This paper discusses the potential of tert-butylhydroquinone (TBHQ), butylated hydroxyanisole (BHA) and alpha-tocopherol (α-T) in preventing palm biodiesel oxidation over an extended storage period of twelve weeks. A combination of Fourier transform infrared spectroscopy (FTIR), gas chromatography-mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC) analyses were carried out. The FTIR spectra showed no significant changes to the carbonyl band at 1740 cm−1 across the storage period for fuels with additives, but the control had increased band intensity, which implied that additives prevented the formation of byproducts which have carbonyl groups or cyclic ether rings. From GC-MS analyses, lower transmittance at C18 methyl esters region was evident of methyl oleate, methyl linoleate and methyl linolenate being consumed to the point where their concentrations dropped below those of methyl palmitate in the sample without additives. Furthermore, the analyses indicated that 3-octyl-oxiraneoctanoic acid ME, E−10,13,13-trimethyl-11-tetradecen-1-ol acetate and 17-octadecynoic acid ME were the major oxidation byproducts. Traces of acetal, carboxylic acid, oxirane and alkylnoic acid were also detected. It was shown through HPLC analyses that TBHQ, BHA and α-T were consumed with α-T samples having the highest consumption compared to TBHQ and BHA. Overall, the findings supported the important role of additives in stabilizing the fuel and preventing oxidation during extended storage.

The excessive and uncontrolled use of pyrethroids, such as cypermethrin (CP), for pest control in Nigeria could adversely affect humans. This study aimed to investigate the oxidative stress response to cypermethrin exposure, focusing on measuring the parameters (i.e., malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT)) and the potential therapeutic effects of single and co-administration of ascorbate and alpha-tocopherol. The lungs and hearts of the animals were histologically examined for cypermethrin-induced cytopathic changes. Twenty-five adult male Wistar rats weighing 180–200 g were randomly assigned to five groups, each consisting of five animals. Group I was the control group that was not subjected to any treatment. Group II was orally exposed to cypermethrin at a dosage of 10 mg/kg bw without any additional treatment. Groups III, IV, and V received cypermethrin at standard doses of 10 mg/kg bw and were orally administered with ascorbate (5,000 mg/kg bw), alpha-tocopherol (3,000 mg/kg bw), and a co-administration of ascorbate (5,000 mg/kg bw) and alpha-tocopherol (3,000 mg/kg bw), respectively. The animals were euthanized after 28 days, and samples were processed for histological analysis using hematoxylin and eosin staining. Analysis of variance (ANOVA) and Duncan's multiple range test were used to compare categorical variables of the biochemical parameters and determine the levels of MDA, SOD, GPx, and CAT. Alpha-tocopherol (vitamin E) is an essential nutrient that functions as a major lipid-soluble antioxidant in humans. The alpha-tocopherol transfer protein (TTP) binds α-tocopherol with high affinity and selectivity and regulates whole-body distribution of the vitamin. Heritable mutations in the TTPA gene result in familial vitamin E deficiency, elevated indices of oxidative stress, and progressive neurodegeneration that manifest primarily in spinocerebellar ataxia. Although the essential role of vitamin E in neurological health has been recognized for over 50 years, the mechanisms by which this essential nutrient is transported in the central nervous system are poorly understood. Here we found that, in the murine cerebellum, TTP is selectively expressed in glial fibrillary acidic protein-positive astrocytes, where it facilitates efflux of vitamin E to neighboring neurons. We also show that induction of oxidative stress enhances the transcription of the TtpA gene in cultured cerebellar astrocytes. Furthermore, secretion of vitamin E from astrocytes is mediated by an ABC-type transporter, and uptake of the vitamin into neurons involves the low-density lipoprotein receptor-related protein 1. Taken together, our data indicate that TTP-expressing astrocytes control the delivery of vitamin E from astrocytes to neurons, and that this process is homeostatically responsive to oxidative stress. These are the first observations that address the detailed molecular mechanisms of vitamin E transport in the central nervous system, and these results have important implications for understanding the molecular underpinnings of oxidative stress-related neurodegenerative diseases.

Almond okara, a by-product of almond milk production, is rich in bioactive components, such as polyphenols, lipids, and alpha-tocopherol, making it a valuable functional food ingredient. This work aimed to investigate its composition while exploring two main aspects: (i) the impact of extraction time, solid-to-solvent ratio, ethanol concentration, and temperature on polyphenol recovery, and (ii) the quantification of okara’s triglycerides (TG) and alpha-tocopherol contents. The polyphenols’ optimal extraction conditions were 90 min, a 1:30 solid-to-solvent ratio (w/v), 50% ethanol, and 60 °C. These conditions achieved a total polyphenol yield of 523 mg GAE, tannin yield of 340 mg GAE, total flavonoid yield of 548 mg CE, and a total antioxidant capacity of 779 mg AAE per 100 g dry okara. The Peleg model effectively described the extraction kinetics. Additionally, TG levels, quantified by UHE/LPSFC-APCI-MS, in okara were comparable to those in almonds, and alpha-tocopherol levels, quantified by LC-UV, were 14,400 µg/100 g in almonds and 15,600 µg/100 g in okara. These findings highlight the potential of okara as a valuable resource, with a straightforward, scalable, and cost-effective solid-liquid extraction (SLE) method for polyphenols and a supercritical fluid extraction method for TG, for use in the functional food, nutraceutical, and cosmetic industries.

