53. Limonene extraction from the zest of Citrus sinensis, Citrus limon, Vitis vinifera and evaluation of its antimicrobial activity

Abstract

Citrus rinds contain essential oils. One of the major constituents of the essential oils in the zest of different fruits like Citrus sinensis, C. limon, and Vitis vinifera is limonene. In this research, limonene was extracted by hydro-distillation method using Clevenger set up and its antimicrobial activity against certain bacterial and fungal strains was determined by using Kirby Bauer’s disc diffusion method. The primary antimicrobial screening of limonene without dilution exhibited a zone of inhibition (mm) comparable to Ampicillin (20mg/ml) and Amphotericin B (20mg/ml). The effect of pure limonene against all strains used was high as compared to the isolated samples. The MIC values also showed an expected decrease in the zone of inhibition from 1:2 to 1:8 dilutions. Based on this study, the cost-effective isolation of limonene and other essential oils is quite possible.

Abstract

The family Nodaviridae's neural necrosis virus (NNV) causes viral encephalopathy and retinopathy in a wide range of fish species, resulting in widespread death. This research suggests a less expensive and more alternative strategy to developing vaccinations against diseases that are causing fisheries productivity to decline. More than 40 species of Teleosts are affected by Betanodavirus in their fries and larval stages. Coat protein and its interactions with neural receptor domains aid viral RNA entrance into the target neuron cytosol. We have selected epitope CRWSVRLSV from coat proteins. As per bio-computational tools greater than ∼1000 epitopes are selected. Six immunogenic epitopes were then further analyzed by molecular docking as well as simulation study to bring out finalized epitope CRWSVRLSV. We generated an epitope derived from Capsid protein that can be easily targeted by fish immune systems, as evidenced by basic laboratory confirmations. From the results represented in the study, it can be considered that CRWSVRLSV (up to 40 pg/mL) showed the antigen and antibody reaction and found concentration of the antibody was in the range of 200.61538 to 946.775. Abstract

The present study aimed to evaluate effects of aging on tensile strength of FRP composite. A hybrid composite is FRP composite that includes any foreign materials. This research aims comparison of tensile strength after more than 1-year manufacturing of GFRP and hybrid composite. The hybrid composite used in this research is generally a GFRP (Glass Fiber Reinforced Plastic) composite which includes a plain weave copper strips mesh which is placed in between the many layers of fiberglass. Both GFRP and hybrid composite were fabricated using CSM fiberglass as reinforcement at 20%. Matrix material was epoxy resin LY556 used. There is a single test is performed on both type of GFRP and hybrid composite which is a tensile strength test with an interval of more than one year. [ABSTRACT FROM AUTHOR]

Abstract

Background: AKT1 is a serine/threonine kinase necessary for the mediation of apoptosis, angiogenesis, metabolism, and cell proliferation in both normal and cancerous cells. The mutations in the AKT1 gene have been associated with different types of cancer. Further, the AKT1 gene mutations are also reported to be associated with other diseases such as Proteus syndrome and Cowden syndromes. Hence, this study aims to identify the deleterious AKT1 missense SNPs and predict their effect on the function and structure of the AKT1 protein using various computational tools. Methods: Extensive in silico approaches were applied to identify deleterious SNPs of the human AKT1 gene and assessment of their impact on the function and structure of the AKT1 protein. The association of these highly deleterious missense SNPs with different forms of cancers was also analyzed. The in silico approach can help in reducing the cost and time required to identify SNPs associated with diseases. Results: In this study, 12 highly deleterious SNPs were identified which could affect the structure and function of the AKT1 protein. Out of the 12, four SNPs—namely, G157R, G159V, G336D, and H265Y—were predicted to be located at highly conserved residues. G157R could affect the ligand binding to the AKT1 protein. Another highly deleterious SNP, R273Q, was predicted to be associated with liver cancer. Conclusions: This study can be useful for pharmacogenomics, molecular diagnosis of diseases, and developing inhibitors of the AKT1 oncogene. [ABSTRACT FROM AUTHOR]

Abstract

Background: Cancer, an uncontrolled multistage disease causing swift division of cells, is a leading disease with the highest mortality rate. Cellular heterogeneity, evading growth suppressors, resisting cell death, and replicative immortality drive the tumor progression by resisting the therapeutic action of existing anticancer drugs through a series of intrinsic and extrinsic cellular interactions. The innate cellular mechanisms also regulate the replication process as a fence against proliferative signaling, enabling replicative immortality through telomere dysfunction. Area Covered: The conventional genotoxic drugs have several off-target and collateral side effects associated with them. Thus, the need for the therapies targeting cyclin-dependent kinases or P13K signaling pathway to expose cancer cells to immune destruction, deactivation of invasion and metastasis, and maintaining cellular energetics is imperative. Compounds with anticancer attributes isolated from plants and rich in alkaloids, terpenes, and polyphenols have proven to be less toxic and highly targetspecific, making them biologically significant. This has opened a gateway for the exploration of more novel plant molecules by signifying their role as anticancer agents in synergy and alone, making them more effective than the existing cytotoxic regimens. Expert Opinion: In this context, the current review presented recent data on cancer cases around the globe, along with discussing the fundamentals of proliferative signaling and replicative immortality of cancer cells. Recent findings were also highlighted, including antiproliferative and antireplicative action of plant-derived compounds, besides explaining the need for improving drug delivery systems.
