It has been advocated that medical imaging systems and reconstruction algorithms should be assessed and optimized by use of objective measures of image quality that quantify the performance of an observer at specific diagnostic tasks. One important source of variability that can significantly limit observer performance is variation in the objects to-be-imaged. This source of variability can be described by stochastic object models (SOMs). A SOM is a generative model that can be employed to establish an ensemble of to-be-imaged objects with prescribed statistical properties. In order to accurately model variations in anatomical structures and object textures, it is desirable to establish SOMs from experimental imaging measurements acquired by use of a well-characterized imaging system. Deep generative neural networks, such as generative adversarial networks (GANs) hold great potential for this task. However, conventional GANs are typically trained by use of reconstructed images that are influenced by the effects of measurement noise and the reconstruction process. To circumvent this, an AmbientGAN has been proposed that augments a GAN with a measurement operator. However, the original AmbientGAN could not immediately benefit from modern training procedures, such as progressive growing, which limited its ability to be applied to realistically sized medical image data. To circumvent this, in this work, a new Progressive Growing AmbientGAN (ProAmGAN) strategy is developed for establishing SOMs from medical imaging measurements. Medical imaging systems are commonly assessed and optimized by use of objective measures of image quality (IQ). The Ideal Observer (IO) performance has been advocated to provide a figure-of-merit for use in assessing and optimizing imaging systems because the IO sets an upper performance limit among all observers. When joint signal detection and localization tasks are considered, the IO that employs a modified generalized likelihood ratio test maximizes observer performance as characterized by the localization receiver operating characteristic (LROC) curve. Computations of likelihood ratios are analytically intractable in the majority of cases. Therefore, sampling-based methods that employ Markov-Chain Monte Carlo (MCMC) techniques have been developed to approximate the likelihood ratios. However, the applications of MCMC methods have been limited to relatively simple object models. Supervised learning-based methods that employ convolutional neural networks have been recently developed to approximate the IO for binary signal detection tasks. In this paper, the ability of supervised learning-based methods to approximate the IO for joint signal detection and localization tasks is explored. Both background-known-exactly and background-known-statistically signal detection and localization tasks are considered. The considered object models include a lumpy object model and a clustered lumpy model, and the considered measurement noise models include Laplacian noise, Gaussian noise, and mixed Poisson-Gaussian noise. The LROC curves produced by the supervised learning-based method are compared to those produced by the MCMC approach or analytical computation when feasible. The objective assessment of image quality (IQ) has been advocated for the analysis and optimization of medical imaging systems. One method of obtaining such IQ metrics is through a mathematical observer. The Bayesian ideal observer is optimal by definition for signal detection tasks, but is frequently both intractable and non-linear. As an alternative, linear observers are sometimes used for task-based image quality assessment. The optimal linear observer is the Hotelling observer (HO). The computational cost of calculating the HO increases with image size, making a reduction in the dimensionality of the data desirable. Channelized methods have become popular for this purpose, and many competing methods are available for computing efficient channels. In this work, a novel method for learning channels using an autoencoder (AE) is presented. AEs are a type of artificial neural network (ANN) that are frequently employed to learn concise representations of data to reduce dimensionality. Modifying the traditional AE loss function to focus on task-relevant information permits the development of efficient AE-channels. These AE-channels were trained and tested on a variety of signal shapes and backgrounds to evaluate their performance. In the experiments, the AE-learned channels were competitive with and frequently outperformed other state-of-the-art methods for approximating the HO. The performance gains were greatest for the datasets with a small number of training images and noisy estimates of the signal image. Medical imaging systems are commonly assessed by use of objective image quality measures. Supervised deep learning methods have been investigated to implement numerical observers for task-based image quality assessment. However, labeling large amounts of experimental data to train deep neural networks is tedious, expensive, and prone to subjective errors. Computer-simulated image data can potentially be employed to circumvent these issues; however, it is often difficult to computationally model complicated anatomical structures, noise sources, and the response of real world imaging systems. Hence, simulated image data will generally possess physical and statistical differences from the experimental image data they seek to emulate. Within the context of machine learning, these differences between the sets of two images is referred to as domain shift. In this study, we propose and investigate the use of an adversarial domain adaptation method to mitigate the deleterious effects of domain shift between simulated and experimental image data for deep learning-based numerical observers (DL-NOs) that are trained on simulated images but applied to experimental ones. In the proposed method, a DL-NO will initially be trained on computer-simulated image data and subsequently adapted for use with experimental image data, without the need for any labeled experimental images. As a proof of concept, a binary signal detection task is considered. Medical image reconstruction is typically an ill-posed inverse problem. In order to address such ill-posed problems, the prior distribution of the sought after object property is usually incorporated by means of some sparsity-promoting regularization. Recently, prior distributions for images estimated using generative adversarial networks (GANs) have shown great promise in regularizing some of these image reconstruction problems. In this work, we apply an image-adaptive GAN-based reconstruction method (IAGAN) to reconstruct high fidelity images from incomplete medical imaging data. It is observed that the IAGAN method can potentially recover fine structures in the object that are relevant for medical diagnosis but may be oversmoothed in reconstructions with traditional sparsity-promoting regularization., Comment: SPIE Medical Imaging 2020

