Within class II bacteriocins, we assume the presence of a separate subfamily of antimicrobial peptides possessing a broad spectrum of antimicrobial activity. Although these peptides are structurally related to the subclass IIa (pediocin-like) bacteriocins, they have significant differences in biological activities and, probably, a mechanism of their antimicrobial action. A representative of this subfamily is acidocin A from Lactobacillus acidophilus TK9201. We discovered the similarity between acidocin A and acidocin 8912 from Lactobacillus acidophilus TK8912 when analyzing plasmids from lactic acid bacteria and suggested the presence of a single evolutionary predecessor of these peptides. We obtained the C-terminally extended homolog of acidocin 8912, named acidocin 8912A, a possible intermediate form in the evolution of the former. The study of secondary structures and biological activities of these peptides showed their structural similarity to acidocin A; however, the antimicrobial activities of acidocin 8912 and acidocin 8912A were lower than that of acidocin A. In addition, these peptides demonstrated stronger cytotoxic and membranotropic effects. Building upon what we previously discovered about the immunomodulatory properties of acidocin A, we studied its proteolytic stability under conditions simulating those in the digestive tract and also assessed its ability to permeate intestinal epithelium using the Caco-2 cells monolayer model. In addition, we found a pronounced effect of acidocin A against fungi of the genus Candida, which might also expand the therapeutic potential of this bacterial antimicrobial peptide. In the Reserva Ecológica del Pedregal of San Ángel, located in the south of Mexico City, Mexico, free-roaming dogs and cats coexist with 148 bird, 33 of mammal, 23 of reptile and seven amphibian species, that represent a remnant of the original fauna of the Mexican Plateau. The negative impact that dogs and cats have on local fauna is unobjectionable, however, the role that these introduced vertebrates play as potential transmitters of infectious diseases for native fauna and humans, is much less understood. Information about parasitic infections in native and introduced animals in this location is scarce. In order to ameliorate this lack of information, the objective of this study is to characterize the helminth fauna of the free-ranging dogs and cats of the ecological reserve. Between 2018 and 2023, 36 Felis silvestris catus and 7 Canis lupus familiaris were studied from the helminthological perspective. Endoparasites were obtained from the digestive tract and were identified to the species level using morphological and molecular evidence. Hosts were parasitized by eight species of helminths: in cats the cestodes Hydatigera taeniaeformis, Mesocestoides sp., Taenia rileyi and the nematode Toxocara cati were recorded, while in dogs, the cestode Taenia pisiformis and the nematodes Ancylostoma caninum, and Uncinaria stenocephala were found. The only species shared between cats and dogs was the cestode Dipylidium caninum. These free-ranging animals act as definitive hosts of 5 species known to have zoonotic potential; their presence in the area may generate a public and animal health problem if programs of dog and cat population control are not continued. Cruciferous vegetables represent a valuable source of bioactive compounds. However, there is currently a deficiency of information regarding the extent to which these compounds remain bioaccessible to the body following thermal treatment and digestion processes within the digestive tract. Accordingly, the aim of this study was to evaluate the impact of heat treatment and in vitro digestion on the level of selected bioactive compounds in Brussels sprouts. The Brussels sprouts samples were subjected to a range of thermal processing techniques, which were then followed by a simulated in vitro digestion. The investigated compounds were analyzed using UV-Vis spectrometry and liquid chromatography coupled with mass spectrometry (LC-MS). The findings revealed that the sous-vide method of cooking Brussels sprouts resulted in significantly higher losses of glucosinolates (GLS) in comparison to conventional cooking methods. No significant differences were observed with regard to isothiocyanates and indoles. The analysis of GLS following digestion revealed that the process was more effective after sous vide and traditional cooking, and slightly less effective after steam cooking. With regard to individual compounds, glucoraphanin (GRA), glucoraphenin (GIV), and gluconasturtiin (GNS) were found to be completely degraded, whereas methoxyglucobrassicin (metGBS) was the most resistant to digestion in both the sous vide and steamed Brussels sprouts. The results indicated that the process of simulating digestion had no significant impact on isothiocyanates and indoles. Background: The association between education, intelligence, and cognition with digestive tract diseases has been established. However, the specific contribution of each factor in the pathogenesis of these diseases are still uncertain. Method: This study employed multivariable Mendelian randomization (MR) to assess the independent effects of education, intelligence, and cognition on gastrointestinal conditions in the FinnGen and UK Biobank European-ancestry populations. A two-step MR approach was employed to assess the mediating effects of the association. Results: Meta-analysis of MR estimates from FinnGen and UK Biobank showed that 1- SD (4.2 years) higher education was causally associated with lower risks of gastroesophageal reflux (OR: 0.58; 95% CI: 0.50, 0.66), peptic ulcer (OR: 0.57; 95% CI: 0.47, 0.69), irritable bowel syndrome (OR: 0.70; 95% CI: 0.56, 0.87), diverticular disease (OR: 0.69; 95% CI: 0.61, 0.78), cholelithiasis (OR: 0.68; 95% CI: 0.59, 0.79) and acute pancreatitis (OR: 0.54; 95% CI: 0.41, 0.72), independently of intelligence and cognition. These causal associations were mediating by body mass index (3.7-22.3%), waist-to-hip ratio (8.3-11.9%), body fat percentage (4.1-39.8%), fasting insulin (1.4-5.5%) and major depression (6.0-12.4%). Conclusion: Our findings demonstrate a causal and independent association between education and six common digestive tract diseases. Introduction: Postpartum period of dairy cattle is an important phase of their life mainly associated with the changes in physiology, rumen function, and energy metabolism. Studies have shown that gut microbial composition undergoes drastic changes during the postpartum period. However, little is known about the temporal variations in digestive tract microbiota in postpartum Tibetan cattle. The aim of this study was to investigate the temporal variations in blood metabolites, ruminal fermentation, and microbial community of oral, rumen, and gut in lactating Tibetan cattle during postpartum. Methods: We collected blood, saliva, rumen fluid, and fecal samples from lactating Tibetan cattle during 1st week (1 W), the 2nd week (2 W), the 1st month (1 M), and the 2nd month (2 M) of the postpartum period. The microbiota of saliva, rumen fluid, and fecal samples were assessed using 16S rRNA sequencing. The rumen volatile fatty acid and blood parameters were also quantified. Results: The content of volatile fatty acids (VFAs) and blood parameters showed opposite tendency to each other and reached to stability at 2 M. Rumen microbiota showed the highest alpha diversity compared to other two sites. At phylum level, the oral cavity was dominated by Proteobacteria, while most dominant phylum in rumen and feces were Firmicutes and Bacteroidetes, respectively. The dominant genera in oral cavity were Moraxella and Bibersteinia, while genera Prevotella 1 and Ruminococcaceae UCG-005 were dominant in rumen and fecal samples, respectively. Discussion: Microbial network analysis revealed that most of the active genera in all networks belonged to phylum Firmicutes, indicating the importance of this phyla during postpartum period of lactating cattle. The