51. Proteins in deep eutectic solvents: Structure, dynamics and interactions with the solvent

Abstract: The structure and dynamics of proteins in solution are closely related to their activity and, ultimately, to their performance in products and technologies, for example, drug formulations and enzymatic catalysis. With this in mind, the study of the fundamental behavior of proteins in deep eutectic solvents not only provides insight into a novel alternative to aqueous and molecular organic solvents, but also contributes to understanding the potential behavior of these macromolecules in non-aqueous environments. In particular, it has relevance for systems with high levels of dehydration, liquid phases for dispersions at cryogenic temperatures, or systems with high ionic strength. In this chapter, we will explore current knowledge of the physicochemical behavior of proteins in deep eutectic solvents. The local and global structure of proteins in these solvents will be presented and compared to those in aqueous solutions, organic solvents and ionic liquids. Furthermore, the characteristic behavior of proteins in deep eutectic solvents will be linked to solvation effects and the interaction of the macromolecules with the solvent, where specific electrostatic and hydrogen bond interactions can affect the structure and dynamics of proteins. Finally, challenges and opportunities in the field will be discussed, with the aim of guiding a common effort to develop a better understanding of the fundamental behavior of proteins in these interesting environments.

Abstract: Deep eutectic solvents (DES) are potentially greener solvents obtained through the complexation of simple precursors which, among other applications, have been investigated in recent years for their ability to support the self-assembly of amphiphilic molecules. It is crucial to understand the factors which influence surfactant solubility and self-assembly with respect to the interaction of the surfactant molecule with the DES components. In this work, small-angle neutron scattering (SANS) has been used to investigate the micellization of cationic (CnTAB) and anionic (SDS) surfactants in a ternary DES comprising choline chloride, urea, and glycerol, where the hydrogen bond donors are mixed in varying molar ratios. The results show that in each case either globular or rodlike micelles are formed with the degree of elongation being directly dependent on the composition of the DES. It is hypothesized that this composition dependence arises largely from the poor solubility of the counterions in the DES, especially at low glycerol content, leading to a tighter binding of the counterion to the micelle surface and giving rise to micelles with a high aspect ratio. This potential for accurate control over micelle morphology presents unique opportunities for rheology control or to develop templated syntheses of porous materials in DES, utilizing the solvent composition to tailor micelle shape and size, and hence the pore structure of the resulting material.

Abstract: MicroRNAs (miRNAs) are a novel class of biomarkers that can be present in different biological samples, which can be related to many diseases. Controlled aggregation of gold nanoparticles (GNPs) in the presence of miRNAs can be used to perform a rapid detection of miRNAs based on color change. Sometimes the color change can be observed with the naked eye; however, this is not always possible. This work proposes the design of a novel instrumental prototype that allows performance of simultaneous transmittance measurements at different wavelengths. Once the signals are obtained, the system calculates the 525nm/650nm transmittance ratio and the miRNA concentration, displaying and recording the results in a smartphone. This prototype has been validated to detect the miRNA "miR146a," which is found in higher concentrations in milk and tissues in cases of bovine mastitis.

55. Prevention of oronasal fistula in primary palatoplasty with acellular dermal matrix

Abstract: Palatal fistula after primary palatoplasty remains one of the most difficult complications that plastic surgeons have to face. Once the fistula has developed, the complexity of the reconstruction increases significantly, and the revisional surgery results are not promising. The recent use of acellular dermal matrix (ADM) provides a new perspective for the reconstructive surgeon. Our goal is to reduce and prevent the postoperative oronasal fistula in patients with cleft palate by placing an ADM in the zone 3 and 4 of Pittsburgh (hard palate and hard–soft palate junction), creating an extra layer for support of the cleft palate. Seven children diagnosed of palatal cleft with high risk of developing oronasal fistula were operated. The technique of choice was two-flap palatoplasty with lateral incisions and intravelar veloplasty and three-layered closure with ADM in the hard palate and hard–soft palate junction. One patient developed a 3-mm oronasal fistula at 3-month follow-up, with spontaneous closure at 12 months. The rest of the children had an uneventful evolution. Our rate of postoperative oronasal fistula was 0% at 12 months. The use of ADM in primary palatoplasty had been of great use to prevent complications in high-risk oronasal fistula patients.