BackgroundNumerous abnormalities in cystic fibrosis (CF) could influence tocopherol absorption, transportation, storage, metabolism and excretion. We hypothesized that the oxidative distress due to inflammation in CF increases vitamin E utilization, which could be positively influenced by supplemental vitamin C administration.MethodsImmediately before and after receiving vitamin C (500 mg) twice daily for 3.5 weeks, adult CF patients (n = 6) with moderately advanced respiratory tract (RT) disease consumed a standardized breakfast with 30% fat and a capsule containing 50 mg each hexadeuterium (d6)-α- and dideuterium (d2)-γ-tocopheryl acetates. Blood samples were taken frequently up to 72 h; plasma tocopherol pharmacokinetics were determined. During both trials, d6-α- and d2-γ-tocopherols were similarly absorbed and reached similar maximal plasma concentrations ~18-20 h. As predicted, during vitamin C supplementation, the rates of plasma d6-α-tocopherol decline were significantly slower.ConclusionsThe vitamin C-induced decrease in the plasma disappearance rate of α-tocopherol suggests that vitamin C recycled α-tocopherol, thereby augmenting its concentrations. We conclude that some attention should be paid to plasma ascorbic acid concentrations in CF patients, particularly to those individuals with more advanced RT inflammatory disease and including those with severe exacerbations.

Objective: To evaluate and compare the preventive role of Nebivolol and Alpha Tocopherol on testosterone-induced benign prostatic hyperplasia in rats. Study Design: Laboratory-based experimental study Place and Duration of Study: Department of Pharmacology and Pathology, Army Medical College, Rawalpindi, in collaboration with National Institute of Health (NIH), Islamabad Pakistan, from Dec 2021 to Jun 2022. Methodology: One hundred fifty healthy male Sprague-Dawley rats were procured from the NIH, Islamabad. They were placed into five groups, each with 30 rats. Results: Out of a total 150 sample of five groups, the mean prostatic index in Group-A, Group-B, Group-C, Group-D, and Group-E were 1.49±0.09, 2.35±0.48, 1.87±0.11, 1.72±0.04 and 1.68±0.08 respectively (p-value< 0.001). Mean Prostate Specific Antigen in Group-A, Group-B, Group-C, Group-D and Group-E was 0.19±0.02, 0.23±0.01, 0.18±0.02, 0.18±0.02 and 0.16±0.02 respectively (p-value< 0.001). On gross examination, in Group-A, all (100%) prostate samples were normal-sized. In Group-B, all (100%) prostate samples were enlarged. In Group-C, 53.3% were mild, and 47.7% of samples were moderately enlarged. In Group-D, 73.3% of samples were mildly enlarged, and 26.7% were moderately enlarged. In Group E, 26.7% of samples were normal, and 73.3% were mildly enlarged (p-value< 0.001). Conclusion: Prostatic index is markedly declined following treatment with the combination of Nebivolol and Alpha-Tocopherol. There is also a declining trend in Prostate Specific Antigen following treatment with a combination of Nebivolol and Alpha-Tocopherol.

