Resumen Introduccion Durante los ultimos anos se ha registrado un aumento creciente de la poblacion inmigrante y de viajeros procedentes de areas endemicas de filariasis producidas por Loa loa (L. loa) , Mansonella perstans (M. perstans) y Wuchereria bancrofti (W. bancrofti) . Esta situacion epidemiologica ha hecho necesario el desarrollo de tecnicas especificas de diagnostico molecular, alternativas al metodo parasitologico clasico. Por lo tanto, el objetivo planteado en este trabajo ha sido la utilizacion de las tecnicas moleculares optimizadas en nuestro laboratorio, nested-PCR e ITS1-RFLP, en el diagnostico de filariasis importadas, y la comparacion de los resultados obtenidos con los derivados del diagnostico parasitologico. Material y metodos Se ha estudiado mediante la tecnica de concentracion de Knott, nested-PCR e ITS1-RFLP un total de 523 muestras (517 sangres, 5 helmintos adultos y un humor vitreo) que fueron remitidas al Servicio de Parasitologia del Centro Nacional de Microbiologia-Instituto de Salud Carlos III, entre los anos 2006 y 2009, por 47 centros sanitarios de las comunidades autonomas. Resultados Las tecnicas moleculares utilizadas demostraron ser mas sensibles que el metodo de concentracion de Knott, tanto en el diagnostico de L. loa (n = 12 frente a n = 4) como en el de M. perstans (n = 57 frente a n = 25) en el total de muestras de sangre estudiadas. Conclusion Los metodos de PCR utilizados permiten un diagnostico especifico y mas sensible de L. loa y M. perstans en muestras clinicas de poblacion inmigrante y viajeros procedentes de areas endemicas, donde estas especies de filarias son coendemicas. El metodo de concentracion de Knott debe emplearse como tecnica complementaria siempre que sea posible.

- Abstract
En este trabajo se desarrollaron películas híbridas, adaptando el proceso sol-gel con un adecuado control en el diseño de las condiciones experimentales para poder incorporar nanopartículas de sílice a una matriz polimérica de poliestireno reciclado (PSR). Para una mejor incorporación entre la sílice y el PSR fue necesario funcionalizar el PSR con grupos carboxilo de ácido abiético.El PSR usado para la preparación de películas nanohíbridas se obtuvo de productos de empaques de comida rápida, y fue evaluada la citoxicidad de las películas sin sustrato, mostrando la viabilidad de ser usadas como películas protectoras en sustrato de vidrio ya que no presentan riesgo para la salud. Los materiales híbridos fueron caracterizados por las espectroscopias Infrarrojo y Raman, y evaluadas sus propiedades mecánicas y térmicas. La temperatura de transición vítrea se incrementó para los HPS-F and HPSR-F por la presencia del ácido abiético y la sílice. Todos los materiales incrementan su ángulo de contacto comparado con el del sustrato sin recubrir y mayor repelencia al agua. La incorporación de partículas de sílice dentro de la matriz polimérica da por resultado interesantes propiedades comparadas con sus análogas hechas con poliestireno comercial (PS), obteniendo resultados satisfactorios permitiendo así su aplicación como recubrimientos en sustratos de vidrio con propiedades de autolimpieza y protección a la radiación UVA que son de gran relevancia tecnológica.

BACKGROUND: Cryptosporidium spp. are enteric parasites that infect humans and animals. In immunocompromised patients infection can be fatal. This study was conducted to identify sub-populations of Cryptosporidium hominis and C. parvum isolates from HIV-seropositive patients in Equatorial Guinea. METHODS: In a previous study conducted in Equatorial Guinea, faecal samples from 171 HIV patients with gastrointestinal symptoms were analyzed. Of these, 13 and 17 were positive for C. hominis and C. parvum, respectively. The isolates were characterized using gp60 gene analysis. RESULTS: The gp60 gene could only be detected in 57% (17/30) of cases (10 C. parvum and 7 C. hominis). Three C. hominis (Ia, Ib and Id) and two C. parvum (IIc and IIe) subtype families were detected, including several subtypes. CONCLUSIONS: The study identified a high diversity of Cryptosporidium subtypes, suggesting that anthroponotic transmission plays an important role in the epidemiology of Cryptosporidium spp. in HIV-seropositive patients in Equatorial Guinea.

