Friedreich ataxia is a hereditary neurodegenerative disorder resulting from reduced levels of the protein frataxin due to an expanded GAA repeat in the FXN gene. This deficiency causes progressive degeneration of specific neuronal populations in the cerebellum and the consequent loss of movement coordination and equilibrium, which are some of the main symptoms observed in affected individuals. Like in other neurodegenerative diseases, previous studies suggest that glial cells could be involved in the neurodegenerative process and disease progression in patients with Friedreich ataxia.In this work, we followed and characterized the progression of changes in the cerebellar cortex in the latest version of Friedreich ataxia humanized mouse model, YG8–800 (Fxnnull:YG8s(GAA)>800), which carries a human FXN transgene containing >800 GAA repeats.Comparative analyses of behavioral, histopathological, and biochemical parameters were conducted between the control strain Y47R and YG8–800 mice at different time points. Our findings revealed that YG8–800 mice exhibit an ataxic phenotype characterized by poor motor coordination, decreased body weight, cerebellar atrophy, neuronal loss, and changes in synaptic proteins. Additionally, early activation of glial cells, predominantly astrocytes and microglia, was observed preceding neuronal degeneration, as was increased expression of key proinflammatory cytokines and downregulation of neurotrophic factors.Together, our results show that the YG8–800 mouse model exhibits a stronger phenotype than previous experimental murine models, reliably recapitulating some of the features observed in humans. Accordingly, this humanized model could represent a valuable tool for studying Friedreich ataxia molecular disease mechanisms and for preclinical evaluation of possible therapies.

Abstract Background To assess whether changes in breakfast and water consumption during the first full school year after the emergence of the COVID-19 pandemic varied based on sex/gender, race/ethnicity, and socioeconomic status among Canadian adolescents. Methods Prospective annual survey data collected pre- (October 2019-March 2020) and post-COVID-19 onset (November 2020-June 2021) the Cannabis, Obesity, Mental health, Physical activity, Alcohol, Smoking, and Sedentary behaviour (COMPASS) study. The sample consisted of 8,128 students; mean (SD) age = 14.2 (1.3) years from a convenience sample of 41 Canadian secondary schools. At both timepoints self-reported breakfast and water consumption were dichotomized as daily or not. Multivariable logistic generalized estimating equations with school clustering were used to estimate differences in maintenance/adoption of daily consumption post-COVID-19 based on demographic factors, while controlling for pre-COVID-19 behaviour. Results Adjusted odds ratios (AOR) with 95% confidence intervals are reported. Females (AOR = 0.71 [0.63, 0.79]) and lower socioeconomic status individuals (AORLowest:Highest=0.41 [0.16, 1.00]) were less likely to maintain/adopt daily breakfast consumption than male and higher socioeconomic status peers in the 2020–2021 school year. Searching for natural alternatives to synthetic fermentation activators has led to the study of bee pollen as a natural alcoholic fermentation activator. This study evaluated the potential of different bee pollen extracts (0.25 g/L) as activators in a Palomino Fino grape must. By analysing the composition of each extract, it was possible to identify the specific bee pollen fractions with the highest efficacy for activating alcoholic fermentation. Four extracts were obtained through sequential extraction using various organic solvents of increasing polarity (hexane, acetone, ethanol, and water), and their compositions were characterised. The effect of each extract was evaluated by monitoring the viable yeast populations and fermentation kinetics throughout the alcoholic fermentation process, along with the physicochemical and colour characterisation of the white wines obtained. The bee pollen fraction extracted with hexane, which was rich in long-chain fatty acids, significantly increased the maximum yeast populations and improved the fermentation kinetics. However, the extracts rich in polyphenolic compounds exhibited slower fermentation rates. Based on the obtained results, the lipid fraction of bee pollen extracted with hexane may be responsible for its ability to activate alcoholic fermentation.

The SARS-CoV-2 pandemic significantly affected the epidemiology of Streptococcus pyogenes, a pathogen associated with various clinical presentations such as pharyngitis, scarlet fever, and invasive diseases. This study analyzed the incidence and characteristics of S. pyogenes infections between 2018 and 2023, examining 915 cases categorized as either respiratory or non-respiratory. Respiratory infections predominantly affected children, accounting for 76% of cases, with a median age of 5 [3, 8] years, while non-respiratory infections were more common in adults, with a median age of 46.5 [34, 64] years. Invasive respiratory infections, such as pneumonia and empyema, were more frequent in children (54.8%), whereas invasive non-respiratory infections, such as primarily cellulitis, were predominantly seen in adults (90.5%). A sharp decline in S. pyogenes infections was observed during the pandemic, with respiratory cases decreasing tenfold in 2020 compared to the previous year, and non-respiratory cases experiencing a twofold reduction. However, infection rates returned to pre-pandemic levels by 2022 and 2023, with a notable resurgence of invasive respiratory infections in children following a public health alert in the United Kingdom in late 2022. These findings highlight distinct infection patterns between pediatric and adult populations and emphasize the significant impact of the pandemic on respiratory infections, particularly in children.