Vitamin E is one of the most important vitamins in aquatic animal breeding, larval growth, and development. It is also considered to be a functional feed addtitive for its antioxidant and immunity-improvement role resulting in better survival and growth during early life stages. However, the role of vitamin E in the diet of milkfish larvae has not been studied in detail. Moreover, the optimal requirement of vitamin E for improved survival and growth in the larval stages of milkfish is yet to be ascertained. A 45-day feeding experiment was conducted to elucidate the effect of dietary vitamin E (alpha tocopherol) supplementation on the growth, survival, and fatty acid composition of milkfish, Chanos chanos, larvae. During the trial, five iso-nitrogenous and iso-energetic (~580 g kg−1 crude protein, 120 g kg−1 crude lipid) experimental larval diets with varying levels of vitamin E 0 (control E0), 100 (E100), 200 (E200), 300 (E300) and 400 (E400) mg kg−1 were tested in 18 days posthatch larvae (mean weight: 48.25 ± 0.08 mg). Milkfish larvae (1,500) were randomly distributed into 15 fiber-reinforced plastic (FRP) rectangular tanks of 100 L capacity (n = 100) in a flow-through rearing system. The results revealed that larvae fed with a diet containing vitamin E supplementation at 200 mg (E200) enhanced (p <.05) the growth performance, final body weight (371.7 ± 12.80 mg), weight gain percentage (628.76 ± 63.12%), and specific growth rate (4.31 ± 0.80%) compared with the E0 fed diet. Also, fish feed with E200 diet had a significantly higher protein efficiency ratio (0.53 ± 0.08) than the control (E0) (0.40 ± 0.01). Vitamin E and lipid content of postfed whole larvae were linearly correlated (R2 = 0.956) with dietary vitamin E inclusions up to 300 mg kg−1. Histological examinations revealed that the length of intestinal villi and the concentration of lymphoid cells in the lamina propria of the treatments E200, E300, and E400 were higher, compared to E0 and E100 treatments. Red blood cell (p < 0.05) and white blood cell (p < 0.01) counts were significantly higher in E200 treatment than in the rest of the treatments. Larvae fed with an E200 and E300 diet showed significantly (p < 0.05) increased total lymphocyte count, while myelocyte count was significantly decreased (p < 0.01). Based on a curvilinear (polynomial) regression analyses, the optimum specific growth rate, final body weight, weight gain, protein efficiency ratio, and feed efficiency ratio were obtained at dietary vitamin E levels of 210, 205, 207, 243, and 242 mg kg−1 diet, respectively. Accordingly, the optimum dietary vitamin E required for milkfish in a range of 205 to 243 mg kg−1 is beneficial for better health, survival, and growth. Background: Antioxidant application soon after bleaching process increases the shear bond strength (SBS) of composite resin to enamel. Aims: The aim of the study was to evaluate the antioxidant effects of selenium alone and in combination with alpha-tocopherol (αT) and green tea (GT) on SBS of composite resin to enamel following in-office bleaching with 38% hydrogen peroxide (HP). Methods: Sixty extracted human single -rooted premolar teeth were cleaned and embedded in acrylic resin blocks at the level of cementoenamel junction(CEJ) followed by bleaching with 38% hydrogen peroxide (HP) and arbitrarily divided into seven groups (n=10) for antioxidant application: Group I (negative control): intact teeth, Group II (positive control): only bleaching, Group III: 10% selenium (Se), Group IV: 10% alpha tocopherol (αT), Group V: 10% αT +10% Se, Group VI: 10% Green tea (GT), Group VII: 10%GT+10% Se. In all groups, self-etch adhesive was applied and composite restoration was done, and specimens were stored in distilled water for 24h followed by SBS evaluation. Statistical Analysis: One-way analysis of variance and post hoc Tukey's tests were used (P < 0.05). Results: The highest SBS was found in negative control Group I (intact teeth) and least in positive control Group II (bleached teeth), whereas in experimental groups, Group VII (GT + Se) showed highest followed by Groups V (αT + Se), III (Se), and VI (GT) and least in Group IV (αT). The ability to readily be designed, manufactured, and described has made cationic lipids one of the highly adaptable vehicles for the transfer of DNA, RNA, and numerous additional therapeutic compounds. In this article, we synthesized a variety of Tris‐(triazolyl)‐s‐triazine linker‐based cationic lipids with different types of anchoring groups, and assessed the transfection efficacy. Specifically, two identical tails like either two alpha‐tocopherol domains or cholesterol domains or aliphatic chains (saturated), and on the other hand, two different tails, alpha‐tocopherol domain and cholesterol domain or alpha‐tocopherol domain and aliphatic chain (saturated) were employed as hydrophobic motifs to synthesize the cationic lipids. The stability, size, and surface charge of liposomes were performed by dynamic light