Thermal effects on turbulent flow and dispersion in and above an idealized street canyon with a street aspect ratio of 1 are numerically investigated using the parallelized large-eddy simulation model (“PALM”). Each of upwind building wall, street bottom, and downwind building wall is heated, and passive scalars are emitted from the street bottom. When compared with the neutral (no heating) case, the heating of the upwind building wall or street bottom strengthens a primary vortex in the street canyon and the heating of the downwind building wall induces a shrunken primary vortex and a winding flow between the vortex and the downwind building wall. Heating also induces higher turbulent kinetic energy and stronger turbulent fluxes at the rooftop height. In the neutral case, turbulent eddies generated by shear instability dominate mixing at the rooftop height and appear as band-shaped perturbations in the time–space plots of turbulent momentum and scalar fluxes. In all of the heating cases, buoyancy-generated turbulent eddies as well as shear-generated turbulent eddies contribute to turbulent momentum and scalar fluxes and band-shaped or lump-shaped perturbations appear at the rooftop height. A quadrant analysis shows that at the rooftop height, in the neutral case and in the case with upwind building-wall heating, sweep events are less frequent but contribute more to turbulent momentum flux than do ejection events. By contrast, in the case with street-bottom and downwind building-wall heating, the frequency of sweep events is similar to that of ejection events and the contribution of ejection events to turbulent momentum flux is comparable to that of sweep events. Urban land surface schemes have been developed to model the distinct features of the urban surface and the associated energy exchange processes. These models have been developed for a range of purposes and make different assumptions related to the inclusion and representation of the relevant processes. Here, the first results of Phase 2 from an international comparison project to evaluate 32 urban land surface schemes are presented. This is the first large-scale systematic evaluation of these models. In four stages, participants were given increasingly detailed information about an urban site for which urban fluxes were directly observed. At each stage, each group returned their models' calculated surface energy balance fluxes. Wide variations are evident in the performance of the models for individual fluxes. No individual model performs best for all fluxes. Providing additional information about the surface generally results in better performance. However, there is clear evidence that poor choice of parameter values can cause a large drop in performance for models that otherwise perform well. As many models do not perform well across all fluxes, there is need for caution in their application, and users should be aware of the implications for applications and decision making. © 2010 Royal Meteorological Society.

A large number of urban surface energy balance models now exist with different assumptions about the important features of the surface and exchange processes that need to be incorporated. To date, no comparison of these models has been conducted; in contrast, models for natural surfaces have been compared extensively as part of the Project for Intercomparison of Land-surface Parameterization Schemes. Here, the methods and first results from an extensive international comparison of 33 models are presented. The aim of the comparison overall is to understand the complexity required to model energy and water exchanges in urban areas. The degree of complexity included in the models is outlined and impacts on model performance are discussed. During the comparison there have been significant developments in the models with resulting improvements in performance (root-mean-square error falling by up to two-thirds). Evaluation is based on a dataset containing net all-wave radiation, sensible heat, and latent heat flux observations for an industrial area in Vancouver, British Columbia, Canada. The aim of the comparison is twofold: to identify those modeling approaches that minimize the errors in the simulated fluxes of the urban energy balance and to determine the degree of model complexity required for accurate simulations. There is evidence that some classes of models perform better for individual fluxes but no model performs best or worst for all fluxes. In general, the simpler models perform as well as the more complex models based on all statistical measures. Generally the schemes have best overall capability to model net all-wave radiation and least capability to model latent heat flux. © 2010 American Meteorological Society.

There are now a number of urban models within the community, which vary in complexity from simple schemes to the detailed representation of momentum and energy fluxes, even distributed within the atmospheric boundary layer. In addition to variations in complexity, these models also differ in parameter requirements and on a more practical level, the cost of running the models. Whilst many of these models have been validated against observational datasets, with some models even using the same observations, these have not been done in a controlled experiment that enables comparisons to be made between the models. Such controlled experiments, however, have been shown in the past to provide scientific insight into both the mechanics of the models and also the physics of the real world, for instance the PILPS experiments. This presentation will describe the progress that has been made so far with regards to an urban model comparison project.

We have reported previously on the expression of recombinant human type X collagen (hrColX) in HEK 293 and HT 1080 cells by using the eukaryotic expression vector pCMVsis (in which CMV stands for cytomegalovirus). Several stably transfected clones secreted full-length triple-helical hrColX molecules in large amounts, but the secreted collagen was underhydroxylated, with a hydroxyproline-to-proline ratio of 0.25 and a melting temperature (T(m)) of 31 degrees C. By comparison, native chicken type X procollagen has a T(m) of 46 degrees C. To stabilize the triple helix of hrColX, an hrColX-expressing clone (A6/16) was co-transfected with both alpha and beta subunits of human prolyl 4-hydroxylase. Clones were selected that secreted proalpha1(X) collagen chains with an apparent molecular mass of 75 kDa and an increased hydroxyproline-to-proline ratio of close to 0.5. As a result of enhanced prolyl hydroxylation, the T(m) of the hrColX was increased to 41 degrees C as measured by CD analysis at various temperatures. The CD spectra indicated a minimum ellipticity at 198 nm and a peak at 225 nm at 20 degrees C, confirming the presence of a triple helix. The same T(m) of 41 degrees C was measured for the triple-helical core fragments of hrColX of 60-65 kDa that were retained after brief digestion with chymotrypsin/trypsin at increasing temperatures. This shows that the human cell line HEK-293 is suitable for the simultaneous expression of three genes and the stable production of substantial amounts of recombinant, fully hydroxylated type X collagen over several years.