A pesar de que la Planta Internacional de Tratamiento de Aguas Residuales de Nuevo Laredo, Tamaulipas tiene una buena calidad en el efluente y un consumo energético bajo, el manejo actual de sus lodos residuales, genera algunos problemas operativos y de sostenibilidad. Por ejemplo, emite grandes cantidades de gases de efecto invernadero, así como que los lodos son considerados desechos. Por esa razón se propone el aprovechamiento de los lodos residuales para la generación de energía, a partir del biogás por medio de la digestión anaerobia. Con la implementación de esta propuesta no solamente se vería mejorado el manejo de los lodos, al disminuir su volumen, sino que disminuirían las emisiones de gases de efecto invernadero y mejoraría la eficiencia de la planta. Aunado a esto se vería beneficiado el ciclo urbano del agua de Nuevo Laredo y el ciclo hidrológico del Río Bravo por la reincorporación de los lodos a procesos productivos lo que favorece la sostenibilidad del ciclo urbano del agua en Nuevo Laredo.

The aim of the study was to perform a molecular characterization of clinical isolates of Cryptosporidium species from Equatorial Guinea. Standard laboratory methods were used to identify 35 cryptosporidiosis cases among 185 patients. PCR-RFLP successfully identified 34 Cryptosporidium species from these 35 cases, comprising C. parvum (52.9%), C. hominis (44.1%) and C. meleagridis (2.9%); over 90% of the species were isolated from HIV-positive patients. This is the first report of the molecular characterization of Cryptosporidium species isolated from humans in Equatorial Guinea and shows that zoonotic and anthroponotic transmission is present in this country.

The prevalence of Giardia duodenalis was determinate in faecal samples from dogs and cats in Madrid, Spain and molecular characterisation of isolates. A total of 604 and 144 faecal samples from dogs and cats, respectively, were analysed by routine coprological methods. The prevalence of G. duodenalis was 16.4 % (99/604) in dogs and 4.2 % (6/144) in cats. Sixty-four G. duodenalis isolates (63 from dogs and 1 from a cat) were characterised using glutamate dehydrogenase and β-giardin genes by PCR-RFLP. The single cat sample showed a mixed infection by assemblages A + F. The assemblages found in the dog samples were A, B, C, D and E, both as single and as mixed infections. The zoonotic assemblages A and B were found in 56 (88.8 %) G. duodenalis-positive samples with 15.9 % of samples having assemblage A (10/63) and 73 % of samples with assemblage B (46/63), indicating high potential zoonotic risk and public health significance.

Cryptosporidium is a significant cause of diarrheal disease in developing and industrialized nations. Cryptosporidium hominis and Cryptosporidium parvum are the main agents of cryptosporidiosis in humans. In Egypt, very little is known about genetic structure of Cryptosporidium spp. Therefore, this study was designed to examine samples from sporadic cases of cryptosporidiosis in Egyptians in order to identify the species involved in infection as well as the transmission dynamics and distribution of the parasite in the Great Cairo area. A total of 391 human faecal samples were collected, between May 2008 and March 2009, from ten public hospitals in Great Cairo. Initial screening by immunochromatographic detection kit “the Stick Crypto-Giardia; Operon” showed 23 possible positive cases. Twenty of them were confirmed by microscopic examination. PCR was performed by amplification of the oocyst wall protein (COWP) gene where 18 out of 23 samples were positive, one not detected by microscopy. Cryptosporidium genotyping was performed by RFLP analysis of PCR products of the diagnosis PCR. Only 15 samples rendered a digestion pattern. The genotyping distribution was nine cases showing C. hominis genotype, three showing C. parvum genotype and three showing mixed infection by C. hominis and C. parvum. The data showed an elevated prevalence of C. hominis (80.0%), the most anthroponotic species, suggesting a human–human transmission. Furthermore, the presence of up to 40% of samples infected with C. parvum shows that further investigations are required to determine the subgenotypes of C. parvum to clarify the mode of transmission in order to improve the control measures.