functional analysis revealed distinct division of labor among three gastrointestinal sites associated with defense, fatty acid synthesis, and maintaining health of host. Background: Evaluation of gut motility in clinical practice is currently limited. A novel medical system (MoPill™) consisting of a capsule that wirelessly transmits radiofrequency signals to assess motility via 3D location, was used to conduct this study. The objectives were to: (1) confirm the safety of the MoPill™ system; (2) compare the 3D location transmitted by the capsule to its location captured by abdominal x‐rays; 3 determine gastric emptying (GE), whole gut transit time (WGTT) and segmental transit times. Methods: The MoPill™ system consists of an electronic capsule (2 × 1.2 cm), eight color‐coded adhesive sensors (6 × 5.5 cm), a recorder (15 × 11 × 2 cm), and software on a laptop. Four sensors were applied to the abdomen and four to the back. Healthy subjects who had fasted overnight ingested a 250‐calorie protein bar, 17 oz. of water, followed by an activated capsule. No further caloric contents were permitted for the next 5 h. At 1, 5, and 24 h (if the capsule had not been expelled), upright abdominal X‐rays (AP and lateral) were obtained to assess the location of the capsule, which was compared to the gastrointestinal positioning system (GPS) location determined by the MoPill™ system. Identification of the capsule's anatomical location by the MoPill™ system was based on (1) the 3D (x, y, z) location; (2) time; (3) trajectory (e.g., going up the right side of the body signified ascending colon); (4) frequency of contractions (e.g., 3 cycles/min for the stomach); and (5) milestone relationship (e.g., pyloric passage must follow the end of gastric contractions). GE was determined first by the end of the 3 cycles/min rhythmic movement of the stomach and then again by pyloric expulsion on 3D location. Small intestine transit was taken as the duration from pyloric expulsion to arrival in the cecum. Colon transit time was determined by calculating the duration from 3D arrival in the cecum to passage of the capsule out of the body (i.e., loss of signal accompanying a bowel movement). Key Results: Ten healthy subjects (five women; mean age 34; mean BMI 24) were enrolled, and nine provided reliable data. The variation between the x‐ray and the estimated (i.e., identified by the MoPill™ system) location of the capsule was within an average of 3.5 cm (range 0.9–9.4 cm). The mean GE was 3.1 h. The small intestine's mean transit time was 4.3 h. The mean colonic transit time was 17.6 h. There were no adverse events recorded during the study. Conclusions & Inferences: MoPill™ is a novel gastrointestinal positional system that accurately identifies the location of a capsule compared to an X‐ray. MoPill™ system also recognizes GE, small bowel, colonic, and WGTT as well as segmental gut location and movement characteristics. In this study, we utilized the larvae of Cheumatopsyche sp. (Insecta, Trichoptera, Hydropsychidae) as a key component to assess heavy metal contamination in the waters of the Bone River, Gorontalo, Indonesia. To measure the concentrations of heavy metals, specifically lead (Pb), cadmium (Cd), and mercury (Hg), an atomic absorption spectroscopy (AAS) was employed on both river sediments and the entire body of the Cheumatopsyche sp. larvae. Additionally, analysis was conducted using the SEM-EDX method to confirm the absorption of heavy metals in the larvae, particularly in the digestive tract. Sediment and larvae samples were collected from five sampling points along the main river, including reference sites in the upstream area. Our findings revealed that Hg and Cd levels were lower in the sediment compared to those in the larvae, while Pb levels were higher in the sediment. Examination of heavy metals in the digestive tract showed morphological abnormalities in larvae from the most contaminated sites. This research confirmed that Cheumatopsyche sp. larvae effectively absorb heavy metals, particularly Hg, validating their use as bioindicators in heavy metal bioassays. [ABSTRACT FROM AUTHOR]

Meat animals harbor diverse E. coli populations in their digestive tracts and can serve as sources of pathogenic E. coli. The consumption of meat and produce contaminated with virulent E. coli from animal sources is associated with human illnesses and outbreaks. Heat treatment is an antimicrobial intervention that is commonly used during meat processing to ensure effective reductions in microbial load. Extreme heat resistance (XHR) has been reported among meat-borne E. coli and is mainly attributed to an ~15–19 kb genetic element known as the transmissible locus of stress tolerance (tLST). XHR E. coli can resist treatments used during meat processing and cooking. Therefore, the detection of heat-resistant E. coli is important for devising effective control measures to prevent meat spoilage and ensure meat safety. Here, we present methods used to (1) screen for tLST genes by multiplex PCR and (2) screen and isolate XHR E. coli from meat sources. The mode of heat exposure affects the outcome of XHR testing. Simple Summary: Mystus macropterus, a commercially significant Mystus fish species found in the Yangtze River and Pearl River, is currently experiencing a rapid decline attributed to the construction of hydraulic engineering projects, water pollution, and overfishing. Consequently, extensive artificial breeding efforts are being undertaken for large-scale commercial aquaculture purposes as well as for reintroducing this fish back into its natural habitat. Our aim was the evaluation of the effect of different opening diets on the growth, the structure of the digestive tract, and digestive enzyme activity of larval and juvenile M. macropterus. It is recommended to include Tubifex in the diet of M. macropterus larvae during the standardized farming process. One of the crucial factors influencing the growth and viability of larvae and juveniles is their opening diets. The objective of this study was to identify suitable initial feed options for M. macropterus larvae and juveniles. A total of 1200 newly hatched M. macropterus with an average weight of 18.3 mg and an average length of 11.58 mm were selected and randomly divided into four groups. The fish were fed with different opening diets, including rotifer, Artemia nauplii, Tubifex, and micro-diet from six days after hatching (dahs), respectively. Growth indices and activities of digestive enzymes were assessed at 10, 15, 20, 25, 30, 35, and 40 dahs. Histological examination of the structure of the digestive tract was performed at 40 dahs, while survival rates were also documented. The results demonstrated that different diets had no effect on the survival rate of larvae and juveniles of M. macropterus. The growth performance indices were ranked as follows: Tubifex group > Artemia nauplii group > micro-diet group > rotifer group. Remarkably, the Tubifex group exhibited superior growth performance, which was also reflected in the structure of the digestive tract and digestive enzyme activity. Simple Summary: This study tested the use of Galleria mellonella larvae as a biodegradation strategy for various types of plastics that were provided to larvae as their sole food source. The physical and enzymatic action of the larvae fragmented and consumed the plastics in various proportions, but none were digested. This was confirmed by the presence of microplastics in excreta and in larval digestive tracts, which affected their life cycle continuity by inducing the early formation of pupae and reducing the number of eggs produced. The accumulation and unsustainable management of plastic waste generate environmental pollution that affects ecosystems, wildlife, and human health. We studied the possibility of using the consumption and digestion of oxo-biodegradable, compostable plastics and polypropylene from face masks by the fifth-instar larvae of G. mellonella as a strategy for the sustainable management of plastic