Background: Recent open-label trials show that psychedelics, such as ayahuasca, hold promise as fast-onset antidepressants in treatment-resistant depression. Methods: To test the antidepressant effects of ayahuasca, we conducted a parallel-arm, double-blind randomized placebo-controlled trial in 29 patients with treatment-resistant depression. Patients received a single dose of either ayahuasca or placebo. We assessed changes in depression severity with the Montgomery-Åsberg Depression Rating Scale (MADRS) and the Hamilton Depression Rating scale at baseline, and at 1 (D1), 2 (D2), and 7 (D7) days after dosing. Results: We observed significant antidepressant effects of ayahuasca when compared with placebo at all-time points. MADRS scores were significantly lower in the ayahuasca group compared with placebo at D1 and D2 (p = 0.04), and at D7 (p < 0.0001). Between-group effect sizes increased from D1 to D7 (D1: Cohen's d = 0.84; D2: Cohen's d = 0.84; D7: Cohen's d = 1.49). Response rates were high for both groups at D1 and D2, and significantly higher in the ayahuasca group at D7 (64% v. 27%; p = 0.04). Remission rate showed a trend toward significance at D7 (36% v. 7%, p = 0.054). Conclusions: To our knowledge, this is the first controlled trial to test a psychedelic substance in treatment-resistant depression. Overall, this study brings new evidence supporting the safety and therapeutic value of ayahuasca, dosed within an appropriate setting, to help treat depression. This study is registered at http://clinicaltrials.gov (NCT02914769). [ABSTRACT FROM AUTHOR]

- Abstract

In this paper, we discuss synchronously the status of the following morphological elements, often used in new word formations in Brazilian Portuguese: affixoids (bio-combustível, eco-sustentabilidade), splinters (choco-tone; sogra-drasta) and borrowed constituents (cyber-café; e-professor). In our description, we observe the extent to which these constituents behave as radicals and in what ways are equivalent to affixes. We intend, thereby, ratify the continuum proposed by Baker (2000) and Ralli (2007), at same time we show that other morphological units, as well as radicals and affixes, should be part of this scale.

In the first part of this work, simultaneous fMRI/EEG recordings were applied to analyze the functional connectivity of the hippocampus with the neocortex during human sleep. This provided relevant differentiation of sleep stage specific contributions to neural plasticity as proposed in models of sleep dependent memory consolidation. In the second part, more methodological aspects were addressed, evaluating the potential of a non-linear analysis method called recurrence quantification analysis (RQA) to extract rapidly changing dynamic features of EEG data, which may be used to better study fMRI models. According to the present results, RQA may be used as an additional tool to obtain new insights in neurophysiological and neuroanatomical correlates of stimulus habituation and novelty processing even in combined trial-by-trial functional fMRI/EEG experiments.

ddc:330 - Abstract
Around the world, cycling has mostly been used for two main purposes. Some societies view and utilize the bicycle as an integral part of the transportation system. Even if not used as the main transportation mode, it can still be used efficiently as a complementary transportation mode to public transit modes. The Netherlands and Denmark are examples of European countries where cycling accounts for high daily shares. Among Asian countries, Japan and China present significant cycling rates. However, as the contemporary societies grow more auto-dependent, cycling has been merely seen as a means of recreation and rarely used for daily travel in many other countries. Examples are the United States, Australia and Great Britain where nationwide statistics suggest insignificant bicycle shares. As part of a larger research project, this paper presents a preliminary discussion on the use of bicycle in two countries with significantly different cycling characteristics, Great Britain and Japan. In most British cities, cycling is not an integral part of the transportation system, despite considerable efforts from government. Conversely, cycling is a widely accepted transportation mode throughout Japan, even though Japanese cities do not yet present extensive cycling networks. Traditionally, Japanese urban plans are limited to two main actions with regards to cycling; either implementing bicycle parking facilities around railway/subway stations, or establishing bicycle zones within road intersections. The main objective of this paper is to present an overview of each country's cycling system, with a focus on travel patterns and bicycle facilities. Initially, benefits from cycling are pointed out. Cycling trends are then compered, whilst bicycle infrastructure and service are summarized. The paper concludes by comparing transport policies and strategies towards cycling in these countries and discussing the relative merits of each approach.