scattering technique. Cytotoxicity study on cell survival showed that the synthesized lipids are less hazardous to the cells despite the fact that cationic lipids have large molecular structure. The lipid having di alpha‐tocopherol domains demonstrated phenomenal β‐galactosidase protein expression and enhanced green fluorescence expression in the hepatocellular carcinoma cell line. Findings from the physico‐chemical and biological evaluations the developed lipids are compatible in various forms while maintaining low cytotoxicity and efficient carriers for nucleic acid delivery. Objective: This study evaluated the effects of antioxidants, 10% sodium ascorbate (SA) or 20% alpha‐tocopherol (AT), after post‐space irrigation with 2.5% sodium hypochlorite +17% EDTA (SH) or 1% peracetic acid (PA) on the adhesive interface after glass fiber post cementation. Materials and Methods: Sixty bovine roots were endodontically treated. After preparation, the post‐space was irrigated with SH or PA followed or not by the use of antioxidants (SA or AT) (n = 10). Push‐out bond strength test, failure mode, and dentin penetrability analysis using confocal laser microscope were performed in the cervical, middle, and apical thirds. Data from bond strength and dentinal penetrability were evaluated by one‐way ANOVA and Tukey post hoc test (p < 0.05). Results: SH showed the lowest bond strength regardless of the third (p < 0.05). In apical third, mixed failure was the most incident in all groups. Only in the cervical third of the post‐space, SH‐AT provided the greatest tag extension of the cementation system into dentin (p < 0.05). However, in the middle and apical thirds, SH‐AT, SH‐SA, and PA‐SA provided the largest tag extensions (p < 0.05), but similar to each other (p > 0.05). Conclusions: The use of antioxidants only favored bond strength when SH was used and dentin penetrability of the adhesive and conventional resin cementation, regardless of the solution used to irrigate the post‐space. Clinical Significance: The use of antioxidants (10% sodium ascorbate and 20% alpha‐tocopherol) after post‐space irrigation with sodium hypochlorite appears to increase the bond strength favoring the glass fiber post‐cementation. Vitamin E is essential for neuromuscular function. The primary treatment, oral supplementation with natural ('RRR') α-tocopherol, is not effective in all horses. The objectives of this pilot study were to evaluate the safety and efficacy of a subcutaneously administered RRR-α-tocopherol preparation. Horses were randomly assigned in a cross-over design to initially receive RRR-α-tocopherol (5000 IU/450 kg of 600 IU/mL) subcutaneously (n = 3) or orally (n = 3) or were untreated sentinels (n = 2). Tissue reactions following injection in Phase I of the study necessitated adjustment of the preparation with reduction of the RRR-α-tocopherol concentration to 500 IU/mL in Phase 2. Following an 8-week washout period, horses received the reciprocal treatment route with the new preparation (5000 IU/450 kg of 500 IU/mL). Serum, CSF and muscle α-tocopherol concentrations were determined by high-performance liquid chromatography over a 14-day period during each phase. To assess the potential of exogenously-applied vitamin E commonly known as alpha-tocopherol (α-TOC) in drought stress tolerance, an experiment was performed on cauliflower (Brassica oleracea L. var. botrytis) plants. Two cultivars (TCF-605 and White Figer) of cauliflower were subjected to control and drought stress conditions. After thirty days of drought stress treatment, cauliflower plants were treated with α-TOC (150 and 300 mg/L) as a foliage spray. Drought stress markedly inhibited the plant biomass, leaf characteristics such as relative water contents (RWC), chlorophyll pigments (a and b), proline, ascorbic acid (AsA), GB, however it elevated RMP, total phenolics, activities of antioxidant enzymes e.g., CAT, SOD and POD. Drought stress also adversely affected curd characteristics including its fresh weight, GB, proline, total phenolics and enzymes activities (SOD and CAT). Exogenously applied α-TOC considerably suppressed RMP, while it improved plant growth, yield, RWC, total phenolics, ascorbic acid and POD, SOD, and CAT enzyme activities in the leaves of the cauliflower. Exogenous application of α-TOC also raised curd total phenolics, AsA and the activities of all observed enzymes under water deficiency. The cv. TCF-605 was better in shoot weight, shoot length, RWC, proline as well as GB contents, while cv. White Figer was higher in leaf RMP, proline and AsA levels under drought stress conditions. Overall, the results exhibited that cauliflower cv. TCF-605 was relatively tolerant to water stress in relation to growth and yield outcomes, which was interlinked with α-TOC-induced improved leaf and curd osmoprotection and antioxidative defense system. So, exogenous application of α-TOC can be suggested for improving growth and yield potential of crops subjected to drought stress.