waste. We used Fourier transform infrared spectrophotometry (FTIR) to determine the percentage of consumption and presence of microplastics in the digestive tract and excreta for 10 treatments evaluated for 135 h. The effects of plastics on the continuity of the life cycle of the greater hive moth were also determined. We established that the larvae fragmented and consumed 35.2 ± 23% of the plastics evaluated, with significant differences between treatments. Larvae were able to consume more of the intermediate layers of masks (86.31%) than the other plastics. However, none of the plastics were digested. Instead, microplastics accumulated in the excreta, resulting in nutritional deficits that affected the continuity of the life cycle, including the induction of the early formation of pupae after 24 h and a reduction in the number of eggs laid by the females. Curcumin, a naturally occurring poorly water-soluble polyphenol with a broad spectrum, is a typical BCS IV drug. The objective of this study was to develop curcumin nanocrystals liposomes with the aim of improving bioavailability. In this study, we prepared cationic curcumin nanocrystals with a particle size of only 29.42 nm; such a phenomenal range of particle sizes is very rare. Moreover, we summarized and evaluated the parameters of the nanocrystal preparation process, including methods, formulations, etc., and the rules we concluded can be generalized to other nanocrystal preparation processes. To counteract the instability of the nanocrystals in the digestive tract, cationic curcumin nanocrystals were loaded into negatively charged liposomes through gravitational force between different charges. Unexpectedly, chitosan oligosaccharide was found to promote the self-assembly process of curcumin nanocrystal liposomes. In vitro and in vivo experiments demonstrated that chitosan-modified curcumin nanocrystal liposomes exhibited enhanced resistance to enzyme barriers, mucus barriers, and cellular barriers, resulting in a 5.4-fold increase in bioavailability compared to crude powder formulations. It can be concluded that cationic nanocrystals liposomes represent an appropriate novel strategy for improving the dissolution rate and bioavailability of poorly soluble natural products such as curcumin. With the widespread use of plastic products globally, plastic pollution is becoming more and more serious. Plastic particles formed by the breakdown of plastics have received significant attention in recent years. There are various sources of plastic particles. Plastic particles can enter the human body through respiratory tract inhalation, digestive tract intake, direct skin contact and other ways, and then affect multiple systems of the human body through oxidative stress, inflammatory response and other mechanisms. In addition, plastic particles can also be co-exposed with additives, heavy metals, bacteria, viruses and other toxic pollutants, which have an impact on the human body. This review summarizes the definition and sources of plastic particles, their entry into the human body, detection methods and their effects on various human systems. The aim of this study was to investigate the characteristics of fructophilic lactic acid bacteria isolated from fructose-rich niches (bee digestive tract, yam fruit, hazelnut leaf, grape, tomato, white mulberry, pear, kiwi) for their use as probiotics. Among a large number of isolates from these sources, 10 catalase-negative and Gram-positive bacterial strains were initially selected. Cell morphology and genotypic characteristics (RAPD-PZR, 16S rRNA sequencing) of the selected strains were determined in the first stage. The fructophilic isolates included in the study were identified as Enterococcus faecalis. Phylogenetic analyses of the related isolates were performed and isolates isolated from the bee gastrointestinal tract and isolates isolated from different plant sources were grouped into separate clades. Isolates whose phenotypic and genotypic characteristics were defined were then tested for their potential probiotic properties. In this context, acid, pepsin and pancreatin resistance profiles of the isolates were determined. In addition, isolates coded A6, 7, 9 and 10 were proven to be potential bacteriocin producers. In this study, it was shown that fructophilic lactic acid bacteria can be isolated from different plant sources and may have probiotic potential. [ABSTRACT FROM AUTHOR]

Bile acids (BAs) are cholesterol derivatives synthesized in the liver and released into the digestive tract to facilitate lipid uptake during the digestion process. Most of these BAs are reabsorbed and recycled back to the liver. Some of these BAs progress to other tissues through the bloodstream. The presence of BAs in the central nervous system (CNS) has been related to their capacity to cross the blood–brain barrier (BBB) from the systemic circulation. However, the expression of enzymes and receptors involved in their synthesis and signaling, respectively, support the hypothesis that there is an endogenous source of BAs with a specific function in the CNS. Over the last decades, BAs have been tested as treatments for many CNS pathologies, with beneficial effects. Although they were initially reported as neuroprotective substances, they are also known to reduce inflammatory processes. Most of these effects have been related to the activation of the Takeda G protein-coupled receptor 5 (TGR5). This review addresses the new challenges that face BA research for neuroscience, focusing on their molecular functions. We discuss their endogenous and exogenous sources in the CNS, their signaling through the TGR5 receptor, and their mechanisms of action as potential therapeutics for neuropathologies. Immunoglobulin A vasculitis, historically known as Henoch-Schönlein purpura, is a rare form of autoimmune-induced vasculitis most common in children. This disease is characterized by a purpuric rash, arthritis, digestive tract complication, and renal inflammation (Hopkins). We present the case of a 78-year-old man in the emergency department with findings of weakness, abdominal pain, and bloody diarrhea for 3 days and a new-onset bilateral lower extremity rash. Diagnostic imaging and labs diagnosed this patient with immunoglobulin A vasculitis (IgAV) with associated acute kidney injury and abdominal mesenteric edema. Why Should an Emergency Physician be Aware of This? Recognition of IgAV by emergency physicians and assessment of multiple organ involvement is critical to expedite treatment and minimize complications. Particularly, physicians should consider and recognize the increased severity and different presentation of IgAV in adults in comparison with the more widely known manifestation in children. Eimeria bovis is a pathogenic protozoan that causes cattle digestive tract infections, which can cause economic losses to farmers. It is necessary to develop specific and accurate detection methods to conserve livestock and prevent coccidiosis in Indonesia. This study aims to detect E. bovis by nested PCR and determine the relationship with reference sequences. A total of 167 samples of beef cattle feces were taken randomly from community farms spread across 18 provinces in Indonesia. The feces were examined natively, and then the oocysts were purified by the sugar flotation method, extracted by KIT extraction, and amplified by the nPCR technique. Positive samples were followed by sequencing and phylogenetic analysis using MEGA 11 software. This study used two pairs of primers (outer and inner) taken from ITS-1 molecular markers. As many as 96 out of 167 samples (57.5%) were positive for Eimeria spp., and 48 of the 96 samples were positive for Eimeria spp. (50%) were detected to be positive for E. bovis based on the presence of a 238 bp DNA fragment. The results of the phylogenetic analysis showed that the study sample formed a separate cluster from the E. bovis cluster from abroad. In conclusion, E. bovis was detected in 16 out of 18 provinces in this study, and the nPCR technique proved to have better sensitivity and specificity. [ABSTRACT FROM AUTHOR]

