Abstract

Introduction: Human ABO blood groups antigens exhibit alternative phenotypes and genetically derived glycoconjugate structures that are present on the surface of red blood cells and play an active role in the cellular Physiology and Pathology. Blood groups antigens are thought to be among hereditary determinate and play a vital role to understand genetics and disease susceptibility. Biological sample: Human Blood Results: Shows highest prevalence of T2DM patients having blood group B followed by blood group O, A & AB and Table.7 show prevalence of blood group in general population. Abstract

While risk of fomite transmission of SARS-CoV-2 is considered low, there is limited environmental data within households. This January—April 2021 investigation describes frequency and types of surfaces positive for SARS-CoV-2 by real-time reverse transcription polymerase chain reaction (RT-PCR) among residences with ≥1 SARS-CoV-2 infection, and associations of household characteristics with surface RT-PCR and viable virus positivity. Of 1232 samples from 124 households, 27.8% (n = 342) were RT-PCR positive with nightstands (44.1%) and pillows (40.9%) most frequently positive. SARS-CoV-2 lineage, documented household transmission, greater number of infected persons, shorter interval between illness onset and sampling, total household symptoms, proportion of infected persons ≤12 years old, and persons exhibiting upper respiratory symptoms or diarrhea were associated with more positive surfaces. Viable virus was isolated from 0.2% (n = 3 samples from one household) of all samples. This investigation suggests that while SARS-CoV-2 on surfaces is common, fomite transmission risk in households is low. [ABSTRACT FROM AUTHOR]

Abstract

The brain is a sexually dimorphic organ that implies different functions and structures depending on sex. Current pharmacological approaches against different neurological diseases act distinctly in male and female brains. In all neurodegenerative diseases, including Alzheimer’s disease (AD), sex-related outcomes regarding pathogenesis, prevalence, and response to treatments indicate that sex differences are important for precise diagnosis and therapeutic strategy. Pathogenesis of AD includes vascular dementia, and in most cases, this is accompanied by metabolic complications with similar features as those assembled in diabetes. This review discusses how AD-associated dementia and diabetes affect cognition in relation to sex difference, as both diseases share similar pathological mechanisms. We highlight potential protective strategies to mitigate amyloid-beta (Aβ) pathogenesis, emphasizing how these drugs act in the male and female brains.

Abstract

A reversible decrease or increase of Candida albicans chromosome copy number was found to be a prevalent means of survival of this opportunistic pathogen, under conditions that kill cells or inhibit their propagation. The utilization of a secondary carbon source, L-sorbose, by reversible loss of chromosome 5, serves as a model system. We have determined that an [approximately equal to] 209-kbp portion of the right arm of chromosome 5 contains at least five spatially separated, functionally redundant regions that control utilization of L-sorbose. The regions bear no structural similarity among themselves, and four of them contain sequences that bear no similarity with any known sequence. We identified a regulatory gene in region A that encodes a helixloop-helix protein. Most important, the multiple redundant regulators scattered along chromosome 5 explain, in a simple, elegant way, why the loss of the entire homologue is usually required for growth on sorbose. Thus, an entire chromosome acts as a single regulatory unit, a feature not previously considered. Our finding appears to be a paradigm for the control of other phenotypes in C. albicans that also depend on chromosome loss, thus implying that C. albicans genes are not distributed randomly among different chromosomes. chromosome-5 monosomy | secondary carbon sources | helix-loop-helix proteins

86. RP-HPLC estimation of thermal oxidation of thymoquinone in Nigella sativawith reference to its antioxidant potential and vis-a-vis in-vitroDNA damage inhibition

Abstract

Nigella sativa(NS), a popular herbal plant is known for its imperative therapeutic potentials. NS possesses active phytochemicals with unique antioxidant properties. Thymoquinone (TQ) is one of the active phytochemical of NS. Our investigation aimed to elucidate the effect of thermal oxidation of NS and TQ for antioxidant property and its effect on DNA damage inhibition (DDI). In the present study methanolic NS extract (NSE) and standard TQ were treated at different temperatures, at a range of 0 to 100 °C. The antioxidant potential was correlated to in-vitroDDI activity. The results elucidated that maximum TQ content was observed at 0 °C for NSE (18.545 ±  2.063%), while nil (0.000 ±  0.000%) was found at 100 °C for NSE. However, for standard TQ maximum content (99.086  ±  0.470%) was observed at 0 °C and minimum content (42.783 ±  3.233%) was observed at 100 °C. When the antioxidant activity was estimated, again a linear decline was observed. NSE kept at 0 °C showed maximum activity (4.252 ±  0.406%), while, at 100 °C showed minimum activity (0.621 ±  0.158%). For standard TQ maximum activity was observed at 0 °C (3.295 ±  0.265%), while the minimum was recorded at 100 °C (0.337 ± 0.087%). With consideration to positive antioxidant activity, in-vitroDDI was targeted. NSE kept at 0 °C demonstrated maximum DDI activity (24.145 ± 2.508%), while the minimum was observed at 100 °C (11.964 ±  1.768%). Standard TQ showed abridged results when compared to NSE. It showed only 17.991 ±  2.077% DDI activity with the sample kept at 0 °C, while with the sample at 100 °C was least effective for DDI (3.442 ±  0.608%). Electrophoresis study clearly reveals the degradation of calf thymus DNA upon oxidative stress. However, the antioxidant potential of NSE and standard TQ inhibited the calf thymus DNA damage. Statistically, a significant variation with p-value≤ 0.05 was observed when two-way ANOVA was applied between the various parameters studied. When correlation was established between the various parameters, a strong positive correlation of approximately 0.9 was observed. Thus, it can be concluded that variation in temperature causes an effect on TQ content, thereby, affecting its antioxidant property due to thermal oxidation. It was observed that within the temperature range of 60 to 100 °C, TQ was quite stable but antioxidant potential was deprived as the temperature increased, thereby affecting DDI activity. Overall, our study is an open-end finding for future investigation in the development of drug formulation using NSE or its active phytochemical TQ.

Abstract

Background Candida auris is an emerging multidrug-resistant yeast that contaminates healthcare environments causing healthcare-associated outbreaks. The mechanisms facilitating contamination are not established. Methods C. auris was quantified in residents’ bilateral axillary/inguinal composite skin swabs and environmental samples during a point-prevalence survey at a ventilator-capable skilled-nursing facility (vSNF A) with documented high colonization prevalence. Environmental samples were collected from all doorknobs, windowsills and handrails of each bed in 12 rooms. C. auris concentrations were measured using culture and C. auris-specific quantitative polymerase chain reaction (qPCR) The relationship between C. auris concentrations in residents’ swabs and associated environmental samples were evaluated using Kendall’s tau-b (τ b) correlation coefficient. Results C. auris was detected in 70/100 tested environmental samples and 31/57 tested resident skin swabs. The mean C. auris concentration in skin swabs was 1.22 × 105 cells/mL by culture and 1.08 × 106 cells/mL by qPCR. C. auris was detected on all handrails of beds occupied by colonized residents, as well as 10/24 doorknobs and 9/12 windowsills. A positive correlation was identified between the concentrations of C. auris in skin swabs and associated handrail samples based on culture (τ b = 0.54, P = .0004) and qPCR (τ b = 0.66, P = 3.83e−6). Two uncolonized residents resided in beds contaminated with C. auris. Conclusions Colonized residents can have high C. auris burdens on their skin, which was positively related with contamination of their surrounding healthcare environment. These findings underscore the importance of hand hygiene, transmission-based precautions, and particularly environmental disinfection in preventing spread in healthcare facilities.