Abstract: The interactions between protein and surfactants play an important role in the stability and performance of formulated products. Due to the high complexity of such interactions, multi-technique approaches are required to study these systems. Here, an integrative approach is used to investigate the various interactions in a model system composed of human growth hormone and sodium dodecyl sulfate. Contrast variation small-angle neutron scattering was used to obtain information on the structure of the protein, surfactant aggregates and surfactant–protein complexes. 1H and 1H–13C HSQC nuclear magnetic resonance spectroscopy was employed to probe the local structure and dynamics of specific amino acids upon surfactant addition. Through the combination of these advanced methods with fluorescence spectroscopy, circular dichroism and isothermal titration calorimetry, it was possible to identify the interaction mechanisms between the surfactant and the protein in the pre- and post-micellar regimes, and interconnect the results from different techniques. As such, the protein was revealed to evolve from a partially unfolded conformation at low SDS concentration to a molten globule at intermediate concentrations, where the protein conformation and local dynamics of hydrophobic amino acids are partially affected compared to the native state. At higher surfactant concentrations the local structure of the protein appears disrupted, and a decorated micelle structure is observed, where the protein is wrapped around a surfactant assembly. Importantly, this integrative approach allows for the identification of the characteristic fingerprints of complex transitions as seen by each technique, and establishes a methodology for an in-detail study of surfactant–protein systems.

Abstract: Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

58. Imported schistosomiasis in travelers: Experience from a referral tropical medicine unit in Barcelona, Spain

Abstract: Background: Acute schistosomiasis occurs most often in travelers to endemic regions. The aim of the study is to describe the epidemiological, clinical and parasitological characteristics of patients with schistosomiasis acquired during an international travel. Methods: Observational retrospective study including all travel-related schistosomiasis cases seen at the International Health Unit Vall d’Hebron-Drassanes (Barcelona, Spain) from 2009 to 2022. Diagnosis of schistosomiasis was defined by the presence of Schistosoma eggs in stools or urine or the positivity of a serological test. We collected demographic, epidemiological, clinical, parasitological, and therapeutic information. Results: 917 cases of schistosomiasis were diagnosed, from whom 96 (10.5 %) were travel-related. Mean age of the patients was 34.9 years, and 53.1 % were women. Median duration of the travel was 72 days, and geographical areas where travelers had contact with fresh water were Africa (82.3 %), Asia (12.5 %), and South America (5.2 %). Twenty (20.8 %) patients reported having had some clinical symptom, being gastrointestinal symptoms the most frequent. Two patients developed the classical Katayama syndrome. In eleven (11.5 %) cases eggs were observed in urine or feces samples, and 85 (88.5 %) cases were diagnosed by a positive serology. Ninety-one (94.8 %) patients received treatment with praziquantel with different therapeutic schemes. The two patients with Katayama syndrome received concomitant treatment with corticosteroids. Conclusions: Schistosomiasis in travelers represented 10 % of the overall schistosomiasis cases in our center. Increasing the awareness in the pre-travel advice and implementing specific screening in those travelers at risk (long travelers, contact with fresh water) could reduce the incidence and associated morbidity in this group.

Abstract: AIM: To assess the efficacy and safety of non-penetrating deep sclerectomy (NPDS) with uveoscleral implant plus subconjunctival and intrascleral collagen matrix overcoming the superficial scleral flap lips (modified deep sclerectomy technique, DS) and minimal use of mitomycin C in glaucoma surgery. METHODS: A retrospective review of 47 consecutive glaucoma patients who underwent NPDS with DS between January 2017 and May 2018. Best-corrected visual acuity, intraocular pressure (IOP), post-operative need for glaucoma medications, visual field mean deviation (MD), re-interventions, needling revisions and laser goniopuncture were noted. Absolute success was defined as IOP≤18 mm Hg without topical medication. Relative success was defined as the same criteria but with the addition of any antihypertensive medication. IOP over 18 mm Hg on two consecutive follow-up visits was considered as a failure. RESULTS: Fifty-two eyes of 47 patients were evaluated. Abstract: Introduction: Treatment of MDRTB is complex: regimens require microbiological data; adverse events are frequent; availability of drugs and authorization for new drugs varies. The aim of this study was to scope the available national resources. Method: A survey to determine whether practising physicians could access MDRTB advice was sent to TBnet members by email. The ERS Office also contacted national respiratory societies. Questions included the name and contact details for their national advisory service, whether it was national policy to use the service for each patient with MDRTB and whether advice was required to access bedaquiline and delaminid. Results: 65 replies were received (14 were uninformative). 26/31 EU/EEA and 10/19 other countries in the WHO European Region were represented. 7 countries referred all MDRTB to a tertiary centre; 12 countries had tertiary referral centres that also gave advice to physicians treating MDRTB. 11 countries had an electronic system for advice, 6 with multidisciplinary team meetings to review patients’ progress. Lead clinicians were identified for a further 8 countries, one of which had no national advisory service and the other 7 had not responded by the time of abstract submission. For 18 (58%) countries, discussion of MDRTB by a national /regional committee was national policy and most (15/18) required consultation to use bedaquiline or delamanid. Electronic platforms had a wide range of functionality but few retained anonymised data and audited patient outcomes. Conclusion: MDRTB management is often concentrated in tertiary centres. Clinical governance, regarding audit and outcome, are at an early stage in managing MDRTB.