Background: The responsiveness of the Early Childhood Oral Health Impact Scale (ECOHIS) has varied greatly across studies; hence, we hypothesized that this discrepancy could be related to the complexity of dental treatment received. Thus, we aimed to evaluate the responsiveness of the ECOHIS to changes in oral health-related quality of life (OHRQoL) following dental treatments of varying complexity in preschool children. Methods: Preschool children aged 3 to 6 years were selected; their parents responded to the ECOHIS at baseline. The parents responded to the ECOHIS again and a global transition question 30 days after the children were treated. The type of treatment received by the children was categorized according to complexity, as follows: 1) non-operative treatment only, 2) restorative treatment, and 3) endodontic treatment and/or tooth extraction. Change scores and effect sizes (ES) were calculated for total scores, as well as considering the different treatment types and global transition question responses. Results: Of the 152 children who completed the study, the ECOHIS yielded large ES for total scores (0.89). The children showed increasing ES values associated with better perception of improvement, assessed by the global transition question. The magnitude of ES after treatment was related to treatment complexity (0.53, 0.92 and 1.43, for children who received non-operative treatment only, restorative treatment, and endodontic treatment and/or tooth extraction, respectively). Conclusions: Parents whose children required more complex dental treatment are more likely to perceive treatment-related changes to OHRQoL assessed with the ECOHIS. - Abstract
We studied visuospatial performance and obtained brain perfusion scintigraphy in 27 patients with posterior cortical atrophy (PCA) and 24 healthy controls, with two aims: (1) to determine inter-hemispheric perfusion imbalances underlying signs of spatial neglect and (2) to establish the functional substrates of patients' performance on distinct visuospatial tasks (line bisection and target cancellation). Between-groups and correlation analyses were performed on a voxel-wise basis with Statistical Parametric Mapping, and right-to-left hemispheric perfusion ratios were calculated in anatomical regions of interest. Nineteen patients had pathological spatial biases. Compared with controls, patients with signs of left-sided and right-sided neglect presented prominent hypoperfusion in the right and left parietal cortex, respectively. Importantly, hypoperfusion extended to the ipsilateral prefrontal regions. Correlation analyses between task scores and brain perfusion showed that shifts in line bisection correlated with hypoperfusion in parieto-frontal regions, whereas omissions on target cancellation mainly correlated with hypoperfusion in prefrontal structures. Overall, the results indicate that spatial neglect in PCA is related to inter-hemispheric perfusion asymmetries in fronto-parietal networks, with partially different neural correlates for line bisection and target cancellation.

- Abstract
In posterior cortical atrophy (PCA), there is a progressive impairment of high-level visual functions and parietal damage, which might predict the occurrence of visual neglect. However, neglect may pass undetected if not assessed with specific tests, and might therefore be underestimated in PCA. In this prospective study, we aimed at establishing the side, the frequency and the severity of visual neglect, visual extinction, and primary visual field defects in an unselected sample of PCA patients.