Alpha‐Gal/α‐Gal is an oligosaccharide produced by non‐primate mammals. Humans have developed an immune response mediated by anti‐α‐Gal antibodies that can trigger an allergic reaction and cause anaphylaxis. In recent years, cases of patients with delayed allergic reaction to mammalian meat have been reported worldwide. In Spain, these cases have been related to the species Ixodes ricinus L. (Ixodida: Ixodidae), whose distribution is located in the north of the country. In this work, the presence of α‐Gal in water‐soluble extracts from samples of salivary glands and digestive tracts of Hyalomma lusitanicum Koch (Ixodida: Ixodidae) both engorged and collected from vegetation were studied. The presence of that epitope was confirmed by the presence of reactive proteins of >250 kDa in both samples. The highest concentrations of α‐Gal were detected in salivary glands. Neither sex nor diet influenced the concentration of α‐Gal, which seems to indicate its endogenous production and its possible inoculation to the host during tick feeding. [ABSTRACT FROM AUTHOR]

Lecithotrophic larvae utilize extensive yolk reserves for early development. In this study, the effect of egg docosahexaenoic acid (DHA):eicosapentaenoic acid (EPA) ratios (i.e., 5.92, 10.08, 11.66, and 14.53) on the emerging larvae foregut development of rainbow trout was examined. Larvae samples were taken from day 22 to 36 post-fertilization. Thin whole body longitudinal sections were prepared and stained by Hematoxylin and Eosin and Alcian blue procedure. The sections were examined regarding epithelial layer thickness, intestinal fold height and mucosal layer thickness along with number of enterocytes and goblet cells. Results indicated that maximum thickness of the epithelium was observed on day 36 post fertilization in larvae hatched from eggs with DHA:EPA ratios of 14.53 and 10.08. The highest and lowest intestinal folds height were also observed in larvae hatched from eggs with DHA:EPA ratios of 10.08 and 14.53, respectively. The mucosal-submucosa layer thickness was the highest in larvae hatched from eggs with DHA:EPA ratio of 10.08. Enterocyte's count was the highest in larvae obtained from eggs with DHA:EPA ratio of 10.08 on day 36 post-fertilization. The highest and lowest number of goblet cells were enumerated in larvae obtained from eggs with DHA:EPA ratios of 5.53 and 14.53, respectively. In conclusion, our results revealed that feeding rainbow trout broodstock with diet contained highly unsaturated fatty acids (HUFA):polyunsaturated fatty acids (PUFA) ratio of 0.28 could result in the egg with DHA:EPA ratio of 10.08 which in turn yielded larvae with better foregut development parameters compared to those larvae emerged from the eggs with increased DHA: EPA ratio. Purpose: This study describes the composition and infection patterns of macro endoparasites associated with the lizard Phyllopezus periosus. Methods: We collected 115 P. periosus over 12 expeditions on the rocky cliffs of the Missão Velha waterfall canyon in northeastern Brazil. Specimens were collected by hand, during active searches conducted between 18:00 h and 23:30 h. The infection patterns were estimated using prevalence, total abundance, and mean intensity of infection for all parasites found. Results: A total of 4256 parasites were collected from 104 hosts, resulting in an exceptionally high overall prevalence of 90.4%. No significant ontogenetic difference in the prevalence of infection was detected. Although juveniles exhibited a high parasite prevalence their parasite loads were significantly lower compared to adults. Six species of endoparasites were found; among these, the nematode Spauligodon oxkutzcabiensis was the most common endoparasite in the digestive tract, while the pentastomid Raillietiella mottae was the only species found in the respiratory tract. Conclusions: Infection rates in Gekkota species appear to correlate with body size, with larger individuals generally exhibiting a higher prevalence. This pattern was also observed across separate populations of P. periosus and other geckos from distinct ecological domains. The dominance of the monoxenous S. oxkutzcabiensis highlights potential transmission mechanisms, suggesting that behaviors such as limb licking after feeding may contribute to its prevalence. The occurrence of Physalopteroides venancioi, Oswaldocruzia sp., Parapharyngodon sp., Physaloptera sp., infecting P. periosus are new records for this host. Gallbladder cancer (GBC) mortality in Chile is among the highest worldwide. In 2006, the Chilean government launched a programme guaranteeing access to gallbladder surgery (cholecystectomy) for patients aged 35–49 years. We evaluated the impact of this programme on digestive cancer mortality. After conducting an interrupted time series analysis of hospitalisation and mortality data from 2002 to 2018 publicly available from the Chilean Department of Health Statistics and Information, we calculated the change in the proportion of individuals without gallbladder since 10 years. We then estimated age, gender, region, and calendar‐year standardised mortality ratios (SMRs) as a function of the change in the proportion of individuals without gallbladder. The cholecystectomy rate increased by 45 operations per 100,000 persons per year (95%CI 19–72) after the introduction of the health programme. Each 1% increase in the proportion of individuals without gallbladder since 10 years was associated with a 0.73% decrease in GBC mortality (95% CI −1.05% to −0.38%), but the negative correlation was limited to women, southern Chile and age over 60. We also found decreasing mortality rates for extrahepatic bile duct, liver, oesophageal and stomach cancer with increasing proportions of individuals without gallbladder. To conclude, 12 years after its inception, the Chilean cholecystectomy programme has markedly and heterogeneously changed cholecystectomy rates. Results based on aggregate data indicate a negative correlation between the proportion of individuals without gallbladder and mortality due to gallbladder and other digestive cancers, which requires validation using individual‐level longitudinal data to reduce the potential impact of ecological bias. Resveratrol (RSV), the primary polyphenol found in grapes, has been revealed to have anti-inflammatory properties by reducing the capacity of the peripheral blood mononuclear cells to produce pro-inflammatory cytokines, including IL-1β, IL-6, IL-1ra and TNFα. Considering the close association between chronic inflammation and cancer development, RSV's immunomodulatory properties are one way by which the polyphenol may inhibit cancer initiation, proliferation, neovascularization, and migration. Resveratrol influences the generation of microtumor environment which is one of the key factors in cancer progress. In addition to immunomodulation, RSV inhibits cancer development by expressing anti-oxidant effects, causing cell cycle arrest, stimulating the function of certain enzymes, and activating cell signaling pathways. The end outcome is one of the various forms of cell death, including apoptosis, pyroptosis, necroptosis, and more, as it has been observed in vitro. RSV has been shown to act against cancer in practically every organ, while its effects on colon cancer have been documented more frequently. It is remarkable that longer-term clinical studies that may have established the potential for this natural substance to serve as a therapeutic adjuvant to traditional anti-cancer medications were not prompted by the encouraging outcomes seen with cancer cells treated with non-toxic doses of resveratrol. The current review aims to assess the recent findings about the immunological and anti-cancer characteristics of RSV, with a particular emphasis on cancers of the digestive tract, as a challenge for future clinical research that may contribute to the better prognosis of cancer. Ulcerative colitis (UC) is a chronic, recurring inflammatory condition triggered by immunological imbalances in the digestive tract, leading to weight loss, diarrhea, rectal bleeding, and an increased risk of colon cancer. Existing UC treatments encounter significant limitations, such as primary non‐responsiveness, secondary loss of efficacy, and adverse effects. This necessitates the development of drugs and drug formulations to broaden UC treatment options. This study describes the extended retention of poly(maleic anhydride)‐drug conjugates in the large intestine of a DSS‐induced acute colitis mouse model and highlights their potential for treating UC. Anti‐inflammatory sirolimus (Siro) is considered an alternative drug for UC treatment, which however also has side effects due to nonspecific systemic delivery. Accordingly, poly(malic anhydride)‐sirolimus (pSiro) is synthesized by linking Siro, a representative immunosuppressant and anti‐inflammatory drug used in clinical practice, to anhydride groups of poly(maleic anhydride) via ester bonds. In a biodistribution study, poly(maleic anhydride) increases drug retention in the large intestine. Histochemical staining reveals the reduced inflammation degree in the treatment of pSiro, which leads to the decline of systemic inflammatory markers such as plasma TNF‐α, NO, and LPS levels. Abstract Extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma) are a group of diseases with marked heterogeneity, including clinical, immunohistochemical, and molecular heterogeneity. The disease remains unspecified in the genetic landscape with only a few sequencing studies to date; however, systematic studies of alimentary canal MALT lymphoma have not been reported. To better understand the genetics of this tumor, targeted sequencing in a group of 31 cases (including 2 esophageal, 2 colonic, 4 small intestinal, and 23 gastric cases) and two cases of lymph node hyperplasiawere performed. We found epigenetic regulation (DNMT3A, KMT2D, KMT2A, EP300, TET2, etc.), signaling pathways (APC, CHD8, TNFAIP3, TNFRSF14, ZAP70, NF1,), and tumor suppressor genes (TP53, BCORL1, FOXO1, ATM, etc.) involved. Moreover, we found MTOR gene mutations in 16% of the cases that made these patients more prone to recurrence and metastasis than those with MTOR wild type genes. More interestingly, ARID2 mutations were detected in 32% of all the cases, and the mutation rate was higher and statistically significant in Helicobacter pylori (Hp)-negative patients in the gastric group. Therefore, this study found that MTOR and ARID2 gene mutations have pathogenic and prognostic implications.

Tumors present a formidable health risk with limited curability and high mortality; existing treatments face challenges in addressing the unique tumor microenvironment (hypoxia, low pH, and high permeability), necessitating the development of new therapeutic approaches. Under certain circumstances, certain bacteria, especially anaerobes or parthenogenetic anaerobes, accumulate and proliferate in the tumor environment. This phenomenon activates a series of responses in the body that ultimately produce anti-tumor effects. These bacteria can target and colonize the tumor microenvironment, promoting responses aimed at targeting and fighting tumor cells. Understanding and exploiting such interactions holds promise for innovative therapeutic strategies, potentially augmenting existing treatments and contributing to the development of more effective and targeted approaches to fighting tumors. Importance: Obesity affects approximately 19% of women and 14% of men worldwide and is associated with increased morbidity. Antiobesity medications (AOMs) modify biological processes that affect appetite and significantly improve outcomes, such as type 2 diabetes, hypertension, and dyslipidemia. Observations: AOMs should be administered in combination with lifestyle interventions and can be classified according to their mechanisms of action. Orlistat modifies digestive tract absorption and causes gastrointestinal adverse effects, such as oily fecal spotting and urgency, in more than 25% of patients. Centrally acting drugs, such as phentermine-topiramate and naltrexone-bupropion, regulate appetite in the brain and are associated with constipation in approximately 20% of patients, although the incidence of other adverse effects (eg, paresthesia, nausea) varies by medication. Nutrient-stimulated hormone-based medications, such as liraglutide, semaglutide, and tirzepatide, mimic the actions of enteropancreatic hormones that modify central appetite regulation and provide multiple cardiometabolic weight-loss benefits. Adverse effects of these drugs include nausea (28%-44%), diarrhea (21%-30%), and constipation (11%-24%). The relative potency of adult obesity medications has been studied in meta-analyses. Compared with placebo, orlistat was associated with 3.1% greater weight loss (52 randomized clinical trials [RCTs]; 16 964 participants), phentermine-topiramate was associated with 8.0% greater weight loss (5 RCTs; 3407 participants), naltrexone-bupropion was associated with 4.1% greater weight loss (6 RCTs; 9949 participants), liraglutide was associated with 4.7% greater weight loss (18 RCTs; 6321 participants), semaglutide was associated with 11.4% greater weight loss (5 RCTs; 4421 participants), and tirzepatide 15 mg was associated with 12.4% greater weight loss (6 RCTs; 1972 participants). Conclusion and Relevance: Obesity is associated with increased morbidity. Antiobesity medications are effective adjunctive therapy to lifestyle changes for improved weight loss and health outcomes. This review examines the safety and efficacy of various antiobesity medications in conjunction with lifestyle modification according to their mechanism of action, including intragastrointestinal, centrally acting, and nutrient-stimulated hormone-based medications. Background: Ingestion of foreign bodies may be seen unconsciously or intentionally in patients with mental health problems. Most cases pass through the esophagus slowly; however, in some cases, the tumor may be located in narrower areas of the digestive tract that require endoscopic or surgical intervention. This study describes a rare case of successful removal of more than 450 pieces of metal objects from the stomach of a 36-year-old man via ingestion of foreign bodies at Imam Khomeini Hospital in Ahvaz. Case presentation: A 36-year-old male patient (Aryan race) presented with complaints of chronic abdominal pain, frequent vomiting, and intolerance to liquids and food. The patient's companions mentioned a history of gradual ingestion of small metal objects 3 months prior. The patient was conscious and had stable vital signs. In the patient's X-ray and endoscopy, multiple metal objects inside the patient's stomach were observed, causing gastric outlet obstruction. The patient underwent gastrostomy surgery, and 452 screws, nuts, keys, stones, and other metal parts weighing 2900 g were removed from the stomach. Five days after the operation, the patient was transferred to the psychiatric service in good general condition and was diagnosed with psychosis, and her condition returned to normal at follow-up. Conclusion: Successful removal of this foreign body is rare. In chronic abdominal pain, especially in the context of psychiatric disorders, attention should be given to the ingestion of foreign bodies. In swallowing large amounts of sharp and metallic foreign objects, surgical intervention is necessary, especially in cases of obstruction, and saves the patient's life. [ABSTRACT FROM AUTHOR]

