Abstract

The application of digital technologies such as 3D modeling, 3D printing, and virtual surgery implemented in the medical field makes it possible to improve the representation of the patient's anatomy and to analyze the clinical case in more detail than the two-dimensional visualization of images from CT, MRI, and ultrasound. In particular, the use of technologies such as 3D printing in cardiac surgery is useful for studying patient-specific cardiac anatomy and manipulating the object of study preoperatively. The integration of a preoperative planning methodology integrated with the new advanced tools facilitates the cardiac surgeon in developing a more informed and precise surgical strategy. Indeed, new technologies allow surgery to be designed and planned in a virtual environment before it is performed in the operating room. This study reports the application of advanced technologies for aortic arch reconstruction in a virtual and physical environment as a tool for the analysis of cardiac pathologies, e.g., aortic dissection. The process involves the use of software that can convert the patient's medical information into a three-dimensional digital model that can be imported into a virtual environment and then 3D printed. Abstract

The following paper focuses on designing an innovative method for connecting 3D printed components. This is an important topic to weakness the problem of printer dimensions. The idea of connecting different parts in a larger one using compliant mechanism starts from a real application concerning the building of molds with dimensions beyond the desktop size for carbon fiber lamination. The goal is to maximize the mold quality without the need of excessively increase in printing time. These aspects provided an opportunity to delve into the topic of dividing 3d printed molds and the subsequent assembly, as well as disassembly for the extraction stages of the laminated part. The goal of the paper is to propose a mechanism capable of facilitating all stages of part production, from mold assembly to component extraction. Abstract

This project consists in the set up and preparation of an educational laboratory where the study and implementation of all the techniques and methodologies of Style Design Engineering (SDE) method were fitted. The SDE method, represents a design methodology widely used in the industrial field that proved to add value in the quality of project outcomes. Through numerous previous scientific exercises published and the various projects carried out, the effectiveness of this methodology demonstrated to lead to higher quality project results in the automotive field. It is therefore necessary for the department to have spaces for the exclusive use of this activity, structured in such a way as to be equipped for the training and support of the various students who want to try their skills on car design projects within their final graduation projects. The various phases that make up this activity from a theoretical point of view will then be analyzed and deconstructed to understand which material support they need for their complete realization throughout the process. Implementation of innovative technologies such as AR/VR and 3D printing will be explored to reach the best possible result in the student's works. Abstract

Fused Deposition Modeling (FDM) 3D printing is the most widespread technology in additive manufacturing worldwide acknowledging its low cost, finished component applications, and the production process of other parts. The aim of this study is to perform a process for checking the reproducibility capabilities of an FDM-created part by means of state of the art, 3D scanning technology and dedicated software for analysis. One of the main challenges that FDM processes have found to become a trustful manufacturing process in the industry, is the high variability of the outcome, given the printing parameters choice due to the wide variety of material choices, and different suppliers. Additionally, there is the variability of the FDM manufacturing elements, that translates into a different interlayer bond quality, same of which could punish the capability of the printed element to sustain mechanical tests with accuracy. This procedure is a step further into reaching a proper method for FDM manufacturing that takes into account these challenges and help to turn FDM manufacturing into a mainstream, trustworthy manufacturing process for engineering prototyping. Abstract

Computer Aided Design (CAD) and 3D printing are increasingly bringing innovation to orthopedic surgery by allowing customization of surgical procedures. This study proposes the design and 3D printing of patient-specific surgical cutting guides (PSCG) that can be implemented in orthopedic surgery. Cutting guides are surgical tools that allow surgeons to perform cutting operations more accurately, achieving the desired result with high accuracy. During an osteotomy, for example, the surgeon will be able to better determine the position of the cutting line and its direction. This study aims to propose an advanced method of designing cutting guides for an orthopedic surgery procedure. Proposed tools are obtained by an additive manufacturing process that starts with a 3D model of the patient's bone obtained from CT scan images and ends by designing the patient-specific cutting guide model in a CAD environment. Virtual surgical planning (VSP) allows for increasing the surgeon's awareness for developing an orthopedic surgical strategy and allows for the design of cutting guides designed around the patient's specific anatomy. Abstract

3D printing technologies are proving to be an increasingly useful tool not only for the creation of prototypes, but also of finished components. 3D printing has become a mature technology able to product finished objects and non finished parts intended to support the production. This is possible because of the continuous innovation in materials that are now able to achieve specific requests. Moreover the lowering of the hardware's costs played a central role in the expansion of 3D printing technology. Another important aspect of 3D printing processes is in fact their application within the production process of other components, for example, by considering less inexpensive technologies such as FDM and mSLA. The goal of this research is to take advantage of the great variety that 3D printing offers in order to implement this process across different mechanical and prototyping applications to emphasize the importance of spreading this technology. Abstract

Whole metagenomic shotgun (WMS) sequencing has dramatically enhanced our ability to study microbial genomics. The possibility to unveil the genetic makeup of bacteria that cannot be easily isolated has significantly expanded our microbiological horizon. Here, we report an approach aimed at uncovering novel bacterial species by the use of targeted WMS sequencing. Employing in silico data retrieved from metabolic modelling to formulate a chemically defined medium (CDM), we were able to isolate and subsequently sequence the genomes of six putative novel species of bacteria from the gut of non-human primates.