Abstract: Secondary bacterial pneumonias following influenza infections consistently rank within the top ten leading causes of death in the United States. To date, murine models of co-infection have been the primary tool developed to explore the pathologies of both the primary and secondary infections. Despite the prevalence of this model, considerable discrepancies regarding instillation procedures, dose volumes, and efficacies are prevalent among studies. Furthermore, these efforts have been largely incomplete in addressing how the pathogen may be directly influencing disease progression post-infection. Herein we provide a precise method of pathogen delivery, recovery, and analysis to be used in murine models of secondary bacterial pneumonia. We demonstrate that intratracheal instillation enables an efficient and accurate delivery of controlled volumes directly and evenly into the lower respiratory tract. Lungs can be excised to recover and quantify the pathogen burden. Following excision of the infected lungs, we describe a method to extract high quality pathogen RNA for subsequent transcriptional analysis. This procedure benefits from being a non-surgical method of delivery without the use of specialized laboratory equipment and provides a reproducible strategy to investigate pathogen contributions to secondary bacterial pneumonia.

Abstract: In this work, we present the first example of the self-assembly of phospholipid monolayers at the interface between air and an ionic solvent. Deep eutectic solvents are a novel class of environmentally friendly non-aqueous room temperature liquids with tunable properties, that have wide ranging potential applications and are capable of promoting the self-assembly of surfactant molecules. We use a chemically-consistent Bayesian modelling of X-ray and neutron reflectometry measurements to show that these monolayers broadly behave as they do on water. This method allows for the monolayer structure to be determined, alongside the molecular volumes of the individual monolayer components without the need for water-specific constraints to be introduced. Furthermore, using this method we are able to better understand the correlations present between parameters in the analytical model. Abstract: Este es un libro colectivo. En más de un sentido. En primer lugar, recoge dinámicas y estrategias enfocadas a la creación literaria entre bachilleres, diseñadas e implementadas por pasantes o egresados de la carrera de Letras Hispánicas de la Universidad de Guadalajara que participan en el proyecto Luvina Joven, que coordina talleres de creación para estudiantes de preparatoria del Sistema de Educación Media Superior (sems) y algunos centros de la red universitaria. Pero a la vez estas actividades se nutren de un gran número de fuentes, muchas de ellas anónimas, porque derivan de las experiencias individuales de cada uno de los coordinadores de los talleres, recogidas a su vez de la participación tanto sistemática como informal en diversos espacios que se enfocan a estos mismos propósitos. La buena educación, a final de cuentas, es colectiva, y habría que remontarnos a nuestros maestros, y a los maestros de nuestros maestros hasta llegar al Primer Maestro, y seguir el proceso para identificar quién enseñó qué, con la consideración de que aquello que yo aprendí lo enseñaré a mi manera, y si mi alumno se vuelve maestro, continuará este proceso sin fin.

Abstract: Food safety control is a key issue in the food and agriculture industries. For such purposes, developing miniaturized analytical methods is critical for enabling the rapid and sensitive detection of food supplements, allergens, and pollutants. Here, a novel bioanalytical methodology based on DNA-functionalized gold nanoparticles (AuNPs) and colorimetric detection was developed to detect the presence of sesame (a major allergen) through sesame seed DNA as a target, in food samples. The presence of sesame DNA induces controlled nanoparticle aggregation/desegregation, resulting in a color change (from blue to red) proportional to sesame DNA concentration. The incorporation of multicomponent nucleic acid enzymes (MNAzymes) in this strategy has been carried out to perform an isothermal signal amplification strategy to improve the sensitivity of detection. Also, open-source software for color analysis was used to ensure an unbiased visual color-change detection, enhancing detection accuracy and sensitivity and opening the possibility of performing a simple and decentralized analyte detection. The method successfully detected the presence of sesame DNA in sesame seed, sesame oil, olive oil, and sunflower oil. In brief, the developed approach constitutes a simple and affordable alternative to perform a highly sensitive detection of DNA in food without complex methodologies or the requirement of expensive instrumentation.