The last few years has seen an increase in the number of immigrants and travellers from endemic areas where filariasis are mainly caused by Loa loa (L. loa), Mansonella perstans (M. perstans) and Wuchereria bancrofti (W. bancrofti) species. These demographic changes has led to the need for better filariae species-specific molecular diagnostic tests to solve problems, as alternatives to the more time consuming classic parasitology methods. Thus, the objective of the present work was the implementation of optimised molecular protocols (nested-PCR and ITS1-RFLP) developed in our laboratory, for the differential diagnosis of filarial parasites. The results obtained were compared with those obtained using the conventional parasitological methods.A total of 523 samples (517 peripheral blood, 5 adult worms and one vitreous body) were sent to Parasitology Department of the National Microbiology Centre, Carlos II Research Institute (ISCIII), from 47 Health Centres in the Autonomous Regions of Spain, from 2006 to 2009. The samples were studied by the Knott technique, nested-PCR and ITS1-RFLP.The molecular techniques applied on blood samples showed to be more sensitive that Knott's concentration technique in the diagnosis of both L. loa (n=12 versus n=4) and M. perstans (n=57 versus n=25) infections.The nested-PCR and ITS1-RFLP are potential diagnostic tools for daily routine laboratory species-specific and sensitive detection of L. loa and M. perstans filarial species in immigrant population and travellers from endemic areas where these filarial species are co-endemic. Knott's concentration technique was less sensitive than molecular methods and should be carried out as a complementary diagnostic assay.

There are different protocols of molecular diagnosis methods available including DNA extraction methods to diagnose of Toxoplasma gondii, being necessary to perform comparative studies in biological samples. The aim of this study is to compare real-time PCR (rtPCR) and nested PCR (nPCR) to evaluate the detection of T. gondii in naturally infected cats. Biological samples of Toxoplasma-seropositive cats were assayed for detection of T. gondii DNA--extracted by both the lysis buffer and proteinase K (LB proteinase K) method and the acid guanidinium thiocyanate (GuSCN) method--using rtPCR and nPCR. T. gondii DNA was detected by nPCR in 43.6% and 40.8% of the samples from which it was extracted by the LB proteinase K and the GuSCN method, respectively. With rtPCR these figures fell significantly to 33.8% and 14.1%. Despite of nPCR showed higher sensitivity, the agreement observed between two PCRs was good; this agreement, however, was affected by the DNA extraction method used, LB proteinase K method showed better results.

The effect of 100 mg IM of vitamin B12, monthly, during 4 months, over anemia in premature infants was evaluated. A controlled, randomized and blinded study in 55 less than 33 weeks of gestational age babies was conduced. There was no differences in hematologyc evolution neither growth in both studied groups. The number of transfusion was significatively lower in both groups than those reported in other studies.

Las competencias ambientales son imprescindibles en el desarrollo de comportamientos que permiten el tratamiento de circunstancias complejas vinculadas al Cambio Climático global. El estudio tuvo el objetivo de identificar el nivel de competencias ambientales que poseen los estudiantes universitarios frente al Cambio Climático. Se configuró la investigación dentro del paradigma positivista, de tipo exploratorio descriptivo, diseño no experimental trasversal. La muestra quedó representada por 180 estudiantes de la carrera educación integral y educación mención computación, de una universidad pública, a este grupo se les aplicó dos cuestionarios. Los resultados evidenciaron que el nivel de competencias ambientales en los estudiantes fueron moderadas, bajas y muy bajas en sus diferentes dimensiones, conceptual, procedimental y actitudinal además de poseer bajos conocimientos sobre las causas y consecuencias del CC. Se concluye que los aprendices no están preparados para enfrentar el CC, por lo que se recomienda incorporar en los planes de la carrera de educación de ambos programas actividades que desarrollen competencias ambientales para la acción que transforme los conocimientos en función de favorecer el cambio de actitudes y patrones de vida que esten alineados a pautas de la Educación para el Desarrollo Sostenible, el cual persigue el empleo adecuado de la energía, el reconocimiento, valor y preservación de los recursos de la naturaleza en pos de la mitigación de los eventos naturales peligrosos vinculados al Cambio Climático, así como también a su adaptación.

El objetivo del estudio fue determinar la influencia de las acciones gerenciales y el rol del docente universitario en el área de investigación en las carreras de Educación de una Universidad Pública Venezolana. Se configuró metodológicamente dentro de un enfoque cuantitativo, tipo descriptivo-correlacional, diseño no experimental transeccional, la muestra quedó representada por 58 docentes del área Ciencias de la Educación. Se empleó la encuesta y dos cuestionarios. Los resultados permitieron identificar que las acciones gerenciales desarrolladas por la coordinación de investigación de la institución son moderadas y bajas, lo que condiciona el logro de objetivos organizacionales con relación al área de investigación, asimismo, se identificó que el rol docente en el área de investigación de la Carrera de Educación, programas educación integral y computación arrojó funciones deficientes, por lo que se requiere de acciones gerenciales que contribuyan a fortalecer el rol del docente en el área de investigación. Por otro lado, se determinó que existe una correlación positiva media entre las variables, por lo tanto, si influye las acciones gerenciales en el rol del docente en el área de investigación, de esta manera es necesario reestructurar y proponer acciones gerenciales en el área de investigación para que el docente sea más competitivo y productivo en el desarrollo de su función como docente investigador.