Abstract

Ten Bifidobacterium strains, i.e., 6T3, 64T4, 79T10, 80T4, 81T8, 82T1, 82T10, 82T18, 82T24, and 82T25, were isolated from mantled guereza (Colobus guereza), Sumatran orangutan (Pongo abeli), silvery marmoset (Mico argentatus), golden lion tamarin (Leontopithecus rosalia), pied tamarin (Saguinus bicolor), and common pheasant (Phaisanus colchinus). Cells are Gram-positive, non-motile, non-sporulating, facultative anaerobic, and fructose 6-phosphate phosphoketolase-positive. Phylogenetic analyses based on the core genome sequences revealed that isolated strains exhibit close phylogenetic relatedness with Bifidobacterium genus members belonging to the Bifidobacterium bifidum, Bifidobacterium longum, Bifidobacterium pullorum, and Bifidobacterium tissieri phylogenetic groups. Phenotypic characterization and genotyping based on the genome sequences clearly show that these strains are distinct from each of the type strains of the so far recognized Bifidobacterium species. Thus, B. phasiani sp. nov. (6T3 = LMG 32224T = DSM 112544T), B. pongonis sp. nov. (64T4 = LMG 32281T = DSM 112547T), B. saguinibicoloris sp. nov. Abstract

The following paper aims to take a detailed look at the various stages of IDeS (Industrial Design Engineering), a prime example of a modern, cutting-edge methodology useful for laying the groundwork for new product development within an organization. Its general outline interfaces with any type of product and is applicable in almost any context, from food production to the world of cosmetics, from the biomedical sector to car design. The methodology described is orderly and perfectly schematic, so it can govern all departments of a generic company with the goal of product development, to the point that its very architecture could be built following the logic of IDeS. From an initial market analysis useful for understanding the needs required by the future users of the product, through benchmarking applied to competitors in the target market segment, arriving at the study of the actual architecture of the product, all the steps of the IDeS will be considered. In detail, the steps of "Stylistic Design Engineering," a purely design approach to develop automotive design projects in the industrial world, will be applied to arrive at the final product configuration. Serving first Quality Function Deployment (QFD), a methodology involving market and competitive analysis, and SDE, the goal was to design a new sports car model that can be competitive and aesthetically cutting-edge. The model creation process is divided into several stages that required in-depth knowledge in the use of various programs. Both the aesthetics and performance of the vehicle must be perfectly optimized. Abstract

Members of the Bifidobacterium dentium species are usually identified in the oral cavity of humans and associated with the development of plaque and dental caries. Nevertheless, they have also been detected from fecal samples, highlighting a widespread distribution among mammals. To explore the genetic variability of this species, we isolated and sequenced the genomes of 18 different B. dentium strains collected from fecal samples of several primate species and an Ursus arctos. Thus, we investigated the genomic variability and metabolic abilities of the new B. dentium isolates together with 20 public genome sequences. Comparative genomic analyses provided insights into the vast metabolic repertoire of the species, highlighting 19 glycosyl hydrolases families shared between each analyzed strain. Phylogenetic analysis of the B. dentium taxon, involving 1140 conserved genes, revealed a very close phylogenetic relatedness among members of this species. Furthermore, low genomic variability between strains was also confirmed by an average nucleotide identity analysis showing values higher than 98.2%. Investigating the genetic features of each strain, few putative functional mobile elements were identified. Besides, a consistent occurrence of defense mechanisms such as CRISPR–Cas and restriction–modification systems may be responsible for the high genome synteny identified among members of this taxon

Abstract

In recent years, bifidobacterial populations in the gut of various monkey species have been assessed in several ecological surveys, unveiling a diverse, yet unexplored ecosystem harbouring novel species. In the current study, we investigated the species distribution of bifidobacteria present in 23 different species of primates, including human samples, by means of 16S rRNA microbial profiling and internal transcribed spacer bifidobacterial profiling. Based on the observed bifidobacterial‐host co‐phylogeny, we found a statistically significant correlation between the Hominidae family and particular bifidobacterial species isolated from humans, indicating phylosymbiosis between these lineages. Furthermore, phylogenetic and glycobiome analyses, based on 40 bifidobacterial species isolated from primates, revealed that members of the Bifidobacterium tissieri phylogenetic group, which are typical gut inhabitants of members of the Cebidae family, descend from an ancient ancestor with respect to other bifidobacterial taxa isolated from primates.