Abstract: Background: Imported strongyloidiasis in non-endemic countries has increasingly been diagnosed. The aim of the present study is to describe the main epidemiological and clinical characteristics of patients with imported strongyloidiasis attended in a referral International Health Unit and to detect trend changes over a 12-year period. Methods: This is an observational retrospective study including all imported strongyloidiasis cases seen at the International Health Unit Vall d’Hebron-Drassanes (Barcelona, Spain) from January 2009 to December 2020. Epidemiological and clinical characteristics from included patients were collected. Results: Overall, 865 cases of imported strongyloidiasis were diagnosed, of whom 472 (54.6 %) were men and mean age was 38.7 (SD 13.4) years. Most cases were diagnosed in migrants (830, 96 %). The distribution of the geographic origin was: Latin America (561, 67.6 %), Sub-Saharan Africa (148, 17.8 %), Asia (113, 13.6 %), North Africa (5, 0.6 %), Eastern Europe (2, 0.2 %), and North America (1, 0.1 %). The main reasons for consultation at the Unit were screening of health status (371, 42.9 %), laboratory test alteration (367, 42.4 %), gastrointestinal symptoms (56, 6.5 %), cutaneous symptoms (26, 3 %), and other clinical symptoms (45, 5.2 %). An increase in the number of cases was observed in the last years of the study period. Conclusions: Imported strongyloidiasis has increasingly been diagnosed in our referral unit, mostly due to screening strategies implementation. Most of the patients were young migrants coming from Latin America, with no symptoms at the time of diagnosis. The optimization of screening strategies will increase the detection and treatment of cases, reducing potential complications.

Abstract: Resumen Introducción y Objetivo El desbridamiento enzimático es hoy una herramienta útil en el tratamiento precoz de las quemaduras profundas. Tras el mismo, existen diferentes protocolos de manejo de la herida en las distintas unidades de quemados, consiguiendo en algunos casos la curación espontánea y en otros necesitando injertos de piel. A día de hoy no existe ninguna herramienta objetiva para asegurar el éxito del desbridamiento, ya que el lecho de herida que deja se asemeja a una escara que puede generar confusión sobre la eficacia del producto y el manejo del paciente. El desarrollo de una técnica fiable y objetiva para la medición de la dermis remanente podría ayudar en la toma de decisiones postdesbridamiento y en consecuencia a lograr mejores resultados. Material y Método Realizamos ultrasonografía (US) cutánea a tiempo real con sondas de alta frecuencia en 15 pacientes, 12 horas tras desbridamiento enzimático con Nexobrid®, proporcionando imágenes precisas del remanente dérmico. Medimos el grosor de la dermis en regiones que no han sufrido quemaduras y las comparamos con áreas similares desbridadas. Resultados Todas las US de piel demostraron menor grosor dérmico en las áreas desbridadas que en las zonas no quemadas del mismo paciente. A pesar de que en todos los casos el diagnóstico visual del lecho fue similar a una escara, en un caso la US demostró la práctica ausencia de dermis y en otro persistencia de la escara. A los pocos días, ambos pacientes necesitaron injertos de piel. Todos los casos con suficiente dermis remanente curaron por epitelización espontánea. Conclusiones La evaluación del lecho de la herida con ultrasonografía tras desbridamiento enzimático, es un método objetivo para valorar la respuesta al tratamiento. Asimismo, la medición de la dermis remanente podría proporcionar parámetros reales para predecir las oportunidades de epitelización espontánea o la necesidad de injertos de piel en pacientes quemados.

Abstract: The self-assembly of the two anomeric forms of n-hexadecyl-d-maltopyranoside (denoted α-C16G2 and β-C16G2) has been studied in dilute aqueous solution by means of surface tension measurements, scattering methods (dynamic light scattering, static light scattering, and small-angle X-ray and neutron scattering), and cryo-transmission electron microscopy at different surfactant concentrations and temperatures. Surface tension measurements demonstrate differences in the surfactant adsorption at the air–water interface, where α-C16G2 shows a lower CMC than β-C16G2. Similarly, micelle morphology was found to profoundly depend on anomerism. β-C16G2 preferentially forms very elongated micelles with large persistence lengths, whereas α-C16G2 assembles into smaller micelles for which the structure varies with concentration and temperature. The differences between the two surfactant anomers in terms of self-assembly can be attributed to the interaction between neighboring headgroups. Specifically, β-C16G2 allows for a closer packing in the palisade layer, hence reducing the micelle curvature and promoting the formation of more elongated micelles. Strong intermolecular headgroup interactions may also account for the observed rigidity of the micelles.

Abstract: Micellisation of surfactants in deep eutectic solvents has been recently demonstrated to provide a controllable way to modify micelle morphology. Ion-pair interactions between the solvent and the surfactant headgroup were identified as affecting the micellisation by modifying the charge density of the micelle. Here we explore the micellisation of dodecylsulfate surfactants with different counterions (Li+, Cs+, Mg2+, Bmim+, Emim+, cholinium+) dissolved in two deep eutectic solvents: choline chloride:urea and choline chloride:glycerol. Surface tension results show a solvent and counterion dependence of the CMC of the surfactants. Small-angle neutron scattering was subsequently used to investigate the morphology of the micelles formed. The results show that the elongation of the micelles is strongly dependent on the solvent, showing more elongated aggregates in choline chloride:urea than in choline chloride:glycerol. The morphology of micelles in DES was also found to depend on the counterion, where the affinity of binding showed similarities to that in water.