Under dysbiosis, a gut metabolic disorder, short-chain carboxylic acids (SCCAs) are secreted to the lumen, affecting colorectal cancer (CRC) development. Butyrate and propionate act as CRC growth inhibitors, but they might also serve as carbon source. In turn, the roles of acetate as metabolic fuel and protein acetylation promoter have not been clearly elucidated. To assess whether acetate favors CRC growth through active mitochondrial catabolism, a systematic study evaluating acetate thiokinase (AcK), energy metabolism, cell proliferation, and invasiveness was performed in two CRC cell lines incubated with physiological SCCAs concentrations. In COLO 205, acetate (+glucose) increased the cell density (50%), mitochondrial protein content (3–10 times), 2-OGDH acetylation, and oxidative phosphorylation (OxPhos) flux (36%), whereas glycolysis remained unchanged vs. glucose-cultured cells; the acetate-induced OxPhos activation correlated with a high AcK activity, content, and acetylation (1.5–6-fold). In contrast, acetate showed no effect on HCT116 cell growth, OxPhos, AcK activity, protein content, and acetylation. However, a substantial increment in the HIF-1α content, HIF-1α-glycolytic protein targets (1–2.3 times), and glycolytic flux (64%) was observed. Butyrate and propionate decreased the growth of both CRC cells by impairing OxPhos flux through mitophagy and mitochondrial fragmentation activation. It is described, for the first time, the role of acetate as metabolic fuel for ATP supply in CRC COLO 205 cells to sustain proliferation, aside from its well-known role as protein epigenetic regulator. The level of AcK determined in COLO 205 cells was similar to that found in human CRC biopsies, showing its potential role as metabolic marker.

Los estudios organizacionales corresponden a las diversas realidades sociales que conciernen a las relaciones e interacciones dinámicas que se presentan entre éstas y el entorno en el que se desenvuelven. Por lo tanto, de acuerdo con el criterio de las investigadoras se plantea como premisa principal entender, comprender y explicar a toda organización, desde el enfoque holístico y sistémico que conforman el todo y el entorno de la organización puesto que se requieren de nuevos paradigmas organizacionales adaptados a la realidad. En tal sentido, se presenta como propósito general analizar desde la visión epistemológica el enfoque holístico y sistémico en los estudios organizacionales. Asimismo, desde la mirada epistemológica de la presente investigación se encuentra contextualizado desde el diseño de investigación documental y del paradigma cualitativo, puesto que el abordaje metodológico de la investigación responde a un determinado enfoque o visión de la realidad, que permitieron realizar un proceso de análisis a través de una matriz ilustrativa que proporcionará las reflexiones en la investigación. Por lo que, como principal reflexión investigativa se resalta que la visión epistemológica de los estudios organizacionales se debe a un proceso de integración entre investigar, comprender, analizar y reflexionar acerca del fenómeno organizacional.

Quantitative measurements of performance parameters have the potential to increase consistency and enhance performance of the surfaces as well as to contribute to the safety of horses and riders. This study investigates how factors known to influence the performance of the surface, incorporation of a drainage package, control of the moisture control, and introduction of a geotextile reinforcement, affect quantitative measurements of arena materials. The measurements are made by using affordable lightweight testing tools which are readily available or easily constructed. Sixteen boxes with arena materials at a consistent depth were tested with the Going Stick (GS), both penetration resistance and shear, the impact test device (ITD), and the rotational peak shear device (RPS). Volumetric moisture content (VMC %) was also tested with time–domain reflectometry (TDR). Results obtained using GS, RPS, ITD, and TDR indicate that the presence of the drainage package, moisture content, and geotextile addition were detected. Alterations due to combinations of treatments could also be detected by GS, ITD, and TDR. While the testing showed some limitations of these devices, the potential exists to utilize them for quality control of new installations as well as for the monitoring of maintenance of the surfaces.

