Abstract

The influence of inositol phosphates and phosphoinositides on the α isoform of the RAC-protein kinase B (RAC/PKB) was studied using purified wild type and mutant kinase preparations and a recombinant pleckstrin homology (PH) domain. Binding of inositol phosphates and phosphoinositides to the PH domain was measured as the quenching of intrinsic tryptophan fluorescence. Inositol phosphates and D3-phosphorylated phosphoinositides bound with affinities of 1-10 μM and 0.5 μM, respectively. Similar values were obtained using RAC/PKB expressed and purified from baculovirus-infected Sf9 cells in the fluorescence assay. The influence of synthetic dioctanoyl derivatives of phosphatidylinositol 3,4-bisphosphate and phosphatidylinositol 3,4,5-trisphosphate on the activity of RAC/PKB purified from transfected COS- 1 cells was studied. Phosphatidylinositol 3,4,5-trisphosphate was found to inhibit the RAC/PKB kinase activity with half-maximal inhibition at 2.5 μM. In contrast, phosphatidylinositol 3,4-bisphosphate stimulated kinase activity (half-maximal stimulation at 2.5 μM). A mutant RAC/PKB protein lacking the PH domain was not affected by D3-phosphorylated phosphoinositides. These results demonstrate that the PH domain of RAC/PKB binds inositol phosphates and phosphoinositides with high affinity, and suggest that the products of the phosphatidylinositide 3-kinase can act as both a membrane anchor and modulator of RAC/PKB activity. The data also provide further evidence for a link between phosphatidylinositide 3-kinase and RAC/PKB regulation.

Abstract

This study characterised and evaluated the phenolic composition and antioxidant activity of the red wine grape Vranac (Vitis vinifera L.) from the southern Serbian vineyard region during grape ripening. Polyphenol composition at different harvest dates was determined by HPLC-DAD analysis. Antioxidant activity was estimated by DPPH assay. The study demonstrates that the Vranac variety represents important sources of dietary antioxidants. The results show that (+)-catechin, (-)-epicatechin and procyanidin dimer B2 were the most prevalent in the seeds, and quercetin and malvidin glucosides in the grape skins. All grape extracts were shown to have high radical-scavenging activity. Strong correlations between radical-scavenging activity and polyphenols suggest that the phenolic composition of the Vranac variety contributes significantly to the antioxidant capacities of grape extracts. During grape ripening there were significant changes in physiological properties and phenolic content, and it is important to determine optimal harvest time, which will ensure grapes with very good quality parameters (in our study at the 30th day after véraison). Abstract

Introduction: Modern dental procedures, especially those in prosthodontics, restore the functional capacity of the orofacial system and improve the patient's appearance. The modern concept of aesthetic dentistry is the most natural appearance of the teeth, and the success of the therapy is closely connected with the respect of anthropometric parameters and the correct ratio of hard and soft oral tissues. Orofacial elements should not be viewed separately from the composition of the face because they form an inseparable whole with it. The aim of this study was to analyze the parameters of the orofacial system that affect the aesthetics of fixed prosthodontic restorations based on literature data and clinical experience. Conclusion: The beauty of a smile is determined by the shape, color and proportions of the upper front teeth. The harmony in creating a smile is not significantly affected by individual numerical values of tooth width and length, but by their mutual relationship. A beautiful smile implies the convergence of the longitudinal axes of the front teeth in relation to the midline of the dental arch, which progresses from the central incisor to the canine. In addition to these criteria, the visibility of the incisors, the midline of the face and dental arch, interdental points and contacts, incisal free spaces as well as the color of the teeth should be taken into account in order to achieve maximum aesthetic effects.