Abstract: In recent years many studies into green solvents have been undertaken and deep eutectic solvents (DES) have emerged as sustainable and green alternatives to conventional solvents since they may be formed from cheap non-toxic organic precursors. In this study we examine amphiphile behaviour in these novel media to test our understanding of amphiphile self-assembly within environments that have an intermediate polarity between polar and non-polar extremes. We have built on our recently published results to present a more detailed structural characterisation of micelles of sodium dodecylsulfate (SDS) within the eutectic mixture of choline chloride and urea. Here we show that SDS adopts an unusual cylindrical aggregate morphology, unlike that seen in water and other polar solvents. A new morphology transition to shorter aggregates was found with increasing concentration. The self-assembly of SDS was also investigated in the presence of water; which promotes the formation of shorter aggregates.

Abstract: Deep eutectic solvents have shown the ability to promote the self-assembly of surfactants in solution. However, some differences have been found compared with self-assembly in pure water and other polar organic solvents. The behaviour of alkyltrimethylammonium bromides in choline chloride:glycerol deep eutectic solvent has been studied by means of surface tension, X-ray and neutron reflectivity and smallangle neutron scattering. The surfactants were found to remain surface active and showed comparable critical micelle concentrations to the same surfactants in water. Our scattering studies demonstrate that these surfactants form globular micelles with ellipsoidal shape in solution. The size, shape and aggregation number of the aggregates were found to vary with the chain length of the surfactant. Specific solventheadgroup interactions were not found in this system, unlike those we have previously postulated for anionic surfactants in choline chloride deep eutectic solvents.

Abstract: Abstract Sarcomas constitute a heterogeneous group of rare and difficult-to-treat tumors that can affect people of all ages, representing one of the most common forms of cancer in childhood and adolescence. Little is known about the molecular entities involved in sarcomagenesis. Therefore, the identification of processes that lead to the development of the disease may uncover novel therapeutic opportunities. Here, we show that the MEK5/ERK5 signaling pathway plays a critical role in the pathogenesis of sarcomas. By developing a mouse model engineered to express a constitutively active form of MEK5, we demonstrate that the exclusive activation of the MEK5/ERK5 pathway can promote sarcomagenesis. Histopathological analyses identified these tumors as undifferentiated pleomorphic sarcomas. Bioinformatic studies revealed that sarcomas are the tumors in which ERK5 is most frequently amplified and overexpressed. Moreover, analysis of the impact of ERK5 protein expression on overall survival in patients diagnosed with different sarcoma types in our local hospital showed a 5-fold decrease in median survival in patients with elevated ERK5 expression compared with those with low expression. Pharmacological and genetic studies revealed that targeting the MEK5/ERK5 pathway drastically affects the proliferation of human sarcoma cells and tumor growth. Interestingly, sarcoma cells with knockout of ERK5 or MEK5 were unable to form tumors when engrafted into mice. Taken together, our results reveal a role of the MEK5/ERK5 pathway in sarcomagenesis and open a new scenario to be considered in the treatment of patients with sarcoma in which the ERK5 pathway is pathophysiologically involved.

Abstract: This study presents a novel expansion of the clay-fraction (CF)/resistivity clustering method aiming at developing realizations of subsurface structures based on multiple point statistics (MPS). The CF-resistivity clustering method is used to define a data driven training image (TI) for MPS simulations. By combining this TI with uncertainty estimates obtained from correlation between the resistivity models and the unique categories in the TI, subsurface realizations are generated honoring geophysical and lithological data. The generated subsurface realizations were calibrated in a steady state groundwater model. Forecasts of well catchment zones were derived based on two wells located in areas with different levels of structural uncertainty. The catchment probability maps derived from the structural realizations were compared with the well catchment forecasted by a deterministic subsurface structure, and we are able to capture this catchment within the estimated uncertainties. We believe that this study is the first to combine MPS methods with a complete data driven workflow going directly from lithological and geophysical data to realizations of the subsurface structures. The main benefits of this is that it is data driven, fast, reproducible, and transparent.