Aims: Alpha-tocopherol (AT) and tocotrienol (TT) are both naturally occurring forms of vitamin E found in palm oil. Previous studies found beneficial effects of AT in multiple sclerosis (MS) animal models. In the present study, the effects of palm oil-derived TT-rich fraction (TRF) and AT on the locomotor function, lipid peroxidation, and histological study of demyelination were compared in cuprizone (CPZ)-induced rat model of MS. Methods: Female Sprague-Dawley rats aged 8 weeks received pellets mixed with CPZ for a duration of 6 weeks, followed by 100 mg/kg AT (group AT, n = 10) daily and 60 mg/kg TRF (group TT, n = 10) daily and vehicle (group V, n = 10) through oral gavage for 2 weeks. Open field test (OFT) and beam walking test (BWT) were carried out every 10th day. The status of myelination in the corpus callosum was evaluated by Luxol fast blue (LFB) staining and immunohistochemical staining of myelin basic protein (MBP). Malondialdehyde (MDA) level was estimated using brain homogenate. Results: Compared with group V, total distance moved and movement speed increased by 42.6 and 52.9%, respectively in group TT (P < 0.05). Line crossing frequency increased by 39% (P < 0.05) and 36% (P < 0.05), respectively in groups TT and AT. Corpus callosum showed a maximum reduction of demyelinated areas and higher expression of MBP in group TT. MDA level in the cortex was significantly reduced in group AT. Background:The combined supplementation of vitamins C and E potentially can mitigate oxidative stress (OS) and accelerate recovery following exercise. However, there is little evidence and a lack of consensus on the effects of these vitamins for this purpose. The objective of this systematic review was to summarize the evidence on the effects of the combined supplementation of vitamins C and E in OS, inflammatory markers, muscle damage, muscle soreness, and musculoskeletal functionality following acute exercise. Methods: The search was carried out from inception until March 2021, on MEDLINE, EMBASE, Cochrane CENTRAL, Web of Science, and SPORT Discus. We included placebo-controlled randomized clinical trials (RCTs) that evaluated the effects of combined supplementation of vitamins C and E in OS, inflammatory markers, muscle damage, muscle soreness, and muscle strength following a single bout of exercise. Random-effect meta-analyses were used to compare pre to post-exercise mean changes in subjects who received supplementation with vitamins C and E or placebo versus controls. Data are presented as standard mean difference (SMD) and 95% confidence interval (95% CI). Results: Eighteen RCTs, accounting for data from 322 individuals, were included. The use of vitamins attenuated lipid peroxidation (SMD= −0.703; 95% CI= −1.035 to −0.372; p < 0.001), IL-6 (SMD= −0.576; 95%CI= −1.036 to −0.117; p = 0.014), and cortisol levels (SMD= −0.918; 95%CI= −1.475 to −0.361; p = 0.001) immediately, and creatine kinase levels 48 h following exercise (SMD= −0.991; 95%CI= −1.611 to −0.372; p = 0.002). Supplementing the combination of vitamins had no effects on protein carbonyls, reduced/oxidized glutathione ratio, catalase, interleukin-1Ra, C-reactive protein, lactate dehydrogenase, muscle soreness, and muscle strength. Conclusion: Prior supplementation of the combination of vitamins C and E attenuates OS (lipid peroxidation), the inflammatory response (interleukin-6), cortisol levels, and muscle damage (creatine kinase) following a session of exercise. Oral vaccines represent many advantages compared to standard vaccines. They hold a simple method of administration and manufacturing process. In addition to these, the way they can induce immune responses makes these a promising technology for the pharmaceutical industry and represents a new hope to society. Physiologically based pharmacokinetics (PBPK) has been used in support of drug development to predict the pharmacokinetics of the compound, considering the patient's physiology. Despite PBPK studies now being widely used, there are very few models in the literature that support vaccine development. Therefore, the goal of this article was to determine how PBPK could support vaccine development. The first PBPK model for an oral vaccine using alpha-tocopherol as a vaccine adjuvant was built. LogP is the parameter that influences the delivery of alpha-tocopherol into the tissues more. Having a high LogP means it accumulates in adipose tissue and is slowly metabolized. The ideal formulation to include alpha-tocopherol in an oral vaccine would incorporate nanoparticles in a capsule, and the dosage of the compound would be 150 mg in a volume of 200 mL. This article aims to determine if alpha-tocopherol, as a well-known adjuvant for intramuscular injection vaccines, could be used as an adjuvant to oral vaccines. This model was built considering the conditions and requirements needed for designing an oral vaccine. This implies making sure the antigen and adjuvants reach the main target by overcoming the challenges of the gastrointestinal tract. Vitamin E is an essential nutrient usually recommended in post-weaning piglets, when a decline in the serum vitamin E concentration is observed. Selected polyphenols have the potential to partially replace vitamin E in animal feed. The aim of this study was to investigate the effect of the dietary inclusion of some commercial polyphenol products (PPs) on the growth performance, antioxidant status and immunity of post-weaning piglets. A total of 300 piglets (BW 7.18 kg ± 1.18) were randomly assigned to six dietary groups: CON− (40 mg/kg vitamin E); CON+(175.8 mg/kg vitamin E); and PP1, PP2, PP3 and PP4, in which 50% vitamin E of CON+ was replaced with PP with equivalent vitamin E activity. The PP1 group exhibited lower performance (p < 0.05) than the other dietary groups, but a similar performance to that commonly registered in pig farms. Dietary polyphenols did not influence the IgG concentration or the IL-6, IL-10, IFN-γ and TNF-α cytokine concentrations. A lower IL-8 level was found in the PP4 group than in the other groups. The diets that affected the vitamin A content showed the highest value (p < 0.05) in the PP1 