GO), and (iii) layered double hydroxides (LDHs) with different compositions into the polymer matrix of Nafion, for use as electrolytes in Proton Exchange Membrane fuel cells. The characteristics of the membranes were studied mainly, in terms of transport properties by NMR spectroscopy, in order to study the water dynamics inside the electrolyte membranes. For this purpose the Pulse-Field-Gradient Spin-Echo NMR (PFGSENMR) method was employed to obtain a direct measurement of water self-diffusion coefficients on the water-swelled membranes in a wide temperature range (25-140 °C). This technique together with the 1H-NMR spectral analysis and NMR spin-lattice relaxation times (T1) conducted under variable temperature. Furthermore, both pristine materials (fillers and Nafion) as well as the resulted nanocomposite membranes were characterized by a combination of X-ray diffraction, FTIR spectroscopy, thermal analysis (DTA/TGA), Raman spectroscopies and scanning electronic microscopy (SEM). In dentistry, a correct detection of caries severity is still a challenging descision-making task that crucially a ects the choice for the best treatment plan. The challenge is to nd both the most objective parameters to detect caries at di erent stages (from an early reversibile stage to a severe one) and the most reliable method(s) that should be used to distinguish these stages. Currently, methods used in clinics are visual inspection, aided with light probe and pick inspection tools, and radiography. The main issue rising by the use of these methods is that both of them are subjective, with possibility for intra- and inter-examiner variability. For this reason, radiography needs an extreme care of interpretation especially when assessing occlusal caries. Visual methods, instead, are a ected by confounding factors, such as stain or uorosis, a ecting the accurate assessment of early caries lesions. Radiography, moreover, should be performed with care considering that the emission of ionising radiation may cause malignant change in tissues, especially for young age patients and are counter-indicated during pregnancy. They are also inadequate for the detection of initial caries and to locate the lesions looking at the superimposition of the tooth along its buccal-lingual axis. The aim of this study was to overcome the limits of the current detection techniques, o ering a non-invasive, objective method for the detection of caries at any stage of the demineralisation process. The proposed method measures the near-infrared (NIR) re ectance response of the tooth at three speci c wavelengths. It is then possible to investigate properties of the sample at the surface and in depth and get an image that maps the lesions on the occlusal view of the sample when combining these wavelengths. Due to the properties of the NIR light, this method is non-invasive, non-contact and allows for detection both at the enamel and at the dentine level. The NIR method o ers objective supporting information to quantify and detect dental caries and is especially suitable for areas a ected by confounding factors, such as stain. The objective of the study was to design and implement a NIR multispetral imaging system, developing e cient image analysis algorithms. In order to prove this objective, an in vitro validation of the technique against gold standard histology was performed together with a comparison to other detection methods - International Caries Detection and Assessment System (ICDAS - clinical visual inspection), bre optic transillumination method (FOTI - visual inspection with light probe), radiography and Quantitative Light-induced Fluorescence method (QLF), used in clinics or in research. A total of 112 teeh, molars and premolars, with di erent lesion severities were used for this study. Histologcal sections were obtained to con rm the lesion severities and used as a gold standard to compare the sensitivity and speci cty among techniques. Visual inspection methods recorded the highest values of sensitivity (ICDAS: >99%, FOTI: 93%) and speci city to dental caries (FOTI: >99%, ICDAS: 90%). However, these methods could have been highly facilitated by the in-vitro viewing of the samples. Sensitivity to dental caries was higher for NIR (91%) than for QLF (88%) and radiography (63%) while speci