Cholangiocarcinoma (CCA) is a rare type of digestive tract cancer originating from the epithelial cells of the liver and biliary tract. Current treatment modalities for CCA, such as chemotherapy and radiation therapy, have demonstrated limited efficacy in enhancing survival rates. Despite the revolutionary potential of immunotherapy in cancer management, its application in CCA remains restricted due to the minimal infiltration of immune cells in these tumors, rendering them cold and unresponsive to immune checkpoint inhibitors (ICIs). Cancer cells within cold tumors deploy various mechanisms for evading immune attack, thus impeding clinical management. Recently, combination immunotherapy has become increasingly essential to comprehend the mechanisms underlying cold tumors to enhance a deficient antitumor immune response. Therefore, a thorough understanding of the knowledge on the combination immunotherapy of cold CCA is imperative to leverage the benefits of immunotherapy in treating patients. Moreover, gut microbiota plays an essential role in the immunotherapeutic responses in CCA. In this review, we summarize the current concepts of immunotherapy in CCA and clarify the intricate dynamics within the tumor immune microenvironment (TIME) of CCA. We also delve into the evasion mechanisms employed by CCA tumors against the anti-tumor immune responses. The context of combination immunotherapies in igniting cold tumors of CCA and the critical function of gut microbiota in prompting immune responses have also been annotated. Three hyperimmune egg-based formulations rich in immunoglobulin Y (IgY) were orally administered (daily, for up to 90 days) to C57BL/6 mice that were not microbially challenged. The serum levels of 32 cytokines were quantified every 30 days. Histopathology, hematology, and serum biochemistry investigations were also performed. As a sign of increased immune activity, lymphohistiocytic infiltrates were detected in the digestive tract and the liver after 30, 60, and 90 days of treatment. These infiltrates were also present in the lungs after 30 and 60 days, but not at 90 days. Blood analysis indicated systemic inflammation after 30 days of treatment: increases in pro-inflammatory cytokines, glycemia, total serum proteins, ALT, and ALP. After 60 and 90 days of treatment, the analyzed blood parameters showed mixed signs of both increased and decreased inflammation. The increased cytokines, which varied with formulation and time of exposure, indicated a combination of mostly Th17- and Th2-type immune responses. As the mice were healthy and housed in standardized sanitary conditions, and were not microbially challenged, the data were consistent with an interaction of IgY with the gut-associated lymphoid tissue as the main mechanism of action. Programmed cell death (PCD) plays a crucial role in maintaining the normal structure and function of the digestive tract in the body. Infection with Helicobacter pylori (H. pylori) is an important factor leading to gastric damage, promoting the Correa cascade and accelerating the transition from gastritis to gastric cancer. Recent research has shown that several PCD signaling pathways are abnormally activated during H. pylori infection, and the dysfunction of PCD is thought to contribute to the development of gastric cancer and interfere with treatment. With the deepening of studies on H. pylori infection in terms of PCD, exploring the interaction mechanisms between H. pylori and the body in different PCD pathways may become an important research direction for the future treatment of H. pylori infection and H. pylori-related gastric cancer. In addition, biologically active compounds that can inhibit or induce PCD may serve as key elements for the treatment of this disease. In this review, we briefly describe the process of PCD, discuss the interaction between different PCD signaling pathways and the mechanisms of H. pylori infection or H. pylori-related gastric cancer, and summarize the active molecules that may play a therapeutic role in each PCD pathway during this process, with the expectation of providing a more comprehensive understanding of the role of PCD in H. pylori infection. Alterations in seawater chemistry posed by acidification may lead to immunological and antioxidant defence impairment in sea urchins, with differences among local populations. Here, we analyzed the effects of reduced pH on Paracentrotus lividus, with a multibiomarker approach, and the possible intraspecific variations in sea urchin responses. Two groups of animals with different ecological histories (i.e., the pattern of environmental characteristics and pressures experienced throughout the organism's lifetime) were maintained at ambient pH and pH reduced of 0.4 units for 8 months. Changes in gonadosomatic index (GSI), immunological, and oxidative stress biomarkers were assessed in coelomic fluid, gonads, and digestive tract. Animals maintained at reduced pH showed limited impact of seawater acidification compared to the ambient pH condition. However, sea urchins from the two sites were differently influenced by the seawater pH (as shown by multivariate analyses). GSI and immunological and antioxidant status were differentially modulated between the two sexes, with generally higher values in females, but differences between sexes in relation to the pH of exposure were limited. Overall, our findings highlight that the impact of environmental stressors may differ in sea urchins from different locations. Botulinum neurotoxins (BoNTs) are the most potent toxins known and are used to treat an increasing number of medical disorders. All BoNTs are naturally co-expressed with a protective partner protein (NTNH) with which they form a 300 kDa complex, to resist acidic and proteolytic attack from the digestive tract. We have previously identified a new botulinum neurotoxin serotype, BoNT/X, that has unique and therapeutically attractive properties. We present the cryo-EM structure of the BoNT/X-NTNH/X complex and the crystal structure of the isolated NTNH protein. Unexpectedly, the BoNT/X complex is stable and protease-resistant at both neutral and acidic pH and disassembles only in alkaline conditions. Using the stabilizing effect of NTNH, we isolated BoNT/X and showed that it has very low potency both in vitro and in vivo. Given the high catalytic activity and translocation efficacy of BoNT/X, low activity of the full toxin is likely due to the receptor-binding domain, which presents very weak ganglioside binding and exposed hydrophobic surfaces. Adult female and male Gongylonema nematodes were found in the oesophagus of a free-living roe deer (Capreolus capreolus) in Slovenia during passive health surveillance of wildlife. The genus Gongylonema was determined by light microscopy based on the genus-specific cuticular bosses in the anterior part of the parasite. Molecular methods were used to confirm the species Gongylonema pulchrum, which has zoonotic potential. Although Gongylonema species are considered common and distributed worldwide, this is the first report of G. pulchrum in an animal on the territory of Slovenia and the first molecular report in a roe deer worldwide. The parasite is likely to be underdiagnosed, misdiagnosed or goes unnoticed as the animals show little or no clinical signs and minor pathological lesions. Slaughterhouse workers, hunters and veterinarians should be aware of this elusive parasite. Roux‐en‐Y gastric bypass (RYGB) involves creating a small stomach pouch, bypassing part of the small intestine, and rerouting the digestive tract. These alterations can potentially change the drug exposure and response. Our primary aim was to assess the impact of RYGB on the pharmacokinetics of simvastatin lactone (SV) and its active metabolite, simvastatin hydroxy acid (SVA). Ultimately, we aimed to optimize dosing for this understudied population by employing a population pharmacokinetic–pharmacodynamic link approach. The study comprised patients who had undergone RYGB surgery and individuals without a previous history of RYGB. All participants received a single oral dose of simvastatin. Plasma concentration data were analyzed with a nonlinear mixed‐effect modeling approach. A parent–metabolite model with first‐order absorption, 2‐compartments for SV and 1‐compartment for SVA, linear elimination, and enterohepatic circulation best described the data. The model was linked to the turnover pharmacodynamic model to describe the SVA inhibition