The Ideal Observer (IO) performance has been advocated when optimizing medical imaging systems for signal detection tasks. However, analytical computation of the IO test statistic is generally intractable. To approximate the IO test statistic, sampling-based methods that employ Markov-Chain Monte Carlo (MCMC) techniques have been developed. However, current applications of MCMC techniques have been limited to several object models such as a lumpy object model and a binary texture model, and it remains unclear how MCMC methods can be implemented with other more sophisticated object models. Deep learning methods that employ generative adversarial networks (GANs) hold great promise to learn stochastic object models (SOMs) from image data. In this study, we described a method to approximate the IO by applying MCMC techniques to SOMs learned by use of GANs. The proposed method can be employed with arbitrary object models that can be learned by use of GANs, thereby the domain of applicability of MCMC techniques for approximating the IO performance is extended. In this study, both signal-known-exactly (SKE) and signal-known-statistically (SKS) binary signal detection tasks are considered. The IO performance computed by the proposed method is compared to that computed by the conventional MCMC method. The objective optimization of medical imaging systems requires full characterization of all sources of randomness in the measured data, which includes the variability within the ensemble of objects to-be-imaged. This can be accomplished by establishing a stochastic object model (SOM) that describes the variability in the class of objects to-be-imaged. Generative adversarial networks (GANs) can be potentially useful to establish SOMs because they hold great promise to learn generative models that describe the variability within an ensemble of training data. However, because medical imaging systems record imaging measurements that are noisy and indirect representations of object properties, GANs cannot be directly applied to establish stochastic models of objects to-be-imaged. To address this issue, an augmented GAN architecture named AmbientGAN was developed to establish SOMs from noisy and indirect measurement data. However, because the adversarial training can be unstable, the applicability of the AmbientGAN can be potentially limited. In this work, we propose a novel training strategy---Progressive Growing of AmbientGANs (ProAGAN)---to stabilize the training of AmbientGANs for establishing SOMs from noisy and indirect imaging measurements. An idealized magnetic resonance (MR) imaging system and clinical MR brain images are considered. It is widely accepted that optimization of medical imaging system performance should be guided by task-based measures of image quality (IQ). Task-based measures of IQ quantify the ability of an observer to perform a specific task such as detection or estimation of a signal (e.g., a tumor). For binary signal detection tasks, the Bayesian Ideal Observer (IO) sets an upper limit of observer performance and has been advocated for use in optimizing medical imaging systems and data-acquisition designs. Except in special cases, determination of the IO test statistic is analytically intractable. Markov-chain Monte Carlo (MCMC) techniques can be employed to approximate IO detection performance, but their reported applications have been limited to relatively simple object models. In cases where the IO test statistic is difficult to compute, the Hotelling Observer (HO) can be employed. To compute the HO test statistic, potentially large covariance matrices must be accurately estimated and subsequently inverted, which can present computational challenges. This work investigates supervised learning-based methodologies for approximating the IO and HO test statistics. Convolutional neural networks (CNNs) and single-layer neural networks (SLNNs) are employed to approximate the IO and HO test statistics, respectively. Numerical simulations were conducted for both signal-known-exactly (SKE) and signal-known-statistically (SKS) signal detection tasks. The performances of the supervised learning methods are assessed via receiver operating characteristic (ROC) analysis and the results are compared to those produced by use of traditional numerical methods or analytical calculations when feasible. It is widely accepted that optimization of imaging system performance should be guided by task-based measures of image quality (IQ). It has been advocated that imaging hardware or data-acquisition designs should be optimized by use of an ideal observer (IO) that exploits full statistical knowledge of the measurement noise and class of objects to be imaged, without consideration of the reconstruction method. In practice, accurate and tractable models of the complete object statistics are often difficult to determine. Moreover, in imaging systems that employ compressive sensing concepts, imaging hardware and sparse image reconstruction are innately coupled technologies. In this work, a sparsity-driven observer (SDO) that can be employed to optimize hardware by use of a stochastic object model describing object sparsity is described and investigated. The SDO and sparse reconstruction method can therefore be "matched" in the sense that they both utilize the same statistical information regarding the class of objects to be imaged. To efficiently compute the SDO test statistic, computational tools developed recently for variational Bayesian inference with sparse linear models are adopted. The use of the SDO to rank data-acquisition designs in a stylized example as motivated by magnetic resonance imaging (MRI) is demonstrated. Photoacoustic computed tomography (PACT), also known as optoacoustic tomography, is an emerging imaging technique that holds great promise for biomedical imaging. PACT is a hybrid imaging method that can exploit the strong endogenous contrast of optical methods along with the high spatial resolution of ultrasound methods. In its canonical form that is addressed in this article, PACT seeks to estimate the photoacoustically-induced initial pressure distribution within the object. Image reconstruction methods are employed to solve the acoustic inverse problem associated with the image formation process. When an idealized imaging scenario is considered, analytic solutions to the PACT inverse problem are available; however, in practice, numerous challenges exist that are more readily addressed within an optimization-based, or iterative, image reconstruction framework. In this article, the PACT image reconstruction problem is reviewed within the context of modern optimization-based image reconstruction methodologies. Imaging models that relate the measured photoacoustic wavefields to the sought-after object function are described in their continuous and discrete forms. The basic principles of optimization-based image reconstruction from discrete PACT measurement data are presented, which includes a review of methods for modeling the PACT measurement system response and other important physical factors. Non-conventional formulations of the PACT image reconstruction problem, in which acoustic parameters of the medium are concurrently estimated along with the PACT image, are also introduced and reviewed.