Graph theoretical analysis of functional magnetic resonance imaging (fMRI) time series has revealed a small-world organization of slow-frequency blood oxygen level-dependent (BOLD) signal fluctuations during wakeful resting. In this study, we used graph theoretical measures to explore how physiological changes during sleep are reflected in functional connectivity and small-world network properties of a large-scale, low-frequency functional brain network. Twenty-five young and healthy participants fell asleep during a 26.7 min fMRI scan with simultaneous polysomnography. A maximum overlap discrete wavelet transformation was applied to fMRI time series extracted from 90 cortical and subcortical regions in normalized space after residualization of the raw signal against unspecific sources of signal fluctuations; functional connectivity analysis focused on the slow-frequency BOLD signal fluctuations between 0.03 and 0.06 Hz. We observed that in the transition from wakefulness to light sleep, thalamocortical connectivity was sharply reduced, whereas corticocortical connectivity increased; corticocortical connectivity subsequently broke down in slow-wave sleep. Local clustering values were closest to random values in light sleep, whereas slow-wave sleep was characterized by the highest clustering ratio (gamma). Our findings support the hypothesis that changes in consciousness in the descent to sleep are subserved by reduced thalamocortical connectivity at sleep onset and a breakdown of general connectivity in slow-wave sleep, with both processes limiting the capacity of the brain to integrate information across functional modules. Background: A condition vital for the consolidation and maintenance of sleep is generally reduced responsiveness to external stimuli. Despite this, the sleeper maintains a level of stimulus processing that allows to respond to potentially dangerous environmental signals. The mechanisms that subserve these contradictory functions are only incompletely understood. Methodology/Principal Findings: Using combined EEG/fMRI we investigated the neural substrate of sleep protection by applying an acoustic oddball paradigm during light NREM sleep. Further, we studied the role of evoked K-complexes (KCs), an electroencephalographic hallmark of NREM sleep with a still unknown role for sleep protection. Our main results were: (1) Other than in wakefulness, rare tones did not induce a blood oxygenation level dependent (BOLD) signal increase in the auditory pathway but a strong negative BOLD response in motor areas and the amygdala. (2) Stratification of rare tones by the presence of evoked KCs detected activation of the auditory cortex, hippocampus, superior and middle frontal gyri and posterior cingulate only for rare tones followed by a KC. (3) The typical high frontocentral EEG deflections of KCs were not paralleled by a BOLD equivalent. Conclusions/Significance: We observed that rare tones lead to transient disengagement of motor and amygdala responses during light NREM sleep. We interpret this as a sleep protective mechanism to delimit motor responses and to reduce the sensitivity of the amygdala towards further incoming stimuli. Evoked KCs are suggested to originate from a brain state with relatively increased stimulus processing, revealing an activity pattern resembling novelty processing as previously reported during wakefulness. The KC itself is not reflected by increased metabolic demand in BOLD based imaging, arguing that evoked KCs result from increased neural synchronicity without altered metabolic demand. Background Current methods of amyloid PET interpretation based on the binary classification of global amyloid signal fail to identify early phases of amyloid deposition. A recent analysis of 18F-florbetapir PET data from the Alzheimer’s disease Neuroimaging Initiative cohort suggested a hierarchical four-stage model of regional amyloid deposition that resembles neuropathologic estimates and can be used to stage an individual’s amyloid burden in vivo. Here, we evaluated the validity of this in vivo amyloid staging model in an independent cohort of older people with subjective memory complaints (SMC). We further examined its potential association with subtle cognitive impairments in this population at elevated risk for Alzheimer’s disease (AD). Methods The monocentric INSIGHT-preAD cohort includes 318 cognitively intact older individuals with SMC. All individuals underwent 18F-florbetapir PET scanning and extensive neuropsychological testing. We projected the regional amyloid uptake signal into the previously proposed hierarchical staging model of in vivo amyloid progression. We determined the adherence to this model across all cases and tested the association between increasing in vivo amyloid stage and cognitive performance using ANCOVA models. Results In total, 156 participants (49%) showed evidence of regional amyloid deposition, and all but 2 of these (99%) adhered to the hierarchical regional pattern implied by the in vivo amyloid progression model. According to a conventional binary classification based on global signal (SUVRCereb = 1.10), individuals in stages III and IV were classified as amyloid-positive (except one in stage III), but 99% of individuals in stage I and even 28% of individuals in stage II were classified as amyloid-negative. Neither in vivo amyloid stage nor conventional binary amyloid status was significantly associated with cognitive performance in this preclinical cohort. Conclusions The proposed hierarchical staging scheme of PET-evidenced amyloid deposition generalizes well to data from an independent cohort of older people at elevated risk for AD. Future studies will determine the prognostic value of the staging approach for predicting longitudinal cognitive decline in older individuals at increased risk for AD. Ayahuasca is a brew with psychedelic properties largely used by indigenous populations from the Amazon basin. It contains the psychedelic tryptamine N,N-dimethyltriptamine (DMT), and monoamine oxidase inhibitors (iMAO), such as harmine and harmaline. Ayahuasca is consid-ered to be a serotonergic psychedelic, capable of inducing an altered state of consciousness with similarities to an oneiric experience, with intense alterations in perception, though, humor, emo-tion, and mystical-type experiences. The neural correlates of its acute effects have been inves-tigated by different neuroimaging techniques, including electroencephalography (EEG). In this study, we explored EEG spectral changes in 48 healthy volunteers using a randomized double-blind placebo-controlled trial. Half of the volunteers received ayahuasca, half received placebo. We used an EEG system to monitor all volunteers throughout the dosing session, which lasted approximately 4-hours. Aiming to improve data quality, the volunteers were asked to perform two controlled tasks at three specific moments: before intake, 2h and 4h after intake. For the first task, they alternated moments of eyes open (20 seconds) with eyes closed (40 seconds) for 5 minutes, avoiding falling asleep. During the second task they should keep their eyes closed for another 5 minutes, again avoiding falling asleep. Spectral analysis at 2h after intake shows reduced alpha power, and increased theta and beta in the ayahuasca group with respect to the placebo, mainly in occipital and right temporoparietal regions. Correlation analysis revealed correspondences between the alpha power (2h) and individual scores on two scales used to measure psychedelic effects – the Hallucinogen Rating Scale (HRS) and the Mystical Experi-ence Questionnaire (MEQ). Additionally, we also present EEG traces with electrophysiological events that might be of importance for the representation of the psychedelic experience. Over-all, our results suggest that the inhibition of alpha oscillations, increased theta and beta in right posterior brain regions play an important role on the psychedelic experience, maybe sharing mechanisms present during the oneiric experience.