on LDL‐cholesterol production. Our simulations indicated that following RYGB surgery, the exposure to SV and SVA decreased by 40%. Consequently, for low‐intensity statin patients, we recommend increasing the dose from 10 to 20 mg in post‐RYGB patients to maintain a comparable response to that of non‐operated subjects. Moderate‐intensity statin patients should require increasing doses to 40 or 60 mg or the addition of a non‐statin medication to achieve similar therapeutic outcomes. In conclusion, individuals post‐RYGB exhibit diminished exposure to SV and may benefit from increasing the dose or adjunctive therapy with non‐statin drugs to attain equivalent responses and mitigate potential adverse events. Characteristics of the digestive enzymes of the fifth instar larvae of the beet moth were determined to achieve a superior understanding of the digestive physiology of the pest. After dissecting the larval digestive tube, optimum acidity for the activity of α-amylase and α-glucosidase was obtained 8–9, and 8 for β-glucosidase, respectively. The optimum temperatures were 35 °C and 35–40 °C for α-amylase activity and for α- and β-glucosidase, respectively. The zymogram displayed the existence of three α-amylase isoforms in the digestive tract of this insect. The maximum activity of general protease was determined in the digestive tract of this pest using azocasein substrate in the pH range of 10–11. Furthermore, the optimum temperature was 35 °C for general protease. The metal ions including iron, magnesium, manganese, zinc, and copper at 5 and 10mM concentrations had a significant influence on the activity of α-amylase, α- and β-glucosidase, trypsin, chymotrypsin, and elastase enzymes. Zymogram confirmed the significant inhibitory effect of PMSF and TLCK on the activity of serine proteases, especially trypsin. The results of this research have determined to some extent the digestive system physiology and the digestive enzymes activity range in S. ocellatella. Certainly, Study on the characteristics of digestive enzymes of beet moth can be the starting point for further studies on the use of enzyme inhibitors and the possibility of preparing new methods to control this key sugar beet pest. Scalloped hammerhead sharks (Sphyrna lewini) are critically endangered, according to the International Union for Conservation of Nature Red List, likely due to anthropogenic activities such as intense fishing and pollution. Nowadays, plastic debris contamination is a subject of concern due to its extensive presence in the sea and the digestive tracts of many fish species. The possible effects of plastic debris as a vector of other pollutants are still unknown. We analyzed the digestive tract of 58 hammerhead sharks to investigate the correlation between plastic and other anthropogenic microparticle contamination and their feeding habits in the eastern region of the Gulf of California, revealing a debris contamination occurrence of 79.3%. Out of these, 91.4% corresponded to fibers, and the remaining 8.6% to fragments. The main component of the debris was cellulose (64.4%). According to their diet, these organisms exhibit benthopelagic habits, feeding both in the water column and on the seabed. These results indicate a high level of contamination of anthropogenic cellulosic microfibers in the area. Although cellulosic microfibers are recognized as a biomaterial, they can be harmful to marine species, posing an additional threat to this iconic shark. This changed according to the year, indicating that the anthropogenic microparticle ingestion is related to the discharges of human activities and their seasonality rather than to a selection process by the sharks. The passage of food through the digestive tract in an herbivorous rodent with a single-chamber semiglandular stomach, the fat sand rat Psammomys obesus, was studied by multiple marking of the food with inert plastic markers. The total mean retention time of markers in the digestive tract (DT), as well as in the stomach and caecum separately, was determined. The peculiarities of the digesta kinetics depending on the morphology of the digestive tract, diet, and the time of marker intake were clarified. The total time for removal of markers from the DT in the fat sand rat reaches 36 h. The mean retention time of markers in the DT was determined as a whole (17–18 h), as well as separately in the forestomach (7–9 h) and caecum (7–12 h). The uneven passage of feed has been revealed, which is considered as an important mechanism for maximizing the extraction of nutrients from plants, including microbial fermentation of structural carbohydrates of the food. Trichomycterus bogotensis is one of the three species of fish that inhabit the Colombian Andes. However, several aspects of its basic biology, including its digestive system, are unknown. The morphology and histology of the digestive system are of great importance for understanding a species' feeding habits and food preferences. Thus, this study aimed to perform a morphological description of the digestive tract of T. bogotensis. The gastrointestinal tract of seven specimens was processed for paraffin histology, high-resolution optical microscopy, and transmission electron microscopy. Anatomically, T. bogotensis exhibits a terminal mouth with villiform teeth, a narrow esophagus, a large and muscular stomach, and a short intestine. Histology and ultrastructure revealed a tissue organization similar to that reported for other Silurids, composed of four layers: mucosal, submucosal, muscular, and serosa. Background: Colorectal cancer (CRC) is a malignant tumor within the digestive tract with both a high incidence rate and mortality. Early detection and intervention could improve patient clinical outcomes and survival. Methods: This study computationally investigates a set of prognostic tissue and cell features from diagnostic tissue slides. With the combination of clinical prognostic variables, the pathological image features could predict the prognosis in CRC patients. Our CRC prognosis prediction pipeline sequentially consisted of three modules: (1) A MultiTissue Net to delineate outlines of different tissue types within the WSI of CRC for further ROI selection by pathologists. (2) Development of three-level quantitative image metrics related to tissue compositions, cell shape, and hidden features from a deep network. (3) Fusion of multi-level features to build a prognostic CRC model for predicting survival for CRC. Results: Experimental results suggest that each group of features has a particular relationship with the prognosis of patients in the independent test set. In the fusion features combination experiment, the accuracy rate of predicting patients' prognosis and survival status is 81.52%, and the AUC value is 0.77. Conclusion: This paper constructs a model that can predict the postoperative survival of patients by using image features and clinical information. Simple Summary: In symbiotic interactions, microorganisms associated with the intestinal tract, constituting the gut microbiome, are especially important, because they influence the life history and fitness of their host organisms. The gut microbiome of ground beetles, one of the most species-rich animal families, is poorly known, and research on sex-specific differences is almost completely missing. Investigating the gut bacterial microbiome of a widespread European carnivorous species, Carabus convexus, using next-generation high-throughput sequencing, we found 1138 different operational taxonomic units belonging to 21 bacterial phyla, 90 families, and 197 genera. One of the most abundant bacterial families and genera was previously not known from the digestive tracts of other ground beetle species. We also detected carbohydrate-degrading gut bacterial symbionts, which indicated possible plant feeding in this predatory species. Although the composition of the gut bacterial microbiome was not significantly different between the sexes, the gut symbionts showed more between-individual