city was higher for radiography (81%) than for NIR (73%) and QLF (63%). The results from this study suggest that the NIR method has the ability to detect dental caries when other methods fail, providing an alternative to assist in the decision-making process with the further advantage of removing the confounding e ect of stain. This method can enhance patient communication and o ers an objective and safe alternative to ionising radiation methods. Il presente lavoro di tesi relativo all‘attività di ricerca svolta durante il triennio di dottorato ha riguardato la messa a punto di metodi analitici per la determinazione di analiti in due distinte aree di interesse. La prima di ambito clinico ed ha riguardato la quantificazione in urina di metaboliti riconosciuti come marker in diagnostica clinica. In particolare ci si occupati della sarcosina come biomarker del tumore alla prostata, e di tre acidi: acido omovanillico (HVA), acido vanilmandelico (VMA) ed acido 5-idrossindoloacetico (5-HIAA) come marker urinari del neuroblastoma. Il secondo ambito di lavoro ha riguardato la quantificazione di inquinanti in matrici acquose, vale a dire carbammati ed acidi perfluoroalchilici. In particolare gli analiti, previa derivatizzazione con alchilcloroformiati (eccetto i carbammati), sono stati estratti dalle matrici acquose (acqua e urina) tramite la tecnica della microestrazione in fase solida (SPME) e successivamente analizzati mediante un gascromatografo con analizzatore di massa a triplo quadrupolo (GC-QqQ-MS). Le variabili significative della microestrazione in fase solida in ciascun metodo sono state ottimizzate tramite l‘approccio multivariato dell‘ ― Experimental Design‖. L‘utilizzo della tecnica SPME ha consentito di poter estrarre gli analiti direttamente dal campione da analizzare minimizzando i tempi di preparazione dello stesso e riducendo l‘uso di solventi organici, ottenendo metodi poco costosi e basso impatto ambientale. L‘utilizzo dello spettrometro di massa triplo quadrupolo, ha consentito di raggiungere livelli di sensibilità molto elevati e, nel contempo, di identificare gli analiti con maggiore sicurezza. In tutti i metodi sviluppati, sono stati ottenuti ottimi risultati in termini di linearità accuratezza e precisione. Anche i valori dei limiti di rilevabilità (LOD) e dei limiti di quantificazione (LOQ) ottenuti in ciascun metodo possono essere considerati soddisfacenti. Il presente lavoro di Tesi di Dottorato di Ricerca (Scuola di Dottorato in Scienza e Tecnica "Bernardino Telesio") dal titolo "Novel Organic Optoelectronic Materials" è stato svolto all'interno dei laboratori di "Organic Optoelectronic Materials" e "New Syntheses via Organometallic Catalysis" del Dipartimento di Chimica dell'Università della Calabria. Nuovi semiconduttori organici potenzialmente impiegabili in dispositivi optoelettronici quali OLEDs, celle solari, transitors ed elettrocromici sono state progettate, sintetizzate e caratterizzate. La prima parte del lavoro ha riguardato la sintesi e la caratterizzazione di derivati di triarilammine, molecole elettron-donatrici (anodiche), da impiegare in dispositivi elettrocromici ("smart windows") per l'attenuazione della radiazione solare nel vicino infrarosso (NIR). L'intensa ed estesa banda di assorbimento nel NIR prodotta delle specie mono-ossidate ha suggerito l'impiego di queste triarilammine in dispositivi "complementari" in cui vengono utilizzate insieme a molecole elettron-accetrici (catodiche) elettrocromiche nel visibile, per un'ampia modulazione dello spettro solare. Tali sistemi sono stati dispersi all'interno di matrici polimeriche al fine di realizzare dei gel elettrocromici le cui prestazioni sono state ampiamente studiate. La risposta elettrocromica del gel è stata inoltre provata in un dispositivo plastico, dimostrandone le potenziali applicazioni campo dell'elettronica flessibile. La seconda parte del lavoro invece, ha riguardato la sintesi e lo studio delle proprietà di cristalli liquidi semiconduttori di tipo "n" (elettron-accetori) da impiegare in celle solari organiche "bulk heterojunction". Tali molecole, appartenenti alla famiglia dei viologeni estesi, hanno mostrato interessantissime proprietà mesomorfiche fortemente influenzate dalla lunghezza delle catene alchiliche ( a 9, 10 e 11 atomi di carbonio) e