Abstract: Summary: Platform trials bring the promise of making clinical research more efficient and more patient centric. While their use has become more widespread, including their prominent role during the COVID-19 pandemic response, broader adoption of platform trials has been limited by the lack of experience and tools to navigate the critical upfront planning required to launch such collaborative studies. The European Union-Patient-cEntric clinicAl tRial pLatform (EU-PEARL) initiative has produced new methodologies to expand the use of platform trials with an overarching infrastructure and services embedded into Integrated Research Platforms (IRPs), in collaboration with patient representatives and through consultation with U.S. Food and Drug Administration and European Medicines Agency stakeholders. In this narrative review, we discuss the outlook for platform trials in Europe, including challenges related to infrastructure, design, adaptations, data sharing and regulation. Documents derived from the EU-PEARL project, alongside a literature search including PubMed and relevant grey literature (e.g., guidance from regulatory agencies and health technology agencies) were used as sources for a multi-stage collaborative process through which the 10 more important points based on lessons drawn from the EU-PEARL project were developed and summarised as guidance for the setup of platform trials. We conclude that early involvement of critical stakeholder such as regulatory agencies or patients are critical steps in the implementation and later acceptance of platform trials. Addressing these gaps will be critical for attaining the full potential of platform trials for patients. Abstract: Recent investigations have shown that deep eutectic solvents provide a suitable environment for self-organisation of biomolecules, in particular phospholipids and proteins. However, the solvation of complex lyophilic moieties by deep eutectic solvents still remains unclear. Here we explore the behaviour of zwitterionic surfactants in choline chloride:glycerol eutectic mixture. Dodecyl-2-(trimethylammonio)ethylphosphate and N-alkyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (alkyl = dodecyl, tetradecyl) surfactants were investigated by means of surface tension, X-ray reflectivity and small-angle neutron scattering. These surfactants were found to remain surface active and form globular micelles in deep eutectic solvents. Still, the surface behaviour of these species was found to differ depending on the headgroup and tail structure. The morphology of the micelles also slightly varies between surfactants, demonstrating differences in the packing of individual monomers. The characteristics of mixtures of the dodecyl surfactants is also reported, showing a deviation from ideal mixing associated with attractive interactions between sulfobetaine and phosphocholine headgroups. Such non-ideality results in variation of the surface behaviour and self-assembly of these surfactant mixtures. The results presented here will potentially lead to the development of new alternatives for drug-delivery, protein solubilisation and biosensing through a better fundamental understanding of the behaviour of zwitterionic surfactants in deep eutectic solvents.

Abstract: Deep eutectic solvents have been demonstrated to support amphiphile self-assembly, providing potential alternatives as structure-directing agents in the synthesis of nanostructures, and drug delivery. Here we have expanded on this recent research to investigate the self-assembly of alkyltrimethylammonium bromide surfactants in choline chloride:malonic acid deep eutectic solvent and mixtures of the solvent with water. Surface tension and small-angle neutron scattering were used to determine the behavior of the amphiphiles. Surfactants were found to remain active in the solvent, and surface tension measurements revealed changes in the behavior of the surfactants with different levels of hydration. Small-angle neutron scattering shows that in this solvent the micelle shape depends on the surfactant chain length, varying from globular micelles (aspect ratio ∼2) for short chain surfactants to elongated micelles (aspect ratio ∼14) for long chain surfactants even at low surfactant concentration. We suggest that the formation of elongated micelles can be explained through the interaction of the solvent with the surfactant headgroup, since ion-ion interactions between surfactant headgroups and solvent may modify the morphology of the micelles. The presence of water in the deep eutectic solvents promotes an increase in the charge density at the micelle interface and therefore the formation of less elongated, globular micelles.

84. Resilience of Malic Acid Natural Deep Eutectic Solvent Nanostructure to Solidification and Hydration

Abstract: Little is presently known about the unique nanostructure of deep eutectic solvents (DES). The order of the liquid-solid phase transition is contended and whether DES-water mixtures are merely aqueous solutions, or have properties dominated by the eutectic pair, is unclear. Here, we unambiguously show the structure of choline chloride-malic acid (malicine) as a liquid, and also in solid and hydrated forms, using neutron total scattering on D/H isotope-substituted samples, and quasi-elastic neutron scattering (QENS). Data were refined using empirical potential structure refinement. We show evidence for a stoichiometric complex ion cluster in the disordered liquid, with strong choline-chloride bonding and a hydrogen bond donor (HBD) contribution. The 1:1 eutectic stoichiometry makes these ionic domains more well-defined, with less HBD clustering than seen previously for reline. There is minimal structural difference for the solidified material, demonstrating that this DES solidification is a glass transition rather than a first order phase change. QENS data support this by showing a gradual change in solvent dynamics rather than a step change. The DES structure is mostly retained upon hydration, with water acting both as a secondary smaller HBD at closer range to choline than malic acid, and forming transient wormlike aggregates. This new understanding of DES structure will aid understanding of the properties of these novel green solvents on the molecular length scale in chemical processes, as well as giving an insight into the apparent role of natural DESs in plant physiology.

Abstract: On a global scale, sewage represents the main point-source of water pollution and is also the predominant source of nitrogen contamination in urban regions. The present research is focused on the study of the main challenges that need to be addressed in order to achieve a successful inorganic nitrogen post-treatment of anaerobic effluents in the mainstream. The post-treatment is based on autotrophic nitrogen removal. The challenges are classified in terms of operational features and system configuration, namely: (i) the short-term effects of organic carbon source, the COD/N ratio and the temperature on the autotrophic nitrogen removal; the results from this study confirms that the Anammox activity is strongly influenced by temperature, in spite of the COD source and COD/N ratios applied. (ii) The long-term performance of the Anammox process under low nitrogen sludge loading rate (NSLR) and moderate to low temperatures; it demonstrates that NSLR affects nitrogen removal efficiency, granular size and biomass concentration of the bioreactor. (iii) The Anammox cultivation in a closed sponge-bed trickling filter (CSTF) and (iv) the autotrophic nitrogen removal over nitrite in a sponge-bed trickling filter (STF). Both types of Anammox sponge-bed trickling filters offer a plane technology with good nitrogen removal efficiency.