Accurately counting cells in microscopic images is important for medical diagnoses and biological studies, but manual cell counting is very tedious, time-consuming, and prone to subjective errors, and automatic counting can be less accurate than desired. To improve the accuracy of automatic cell counting, we propose here a novel method that employs deeply-supervised density regression. A fully convolutional neural network (FCNN) serves as the primary FCNN for density map regression. Innovatively, a set of auxiliary FCNNs are employed to provide additional supervision for learning the intermediate layers of the primary CNN to improve network performance. In addition, the primary CNN is designed as a concatenating framework to integrate multi-scale features through shortcut connections in the network, which improves the granularity of the features extracted from the intermediate CNN layers and further supports the final density map estimation., Comment: SPIE medical imaging 2019 oral presentation

Accurate cell counting in microscopic images is important for medical diagnoses and biological studies. However, manual cell counting is very time-consuming, tedious, and prone to subjective errors. We propose a new density regression-based method for automatic cell counting that reduces the need to manually annotate experimental images. A supervised learning-based density regression model (DRM) is trained with annotated synthetic images (the source domain) and their corresponding ground truth density maps. A domain adaptation model (DAM) is built to map experimental images (the target domain) to the feature space of the source domain. By use of the unsupervised learning-based DAM and supervised learning-based DRM, a cell density map of a given target image can be estimated, from which the number of cells can be counted. Optical coherence tomography (OCT) can provide high-resolution cross-sectional images for analyzing superficial plaques in coronary arteries. Commonly, plaque characterization using intra-coronary OCT images is performed manually by expert observers. This manual analysis is time consuming and its accuracy heavily relies on the experience of human observers. Traditional machine learning based methods, such as the least squares support vector machine and random forest methods, have been recently employed to automatically characterize plaque regions in OCT images. Several processing steps, including feature extraction, informative feature selection, and final pixel classification, are commonly used in these traditional methods. Therefore, the final classification accuracy can be jeopardized by error or inaccuracy within each of these steps. In this study, we proposed a convolutional neural network (CNN) based method to automatically characterize plaques in OCT images. Unlike traditional methods, our method uses the image as a direct input and performs classification as a single-step process. The experiments on 269 OCT images showed that the average prediction accuracy of CNN-based method was 0.866, which indicated a great promise for clinical translation., Comment: SPIE 2018