dell'anione utilizzato, la bis(triflimmide). Le proprietà elettrochimiche di questi composti sono state investigate sia in soluzione che nelle mesofasi. Un'efficace elettrocromismo dovuta (i) al doppio strato elettrico creato all'elettrodo degli anioni presenti e (ii) all'elevata sovrapposizione degli orbitali di frontiera dell'esteso sistema π- coniugato è stato osservato nelle fasi colonnari e smettiche in cui le molecole si sono auto-assemblate. Infine, le proprietà elettrocromiche di questi composti sono state studiate all'interno di film plastici utilizzabili in dispositivi quali i displays. This PhD work concerns the theoretical photochemistry study of molecules with an interest on Photodynamic Therapy (PDT). PDT is a medical technique for the treatment of different tumor diseases, such as age related macular degeneration, psoriasis or bladder cancer. This technique is increasingly being required, in particular in cases when conventional methods, chemotherapy and radio therapy fail to be successful. It consists of the administration of photosensitizer (a drug) followed by light irradiation and requires the presence of molecular oxygen on tissue to be destructed. On molecular terms the action of the drug is explained by the excitation of the drug to a singlet state after light irradiation, followed by its conversion of to a triplet state. This triplet state, in more common cases, directly transfers its energy to molecular oxygen producing singlet oxygen. In other cases the photophysical parameters of the triplet state are such that it is allowed to participate in electron transfer reactions, where it becomes reduced and after its anion reduces oxygen forming radical oxygen species, that like singlet oxygen, is toxic to the cell, resulting in cellular death by apoptosis or necrosis. As chemists, our main interest is on the proposal of drugs with ideal photophysical and solution properties. In regards to the first aspect, a drug should have an intense absorption on the red part of the visible spectra, known as therapeutic window, where the body tissue has a better penetration. Furthermore, the drug should have ideal photochemistry parameters to participate in the reaction. For the activation of oxygen it should have a triplet energy higher than the 0.98 eV of oxygen triplet and for others photochemical mechanism of action it should have the ideal parameters namely, ionization potentials and electron affinities. The drug should be soluble in water to react on the cells, that is usually provided by an hydrophilic chemical group incorporated on the drug. In order to have fewer side effects and a decreased drug dose, the drug should preferentially be localized on the tumor site. In reality, the question to find an ideal drug goes beyond chemistry crossing the fields of physics and clinical medicine. It involves physical problems like the interaction of light and body tissue with better dispositive or lasers for light irradiation to be developed. On the clinical part, it is evaluated on the in vitro or in vivo toxicity of the drug as well as the drug’s side effects. The organic and inorganic chemists are interested on the synthesis and characterization of the new drugs. The developments of new quantum mechanics methods with a good balance between accuracy and computational cost, namely Density Functional Theory (DFT) allowed the theoretical chemists to contribute to diverse fields like bio-medicine and other fields where the size of the systems can have dimensions large enough to be studied by older quantum mechanics methods. On the particular question of PDT, the simulation of electronic spectrum and calculation of photochemical parameters can be a support to the work of experimentalist on his synthesis strategy and on the interpretation of obtained data. The chemist experimental work on PDT, usually starts with a tetrapyrrolic macrocycle, natural like porphyrin or synthetic like phthalocyanine. Its structure is modified by the incorporation of substituent groups (e.g phenyl groups) that extends the electron conjugation that can shift the maximum absorption wavelength, λmax, to red part. The possibility of predicting the effect of a substituent group can address the synthesis to a molecule than another. Also, the calculation of the photochemical parameters