88. A dataset of human body tracking of walking actions captured using two Azure Kinect sensors

Abstract: A dataset of body tracking information is presented. The dataset consists of 315 captured walking sequences. Each sequence is simultaneously captured by two Azure Kinect devices. The two captures are interleaved to effectively double the frame rate. Fifteen participants partook in this experiment. Each experiment consists of seven walking actions, and having three predefined trajectories per experiment. That results in 21 sequences per participant. The data were collected using the Azure Kinect Sensor SDK. They were later processed using the official tools and libraries provided by Microsoft. For each sequence and trajectory, the positions and orientations of thirty-two tracked joints were obtained and saved.The dataset is structured as follows. The experiments from each subject are saved in a single directory. Each directory contains multiple JSON files of timestamped body tracking information to enable the fusion of the two device streams. A calibration file is also provided, enabling the mapping of the coordinates between the two Azure Kinect devices capturing the data (mapping the coordinates of the device known as the Subordinate device to the Master device coordinate system). This data can be used to train neural networks for human motion prediction tasks or test pre-existing algorithms on Azure Kinect data. This dataset could also aid in gait recognition and analysis, as well as in performing action recognition and other surveillance activities. Abstract: Male pattern hair loss (MPHL) is exceedingly common. It is characterised by onset in early adulthood and progression with age. It has a strong heritable component. The reason for its existence remains unexplained. Given that MPHL is progressive and has its earliest manifestations in young adults it may be a barometer of age. Here we suggest that MPHL may have atavistically allowed women in our species and ancestor species to select younger (but not necessarily the youngest) adult mates. Evidence suggests that conceptions by younger fathers are more likely to lead to live births and less likely to result in miscarriage. Further children fathered by younger men may have improved health and be less likely to suffer from a number of co-morbidities. This is collectively known as the “paternal age affect”. Hence the selection of younger males mediated by the MPHL may improve the fitness of the population and of the species at the expense of the individual. Indeed MPHL may have been an evolutionary “nudge” directing women to favour younger partners. It is conceivable that for a species whose success is predicated upon co-operation, collaboration and altruism the gene cannot be exclusively selfish and must have a selfless allele.

Abstract: In the rapidly evolving domain of conversational agents, the integration of Large Language Models (LLMs) into Chatbot Development Platforms (CDPs) is a significant innovation. This study compares the efficacy of employing generic and fine-tuned GPT-3.5-turbo models for designing dialog flows, focusing on the intent and entity recognition crucial for dynamic conversational interactions. Two distinct approaches are introduced: a generic GPT-based system (G-GPT) leveraging the pre-trained model with complex prompts for intent and entity detection, and a fine-tuned GPT-based system (FT-GPT) employing customized models for enhanced specificity and efficiency. The evaluation encompassed the systems’ ability to accurately classify intents and recognize named entities, contrasting their adaptability, operational efficiency, and customization capabilities. The results revealed that, while the G-GPT system offers ease of deployment and versatility across various contexts, the FT-GPT system demonstrates superior precision, efficiency, and customization, although it requires initial training and dataset preparation. This research highlights the versatility of LLMs in enriching conversational features for talking assistants, from social robots to interactive chatbots. By tailoring these advanced models, the fluidity and responsiveness of conversational agents can be enhanced, making them more adaptable and effective in a variety of settings, from customer service to interactive learning environments.

Abstract: Nowadays, the need to monitor different physical variables is constantly increasing and can be used in different applications, from humidity monitoring to disease detection in living beings, using a local or wireless sensor network (WSN). The Internet of Things has become a valuable approach to climate monitoring, daily parcel monitoring, early disease detection, crop plant counting, and risk assessment. Herein, an autonomous energy wireless sensor network for monitoring environmental variables is proposed. The network’s tree topology configuration, which involves master and slave modules, is managed by microcontrollers embedded with sensors, constituting a key part of the WSN architecture. The system’s slave modules are equipped with sensors for temperature, humidity, gas, and light detection, along with a photovoltaic cell to energize the system, and a WiFi module for data transmission. The receiver incorporates a user interface and the necessary computing components for efficient data handling. In an open-field configuration, the transceiver range of the proposed system reaches up to 750 m per module. The advantages of this approach are its scalability, energy efficiency, and the system’s ability to provide real-time environmental monitoring over a large area, which is particularly beneficial for applications in precision agriculture and environmental management.