Photoacoustic computed tomography (PACT) is an emerging imaging modality that exploits optical contrast and ultrasonic detection principles to form images of the photoacoustically induced initial pressure distribution within tissue. The PACT reconstruction problem corresponds to an inverse source problem in which the initial pressure distribution is recovered from measurements of the radiated wavefield. A major challenge in transcranial PACT brain imaging is compensation for aberrations in the measured data due to the presence of the skull. Ultrasonic waves undergo absorption, scattering and longitudinal-to-shear wave mode conversion as they propagate through the skull. To properly account for these effects, a wave-equation-based inversion method should be employed that can model the heterogeneous elastic properties of the skull. In this work, a forward model based on a finite-difference time-domain discretization of the three-dimensional elastic wave equation is established and a procedure for computing the corresponding adjoint of the forward operator is presented. Massively parallel implementations of these operators employing multiple graphics processing units (GPUs) are also developed. The developed numerical framework is validated and investigated in computer-simulation and experimental phantom studies whose designs are motivated by transcranial PACT applications.

An approach to incorporate deep learning within an iterative image reconstruction framework to reconstruct images from severely incomplete measurement data is presented. Specifically, we utilize a convolutional neural network (CNN) as a quasi-projection operator within a least squares minimization procedure. The CNN is trained to encode high level information about the class of images being imaged; this information is utilized to mitigate artifacts in intermediate images produced by use of an iterative method. The structure of the method was inspired by the proximal gradient descent method, where the proximal operator is replaced by a deep CNN and the gradient descent step is generalized by use of a linear reconstruction operator. It is demonstrated that this approach improves image quality for several cases of limited-view image reconstruction and that using a CNN in an iterative method increases performance compared to conventional image reconstruction approaches. We test our method on several limited-view image reconstruction problems. Qualitative and quantitative results demonstrate state-of-the-art performance.

Photoacoustic computed tomography (PACT) is an emerging computed imaging modality that exploits optical contrast and ultrasonic detection principles to form images of the absorbed optical energy density within tissue. If the object possesses spatially variant acoustic properties that are unaccounted for by the reconstruction method, the estimated image can contain distortions. While reconstruction methods have recently been developed to compensate for this effect, they generally require the object's acoustic properties to be known a priori. To circumvent the need for detailed information regarding an object's acoustic properties, we previously proposed a half-time reconstruction method for PACT. A half-time reconstruction method estimates the PACT image from a data set that has been temporally truncated to exclude the data components that have been strongly aberrated. However, this method can be improved upon when the approximate sizes and locations of isolated heterogeneous structures, such as bones or gas pockets, are known. To address this, we investigate PACT reconstruction methods that are based on a variable data truncation (VDT) approach. The VDT approach represents a generalization of the half-time approach, in which the degree of temporal truncation for each measurement is determined by the distance between the corresponding ultrasonic transducer location and the nearest known bone or gas void location. Computer-simulated and experimental data are employed to demonstrate the effectiveness of the approach in mitigating artifacts due to acoustic heterogeneities.

Ultrasound computed tomography (USCT) holds great promise for breast cancer screening. Waveform inversion-based image reconstruction methods account for higher order diffraction effects and can produce high-resolution USCT images, but are computationally demanding. Recently, a source encoding technique was combined with stochastic gradient descent to greatly reduce image reconstruction times. However, this method bundles the stochastic data fidelity term with the deterministic regularization term. This limitation can be overcome by replacing stochastic gradient descent (SGD) with a structured optimization method, such as the regularized dual averaging (RDA) method, that exploits knowledge of the composition of the cost function. In this work, the dual averaging method is combined with source encoding techniques to improve the effectiveness of regularization while maintaining the reduced reconstruction times afforded by source encoding. It is demonstrated that each iteration can be decomposed into a gradient descent step based on the data fidelity term and a proximal update step corresponding to the regularization term. Furthermore, the regularization term is never explicitly differentiated, allowing non-smooth regularization penalties to be naturally incorporated. The wave equation is solved by use of a time-domain method. The effectiveness of this approach is demonstrated through computer-simulation and experimental studies. The results suggest that the dual averaging method can produce images with less noise and comparable resolution to those obtained by use of stochastic gradient descent.