of the drug allows the evaluation of the feasibility of a mechanism. For example, the electronic energy of the first triplet state furnishes a first estimation on the capability of a drug to generate singlet oxygen. The theoretical methodology in this work is Density Functional Theory (DFT) for the optimization of the structures and its time dependent formalism (TDDFT) for the calculation of electronic excitations. The behavior in solutions is simulated by the solvent implicit methods(C-PCM). On the first part of the study, we focus on two compounds designed to be used on PDT, belonging to the class pentaporphyrins that are porphyrin like molecules containing five pyrrole rings. These compounds were subject to clinical studies were they have shown a PDT action. We predict the electronic spectra and further investigate the mechanism of action of these compounds. On a second part of the study, we focus on corroles - molecules analogues to porphyrin - and its metal complexes, which synthesis were recently reported. We investigate the electronic spectra and evaluate the ability to produce singlet oxygen. The third part of the study consists on a non porphyrin based compounds – the squarines. These molecules are much known to their use in photo cells devices. Their sharp transitions make them as promising drugs to be used on PDT. Il presente lavoro di ricerca, svolto presso il Laboratorio di Chimica Inorganica e di Coordinazione (LaCIC) dell'Università della Calabria, sotto la supervisione del Dott. Massimo La Deda, e in parte nel Laboratoire de Physico-Chimie des Matériaux Luminescents (Université Claude Bernard, Lyon, France), si colloca all'interfaccia tra la Biomedicina, la Chimica di Coordinazione e la Fotochimica, alla ricerca di un comune denominatore. L'obiettivo del nostro lavoro è stato quello di sviluppare una metodologia ed un set-up sperimentale per collegare l'esperienza del LaCIC nella sintesi organometallica, con le applicazioni di composti di coordinazione in campo biomedico. Abbiamo scelto tre aree di ricerca in grado di mettere in evidenza la relazione tra "composti di coordinazione", "luce" e "biomedicina": l'applicazione di complessi metallici incapsulati in polimeri o in nanoparticelle di oro e silice per la generazione di ossigeno di singoletto nella Terapia Fotodinamica (Capitoli 3 e 4), l'utilizzo dei processi a trasferimento di energia che coinvolgono i composti di coordinazione per lo studio delle interazioni farmaco-proteina (applicazioni di “sensing”, capitolo 2), l'utilizzo della luminescenza di nanoparticelle contenenti complessi di metalli di transizione nell’imaging cellulare. Le proprietà uniche dei composti metallici, soprattutto la rilevante fotochimica e fotofisica dei composti di metalli di transizione, li rendono idonei per applicazioni in fotomedicina. Capitolo 2 - Applicazione di “sensing” dei composti di coordinazione: interazione farmaco-proteina. Un nuovo complesso di zinco, recentemente sintetizzato presso il LaCIC, ha evidenziato un’interessante attività antiproliferativa in vitro nei confronti di alcune linee cellulari tumorali. Tuttavia, i test in vitro rappresentano solo il primo step per l’applicazione di questo complesso come farmaco antineoplastico; una fase successiva richiede uno studio della sua biodistribuzione, dunque la sua interazione con biomolecole quali l’ Albumina sierica umana, la proteina più abbondante presente nel torrente circolatorio, la quale aumenta la solubilità di farmaci idrofobici nel plasma e ne modula il rilascio a livello cellulare. Grazie alla fluorescenza della proteina, è stato possibile studiarne il fenomeno di quenching della luminescenza, correlandolo all’interazione di legame con il complesso metallico. Inoltre, la "struttura speciale" del composto di coordinazione, la sua luminescenza intrinseca, ha reso possibile lo studio dell’interazione di legame da un’altra prospettiva, giungendo ad una interessante conclusione, che evidenzia l'aspetto multifattoriale del complesso: terapeutico e sensoristico. Capitolo 3 - Processi attivati dalla luce in composti di coordinazione: fotogenerazione di ossigeno di singoletto. La Terapia Fotodinamica (PDT) fa riferimento all’applicazione di luce al fine di ottenere un effetto terapeutico, in particolare fa riferimento alla capacità