Abstract: With the evolving demographics in the U.S. population, Latinos are the largest racial/ethnic community in the United States and face many barriers to psychological well-being and overall health. Some Latinos present with culturally influenced idioms of distress such as ataque de nervios. Associations between ataque de nervios and a range of social and psychiatric vulnerabilities among Latinos living on the United States mainland have been found. An integrative, trauma-informed, and culturally sensitive psychodynamic perspective can help with understanding and treating ataque de nervios. This case study presents the treatment of a young undocumented Latina mother experiencing ataque de nervios. A psychodynamic approach, integrated with narrative therapy and cognitive-behavioral therapy techniques, is illustrated.

Abstract: The Asian citrus psyllid, Diaphorina citri (Hemiptera: Psyllidae), is an invasive pest of citrus in the United States. The psyllid feeds and reproduces primarily on new flush growth of citrus and other rutaceous plants. Because it vectors the bacterial causal agents of the deadly citrus greening disease, D. citri is potentially a pest of economic importance in all citrus growing areas where it occurs together with the disease. We investigated the diurnal patterns of its flight activity in the field and the effects of light on its host selection and egg laying behaviors. The numbers of adult psyllids caught on yellow sticky traps were 3 to 4-fold higher during daytime than nighttime. Daytime flight activity of D. citri adults also varied with time of the day with peak catches occurring at midday from 1200 to 1500 h. Illumination of the traps at night increased their attractiveness to adult psyllids by 5- fold. Similarly, light significantly increased plant colonization by adults and female egg deposition on potted plants in the laboratory. These results showed that the flight activity and host selection behavior of adult psyllids are regulated by light and circadian rhythms. Thus, adult psyllids utilize light as visual cues in their host-plant selection process.

Abstract: Introduction: Patients with pulmonary tuberculosis (PTB) disease and positive sputum cultures are the main source of infection. Culture conversion time is inconsistent and defining the length of respiratory isolation is challenging. The objective of this study is to develop a score to predict the length of isolation period. Methods: A retrospective study was carried out to evaluated risk factors associated with persistent positive sputum cultures after 4 weeks of treatment in 229 patients with PTB. A multivariable logistic regression model was used to determinate predictors for positive culture and a scoring system was created based on the coefficients of the final model. Results: Sputum culture was persistently positive in 40.6%. Fever at consultation (1.87, 95% CI:1.02–3.41), smoking (2.44, 95% CI:1.36–4.37), >2 affected lung lobes (1.95, 95% CI:1.08–3.54), and neutrophil-to-lymphocyte ratio > 3.5 (2.22, 95% CI:1.24–3.99), were significantly associated with delayed culture conversion. Therefore, we assembled a severity score that achieved an area under the curve of 0.71 (95% CI:0.64–0.78). Conclusions: In patients with smear positive PTB, a score with clinical, radiological and analytical parameters can be used as a supplemental tool to assist clinical decisions in isolation period.

Abstract: Deep eutectic solvents (DES) resemble ionic liquids but are formed from an ionic mixture instead of being a single ionic compound. Here we present some results that demonstrate that surfactant sodium dodecyl sulfate (SDS) remains surface-active and shows self-assembly phenomena in the most commonly studied DES, choline chloride/urea. X-ray reflectivity (XRR) and small angle neutron scattering (SANS) suggest that the behavior is significantly different from that in water. Our SANS data supports our determination of the critical micelle concentration using surface-tension measurements and suggests that the micelles formed in DES do not have the same shape and size as those seen in water. Reflectivity measurements have also demonstrated that the surfactants remain surface-active below this concentration.

Abstract: Vacuum carbonization of organic precursors usually causes considerable structural damage and collapse of morphological features. However, for a polymer with intrinsic microporosity (PIM-EA-TB with a Brunauer-Emmet-Teller (BET) surface area of 1027 m(2)g(-1)), it is shown here that the rigidity of the molecular backbone is retained even during 500 °C vacuum carbonization, yielding a novel type of microporous heterocarbon (either as powder or as thin film membrane) with properties between those of a conducting polymer and those of a carbon. After carbonization, the scanning electron microscopy (SEM) morphology and the small-angle X-ray scattering (SAXS) Guinier radius remain largely unchanged as does the cumulative pore volume. However, the BET surface area is decreased to 242 m(2)g(-1), but microporosity is considerably increased. The new material is shown to exhibit noticeable electrochemical features including two pH-dependent capacitance domains switching from ca. 33 Fg(-1) (when oxidized) to ca. 147 Fg(-1) (when reduced), a low electron transfer reactivity toward oxygen and hydrogen peroxide, and a four-point-probe resistivity (dry) of approximately 40 MΩ/square for a 1-2 μm thick film.
