In most mammals, including humans, the postnatal acquisition of normal social and nonsocial behavior criticallydepends on interactions with peers. Here we explore the possibility that mixed-group housing of mice carrying adeletion of Nlgn3, a gene associated with autism spectrum disorders, and their wild-type littermates induceschanges in each other’s behavior. We have found that, when raised together, male Nlgn3 knockout mice and theirwild-type littermates displayed deficits in sociability. Moreover, social submission in adult male Nlgn3 knockoutmice correlated with an increase in their anxiety. Re-expression of Nlgn3 in parvalbumin-expressing cells intransgenic animals rescued their social behavior and alleviated the phenotype of their wild-type littermates, furtherindicating that the social behavior of Nlgn3 knockout mice has a direct and measurable impact on wild-typeanimals’ behavior. Finally, we showed that, unlike male mice, female mice lacking Nlgn3 were insensitive to theirpeers’ behavior but modified the social behavior of their littermates. Altogether, our findings show that theenvironment is a critical factor in the development of behavioral phenotypes in transgenic and wild-type mice. Inaddition, these results reveal that the social environment has a sexually dimorphic effect on the behavior of micelacking Nlgn3, being more influential in males than females.

Das kleinzellige Bronchialkarzinom (SCLC) gehört zu den malignen Neoplasien mit einer extrem schlechten Prognose. Die Chemo-/Radiotherapie zeigt sich zwar initial als sehr effektiv mit guten Remissionsraten, es kommt aber im Krankheitsverlauf praktisch unausweichlich zu einem Rezidiv auf Grund einer sekundär erworbenen Resistenz gegenüber den eingesetzten Therapeutika. Es gibt verschiedene Modellvorstellungen über die Ursachen dieser Resistenzentwicklung. Da Zytostatika und Gamma-Strahlen einen Großteil ihrer Wirkung durch Induktion des Apoptoseprogramms (Anstoß des programmierten Zelltodes) ausüben, besteht eine Hypothese in der Vorstellung, daß in den resistenten Zellen das Apoptoseprogramm inhibiert oder herunterreguliert ist. Die meisten Daten zu Mechanismen der Resistenzentwicklung wurden bislang in Zellkulturmodellen gewonnen. Auch diese Arbeit basiert auf einem solchen Modell. In unserer Arbeitsgruppe ist es gelungen, von den drei kleinzelligen Bronchialkarzinomzellinien NCI-H69, NIH-N592 und NCI-H82, die normalerweise im Zellkulturmedium schwimmen, also im Suspensionszustand wachsen, Kulturen zu subkultivieren, die sich durch ein am Zellkulturflaschenboden adhärentes Wachstum auszeichneten. Diese adhärenten Zellen zeigten eine völlig veränderte Morphologie im Sinne von flachen, ausgezogenen, epitheloiden Zellen. In der Arbeit wurde nun geprüft, ob die veränderte Morphologie der identischen Linien mit einer neu erworbenen Resistenz gegenüber verschiedenen Zytostatika Etoposid, Cyclophosphamid, Doxorubicin und Taxol) einhergeht. Interessanterweise zeigten die adhärenten Subklone H69ad und N592ad eine deutliche Resistenz gegenüber ihren Suspensions-Ausgangszellinien H69sus und N592sus. Diese Resistenz der adhärenten H69ad und N592ad zeigte sich auch gegenüber der Bestrahlung mit Gamma-Strahlen. Untersuchungen des Anteils der apoptotischen Zellen nach Gamma-Strahlen-Exposition ergaben in den adhärenten H69ad und N592ad eine deutlich geringere Zahl apoptotischer Zellen. Die gefundene Chemo-/Radioresistenz der adhèrenten Zellen ist also am ehesten durch eine Inhibition des Apoptoseprogramms bedingt. In den adhärenten Zellinien H69ad und N592ad fanden sich auf RT-PCR-Ebene differentiell hochregulierte a2-, a3- und b4-Integrine. Entsprechend anderen in der Literatur beschriebenen Adhärenz-induzierten Chemo-/Radioresistenz-Modellen (CAM-DR) könnten diese Integrine die Mediatoren der Adhäenz-induzierten Resistenz in unserem Modell sein. Interessanterweise fanden wir eine konstitutiv aktivierte Proteinkinase B/Akt in den adhäenten Klonen H69ad und N592ad, die in den Suspensionszellinien H69sus und 592sus nicht vorlag. PKB/Akt ist eine Proteinkinase, die an zentraler Stelle eines potenten anti-apoptotischen Signaltransduktionsweges liegt. Die Aktivierung der PKB/Akt in H69ad und N592ad ist direkt Adhärenz-abhängig, da sie durch Rückführung des adhärenten Phänotyps der Linien H69ad