Emotions are crucial for the survivor of the organisms. They allow individuals to recognize what is threatening and what is safe in the environment. Emotions work modulating several cognitive processes, such as perception, attention and memory. At an electrophysiological level, emotions influence the response of neuronal networks measured by Event-Related Potentials (ERP). It has been documented that pleasant and unpleasant pictures increase the amplitude of the visual ERP in the visual areas of the brain. However, little is known about how emotions affect the processing of stimuli of different sensory modalities. The aim of this work was to test how the International Affective Picture System (IAPS) may affect the ERP of an innocuous sound. For this purpose the auditory ERP of 36 healthy students was recorded while they were watching a stream of high arousal unpleasant, high arousal pleasant, neutral images or a white screen. Heart rate variability (HRV), subjective self-report and band frequency power were measured too. Results showed that subjective self-report of the pictures emotional property verified the emotional qualities of these pictures, but it was slightly different form the Brazilian standard scores. We also found a deceleration of the HRV during emotional pictures viewing, and this correlated with the subjective self-report. No effect from emotional valence on the auditory ERP was found, only an effect of the pictures when compared to the white screen condition. In the left hemisphere, band frequency power showed a increase of theta and alpha power in the first moment for the four conditions. In a second moment, there was a increase of the power of this two bands during emotional pictures presentation in the right hemisphere. And then, it was seen a decrease in the right hemisphere of theta band power during unpleasant picture viewing. Scores from the subjective self-report and HRV confirmed that IAPS are a good method to elicit and evaluate emotions, but is necessary to extend the Brazilian standardization to have a more reliable instrument. Findings in the auditory ERP suggest a division of attention between the two kind of stimulation. Finally, the increase in the theta and alpha band power, especially in the right hemisphere, throughout the temporal series suggest a specialization of right hemisphere in the processing of emotional information.

- Abstract
Introduction: Alveolar bone coverage can be diagnosed through cone beam computed tomography (CBCT) and this information can prevent orthodontic tooth movement beyond the biological limit., Objective: This study evaluated the impact of the bone coverage (BC) diagnosis by CBCT in the orthodontists' planning., Methods: One hundred fifty-nine Brazilian orthodontists suggested treatment plans for six patients at two different times, using two sequential questionnaires. The first questionnaire consisted of extra and intra-oral photographs, one panoramic radiograph; one lateral cephalometric radiograph with Steiner and Tweed analysis, and the patient chief complaint. The second questionnaire included the same presentations of cases with tomographic images and the radiologist's report. The McNemar test assessed the difference between the first and the second treatment plans., Results: In all six cases, most participants changed the treatment plan after evaluating the CBCT images and the radiologist's report (93.7% in case 5, 78.6% in case 4, 74.2% in case 3, 69.8% in case 6, 66% in case 2 and 61% in case 1; p≤0.01)., Conclusion: The evaluation of bone coverage through CBCT images has a substantial impact on the orthodontic diagnosis and planning of the Brazilian orthodontists.