group, and a trend was noted for vitamin E with a higher content in PP4 and CON+. The polyphenols-enriched diets, especially the PP3 diet, maintained an antioxidant capacity (whole blood KRL) similar to the CON+ diet. Abstract Background Deterioration in shear bond strength has been reported after immediate bracket bonding following hydrogen peroxide bleaching. This study compared the effectiveness of three antioxidant agents, namely, alpha-tocopherol, green tea extract, and sodium ascorbate, in reversing the bleaching effect and as possible alternatives to delayed bonding. Methods A total of 105 extracted human premolars were arbitrarily assigned to 7 groups (n = 15 each), including group 1 as the unbleached control group and six experimental groups, which were bleached with 40% hydrogen peroxide in three sessions of 15 min each. In experimental group 2, bonding was performed immediately after bleaching, whereas in groups 3 and 4, bonding was delayed for 1 and 2 weeks, respectively; meanwhile, the specimens were immersed in artificial saliva at 37 °C. Groups 5, 6, and 7 were treated immediately after bleaching with 10% of alpha-tocopherol, green tea extract, and sodium ascorbate solutions, respectively, for 15 min. Specimens were processed using 500 thermal cycles between 5 and 55 °C, with a dwell time of 30 s after 24 h of bracket bonding, and then tested for shear bond strength. The adhesive remnant index was examined to evaluate fracture mode. One-way analysis of variance, Kruskal–Wallis H, and post hoc Tukey’s honestly significant difference tests were used to compare the data. Significant results were subjected to pairwise comparisons with Bonferroni’s correction-adjusted of p values ≤ 0.050. Results Shear bond strength was significantly lower (p 0.05). Conclusions Application of 10% alpha-tocopherol, green tea extract, or sodium ascorbate for 15 min could restore shear bond strength after 40% hydrogen peroxide bleaching as an alternative to delay in bracket bonding.

Abstract Objective This study aimed to evaluate and compare the impacts of the postbleaching application of sodium ascorbate, alpha‐tocopherol, and quercetin on the shear bond strength (SBS) of composite resin. Material and Methods 60 extracted intact maxillary first premolars were collected and were randomly divided into five experimental groups as follows (n=12): Group A (negative control): no bleaching, Group B (positive control): bleaching with 40% hydrogen peroxide (HP), Group C: HP±10% sodium ascorbate for 10min, Group D: HP±10% alpha‐tocopherol for 10min, and Group E: HP±1% quercetin for 10min. Composite bonding was done immediately after bleaching for Groups B‐E and without any treatment for Group A. After being stored in distilled water at room temperature for 24h, all specimens were tested for SBS in the universal testing machine. One‐way analysis of variance and Tukey's post‐hoc test were used to analyze the SBS values of all groups. Results The results showed that the bonding of composite to the unbleached group exhibited the highest mean value of SBS (22.68±2.91MPa). A variant of the method of differential pulse voltammetry is proposed for the using in the study of a new group of natural antioxidants, polar lipids with paramagnetic properties, in the total lipids of marine organisms. Voltammetric analysis of the total lipids of the Mediterranean mussel Mytilus galloprovincialis L. (gonads) and the Black Sea katran Squalus acanthias (liver) showed them to have one similar anodic wave with an oxidation peak potential of +0.60 V. The difference between the studied hydrobionts manifested itself in the registration of one more anodic wave with the oxidation peak potentials of +1.20 V in the total lipids of the mussel gonad and +0.90 V in those of the katran liver. The using of the proposed version of the method of differential pulse voltammetry together with ESR-spectroscopy and spectrofluorometry in the study of the physico-chemical properties of the new group of polar lipids with paramagnetic properties is substantiated. This article aimed to evaluate the activity of Vitamin E as an antioxidant in oral pharmaceutical preparations. Vitamin E is an essential nutrition soluble in fat that has the pharmacology effect as an antioxidant. Vitamin E consists of α, β, γ, δ-tocopherol and α, β, γ, δ-tocotrienol. The activity of vitamin E was observed for its ability to inhibit reduction using the DPPH method. The antioxidant activity is calculated as an inhibition concentration of 50% (IC50) which is the concentration of a sample that can reduce 50% of DPPH. It was known that IC50 of vitamin E was very strong with value of 42.86%. Equine neuroaxonal dystrophy/equine degenerative myeloencephalopathy (eNAD/EDM) is an inherited neurodegenerative disorder of unknown etiology. Clinical signs of neurological deficits develop within the first year of life in vitamin E (vitE) deficient horses. A genome-wide association study (GWAS) was carried out using 670,000 SNP markers in 27 case and 42 control Quarter Horses. Two markers, encompassing a 2.5 Mb region on ECA7, were associated with the phenotype (p = 2.05 × 10-7 and 4.72 × 10-6). Within this region, caytaxin (ATCAY) was identified as a candidate gene due to its known role in Cayman Ataxia and ataxic/dystonic phenotypes in mouse models. Whole-genome sequence data in four eNAD/EDM and five unaffected horses identified 199 associated variants within the ECA7 region. MassARRAY® genotyping was performed on these variants within the GWAS population. The three variants within ATCAY were not concordant with the disease phenotype. No difference in expression or alternative splicing was identified using qRT-PCR in brainstem across the ATCAY transcript. Atcayji-hes mice were then used to conduct functional analysis in a second animal model. Histologic lesions were not identified in the central nervous system of Atcayji-hes mice. Additionally, supplementation of homozygous Atcayji-hes mice with 600 IU/day of dl-α-tocopheryl acetate (vitE) during gestation, lactation, and adulthood did not improve the phenotype. ATCAY has therefore been excluded as a candidate gene for eNAD/EDM.