di fotogenerare 1O2, una specie altamente reattiva (il “vero” agente terapeutico) da una molecola cosiddetta “fotosensibilizzante”. Tra gli effetti terapeutici dell’ 1O2 si pongono in evidenza la terapia antimicrobica e, soprattutto, la terapia antitumorale: in entrambe è preferibilmente richiesto l’utilizzo di fotosensibilizzanti solubili in acqua. I Complessi di Metalli di Transizione (TMC), grazie alle loro “speciali” proprietà fotofisiche, sono fotosensibilizzanti eccellenti, ma per la maggior parte scarsamente idrofilici. Per rendere TMC solubili in acqua si può procedere per esempio inserendoli in un polimero biocompatibile, senza che gli stessi perdino la loro capacità di generare ossigeno di singoletto. Seguendo questo criterio, è stato sintetizzato e caratterizzato il primo esempio di un polimero solubile in acqua legante un complesso di Pt(II) in grado di generare ossigeno di singoletto. Capitolo 4 - Il paradigma “theranostic”: complessi di metalli di transizione e nanoparticelle. Un’altra alternativa per ottenere un fotosensibilizzante solubile in acqua con le “speciali” proprietà dei TMC è di incapsularlo all’interno di nanoparticelle (NPs), le quali stanno sempre più acquisendo una crescente importanza in ambito medico, grazie alla capacità di agire da sistema di rilascio e alla loro bassa tossicità. Su questa base, sono state sintetizzate e caratterizzate un certo numero di NPs aventi un “core” d’oro e una “shell” di silice con intrappolati nella matrice complessi di Ir (III) e Ru (II), aventi la capacità di generare ossigeno di singoletto. Come prova preliminare, un campione di NPs contenenti un complesso di Ru (II), è stato caratterizzato in vitro per valutarne la citotossicità in diverse linee di cellule tumorali, con risultati promettenti. Inoltre, le "speciali" proprietà fotofisiche dei TMC consentono una disattivazione non radiativa degli stati eccitati (fenomeno necessario per la generazione di 1O2 mediante un processo a trasferimento di energia) senza perdere la luminescenza. In virtù di questo, è stato possibile localizzare le NPs fotosensibilizzanti all'interno della cellula mediante microscopia a fluorescenza, rendendo le NPs sintetizzate un nuovo materiale per “theranostic purposes”. Il lavoro svolto durante il periodo di dottorato di ricerca è stato suddiviso in due progetti: il primo incentrato sulla sintesi di analoghi nucleosidici di tipo isossazolidinici mediante l‟impiego di cicloaddizioni 1,3-dipolari, il secondo progetto basato sulla determinazione del contenuto di digliceridi negli oli d‟oliva mediante spettroscopia 31P NMR, spostando la nostra attenzione nel campo dell‟agroalimentare. Nel primo progetto come primo step, si è costruito un glicosil nitrone opportunamente protetto, a partire da uno zucchero, il diacetone D-glucosio e, successivamente, si è passati alla sua cicloaddizione 1,3-dipolare con un set di vinilnucleobasi, preparate mediante vinilazione diretta delle seguenti nucleobasi: N-1-viniltimina, N-1-vinil-5-fluorouracile, N-1-viniluracile, N-1- vinilcitosina e N-9-viniladenina. Il metodo di sintesi utilizzato, si è dimostrato soddisfacente viste le buone rese, i tempi brevi di reazione, l‟alta regiospecificità e un rapporto diastereoisomerico in favore prevalentemente dello stereoisomero eso rispetto a quello endo, riscontrando quindi una minore formazione di prodotti collaterali senza dover ricorrere ad alcun tipo di protezione delle vinilnucleobasi usate, per le cicloaddizione condotte in microonde ed in assenza di solvente. Nello stesso tempo si è cercato di sintetizzare nitroni di zuccheri a catena aperta in modo da avere degli ossidrili facilmente disponibili per successive trasformazioni. La sintesi ha previsto l‟iniziale formazione di N-benzil-Nglicosilidrossilammina a partire da N-benzilidrossilammina e 2,3,4,6-tetra-Obelzilglicopiranosio. L‟idrossilammina ottenuta, in equlibrio con la forma nitronica, è stata fatta reagire con la N-1-viniltimina, la nucleobase generalmente più attiva verso questa tipologia di sintesi. I composti ottenuti, i 4'-aza analoghi di dideossinucleosidi, già da tempo fanno parte di una classe di substrati che mostrano una potenziale applicazione nei trattamenti antitumorali e contro la diffusione di patologie di natura virale. In tempi brevi i substrati isossazolidinici