und N592ad zu Suspensionszellen bei Kultivierung auf völlig unbeschichteten Kulturschalen rückgängig zu machen war. Resultierend aus obigen Ergebnissen wurde in dieser Arbeit zusätzlich untersucht, ob über experimentelle Transfektion von PKB/Akt Antisense-Oligonukleotiden das Resistenzverhalten modulierbar ist. Die experimentellen Ergebnisse liessen hier jedoch keine eindeutige Schlussfolgerung zu. Es verbleibt somit die Notwendigkeit, weitere Untersuchungen zum Mechanismus der Adhärenz-induzierten Chemo-/Radioresistenz durch Apoptoseinhibition durchzuführen. Auf der Basis eines besseren Verständnis könnten so in Zukunft eventuell neue Therapeutika entwickelt werden, die zu einer Überwindung dieses bisher ungelösten Therapieproblems beim kleinzelligen Bronchialkarzinom führen., Most small cell lung cancer (SCLC) patients relapse within 12 months of starting combination chemotherapy plus radio-therapy, due to the development of acquired chemo- and radio-resistance. This phenomenon relates to the induction of tumour differentiation, resulting in apoptosis-resistant, morphologically variant (v-SCLC) cells, which lack the neuroendocrine expression of classic (c-) SCLC cells. In this study spontaneously adherent SCLC sublines were shown by differential gene expression analysis to provide an in vitro model of variant differentiation in SCLC, with down-regulation of neuroendocrine markers and up-regulation of epithelial differentiation markers. The sensitivity of adherent SCLC sublines to etoposide, cyclophosphamide and gamma radiation was significantly diminished relative to parent suspension cell lines. Western blot analysis using phosphorylation-specific antibodies to Akt and MAP kinase revealed markedly elevated activation in adherent SCLC sublines, paralleled by increased levels of phosphorylated Bad protein and activated NF-kappaB. Subcultivation of the adherent sublines on uncoated surfaces reversed their adherent phenotype immediately and under these conditions Akt activity reverted to low levels. These results suggest that c-SCLC cells can differentiate spontaneously to v-SCLC and that the associated cellular adhesion may trigger Akt-dependent inhibition of apoptosis in SCLC cells, thus leading to acquired chemo- and radio-resistance.

Autism spectrum disorders (ASD) are more prevalent in male than in female\ud individuals and are characterised by clinical core symptoms such as impaired\ud sociability, verbal communication and ritualistic behaviours. The gene encoding the\ud cytoplasmic FMR1 interacting protein 1 (CYFIP1) has been associated with ASD in\ud humans. At the molecular level, CYFIP1 has been demonstrated to negatively\ud regulate protein synthesis and actin remodelling. At the neuronal level, Cyfip1\ud haploinsufficiency leads to defects in synaptic plasticity associated with alteration of dendritic spine morphology, two pathophysiological features found in numerous\ud mouse models of ASD. However, the consequences of Cyfip1 deletion at the\ud behavioural level remains unclear, limiting our understanding of the relationship\ud between pathophysiology and behavioural phenotypes.\ud With this study, we aimed to characterise the behavioural phenotype of Cyfip1+/- mice and then to identify associated cellular phenotypes. The results we obtained revealed sex-specific defects in social interest and motor learning. In addition, motor learning deficits were observed in adult Cyfip1+/- mice but not earlier in development. Associated with motor learning deficits, we identified a brain region-specific neuronal phenotype with decreased dendritic spine densities and increased dendritic spine turnover in Cyfip1+/- mice. The dendritic spine formation and the in vivo protein synthesis rate were intact in Cyfip1+/- mice.\ud These results identified behavioural deficits in Cyfip1+/- mice, which relate to symptoms and comorbidities of ASD in human. The cellular phenotypes indicated an alteration of dendritic spine density and spine turnover, a phenotype found in several mouse models of ASD and in humans affected by the condition. Altogether, these findings indicate that Cyfip1+/- mice can represent a valuable model for the study of ASD pathophysiology and in particular the relationship between specific neuronal phenotypes and behavioural alterations.