Retinol (vitamin A) and α-tocopherol (vitamin E) concentrations were measured in tissue samples (liver, heart, pectoral muscle, and brain) from Anna's Hummingbirds (Calypte anna). Hummingbirds were after-hatch year birds that were sourced from various rehabilitation centers throughout California. Tissues samples were analyzed using high-performance liquid chromatography (HPLC). Minimum, maximum, mean, standard deviation (SD), and median ppm concentrations were calculated for each vitamin and tissue sample type. A novel analytical method was developed to analyze small mass tissue samples, with the smallest sample mass being 0.05 g for which analysis can be performed. Mean ± standard deviation (SD) concentrations of retinol in hummingbird livers, hearts, and pectoral muscle samples were 269.0 ± 216.9 ppm, 1.8 ± 2.2 ppm, and 0.3 ± 0.1 ppm, respectively. Mean ± SD α-tocopherol concentrations were 6.9 ± 4.6 ppm, 5.5 ± 4.0 ppm, 3.7 ± 2.2 ppm, and 9.1 ± 3.2 ppm for liver, heart, pectoral muscle, and brain samples, respectively. Vitamin concentrations from varying tissue types were compared to determine which were best associated with liver concentrations, the most commonly analyzed tissue for these vitamins. For both retinol and α-tocopherol, heart samples were most strongly associated with the liver samples. The results of this study provide baseline retinol and α-tocopherol concentrations in different tissue types from Anna's hummingbirds. These baseline values may be utilized in conservation efforts to avoid hypervitaminosis and hypovitaminosis of rehabilitated and/or captive hummingbirds by providing guidelines for nutritional targets which could be assessed on post-mortem examinations. Post-mortem examination of birds and measurement of vitamin concentrations in tissues may allow for dietary changes that aid captive hummingbirds.

Ninety percent of Americans consume less than the estimated average requirements of dietary vitamin E (vitE). Severe vitE deficiency due to genetic mutations in the tocopherol transfer protein (TTPA) in humans results in ataxia with vitE deficiency (AVED), with proprioceptive deficits and somatosensory degeneration arising from dorsal root ganglia neurons (DRGNs). Single-cell RNA-sequencing of DRGNs was performed in Ttpa-/- mice, an established model of AVED. In stark contrast to expected changes in proprioceptive neurons, Ttpa-/- DRGNs showed marked upregulation of voltage-gated Ca2+ and K+ channels in mechanosensitive, tyrosine-hydroxylase positive (TH+) DRGNs. The ensuing significant conductance changes resulted in reduced excitability in mechanosensitive Ttpa-/- DRGNs. A highly supplemented vitE diet (600 mg dl-α-tocopheryl acetate/kg diet) prevented the cellular and molecular alterations and improved mechanosensation. VitE deficiency profoundly alters the molecular signature and functional properties of mechanosensitive TH+ DRGN, representing an intriguing shift of the prevailing paradigm from proprioception to mechanical sensation.

Research questionIs a mixture of preconception serum lipids and lipophilic micronutrients associated with clinical pregnancy and live births?DesignIn this prospective cohort study, blood serum was collected on the day of oocyte retrieval for 180 women undergoing IVF at an academic reproductive health centre. Concentrations of lipids (phospholipids, total cholesterol, high- and low-density lipoproteins, and triglycerides) and lipophilic micronutrients (α-, δ-, and γ-tocopherols, retinol, β- and α-carotenes, β-cryptoxanthin, lutein and lycopene) were determined using diagnostic reagent kits and high-performance liquid chromatography. Poisson regression was used with robust variance estimation to evaluate changes in Z-scores for the mixture of serum lipid and lipophilic micronutrient concentrations as predictors of embryo implantation, clinical pregnancy and live birth, adjusted for age, body mass index (BMI), race, smoking status, infertility diagnosis, ovarian stimulation protocol and other measured lipid and lipophilic micronutrient concentrations.ResultsEach SD higher serum triglyceride concentration was associated with a lower chance of live birth (RR 0.54; 95% CI 0.33 to 0.90) whereas a 1 SD higher serum α-tocopherol concentration, as part of a mixture of serum lipids and lipophilic micronutrients, was associated with a higher likelihood for a live birth (RR 1.61; 95% CI 1.11 to 2.36). Serum β-carotene concentrations were associated with live birth in a non-linear fashion; low β-carotene was associated with a lower chance of live birth and high β-carotene with a higher chance of live birth.ConclusionAlthough components of a mixture of lipids and lipophilic micronutrients were associated with live birth outcomes after IVF, a larger investigation is necessary to fully evaluate the potential clinical implications.