ottenuti saranno sottoposti a prove biologiche per verificarne la loro attività. Questo progetto si aggiunge alla ricerca attualmente in atto presso il laboratorio di sintesi organica della Dott.ssa Loredana Maiuolo e il suo gruppo di lavoro, circa lo studio di cicloaddizioni 1,3-dipolari per la costruzione di eterocicli a cinque termini diastereoisomericamente puri. Le conoscenze e la manualità acquisita, nel primo progetto, nel campo della sintesi organica sono state utili per la messa a punto del secondo progetto che prevede, inizialmente, la sintesi di standard per la determinazione di digliceridi in oli vergini d‟oliva. I digliceridi (DG), costituenti minor dell‟olio vergine di oliva, possono essere utilizzati nella determinazione del processo di invecchiamento dell‟olio, in base all‟osservazione che un olio fresco presenta maggiori quantità di 1,2- DG e che la sua concentrazione nel tempo diminuisce per isomerizzazione nell‟1,3-DG. Sebbene non esista una regolamentazione ufficiale che stabilisca il contenuto dei vari DG nei diversi oli di oliva, è tuttavia possibile considerare che oli di oliva freschi della stessa varietà di olive siano quelli che contengono un rapporto di 1,3-DG/1,2-DG basso. La spettroscopia NMR può costituire un metodo di indagine alquanto valido nella determinazione dei digliceridi, anche se l‟applicazione di 1H e 13C NMR risulta abbastanza complessa, considerando grosse sovrapposizione di segnali prevalentemente in alcune zone dello spettro. È possibile, invece, pensare di derivatizzare il gruppo funzionale alcolico presente sulla catena diacilglicerolica mediante reagenti al fosforo e utilizzare la spettroscopia 31P NMR per identificare la presenza di 1,2-digliceridi in oli freschi e verificare analiticamente la formazione di 1,3-digliceridi rispetto all‟1,2-derivato in fase di invecchiamento dell‟olio stesso. In questo lavoro verranno illustrati le sintesi iniziali e i risultati ottenuti da uno screening con diversi composti al fosforo e la successiva applicazione del reagente che ha mostrato i migliori risultati con soluzioni standard di 1,2- DG e 1,3-DG e con alcuni campioni di olio d‟oliva calabrese monitorati mediante 31P NMR anche in diversi periodi dell‟anno. http://dx.doi.org/10.13126/UNICAL.IT/DOTTORATI/1148 Dottorato di Ricerca in Organic Materials of Pharmacological Interest, Ciclo XXV a.a. 2011-2012 Università degli Studi della Calabria

Nel presente lavoro di tesi, è riportato un nuovo approccio alla sintesi di furani-3-carbossilati tramite reazione di carbonilazione ossidativa palladiocatalizzata di composti 3-in-1,2-diolici, aventi un gruppo alcolico primario o secondario sul C-1. Tutte le reazioni sono state condotte in solvente alcolico (metanolo o etanolo) in condizioni relativamente blande (100°C e 40 atm di una miscela CO-aria 4:1). I rispettivi furani carbonilati sono stati ottenuti con rese eccellenti (56-93%) attraverso un processo di etero ciclizzazione-alcossicarbonilazione-deidratazione 5-endo-dig sequenziale, usando l’ossigeno come ossidante esterno. In condizioni simili, i derivati 2- metil-3-in-1,2-diolici, aventi un gruppo alcolico terziario, permettono la sintesi di 4-metilene-4,5-diidrofurani-3-carbossilati con rese soddisfacenti (58-60%). I tiofeni sono una classe molto importante di composti eterociclici. Molte molecule contenenti il nucleo tiofenico mostrano una grande varietà di attività biologiche e trovano applicazione in campo farmaceutico e cosmetico. Inoltre, sono degli intermedi sintetici molto utili nella preparazione di nuovi materiali polimerici. Nel presente lavoro è stato realizzato un nuovo percorso sintetico per la sintesi di tiofeni sostituiti a partire da 1-mercapto-3-in-2-oli, tramite S-eterociclizzazione PdI2/KIcatalizzata. Il sistema catalitico PdI2/KI, sviluppato dal gruppo di ricerca in cui è stato svolto il Dottorato, ha già dimostrato la sua efficacia nel favorire reazioni simili. La reazione di amminocarbonilazione ossidativa di alchini funzionalizzati è uno dei metodi più versatili per la sintesi diretta di eterocicli e carbocicli carbonilati. In particolare, il sistema catalitico PdI2/KI è un ottimo promotore per queste reazioni, in presenza di ossigeno come ossidante. Nell’ultima parte del lavoro è stata sviluppata una nuova sintesi di indanilideni