Optoacoustic tomography (OAT), also known as photoacoustic tomography, is a rapidly emerging hybrid imaging technique that possesses great potential for a wide range of biomedical imaging applications. In OAT, a laser is employed to illuminate the tissue of interest and acoustic signals are produced via the photoacoustic effect. From these data, an estimate of the distribution of the absorbed optical energy density within the tissue is reconstructed, referred to as the object function. This quantity is defined, in part, by the distribution of light fluence within the tissue that is established by the laser source. When performing three-dimensional imaging of large objects, such as a female human breast, it can be difficult to achieve a relatively uniform coverage of light fluence within the volume of interest when the position of the laser source is fixed. To circumvent this, researchers have proposed illumination schemes in which the relative position of the laser source and ultrasound probe is fixed, and both are rotated together to acquire a tomographic data set. A problem with this rotating-illumination scheme is that the tomographic data are inconsistent; namely, the acoustic data recorded at each tomographic view angle (i.e., probe position) are produced by a distinct object function. In this work, the impact of this data inconsistency on image reconstruction accuracy is investigated systematically. This is accomplished by use of computer-simulation studies and application of mathematical results from the theory of microlocal analysis. These studies specify the set of image discontinuities that can be stably reconstructed with a non-stationary optical illumination set-up. The study also includes a comparison of the ability of iterative and analytic image reconstruction methods to mitigate artifacts attributable to the data inconsistency.

Photoacoustic computed tomography (PACT) is a rapidly emerging bioimaging modality that seeks to reconstruct an estimate of the absorbed optical energy density within an object. Conventional PACT image reconstruction methods assume a constant speed-of-sound (SOS), which can result in image artifacts when acoustic aberrations are significant. It has been demonstrated that incorporating knowledge of an object's SOS distribution into a PACT image reconstruction method can improve image quality. However, in many cases, the SOS distribution cannot be accurately and/or conveniently estimated prior to the PACT experiment. Because variations in the SOS distribution induce aberrations in the measured photoacoustic wavefields, certain information regarding an object's SOS distribution is encoded in the PACT measurement data. Based on this observation, a joint reconstruction (JR) problem has been proposed in which the SOS distribution is concurrently estimated along with the sought-after absorbed optical energy density from the photoacoustic measurement data. A broad understanding of the extent to which the JR problem can be accurately and reliably solved has not been reported. In this work, a series of numerical experiments is described that elucidate some important properties of the JR problem that pertain to its practical feasibility. To accomplish this, an optimization-based formulation of the JR problem is developed that yields a non-linear iterative algorithm that alternatingly updates the two image estimates. Heuristic analytic insights into the reconstruction problem are also provided. Photoacoustic computed tomography (PACT) is an emerging computed imaging modality that exploits optical contrast and ultrasonic detection principles to form images of the absorbed optical energy density within tissue. When the imaging system employs conventional piezoelectric ultrasonic transducers, the ideal photoacoustic (PA) signals are degraded by the transducers' acousto-electric impulse responses (EIRs) during the measurement process. If unaccounted for, this can degrade the accuracy of the reconstructed image. In principle, the effect of the EIRs on the measured PA signals can be ameliorated via deconvolution; images can be reconstructed subsequently by application of a reconstruction method that assumes an idealized EIR. Alternatively, the effect of the EIR can be incorporated into an imaging model and implicitly compensated for during reconstruction. In either case, the efficacy of the correction can be limited by errors in the assumed EIRs. In this work, a joint optimization approach to PACT image reconstruction is proposed for mitigating errors in reconstructed images that are caused by use of an inaccurate EIR. The method exploits the bi-linear nature of the imaging model and seeks to refine the measured EIR during the process of reconstructing the sought-after absorbed optical energy density. Ultrasound computed tomography (USCT) holds great promise for improving the detection and management of breast cancer. Because they are based on the acoustic wave equation, waveform inversion-based reconstruction methods can produce images that possess improved spatial resolution properties over those produced by ray-based methods. However, waveform inversion methods are computationally demanding and have not been applied widely in USCT breast imaging. In this work, source encoding concepts are employed to develop an accelerated USCT reconstruction method that circumvents the large computational burden of conventional waveform inversion methods. This method, referred to as the waveform inversion with source encoding (WISE) method, encodes the measurement data using a random encoding vector and determines an estimate of the sound speed distribution by solving a stochastic optimization problem by use of a stochastic gradient descent algorithm. Both computer-simulation and experimental phantom studies are conducted to demonstrate the use of the WISE method. The results suggest that the WISE method maintains the high spatial resolution of waveform inversion methods while significantly reducing the computational burden.