This study shows that the non-steroidal anti-inflammatory drug (NSAID) celecoxib and its non-cyclooxygenase-2 (COX2) analogue dimethylcelecoxib (DMC) exert a potent inhibitory effect on the growth of human cervix HeLa multi-cellular tumor spheroids (MCTS) when added either at the beginning (“preventive protocol”; IC50 = 1 ± 0.3 nM for celecoxib and 10 ± 2 nM for DMC) or after spheroid formation (“curative protocol”; IC50 = 7.5 ± 2 µM for celecoxib and 32 ± 10 µM for DMC). These NSAID IC50 values were significantly lower than those attained in bidimensional HeLa cells (IC50 = 55 ± 9 µM celecoxib and 48 ± 2 µM DMC) and bidimensional non-cancer cell cultures (3T3 fibroblasts and MCF-10A mammary gland cells with IC50 from 69 to >100 µM, after 24 h). The copper-based drug casiopeina II-gly showed similar potency against HeLa MCTS. Synergism analysis showed that celecoxib, DMC, and casiopeinaII-gly at sub-IC50 doses increased the potency of cisplatin, paclitaxel, and doxorubicin to hinder HeLa cell proliferation through a significant abolishment of oxidative phosphorylation in bidimensional cultures, with no apparent effect on non-cancer cells (therapeutic index >3.6). Similar results were attained with bidimensional human cervix cancer SiHa and human glioblastoma U373 cell cultures. In HeLa MCTS, celecoxib, DMC and casiopeina II-gly increased cisplatin toxicity by 41–85%. These observations indicated that celecoxib and DMC used as adjuvant therapy in combination with canonical anti-cancer drugs may provide more effective alternatives for cancer treatment.

Background and Objectives: Maxillary bone defects related to post-extraction alveolar ridge resorption are usual. These defects may lead to failure in further surgical implant phases given the lack of bone volume to perform the dental implant. The objective of this clinical assay was to evaluate the safety and efficacy of an experimental synthetic bone substitute in the preservation of post-extraction maxillary alveoli. Materials and Methods: 33 voluntary patients who had at least one maxillary premolar tooth that was a candidate for exodontia (n = 39) and subsequent implant rehabilitation participated. The regenerated alveoli were monitored by means of periodic clinical examinations (days 9 ± 1, 21 ± 4, 42 ± 6, and 84 ± 6), measuring the height and width of the alveolar crest (days 0 and 180 ± 5), measurement of radiodensity using tomographic techniques (days 0−5 and 175 ± 5), and histological examination of biopsies collected at 180 ± 5 days. Results: No significant differences were observed during the entire follow-up period between the two groups with respect to the safety variables studied. A variation in width of −0.9 ± 1.3 mm and −0.6 ± 1.5 mm, and a variation in height of −0.1 ± 0.9 mm and −0.3 ± 0.7 mm was observed for experimental material Sil-Oss® and Bio-Oss®, respectively. The radiodensity of the alveoli regenerated with the experimental material was significantly lower than that corresponding to Bio-Oss®. However, the histological study showed greater osteoid matrix and replacement of the material with newformed bone in the implanted beds with the experimental material. Conclusions: Both materials can be used safely and proved equally effective in maintaining alveolar flange dimensions, they are also histologically biocompatible, bioactive and osteoconductive. The experimental material showed the advantage of being resorbable and replaced with newformed bone, in addition to promoting bone regeneration.

El objetivo de este trabajo fue analizar el comportamiento germinativo de semillas dePaspalum dilatatum Poir. provenientes de poblaciones de posición topográfica intermedia y baja y de diferentes historias de uso (clausura y pastoreo). Se utilizaron condiciones de incubación de alternancia de luz y temperatura (20oC 16 hs. oscuridad - 35oC 8 hs.luz) y oscuridad y temperatura constante (25oC). Previo a la incubación la mitad de las semillas se colocaron en condiciones de inmersión, por la relación de estas poblaciones con eventos naturales de inundación. Se estimó la germinación total a través del número de semillas germinadas a los 30 días y la germinación inicial a través de un índice basado en el descripto por Maguire.Tanto la inmersión en agua como la incubación en condiciones de luz y alternancia de temperaturas aumentaron significativamente la germinación inicial y total. La germinación inicial y totaltambién fue afectada por la condición de incubación mostrando en condiciones de luz y alternancia de temperatura valores superiores. La germinación inicial mostró interacciones significativas entre la ubicación topográfica y la condición de incubación y entre esta última y la historia de uso.Las diferencias en la germinación inicial entre condiciones de incubación fueron mayores en las poblaciones deposición topográfica intermedia que en las de posición media y en las provenientes de ambientes pastoreados que en las de clausuras. La interacción entre historia de uso y condición de incubación puede ser explicada por las diferencias en los tamaños de semillas generados en ambientes pastoreados y clausurados respectivamente.