Wave setup is a physical process that induces a temporal increase of the mean water level due to wave dissipation by bottom friction and breaking in the surf zone, extending over tens to hundreds of meters in the cross-shore direction. Wave setup contribution to coastal sea level solely induced by wind and atmospheric effects can increase by more than 100% under extreme events and conditions favoring its formation. It is therefore crucial to consider this phenomenon when assessing sea-level-related coastal hazards. Previous studies estimated the wave setup effect by means of numerical modeling and empirical formulations at regional and global scale. Such analyses require either high computational capacity to implement high-resolution numerical models over large domains, and/or accurate information on coastal morphological features from global or regional databases. Although the Mediterranean Sea is a fetch-limited environment, waves generated from extra-tropical cyclones are powerful enough for wave setup to develop, and subsequently for a potential significant wave setup contribution to extreme coastal sea level. Through the use of both numerical and empirical methods, we investigate the uncertainty associated to wave setup representation on the frequency and magnitude of coastal extreme sea levels occurring on sandy beaches in the Mediterranean Sea. Wave setup values are compared at beach scale between process-based modeling and empirical approaches, showing highly variable results. We also quantify the impact of wave setup on return levels of coastal sea level extremes using reconstructed sea levels. We employ various methods to calculate the wave setup component. Results show high spatial dispersion, with clear differences between the numerical and empirical approaches, especially in regions prone to the development of energetic waves. The total inter-method dispersion of 100-year return levels is often higher than 30 cm for average values of 62.4 cm. We emphasize the important limitations related to wave setup modeling (i.e., its underestimation) at large scale, and call for caution when applying empirical formulations (generally developed from local studies) at regional to global scale, which can lead to unrealistic wave setup values.

En este artículo se presenta el desarrollo de la estratégica didáctica Aprendizaje Basado en Proyectos para motivar la adquisición y aplicación del conocimiento de estudiantes de nivel superior del Instituto Tecnológico Superior de Xalapa (ITSX), con apoyo de la Facultad de Biología de la Universidad Veracruzana y el INECOL, A.C. Esta propuesta tuvo como objetivo desarrollar habilidades del saber y el quehacer científico en estudiantes de nivel superior del ITSX a partir de la elaboración de productos de plantas medicinales. El trabajo fue estructurado bajo el enfoque del ABP durante agosto 2021 a julio 2022. La investigación se llevó a cabo en tres etapas: documentación, indagación, propagación de las plantas, experimentación y presentación de resultados. Mediante este trabajo los estudiantes desarrollaron residencias profesionales en Ingeniería Bioquímica, Ingeniería en Sistemas Computacionales, Ingeniería Mecatrónica. A través de la evaluación de sus reportes finales de residencias, así como una indagación anecdótica se evidenció que los estudiantes desarrollaron habilidades científicas y aplicaron conocimiento teórico para el desarrollo de la parte experimental, fortalecieron el análisis crítico al relacionar la teoría con la práctica, además de desarrollar valores como la responsabilidad, compromiso y respeto entre sus compañeros.

The right hemisphere of the brain is often referred to as the non-dominant hemisphere. Though this is meant to highlight the specialized role of the left hemisphere in language, the use of this term runs the risk of oversimplifying or minimizing the essential functions of the right hemisphere. There is accumulating evidence from functional MRI, clinical lesion studies, and intraoperative mapping data that implicate the right hemisphere in a diverse array of cognitive functions, including visuospatial functions, attentional processes, and social cognitive functions. Neuropsychological deficits following right hemisphere resections are well-documented, but there is a general paucity of literature focusing on how to best map these functions during awake brain surgery to minimize such deficits. To address this gap in the literature, a systematic review was conducted to examine the cognitive and emotional processes associated with the right hemisphere and the neuropsychological tasks frequently used for mapping the right hemisphere during awake brain tumor surgery. It was found that the most employed tests to assess language and speech functions in patients with lesions in the right cerebral hemisphere were the naming task and the Pyramids and Palm Trees Test (PPTT). Spatial cognition was typically evaluated using the line bisection task, while social cognition was assessed through the Reading the Mind in the Eyes (RME) test. Dual-tasking and the movement of the upper and lower limbs were the most frequently used methods to evaluate motor/sensory functions. Executive functions were typically assessed using the N-back test and Stroop test. To the best of our knowledge, this is the first comprehensive review to help provide guidance on the cognitive functions most at risk and methods to map such functions during right awake brain surgery.Systematic Review RegistrationPROSPERO database [CRD42023483324].