Optoacoustic tomography (OAT), also known as photoacoustic tomography, is an emerging computed biomedical imaging modality that exploits optical contrast and ultrasonic detection principles. Iterative image reconstruction algorithms that are based on discrete imaging models are actively being developed for OAT due to their ability to improve image quality by incorporating accurate models of the imaging physics, instrument response, and measurement noise. In this work, we investigate the use of discrete imaging models based on Kaiser-Bessel window functions for iterative image reconstruction in OAT. A closed-form expression for the pressure produced by a Kaiser-Bessel function is calculated, which facilitates accurate computation of the system matrix. Computer-simulation and experimental studies are employed to demonstrate the potential advantages of Kaiser-Bessel function-based iterative image reconstruction in OAT.

Purpose: Optoacoustic tomography (OAT) is inherently a three-dimensional (3D) inverse problem. However, most studies of OAT image reconstruction still employ two-dimensional (2D) imaging models. One important reason is because 3D image reconstruction is computationally burdensome. The aim of this work is to accelerate existing image reconstruction algorithms for 3D OAT by use of parallel programming techniques. Methods: Parallelization strategies are proposed to accelerate a filtered backprojection (FBP) algorithm and two different pairs of projection/backprojection operations that correspond to two different numerical imaging models. The algorithms are designed to fully exploit the parallel computing power of graphic processing units (GPUs). In order to evaluate the parallelization strategies for the projection/backprojection pairs, an iterative image reconstruction algorithm is implemented. Computer-simulation and experimental studies are conducted to investigate the computational efficiency and numerical accuracy of the developed algorithms. Results: The GPU implementations improve the computational efficiency by factors of 1, 000, 125, and 250 for the FBP algorithm and the two pairs of projection/backprojection operators, respectively. Accurate images are reconstructed by use of the FBP and iterative image reconstruction algorithms from both computer-simulated and experimental data. Conclusions: Parallelization strategies for 3D OAT image reconstruction are proposed for the first time. These GPU-based implementations significantly reduce the computational time for 3D image reconstruction, complementing our earlier work on 3D OAT iterative image reconstruction.

Existing approaches to image reconstruction in photoacoustic computed tomography (PACT) with acoustically heterogeneous media are limited to weakly varying media, are computationally burdensome, and/or cannot effectively mitigate the effects of measurement data incompleteness and noise. In this work, we develop and investigate a discrete imaging model for PACT that is based on the exact photoacoustic (PA) wave equation and facilitates the circumvention of these limitations. A key contribution of the work is the establishment of a procedure to implement a matched forward and backprojection operator pair associated with the discrete imaging model, which permits application of a wide-range of modern image reconstruction algorithms that can mitigate the effects of data incompleteness and noise. The forward and backprojection operators are based on the k-space pseudospectral method for computing numerical solutions to the PA wave equation in the time domain. The developed reconstruction methodology is investigated by use of both computer-simulated and experimental PACT measurement data.

Photoacoustic computed tomography (PACT), also known as optoacoustic tomography, is an emerging imaging modality that has great potential for a wide range of biomedical imaging applications. In this Note, we derive a hybrid reconstruction formula that is mathematically exact and operates on a data function that is expressed in the temporal frequency and spatial domains. This formula explicitly reveals new insights into how the spatial frequency components of the sought-after object function are determined by the temporal frequency components of the data function measured with a circular or spherical measurement geometry in two- and three-dimensional implementations of PACT, respectively. The structure of the reconstruction formula is surprisingly simple compared with existing Fourier-domain reconstruction formulae. Iterative image reconstruction algorithms for optoacoustic tomography (OAT), also known as photoacoustic tomography, have the ability to improve image quality over analytic algorithms due to their ability to incorporate accurate models of the imaging physics, instrument response, and measurement noise. However, to date, there have been few reported attempts to employ advanced iterative image reconstruction algorithms for improving image quality in three-dimensional (3D) OAT. In this work, we implement and investigate two iterative image reconstruction methods for use with a 3D OAT small animal imager: namely, a penalized least-squares (PLS) method employing a quadratic smoothness penalty and a PLS method employing a total variation norm penalty. The reconstruction algorithms employ accurate models of the ultrasonic transducer impulse responses. Experimental data sets are employed to compare the performances of the iterative reconstruction algorithms to that of a 3D filtered backprojection (FBP) algorithm. By use of quantitative measures of image quality, we demonstrate that the iterative reconstruction algorithms can mitigate image artifacts and preserve spatial resolution more effectively than FBP algorithms. These features suggest that the use of advanced image reconstruction algorithms can improve the effectiveness of 3D OAT while reducing the amount of data required for biomedical applications.