91. G-Protein-coupled receptors and Fcgamma-receptors mediate activation of Akt/protein kinase B in human phagocytes.

Abstract

Activation of the serine/threonine kinase Akt, also called protein kinase B (PKB), was investigated in human neutrophils. Stimulation of the cells with the chemoattractant fMet-Leu-Phe or the chemokines IL-8 and GROalpha leads to the rapid and transient activation of PKB. Maximum PKB activation correlates with the well documented kinetics of respiratory burst and exocytosis. Wortmannin, a selective inhibitor of phosphoinositide 3-kinases (PI 3-kinases) in neutrophils, abrogates PKB activation. Similarly homo and heterotypic cross-linking of FcgammaIIA and FcgammaIIIB causes a transient activation of PKB that is sensitive to wortmannin treatment. Kinase activity measurements in immunoprecipitates from lysates of the myelocytic GM-1 cells or GM-1/CXCR1 cells, which are transfected with the IL-8 receptor 1, confirmed the transient activation of PKB observed in neutrophils. Stimulation of human monocytes with the CC chemokine RANTES (regulated on activation normal T cell expressed and secreted) also results in the activation of PKB. Preincubation of monocytes and neutrophils with Bordetella pertussis toxin inhibits fMet-Leu-Phe and RANTES-stimulated PKB activation, demonstrating that coupling of the receptors to heterotrimeric Gi-protein is required. The data show, that activation of PKB by Gi-protein-coupled receptors is mediated by PI 3-kinase and suggest that PKB is a constituent of neutrophil activating pathways.

92. Frequency dependent selection: I. Rare male phenomenon in D. subobscuradependent on the proportion of Amygenotypes and substrate composition

Abstract

Analysis of the rare male mating advantage in D. subobscura, as a type of frequency dependent selection on maltose and starch media, was done by applying different statistical approaches (?2, cross-product ratio, variance and regression analysis). They reveal that mating occurs at random when proportions of prospective mates are equal, and that mating success of the males homozygous for Amy-locus genotypes (S/S and F/F) depends on their proportion. Regression analysis showed that the F/F males are sexually more active (have higher vigour) than S/S males. Rare male effect is one-sided and appears in F/F males that partake in more heterogamic matings. Comparison of the number of observed and expected homo- and heterogamic matings shows that homogamic matings are more frequent. Multifactorial analysis of variance shows that the number of matings are different for nine pairs of lines and four possible mating types (SfSm, SfFm, FfSm, FfFm). The rare male phenomenon is not dependent on different food composition, but is associated with variations in individual genotypes.

97. Effective population size in Drosophila subobscura: Ecological and molecular approaches

Abstract

The effective population size (Ne) represents the number of individuals that can contribute genes equally to the next generation and is usually smaller then the actual size of a population. The aims of this study were: (i) to assess the Ne for two Drosophila subobscura populations sampled from geographically close, but ecologically and topologically distinct habitats, (ii) to compare the results obtained from two independent approaches for estimating Ne [an ecological (capture-mark-release-recapture) and a molecular (microsatellite data, linkage disequilibrium and heterozygote excess)], and (iii) to obtain a long-term Ne estimation using a variety of mutational models for D. subobscura populations. The ecological method showed that the beech wood population (B) had a significantly larger Ne compared to the oak wood population (O). Observed sex ratio was in favor of females in both populations studied. The microsatellite analysis of populations showed that short-term effective population size in beech wood was larger when compared to population from oak wood, which is in concordance with results obtained by the ecological method. Long term Ne of both natural populations is infinite according to a variety of mutational models. Our results confirmed concordance between ecological and molecular methods in Ne estimation, but also suggested that ecological approach showed less robustness. Molecular approach provides a promising opportunity for more effective monitoring of Ne in Drosophila subobscura. However, the biology, demography and history of the populations may affect different estimators differently so we suggest that ecological and molecular approach should be combined in Ne estimation. 98. Microsatellite variability of Drosophila subobscura populations from the central Balkans

Abstract

Background: Some populations of Drosophila subobscara (Collin) inhabit geographically separate and ecologically distinct habitats of the central Balkans. Hypothesis: Some of the separate populations were genetically isolated in glacial refugia and have subsequently diverged, leading to high genetic diversity. Methods: We sampled five different D. subobscum populations. Using fragment analysis for 11 microsatellite loci, we used standard diversity parameters (expected heterozygosity, allelic richness, allele size range) to estimate population genetic structure and genetic diversity. Results: We found significant differences in the number of alleles, range of allele sizes, and expected heterozygosity between populations from ecologically distinct microhabitats. However, their geographical distance from each other did not contribute to their genetic differences. Analysis of molecular variance showed slight inter-population differentiation (F-ST = 0.00996, P = 0.0215). Conclusions: Microsatellite variability parameters generally match those of other European populations. Drosophila subobscura populations of the Balkan Peninsula likely did not remain isolated in glacial refugia.