Background: Single anastomosis duodenoileal bypass with sleeve gastrectomy (SADI-S) is a predominantly malabsorptive technique that has shown excellent results in morbid obese patients. The aim of this study is to establish a rodent model modifying the SADI-S technique by performing a proximal duodenojejunal anastomosis. This model can be useful for the study of glucose metabolism without malabsorption observed after the SADI-S technique. Methods: Goto-Kakizaki rats, a genetic model of non-obese and non-hypertensive type 2 diabetes mellitus, that develop hyperglycemia at an early age was used. Surgery consisted in a sleeve gastrectomy, duodenojejunal anastomosis and duodenal exclusion using three different techniques: duodenal transection (DT), duodenal ligation with hem-o-lock (DLH), and duodenal ligation with suture (DLS). Surgery time, weight loss, morbidity and mortality were recorded. Results: A total of 16 animals were subjected to surgical intervention and overall mortality was 25 %, with the DT group showing the highest mortality rate (42.9 %). No differences were observed among groups in terms of weight loss. Conclusion: The surgical technique described in this work is feasible and reproducible. Weight loss is comparable regardless of the technique used for duodenal exclusion.

Abstract Background Very little research has examined how perceptions of cannabis access among underage youth in Canada have changed since cannabis was legalized and since the onset of the COVID-19 pandemic. As such, this paper examines the effect of the early and ongoing stages of the COVID-19 pandemic period on youth perceptions of cannabis access over time since the onset of the Cannabis Act in 2018 in a large sample of Canadian youth. Methods Using data from the COMPASS study (T1:2018/19, T2:2019/20, T3:2020/21), we used both repeat cross-sectional data [T1 (n = 38,890), T2 (n = 24,109), and T3 (n = 22,795)] to examine overall trends in perceptions of cannabis access, and sequential cohort longitudinal data [n = 4,677 students linked from T1 to T3] to examine the differential changes in perceptions of cannabis access among students over time. Results In the cross-sectional sample, the frequency of students reporting that cannabis was easy to access decreased by 26.7% from T1 (51.0%) to T3 (37.4%), although respondents who have used cannabis were more likely to report access was easy. In the longitudinal sample, perceptions of cannabis access being easy increased over time, especially among cannabis users. Perceived ease of access appears to have been slightly impeded during the initial pandemic period but rebounded during the ongoing pandemic period. Conclusions While the prevalence of youth reporting that cannabis is easy to access has declined since legalization and throughout the early and ongoing pandemic periods, a substantial number of underage youth continue to report that cannabis is easy to access. This suggest that there is an ongoing need for continued cannabis control efforts to address this issue.

Background: The main objectives of this study were to analyze the effect of early specialization in swimming and to observe the general patterns of success of two different sport specialization models [Spanish (SPA) and United States of American (USA) swimmers] participating in World Championships (WCs) and Olympic Games (OGs) between the years 2006 and 2021 of all swimming strokes and distances. Methods: Descriptive analyses and contingency tables were examined for all the variables. Explanatory models of the z scores were estimated from age depending on the events’ distances and strokes. Quadratic regression models were developed to capture the behavior of the variable time with parabolic functions, and the significance of the global model and the predictor variables (age) were also evaluated. In addition, the optimal age (peak performance) as well as the curvature of the model were analyzed. These models were compared between SPA and USA swimmers. Results: The results showed that the main differences in the patterns to success between SPA and USA were the earlier participation of USA swimmers in high-level competitions, as well as the greater number of participants for all the strokes, events, genders, and competitions. Age peak performance in short distances was lower for Spanish swimmers, obtaining the opposite situation for long distances. Conclusions: Being a finalist in junior WCs did not influence success in the finals of the senior WCs and OGs. Main differences in general patterns of success between SPA and USA showed younger swimmers participating in short-distance events, backstroke, and butterfly for USA swimmers and older swimmers taking part in butterfly and short-distance events for SPA.