variability in females. This difference may result from the different mobility of sexes and the difference in reproductive investment. Biological interactions, including symbiotic ones, have vital roles in ecological and evolutionary processes. Microbial symbionts in the intestinal tracts, known as the gut microbiome, are especially important because they can fundamentally influence the life history, fitness, and competitiveness of their hosts. However, their effects on the ruminal and intestinal digestive tract have not yet been studied. Our study represents the first investigation of the effects of polyethylene terephthalate microplastics on the ability of the ruminal-gastro-intestinal system to degrade and digest mixed hay. Our findings reveal that polyethylene terephthalate significantly reduce the digestibility of crude protein in mixed hay. Notably, low levels of PET MPs negatively impacted the intestinal phase, while medium and high levels disrupted the ruminal phase. Moreover, medium and high concentrations of polyethylene terephthalate hindered the degradation of fiber fractions, specifically of neutral detergent fiber. This study aims to investigate the effects of polyethylene terephthalate (PET) MPs on the ability of the ruminal–gastro-intestinal system to degrade and digest mixed hay. Using a three-step in vitro ruminal–gastro-intestinal incubation system, PET MPs were introduced at concentrations of 0, 5, 10, and 15 g/L in ruminal and gastro-intestinal solutions. Ruminal fluid was collected from three 16-month-old Piedmontese bulls. The experiment was conducted on three mixed hays and was repeated three times, with triplicate incubations in each run. The results reveal that PET MPs reduced the degradability and digestibility of crude protein. Specifically, crude protein degradation was reduced by 9% at medium and 16% at high PET MP concentrations in the ruminal phase, while the crude protein digestibility of undegraded crude protein was reduced by 8% at the lowest PET MPs concentration in the gastro-intestinal tract. Additionally, PET MPs reduced the degradation of neutral detergent fiber at medium and high PET MP concentrations in the ruminal phase by 9% and 13%, respectively. These results highlight the risks of PET MPs contamination on ruminal–gastro-intestinal functions and underscore the urgent need to mitigate MPs contamination in the livestock sector. Bladder leiomyoma is benign neoplasm of smooth muscle and rarely reported in domestic animals. This tumor is most commonly diagnosed in the alimentary and urogenital tracts. This report describes the pathological characteristics of leiomyoma in the urinary bladder of a 4-year-old female sheep during routine necropsy. Urinary bladder showed that two sessile masses firm in consistency and well demarcated were located within the muscle layers and protruded from the mucosal layer. Bladder mucosa was intact. In histopathologic investigation, the masses were composed of densely interwoven bundles of smooth muscle with spindle cells. No malignancy changes including mitotic index, hemorrhage, or necrosis foci were detected. Based on the gross and microscopic findings, leiomyoma was diagnosed. Background Complete digestive disunion due to anastomotic necrosis is considered a contraindication to endoscopic repair. However, recent publications have suggested that endoscopic treatment by insertion of a self-expandable metal stent (SEMS) is possible. The report of this patient series aims to demonstrate the use of endoscopic management in selected cases with complete digestive disunion. Methods Seven consecutive patients with complete and circumferential upper gastrointestinal anastomotic disunion were treated at two European tertiary care centers between 2009 and 2021 by endoscopic insertion of an SEMS. Treatment was performed with a therapeutic gastroscope under general anesthesia, carbon dioxide insufflation, and fluoroscopic guidance, after surgical or percutaneous drainage. Results All patients were successfully treated by endoscopic insertion of fully or partially covered SEMS left in place for a median of 8 weeks, with a median of 3 endoscopic sessions. Digestive neo-epithelialization was associated with a restored circumferential gut lumen in all cases. The rate of stent migration was 23% and three patients (43%) experienced symptomatic strictures, which were successfully treated by endoscopic dilation. Conclusion Complete digestive rupture could be successfully treated by endoscopy in selected cases, adding proof-of-concept data regarding guided tissue regeneration alongside SEMS placement. Obesity is primarily caused by excessive intake as well as absorption of sugar and lipid. Postprandial surge in distention pressure and intestinal motility accelerates the absorption of nutrients. The response of intestinal epithelial cells to mechanical stimulation is not fully understood. Piezo1, a mechanosensitive ion channel, is widely expressed throughout the digestive tract. However, its function in intestinal nutrient absorption is not yet clear. In our study, excessive lipid deposition was observed in the duodenum of obese patients, while duodenal Piezo1–CaMKK2–AMPK α was decreased when compared to normal-weight individuals. Under high-fat diet condition, the Piezo1 iKO mice exhibited abnormally elevated sugar and lipid absorption as well as severe lipid deposition in the duodenum and liver. These phenotypes were mainly caused by the inhibition of duodenal CaMKK2–AMPK α and the upregulation of SGLT1 and DGAT2. In contrast, Yoda1, a Piezo1 agonist, was found to reduce intestinal lipid absorption in diet induced obese mice. Overexpression of Piezo1, stretch and Yoda1 inhibited lipid accumulation and the expression of DGAT2 and SGLT1, whereas knockdown of Piezo1 stimulated lipid accumulation and DGAT2 in Caco-2 cells. Our study reveals a previously unexplored mechanical regulation of nutrient absorption in intestinal epithelial cells, which may shed new light on the therapy of obesity. Linezolid is a critically important antimicrobial used in human medicine. While linezolid is not licensed for food-producing animals, the veterinary use of other antimicrobials, such as phenicols (e.g., florfenicol), could cross/co-select for linezolid-resistant (LR) bacteria. Such LR strains pose a great concern for public health due to their potential transfer between animals and humans. This study explored possible associations between epidemiological risk factors, including phenicol use, and the occurrence of LR bacteria, such as enterococci and staphylococci, in poultry, pigs, and veal calves in Belgium. Florfenicol use significantly increased the likelihood of harboring LR bacteria in veal calves, sows, and fattening pigs, particularly for the digestive tract (odds ratio (OR): [3.19–5.29]) and the respiratory tract (OR: [6.11–9.09]). LR strains from feces from fattening pigs were significantly associated with production type (OR: [3.31–44.14]) and the presence of other animal species (OR: 0.41). The occurrence of LR strains in the respiratory tract from sows was also significantly associated with using antimicrobials other than florfenicol (OR: 10.07) and purchasing animals (OR: 7.28). Our study highlights the potential risks of using certain veterinary antimicrobials, such as florfenicol, in food-producing animals and emphasizes the need for responsible antimicrobial use to safeguard both animal and public health. Gastric cancer (GC) is one of the most common malignant tumors in the digestive tract, and chemotherapy plays an irreplaceable role in the comprehensive treatment of GC. However, chemoresistance makes it difficult for patients with GC to benefit steadily from chemotherapy in the long term, which ultimately leads to tumor recurrence, metastasis, and patient death. Elucidating the detailed mechanism of chemoresistance in GC and identifying specific therapeutic targets will help to solve the difficult problem of chemoresistance and improve the prognosis of patients with GC.