carbossilati, applicando la reazione di amminocarbonilazione ossidativa a esteri 2-etinilbenzilmalonici, per l’ottenimento di prodotti indanilidenici carbonilati con un alto grado di selettività nei confronti del diastereoisomero E che, in alcuni casi, è stato isolato in maniera totalmente selettiva. The ever expanding cutting edge technologies in medicine for the benefit of society, the orthopedic branch is one among those significant branches in medicine pertaining to bone. Bisphosphonates (BPS) are being increasingly and successfully used to prevent bone fractures and the concerning problems of bone diseases such as Paget’s diseases, osteoporosis and tumour bone disease. In view of this specific problem, BPS are well established in the treatment of osteoclast -mediated resorbtive bone diseases including osteoporosis, Paget's disease and tumor-induced osteolysis. Recent studies suggest that, besides inhibiting bone resorbtion, BPS may also exert a direct antitumor effect, and this class of drugs has been shown to inhibit proliferation and to induce apoptosis in vitro in different human tumor cell lines. BPs are classified into two groups according to their chemical structure and mechanism of action: (i) non nitrogen containing BPS such as etidronate and clodronate that are of low potency and inhibit osteoclast function via metabolism into toxic ATPmetabolites and (ii) nitrogen-containing BPS (NBPS), such as pamidronate, alendronate, risedronate, ibandronate and zoledronate which is the most potent antiresorptive agent. Hence in present investigation we synthesized some several bisphosphonates bearing a substituted isoxazolidine ring by direct 1, 3- dipolar cyclization reaction in the absence of solvent and good yield under novel, promising and low cost microwaves catalysis. The method allows the simultaneous incorporation on the geminal position of the bisphosphonate framework, of basic nitrogen and of an oxygen atom, as third hook. The studies on the inhibitory potency of cyclic nitrogen containing bisphosphonates indicate that the presence of two geminal phosphonate groups is responsible for interaction with the molecular target. In addition, basic nitrogen in the heterocyclic side chain affects potency and its orientation is critical for effective inhibition of bone diseases. For the synthetic point of view, different aryl and alkyl substituents on the isoxazolidine ring prompt us to investigate the ring opening of these compounds through cleavage of the N-O bond. This strategy represents a novel access to new gem-hydroxyl bisphosphonates, bearing aryl substituents on the lateral chain. The reductive cleavage of the N-O bond in isoxazolidines represents a simple and direct access to N-substituted aminoalcohols, valuable intermediates in many synthetic strategies. Moreover, additional reaction path way have been envisaged leading to the formation of non-hydroxyl bisphosphonates. Nowadays, intensive efforts have been carried out on the design of novel advanced molecular materials, which can self-assemble in a strong, directional and reversible way to construct supramolecular materials with specific properties. The rational design and preparation of supramolecular assemblies through the coordination of metal ions with organic ligands has attracted attention for developing novel crystalline materials with interesting structural topologies and promising applications, and has evolved as an interesting research. The metals used in these complexes can serve as structural components and/or as a source of properties (e.g., magnetic, catalytic, optoelectronic, etc). Cyclometallated Ir(III) octahedral complexes possess fascinating properties used in various applications such as luminescent and electrochemiluminescent labeling reagents for biological substrates1, sensors2, or electronic devices3,4. Recently, the interest in ionic Ir(III) complexes is growing rapidly because not only high internal quantum efficiency (~100%) can be achieved in principle, but also tunable emission wavelengths over the entire visible spectrum can be successfully obtained through ingenious modification of ligands. In particular, Ir(III) complexes based on the chelating ligand 2,2’-bipyridine (bpy) have been successfully applied in light-emitting electrochemical cells (LECs) and sensors.5 The