Background: Epidemiological evidence on alpha (a)-tocopherol intake and cognitive performance in older individuals is controversial and the effect of periodontitis in this chain is sparse and limited. The goal of this study was to characterize the association between a-tocopherol intake and cognitive performance and the mediating role of periodontitis in a nationally representative sample of older adults. Methods: Data from the National Health and Nutrition Examination Survey (NHANES), 2011-2014, were used. Multivariate logistic regression analysis was performed to explore the association of a-tocopherol intake, periodontal measures (mean attachment loss [AL] and mean probing depth [PD]), and clinical periodontitis defined by the European Workshop in Periodontology with poor cognitive performance evaluated by Consortium to Establish a Registry for Alzheimer's disease (CERAD); the animal fluency test (AFT); and the Digit Symbol Substitution test (DSST) and the correlation between a-tocopherol intake and clinical periodontitis. Multiple linear regression analysis was used to explore the relationship between a-tocopherol intake and periodontal measures. Mediation analysis was used to test the effects of periodontal measures on the association between a-tocopherol intake and cognitive measures. Results: A total of 1,749 older participants (>60 years of age) with complete periodontal diagnosis, dietary retrospective survey, and cognitive tests were included. In the fully adjusted model, the odds ratio (OR) with 95% confidence interval (CI) of CERAD score, AFT score and DSST score were 0.214 (0.137-0.327), 0.378 (0.241-0.585) and 0.298 (0.169-0.512) for the highest versus lowest tertile of a-tocopherol intake, respectively. And participants with clinical periodontitis were more likely to exhibit lower DSST score (OR = 1.689; 95 CI%: 1.018-2.771) than those without periodontitis. Mean AL (OR = 1.296; 95 CI%: 1.102-1.524) and PD (OR = 1.667; 95 CI%: 1.18-2.363) were negatively correlated with DSST, and were estimated to mediate 9.1 and 8.2% of the total association between a-tocopherol intake and cognitive performance, respectively. Conclusion: Finding of the present study suggested that participants with low a-tocopherol intake were at higher risk for developing cognitive decline. Mice with deficiency in tocopherol (alpha) transfer protein gene develop peripheral tocopherol deficiency and sensory neurodegeneration. Ttpa-/- mice maintained on diets with deficient α-tocopherol (α-TOH) had proprioceptive deficits by six months of age, axonal degeneration and neuronal chromatolysis within the dorsal column of the spinal cord and its projections into the medulla. Transmission electron microscopy revealed degeneration of dorsal column axons. We addressed the potential pathomechanism of α-TOH deficient neurodegeneration by global transcriptome sequencing within the spinal cord and cerebellum. RNA-sequencing of the spinal cord in Ttpa-/- mice revealed upregulation of genes associated with the innate immune response, indicating a molecular signature of microglial activation as a result of tocopherol deficiency. For the first time, low level Ttpa expression was identified in the murine spinal cord. Further, the transcription factor liver X receptor (LXR) was strongly activated by α-TOH deficiency, triggering dysregulation of cholesterol biosynthesis. The aberrant activation of transcription factor LXR suppressed the normal induction of the transcription factor retinoic-related orphan receptor-α (RORA), which is required for neural homeostasis. Thus we find that α-TOH deficiency induces LXR, which may lead to a molecular signature of microglial activation and contribute to sensory neurodegeneration.

The development of new vaccine adjuvants represents a key approach to improvingi the immune responses to recombinant vaccine antigens. Emulsion adjuvants, such as AS03 and MF59, in combination with influenza vaccines, have allowed antigen dose sparing, greater breadth of responses and fewer immunizations. It has been demonstrated previously that emulsion adjuvants can be prepared using a simple, low-shear process of self-emulsification (SE). The role of alpha tocopherol as an immune potentiator in emulsion adjuvants is clear from the success of AS03 in pandemic responses, both to influenza and COVID-19. Although it was a significant formulation challenge to include alpha tocopherol in an emulsion prepared by a low-shear process, the resultant self-emulsifying adjuvant system (SE-AS) showed a comparable effect to the established AS03 when used with a quadrivalent influenza vaccine (QIV). In this paper, we first optimized the SE-AS with alpha tocopherol to create SE-AS44, which allowed the emulsion to be sterile-filtered. Then, we compared the in vitro cell activation cytokine profile of SE-AS44 with the self-emulsifying adjuvant 160 (SEA160), a squalene-only adjuvant.