Abstract

The present study wants to bring to light a new type of crutch designed for a chronic patient with perennial limited mobility, who must use this support every time a move is needed. The main purpose of the project consists in recommending a correct use of the crutch through technology, limiting the damage normally caused by a bad use of crutches and giving a support both for the patient and for the doctor. All of the features of the crutches were defined through relationship matrices and a benchmarking, which helped us for defining the requirements; other important features were defined, taking a look to the technological progresses applied to new, patented crutches. The result is a sensorized crutch, functional and oriented to meet the user's needs in order to prevent an incorrect use of the support avoiding the growth of other pains. [ABSTRACT FROM AUTHOR]

Abstract

Bifidobacteria are recognized as health-promoting bacteria that reside in the human gut, helping in the digestion of fiber, preventing infections, and producing essential compounds like vitamins. To date, Bifidobacterium animalis subsp. lactis, together with Bifidobacterium adolescentis, Bifidobacterium bifidum, Bifidobacterium breve, and Bifidobacterium longum, represents one of the species that are used as probiotic bacteria. Despite the extensive and detailed scientific research conducted on this microbial taxon, the molecular mechanisms by which B. animalis subsp. lactis exerts health benefits to its host are still largely unknown. Thus, we dissected the genetic repertoire and phylogenetic relationship of 162 strains of B. animalis subsp. lactis to select a representative reference strain of this taxon suitable for investigating its interaction with the host. The B. animalis subsp. lactis PRL2013 strain, which was isolated by a mucosal sample of a healthy adult, was chosen as the reference of the monophyletic cluster of human origin and revealed a greater adhesion index than that observed for another B. animalis subsp. lactis strain used in the industry as a probiotic supplement. Transcriptomics analyses of PRL2013 strain, when exposed to human cell monolayers, revealed 291 significantly upregulated genes, among which were found genes predicted to encode extracellular structures that may directly interact with human cells, such as extracellular polymeric substances, wall teichoic acids, and pili. IMPORTANCE To date, many Bifidobacterium animalis subsp. lactis strains have been isolated from human fecal samples. However, their presence in these samples does not necessarily suggest an ability to colonize the human gut. Furthermore, probiotics of non-human origin may not effectively interact with the gut epithelium, resulting in transient bacteria of the gut microbiota. In vitro experiments with human cells revealed that B. animalis subsp. lactis PRL2013, an autochthonous member of the human gut, shows colonization capability, leading to future applications in functional foods. Abstract

Members of the genus Lactobacillus represent the most common colonizers of the human vagina and are well-known for preserving vaginal health and contrasting the colonization of opportunistic pathogens. Remarkably, high abundance of Lactobacillus crispatus in the vaginal environment has been linked to vaginal health, leading to the widespread use of many L. crispatus strains as probiotics. Nevertheless, despite the scientific and industrial relevance of this species, a comprehensive investigation of the genomics of L. crispatus taxon is still missing. For this reason, we have performed a comparative genomics analysis of 97 L. crispatus strains, encompassing 16 strains sequenced in the framework of this study alongside 81 additional publicly available genome sequences. Thus, allowing the dissection of the L.crispatus pan-genome and core-genome followed by a comprehensive phylogenetic analysis based on the predicted core genes that revealed clustering based on ecological origin. Subsequently, a genomics-targeted approach, i.e., probiogenomics analysis, was applied for in-depth analysis of the eight L. crispatus strains of human origin sequenced in this study. In detail their genetic repertoire was screened for strain-specific genes responsible for phenotypic features that may guide the identification of optimal candidates for next-generation probiotics. The latter includes bacteriocin production, carbohydrates transport and metabolism, as well as a range of features that may be responsible for improved ecological fitness. In silico results regarding the genetic repertoire involved in carbohydrate metabolism were also validated by growth assays on a range of sugars, leading to the selection of putative novel probiotic strains. Abstract

Human milk is known to carry its own microbiota, of which the precise origin remains obscure. Breastfeeding allows mother-to-baby transmission of microorganisms as well as the transfer of many other milk components, such as human milk oligosaccharides (HMOs), which act as metabolizable substrates for particular bacteria, such as bifidobacteria, residing in infant intestinal tract. In the current study, we report the HMO composition of 249 human milk samples, in 163 of which we quantified the abundance of members of the Bifidobacterium genus using a combination of metagenomic and flow cytometric approaches. Metagenomic data allowed us to identify four clusters dominated by Bifidobacterium adolescentis and Bifidobacterium pseudolongum , Bifidobacterium crudilactis or Bifidobacterium dentium , as well as a cluster represented by a heterogeneous mix of bifidobacterial species such as Bifidobacterium breve and Bifidobacterium longum. Furthermore, in vitro growth assays on HMOs coupled with in silico glycobiome analyses allowed us to elucidate that members of the Bifidobacterium bifidum and B. breve species exhibit the greatest ability to degrade and grow on HMOs. Altogether, these findings indicate that the bifidobacterial component of the human milk microbiota is not strictly correlated with their ability to metabolize HMOs. [ABSTRACT FROM AUTHOR]