theoretically calculated phosphorescence yield (Fp) of the Ir(III) complexes are close to unity in solution.6 The solution investigations have made great contributions to the fundamental understanding of luminescence processes at molecular level. The conclusions drawn from the dilute solution data, however, cannot commonly be extended to the concentrated solutions. Indeed, many Ir(III) complexes show very different light-emitting behaviors in dilute and concentrated solutions and respectively in the solid state. The luminescence is often weakened or quenched at high concentrations, a phenomenon widely known as “concentration quenching”. A main cause for the quenching process is mechanistically associated with the “formation of aggregates”, which is probably why the concentration quenching effect has frequently been referred to as “aggregationcaused quenching” (ACQ). On the other hand “aggregation-induced phosphorescent emission” (AIPE) is an unusual phenomenon existing also in transition metal complexes, which have no emission in solution but enhanced emission in the solid state.7 There are some examples of AIPE, most of them in neutral Ir(III) complexes.8, 9, 10, 11, 12 The main strategies to avoid unpleasant quenching phenomena are based on the dispersion of the chromophore. Mainly, two strategies are employed: engineering at molecular level by introducing functionalities able to electronically disconnect the chromophores (bulky groups or functionalities capable to construct hard crystalline or soft dynamic supramolecular assemblies) or isolating the active molecules in different host matrices (host-guest systems).13 In particular, the dispersion of a chromophore into mesoporous materials not only prevents the aggregation phenomena but also provides increased thermal, chemical and mechanical stability to the final materials. Mesoporous materials are ordered porous materials with periodic distribution of pores, high surface area, controllable large pore sizes in the range of 2 – 50 nm and variable topology of the pores. The inorganic matrixes may be made up of SiO2, TIO2, ZrO2, Al2O3, Nb2O5 etc. Basically, the synthesis of ordered functional mesoporous materials is based on the condensation of an inorganic scaffold on the organised structure formed in water by surfactant molecules. Two different strategies may be employed, the cooperative self-assembly mechanism (CSA) and the true liquid crystal templating’ (TLCT) mechanism.14 The functionalization of the mesoporous material may be done in both cases by inserting the chromophore into the primarily water solution. Therefore, water soluble chromophores may guarantee a better compatibility with the surfactant/water system, whereas a proper functionalization on the molecular structure of the chromophore that permit the self-assembly into supramolecular ordered water assemblies, will allow to use the chromophores directly as structure directing agents (SDAs). Since the photophysical properties of the ionic complexes are influenced profoundly by the surroundings of the molecule both in solution and in condensed states, it is fundamental to study the behavior of such complexes in these different states, in order to achieve a fine tuning of the properties as a function of their structure and order in the final material. The knowledge gained in the assembling of supramolecular materials using non-covalent bonds may be used for the construction of ordered systems in water. This strategy will permit the one-step synthesis of functional mesoporous materials, and to control the order of the final material controlling the order in water of the functional Ir(III) complexes. In particular, the molecular fragments that one can change to achieve the desired properties in the final ionic Ir(III) complexes are the cyclometallating or coordinating ligands, and respectively the counterion. My research therefore is focused on the design and synthesis of hydrophilic ionic Ir(III) complexes with flexible or rigid ancillary ligands and use of different counterions, all suitable for controlling the supramolecular assembly in the solid state, and to transfer the knowledge gained into obtaining ordered structures in water, or water-surfactant systems, necessary for the synthesis of mesoporous materials with defined properties.
